Summary The inheritance of resistance to mungbean yellow mosaic virus (MYMV) was studied in blackgram (Vigna mungo (L.) Hepper). The highly resistant donors Pant U-84 and UPU-2 and a highly susceptible line, UL-2, their F1's, F2's and backcrosses were grown with spreader located every 5 to 6 rows. The resistance was found to be digenic and recessive in all the crosses and free from cytoplasmic effect.  相似文献

Thinopyrum intermedium is a promising source of resistance to wheat streak mosaic virus (WSMV), a devastating disease of wheat. Three wheat germplasm lines possessing resistance to WSMV, derived from Triticum aestivum×Th. intermedium crosses, are analyzed by C-banding and genomic in situ hybridization (GISH) to determine the amount and location of alien chromatin in the transfer lines. Line CI15092 was confirmed as a disomic substitution line in which wheat chromosome 4A was replaced by Th. intermedium chromosome 4Ai?2. The other two lines, CI17766 and A29-13-3, carry an identical Robertsonian translocation chromosome in which the complete short arm of chromosome 4Ai?2 was transferred to the long arm of wheat chromosome 4A. Fluorescence in situ hybridization (FISH) using ABD genomic DNA from wheat as a probe and S genomic DNA from Pseudoroegneria stipifolia as the blocker, and vice versa, revealed that the entire short arm of the translocation was derived from the short arm of chromosome 4Ai?2 and the breakpoint was located at the centromere. Chromosomal arm ratios (L/S) of 2.12 in CI17766 and 2.15 in A29-13-3 showed that the translocated chromosome is submetacentric. The enzyme nitrite reductase (EC 1.6.6.4) is generally assayed in terms of disappearance of nitrite from the assay medium. We describe a technique which allowed estimation of the enzyme level in leaf tissues of Vigna mungo (L). Hepper in terms of the release of the product (NH3) of the enzyme reaction. The technique is offered as an alternative, possibly more convenient method for assay of nitrite reductase in plant tissue in vivo.  相似文献

The Arabidopsis-turnip crinkle virus (TCV) system is one of the few tractable plant-virus systems that allow simultaneous characterization of host components required for basal- and/or resistance (R) protein-mediated defenses. Another unique feature is that hypersensitive response (HR) and resistance can be studied as two distinct phenotypes in this pathosytem. The R protein HRT confers HR to TCV but requires a recessive locus rrt to confer resistance. The pathways leading to HR and resistance are mutually exclusive. HRT interacts with EDS1, which potentiates HR to TCV and is also required for resistance signaling. HRT-mediated signaling is also dependent on the EDS1-interacting proteins PAD4 and SAG101, which form binary and ternary complexes with EDS1. HRT-mediated resistance is also dependent on light and more specifically on the blue-light photoreceptors, cryptochromes (CRY) and phototropins (PHOT). Of these, CRY2 and PHOT2 are required for the stability of HRT. HRT is degraded in a proteasome-dependent manner, which correlates with its interaction with the E3 ubiquitin ligase, COP1. Together, these results suggest that components of light signaling modulate plant defense against TCV by regulating the stability of, and signaling mediated by, the R protein HRT.  相似文献

Four hybrids (4 F1s) were chosen out of crosses in the urdbean [Vigna mungo (L.) Hepper, 2n = 22] having contrasting morphological characters. Zymograms for isozyme peroxidase were drawn from the patterns obtained from parents and their respective F1 hybrids on the basis of relative similarities to parental bands. The selfed or crossed nature of hybrid pods was determined from the zymograms and their analysis. The number of bands and their intensities gave an idea about the extent of crossing in F1 populations. Genetic identity (I) values were indicative of their selfed nature. Dendrograms were constructed on the basis of genetic identity values to display the relative similarities between the populations. Analysis was based on individual pods to confirm their hybrid or selfed nature. Possible use of this technique for identification of F1 pods and elimination of selfed pods might be implemented to shorten the breeding operations during crossing.  相似文献

One of the most severe diseases of cultivated tomato worldwide is caused by tomato yellow leaf curl virus (TYLCV), a geminivirus transmitted by the whitefly Bemisia tabaci. Here we describe the application of antisense RNAs to interfere with the disease caused by TYLCV. The target of the antisense RNA is the rare messenger RNA of the Rep protein, encoded by the C1 gene. Transgenic Nicotiana benthamiana plants expressing C1 antisense RNA were obtained and shown to resist infection by TYLCV. Some of the resistant lines are symptomless, and the replication of challenge TYLCV almost completely suppressed. The transgenes mediating resistance were shown to be effective through at least two generations of progeny.  相似文献

