Hymenolepidid cestodes of synanthropic rodents represent a risk for public health. In order to describe the occurrence of hymenolepidids in children and the role of rodents as a potential source of infection, we conducted a morphological and molecular survey on cestodes in two rural villages from Yucatan, Mexico. One hundred and thirty-five stool samples from children (64 from Paraíso and 71 from Xkalakdzonot), 233 Mus musculus (159 from Paraíso and 74 from Xkalakdzonot) and 125 Rattus rattus (7 from Paraíso and 118 from Xkalakdzonot) were analyzed for the presence of cestodes. Three hymenolepidid species were identified morphologically: Hymenolepis nana in 7.8% of children from Paraíso, Hymenolepis microstoma in 4.4% of M. musculus from Paraíso and Hymenolepis diminuta in 15.3% of R. rattus from Xkalakdzonot. The molecular characterization and phylogenetic analysis based on mitochondrial cytochrome c subunit 1 (CO1) gene and ribosomal internal transcribed spacer 1 (ITS1) region, confirmed the identity of the three cestodes isolated from Yucatan. Phylogeny of the CO1 gene identified intraspecific genetic differences within H. nana ranging from 0 to 5%, in H. microstoma from 0 to 0.4%, and in H. diminuta ranged from 0 to 6.5% which suggests, the presence of complex species within H. nana and H. diminuta infecting humans and rodents, as reported by other authors. Based on the morphological and molecular results, and the epidemiological evidence, infections with H. nana suggest a non-zoonotic transmission; however, the presence of H. microstoma and H. diminuta in synanthropic rodents serve as a possible source for human infection.  相似文献

Complement-mediated lysis in vitro of newly excysted tapeworms: Hymenolepis diminuta, Hymenolepis microstoma, Hymenolepis nana and Hymenolepis citelii. International Journal for Parasitology16: 157–161. Newly excysted worms of Hymenolepis diminuta were lysed in 50% normal serum from all 13 animal species tested, including man. Since H. diminuta was neither lysed in complement inactivated serum—by heat or adding EDTA, LPS or CVF—nor in C5-deficient mouse serum, it is concluded that the lysis was associated with the complement cascade. It is shown that H. diminuta can activate the complement system via both the classical and alternative pathway. Furthermore, it is indicated that the lysis is independent of serum antibodies. Hymenolepis nana and H. citelli were also lysed in all normal sera tested, eight and six respectively, while newly excysted worms of H. microstoma were lysed in normal sera from 10 mammals and birds, but not in sera from its hosts, the mouse, rat and golden hamster. This indicates that the complement system of these three species differs from that of the other species tested in such a way that H. microstoma is able to avoid lysis in these sera.  相似文献

The activities of 5-nucleotidase (E.C. 3.1.3.5), adenosine deaminase (E.C. 3.5.4.4), adenosine kinase (E.C. 2.7.1.20), AMP deaminase (E.C. 3.5.4.6) and adenylate kinase (E.C. 2.7.4.3) were demonstrated in homogenates of Hymenolepis diminuta. The Km values for adenosine and AMP of the above enzymes were determined. The effects of several antibacterial or antifungal antibiotics on the growth of 3-day-old Hymenolepis diminuta cultivated in vitro were investigated. Worms were recovered from the rat and cultured in roller tubes for 4 days without a medium change. The medium contained horse serum, yeast extract, and liver extract; the gas phase was 5% CO2 in N2. It was found that penicillin and streptomycin did not inhibit the growth of worms at concentrations lower than 5000 units and μg/ml of medium, respectively. Cycloheximide was toxic to H. diminuta, retarding or inhibiting growth at levels higher than 1.5 μg/ml. The antifungal antibiotics nystatin and amphotericin B did not affect worm growth at concentrations lower than 1000 units and 123 μ/ml of medium, respectively.The use of penicillin and streptomycin, and nystatin or amphotericin B can be recommended for the control of bacterial and fungal contaminants in the cultivation of H. diminuta removed from the rat.  相似文献

