不同地区库蚊复组群体的同工酶遗传多样性研究   总被引:2,自引:0,他引:2

采用水平切片淀粉凝胶电泳的方法,对分布于我国5省的8个库蚊复组(Culex pipiens complex)野生群体的遗传多样性进行研究,分析了4个酶系统7个基因座(ME、MDH-1、MDH-2、MDH-3、GPD、EST-2、EST-3)的酶谱资料。结果显示:(1)群体内存在不同程度的遗传变异(He为0.098~0.41);(2)较低的基因流水平(Nm=0.64)使遗传漂变起主要作用,造成群体之间的遗传分化(Gst=0.303),而总群体的遗传多样性相对富集于群体之内(Hs/Dst=2)。(3)库蚊群体的遗传结构属于距离隔离模式。(4)群体间的遗传一致性(或遗传距离)反映出群体间的遗传分化程度,也表明与地理位置存在对应关系。Abstract: Eight field populations of Culex pipiens complex collected from five provinces (Guangdong, Henan, Shandong, Beijing and Yunnan) in 2001 were used to study genetic diversity by starch gel electrophoresis. Data from seven loci (ME、MDH-1、MDH-2、MDH-3、GPD、EST-2、EST-3) of four isozymes were analyzed by software Biosys2.0 and FSTAT(Version 2.9.3). The results were as follows: (1) The values of He (from 0.098 to 0.41) indicated genetic variabilities of different degree in populations.(2)The low level of gene flow (Nm=0.64) could not prevent genetic drift to cause the gene differentiation between populations. The genetic diversity between populations attributed to the genetic diversity of total populations is small (Gst =0.303), and the great part is accumulated within populations (Hs/Dst=2). (3) The genetic structure of Culex pipiens complex population was the isolation-by-distance model. (4) The genetic identity (or genetic distance) revealed the scale of genetic differentiation between populations which related to the collection sites.  相似文献

对分布于中国西北地区的11种黄耆属植物进行了细胞学研究,其中9种作了核型分析。结果表明,杯萼黄耆(Astragalus cupulicalycinus)为六倍体(2n=48),酒泉黄耆(A.jiuquanensis)具有2种细胞型:2n=32的四倍体和2n=48的六倍体,其余种类均为二倍体(2n=16)。9个种的核型公式分别为:木黄耆(A.arbuscula),2n=16=14 2sm,“1A”核型;胀萼黄耆(A.ellipsoideus),2n=16=10m 6sm,“2A”核型;粗毛黄耆(A.scabrisetus),2n=16=14m 2sm,“1A”核型;秦岭黄耆(A.henryi),2n=16=8m 8sm(2SAT),“2A”核型;哈巴河黄耆(A.habaheensis),2n=16=10m 6sm(2SAT),“2A”核型;变异黄耆(A.variabilis),2n=16=4m 10sm(2SAT) 2t,“2A”核型;喜沙黄耆(A.ammodytes),2n=16=6m 10sm(2SAT),“2A”核型;密花黄耆(A.densiflorus),2n=16=6m(2SAT) 10sm,“2A”核型;茧荚黄耆(A.lehmannianus),2n=16=14m(2SAT) 2st,“2A”核型。所有研究种中,仅变异黄耆的染色体数目有过报道,其余种的染色体数目及核型均为首次报道。  相似文献

测定了27株中国1968~2000年的人H3N2亚型流行性感冒(流感)病毒分离株HA1基因的核苷酸全序列,其中包括7株流行代表株:A3/Beijing/1/68、A3/Guangdong/243/72、A3/Beijing/39/75、A3/Guangdong/38/77、A3/Beijing/266/85、A3/Beijing/30/95、A3/Shanghai/1/98.阐明了这些流感病毒分离株的基因结构、变异本质、进化发展过程以及这些毒株与其它分离株之间的遗传进化亲缘关系,为应用生物信息学研究流感病毒基因组变异规律等打下了基础.初步的进化分析表明,历年来的人流感病毒H3N2亚型的起源和进化分为两个分支谱系,并在1984/1985年进行了交替转换;且有一部分人的H3N2亚型分离株其HA1核苷酸序列与猪分离株的进化亲缘关系最为接近.  相似文献