Resistance conferred by the Tsw locus from Capsicum chinense against Tomato spotted wilt virus (TSWV) has been widely used in breeding programmes. Nevertheless, this resistance depends on inoculation conditions, and isolates able to overcome it have already been detected. In this work 29 accessions of several Capsicum species have been mechanically inoculated with TSWV to identify new sources of resistance. Five accessions showed variable percentages of resistant plants, two of which did not show local lesions on inoculated leaves, suggesting that the response was not mediated through hypersensitivity. Two of these accessions also had a remarkable reduced viral accumulation compared to susceptible control. ECU‐973., a C. chinense accession, showed the best performance against TSWV, with 100% resistant plants. This response was confirmed after mechanical inoculation with three different TSWV isolates. The resistance was maintained when the accession was inoculated with TSWV using a high pressure of viruliferous thrips. These results open new possibilities in the development of a durable resistance to TSWV in pepper.  相似文献

Inoculation of turnip crinkle virus (TCV) into a (TCV)-resistant line of Arabidopsis thaliana , Di-17, results in the development of a hypersensitive response (HR) on the inoculated leaves. In contrast, an HR does not occur when leaves of the TCV-susceptible Di-3 line or the susceptible ecotypes Columbia (Col-0), or Landsberg erecta ( Ler ) are inoculated. Genetic analysis of progeny from crosses between Di-17 and either Di-3, Col-0 or Ler demonstrates that the development of an HR is regulated by a single dominant nuclear locus, herein designated HRT . Using progeny from a Di-17 X Col-0 cross, HRT was mapped to chromosome 5, where it is tightly linked to the DFR locus. We also demonstrate that a variety of resistance-associated phenomena, including the TCV-induced accumulation of salicylic acid, camalexin and autofluorescent cell-wall material, correlate with the HR, suggesting the possibility that HRT is required for their activation.  相似文献

The role of salicylic acid (SA) in inducing resistance to MYMIV infection in Vigna mungo has been elucidated by proteomics. Twenty-nine proteins identified by MALDI-TOF/TOF, predicted to be involved in stress responses, metabolism, photosynthesis, transport and signal transduction, showed increased abundance upon SA treatment. Susceptible plants showed characteristic yellow mosaic symptoms upon MYMIV infection. A concentration dependent decrease in physiological symptoms associated with MYMIV was observed upon exogenous SA treatment prior to viral inoculation; and no visible symptom was observed at 100 μM SA. SA treatment stimulated SOD and GPX activity and inhibited CAT activity thus preventing ROS mediated damage. Significant increase in chlorophyll, protein, carbohydrate, phenolic content and H(2)O(2) were observed. Involvement of calmodulin for transmission of defense signal by SA is suggested. A metabolic reprogramming leading to enhanced synthesis of proteins involved in primary and secondary metabolisms is necessary for SA mediated resistance to MYMIV. Identification of proteins showing increased abundance, involved in photosynthetic process is a significant finding which restores virus-induced degradation of the photosynthetic apparatus and provides enhanced metabolites required for repartition of resources towards defense.  相似文献

Black gram (Vigna mungo) seeds are shown to contain a lectin with certain unusual features. The lectin agglutinates only trypsinized red cells, and its sugar specificity is complex as none of the common sugars, oligosaccharides or complex polysaccharides exhibit any affinity for the lectin. The purified lectin has a molecular weight of 58 kDa and is a monomer. Unlike other plant lectins, antibodies to the P. mungo lectin do not exhibit any immunological cross reactivity. The clot forming ability of the lectin is unusual in that the clot once formed is rapidly disaggregated indicated that it induces, as yet undefined, certain membrane alterations.  相似文献

针对近年弯孢菌叶斑病日益严重的发生趋势,对1698份玉米种质(自交系、群体、杂交种以及特殊材料)进行了抗弯孢菌叶斑病鉴定.结果表明,中国玉米种质抗性较引进种质抗性好;不同省份所供种质抗性存在差异,北京、四川、广西种质总体抗性较好;在新选育的自交系中,鉴定出12份高抗材料;在当前培育的杂交种中,有22份高抗或抗弯孢菌叶斑病;玉米对弯孢菌叶斑病抗性在相同核基因、不同细胞质种质间无差异;玉米抗大斑病基因对抗弯孢菌叶斑病无效.  相似文献

