Monovalent ion and calcium ion fluxes in sarcoplasmic reticulum   总被引:7,自引:0,他引:7  
Summary The ion permeability of sarcoplasmic reticulum vesicles from skeletal and heart muscle has been characterized by radioisotope flux, osmotic and membrane potential measurements, and by incorporating vesicles into planar phospholipid bilayers. The sarcoplasmic reticulum membrane is uniquely permeable to various biologically relevant monovalent ions. At least two and possibly three separate passive permeation systems for monovalent ions have been identified: 1) a K+, Na+ channel, 2) an anion channel, and 3) a H+ (OH) permeable pathway which may or may not be synonymous with the anion channel. A possible physiological function of these monovalent ion permeation systems is to permit rapid movement of K+, Na+, H+ and Cl across the membrane to counter electrogenic Ca2+ fluxes during Ca2+ release and uptake by sacroplasmic reticulum.  相似文献

Rat gastric membranes enriched in (H+-K+)-ATPase, when prepared in the presence of 1 mM ethyleneglycol-bis-(β-aminoethyl ether)N,N′-tetraacetic acid, showed the ability to accumulate H+ ions upon addition of ATP, KCl, and valinomycin. The membranes were largely impermeable to K+ and Cl?. In contrast, the rat membranes prepared without the Ca2+ chelator lost the ability to develop a pH gradient because of the membrane leakiness to H+. A majority of these membrane vesicles became also permeable to K+. We suggest that the calcium chelator preserved the gastric membrane permeability barrier during isolation by inhibiting various Ca2+-dependent phospholipases in rat gastric mucosa.  相似文献

Cholesterol is an essential component of lysosomal membranes. In this study, we investigated the effects of membrane cholesterol on the permeability of rat liver lysosomes to K+ and H+, and the organelle stability. Through the measurements of lysosomal β-hexosaminidase free activity, membrane potential, membrane fluidity, intra-lysosomal pH, and lysosomal proton leakage, we established that methyl-β-cyclodextrin (MβCD)-produced loss of membrane cholesterol could increase the lysosomal permeability to both potassium ions and protons, and fluidize the lysosomal membranes. As a result, potassium ions entered the lysosomes through K+/H+ exchange, which produced osmotic imbalance across the membranes and osmotically destabilized the lysosomes. In addition, treatment of the lysosomes with MβCD caused leakage of the lysosomal protons and raised the intra-lysosomal pH. The results indicate that membrane cholesterol plays important roles in the maintenance of the lysosomal limited permeability to K+ and H+. Loss of this membrane sterol is critical for the organelle acidification and stability.  相似文献

Summary Above a critical external pH (about 10.5), theChara membrane acquires new propertes. In this state the membrane potential is close to the equilibrium potentials for H+ and OH, hyperpolarizing as external pH increases with a slope of –59 mV/pH unit. The membrane conductance increases by an average factor of 2.4 above the critical pH. These changes are explained by an increase in permeability to OH (or H+). The establishment of a OH (or H+ permeable membrane at high pH suggests that the large fluxes of OH (or H+ which occur in the alkaline band in photosynthesizing cells are passive.  相似文献

In pure phospholipid vesicles, the conductivity of H+/OH ions exceeds that for other simple inorganic ions. Protons achieve electrochemical equilibrium across egg phosphatidylcholine vesicles within tens of minutes. When pH gradients are established across vesicles, transmembrane potentials develop. Conversely, the establishment of transmembrane potentials leads to the formation of pH gradients. When the phenomenological permeability of H+/OH ions in vesicles is estimated, values are obtained that are much greater (six orders of magnitude larger) than those for Na+ or K+. A wide range in the values for this permeability has been reported; however, much of the discrepancy can be attributed to differences in the vesicle systems and experimental conditions. The H+/OH current appears to be modulated by changes in membrane dielectric constant. However, the dependence of this current on the pH gradient and on the membrane voltage argues against simple diffusion mechanisms as the source of the H+/OH current. In addition, in vesicle systems the H+/OH current shows a surprising invariance to changes in the membrane dipole potential, an observation that argues against the role of simple carriers for H+ and OH ions.  相似文献

