“微生物”这一名词指非常小的生物,如古菌、细菌、原生生物、真菌和病毒,肠道“微生物组”表示的是肠道微生物集合体。它们实际上共享宿主的身体空间,但作为宿主健康和疾病的决定因素却几乎被忽视。作为信息的集合,微生物组包括微生物的基因组数据、结构元件、代谢物和环境条件。最近对肠道微生物组的研究表明,微生物群落在维持宿主稳态和调节宿主表型上发挥着重要作用。随着包括二代测序(next-generation sequencing, NGS)在内的新技术的出现以及微生物群落序列谱等深入测定技术出现,人们对肠道微生物组与宿主遗传背景之间的关系有了许多见解。本文通过肠道微生物组学的概述,基于全基因组关联分析技术建立肠道微生物组学与宿主遗传之间联系,并对宿主遗传学与肠道微生物组的关系及未来发展前景进行探讨。  相似文献

肠道微生物与肠道细胞线粒体功能之间的关系十分密切。一方面,肠道微生物可直接或通过短链脂肪酸、硫化氢和一氧化氮等代谢产物间接影响与线粒体相关的能量代谢过程,调节线粒体活性氧的产生,调控线粒体甚至整个机体的免疫反应。另一方面,肠道细胞线粒体功能紊乱和基因组的遗传变异也会影响肠道微生物的组成和功能。本文主要介绍了肠道微生物和线粒体之间的互作关系的最新研究进展,为靶向作用于肠道菌群和线粒体以调节肠道健康提供理论依据。  相似文献

无菌动物是指通过现代技术手段在其体内外的任何部位均检测不出细菌、真菌、放线菌、支原体、衣原体、螺旋体、立克次氏体、病毒、原生动物和寄生虫的动物。无菌动物因其不携带任何微生物,可转化为携带特定微生物的动物,同时因其免疫系统处于休眠状态,对微生物感染异常敏感,可建立多种悉生动物模型,用于特定微生物感染实验和致病机制研究。此外,无菌动物作为关键工具,是研究菌群与疾病关系的核心,在微生物与宿主健康、疾病和感染机制研究过程中,起着不可替代的作用。本文将对无菌动物及其在微生物与宿主互作机制研究中的应用进行简要综述。  相似文献

通过回顾过敏性疾病以及高脂饮食、肥胖等的大量研究报告,探究高脂饮食以及高脂饮食带来的肥胖是否会对食物过敏的发生产生影响以及可能的机制。研究发现,高脂饮食可能会增加食物过敏发生率,降低食物过敏阈值,增加食物过敏的风险,并且高脂饮食对肠道微生物的影响是其中的一个原因。高脂饮食改变肠道微生物结构,导致短链脂肪酸产生差异,而短链脂肪酸通过G蛋白偶联受体、组蛋白乙酰化等途径引起Treg细胞分化、IgA产生以及致耐受性的DC细胞产生,影响肠道屏障的健康,因此,高脂饮食对肠道微生物结构的改变带来的短链脂肪酸的产生差异影响了肠道屏障的健康,进而导致食物过敏的阈值降低,易感性增加。  相似文献

肠道上皮是肠上皮细胞及其分泌物有机构成的黏膜界面。随着技术的进步和对肠道菌群作用的逐渐重视,研究者对肠道上皮与肠道微生物相互作用的认识也不断深入。研究表明,肠道上皮调节并维持肠道微生物的定殖与分布,肠道微生物也影响肠道上皮的多种屏障功能,二者通过一系列细胞分子机制紧密联系,共同维持肠道稳态。此外,其过程中产生的宿主-肠道菌群共代谢物被发现可以反映宿主的生理病理状态,作为指标被应用于临床疾病诊断、治疗效果评估和预后推测。本文基于近年的研究,综述了肠道上皮与肠道微生物的相互作用及其细胞分子机制,为进一步研究和临床应用总结了理论基础,并探讨了未来可能的研究方向。  相似文献