The inter-relationship between the amino acid serine and lipid metabolism in the rat tapeworm Hymenolepis diminuta has been studied under in vitro conditions. The label from U-14C-serine, U-14C-glucose and 1-14C-oleic acid was rapidly incorporated into worm tissue phospho- and glycolipids, the latter illustrating the synthesis of cerebrosides by H. diminuta. Activity from U-14C-serine was recovered in phosphatidylserine, phosphatidylethanolamine, cerebrosides and several unidentified lipid-like compounds. The majority of the label recovered in phosphatidylethanolamine was associated with the ethanolamine moiety; in the cerebrosides with the sphingosine moiety. The sugar moiety of the cerebrosides was galactose.Pulse label studies showed a serine flux phenomenon, and a rapid rate of turnover of some of the unidentified compounds.Exogenous ethanolamine had no detectable effect upon absorption and conversion of serine to tissue phosphatidylethanolamine. Incubation of H. diminuta homogenates with phosphatidyl U-14C-serine resulted in the recovery of considerable activity in phosphatidylethanolamine. The results show that the major pathway of phosphatidylethanolamine synthesis is by decarboxylation of phosphatidylserine.  相似文献

We report a case of Hymenolepis diminuta infection in a 2-year-old child living in a suburban area of Catania, Italy. This case was initially referred to us as Dipylidium caninum infection, which was not cured after being treated twice with mebendazole. However, by analyzing the clinical presentation and stool samples we arrived to the diagnosis of H. diminuta infection. The case presented with atypical allergic manifestations which had never been reported as clinical features of symptomatic H. diminuta infection; remittent fever with abdominal pain, diffuse cutaneous itching, transient thoracic rash, and arthromyalgias. The patient was treated with a 7-day cycle of oral niclosamide, which proved to be safe and effective. This case report emphasizes that a correct parasitological diagnosis requires adequate district laboratories and trained personnel. In addition, we recommend the importance of reporting all H. diminuta infection cases, in order to improve knowledge on epidemiology, clinical presentation, and treatment protocols.  相似文献

In this study, we examine the effect of Hymenolepis diminuta on ion transport in the ileum and on tight junctions in the ileum and colon of rats. We also evaluate the effect of H. diminuta on C-fiber endings in the ileum, the direct habitat of H. diminuta, before and after mechanical stimulation and pharmacological modification by capsaicin (C-fiber irritant).Wistar rats were orally infected with five cysticercoids of H. diminuta. Using a modified Ussing chamber, electrophysiological parameters of the ileum were measured (transepithelial electrical potential difference and transepithelial electrical resistance) as well as the deposition of occludin (a tight junction protein) in the ileum and colon of the rats 8, 16, 25, 35, 40 and 60 days post infection.We observed a significant reduction in transepithelial electrical potential difference in the ileum of rats infected with H. diminuta. In both the ileum and colon of rats infected with H. diminuta we also observed a decrease in occludin deposition, which indicates leakage of tight junctions, correlating with the decrease in transepithelial electrical resistance of these tissues. The application of capsaicin confirmed the hypothesis that H. diminuta in rats affects the C-fiber sensory receptors, causing changes in ion transport in the ileum.The results of the performed electrophysiological and immunohistochemical examinations indicate hymenolepidosis-related changes in the active transport of ions and the passive movement of ions.  相似文献

To determine geographical patterns of natural parasite infections among wild rodents, a total of 46 wild rodents from 3 different localities in northern Gangwon-do (Province), Korea were examined for intestinal parasite infections. Along with nematodes such as hookworms and Syphacia spp., Plagiorchis muris (2 specimens) (Trematoda) were collected from striped field mice, Apodemus agrarius. In a Korean wood mouse, Apodemus peninsulae, the overall nematode infections were similar to A. agrarius, but an adult worm of Echinostoma hortense (Trematoda) was collected. In addition, 2 species of cestodes, i.e., Hymenolepis nana and Hymenolepis diminuta, were collected from A. agrarius. Through this survey, A. agrarius and A. peninsule were confirmed as the natural definite hosts for zoonotic intestinal helminths, i.e., P. muris, E. hortense, H. nana, and H. diminuta, in northern Gangwon-do, Korea. Considering increased leisure activities around these areas, seasonal and further comprehensive surveys on wild rodents seem to be needed to prevent zoonotic parasite infections.  相似文献

Background

The aim of the present paper was to describe the enzymatic antioxidant system in Hymenolepis diminuta collected from rats exposed to chronic cestode invasion.