Prooxidants induce release of Ca2+ from mitochondria through the giant solute pore in the mitochondrial inner membrane. However, under appropriate conditions prooxidants can induce Ca2+ release without inducing a nonspecific permeability change. Prooxidant-induced release of Ca2+ isselective. Presumably, this is the result of the operation of a permeability pathway for H+ coupled to the reversal of the Ca2+ uniporter, the latter generating the selectivity. The solute pore and prooxidant-induced Ca2+-specific pathways exhibit common sensitivities to a set of inhibitors and activators. It is proposed that the pore can operate in two open states: (1) permeable to H+ only and (2) permeable to solutes of Mr<1500. Under some conditions, prooxidants induce the H+-selective state which, in turn, collapses the inner membrane potential and permits selective loss of Ca2+ via the Ca2+ uniporter.  相似文献

The permeability of the inner membrane of turnip mitochondria to H+ and OH- ions has been investigated using an acid pulse technique. The rate of decay of a H+ pulse across the inner membrane is exponential having first-order kinetics and gives t 1/2 values of approx 54 s at neutral pH and at 25° C. Valinomycin or 1799 alone have little effect on t 1/2 values, whereas in combination, values of <15 s are observed. Nigericin produces a similar effect. The effective proton conductance of the inner membrane near pH 7 at 25° C is 0.27 nmol H+ min-1 mg protein-1 mV-1. The results suggest that at neutral pH, the inner membrane of plant mitochondria is relatively impermeable to H+ and OH- ions.  相似文献

The permeability transition pore (PTP) is a Ca2+‐dependent mitochondrial channel whose opening causes a permeability increase in the inner membrane to ions and solutes. The most potent inhibitors are matrix protons, with channel block at pH 6.5. Inhibition is reversible, mediated by histidyl residue(s), and prevented by their carbethoxylation by diethylpyrocarbonate (DPC), but their assignment is unsolved. We show that PTP inhibition by H+ is mediated by the highly conserved histidyl residue (H112 in the human mature protein) of oligomycin sensitivity conferral protein (OSCP) subunit of mitochondrial F1FO (F)‐ATP synthase, which we also show to undergo carbethoxylation after reaction of mitochondria with DPC. Mitochondrial PTP‐dependent swelling cannot be inhibited by acidic pH in H112Q and H112Y OSCP mutants, and the corresponding megachannels (the electrophysiological counterpart of the PTP) are insensitive to inhibition by acidic pH in patch‐clamp recordings of mitoplasts. Cells harboring the H112Q and H112Y mutations are sensitized to anoxic cell death at acidic pH. These results demonstrate that PTP channel formation and its inhibition by H+ are mediated by the F‐ATP synthase.  相似文献

This paper examines the applicability of the Nernst-Planck approach in treating the relationship between the initial rates at which critical ions interexchange across permselective membranes in bi-ionic systems and the rates of self-exchange of these ions across the same membranes. Data are presented for five species of univalent cations with two types of cation permeable membranes, a polystyrene sulfonic acid-collodion matrix membrane, and an oxidized collodion membrane; five species of univalent anions were studied with a protamine-collodion matrix anion permeable membrane. Except with systems involving H+ ion, the experimentally found relationships between the rates of interexchange and the rates of self-exchange were in agreement, in most cases within ±5%, with the values calculated from an expression in which interaction between the critical ions in the membrane is not taken into account. In systems with H+ ion, the experimental rates of interexchange were from 27% to 40% less than calculated values.  相似文献

A topic emerging roughly 30 years ago and engendering an incompletely resolved controversy is reviewed in this article: the relatively high permeability and pH independence associated with H+/OH passive movements across lipid membranes. We summarize the expected characteristics of simple H+/OH diffusion and those of a reaction between H+ and OH being attracted from opposite surfaces and condensing in an interfacial zone of the membrane. An interfacial H+/OH reaction mechanism gives the experimentally observed behavior of an H+/OH flux that is independent of the pH measurement range. This mechanism assumes that H+ and OH within the interfacial zone become electrostatically aligned on opposite sides of the hydrophobic membrane core. Electrostatic attraction and charge delocalization among a small cluster of water molecules surrounding the ions reduce the Born energy for H+/OH insertion into lipid. This transmembrane condensation model predicts the magnitude of the experimentally determined H+/OH flux, which is significantly greater than that of other monovalent ions. The consequences of an elevated H+/OH permeability compared to other ions and the relative pH independence of this flux have consequences for understanding the chemical evolution of life.  相似文献