Eighty three wild Arachis germplasm accessions, belonging to 24 species of five sections and one natural hybrid derivative of a cross between the cultivated and a wild Arachis species, were evaluated along with a susceptible groundnut cultivar for resistance to Peanut bud necrosis virus (PBNV) in a replicated field trial at ICRISAT, Patancheru, India. Thirty days after sowing, the percentage of infected plants were recorded for all the accessions and subsequently young leaflets from all these accessions were tested for the presence of the virus by enzyme linked immunosorbent assay (ELISA). One accession each of A. benensis and A. cardenasii, and two accessions of A. villosa, in the section Arachis, two accessions of A. appressipila in the section Procumbentes, and one accession of A. triseminata under section Triseminatae were not infected by PBNV. These seven field‐resistant accessions were tested under glasshouse conditions for virus resistance by mechanical sap inoculations. One accession of A. cardenasii and two accessions of A. villosa did not show systemic infection. Similarly, in another glasshouse test, where 13 A. cardenasii accessions of section Arachis were evaluated, two accessions did not show systemic infection. In all these resistant accessions, the inoculated leaves showed infection, but the systemic leaves did not show the presence of virus in spite of repeated mechanical sap inoculations. So, the resistance in these accessions appears to be due to a block in systemic movement of the virus. To our knowledge this is the first report on the identification of resistance to PBNV in wild Arachis species. Since both A. cardenasii and A. villosa are the progenitors of cultivated groundnut and can be hybridised with the latter, the resistant accessions are being utilised in conventional breeding programmes to transfer PBNV resistance to widely adapted groundnut cultivars.  相似文献

Transformed Vigna mungo (blackgram) calli were obtained by cocultivating segments of primary leaves with Agrobacterium tumefaciens vir helper strains harbouring the binary vector pGA472 having kanamycin resistance gene as plant transformation marker. Transformed calli were selected on Murashige and Skoog medium supplemented with 50 mg/l kanamycin and 500 mg/l carbenicillin. Transformed calli were found to be resistant to kanamycin up to 900 mg/l concentration. Expression of kanamycin resistance gene in transformed calli was demonstrated by neomycin phosphotransferase assay. Twenty-five isolates of urd bean ( Vigna mungo ) Rhizobium were tested for the presence of an H2-uptake system using triphenyl tetrazolium chloride reduction as the screening procedure. The isolates which reduced the dye rapidly at early stages of growth were found to recycle H2 both in culture as well as in nodules. H2-uptake positive, H2-uptake negative strains and H2-uptake negative mutants were compared on the basis of their effect on dry matter accumulation and N2-fixation. Greenhouse experiments have shown the beneficial effect of the H2-uptake positive system in nodules on N2-fixation. Plants inoculated with H2-uptake positive strains produced higher N content and dry matter over the plants inoculated with H2-uptake negative strains.  相似文献

The present experiment was undertaken to examine whether shading affects the critical boron (B) concentration for leaf blade elongation in black gram. Six days after germination (D6), black gram seedlings were transferred to 8 pairs of pots containing basal nutrient solution: one pot of each pair contained 1 M H3BO3 and the other 10 M H3BO3. On D10, one day after the emergence of the first trifoliolate leaf blade (TF1) and one day before the emergence of TF2, four pairs of pots were shaded, decreasing the light intensity they received in the glasshouse to about 35% of full sunlight compared with 70% received by the unshaded plants. The response to B supply of dry matter (DM) and elongation rate (LBER) of TF2 were less pronounced in shaded than in unshaded plants. Critical B concentrations for LBER in TF2 of black gram were 10 mg B kg-1DM in shaded and 15 mg B unshaded plants, suggesting that shading may have decreased the B requirement for LBER. Thus the present results suggest that light may need to be considered when setting critical values for the diagnosis of B deficiency in black gram.  相似文献

Summary The inheritance of the dwarf plant type was studied in blackgram (V. mungo (L.) Hepper). Type 9 has erect plant type with normal internode length. The mutant line, EMSD has reduced internode length. The F1, F2 and F3 generations of a cross between Type 9 and EMSD and its reciprocal were studied. Lebanon, an East Mediterranean country, does not have a comprehensive reference national olive collection. A report published 30 years ago indicated the presence of four Lebanese varieties, but the confusion regarding these varieties was discussed then and is still prevalent today hindering prospects for conserving and exploring the national germplasm. This study assessed the existing in situ diversity of old Lebanese olive groves using simple sequence repeat (SSR) and amplified fragment length polymorphisms (AFLP). Trees (140) were collected from 14 groves located in the four traditional olive areas. Analysis was based on 22 polymorphic alleles produced from six SSR primers, and on 135 polymorphic AFLP fragments. SSR and AFLP did not yield consistent results in terms of level of polymorphism, with SSR detecting higher variation than AFLP. On the other hand, both clustered trees based on geographic provenance. AFLP coefficient of similarity between trees ranged between 0.70 and 0.99 indicating the possible presence of varieties since some values fall within reported intervarietal ranges of 0.65 to 0.84. SSR unveiled the presence of nine different genotypes: four corresponding each to a provenance and five consisting of single trees characterized by the presence of less frequent alleles with an average of 0.33. Results also revealed a high incidence of clones greater than 90% in three of the four provenances. The findings of this study point for the need to plan for a sampling strategy that takes into consideration geographic provenances.  相似文献
