A method of testing for whether inherited deletions are a cause of a single-locus dominant disease was derived, involving analysis of the marker segregation within the pedigree of a single family that segregates for the disease. It is shown that markers can be used to test deductively for the presence of an inherited deletion. The probabilities of confirming or rejecting the presence of a deletion in an arbitrary pedigree without inbreeding are then derived. The power of the test is shown to be limited in single trios but to increase rapidly as the size of the pedigree increases. For larger pedigrees, the probabilities of confirming or rejecting a deletion are higher than 0.9 for SNPs having a minor allele frequency greater than 0.4. The probabilities are higher using multiallelic markers such as microsatellites, reaching levels as high as 0.9 in even rather small pedigrees. In certain cases the test outcome is not deductive, a deletion being neither confirmed nor rejected. It is shown to still be possible then to employ a statistical test for the presence of a deletion by use of an a priori probability for a deletion.  相似文献

Horse trailers are a common form of transportation for horses and ponies and often require the animal to travel alone or with a single companion. The current study investigated the effect of transporting horses alone, in company or with an acrylic safety mirror (measuring 81 cm × 61.5 cm) that provided surrogate companionship. The behavioural and physiological responses of 12 mature horses during a 30-min journey by trailer under the three treatments were compared. Behaviours (vocalisation, eating, head-tossing, pawing, and head-turning) were recorded. In order to assess circulatory changes that occur as part of the response to transport, heart rate (HR), rectal (Tr) and ear-pinna (Tp) temperatures were recorded. When travelling with a live companion significantly less time was spent vocalising (p < 0.001), head-turning (p < 0.001), head-tossing (p < 0.01) and pawing (p < 0.01); eating behaviour increased (p < 0.05). Physiological responses (increases in HR and Tr and decreases in Tp) were also significantly reduced when travelling with a live companion (p < 0.01). Travelling with the mirror did not significantly affect physiological responses compared with travelling alone, but the rise in Tr and fall in Tp was reduced (p = 0.052 and p = 0.051, respectively) and can be considered a trend. When travelling with a mirror significantly less time was spent turning the head (p < 0.01), vocalising (p < 0.05) and head-tossing (p < 0.05); eating behaviour increased (p < 0.05). The only significant difference between travelling with a live companion and a mirror was that the time spent turning the head round was less with a live companion (p < 0.05). The provision of surrogate companionship in the form of a mirror was found to be preferable to travel alone, but where possible a live companion is recommended. Isolation during transportation was found to suppress feeding behaviour. Although peripheral blood flow (Tp) has been used to assess transport stress in other species it has not previously been used in the horse. Further evaluation of this non-invasive measure is now required.  相似文献

Ecological dominance in ants is often fuelled by carbohydrate intake. Most studies have focused on the importance of invasive ant mutualistic associations with trophobionts whereas few studies have investigated the importance of floral nectar on invasion success. In this study, utilisation of temporarily available floral nectar by the invasive Argentine ant, Linepithema humile, was compared to that of the dominant native ant, Anoplolepis custodiens, within the Cape Floristic Region (CFR), a biodiversity hotspot. The effect of these two focal ant species on species composition and abundance of ground foraging ants as well as floral arthropod visitors in inflorescences of Proteacea species was assessed. Foraging activity, and trophic ecology inferred from the abundance of natural stable isotopes of Carbon (δ13C) and Nitrogen (δ15N), and the ratio of Carbon to Nitrogen (C:N) were compared between the two ant species during three flowering periods. Linepithema humile significantly reduced the abundance and species diversity of both above-ground and floral arthropod species abundance and composition. Linepithema humile increased its foraging activity with increasing nectar availability, switching its diet to a more herbivorous one. Anoplolepis custodiens did not respond as effectively to increasing floral nectar or negatively impact floral arthropod visitors. This study showed that the availability of floral nectar and ability of L. humile to more effectively utilise this temporarily available resource than native ants, can contribute significantly to the further spread and persistence of L. humile in natural environments in the CFR.  相似文献