Methodology

We dissected different tissues of H. diminuta (immature proglottids, genital primordia, hermaphroditic proglottids, early uterus, and gravid uterus) and studied activity of: superoxide dismutases (SOD1 and SOD2), catalase (CAT), glutathione peroxidases (non-Se-dependent GSHPx and Se-dependent GSHPx), glutathione-S-transferase (GST) and glutathione reductase (GSHR), and oxidative stress markers ?? reduced glutathione (GSH), and the lipid peroxidation level (TBARS).

Results

We demonstrated changes in antioxidant enzyme activities and levels of oxidative stress markers in different tissues of the parasite. The levels of TBARS and GSH indicate that oxidative stress occurred in tissues located proximal to the intestine wall. Activity of SOD1 was high in all parts of H. diminuta, but the GST activity was the highest of all studied antioxidant enzymes. SOD2 activity differed significantly in various parts of H. diminuta. Significant differences were observed for nonSeGSHPx and activity of other GSH-dependent enzymes was generally similar in all the tissues.

Conclusions

Our results show that the enzymatic antioxidant system of H. diminuta, allows the parasite to adapt and live under conditions of chronic oxidative stress. It suggests an oxidative-antioxidative balance during interactions between parasite and host.  相似文献

Immunological damage to Hymenolepis diminuta following a challenge infection in C57 mice. International journal for Parasitology17: 795–803. Ultra-structural observations were made on the scpleces of Hymenolepis diminuta from 10-worm 2° infections in C57 mice to determine whether the immune stress causing reduced establishment, stunting of growth and early rejection was accompanied by significant damage to the worm fine structure. In almost all of the worms recovered there was extensive damage to tissues throughout the scolex, even in those examined 24 h post-secondary-infection (p-2°-i). Many of the muscle, tegumental and parenchyma cells were completely disrupted and dispersed, leaving only isolated strands of cytoplasm and cell debris. A number of excretory ducts were breached, and large and apparently empty spaces were evident throughout the scolex tissues. There was an increase in lipid accumulation in the tegumental syncytium, underlying cytons and in the suckers. Extensive vacuolation also occurred in the sucker tegument. Although some scoleces showed certain regions to be apparently undamaged, close examination revealed a fine structure consistent with enhanced metabolic activity. The structural abnormalities observed are discussed in relation to host immunity.  相似文献

We provided the first known evidence of the presence and release of extracellular vesicles in adults of important model tapeworm Hymenolepis diminuta. Two different subtypes have been observed on the surface of the worm and among the secretory products confirmed by several microscopical methods. Proteomic analysis revealed the presence of parasite-specific proteins as well as those of the host in purified extracellular vesicles. Among the protein cargo, we identified potential drug targets, vaccine candidates and H. diminuta antigens. Finally, the protein composition further revealed proteins participating in the endosomal complex required for transport-dependent biogenesis pathway.  相似文献

An investigation of the chemotherapeutic and biochemical effects of a new broad-spectrum anthelmintic, fenbendazole, (FBZ, methyl 5-[phenylthio]-benzimadazole-2-carbamate), on Hymenolepis diminuta in experimentally infected rats is reported. FBZ proved to be highly active against H. diminuta; a single oral dose of 12.5, 25 and 50 mg/kg body wt. on day 15 of infection eliminated 77, 100 and 88% of the tapeworms respectively as determined at necropsy 24 h after treatment. The chemotherapeutic actions of FBZ on H. diminuta in vivo were accompanied by marked changes in worm wt. and chemical composition. Tapeworms recovered from rats that had received a therapeutically effective dose of FBZ 18 h earlier were significantly smaller and contained much less glycogen (as a percent of the fresh wt.) than worms from unmedicated controls. Protein concentrations rose in FBZ-treated worms, but more slowly than the rate of decline in glycogen concentration. Glycogen/protein ratios in FBZ-treated worms were considerably lower than the corresponding control values. Differences in the absolute amounts of glycogen and protein between control and drug-treated worms were even more profound. Administration of a single oral dose of FBZ (14 mg/kg) to the rat produced in H. diminuta another change, the onset of which coincided with, or preceded, the gross alterations in worm wt. and chemical composition. In vitro studies, carried out 16 h after treatment, revealed that FBZ-treated worms absorbed and metabolized much smaller quantities of exogenous glucose than did the controls, and the ability to the worm to accumulate glucose against a concentration difference was significantly depressed.  相似文献

