Summary The gastrodermis and mesoglea of the basal disk of Hydra were investigated to conclude a three-part series of papers. The gastrodermis is composed of digestive cells (most predominant cell type), mucous and nerve cells (both immature and fully differentiated). The principal function of the digestive cells appears to be storage of protein, lipid and glycogen reserves which are utilized by neighboring cells. Mucous cells apparently use some of the reserves to synthesize their secretions which lubricate cells and prevent cell damage during egestion of waste through the aboral pore. The function of the gastrodermal nerve cells is uncertain.The mesoglea of the basal disk, contains the same structural components as seen in other regions of the polyp. It is reasonable to assume that it maintains the same function of cell adhesion and migration. As the mesoglea converges on the aboral pore, it loses its structural integrity and cells are sloughed off the column.This investigation was supported by The National Science Foundation, Grant Number GB-27395.  相似文献

Summary A variety of inclusion bodies occur in the mitochondria of several cell types of the snake, Elaphae quadrivirgata. These lie in the mitochondrial matrix or within the space of the cristae. The inclusions in the matrix are as follows: dense homogeneous and fine granular materials, structures with finger-print appearance and with filamentous and/or crystalloid pattern and fine ring-shaped and/or microtubular structures. The inclusions within mitochondrial cristae are glycogen particles, globular materials, and strand-like structures.These inclusions occur not only during the hibernation period of the snake, but also in the arousal period. Furthermore, some inclusions are encountered in fetal tissues. The functional significance of these inclusions is unknown; however, the present study suggests that they are related to the metabolic activity of the cells.  相似文献

Summary The zymogenic secretory cells of Hydra viridis are scattered between the digestive muscle cells of the gastric region. The mature zymogenic cells are located along the apical surface of the gastrodermal epithelium and contain numerous spherical secretory droplets. They appear to differentiate from stem cells located near the mesoglea. These stem cells resemble epidermal interstitial cells and are filled with free ribosomes. They differ from the interstitial cell in that they usually possess a small amount of granular endoplasmic reticulum. During the process of differentiation they elaborate a highly organized system of granular endoplasmic reticulum. This system becomes dispersed into vesicles as the secretory product is synthesized. There is no indication that the Golgi apparatus participates directly in the formation of the secretory droplets, and there is no indication of a membrane bounding the mature secretory droplet.The fate of the zymogenic cell following the discharge of its secretory product was not determined. It is possible that these cells revert back to a stage resembling the stem cell before resynthesizing a new supply of secretion. In this case the normal secretory process would be very similar to the events described in the dedifferentiation of the zymogenic cells during regeneration.This work was supported by Grant number GB-3262 from the National science Foundation.  相似文献

Using an affinity purified antibody raised against the RI-H fragment of rat intestinal lectin L-36, the latter protein has been identified within the esophageal epithelium by means of ultracryotomy followed by immunogold labeling. The epithelium consists of 4 morphologically distinct cell-types, namely, the basal, spiny, granular and squamous cells, and each of these exhibits a different immunolabeling pattern. The basal cells form a layer on the basal lamina, and in these a diffuse cytoplasmic staining is observed. This basal cell layer is overlaid by spiny cells that extend many cell processes into wide intercellular spaces. In these cells, immunogold particles are found only on small granular inclusions consisting of an electron-lucent homogeneous substance. The granular cells from a third layer over the spiny cells, and are characterized by a number of large granular inclusions with an electron-dense core rimmed by a less electron-dense substance. Immunogold labeling is found on these granules, both on the core and peripheral region. Squamous cell-types constitute the most superficial layer of the epithelium. They are without granular inclusions, and immunogold labeling is confined to the cytoplasmic surface of the thickened plasma membrane. These findings suggest that L-36 is produced in the basal cells as free cytosolic protein, then becomes progressively aggregated into the granular inclusions of the spiny and granular cells, and is eventually transferred onto the cytoplasmic surface of the squamous cell plasma membrane where it may interact with complementary glycoconjugate(s) located at this site. The membrane lining substance thus formed may play a role in stabilizing the squamous cell membranes, thereby maintaining the structural integrity of the epithelium against mechanical stress coming from the esophageal lumen.  相似文献

