在低盐介质中,含有垛叠类囊体(基粒)与不垛叠类囊体(间质片层)结构的叶绿体进行的光系统I电子传递与磷酸化反应(PSP)都为低浓度的尼日利亚菌素所促进,低浓度的氯化铵对基粒结构叶绿体的系统I与包括两个系统的电子传递以及与之相偶联的磷酸化反应有促进效应,而对间质片层膜上进行的PSP反应无影响。尼日利亚菌素或氯化铵对PSP的促进效应在高盐介质中消失,且它们对高能态(Z)形成的抑制效应在低盐介质中较高盐介质中大。当存在吡啶时,在低盐介质中它们对Z形成的影响是抑制作用,在高盐介质中则是促进效应。这都表明不同结构的膜上均存在区域化质子。  相似文献

目的:观察低浓度哇巴因对人白血病细胞株Jurkat生长的影响并初步探讨哇巴因特异性调节Jurkat细胞生长的机制。方法:分别用不同低浓度哇巴因作用于人白血病细胞株Jurkat后,采用四甲基偶氮唑盐MTT法检测细胞增殖情况、流式细胞学FCM技术检测细胞凋亡情况以及细胞内线粒体膜电位变化情况,Western blot法观察哇巴因对Jutkat细胞膜表面钠钾ATP酶的表达调节作用。结果:MTT及FCM检测结果表明随着哇巴因浓度的增高,哇巴因对Jurkat细胞的增殖抑制及促凋亡作用越明显。WesternBlot结果显示30nM及50nM哇巴因作用于Jurkat细胞株48h后引起细胞钠钾ATP酶表达下调,[3H]-哇巴因结合实验结果显示在Jurkat细胞株随着哇巴因作用浓度升高,细胞膜钠泵对哇巴因的亲和力逐渐下降。结论:低浓度哇巴因即可抑制白血病细胞株Jurkat增殖并诱导其凋亡。这种特异性细胞生长调控作用与哇巴因引起的细胞膜钠钾ATP酶表达变化相关,最终引起细胞内线粒体膜电位发生变化,释放相关凋亡蛋白,诱导细胞凋亡。  相似文献

Application of zero-valent iron nanoparticles (nZVI) for Zn2+ removal and its mechanism were discussed. It demonstrated that the uptake of Zn2+ by nZVI was efficient. With the solids concentration of 1 g/L nZVI, more than 85% of Zn2+ could be removed within 2 h. The pH value and dissolved oxygen (DO) were the important factors of Zn2+ removal by nZVI. The DO enhanced the removal efficiency of Zn2+. Under the oxygen-contained condition, oxygen corrosion gave the nZVI surface a shell of iron (oxy)hydroxide, which could show high adsorption affinity. The removal efficiency of Zn2+ increased with the increasing of the pH. Acidic condition reduced the removal efficiency of Zn2+ by nZVI because the existing H+ inhibited the formation of iron (oxy)hydroxide. Adsorption and co-precipitation were the most likely mechanism of Zn2+ removal by nZVI. The FeOOH-shell could enhance the adsorption efficiency of nZVI. The removal efficiency and selectivity of nZVI particles for Zn2+ were higher than Cd2+. Furthermore, a continuous flow reactor for engineering application of nZVI was designed and exhibited high removal efficiency for Zn2+.  相似文献

The growth of white clover (Trifolium repens L.) in conditionstypical of April in Southern England (8 °C day/4 °Cnight, 12 h photoperiod of 90 J m–2 s–1 visibleradiation) was extremely slow, whether the plants were dependentfor nitrogen on fixation by their root nodules or were suppliedwith abundant nitrate; although growth was slower in the nodulatedplants. The reasons for slow growth were a large root: shootratio and a small leaf area, particularly in the nodulated plants,and a low photosynthetic rate in all plants. The probable effectsof these characteristics on the growth of white clover withgrasses in mixed pastures are discussed. The ability of an Aeromonas hydrophila isolate obtained from filtered river water to grow at low substrate concentrations was studied in batch experiments with tap water supplied with low concentrations of substrates. Growth was assessed by colony count determinations. The isolate only multiplied in the used tap water (2 to 3 mg of dissolved organic carbon per liter) after the addition of a small amount of an assimilable carbon compound. d-Glucose especially caused growth of the organism even at initial concentrations below 10 mug of C per liter. At initial glucose concentrations below the K(s) value (12 mug of C per liter), generation times and yield (colony-forming units per milligram of substrate-C) were nonlinear with 1/initial glucose concentrations and initial glucose concentrations, respectively. From these observations, the maintenance coefficient m was calculated (m = 0.015 mg of glucose per mg [dry wt] per h at 12 degrees C). At initial concentrations below the K(s) value of starch (73 mug of C per liter), no growth was observed, but complete use of starch occurred in these situations after the addition of 10 mug of glucose-C per liter. The results of this study show that information of ecological significance may be obtained by very simple batch experiments. Moreover, the isolate studied may be used in growth experiments to assess the maximum concentration of glucose which might be present in water, particularly tap water.  相似文献

