To investigate the expression and biological significance of Leptin, Leptin receptor, Vascular Endothelial Growth Factor (VEGF), and CD34 protein in colorectal carcinoma tissues. The expression of Leptin, Leptin receptor, VEGF, and CD34 was detected in 68 cases of colorectal carcinoma tissues, paired para-carcinoma tissues and normal colorectal tissues by Immunohistochemical SP Method. The results and related clinicopathological data were analyzed. The positive rate of Leptin, Leptin receptor, and VEGF was significantly higher in colorectal carcinoma tissues than that in paired para-carcinoma tissues and normal colorectal tissues. The expression of Leptin, Leptin receptor, and VEGF was correlated with grade of tumor differentiation, depth of bowel wall invasion, lymph node metastasis, Dukes stage, distant metastasis, and lympho/vascular tumor embolization. Microvessel density (MVD) value in colorectal carcinoma was significantly higher than that in para-carcinoma tissues and normal colorectal tissues, and the density in para-carcinoma tissues was higher than that in normal colorectal tissues. The expression of Leptin, Leptin receptor, VEGF, and MVD value in colorectal carcinoma was positively correlated. In conclusion, microvessel density value is an important index of the growth, invasion, and metastasis of colorectal carcinoma. The binding of Leptin and Leptin receptor promotes the proliferation of colorectal carcinoma cells. The synergy between Leptin and VEGF accelerates the angiogenesis in colorectal carcinoma and accelerates the invasion and metastasis of the tumor cells.  相似文献

Background

There is compelling evidence in humans that peripheral endocannabinoid signaling is disrupted in obesity. However, little is known about the corresponding central signaling. Here, we have investigated the relationship between gender, leptin, body mass index (BMI) and levels of the endocannabinoids anandamide (AEA) and 2-arachidonoylglycerol (2-AG) in the serum and cerebrospinal fluid (CSF) of primarily overweight to obese patients with osteoarthritis.

Methodology/Principal Findings

Patients (20 females, 15 males, age range 44-78 years, BMI range 24-42) undergoing total knee arthroplasty for end-stage osteoarthritis were recruited for the study. Endocannabinoids were quantified by liquid chromatography – mass spectrometry. AEA and 2-AG levels in the serum and CSF did not correlate with either age or BMI. However, 2-AG levels in the CSF, but not serum, correlated negatively with CSF leptin levels (Spearman’s ρ -0.48, P=0.0076, n=30). No such correlations were observed for AEA and leptin.

Conclusions/Significance

In the patient sample investigated, there is a negative association between 2-AG and leptin levels in the CSF. This is consistent with pre-clinical studies in animals, demonstrating that leptin controls the levels of hypothalamic endocannabinoids that regulate feeding behavior.  相似文献

目的:通过检测人膝骨关节炎裸露软骨下骨中OPN的表达,探讨OPN在OA发病及病情进展中的意义。方法:选取接受膝关节置换手术的膝关节骨关节炎患者软骨下骨标本50例,采用综合评分法对OA患者进行严重程度分级,分为轻、中、重度三组,取正常膝关节软骨下骨(股骨髁关节面)10例作为正常软骨下骨对照;对标本进行免疫组织化学染色,用SPSS17.0统计软件包分析各组间OPN表达的差异及OA患者OPN表达与综合评分、K-L分期的相关性。结果:人膝骨关节炎裸露软骨下骨OPN表达明显高于正常软骨下骨组,差异有统计学意义(P〈0.01);OPN在轻、中、重度膝骨关节炎裸露软骨下骨的表达差异有统计学意义(P〈0.05);膝骨关节炎裸露软骨下骨OPN的表达与骨关节炎的综合评分、K—L分期呈正相关。结论:OA患者膝关节软骨下骨OPN表达与疾病严重程度呈正相关,提示OPN在骨关节炎发病及病情进展中可能起作用。  相似文献

