HCMV感染抑制人海马神经干细胞分化   总被引:1,自引:0,他引:1  
研究HCMV感染对体外培养的人海马源性神经干细胞(Neural stem cells,NSCs)分化的影响。体外分离、培养人海马NSCs,应用免疫荧光方法检测其NSCs标记物-巢蛋白(Nestin)的表达。10%胎牛血清诱导NSCs贴壁分化,同时用MOI为5的HCMV AD169株感染NSCs,7d后使用激光共聚焦显微镜免疫荧光方法检测Nestin、神经胶质纤维酸性蛋白(GFAP)和HCMV即刻早期蛋白(IE)的表达,计算阳性细胞比率。本实验所培养的细胞(4~6代)95±8%表达Nestin;分化诱导7d后,感染组86±12%细胞表达IE,未感染组和感染组Nestin阳性率分别为50±19%和93±10%(t=6.03,P<0.01),GFAP阳性细胞率分别为81±11%和55±17%(t=3.77,P<0.01)。以上结果表明分化过程中的NSCs是HCMV的容许细胞;HCMV感染可以抑制NSCs的分化。  相似文献

The human brain may be regarded as an irreversible system which is constrained by a fixed inflow of free energy in the form of chemical nourishment from within the body and information from the environment. The evolution of the internal spontaneous process may be described as a path on the saddle surface of entropy production rate in the configuration space of the process variables. From any initial state of high entropy production the system evolves towards the saddle point by a series of regessions to temporary minima alternating with fluctuations which introduce new internal constraints and open new channels for regression. The spontaneous regression steps of the process are to be associated with the learning process and the deductive processes of thought since information is necessarily being stored, whereas the fluctuation or nucleation steps are to be associated with the inductive or creative part of the thought process. The state of consciousness is to be associated with the system undergoing regression. If, under certain boundary conditions, the system attains the stable saddle point, a state of unconsciousness is attained in which no change of state variables occurs in time. The stability of this state is indicated by its resistance to perturbation.  相似文献

Five patients with traumatic colobomas of the eyelid secondary to human bites were surgically repaired with retrieved autogenous tissue. All patients were treated with prophylactic intravenous antibiotics. Surgical repair consisted of debridement of the autograft, meticulous layered closure of the autograft to the wound, and placement of a lid margin suture. In two of the patients, mild upper eyelid retraction was noted, and two patients had loss of cilia.  相似文献

Electrical activity was recorded in the human hypophysis from the neurosecretory fibres of the posterior lobe. In seven patients undergoing trans-sphenoidal hypophysectomy, electrophysiological recording was carried out with a special fine-shielded bipolar concentric electrode with a tip diameter of 30 microns. The rate of unit impulses was modified by anesthetic agents and after intravenous injections of drugs acting at the hypothalamic level.This technique was used to detect the limit between the anterior and posterior lobe in order to perform a selective adeno-hypophysectomy by implantation of yttrium-90 seeds.  相似文献

Background

MicroRNAs (miRNAs) are highly conserved, short (18–22 nts), non-coding RNA molecules that regulate gene expression by binding to the 3′ untranslated regions (3′UTRs) of mRNAs. While numerous cellular microRNAs have been associated with the progression of various diseases including cancer, miRNAs associated with retroviruses have not been well characterized. Herein we report identification of microRNA-like sequences in coding regions of several HIV-1 genomes.

Results

Based on our earlier proteomics and bioinformatics studies, we have identified 8 cellular miRNAs that are predicted to bind to the mRNAs of multiple proteins that are dysregulated during HIV-infection of CD4+ T-cells in vitro. In silico analysis of the full length and mature sequences of these 8 miRNAs and comparisons with all the genomic and subgenomic sequences of HIV-1 strains in global databases revealed that the first 18/18 sequences of the mature hsa-miR-195 sequence (including the short seed sequence), matched perfectly (100%), or with one nucleotide mismatch, within the envelope (env) genes of five HIV-1 genomes from Africa. In addition, we have identified 4 other miRNA-like sequences (hsa-miR-30d, hsa-miR-30e, hsa-miR-374a and hsa-miR-424) within the env and the gag-pol encoding regions of several HIV-1 strains, albeit with reduced homology. Mapping of the miRNA-homologues of env within HIV-1 genomes localized these sequence to the functionally significant variable regions of the env glycoprotein gp120 designated V1, V2, V4 and V5.

Conclusions

We conclude that microRNA-like sequences are embedded within the protein-encoding regions of several HIV-1 genomes. Given that the V1 to V5 regions of HIV-1 envelopes contain specific, well-characterized domains that are critical for immune responses, virus neutralization and disease progression, we propose that the newly discovered miRNA-like sequences within the HIV-1 genomes may have evolved to self-regulate survival of the virus in the host by evading innate immune responses and therefore influencing persistence, replication and/or pathogenicity.  相似文献

Since 1985, UNESCO studies ethical questions arising in genetics. In 1992, I established the International Bioethics Committee at UNESCO with the mission to draft the Universal Declaration on the Human Genome and Human Rights, which was adopted by UNESCO in 1997 and the United Nations in 1998. The Declaration relates the human genome with human dignity, deals with the rights of the persons concerned by human genome research and provides a reference legal framework for both stimulating the ethical debate and the harmonization of the law worldwide, favouring useful developments that respect human dignity.  相似文献

DNA replication in bacteria and eukaryotes requires the activity of DNA primase, a DNA-dependent RNA polymerase that lays short RNA primers for DNA polymerases. Eukaryotic and archaeal primases are heterodimers consisting of small catalytic and large accessory subunits, both of which are necessary for RNA primer synthesis. Understanding of RNA synthesis priming in eukaryotes is currently limited due to the lack of crystal structures of the full-length primase and its complexes with substrates in initiation and elongation states. Here we report the crystal structure of the full-length human primase, revealing the precise overall organization of the enzyme, the relative positions of its functional domains, and the mode of its interaction with modeled DNA and RNA. The structure indicates that the dramatic conformational changes in primase are necessary to accomplish the initiation and then elongation of RNA synthesis. The presence of a long linker between the N- and C-terminal domains of p58 provides the structural basis for the bulk of enzyme''s conformational flexibility. Deletion of most of this linker affected the initiation and elongation steps of the primer synthesis.  相似文献
