One Helicosporium strain, isolated from a wilted chestnut tree, evidenced in vitro antimicrobial activity against various types of bacteria and fungi, and generated a diffusible pigment. The antimicrobial compounds and the diffusible pigment of the Helicosporium sp. isolate were purified via solvent fractionation, column chromatography, and recycling preparative chromatography. Both the major antimicrobial compound and the diffusible pigment were identified as 2-methylresorcinol via nuclear magnetic resonance spectroscopy. Therefore, 2-methylresorcinol, a diffusible pigment generated by Helicosporium sp., appears to be an active antimicrobial principle. This pigment also exhibited considerable cytotoxicity against mammalian cells.  相似文献

The effect of Paxillus involutus, Laccaria laccata, Suillus luteus, S. bovinus, Hebeloma crustuliniforme and a strain of the ectendomycorrhizal fungus Mrg X (Ascomycotina) on the content of phenolic compounds: ortho-diphenols and total soluble phenols in roots of Pinus sylvestris seedlings grown in vitro was investigated. Mycorrhizal fungi varied significantly in the ability to influence the concentration of phenolics in Scots pine roots. Plants inoculated with S. luteus accumulated the lowest amount of the phenolics, whereas the amount of phenolics in pine roots inoculated with L. laccata was the highest.  相似文献

The effects of naturally-occurring organic compounds on ferrous iron oxidation by the bacterium Thiobacillus ferrooxidans were examined with a view to using these compounds to treat or prevent acid mine/rock drainage. The compounds glucose, cellobiose, galacturonic acid, and citric acid were added to the growth medium of five different strains of the bacterium and growth studies were done to determine whether or not strain differences existed with respect to organic compound sensitivity. The effects of these compounds were compared to the effects of sodium dodecyl sulfate (SDS) an anionic detergent. Each of the compounds tested had an inhibitory effect on the strains of the bacterium and sensitivity to these compounds was strain dependent. All strains appeared to be equally susceptible to SDS. Inhibitory concentrations ranged from 70 mM to >280 mM for glucose, 7.5 mM to 150 mM for cellobiose, 20 mM to 230 mM for galacturonic acid, and 50 mM to 130 mM for citric acid. SDS effectively inhibited iron oxidation for all strains at a concentration of 0.3 mM, the lowest concentration tested. Some naturally-occurring organic compounds, therefore, might be candidates for the growth control of T. ferrooxidans.  相似文献

Pseudomonas corrugata, a soil bacterium originally isolated from a temperate site of Indian Himalayan Region (IHR) is examined for its antagonistic activities against two phytopathogenic fungi, Alternaria alternata and Fusarium oxysporum. Although the bacterium did not show inhibition zones due to production of diffusible antifungal metabolites, a reduction in growth between 58% and 49% in both test fungi, A. alternata and F. oxysporum, was observed in sealed Petri plates after 120 h of incubation due to production of volatile antifungal metabolites. Reduction in biomass of A. alternata (93.8%) and F. oxysporum (76.9%) in Kings B broth was recorded after 48 h of incubation in dual culture. The antagonism was observed to be affected by growth medium, pH and temperature. The reduction in fungal biomass due to antagonism of bacteria was recorded maximum in the middle of the stationary phase after 21 h of inoculation. The production of siderophore, ammonia, lipase and chitinase in growth medium by P. corrugata were considered contributing to the antagonistic activities of the bacterium.  相似文献

[目的]对实验室分离到的菌株ZH-356进行鉴定并评价其对植物病原真菌的生物防治效果,为研发针对植物真菌病害的生防菌剂提供理论指导。[方法]通过平板对峙法确定菌株ZH-356抗菌谱,并通过16S rRNA基因序列分析确定其种属,利用离体枝条的苹果树腐烂病菌感染预防试验和患腐烂病苹果树的防治试验评价其生防效果。[结果]菌株ZH-356鉴定为链霉菌属,与直丝紫链霉菌(Streptomyces rectiviolaceus)相似性最高,为99.71%。抗菌谱试验表明,菌株ZH-356对苹果树腐烂病菌、小麦赤霉病菌、小麦根腐病菌和番茄早疫病菌等多种植物病原真菌均具有较强的抑制作用,这种抑制作用可导致苹果树腐烂病菌菌丝变粗、交叉扭曲、分支变少且容易断裂。此外,ZH-356产生的抑菌活性物质对温度和酸碱度具有高度稳定性,并且该活性物质只存在于其胞内,只有当ZH-356遇到植物病原真菌时才会被分泌出来以抑制它们的生长。在离体枝条的苹果树腐烂病菌感染预防试验中,ZH-356对苹果树腐烂病防效可达94%以上,而在患腐烂病苹果树的防治试验中,ZH-356菌制剂对苹果树腐烂病的防效高达100%。[结论]链霉菌ZH-356抑菌谱广,对多种植物病原真菌均具有良好的拮抗活性,可作为防治植物真菌病害的生防菌株,为基于ZH-356菌株的生防菌剂的开发和防治苹果树腐烂病等植物真菌病害奠定了基础。  相似文献