In poultry, HyLine (HL) Hens are known for their excellent laying performance. However, ZhenNing (ZN) Hens, a native chicken breed in China, are known for their unique flavour. The intestinal mucosa, which is the main organ for nutrient absorption, could affect livestock product quality. In ZN Hens’ intestinal mucosa, we found more villus wrinkles, larger villus circumference and higher amino acid transporters mRNA abundance compared with HL Hens. Among three laying periods of ZN Hens, in the intestinal lumen, Lactobacillus salivarius (L. sa.), Lactobacillus agilis (L. ag.) and Lactobacillus aviarius were the predominant species in the laying peak period. Furthermore, multiple-antibiotics feeding in ZN Hens and predominant Lactobacillus feeding in HL Hens suggested that these Lactobacilli could indeed increase villus wrinkles and improve intestinal absorption. In HL Hens, L. sa. + L. ag. treatment could promote organoids budding in vitro, and promote epithelial proliferation in vivo. Collectively, the unique intestinal mucosa morphology in ZN Hens was due to the high abundance of intestinal L. sa. and L. ag. Transplant these Lactobacilli to HL Hens could increase their intestinal probiotics abundance, fine adjust the intestinal stem cell function and promote the epithelial proliferation, in turn, increase villus winkles and mucosal absorption area.  相似文献

The exposure of mammalian cells or tumors for weeks or months to low non-lethal doses of cytostatic drugs may induce multi-drug resistance, which can be enhanced by a variety of DNA-damaging agents. In yeast multi-drug resistance to a variety of drugs has been observed. DNA-damaging agents have not yet been tested. As the appearance of resistance is the result of long-term exposure, we decided to extend the application of test substances to a period of up to 400 days. In such long-term experiments S. cerevisiae MP1 adapted to treatment with low doses of mutagens. Consistent results were obtained for both alkylating and non-alkylating mutagenic substances. Furthermore, the adaptive resistance to the alkylating agent also adapted cells to the non-alkylating agent, which implies that there may be a single pathway for mutagens with different modes of action. Random spore analysis of adapted yeast cells and the back-cross to the parental wild type indicates that a single dominant mutation is responsible for the adaptive resistance.  相似文献

Tests assessing the fear of humans by measuring avoidance or approach reactions of the test animals towards humans can be useful tools in welfare assessment schemes. In this study, we wanted to compare tests assessing the reactions of laying hens towards humans in the home environment to a test performed in a novel environment. As well we investigated inter-test correlations in the home environment. We performed several tests to assess the hens’ approach and avoidance reactions towards an unfamiliar human inside the home environment of laying hens in 14 caged flocks and 10 free-range flocks. We measured the reactions of hens to a stationary person and a moving person approaching individual hens in both systems, as well as reactions to a stationary person trying to touch individual hens in non-cage systems and reactions of caged hens to a person passing by. Additionally, a standardised arena test was performed outside the home environment to compare the birds’ avoidance reactions towards an approaching human in a novel environment. In caged hens no significant correlations of tests performed inside the home environment with the arena test could be found, but there were moderate to high correlations (rs > 0.6) with one parameter of the arena test in non-caged hens. The different tests applied inside the home environment correlated significantly and moderately to highly with each other, supporting the validity of these tests to measure fear of humans. The caged laying hens in our study reacted differently in the novel environment from the non-caged birds, indicating that the birds’ reactions towards humans in an unfamiliar environment depend on the housing system. In summary, a comparison of the level of fear of humans between flocks in different housing systems by using an arena test outside the home environment seems difficult and was not possible in the present study.  相似文献

By the method of spectral-coherent analysis the dynamics was studied of successive changes in the structure the rabbit brain electrical activity coherent relations in the process of formation of motor polarization dominant created by DC anode action on the sensorimotor cortical region. It has been shown that at earlier stages, when the motor "dominant" reaction is absent, there appears an interhemispheric asymmetry in Coh spectra of electrical activity of the sensorimotor cortex and of the thalamus VPL. On the contrary, interhemispheric asymmetry in Coh spectra of electrical activity of the sensorimotor cortex and dorsal hippocampus CA3 field appears only at the stage when the motor "dominant" reaction is recorded. Asymmetry in alpha- and beta-frequencies ranges in biopotentials Coh spectra of the studied regions coinciding with the motor "dominant" reaction realization is connected with processes of movement organization.  相似文献

