Striga hermonthica and S. asiatica are root parasitic weeds that infect the major cereal crops of sub-Saharan Africa causing severe losses in yield. The interspecific upland NEw RICe for Africa (NERICA) cultivars are popular amongst subsistence farmers, but little is known about their post-attachment resistance against Striga. Here, we evaluate the post-attachment resistance levels of the NERICA cultivars and their parents against ecotypes of S. hermonthica and S.asiatica, characterize the phenotype of the resistance mechanisms and determine the effect of Striga on host biomass. Some NERICA cultivars showed good broad-spectrum resistance against several Striga ecotypes, whereas others showed intermediate resistance or were very susceptible. The phenotype of a resistant interaction was often characterized by an inability of the parasite to penetrate the endodermis. Moreover, some parasites formed only a few connections to the host xylem, grew slowly and remained small. The most resistant NERICA cultivars were least damaged by Striga, although even a small number of parasites caused a reduction in above-ground host biomass. The elucidation of the molecular genetic basis of the resistance mechanisms and tolerance would allow the development of cultivars with multiple, durable resistance for use in farmers' fields.  相似文献

Tillering in cereals is a complex process in the regulation of which also signals from the roots in the form of strigolactones play an important role. The strigolactones are signalling molecules that are secreted into the rhizosphere where they act as germination stimulants for root parasitic plants and hyphal branching factors for arbuscular mycorrhizal fungi. On the other hand, they are also transported from the roots to the shoot where they inhibit tillering or branching. In the present study, the genetic variation in strigolactone production and tillering phenotype was studied in twenty rice varieties collected from all over the world and correlated with S. hermonthica infection. Rice cultivars like IAC 165, IAC 1246, Gangweondo and Kinko produced high amounts of the strigolactones orobanchol, 2′-epi-5-deoxystrigol and three methoxy-5-deoxystrigol isomers and displayed low amounts of tillers. These varieties induced high S. hermonthica germination, attachment, emergence as well as dry biomass. In contrast, rice cultivars such as Super Basmati, TN 1, Anakila and Agee displayed high tillering in combination with low production of the aforementioned strigolactones. These varieties induced only low S. hermonthica germination, attachment, emergence and dry biomass. Statistical analysis across all the varieties confirmed a positive correlation between strigolactone production and S. hermonthica infection and a negative relationship with tillering. These results show that genetic variation in tillering capacity is the result of genetic variation in strigolactone production and hence could be a helpful tool in selecting rice cultivars that are less susceptible to S. hermonthica infection.  相似文献

The surface features of the seed of the parasitic floweringplant Striga hermonthica were examined with the scanning electronmicroscope. The details of ornamentation were constant on seedsfrom one plant but varied within and between populations andare probably due to out-breeding. 从非洲水稻研究中心引进了78份非洲新稻品种资源,2009年在武汉大田条件下种植,对生育期、水稻白叶枯抗性和稻米品质进行了初步评价。观察表明,生育期可以分为4个类型:短生育期品种13份,中生育期品种37份,长生育期11份,生育期特长在武汉不能安全抽穗的品种17份。白叶枯抗性鉴定表明,57.7%的品种高抗或抗长江中下游籼稻区的优势白叶枯病菌株ZHE173,76.9%的品种高抗或抗来自云南稻区的菌株YN18,对中国新发现的强致病力菌株FuJ能达到抗级水平的仅有3个品种,而达到中抗水平的有5个品种,凡是对FuJ达到中抗以上的品种,均高抗或抗ZHE173和YN18。对50个品种的米质分析表明,非洲新稻品种的稻米加工品质和粒型品质特别优良。非洲新稻品种资源可以用于改良国内新品种的白叶枯病抗性和稻米加工品质。  相似文献

Striga is a parasitic weed attacking mainly maize, sorghum, millet and cowpea. Studying the interaction between rice and Striga is valuable since rice is a model monocot. In this paper, the susceptibility of different rice cultivars to S. hermonthica was tested and quantitative trait loci (QTL) for Striga tolerance mapped on the Bala x Azucena F(6) population. Seven rice cultivars were grown with and without S. hermonthica for 14 wk. For the mapping experiment, 115 recombinant inbred lines (RILs), along with Azucena and Bala, were grown with and without Striga for 11 wk. Rice cultivars tested had different susceptibilities to Striga, ranging from highly susceptible to completely resistant. Azucena and Bala differed in the speed of Striga emergence and the impact on host growth. A genomic region between positions 139 and 166 cM on chromosome 1 was identified containing strong QTL (LOD = 4.9-15.7) for all traits measured. This indicates that genes for Striga tolerance exist in rice germplasm and the mapped QTL can be further studied to promote understanding of the nature of resistance/tolerance and breeding for Striga-resistant crop plants.  相似文献