Summary The epidermal cell layer of the apical end of the ceras was investigated in two species of aeolid nudibranchs. Based on cellular inclusions, mostly two cell types were found: mucoid and ellipsoid-vacuolate cells. Mucoid cells ofCoryphella rufibranchialis have large heterogeneous and fibrillar secretory granules whereas inAeolidia papillosa, the granules are homogeneous, but vary in electron density from one cell to another. Ellipsoid-vacuolate cells contained large quantities of small vacuoles with an included ellipsoidal structure. Both species contained very numerous ellipsoid-vacuolate cells. Secretory granules and ellipsoid-vacuoles appear to arise from the Golgi apparatus and these contents stain with PAS, suggesting a polysaccharide composition. Mucoid cells contained both secretory granules and ellipsoid-vacuoles which may arise from the same Golgi apparatus.  相似文献

Hydra are characterized by having their body wall organized as an epithelial bilayer with an intervening acellular layer termed the mesoglea. As an extension of the previous study which indicated that mesoglea is a primitive basement membrane which has retained some characteristics of interstitial extracellular matrix, the present study was undertaken to analyze the role of mesoglea components during head regeneration in Hydra vulgaris. Studies were conducted that utilized drugs that affect collagen processing or secondary collagen structure (beta-aminoproprionitrile; 2,2'-dipydridyl; and cis-4-hydroxy-L-proline) and a drug that inhibits addition of glycosaminoglycan chains to proteoglycan core proteins (p-nitrophenyl-beta-D-xylopyranoside). These studies indicated that alterations in the structure of collagens or proteoglycans caused blockage of head regeneration in Hydra as monitored over a 48-hr period. Blockage of head regeneration was reversible once the drugs were removed, indicating that the drugs were not having a general toxic effect on the organism. Radiotracer studies also indicated that blockage of head regeneration was not simply due to a general depression of protein synthesis by the drugs. Various controls indicated that each drug was affecting mesoglea components under the conditions utilized in these studies. These observations indicate that preservation of normal mesoglea structure is required for Hydra head regeneration to proceed.  相似文献

The ventriculus and the midgut caeca of the fed females of Anystis baccarum (L.) were investigated by using light and electron microscopy. In addition to the main type of polyfunctional digestive cells, special secretory cells were detected in the anterior region of the ventriculus. The shape and the ultrastructure of the digestive cells vary depending on their physiological state. Intracellular digestion, absorption or excretion processes prevail at different stages of the cell cycle. The secretory cells are characterized by the presence of extensive rough endoplasmic reticulum, filling whole space of the cell. These cells do not contain the apical network of pinocytotic canals, which are typical for the digestive cells. Three types of secretory granules were found in the cytoplasm of the secretory cells that probably correspond to three sequential stages of granulogenesis. The primary secretory granules are formed by the fusion of Golgi vesicles. The primary granules fuse to form complex vesicles with heterogeneous contents. These secondary granules aggregate to form very large inclusions of high electron density (tertiary secretory granules), which probably represent the storage of the secretory product. All types of secretory granules were observed close to the apical plasmalemma.  相似文献

Changes in the structure of the nucleolus during the cell cycle of the Chinese hamster cell in vitro were studied. Quantitative electron microscopic techniques were used to establish the size and volume changes in nucleolar structures. In mitosis, nucleolar remnants, "persistent nucleoli," consisting predominantly of ribosome-like granular material, and a granular coating on the chromosomes were observed. Persistent nucleoli were also observed in some daughter nuclei as they were leaving telophase and entering G1. During very early G1, a dense, fibrous material characteristic of interphase nucleoli was noted in the nucleoplasm of the cells. As the cells progressed through G1, a granular component appeared which was intimately associated with the fibrous material. By the middle of G1, complete, mature nucleoli were present. The nucleolar volume enlarged by a factor of two from the beginning of G1 to the middle of S primarily due to the accumulation of the granular component. During the G2 period, there was a dissolution or breakdown of the nucleolus prior to the entry of the cells into mitosis. Correlations between the quantitative aspects of this study and biochemical and cytochemical data available in the literature suggest the following: nucleolar reformation following division results from the activation of the nucleolar organizer regions which transcribe for RNA first appearing in association with protein as a fibrous component (45S RNA) and then later as a granular component (28S and 32S RNA).  相似文献