The incorporation of porin protein F from the outer membrane of Pseudomonas aeruginosa into artificial lipid bilayers results in an increase of the membrane conductance by many orders of magnitude. The membrane conductance is caused by the formation of large ion-permeable channels with a single-channel conductance in the order of 5 nS for 1 M alkali chlorides. The conductance has an ohmic current vs. voltage relationship. Further information on the structure of the pore formed by protein F was obtained by determining the single-channel conductance for various species differing in charge and size, and from zero-current potential measurements. The channel was found to be permeable for large organic ions (Tris+, N(C2H5)4+, Hepes?) and a channel diameter of 2.2 nm could be estimated from the conductance data (pore length of 7.5 nm). At neutral pH the pore is about two times more permeable for cations than for anions, possibly caused by negative charges in the pore. The consistent observation of large water filled pores formed by porin protein F in model membrane systems is discussed in the light of the known low permeability of the Ps. aeruginosa outer membrane towards antibiotics. It is suggested that this results from a relatively low proportion of open functional porin protein F pores in vivo.  相似文献

Increasing the hydrogen ion concentration of the bathing medium reversibly depresses the sodium permeability of voltage-clamped frog nerves. The depression depends on membrane voltage: changing from pH 7 to pH 5 causes a 60% reduction in sodium permeability at +20 mV, but only a 20% reduction at +180 mV. This voltage-dependent block of sodium channels by hydrogen ions is explained by assuming that hydrogen ions enter the open sodium channel and bind there, preventing sodium ion passage. The voltage dependence arises because the binding site is assumed to lie far enough across the membrane for bound ions to be affected by part of the potential difference across the membrane. Equations are derived for the general case where the blocking ion enters the channel from either side of the membrane. For H+ ion blockage, a simpler model, in which H+ enters the channel only from the bathing medium, is found to be sufficient. The dissociation constant of H+ ions from the channel site, 3.9 x 10-6 M (pKa 5.4), is like that of a carboxylic acid. From the voltage dependence of the block, this acid site is about one-quarter of the way across the membrane potential from the outside. In addition to blocking as described by the model, hydrogen ions also shift the responses of sodium channel "gates" to voltage, probably by altering the surface potential of the nerve. Evidence for voltage-dependent blockage by calcium ions is also presented.  相似文献

The permeability properties of frog rod photoreceptor outer segment discs were investigated in preparations of purified, dark-adapted, outer segment fragments by the techniques of direct volume measurement and electron microscopy. Outer segment discs were found to swell and contract reversibly in response to changes in the osmotic pressure of the bathing medium in accordance with the Boyle-van't Hoff law. By use of the criterion of reversible osmotic swelling, the disc membrane is impermeable to Na+, K+, Mg+2, Ca+2, Cl-, and (PO4)-3 ions, whereas it is freely permeable to ammonium acetate. The disc membrane is impermeable to sucrose, although its osmotic behavior towards this substance is different from its behavior towards impermeable ions. Electron microscopy showed that the osmotic effects on the rod outer segment fragments represent changes in the intradiscal volume. Fixation with glutaraldehyde did not abolish the permeability properties of the disc membrane, and fixed membranes were still capable of osmotic volume changes. It is concluded from this study that the frog's rod photoreceptor outer segment discs are free-floating membranous organelles with an inside space separate and distinct from the photoreceptor intracellular space.  相似文献

The simulated system consisted of a fatty acid bilayer membrane dividing two electrolyte layers each containing ions, and a channel composed of linked 15-crown-5 ether rings. The Na+ and F ions in the aqueous electrolyte layers were too large to enter the channel, but the Li+ ions entered and were transported. Conditions that optimised the passive, electric-field-induced transport of Li+ ions through the channel were investigated. It was calculated and rationalised that the higher the numerical value of the electrostatic charge on the oxygen atoms of the crown ether rings, the more easily does the channel convey the Li+ ions.  相似文献

Summary Measurements of the transepithelial potential (Vint-Vext) across the gills of Brown Trout,Salmo trutta, were made in solutions of a range of pH and calcium concentrations. The potential was strongly dependent on external pH, being negative in neutral solutions but positive in acid solutions. The addition of calcium to the external medium produced a positive shift in potential in all but very acid media (pH 4.0–3.5), where very little change was seen. The gill membrane appears to act as a hydrogen electrode having a very high permeability to H+ ions, and the potential behaves as a diffusion potential. The presence of calcium reduced the permeability to both H+ and Na+ ions but even at a calcium concentration of 8.0 mM/l the permeability ratio H+/Na+ was still more than 900. The transepithelial potential is shown to be diffusional in origin and is discussed in terms of the relative permeability of the gill to H+, Na+ and Cl ions. Sodium fluxes across the gills were measured and provide the basis for a theoretical consideration of Na+, Cl and H+ fluxes across the gills in neutral and acid solutions. The positive potential at low pH largely accounts for the increased loss of sodium from fish in these conditions.  相似文献