目的探究孤独症谱系障碍(autism spectrum disorder,ASD)患儿与健康儿童的肠道菌群利用不同碳水化合物产生短链脂肪酸(short-chain fatty acids,SCFAs)的差异。方法前瞻性病例对照研究。选择2016年12月至2017年10月解放军总医院儿科门诊的15例ASD患儿为研究对象,并匹配15名健康儿童为对照组;收集研究对象粪便,稀释成10%悬浊液,然后分别接种到以乳果糖(LAU)、乳糖(LAT)、棉籽糖(RAF)、低聚半乳糖(GOS)、低聚异麦芽糖(IMO)和低聚甘露糖(MOS)为单碳源的肠道微生态小型模拟发酵系统中批量发酵24 h,检测SCFAs浓度、底物降解率和产气气压。结果 ASD组患儿粪便SCFAs浓度与对照组差异无统计学意义,在YCFA(无碳源对照培养基,蛋白发酵为主)培养基发酵后总SCFAs、乙酸和丙酸均低于对照组,差异有统计学意义(Z=-2.509、-2.509、-3.007,P=0.011、0.011、0.002);在添加LAT、RAF、GOS、IMO和MOS的培养基发酵后总SCFAs浓度显著高于对照组,差异有统计学意义(Z=2.385、2.344、2.675、2.344、2.302,P=0.016、0.019、0.007、0.019、0.021),而含LAU的培养基发酵后,两组研究对象总SCFAs浓度比较差异无统计学意义。结论 ASD患儿肠道菌群利用蛋白发酵产SCFAs能力显著低于健康对照,利用碳水化合物产SCFAs能力显著高于健康对照。6种发酵底物中,乳果糖是最适合ASD患儿的碳水化合物,有改善ASD患儿肠道菌群产SCFAs的潜力。  相似文献

肠道菌群和宿主健康之间有着密切的关系,其与宿主之间存在着复杂的相互作用,如菌群及其代谢产物与免疫系统的互作、脑-肠轴、肺-肠轴等.肠道菌群紊乱与多种疾病的发生和发展存在相关性,且部分微生物菌株与一些疾病的发生存在着因果关系.肠道菌群还会影响药物代谢,个体差异的肠道菌群使得不同个体对于同种药物的代谢具有很大差别;解析个体肠道菌群的状态及其与宿主之间的关系是实施个性化精准诊疗的重要环节.肠道菌群具有可塑性,通过饮食调控、益生菌/益生元/合生元补充、粪菌移植等干预手段可以使肠道菌群处于健康状态,应用肠道菌群编辑和合成肠道微生物组等新技术调控、合成肠道菌群的研究已有报道.目前,利用合成生物学等方法调控肠道菌群已成为改善和治疗疾病的有效方法之一.本文综述了肠道菌群与人体等宿主的相互作用、肠道菌群与部分疾病的相关性和因果性,以及通过肠道菌群调控改善人体健康状态的策略,展望了微生物组学和合成生物学在肠道菌群调控与合成方面的应用.  相似文献

The small intestine is an important digestive organ and plays a vital role in the life of a pig. We tested the hypothesis that the length of the small intestine is related to growth performance and intestinal functions of piglets. A total of 60 piglets (Duroc × Landrace × Yorkshire), weaned at day 21, were fed an identical diet during a 28-day trial. At the end of the study, all piglets were sacrificed, dissected and grouped according to small intestine lengths (SILs), either short small intestine (SSI), middle small intestine (MSI) or long small intestine (LSI), respectively. Positive relationships between SIL and BW, average daily gain (ADG), average daily feed intake (ADFI) and gain-to-feed ratios (G : F) were observed. Final BW, ADG, ADFI and G : F significantly increased (P < 0.05) in MSI and LSI piglets compared with SSI piglets. Short small intestine and MSI had greater jejunal mucosa sucrase and alkaline phosphatase activities (P < 0.05) than LSI piglets. The mRNA level of solute carrier family 2 member 2 (Slc2a2) in the jejunal mucosa of SSI piglets was the greatest. The MSI piglets had a greater (P < 0.05) ileal villus height than other piglets and greater (P < 0.05) villus height-to-crypt depth ratios than LSI piglets. However, the LSI piglets had a greater (P < 0.05) ileal crypt depth than SSI piglets. No significant differences in duodenal, jejunal, caecal and colonic morphologies were detected among the groups. Moreover, luminal acetate, propionate, butyrate and total short-chain fatty acid contents were greater (P < 0.05) in SSI and MSI piglets than those in LSI piglets. In addition, there was greater serum glucose concentration in MSI piglets than other piglets. Serum albumin concentration in SSI piglets was the lowest. In conclusion, these results indicate that SIL was significantly positively associated with growth performance, and in terms of intestinal morphology and mucosal digestive enzyme activity, the piglets with a medium length of small intestine have better digestion and absorption properties.  相似文献