Human small intestine accounts for 75% of the gastrointestinal (GI) length but for only 1–5% of GI tumors. The reason remains as yet unclearly understood. Our study was designed to examine whether increased apoptosis and expression of related genes/proteins, especially those of the Bcl-2 family, contribute to this difference. For this purpose, 77 samples from patients were examined by terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling and immunohistochemistry, including 40 cases from normal small intestine (jejunum), 7 cases from jejunum and ileum adenocarcinomas, and 30 cases from normal colon. The results showed that a significantly higher level of enterocyte apoptosis was observed in normal small intestine compared with small intestinal adenocarcinomas and normal colon (median of apoptotic index, 15.2% vs 0.1% and 1.6%, p<0.01). A similar pattern was observed for Bax (expression-positive, 77.5% vs 28.6% and 53.3%, p<0.05) but not for Bcl-2 (42.5% vs 42.9% and 46.7%, p>0.05) or Bax/Bcl-2 ratio (percent of samples having a ratio ≥1, 45.0% vs 14.3% and 36.7%, p>0.05). In conclusion, increased apoptosis and expression of Bax, not Bcl-2 or the Bax/Bcl-2 ratio, may play some role in the relatively lower incidence of human small intestinal carcinomas. The mechanisms of epilepsy remain incompletely understood. Rac1 (ras-related C3 botulinum toxin substrate 1) belongs to the Rho family of small GTPases. Rac1 play important roles in cytoskeleton rearrangement and neuronal synaptic plasticity, which had also been implicated in epilepsy. However, little is known regarding the expression of Rac1 in the epileptic brain or whether Rac1-targeted interventions affect the progression of epilepsy. The aim of this study was to investigate the expression profile of Rac1 in brain tissues from patients suffering from temporal lobe epilepsy (TLE) and experimental epileptic rats and determine the possible role of Rac1 in epilepsy. We demonstrated that the expression of Rac1 is significantly increased in TLE patients and in lithium-pilocarpine epilepsy model animals compared to the corresponding controls. Rac1 inhibitor NSC23766 reduced the severity of status epilepticus during the acute stage in a lithium-pilocarpine animal model. Consistent with these results, the latent period of a PTZ kindling animal model also increased. Our results demonstrated that the increased expression of Rac1 may contribute to pathophysiology of epilepsy.

The adipocyte hormone, leptin has been demonstrated to have profibrogenic actions in vitro and in animal models. However, no correlation was found between plasma leptin levels and fibrosis stage in humans. Thus, our aim was to study whether soluble leptin receptor (SLR) or free leptin index (FLI; calculated as the ratio of leptin to SLR), may correlate better with the features of metabolic syndrome and with the histological grade and stage of nonalcoholic fatty liver disease (NAFLD)/nonalcoholic steatohepatitis (NASH). We studied a population (n = 104) of morbidly obese patients undergoing bariatric surgery. Data including BMI, type 2 diabetes mellitus, hypertension, and hyperlipidemia were obtained. Plasma fasting leptin and SLR, fasting glucose and insulin were measured, and homeostasis model of assessment insulin resistance (HOMAIR) index and FLI were calculated. All patients had intraoperative liver biopsies. Leptin levels correlated with the BMI. The multiple regression analysis indicated that increasing HOMA and decreasing FLI were predictors of steatosis in the liver (P < 0.0003). SLR levels were positively correlated with the presence of diabetes mellitus and the stage of fibrosis. In conclusion, increased SLR levels in morbidly obese patients with diabetes are correlated with the stage of liver fibrosis, and may reflect progressive liver disease.  相似文献

In the central nervous system, zinc is released along with glutamate during neurotransmission and, in excess, can promote neuronal death. Experimental studies have shown that metallothioneins I/II (MT-I/II), which chelate free zinc, can affect seizures and reduce neuronal death after status epilepticus. Our aim was to evaluate the expression of MT-I/II in the hippocampus of patients with temporal lobe epilepsy (TLE). Hippocampi from patients with pharmacoresistant mesial temporal lobe epilepsy (MTLE) and patients with TLE associated with tumor or dysplasia (TLE-TD) were evaluated for expression of MT-I/II, for the vesicular zinc levels, and for neuronal, astroglial, and microglial populations. Compared to control cases, MTLE group displayed widespread increase in MT-I/II expression, astrogliosis, microgliosis and reduced neuronal population. In TLE-TD, the same changes were observed, except that were mainly confined to fascia dentata. Increased vesicular zinc was observed only in the inner molecular layer of MTLE patients, when compared to control cases. Correlation and linear regression analyses indicated an association between increased MT-I/II and increased astrogliosis in TLE. MT-I/II levels did not correlate with any clinical variables, but MTLE patients with secondary generalized seizures (SGS) had less MT-I/II than MTLE patients without SGS. In conclusion, MT-I/II expression was increased in hippocampi from TLE patients and our data suggest that it is associated with astrogliosis and may be associated with different seizure spread patterns.  相似文献