Elevated levels of intracellular Ca2+ activate a K+-selective permeability in the membrane of human erythrocytes. Currents through single channels were analysed in excised inside-out membrane patches. The effects of several ions that are known to inhibit K+ fluxes are described with respect to the single-channel events. The results suggest that the blocking ions can partly move into the channels (but cannot penetrate) and interact with other ions inside the pore. The reduction of single-channel conductance by Cs+, tetraethylammonium and Ba2+ and of single-channel activity by quinine and Ba2+ is referred to different rates of access to the channel. The concentration- and voltage-dependent inhibition by ions with measurable permeability (Na+ and Rb+) can be explained by their lower permeability, with single-file movement and ionic interactions inside the pore.  相似文献

Permeability properties of reconstituted rabbit skeletal muscle sarcoplasmic reticulum vesicles were characterized by measuring efflux rates of [3H]inulin, [3H]choline+, 86Rb+, and 22Na+, as well as membrane potential changes using the voltage-sensitive probe, 3,3′-dipentyl-2,2′-oxacarbocyanine. Native vesicles were dissociated with deoxycholate and were reconstituted by dialysis. Energized Ca2+ accumulation was partially restored. About 12 of the reconstituted vesicles were found to be ‘leaky’, i.e., permeable to choline+ or Tris+ but not to inulin. The remaining reconstituted vesicles were ‘sealed’, i.e., impermeable to choline+, Tris+ and inulin. Sealed reconstituted vesicles could be further subdivided according to their K+, Na+ permeability. About 12, previously designated Type I, were readily permeable to K+ and Na+, indicating the presence of the K+, Na+ channel of sarcoplasmic reticulum. The remaining sealed vesicles (Type II) formed a permeability barrier to K+ and Na+, suggesting that they lacked the K+, Na+ channel. These studies show that the K+, Na+ channel of sarcoplasmic reticulum can be solubilized with detergent and reconstituted with retention of activity. Furthermore, our results suggest that part or all of the decreased Ca2+-loading efficiency of reconstituted vesicles may be due to the presence of a significant fraction of leaky vesicles.  相似文献

The effect of pH on the interfacial tension of a sphingomyelin membrane in aqueous solution has been studied. Three models describing H+ and OH ion adsorption on the bilayer lipid surface are presented. In models I and II, the membrane surface is continuous, with uniformly distributed functional groups as centers of H+ and OH ion adsorption. In model III, the membrane surface is composed of lipid molecules, with and without adsorbed H+ and OH ions. The contribution of each individual lipid molecule to the overall interfacial tension of the bilayer was assumed to be additive in models I and II. In model III, the Gibbs isotherm was used to describe adsorption of H+ and OH ions at the bilayer surface. Theoretical equations are derived to describe the interfacial tension as a function of pH for all three models. Maximum interfacial tension was observed experimentally at the isoelectric point.  相似文献

This article will cover historical and recent aspects of reactions and mechanisms involved in the auxin-induced signalling cascade that terminates in the dramatic elongation growth of cells and plant organs. Massive evidence has accumulated that the final target of auxin action is the plasma membrane H+-ATPase, which excretes H+ ions into the cell wall compartment and, in an antiport, takes up K+ ions through an inwardly rectifying K+ channel. The auxin-enhanced H+ pumping lowers the cell wall pH, activates pH-sensitive enzymes and proteins within the wall, and initiates cell-wall loosening and extension growth. These processes, induced by auxin or by the "super-auxin" fusicoccin, can be blocked instantly and specifically by a voltage inhibition of the H+-ATPase due to removal of K+ ions or the addition of K+-channel blockers. Vice versa, H+ pumping and growth are immediately switched on by addition of K+ ions. Furthermore, the treatment of segments either with auxin or with fusicoccin (which activates the H+-ATPase irreversibly) or with acid buffers (from outside) causes an identical transformation and degradation pattern of cell wall constituents during cell-wall loosening and growth. These and other results described below are in agreement with the acid-growth theory of elongation growth. However, objections to this theory are also discussed.  相似文献