Increased carbohydrate fermentation, compared with protein fermentation, could benefit gut health. In two in vitro experiments, the effect of carbohydrate-rich feed ingredients on fermentation characteristics of ileal chyme from pigs was assessed, using the cumulative gas production technique. Ingredients of the first experiment included gums, inulins, pectins, transgalacto-oligosaccharides, lactose and xylan. In the second experiment, a gum, pectin and transgalacto-oligosaccharides were added at different starting weights, to determine their effects on fermentation characteristics of chyme, in relation to differences in the carbohydrate concentrations. In comparison to fermentation of chyme alone, added carbohydrates led to higher total gas production (p < 0.05), faster maximum rate of gas production (except for xylan) (p < 0.05), and a decreased branched-chain fatty acids to straight chain fatty acids ratio (BCR) (p < 0.05). In the second experiment, for all carbohydrate ingredients, the BCR decreased with increasing starting weights (p < 0.05). If these supplemented dietary carbohydrates were to reach the terminal ileum of the living animal, carbohydrate fermentation in the large intestine could be stimulated, which is known to have beneficial effects on host health.  相似文献

Consumption of reishi mushroom has been reported to prevent colon carcinogenesis in rodents, although the underlying mechanisms remain unclear. To investigate this effect, rats were fed a high-fat diet supplemented with 5% water extract from either the reishi mushroom (Ganoderma lingzhi) (WGL) or the auto-digested reishi G. lingzhi (AWGL) for three weeks. Both extracts markedly reduced fecal secondary bile acids, such as lithocholic acid and deoxycholic acid (colon carcinogens). These extracts reduced the numbers of Clostridium coccoides and Clostridium leptum (secondary bile acids-producing bacteria) in a per g of cecal digesta. Fecal mucins and cecal propionate were significantly elevated by both extracts, and fecal IgA was significantly elevated by WGL, but not by AWGL. These results suggest that the reishi extracts have an impact on colon luminal health by modulating secondary bile acids, microflora, mucins, and propionate that related to colon cancer.  相似文献

蛋白质不仅是构建机体组织的主要原料,而且对动物新陈代谢活动至关重要。数目庞大的肠道细菌在机体营养素,尤其是氮营养素的代谢过程中发挥重要作用。小肠细菌能代谢部分氨基酸,进而影响宿主整体氨基酸的代谢。与小肠相比,大肠拥有更为丰富的菌群和更长的蠕动时间。一方面,进入大肠的氮营养素会影响大肠菌群的代谢和群落结构;另一方面,大肠菌群也能广泛参与氮营养素的代谢与利用,生成许多代谢产物,进而影响机体健康。本文主要综述了日粮蛋白质对大肠菌群的影响、大肠菌群代谢氨基酸的产物及其对肠道生理和机体健康的影响。  相似文献

越来越多的研究表明,肠道微生物可以影响大肠癌的发生发展。例如,产肠毒素脆弱拟杆菌、具核梭杆菌等已被证实与晚期的大肠癌和患者生存率降低相关。肠道微生物变化可以导致肠道稳态破坏,菌群数量以及类别的变化会导致宿主产生复杂的病理生理反应过程,促进大肠癌的发生发展。因此需要研究肠道微生物如何破坏肠道屏障、介导物质代谢、产生炎症因子及激活信号转导通路以及如何造成肠道微生物生态失调从而加速疾病进程。通过研究肠道微生物与大肠癌之间的相互作用,可以对大肠癌的早期诊断、治疗和预后有所帮助。本文就目前肠道微生物与大肠癌相关机制和前沿治疗的研究现状作一综述。  相似文献

Fermentation pattern and yields of microbial protein were investigated in cultures of the rabbit caecal contents supplied with glucose, xylose, starch, pectin and xylan. Rabbits at the age of 4 weeks (before weaning) and 3 months were slaughtered, their caecal contents added at 1.1% to growth media and incubated anaerobically at 39°C for 18 h. Caecal microorganisms of 4‐week‐old rabbits produced no methane and caproate, less butyrate, but more propionate than microorganisms of 3‐month‐old rabbits. In both groups of rabbits, fermentation of xylose produced significantly more propionate and less butyrate than fermentation of glucose. More propionate and less acetate was formed from starch than from pectin. In caecal cultures from 4‐week‐old rabbits with pectin, the molar percentages of acetate was significantly higher and percentages of other short‐chain fatty acids (SCFA) lower than in cultures with starch or xylan. In cultures from 3‐month‐old rabbits, fermentation of pectin and xylan produced similar SCFA profiles, different from SCFA molar composition in cultures with starch. Average production of microbial protein was 129mg per lg of carbohydrate digested (range 110 to 141mg/g). Protein yields were the same on glucose and xylose, but nonsignificantly higher on starch than on pectin and xylan. It can be concluded that the characteristics of substrate affected fermentation pattern in mixed cultures of rabbit caecal microorganisms. Substrate effects on protein yields were not statistically significant, due to high variation.  相似文献