Summary The epineural connective tissue sheath investing the subesophageal ganglion of Helix aspersa consists of a superficial region and a deeper region. The superficial region contains masses of globular cells intermingled with smooth muscle cells and nerve fibers all embedded in a connective tissue matrix. The histochemical and fine structural features of the globular cells show seasonal changes. During autumn to winter glycogen accumulates in their cytoplasm; this accumulation is accompanied by the appearance of dense, cytoplasmic globules which fuse together and ultimately form large pools of granular material. All the organelles and cytoplasm are displaced towards the cell periphery. Various cell-membrane invaginations containing dense material are prominent but there is no direct evidence to link these structures with the uptake of metabolites for glycogenesis. In winter there is a concentration of homogeneous, membrane-bound inclusions in the vicinity of the Golgi bodies. It is suggested that these inclusions constitute a lipid store. They decrease in number during summer. The globular cells do not bear any intimate relation to neurons and there is no reason to include these cells in the neuroglia. The muscle cells often weave around the globular cells but there is no direct contact. Nerve fibers innervate at least some of the muscle cells. The connective tissue consists of large and small diameter fibers suggesting that maturation of the fibrous components of the intercellular matrix is taking place in the superficial regions of the epineural sheath.This work has been supported by the Australian Research Grants Committee.  相似文献

The fine structure of the ectodermal and endodermal muscle layers of Hydra magnipapillata has been analyzed by scanning electron microscopy after hydrolytic removal of the mesoglea with NaOH and subsequent exposure of the basal and lateral aspects of the layers by mechanical dissection. The ectodermal muscle layer consists of fibrous processes of epithelial cells extending longitudinally to the body axis, whereas the endodermal muscle layer comprises cells with hexagonal bases and several strands of myonemes oriented circularly. In each layer, the muscular elements tightly interdigitate, extending a continuous muscle sheet along the mesoglea. The ectodermal and endodermal muscle sheets communicate with each other via foliate microprojections penetrating the mesoglea. On the lateral aspect of the ectodermal epithelium, spiny nerve fibers run along the upper surface of the muscle processes. The spines are often attached to muscle processes, suggesting that the former monitor muscle contraction. Nerve fibers occasionally come into contact with the mesoglea through narrow gaps between the muscle processes. In the hypostomal ectoderm, a small spindle-shaped cell, probably sensory in nature, extends an apical cilium and a long basal process.  相似文献

The cytology and cytochemistry of gemule formation in the freshwater sponge Eunapius fragilis (Leidy) is investigated. The aggregation of ameboid cells which initiates gemmulation is composed of three distinct cell types, two of these fitting the morphological criteria of the archeocyte class. The third cell type, the granular cell, is a differentiated cell type which undergoes cytolysis during gem-mule coat synthesis. Following early polarisation of the aggregate into an internal reproductive area and a peripheral coat region, a structurally and chemically heterogeneous gemmule coat is synthesis-ed. The coat is formed through secretory activities involving several cell types including, possibly, the internal endothelium. The coat is composed of "collagenous" fibers embedded in a matrix of varying degrees of density–showing close structural and cytochemical similarities to vertebrate chondroid tissue. Vitelline platelets of thesocytes of E. fragilis do not cytochemically resemble any previously reported vitelline inclusions from other sponges. Outer valves of the platelets contain components stained by basic dyes at acidic pH which are not extracted by RNase. Distinct differences in developmental processes and morphological–including cytochemical–characteristics of gemmules of different spongillid species are apparent.  相似文献

The monostratified clitellar epidermis of Lumbricillus rivalis consists of supporting cells, granular secretory cells, and globular secretory cells, together with the acid mucous cells typical of normal skin in the anterior and posterior transition zones. The secretion of the granular cells is a neutral glycoprotein with low levels of bound lipid, and that of the globular cells is a sulphated acid mucopolysaccharide lacking detectable protein or lipid.
Ultrastructurally the granular cells possess 1 μm, membrane-bound granules of variable electron density suggestive of maturation changes. The formative granules arise from Golgi vesicles and are moderately electron translucent. They contain parallel-aligned, tubule-like inclusions of 14 nm diameter, observed also in the mature regions of the Golgi. Granules in the mid or apical part of the cell show increased electron density, the formative pattern being wholly or partially obscured and the subunit alignment, when discernable, no longer uniform. Moiré fringe patterns are evident in some granules. The patterns and electron density are lost after pronase digestion. The globular cells contain electron translucent, membrane-bound globules of 2–3 μm diameter with finely fibrous contents. They arise from mature Golgi vesicles and are unaffected by pronase treatment.
The results suggest that the granular cells secrete the cocoon wall and the globular cells the material that surrounds the developing embryos, and are compared with published accounts of other microdrile clitella and with those of the clitellum of lumbricid earthworms.  相似文献

