Summary Penetration of the stems ofAraujia sericofera, Asclepias curassavica, Cynanchum ellipticum andSarcostemma viminale by stylets of the aphidAphis nerii was studied with light and differential interference contrast microscopes. Of a total of 118 stylets and 446 stylet tracks observed in cross-sections of stems of the fourAsclepiadaceae, 97 stylets and 372 stylet tracks terminated within the internal primary phloem. Of the remainder, 15 stylets and 74 tracks terminated within the external primary phloem. 22 stylets and 179 of the stylet tracks penetrated the external phloem on the way to the internal phloem. Of these, only four stylets and 32 of the stylet tracks showed signs of attempted probes of the external phloem. It is suggested thatAphis nerii may obtain not only its essential food requirements but also cardiac glycosides as a basis for chemical aposematism. 相似文献

A detailed karyotype analysis of the oleander aphid Aphis nerii focusing on the distribution, molecular composition and epigenetic modifications of heterochromatin was done in order to better understand the structure and evolution of holocentric/holokinetic chromosomes in aphids. The female karyotype (2n = 8) consisted of 3 pairs of autosomes and a pair of X chromosomes that were the longest elements in the karyotype and carried a single, terminally located nucleolar organizer region. Males showed 2n = 7 chromosomes due to the presence of a single X chromosome. Heterochromatin was located in the X chromosomes only and consisted of 4 satellite DNAs that have been identified. A. nerii constitutive heterochromatin was enriched in mono-, di- and tri-methylated H3 histones and HP1 proteins but, interestingly, it lacked DNA methylation that was widespread in euchromatic chromosomal regions. These results suggest that aphid heterochromatin is assembled and condensed without any involvement of DNA methylation. 相似文献

Leaves ofNerium oleander L. plants, which had been previously kept in a shaded glasshouse for at least two months, were fed 1 mM dithiothreitol (DTT)
through their petioles, either for 12h in darkness (overnight) or for 2h in low light (28 μmol photons·m−2·s−1), in each case followed by a 3-h exposure to high light (1260 μmol photons·m−2·s−1). During exposure to high light, violaxanthin became converted to zeaxanthin in control leaves, to which water had been fed,
whereas zeaxanthin did not accumulate in leaves treated with DTT. Total carbon gain was not reduced by DTT during the photoinhibitory
treatment. Exposure to high light led to a decrease in the photochemical efficiency of photosystem II, measured as the ratio

Two clones of Aphis craccivora, one from Nigeria and one from Kenya, differed in their ability to transmit the GRVW1 strain of the rosette virus (Okusanya, 1965). Comparison of body measurements of the two clones showed them to be two distinct but overlapping populations. The Nigerian clone formed colonies on Onobrychus viciifolia, Gomphrena globosa and young Soya max more readily than the Kenya clone. The fecundity of single viviparous females of the two clones on groundnut and bean does not differ significantly. Alatae produce half as many young as apterae.
Zusammenfassung Zwei Klone von Aphis craccivora, einer von Nigeria und einer aus Kenia, unterscheiden sich in der Fähigkeit, Erdnußvirus-Stämme zu übertragen (Okusanya, 1965). Maße der Körpergröße und die Längen der Siphonen, Hintertibien, Antennenglieder und Vordertibien zeigen, daß sie zwei sich überlappenden Populationen angehören. Jungpflanzen von Soya max, Gomphrena globosa und Onobrychis viciifolia werden von dem nigerischen Stamm viel leichter besiedelt als von dem aus Kenia. Einzelne Aptere der beiden Klone produzierten an Erdnuß und Ackerbohnen ähnliche Nachkommenzahlen. Eine einzelne Aptere gebar an Erdnuß 230 Junge; die meisten Larven wurden während der ersten 12 Tage der Fortpflanzungsperiode geboren. Geflügelte beider Klone produzierten nur halb so viele Larven (im Mittel 57 und 52) wie Ungeflügelte (im Mittel 118 und 110). Die beiden Klone sind biologische Rassen von A. craccivora.相似文献