Dehydration-responsive-element-binding protein 1 genes have important roles in response to stress. To improve the drought tolerance of an upland rice cultivar NERICA1, we introduced Arabidopsis AtDREB1C or rice OsDREB1B driven by a stress-inducible rice lip9 promoter. Plants of some transgenic lines survived better than non-transgenic plants under severe drought. AtDREB1C transgenic plants had higher dry weights than non-transgenic plants when grown under moderate drought until the late vegetative growth stage. On the other hand, OsDREB1B transgenic plants had lower dry weights than non-transgenic plants under the same condition. Similar results were obtained under osmotic stress. The AtDREB1C transgenic plants headed earlier, had a larger sink capacity, and had more filled grains than non-transgenic plants. These results suggest that AtDREB1C expressed in NERICA1 improves not only survival under severe drought, but also growth and yield under moderate drought.  相似文献

A fungus isolated from wilted Striga hermonthica plants was identified as Fusarium oxysporum. A comparison of this isolate (PSM-197) with four other Fusarium spp. for control of S. hermonthica using conidial foliar sprays, showed that it was the most pathogenic and virulent. The isolate, grown on sorghum grain and incorporated into soil, completely inhibited the emergence of S. hermonthica compared with other substrates.  相似文献

Autotrophic photosynthesis in developing plants of Striga hermonthica, a C3 plant, parasitizing Sorghum bicolor, a C4 plant, has been quantified by measuring the 13C/12C isotope ratio (δ13C). This parameter has been determined in cellulose and chlorophyll extracted from plants 8 weeks after emergence. It is shown that these components are both proportionately high in autotrophic carbon in the leaf material, thus confirming the previous hypothesis that St. hermonthica photo-assimilate is directed primarily towards the synthesis of new photosynthetic capacity.  相似文献

Data were obtained from the research done in the Guinea Savanna (Zaria 11°11′N; 070 38′E) and Sudan Savanna (Maiduguri ?11°51′N; 13°15′E) regions of Nigeria, respectively on different cultural methods of Striga control and management. In the Guinea Savanna, trials on the effects of nitrogen on the response of resistant and susceptible upland rice varieties to Striga hermonthica infestation and the effect of resistant and susceptible varieties of maize and crop rotation on Striga infestation was carried out, while the effect of inter-cropping trap crop (Bambaranut) with resistant sorghum varieties on S. hermonthica was studied in the Sudan Savanna Zone of Nigeria. In the Guinea Savanna, it was observed that a combination of upland rice variety, Faro 40 and an application of 90 kg N/ha in the wet season and WAB 56-50 upland rice variety and 120 kg N/ha in the dry season, respectively reduced Striga infestation and produced maximum grain yield. Also, the growing of resistant variety of maize (Across 97ITZ comp. I-W) after 1 or 2 years' rotation with cowpea or soybean was observed not only to be effective in Striga control, but resulted in higher grain yield of maize. In the Sudan Savanna, the use of resistant varieties of sorghum, 1CSV1002 and 1CSV1007 intercropped with bambaranut significantly reduced Striga infestation, but the grain yield of the resistant varieties was low. From these studies, Faro 40 with 90 kg N/ha application rate and WAB56–50 with 120 kg N/ha were suitable for the management of Striga and for higher grain yield of upland rice in both wet and dry seasons, respectively, while Across 97ITZ comp. I-W, resistant maize variety and 1 or 2 years rotation with cowpea or soybean were also the best for the management of S. hermonthica and for higher maize yield in the Guinea Savanna zone. Further research needs to be carried out in the Sudan Savanna to select a high yielding resistant variety of sorghum which when intercropped with bambaranut will not only control Striga infestation but will also give high grain yield.  相似文献

Jasmonates and related compounds were found to elicit the seed germination of the important root parasites, clover broomrape (Orobanche minor Smith) and witchweed [Striga hermonthica (Del.) Benth]. The stimulation of seed germination by the esters was more effective than by the corresponding free acids, and methyl jasmonate (MJA) was the most active stimulant among the compounds tested.  相似文献