Cytoplasmic annylate lamellae were found in the islet organ of a cyclostome, the hagfish (Myxine glutinosa), predominantly in cells interpreted as young proliferating beta-cells, and also in endocrine cells and enterocytes of the bile duct and gut and in the endothelial cells of small blood vessels. A close association was observed annulate lamellae and granular endoplasmic reticulum. Both in cells with and in those without annulate lamellae, crystalline inclusions of proteinaceous nature were seen in granular endoplasmic reticulum. These inclusions were occasionally closely associated to annulate lamellae, and a direct continuity could be seen between granular endoplasmic reticulum and the outer nuclear membrane surrounding an inclusion partially situated in the perinuclear cisterna. Rod-shaped structures and rounded electron dense bodies were seen in the nuclei of some islet parenchymal cells. The presence of annulate lamellae in the islet organ and associated tissues of Myxine glutinosa is believed to be related to the very high phylogenetic age of this species. The close association observed between annulate lamellae, granular endoplasmic reticulum, crystalline inclusions, and sometimes also nuclear membranes, may be of functional significance.  相似文献

Hydrozoans such as Hydra vulgaris, as with all classes of Cnidaria, are characterized by having their body wall organized as an epithelial bilayer with an intervening acellular layer termed the mesoglea. The present study was undertaken to determine what extracellular matrix (ECM) components are associated with Hydra mesoglea. Using polyclonal antibodies generated from vertebrate ECM molecules, initial light and electron microscopic immunocytochemical studies indicated the presence of type IV collagen, laminin, heparan sulfate proteoglycan, and fibronectin immunoreactive components in Hydra mesoglea. These immunocytochemical observations were in part supported by biochemical analyses of isolated Hydra mesoglea which indicated the presence of fibronectin and laminin based on Western blot analysis. Amino acid analysis of total mesoglea and some of its isolated components confirmed the presence of collagen molecules in mesoglea. Additional studies indicated the presence of (1) a gelatin binding protein in Hydra which was immunoreactive with antibodies raised to human plasma fibronectin and (2) a noncollagen fragment extracted from mesoglea which was immunoreactive to antibodies raised to the NC1 domain (alpha 1 subunit) of bovine glomerular basement membrane type IV collagen. These observations indicate that Hydra mesoglea is evolutionarily a primitive basement membrane that has retained some properties of interstitial ECM.  相似文献

A fraction defined as the inclusions was isolated by banding in CsCl gradients from nuclei of adenovirus 12-infected KB cells. When examined by electron microscopy, the isolated inclusions were relatively homogeneous, finely granular materials of moderate electron density, possibly representing the disintegrated type II or IV inclusions. The conditions of endogenous DNA synthesis in vitro with the inclusions were determined. The product of DNA synthesis in vitro with the inclusions was mainly viral and scarcely cellular, as revealed by DNA-DNA hybridization and methylated albumin kieselgur column chromatography. However, viral DNA synthesized in vitro was smaller (18 S, 22 S) than viral DNA in virions (31 S, 34 S) in neutral and alkaline sucrose gradients. Effects of various treatment of the inclusions on the DNA-synthesizing activity showed that phospholipase C inhibited the activity efficiently. The in vitro DNA synthesis was stimulated by addition of the cytoplasmic extract from adenovirus 12-infected cells and not that from unifected cells. The analysis of the composition of the inclusions showed that the inclusions contained DNA, protein, phospholipid and a small amount of RNA and carbohydrate.  相似文献