Background
The importance of genetic diversity in successful biological invasions is unclear. In animals, but not necessarily plants, increased genetic diversity is generally associated with successful colonization and establishment of novel habitats. The Oleander aphid, Aphis nerii, though native to the Mediterranean region, is an invasive pest species throughout much of the world. Feeding primarily on Oleander (Nerium oleander) and Milkweed (Asclepias spp.) under natural conditions, these plants are unlikely to support aphid populations year round in the southern US. The objective of this study was to describe the genetic variation within and among US populations of A. nerii, during extinction/recolonization events, to better understand the population ecology of this invasive species.Methodology/Principal Findings
We used five microsatellite markers to assess genetic diversity over a two year period within and among three aphid populations separated by small (100 km) and large (3,700 km) geographic distances on two host plant species. Here we provide evidence for A. nerii “superclones”. Genotypic variation was absent in all populations (i.e., each population consisted of a single multilocus genotype (MLG) or “clone”) and the genetic composition of only one population completely changed across years. There was no evidence of sexual reproduction or host races on different plant species.Conclusions/Significance
Aphis nerii is a well established invasive species despite having extremely low genetic diversity. As this aphid appears to be obligatorily asexual, it may share more similarities with clonally reproducing invasive plants, than with other animals. Patterns of temporal and geographic genetic variation, viewed in the context of its population dynamics, have important implications for the management of invasive pests and the evolutionary biology of asexual species. 相似文献10.
Summary Penetration of leaves of barley,Hordeum vulgare L., by the corn leaf aphid,Rhopalosiphum maidis (Fitch), was studied with light, phase, and electron microscopes. Penetration of epidermis and mesophyll was largely intercellular, that of vascular bundles or veins largely intracellular. Like other aphids,R. maidis secretes a salivary sheath which surrounds the stylets. When mesophyll cells and parenchymatous elements of the veins were penetrated by stylets, their protoplasts were pushed to one side by intruding sheath material; hence, the protoplasts were not punctured by the stylets, although sometimes the plasmalemma of penetrated cells was ruptured by sheath material. The salivary sheaths ended more or less abruptly outside the walls of sieve elements being fed upon, the maxillary stylets projecting beyond the sheaths and into the sieve elements. Before penetrating a functional sieve element the aphid apparently flushes its stylets in order to clear them for ingestion of food. Salivary and food canals merge near the tips of the maxillary stylets to form a single canal, which ends short of the tips.This research was supported by the U.S. National Science Foundation (GB-8330). 相似文献

Aphids are important sucking insects that attack many crops and cause huge economic loss. Essential oils have been proposed to be appropriate alternative to synthetic insecticides due to their low impact on environment and human health. In this work, the chemical compositions of eight essential oils extracted from plants growing in Egypt were determined by gas chromatography/mass spectrometry (GC–MS). The aphicidal activities of the isolated oils were examined against the adults of oleander aphid, Aphis nerii by a leaf-dipping assay. The inhibitory effect of essential oils on acetylcholinesterase (AChE) activity was also tested in vitro. Chemical analyses showed the essential oils consisted mainly of monoterpene hydrocarbons, oxygenated monoterpenes, sesquiterpene hydrocarbons and oxygenated sesquiterpenes except the oil of Artemisia monosperma which contained high content of benzenes. The results of toxicity assay showed that the oils of A. monosperma and Citrus sinensis were the most potent toxicants displaying LC50 value of 0.06 mg/L. The oils of Callistemon viminals, Schinus terebinthifolius and Schinus molle were also highly effective against aphid as their LC50 values were lower than 1 mg/L. In addition, the highest inhibitory effect on AChE was recorded by 0.5 mg/L C. sinensis with inhibition of 49.33%, while 0.1 mg/L Citrus lemon oil recorded the lowest inhibitory effect with inhibition of 0.4%. In general, six of tested essential oils showed high toxicity against A. nerii and they might be applied in the IPM programs for this insect. 相似文献