The present study sought to assess the potential of the cestode Hymenolepis diminuta as a bioindicator for lead accumulation in two industrial areas of the city of Riyadh, Saudi Arabia. Rats (Meriones libycus) were collected from two sites (industrial area II and Salbukh) in Riyadh. In the industrial area II, the mean levels of lead concentrations were found to be 1.96, 1.92, 1.4 and 30.72 μg/g in the rats’ liver, kidney and intestine, and in H. diminuta, respectively. In Salbukh, meanwhile, the lead concentrations were 1.63, 1.52, 1.20 and 21.31 μg/g in the rats’ liver, kidney, and intestine, and in H. diminuta, respectively. In addition, in industrial area II, compared with the liver, kidney and intestine of their host, the bioconcentration factors of lead were found to be, respectively, 15.6, 16 and 21.9 times higher in H. diminuta, and were 7.5, 8, and 10.2 times higher in the same organs compared to H. diminuta in Salbukh. The present study, therefore, proved that H. diminuta could be used as a bioindicator for heavy metal contamination in the industrial areas of the city of Riyadh.  相似文献

Roepstorff A. and Andreassen J. 1982. Course of heavy primary infections and earlier immunologically mediated rejection of secondary infections of Hymenolepis diminuta in mice. International Journal for Parasitology12: 23–28. The worms of heavy (50–100 worms) primary Hymenolepis diminuta infections in inbred C57-mice were 1–2 mm long when growth ceased about day 4. Thereafter the mean length decreased by shrinkage and/or ‘decollation’, the worms moved backwards in the small intestine and were rejected from day 6 to day 10. Heavy secondary infections given 14 days after a heavy primary infection were severely stunted (0.2–0.3 mm) but normally situated in the intestine on day 2 and nearly all were rejected by day 4. Even when the time between the primary and secondary infections was increased to 21 or 42 days, therecovery, position and length of the secondary worms were significantly different from primary infections. These results show that an immunologically mediated memory was involved, and that functional antigens can be released from the scolex and/or the neck alone.  相似文献

The metabolism in vitro of U-13C-glucose and NaH13CO3 by two strains of adult Hymenolepis diminuta, the ANU and UT strains, was examined using 13C n.m.r. spectroscopy. The incubation medium and perchlorate extracts from worms incubated in vitro with U-13C-glucose showed incorporation of significant quantities of label into the end products succinate, lactate and acetate, and also into glycogen. Similar experiments with NaH13CO3 showed incorporation principally into succinate C-1,4, plus significant labelling also in lactate C-1. This shows that nutochondrial malate or pyruvate contributes to the cytosolic pyruvate pool in H. diminuta. The metabolism of U-13C-glucose was followed directly by incubating live worms directly in the spectrometer. Worms from 24 h-fasted hosts metabolised the added glucose completely during an experimental period of 2 h and incorporation of label was evident in the time course spectra. Parasites from fed hosts metabolised the added glucose more slowly. This work confirms the accepted routes of glucose metabolism in H. diminuta and demonstrates the utility of the n.m.r. technique in investigating the metabolism of parasites.  相似文献

Peroxidase and phosphatase activities have been reported to be localized in the tegument of adult hymenolepidid tapeworms. In order to localize adenyl cyclase (EC 4.6.1.1) activity in the tegument of mature and gravid sections of Hymenolepis microstoma, 5-adenyl-imidodiphosphate was used as a substrate, and lead was used as a capturing agent. Results indicate that adenyl cyclase activity is present in the crypts between the microtriches of the mature sections and that activity is absent from the gravid sections.  相似文献

