A large number of Penicillium nalgiovense strains were found to be as good penicillin producers on optimal production media as strains of Penicillium chrysogenum. It is not known whether penicillin can be produced on fermented sausages, but selection for non-penicillin producing fermenting strains of P. nalgiovense is suggested.  相似文献

Summary Penicillin was produced by a non-sterile solid state fermentation (SSF) on bagasse impregnated with culture medium. The use of concentrated media greatly enhanced the antibiotic production in this system. It was observed that adequate initial moisture content (70%) of the impregnated solid medium results in higher production. A comparison between solid and liquid fermentation showed superior yield and productivity.  相似文献

Enzyme production with E. coli ATCC 11105, in a complex medium using phenylacetic acid as inducer is carried out in a stirred-tank reactor of 10 dm3 and an airlift tower-loop reactor of 60 dm3 with outer loop at a temperature of 27 °C. The optimum inducer concentration was 0.8 kg/m3, which was kept constant by fed-batch operation. The optimum of the relative dissolved O2-concentration with regard to saturation is below 10% in a stirred-tank reactor and at 35% in a tower-loop reactor. It was kept constant by parameter-adaptive control of the aeration rate. Summary The activity of penicillin G production was compared in a well defined medium by native vesicles as well as by calcium alginate gel immobilized vesicles isolated from the protoplasts of Penicillium chrysogenum PQ-96. The activity yield of the immobilized vesicles was 44% in comparison with native vesicles. After 60 h of storage at 4° C the native vesicles showed a rapid decrease in penicillin G production. The storage stability of these vesicles was improved after entrapment inside the calcium alginate gel. After 240 h of storage 1 mg of immobilized vesicular protein catalyzed the production of about 140 nmoles of penicillin G in 1 h.  相似文献

Summary Wild-type strains ofPenicillium chrysogenum produce lower penicillin V titers in media containing excess glucose. Two mutant strains were isolated and shown to produce normal penicillin V titers in the presence of excess glucose. These strains, designated as glucose-repression insensitive (GRI) mutants, produced higher penicillin V titers than the wild-type strain in media containing lactose as the main carbohydrate source. In lactose-based media, the production of penicillin V was depressed to a much lesser extent by in-cycle additions of glucose with the GRI mutants when compared to the wild-type strain. In short-term biosynthesis experiments using washed cells in a medium containing glucose as the sole carbon source, the GRI mutants produced penicillin V at a faster rate than the wild-type strain. In fed-batch fermentations in 14-liter fermentors, where glucose was fed continuously and pH controlled, both GRI mutants produced more than 10% higher penicillin V titers than the wild-type strain. These results suggest that isolation of GRI mutants is an effective way to select for higher producing strains and that the synthesis of penicillin synthesizing enzymes in GRI mutants may be less repressed by glucose than in wild-type strains.  相似文献