Summary Populations of the milkweed-oleander aphid,Aphis nerii, were sampled in California, Iowa and Puerto Rico. Among these localities the aphid's host plants differ greatly in permanence. I compared populations for migratory potential, measured as the proportion of winged offspring produced in response to being crowded, and for life history and morphometric traits of the subsequent adult winged aphids. I predicted a negative correlation between degree of host plant permanence and migratory potential. As predicted, aphids from Iowa, where migration on to temporary hosts must occur each year, produce a greater proportion of winged offspring (37.7%) than those from California (25.7%) or Puerto Rico (31.6%) where hosts are more permanent. However, hosts in Puerto Rico appear to be more permanent than those in California, yet the difference between populations for migratory potential was opposite to that predicted. Within California the prediction again held: aphids collected from the most impermanent sites produce the greatest proportion of winged offspring. There were no population differences for any life history or morphometric traits of winged aphids that are important contributors to fitness or migratory ability such as time to reproductive maturity, fecundity or wing length. Nor did any traits covary with migratory potential. Thus, there does not appear to be an association of life history and morphology with migratory potential that could enhance the colonizing ability of migrant aphids. I was unable to detect population differentiation for life history and morphology even though there is ample genetic variation within populations on which selection could act and an absence of constraints arising from genetic correlations that could prevent appropriate evolution of traits within populations. The exploitation of temporary host plants therefore occurs by an increase in the number of colonists produced and not by change in life history or morphology of those colonists. 相似文献

The sensory innervation of the mandibular stylets of the aphid Brevicoryne brassicae (L.) has been examined by electron microscopy. Two groups of sensory neurones are present in the mandible. Each has two neurones, one with a short dendrite extending into the base of the mandible and ending in the base and another with a long microtubular process which extends 500 m? down to the distal tip of the mandible. The two neurones are enclosed by an ensheathing cell comparable to the trichogen cell enveloping the group of neurones innervating pegs and hairs. This ensheathing cell is supported by extensive electron-dense filaments to form a scolopale and is embedded in the mass of stylet-forming cells at the base of the mandible. The inner segments of the dendrites are anchored to the ensheathing cell by desmosome junctions. Desmosome junctions also bind the microtubular outer segments of the short and long dendrite to each other. There is no evidence of a dendritic sheath enclosing the distal portion of the short dendrite which ends while still in the extracellular space within the ensheathing cell. The microtubular process of the long dendrite extends down the lumen of the mandible enclosed by a close-fitting extracellular sheath which penetrates and is attached to the cuticular wall of the mandible tip. Distally this sheath is thickened on one side. Deflection of the mandible would therefore deform the dendritic membrane asymmetrically because the thin walls of the sheath would bend more than the thick walls. This would exert an unequal mechanical strain on the dendritic membrane which could result in depolarization in response to deflection in a particular direction. The arrangement of the dendrites and their sheaths within the mandible is such that deflection to the right would distort one dendrite in the same way as deflection to the left would distort the other. 相似文献

Cowpea, Vigna unguiculata was infested at the young, mid-fill and mature pod growth stages with, respectively, one, two and three pre-reproductive adult apterae of Aphis craccivora per pod in a screen house. A. craccivora caused pod shrivelling at the young and mid-fill pod stages, on which the aphid was more fecund. The economically important grain aphid, Sitobion avenae (F.) shows colour polymorphism, with brown and green forms predominating. Colour is determined both genetically and in response to environmental factors, including nutrition. The biological significance of the colour polymorphism is unknown, although seasonal changes occur in the frequency of colour morphs in the field, whilst the brown morph may have adaptive significance in terms of hymenopterous endoparasitism. The ground colour of aphids is produced by haemolymph pigments, aphins (glucosides) and carotenoids. The latter may be under the synthetic control of intracellular endosymbiotic bacteria. In this study, the major carotenoid pigments of a brown and a green clone of S. avenae were examined using thin layer chromatography (TLC) and high-performance liquid chromatography (HPLC), and their absorbance spectra recorded. Using TLC, the brown clone produced five bands of different Rf, ranging from yellow, to orange-pink to pink in colour. In contrast, the green clone gave only a single yellow band of higher Rf than any of the bands of brown aphids. Following separation of carotenoids by HPLC, brown aphids gave seven peaks and green aphids five. Comparison of absorbance maxima with known published values for carotenoids provides strong evidence for the identification of four of the carotenoid pigments from brown aphids (RB-4, 3,4-didehydrolycopene; RB-5, torulene; RB-6; lycopene; RB-7, γ-carotene) and one from green aphids (RG-2, α-carotene). The other carotenoids remain unidentified. The biosynthesis and possible biological relevance of the various pigments of S. avenae are briefly discussed. 相似文献