Lethbridge R. C. 1976. The architecture of the eggshell of Hymenolepis diminuta. International Journal for Parasitology6: 87–90. Scanning and transmission electron microscopy demonstrated that the eggshell of H. diminuta consisted of an inner continuous band from which arose an outer zone of finger-like projections with rounded tips. These tips were revealed as globule-like units lacking a regular spatial distribution but entirely covering the eggshell's surface. The appearance of these structures suggested that the shell constituents may have been deposited in an initially plastic state and possible methods of attaining the final architecture of the hardened eggshell have been discussed on the basis of this hypothesis.  相似文献

Interleukin (IL)-22, an immune cell-derived cytokine whose receptor expression is restricted to non-immune cells (e.g. epithelial cells), can be anti-inflammatory and pro-inflammatory. Mice infected with the tapeworm Hymenolepis diminuta are protected from dinitrobenzene sulphonic acid (DNBS)-induced colitis. Here we assessed expulsion of H. diminuta, the concomitant immune response and the outcome of DNBS-induced colitis in wild-type (WT) and IL-22 deficient mice (IL-22-/-) ± infection. Interleukin-22-/- mice had a mildly impaired ability to expel the worm and this correlated with reduced or delayed induction of TH2 immunity as measured by splenic and mesenteric lymph node production of IL-4, IL-5 and IL-13 and intestinal Muc-2 mRNA and goblet cell hyperplasia; in contrast, IL-25 increased in the small intestine of IL-22-/- mice 8 and 12 days post-infection compared to WT mice. In vitro experiments revealed that H. diminuta directly evoked epithelial production of IL-25 that was inhibited by recombinant IL-22. Also, IL-10 and markers of regulatory T cells were increased in IL-22-/- mice that displayed less DNBS (3 mg, ir. 72h)-induced colitis. Wild-type mice infected with H. diminuta were protected from colitis, as were infected IL-22-/- mice and the latter to a degree that they were almost indistinguishable from control, non-DNBS treated mice. Finally, treatment with anti-IL-25 antibodies exaggerated DNBS-induced colitis in IL-22-/- mice and blocked the anti-colitic effect of infection with H. diminuta. Thus, IL-22 is identified as an endogenous brake on helminth-elicited TH2 immunity, reducing the efficacy of expulsion of H. diminuta and limiting the effectiveness of the anti-colitic events mobilized following infection with H. diminuta in a non-permissive host.  相似文献

Absorption kinetics of [14C]glucose and [β-methyl-14C]glucoside in Hymenolepis diminuta are reported. β-Methylglucoside (βMG) is a pure competitive inhibitor of [14C]glucose transport and has kinetic parameters, Vmax and Kt, for transport similar to those reported for glucose. While absorbed 14C-βMG is not metabolized, transport of this glucose analog retains the general characteristics which have been established for glucose transport including: (1) Na+ dependence, (2) inhibition by K+, (3) sensitivity to phlorizin and various hexoses, (4) transport against an apparent concentration gradient, and (5) increase in worm water during accumulation. It is concluded that glucose and βMG are transported by the same system. The value of using βMG to study the mechanism of hexose transport and accumulation in H. diminuta is suggested.  相似文献

In the Canary Islands two invasive rat species, Rattus rattus and Rattus norvegicus are present, but little is known about the origin and colonization. To this end, a molecular study was performed on R. rattus from the Archipelago and from the nearest continents. Partial cytochrome b gene sequencing offered very low levels of haplotype and nucleotide diversities, with only seven haplotypes identified. All of them belong to the European Lineage I, specifically to the “ship rat” cluster. The haplotype network showed a star-like topology. Haplotype distribution showed a genetic subdivision between eastern and central/western islands, suggesting a double colonization event. This hypothesis is congruent with historical human colonization and it is similar to that proposed for the rodent parasite Hymenolepis diminuta. In addition, a possible role of the Canary Islands as a faunal link with the European and American continents is discussed.  相似文献