Striga hermonthica is a root hemiparasite that attacks onlyGramineae, includingSorghum and millet for which it is a principal cause of lowered yield. Enzyme electrophoresis was used to investigate genetic diversity inStriga hermonthica and to determine the level of differentiation between host-specialized populations. Nine genetic loci coding eight enzymes were interpreted and data obtained from three populations: oneSorghum-adapted population from Sudan and two populations from Burkina Faso, oneSorghum-adapted and the other millet-adapted. Levels of polymorphism were similarly high in all three populations (P=0.625, A=2.6–2.8, H=0.293–0.401). Genotypic frequencies at most loci conformed to Hardy-Weinberg expectations in each population, consistent with outcrossing as predicted from previous studies of floral biology. Occasional heterozygote deficiencies were probably the result of Wahlund effect. The mean value of FST over the three populations was 0.068, indicating a slight to moderate level of genetic differentiation among the populations. The two Burkina Faso populations were more closely related (S=0.940, D=0.006) than either was to the Sudan population, suggesting that geographic separation is more important than host specialization in contributing to population differentiation. TheSorghum-adapted population was slightly closer to the Burkina FasoSorghum-adapted population (S=0.873, D=0.047) than to Burkina Faso millet-adapted population (S=0.851, D=0.074). The absence of substantial genetic divergence between host-specific populations ofStriga could result either from recent evolution of host-specialized strains or from strong selection for physiological specialization in the face of substantial gene flow between the populations.  相似文献

Summary The contribution of introgressed regions derived from wild species to the genetic variation within the species of Lycopersicon esculentum was investigated by comparing the RFLP patterns of 2 introgression-free, obsolete cultivars (Moneymaker and Premier) and a modern cultivar (Sonatine) that carries at least 5 introgressed resistance genes. In this analysis 195 mapped nuclear markers were used in combination with 6 restriction enzymes. Among the 1170 probe-enzyme combinations tested, only 3 showed a polymorphism between the 2 introgression-free cultivars. On the other hand 24 probe-enzyme combinations were found to exhibit polymorphisms between Moneymaker and Sonatine. These represented ten polymorphic loci distributed among 5 linkage groups on chromosomes 1, 3, 4, 6, and 9.On the assumption that most of the polymorphic loci corresponded to introgressed chromosome segments of wild species carrying resistance genes, linkages between these loci and the component resistance genes were examined by RFLP analysis of pairs of near-isogenic lines differing only for one particular resistance gene, and a variety of commercial cultivars having different resistance gene compositions. Two of the polymorphic linkage groups could thus be ascribed to resistance genes whose map positions were already known: Cf2 on chromosome 6 and Tm2a on chromosome 9, whereas another marker, TG301 on chromosome 1, could be assigned to the Cladosporium fulvum resistance gene Cf9 with a hitherto disputable map position. By linkage analysis of a segregating F2 population the genetic distance between the Cf9 gene and the marker TG301 was estimated at 5.5 ± 2.3 cM.  相似文献

The threshold of tolerance for nitrate of the parasitic weed Striga hermonthica (Del.) Benth and the host plant Sorghum bicolor L. was determined by estimating the impact of increasing nitrate loads on plant growth and various parameters of C and N assimilation. Nitrate supply improved chlorophyll (Chl) content and photosystem II (PSII) photochemistry of infected S. bicolor that, in comparison to S. hermonthica, displayed a low imbalance between C and N assimilation when nitrate was supplied up to 1500 mg N per plant. Indeed, nitrate supplies increased strongly the leaf N:C ratio of the parasite. The higher nitrate load induced strong accumulation of nitrate, nitrite and ammonium, and consequently the death of S. hermonthica. Nevertheless, lower nitrate loads (up to 500 mg N per S. bicolor in this study) promoted leaf expansion, PSII photochemistry and N metabolism of S. hermonthica mature (M) plants, as attested by the significant rise in soluble protein and free amino-acid contents. Following these N supplies, the nitrate tolerance of S. hermonthica was correlated with an increase in PSII activity and a high incorporation of N excess into asparagine. This confirmed the central role of asparagine in the N metabolism of S. hermonthica, although this detoxification pathway was insufficient to limit ammonium accumulation under higher nitrate loads.  相似文献