Chinese hamster cell strains in the early passages in culture display wide variation in number of nucleolus-like bodies per cell, though such strains are characteristically euploid. A variety of criteria indicate that the nucleolus-like bodies are true nucleoli. Their Azure B- and fast green-staining properties indicate the presence of RNA and protein; they have typical nucleolar fine structure, including both fibrous and granular components; radioautography reveals that their patterns of uptake of uridine-3H into RNA are similar to those reported for nucleoli of other cell types; actinomycin D, at a level which selectively inhibits ribosomal RNA synthesis, greatly reduces their RNA synthesis and also causes segregation of fibrous and granular nucleolar components. Colchicine was used to experimentally fragment the nuclei of these cells into a number of separate karyomeres, each presumably containing some, or only one, of the chromosomes of the complement. Almost all the karyomeres contain nucleolus-like bodies which, by the same criteria applied to the multiple nucleolus-like bodies of uninuclear cells, appear to be true nucleoli. The nucleoli of individual karyomeres of the same cell often differ from each other in fine structure while the multiple nucleoli of a uninuclear cell generally resemble each other. The evidence presented in this study indicates that Chinese hamster cells contain many nucleolus-producing sites scattered through the genome.  相似文献

The gastrodermis of the black coral Antipathes aperta is associated with eight distinct types of cells, including two types of microbasic b-mastigophores (nematocysts), spumous and vesicular mucus cells, and ganglion cells that are essentially the same as in the epidermis. Three additional types of cells are unique to the gastrodermis, and are readily distinguished from those of the epidermis by their electron-opaque inclusions. These include lipoidal cells, zymogen digestive cells, and an unusual type of epitheliomuscular collar cell. The pharyngeal region is characterized by the presence of electron-opaque nematocysts, a scattering of zymogen cells, and a large number of collar cells. The latter are distinguished in part by the presence of dense microfibrillar processes that surround the microvilli and extend into adjacent collars. This interconnection results in the formation of an extensive pharyngeal meshwork. These collar cells are additionally distinguished by the placement of the collar and flagellum adjacent to a flared cup of cytoplasm. This portion of the cell is capable of endocytosis of relatively large unicellular prey, and apparently is capable of forming digestive vesicles as well. The pharyngeal gastrodermis grades into simple lobate septal filaments toward the base of the coelenteron, where large, granular nematocysts all but replace the smaller electron-opaque types Collar cells are found here as well, but in fewer numbers compared to the zymogen cells. Ultrastructural results are compared with those of other coelenterates and discussed in terms of food and modes of nutrition.  相似文献

Summary Ultrastructural evidence is given of the occurrence of nervous elements in the mesoglea of Ctenophores based on the presence of the typical synapses of this phylum.In Beroids, nervous fibers from the ectodermal nerve-net cross the epithelial basal membrane and run through the mesoglea; they are devoid of any ensheathing cell. These neurites build highly differentiated synapses upon the muscles and upon peculiar cells, tentatively named mesenchymal cells.In Cydippids, nerve fibers and nerve cell-bodies have been observed in the mesoglea of the tentacles. The mesogleal core of each tentacle contains mesenchymal cells and a thick strand of neurons and neurites, forming a kind of elongated ganglion. Neurites of either the axial neurones or the epithelial nerve-net neurones form numerous radial nerve strands across the tentacular muscles. Interneural, neuro-muscular and neuro-mesenchymal junctions are very frequent in the tentacle.As far as the organization of the mesoglea is concerned, the Ctenophora thus appear closer to Turbellaria than to Cnidaria.

By means of histological methods and DNA cytophotometry, a study was made of the salivary glands of 16 species of gastropod molluscs belonging to the subclass Pectinibranchia and making a group of Mesogastropoda. Four cell types of salivary glands were distinguished: granular cells (with glycoproteid granular inclusions), mucocytes-I (with sulfatic acid mucopolysaccharides), mucocytes-II (with neutral and acid polysaccharides and proteins), and also the epithelial ciliated cells and mucous duct cells. Data of experiments on starvation and synchronous feeding of molluscs have testified that all described cell types are independent. In some species differentiation on protein and mucous departments within the glandular epithelium was shown. In some marine representatives of the orders Discopoda and Aspidophora polyploid cells with the ploidy levels from 8c to 32c were revealed along with diploid cells. The ecological and phylogenetic regularities of somatic polyploidy manifestation in Mesogastropoda are discussed.  相似文献