We explore the possibility for the native structure of a protein being inherently multiconformational in an ab initio coarse-grained model. Based on the Wang-Landau algorithm, the complete free energy landscape for the designed sequence 2DX4: INYWLAHAKAGYIVHWTA is constructed. It is shown that 2DX4 possesses two nearly degenerate native structures: one is a helix structure with the other a hairpinlike structure, and their free energy difference is <2% of that of local minima. Two degenerate native structures are stabilized by an energy barrier of ~10 kcal/mol. Furthermore, the hydrogen-bond and dipole-dipole interactions are found to be two major competing interactions in transforming one conformation into the other. Our results indicate that two degenerate native structures are stabilized by subtle balance between different interactions in proteins. In particular, for small proteins, balance between the hydrogen-bond and dipole-dipole interactions happens for proteins of sizes being ~18 amino acids and is shown to the main driving mechanism for the occurrence of degeneracy. These results provide important clues to the study of native structures of proteins.  相似文献

The benefits of dominance may not come without costs, particularly for males. For example, the “immunocompetence handicap hypothesis” states that males with enhanced mating success allocate resources to enhance reproductive output at a cost to their current health, whereas the “resource quality hypothesis” predicts that high-ranking males may benefit from increased reproduction and good health. Whereas the predictions from each have been well tested in captive animals and in a variety of highly social primates, fewer studies have been carried out in free-living, facultatively social animals. Using adult male yellow-bellied marmots (Marmota flaviventer), we evaluated predictions of these hypotheses by examining the relationship between social rank and 2 health indicators—fecal glucocorticoid metabolite (FCM) levels, and neutrophil/lymphocyte (N/L) ratios—after accounting for variation explained by age, body mass, and seasonality. We found that higher-ranking males tended to have a lower N/L ratio (reflecting good health) than lower-ranking individuals, whereas FCM levels were not significantly related to rank. In addition, heavier male marmots had lower N/L ratios, whereas body mass was not associated with FCM levels. We also found that older adult males had lower FCM levels (reflecting less physiological stress) but higher N/L ratios than younger adults. Finally, we found that FCM levels decreased as the active season progressed and FCM levels were associated with the time of the day. Overall, our results suggest that socially-dominant male marmots enjoyed better, not worse health in terms of lower N/L ratios.  相似文献

In this study, we take advantage of a natural experiment--a 2004 mass die-off of the Common Murre in Alaska to determine whether closely related mtDNA haplotypes differ in their probability of being eliminated during such a short term but a marked event removing hundreds of thousands of individuals. We sequenced complete mtDNA ND2 gene (1041 bp) for 168 Common Murres sampled from seven breeding colonies across Alaska before the 2004 die-off and 127 dead murres washed ashore during the die-off. We found little mtDNA variation and lack of geographical structuring among the seven Common Murre breeding colonies in Alaska. A comparison of the single-dominant mtDNA haplotype's frequency between live murres sampled on breeding colonies before the die-off (73.2%; 95% confidence interval 66.3-79.9%) and dead murres sampled during the die-off (59.1%; 95% confidence interval 50.4-67.4%; Fisher's exact P=0.01) showed that carriers of the dominant haplotype were significantly less likely to die than carriers of other haplotypes. At the same time, the ratio of non-synonymous to synonymous substitutions did not differ between live (10:35) and dead birds (18:34; Fisher's exact P=0.26), indicating that non-synonymous substitutions were as likely to be eliminated as synonymous substitutions. These results are consistent with the possibility of positive selection on the dominant mtDNA haplotype during the die-off.  相似文献

Habitat use and the processes which determine fish distribution were evaluated at the reef flat and reef crest zones of a tropical, algal-dominated reef. Our comparisons indicated significant differences in the majority of the evaluated environmental characteristics between zones. Also, significant differences in the abundances of twelve, from thirteen analyzed species, were observed within and between-sites. According to null models, non-random patterns of species co-occurrences were significant, suggesting that fish guilds in both zones were non-randomly structured. Unexpectedly, structural complexity negatively affected overall species richness, but had a major positive influence on highly site-attached species such as a damselfish. Depth and substrate composition, particularly macroalgae cover, were positive determinants for the fish assemblage structure in the studied reef, prevailing over factors such as structural complexity and live coral cover. Our results are conflicting with other studies carried out in coral-dominated reefs of the Caribbean and Pacific, therefore supporting the idea that the factors which may potentially influence reef fish composition are highly site-dependent and variable.  相似文献