The root‐lesion nematode Pratylenchus thornei is a major pathogen of wheat and other field crops, particularly in the northern grain region of sub‐tropical eastern Australia. Research was conducted into the temperature requirements of P. thornei for reproduction on wheat to increase the reliability of selection in resistance tests for wheat breeding. Final population densities (Pf) of P. thornei were determined on four wheat cultivars (Gatcher, GS50a, Potam and Suneca) at fortnightly intervals from 8 to 18 weeks at a range of six soil temperatures (15°C, 20°C, 22.5°C, 25°C, 27.5°C and 30°C) in a glasshouse experiment. Pratylenchus thornei had the highest Pf in the temperature range of 20–25°C on all wheat cultivars at all growth times after sowing, with no nematode reproduction measured at 30°C and very little at 15°C. The wheat cv. GS50a consistently produced lower Pf than cvs Gatcher, Potam and Suneca in the optimum temperature range of 20–25°C. In carrot disc cultures, P. thornei had an optimum temperature of 25°C with little reproduction at 17.5°C and none detectable at 30°C. A standard soil temperature of 23°C was chosen to maximise differences in nematode reproduction between resistant and susceptible wheat genotypes for selection in wheat breeding, and to improve reproducibility among successive experiments. The relationships derived from these experiments will be valuable for simulation of P. thornei reproduction in crop growth models. They also indicate that early sowing of wheat into cool soil (≤15°C) in farmers' fields of the northern grain region should favour wheat growth over nematode reproduction and increase grain yield.  相似文献

Potential resistance to the twolined spittlebug, Prosapia bicincta (Say), was evaluated among 56 turfgrass genotypes. Greenhouse, laboratory, and field bioassays identified differences in spittlebug survival and development, host preference and damage levels, and turfgrass tolerance to and ability to recover from pest induced injury. All centipede grasses demonstrated high levels of susceptibility, followed by bermudagrasses, seashore paspalums, and zoysiagrasses. Average nymphal survival to the adult stage ranged from 1.5 to 78.1%. Development required 38.1-62.0 d under greenhouse conditions, depending on plant taxa. Among seashore paspalums, nymphal survival to the adult stage was lowest and duration of development was longest on HI-1, 'Sea Isle 2000', 561-79, and 'Mauna Kea'. Reduced spittlebug survival and increased developmental times were also observed on the bermudagrasses BERPC 91-15 and 'Tifway'. Although zoysiagrasses supported spittlebug development and survival to the adult stage, developmental times were extended on the zoysiagrass cultivars 'Emerald' and 'El Toro'. Spittlebug preference varied with generation evaluated. First-generation spittlebugs inflicted the greatest damage on TC201 (centipede grass), 'Primavera' (bermudagrass), and 'Emerald' (zoysiagrass) in choice tests. In the fall, second-generation spittlebugs damaged TC201 (centipedegrass) and 'Sea Isle 1' (paspalum) most severely, whereas 561-79 (paspalum) and 'Emerald'(zoysiagrass) were less severely affected. Among taxa included in field trials, HI-1, 'Mauna Kea', 'Sea Isle 2000',and AP-14 paspalums, 'Tifway' bermudagrass, and 'Emerald' zoysiagrass were most tolerant (demonstrated the best regrowth potential following twolined spittlebug feeding).  相似文献

Olea europaea is one of the oldest species of domesticated trees. We used microsatellite markers for fingerprinting and for evaluation of genetic similarity and structure of 26 Greek olive cultivars, which cover most of the olive cultivation regions of Greece, including previously undescribed denominations from northern Greece. Eighty-one alleles were revealed with six SSR loci that were selected as most informative of 10 SSR primers that were initially investigated. The number of alleles per locus varied from 7 to 20 (mean, 13.5). Heterozygosity ranged from 0.240 at locus DCA-3 to 0.826 at locus UDO99-9, with a mean value of 0.600. Analysis of 104 trees representing 26 denominations (four trees per denomination) revealed 26 distinct SSR profiles, indicating 26 olive cultivars; no intracultivar variability was observed. Genetic and geographic distances were not significantly correlated, based on the Mantel test. These SSR loci allowed unequivocal identification of all the cultivars and will be useful for future breeding and olive germplasm management efforts.  相似文献