中外11个猪种A-FABP基因微卫星遗传变异的研究   总被引:8,自引:2,他引:6  
利用微卫星技术对五指山猪、沂蒙黑猪、汉江黑猪、莱芜猪、北京黑猪、民猪、成华猪、内江猪、二花脸猪、巴马香猪和大白猪11个猪种共420头猪的脂肪细胞脂肪酸结合蛋白基因内含子的微卫星序列的遗传变异进行了研究。研究表明:(1)中国猪种在A-FABP基因微卫星基因座上有极高的的多态性,五指山猪的PIC最高(PIC=0.7904)并检测到11个等位基因,但北京黑猪仅检测到两个等位基因。(2)本实验中,只有北京黑猪、民猪、巴马香猪和大白猪A-FABP基因微卫星在群体中的分布达到平衡状态。(3)群体间的基因分化系数分析表明,中国猪种在该微卫星基因座上分化程度平均约为40.83%。Abstract:The genetic variations of microsatellites in intron 2 of the porcine adipocyte fatty acid-binding protein (A-FABP) genes were investigated in 420 pigs including Wuzhishan pig,Yimeng black pig,Hanjiang black pig,Laiwu pig,Beijing black pig,Min pig,Chenghua pig,Neijiang pig,Erhualian pig,Bama xiang pig and Large White pig.The results suggested as follows:(i)PIC of the Wuzhishan pig breed is the highest(0.7904) and 11 alleles were detected. Compared with Large White pig,Chinese pig breeds showed a great polymorphism of A-FABP microsatellites except Beijing black pig in which only 2 alleles were detected.(ii)Only Min pig, Bama xiang pig, Beijing black pig and Large White pig were in Hardy-Weinberg equilibrium. (iii)The analysis of genetic differentiation showed that the average value of the A-FABP gene differentiation of 10 Chinese pigs is about 40.83%.  相似文献

美洲黑杨抗黑斑病基因的RAPD标记筛选和连锁分析   总被引:7,自引:0,他引:7  
以美洲黑杨(Populus deltoides Bartr.cv.“Lux”(I-69/55))为母本,欧美杨(P.euramericana cv.I-45)为父本得到的F1为材料,其中I-69对黑斑病表现为高度抗病,I-45为高度感病。利用分离群体混合分析(bulked segregrants analysis,BSA)技术建立两个DNA池(感病池和抗病池),筛选出了与抗黑斑病性状基因相连锁的RAPD标记:OPAI17―1550、OPAI13―900。利用选择性基因型分析法进行标记―抗黑斑病基因连锁分析,两标记与抗黑斑病基因遗传距离分别为29.9cM和37.4cM。 Identification of Markers Linked to Resistance Locus of Marssonina Leaf Spot in Poplars by Bulked Segregant Analysis(BSA) ZHANG Bo1,HUANG Min-ren1,ZHUGE Qiang1,HAN Zheng-min1,YIN Tong-ming1,PAN Hui-xin1,ZHU Li-huang2,WU Rong-ling3,WANG ming-xiu1 1.The Key Laboratory of Forest Tree Genetic Engineering,Nanjing Forestry University,Nanjing,210037,China; 2.The Institute of Genetics and Developmental Biology,Chinese Academy of Science,Beijing 100101,China; 3.Department of Statistics,University of Florida,Gainesville,Florida 32611,USA Abstract:DNA markers linked to resistance locus of Marssonina leaf spot in poplars were found by bulked segregant analysis(BSA).The bulks consisted of individual with a extreme phenotype taken from a population of 91 F1 clones,which is a progeny of Populus deltoides Bartr.cv.“Lux”(I-69/55)(Resistance) and P.euramericana cv.I-45(Susceptible).Out of 114 RAPD primers,four markers showed polymorphisms between the resistance-bulk and the susceptible-bulk.By using selective genotype linkage analysis,OPAI17-1550 and OPAI13-900 were found linked to the resistance locus.The genetic distances between the two markers and the resistance locus were 29.9cM and 37.4cM,respectively. 一株茄病镰刀菌(Fusarium solani)固体发酵培养物经柱层析分离得到10个化合物。通过波谱分析,分别鉴定为对羟基苯甲酸甲酯(1)、水杨酸甲酯(2)、水杨酸戊酯(3)、香草乙酮(4)、草夹竹桃苷(5)、2-甲氧基-4-乙烯苯基-β-D-吡喃葡萄糖苷(6)、1-O-β-D-glucopyranosyl-(2S,3R,8E)-2-[(2R)-2-hydroxylpalmitoylamino]-8-octadecene-1,3-diol(7)、1-O-β-D-glucopyranosyl-(2S,3R,4E,8E)-2-[(2R)-2-hydroxyhexadecanoylaino]-8-octadecene -1,3-diol (8)、脑苷脂D(9)和脑苷脂C(10)。所有化合物均为首次从茄病镰刀菌中分离得到,其中化合物6首次作为天然产物报道。  相似文献