目的:研究子痫前期患者胎盘组织中色素上皮衍生因子(pigment epithelium-derived factor,PEDF)的表达,探讨PEDF在子痫前期发病中的作用。方法:选取2012年3月至2013年3月在我院产科住院剖宫产的20例子痫前期孕妇作为研究对象,另选取同期正常分娩的孕妇20例作为对照组。采用Western blot、免疫荧光组织化学方法检测子痫前期患者和正常对照组妇女胎盘组织中PEDF和血管内皮生长因子(vascular endothelial growth factor,VEGF)的表达,并通过免疫荧光方法计数胎盘微血管密度(MVD)。结果:同正常对照组相比,子痫前期患者胎盘组织中PEDF表达升高,而VEGF表达则减少。PEDF与VEGF组织表达位置大致相同。子痫前期患者胎盘组织中PEDF表达与24h尿蛋白定量呈正相关,与VEGF表达及MVD计数呈负相关;VEGF表达与MVD计数呈正相关。结论:PEDF与子痫前期疾病发生发展及病情轻重程度有关;PEDF可能是通过影响胎盘血管的重铸,而参与子痫前期疾病的发生发展。  相似文献

目的:探讨原发性肝细胞癌(HCC)组织中Beclin 1的表达及其在肝细胞癌发生发展中的作用。方法:应用免疫组化EnVision法检测54例肝细胞癌组织及其癌旁组织中Beclin 1的表达,分析其与肝细胞癌的病理分级、临床分期、侵袭及转移等临床病理因素的关系。结果:HCC组织中Beclin 1阳性表达率明显低于癌旁组织(X2=7.53,P〈0.01)。病理分级Ⅲ~Ⅳ、TNM分期Ⅲ~Ⅳ、存在静脉浸润和淋巴结转移的肝癌组织中的Beclin 1表达水平明显低于病理分级Ⅰ~Ⅱ、TNM分期Ⅰ~Ⅱ、无静脉浸润和淋巴结转移的肝癌组织(P〈0.01)。结论:HCC组织中存在Beclin 1表达缺失,可能与HCC的发生有着密切的关系,且对HCC的恶性生物学行为有重要影响。  相似文献

目的:研究脑胶质瘤患者血浆中miR-21的水平,并评价其作为临床诊断标志物的可能性。方法:收集临床确诊的胶质瘤患者和健康对照人群的血浆标本,常规方法提取血浆中RNA,采用茎环结构的引物将其逆转录成cDNA,实时定量PCR的方法分别检测miR-21在胶质瘤患者和健康对照人群中的水平,根据ROC分析评价miR-21作为诊断标志物的可能性。结果:分别收集了胶质瘤患者和健康对照人群血浆标本52例和43例,实时定量PCR结果显示,相对于健康对照人群,miR-21在胶质瘤患者血浆中呈高水平状态。ROC分析说明血浆miR-21水平作为诊断胶质瘤标志物具有较高的灵敏度和特异度。结论:血浆miR-21水平具有作为胶质瘤诊断的生物标志物的潜能。  相似文献

目的:研究妊娠高血压患者血清瘦素、胱抑素C(Cys C)以及人绒毛膜促性腺激素(β-HCG)水平检测的临床意义。方法:选取2014年4月到2015年4月我院收治的妊娠高血压患者90例(妊娠高血压组),另选同期正常妊娠者90例(正常妊娠组),健康体检非妊娠妇女90例(对照组),测量三组血清瘦素、Cys C以及β-HCG水平,并分析各指标的相关性。结果:妊娠高血压组和正常妊娠组血清瘦素、Cys C以及β-HCG水平均显著高于对照组,比较差异具有统计学意义(P0.05),妊娠高血压组血清瘦素、Cys C以及β-HCG水平均显著高于正常妊娠组,比较差异具有统计学意义(P0.05);妊娠高血压越严重血清瘦素、Cys C以及β-HCG水平越高;妊娠高血压患者血清瘦素与Cys C和β-HCG均呈正相关关系(P0.05),Cys C与β-HCG也呈正相关关系(P0.05)。结论:检测血清中瘦素、Cys C和β-HCG水平有利于妊娠高血压的诊断,体检时发现上述物质异常升高要高度重视。  相似文献

目的:探讨ADAM10在胰腺癌患者外周血中的表达及其临床意义。方法:应用酶联免疫吸附试验法(ELISA)检测40例胰腺癌患者和20例健康体检者的外周血ADAM10的表达水平,分析其与临床病理特征的关系。结果:胰腺癌患者血清ADAM10水平显著高于正常对照组(P〈0.01);胰腺癌患者血清中ADAM10的表达水平与胰腺癌淋巴结转移、远处转移及TNM分期密切相关(P〈0.05),且行根治性手术切除的胰腺癌患者ADAM10表达水平低于姑息性手术切除的患者(P〈0.05);ADAM10对胰腺癌诊断的敏感度、特异性分别为51.7%、76.9%,联合检测CA19-9有助于提高胰腺癌诊断敏感度,但特异性有所下降;根治性切除后胰腺癌患者血清中ADAM10水平明显下降。结论:胰腺癌癌患者血清中ADAM10水平明显增高,检测血清ADAM10有助于胰腺癌的诊断和治疗。  相似文献