Functional organization of neurons in rabbit's sensorimotor cortex was studied before and within several days after formation of the rhythmical dominant focus. Functional reorganization of neurons in cortical microareas took place during actualization of the dominant. The number of functional interneuronal relations within neuronal pairs of a certain type could be increased in comparison with the control values and decreased within pairs of another type. As a result, the total percent of the interneuronal correlations in cortical microareas in the control animals and rabbits with the acting dominant was approximately equal. The total percent of correlations between neurons of the adjacent cortical areas during the actualization of the dominant was significantly higher than in the control due to increased number of correlations with participation of small and medium-sized neurons. A possibility of information circulation about the "stimulus image" in the closed chain of neurons was exemplified by the real micronetwork. The data suggest the reverberation of encoded information between adjacent microareas of the sensorimotor cortex within several days after application of the stimulus, which has formed the excitation focus.  相似文献

The purpose of this study was to investigate the prevalence of dietary supplement use among adolescent athletes. The project was also directed at identifying the sources these student-athletes used for acquiring information about dietary supplements. One hundred thirty nine high school athletes (99 males; 34 females; mean age = 15.8 +/- 1.19 years) volunteered to participate in this study. A 16-question anonymous survey instrument examined use of dietary supplements, reasons for use, type of sport participation, and sources of information regarding dietary supplements. Of the participants in this study, 22.3% (N = 31) reported currently taking dietary supplements. There was no relationship found between dietary supplement use and age. There were a significantly higher number of males reporting current dietary supplement use. Of those who reported to be currently taking dietary supplements, sports performance (N = 25) was the most reported reason for use. There were no significant differences found in reported dietary supplement use between any of the sports. Of the participants, 38.1% (N = 53) listed their coach as their best source of information on dietary supplements. The results of this study offer the current literature some additional insight into trends in supplement use among high school student athletes. Practical implications suggest that it may be necessary to ensure coaches have sufficient knowledge about dietary supplements so that adolescent athletes are receiving accurate information.  相似文献

The topography of the somatosensory maps of our body can be largely shaped by alterations of peripheral sensory inputs. Following hand amputation, the hand cortical territory becomes responsive to facial cutaneous stimulation. Amputation-induced remapping, however, reverses after transplantation, as the grafted hand (re)gains its sensorimotor representation. Here, we investigate hand tactile perception in a former amputee by touching either grafted hand singly or in combination with another body part. The results showed that tactile sensitivity recovered rapidly, being remarkably good 5 months after transplant. In the right grafted hand, however, the newly acquired somatosensory awareness was strikingly hampered when the ipsilateral face was touched simultaneously, i.e., right face perception extinguished right hand perception. Ipsilateral face-hand extinction was present in the formerly dominant right hand 5 months after transplant and eventually disappeared 6 months afterwards. Control conditions' results showed that right hand tactile awareness was not extinguished either by contralateral left face and left hand stimulation or ipsilateral stimulation of the arm, which is bodily close to, but cortically far from, the hand. We suggest that ipsilateral face-hand extinction is a perceptual counterpart of the remapping that occurs after allograft and eyewitnesses the inherently competitive nature of sensory representations.  相似文献

Salt tolerance in Saccharomyces cerevisiae is a complex trait, involving regulation of membrane polarization, Na(+) efflux and sequestration of Na(+) in the vacuole. Since transmembrane transport energized by H(+)-adenosine triphosphatases (ATPases) is common to all of these tolerance mechanisms, the objective of this study was to characterize the responses of the plasma membrane H(+)-ATPase, vacuolar H(+)-ATPase and mitochondrial F(1)F(0)-ATPase to NaCl stress. We hypothesized that since the vacuolar ATPase is responsible for generating the proton motive force required for import of cations (such as Na(+)) into the vacuole, strains lacking this activity should be hypersensitive to NaCl. We found that strains lacking vacuolar ATPase activity were in fact hypersensitive to NaCl, while strains lacking ATP synthase were not. This effect was specific to the ionic component of NaCl stress, since the mutant strains were indistinguishable from wild-type and complemented strains in the presence of sorbitol.  相似文献