The objective of this research is to investigate Fenton and persulfate oxidation with zero-valent iron [Fe(0)] as a batch type ex-situ remediation technology for the treatment of diesel-contaminated soil. Results from batch experiments indicate that Fe(0) is a better catalyst for H2O2 and persulfate than Fe2+ for the enhancement of Fenton and persulfate oxidation in a batch system. Maximum removal was obtained after 12 h when 1 and 2 g of Fe(0) were added to hydrogen peroxide (250 mg/L) and persulfate (250 mg/L), respectively, in a soil-water system. As the amounts of Fe(0) and persulfate were increased three times at the optimal ratio, the removal of total petroleum hydrocarbon (TPH) was enhanced accordingly. More than 90% of the TPH was removed in 3 h, and the treated soil met the Korean regulation level (500 mg/kg) for TPH. Increased amounts of Fe(0) and hydrogen peroxide (up to 10 g and 1250 mg/L, respectively) also significantly enhanced degradation under the optimal conditions. The results of our study suggest that Fe(0)-assisted Fenton and persulfate oxidation in a batch reactor may be an alternative option to treat diesel-contaminated soil.  相似文献

A series of iron intercalated montmorillonites (Fe‐Monts) were prepared using (i) ion exchange of native sodium and calcium ions with iron ions, (ii) base hydrolysis of inserted iron ions in montmorillonite suspension, and (iii) insertion of pre‐hydrolyzed iron colloid in montmorillonite. The materials were characterized by X‐ray diffraction and gas adsorption‐desorption techniques. The basal d(001)‐spacing and BET specific surface area increased after the intercalation of iron species in montmorillonite. Local iron structure studied by X‐ray absorption fine structure (XAFS) spectroscopy showed an unsaturation of the Fe···Fe coordination number (N 2.5) of the intercalated iron species as compared to the bulk iron oxyhydroxides (N 6). The Fe‐Monts were employed for arsenic removal from aqueous solutions at low concentration (0.2–16 mg/L). Among the Fe‐Monts, the one prepared by the hydrolysis of inserted iron ions, was the best in performance. The saturation adsorption amount of the optimized iron‐montmorillonite was 4 and 28 times higher for the removal of arsenite and arsenate, respectively, as compared to bulk iron oxyhydroxide (goethite). Compared with bulk iron oxyhydroxide, the Fe‐Monts were superior for arsenate uptake and comparable for arsenite. In addition, arsenite adsorbed on the Fe‐Monts was found to be oxidized to arsenate based on XAFS spectroscopy.  相似文献

氮素形态和铁营养对玉米苗期生长及体内铁分布的影响   总被引:1,自引:0,他引:1  
以玉米(Zea mays)品种‘豫玉-22’为材料,采用营养液培养方法,研究了低铁和正常供铁条件下供应不同形态氮素对玉米苗期生长及体内铁分布的影响。结果表明:(1)与低铁介质相比,常铁介质增加了各氮素处理玉米幼苗的株高、地上部干重、全株干重,降低了根冠比,其中硝态氮处理表现得尤其突出;与供应硝态氮(NO3--N)相比,增施铵态氮(1/2 NO3--N 1/2 NH4 -N,NH4 -N)能明显促进低铁介质中玉米生长,但在常铁介质下作用不明显。(2)相比于低铁介质,正常供铁显著提高了相应处理玉米新叶叶绿素含量及净光合速率;2种供铁介质中,NH4 -N处理的新叶叶绿素含量以及净光合速率均高于其它氮素处理。(3)相比于低铁介质,正常供铁处理总体上增加了玉米各部分活性铁含量和全铁含量,对NO3--N处理的新叶活性铁含量增加尤其明显;2种供铁介质中,NH4 -N均有利于提高新叶活性铁含量和植株地上部全铁含量。(4)玉米新叶活性铁含量不仅与其叶绿素含量显著正相关(r=0.979**),也与叶片净光合速率显著正相关(r=0.950**)。研究发现,供铁状况显著影响玉米新叶的叶绿素含量及其净光合速率且与供氮形态存在互作;供应铵态氮有利于提高缺铁条件下玉米新叶活性铁含量,增强玉米植株的光合能力,进而促进其正常生长。  相似文献

The effect of ethephon on the growth of leaves and stems ofP. pratensis, Agrostis sp., and A. sativa was investigated.The length of the leaves of all species as well as that of theinternodes of stems of vegetative tillers of Agrostis sp. andof stems of reproductive A. sativa plants was reduced. Epidermalcell length was unchanged or somewhat increased, indicatinga reduction in cell number. The rate of cell elongation maybe decreased, however. A reduction in cell length was observedin the internode of generative oat plants that just startedto elongate at the time of treatment. In vegetative tillers of P.pratensis and A. sativa ethephonpromoted growth of the internodes by increasing cell length.Cell number was little affected. GA3 also promoted internodegrowth, causing an increase in cell length as well as in cellnumber. Application of ethephon and GA3 together gave an additiveeffect on cell length but a reverse effect on cell number. Effects of ethephon were observed for 5 or more weeks aftertreatment. Ethylene production from plants of A sativa increased,but the extra production had already decreased by 90% after11 d. No stimulation of the ethylene production by the plantsother than from the breakdown of ethephon was observed.  相似文献

Micronutrient nanoparticles (NPs) are currently an option for chemical fertilization and biostimulation in crops. However, there is little information on the phytotoxic or biostimulatory effects of NPs at low concentrations of some elements, such as Zn. In this study, the effect of low concentrations of Zn oxide (ZnO) NPs on germination, growth variables, and nutritional attributes of lettuce (Lactuca sativa L.) was evaluated in comparison to Zn sulfate. Romaine lettuce seeds were treated with ZnSO4-- × 7H2O and ZnO NPs at Zn molar concentrations of 1 × 10−3, 5 × 10−3, 1 × 10−4, 5 × 10−4, 1 × 10−5, 5 × 10−5, 1 × 10−6, and 5 × 10−6. The seeds treated with ZnSO4 at 5 × 10−6 registered the highest radicle length, 73% more than the control treatment. The seeds treated with ZnSO4 at 5 × 10−3 registered the lowest values, with 50% less than the control treatment. ZnO NPs at 5 × 10−6 significantly increased content of chlorophyll A and B and total phenolics. These results indicate the possible existence of a mechanism related to the intrinsic nanoparticle properties, especially at low concentrations.

Low temperature of 2–5° C. accelerates the rate offormation of amino acids from reserve proteins in the endospermof Heracleum sphondylium. It also brings about the accumulationof glycine and arginine in abundance. Experimental culture ofexcised embryos on synthetic media shows that these acids areparticularly beneficial to embryo growth whilst alanine, thepredominating acid at room temperature, is not. S ummary : Growth in soils of Mycobacterium paraffinicum with 0, 5, 15, 45 and 270 p/m of ethane was assayed by adding ethane-1,2-14C and measuring the 14CO2 produced. With < 45 p/m of ethane the lag period lasted several months. In nature, ammonia-oxidizing bacteria have to compete with heterotrophic bacteria and plants for limiting amounts of ammonium. Previous laboratory experiments conducted with Nitrosomonas europaea suggested that ammonia-oxidizing bacteria are weak competitors for ammonium. To obtain a better insight into possible methods of niche differentiation among ammonia-oxidizing bacteria, we carried out a growth experiment at low ammonium concentrations with N. europaea and the ammonia oxidizer G5-7, a close relative of Nitrosomonas oligotropha belonging to Nitrosomonas cluster 6a, enriched from a freshwater sediment. Additionally, we compared the starvation behavior of the newly enriched ammonia oxidizer G5-7 to that of N. europaea. The growth experiment at low ammonium concentrations showed that strain G5-7 was able to outcompete N. europaea at growth-limiting substrate concentrations of about 10 μM ammonium, suggesting better growth abilities of the ammonia oxidizer G5-7 at low ammonium concentrations. However, N. europaea displayed a more favorable starvation response. After 1 to 10 weeks of ammonium deprivation, N. europaea became almost immediately active after the addition of fresh ammonium and converted the added ammonium within 48 to 96 h. In contrast, the regeneration time of the ammonia oxidizer G5-7 increased with increasing starvation time. Taken together, these results provide insight into possible mechanisms of niche differentiation for the ammonia-oxidizing bacteria studied. The Nitrosomonas cluster 6a member, G5-7, is able to grow at ammonium concentrations at which the growth of N. europaea, belonging to Nitrosomonas cluster 7, has already ceased, providing an advantage in habitats with continuously low ammonium concentrations. On the other hand, the ability of N. europaea to become active again after longer periods of starvation for ammonium may allow better exploitation of irregular pulses of ammonium in the environment.  相似文献

Metal-containing nanoparticles (NPs) are currently used for various biomedical applications. Since such NPs are able to enter the brain, the cells of this organ have to deal with NPs and with NP-derived metal ions. In brain, astrocytes are considered to play a key function in regulating metal homeostasis and in protecting other brain cells against metal toxicity. Thus, among the different types of brain cells, especially astrocytes are of interest regarding the uptake and the handling of metal-containing NPs. This article summarizes the current knowledge on the consequences of an exposure of astrocytes to NPs. Special focus will be given to magnetic iron oxide nanoparticles (IONPs) and silver nanoparticles (AgNPs), since the biocompatibility of these NPs has been studied for astrocytes in detail. Cultured astrocytes efficiently accumulate IONPs and AgNPs in a time-, concentration- and temperature-dependent manner by endocytotic processes. Astrocytes are neither acutely damaged by the exposure to high concentrations of NPs nor by the prolonged intracellular presence of large amounts of accumulated NPs. Although metal ions are liberated from accumulated NPs, NP-derived iron and silver ions are not exported from astrocytes but are rather stored in proteins such as ferritin and metallothioneins which are synthesized in NP-treated astrocytes. The efficient accumulation of large amounts of metal-containing NPs and the upregulation of proteins that safely store NP-derived metal ions suggest that astrocytes protect the brain against the potential toxicity of metal-containing NPs.  相似文献

The effect of abscisic acid on growth, ultrastructure and nucleic acid biosynthesis was studied in tissue culture of spinach (Spinacia oleracea L.). Low concentration (0.01 mg l?1) of abscisic acid increased fresh and dry weight of calluses, whereas 1.0 mg l?1 was inhibitory. The stimulating effect was observed only in the presence of a relatively high concentration of kinetin (1 mg l?1). The inhibitory effect was partly overcome by the same kinetin concentration. The low concentration of abscisic acid probably accelerated the induction of callus growth after subculture and stimulated cell division in the exponential phase of growth. Electron microscopy showed the presence of numerous polysomes and rough endoplasmic reticulum in callus cells grown at the stimulating abscisic acid concentration. Control cells and cells at the inhibitory concentration had slightly hyaline cytoplasm and were more vacuolated. Incubation of callus tissue with 32P in the presence of stimulating concentration of abscisic acid showed a significant increase in the rate of biosynthesis of all nucleic acid classes after 8 h, whereas inhibitory concentration produced a decrease in 32P incorporation. However, when the tissue was grown in the presence of abscisic acid for 20 days, both concentrations decreased the rate of nucleic acid biosynthesis, as compared to the controls.  相似文献

以2个耐低钾基因型水稻N18、N19和2个低钾敏感基因型水稻N27、N28为材料,采用溶液培养试验,研究低钾胁迫对其苗期根系生长和内源激素含量的影响。结果表明,低钾胁迫下,水稻根长、地上部干重和根干重均降低,但N18和N19显著高于N27和N28。低钾胁迫使4个基因型水稻的根冠比增大,而各基因型之间差异不显著。低钾胁迫下,水稻根中IAA、GA1和ZR含量均减少,ABA含量增加;N18、N19根中IAA、GA1和ZR含量都高于N27、N28。此外,低钾胁迫使水稻根中IAA/ABA、ZR/ABA、GA1/ABA值降低,但N18、N19的上述比值高于N27、N28。  相似文献