The effect of Paxillus involutus, Laccaria laccata, Suillus luteus, S. bovinus, Hebeloma crustuliniforme and a strain of the ectendomycorrhizal fungus Mrg X (Ascomycotina) on the content of volatile organic compounds in roots of Pinus sylvestris seedlings grown in vitro was investigated. Volatile compounds extracted with a supercritical fluid extraction were primarily terpenes and sesquiterpenes and qualitatively were the same in roots of mycorrhizal and nonmycorrhizal plants. The major monoterpenes were α-pinene, Δ3-carene and β-pinene. Inoculation of plants with the fungi resulted in statistically non-significant increases in the total amount of the volatiles. The mycorrhizal fungi showed diversified effect on the concentrations of several terpenoids.  相似文献

A perfusion technique has been described and used to study the effect of diffusible metabolites of cellulolytic fungi on one another. The effect of non-diffusible metabolites of these fungi on their cellulolytic ability has also been studied by growing them in mixed cultures and then observing the clearing of the cellulose agar. The results thus obtained with diffusible and non-diffusible metabolites are then correlated.  相似文献

An antifungal substance produced by Paenibacillus brasilensis strain Sa3 was preliminary characterized and showed to be stable after treatment with different enzymes and organic solvents and at a wide range of pH, and presented a molecular weight between 3 and 10 kDa. In vitro antagonism of this strain towards Cryptococcus neoformans was investigated by optical and electronic microscopic analyses and a fungicidal effect on C. neoformans was observed. Ultrastructural analysis showed intense changes on the fungus when it was paired cultured with strain Sa3, mainly the detachment of the capsule from the cell wall and the presence of altered organelles in the cytoplasm. This novel antifungal substance produced by P. brasilensis Sa3 may represent a new insight in antifungal therapy mainly against emergent fungi. Also, prospective studies on rhizobacteria of plants as Kalanchoe brasiliensis may offer a potential source for the discovery of bioactive compounds with medical value.  相似文献

An actinomycete strain NM94 was isolated from a Saharan soil sample by a dilution agar plating method using chitin-vitamins B medium supplemented with penicillin. The strain presented the morphological and chemical characteristics of the genus Nonomuraea. On the basis of 16S rDNA analysis and physiological tests, this isolate was found to be quite different from the known species of Nonomuraea and might be new. The strain NM94 secreted several antibiotics on yeast extract malt extract glucose medium that were active against some Gram-positive bacteria, yeast, and fungi. The antibiotics were extracted with dichloromethane and detected by bioautography on silica gel plates using Mucor ramannianus and Bacillus subtilis as the test organisms. Among these antibiotics, a complex called 94A showed interesting antifungal activity. It was selected and purified by reverse-phase HPLC. This complex was composed of five compounds. Spectroscopic studies by infrared, mass, and 1H NMR of the compounds were carried out. Initial results showed that these molecules differed from the known antibiotics produced by other Nonomuraea species.  相似文献

FJAT-4748 is a bacterial strain isolated from forest soil samples taken from Dongba Valley, Lijiang, Kunming, Yunnan Province, PR China. This strain was identified as Lysinibacillus sp. based on a 16S rRNA gene sequence analysis. FJAT-4748 has been shown to possess antifungal activity against different fungi, including Colletotrichum acutatum, Aspergillus niger, Fusarium solani, Fusarium moniliforme and Fusarium oxysporum. The results of the present study indicate that this antifungal activity results from volatile organic compounds (VOCs) produced by this strain. The observed inhibition rates of VOCs from FJAT-4748 against these fungi were 100%, 100%, 37.20%, 18.94% and 7.64%, respectively. GC-MS analysis identified 24 VOCs from FJAT-4748, which included different categories of compounds, such as aldehydes, ketones, alcohols, aromatic hydrocarbons and alkanes. Of these 24 VOCs, the most abundant compound was 2-ethyl-1-hexanol, which constituted 36.24% of the total VOCs based on the relative peak area. In the in vitro C. acutatum mycelial growth assay, 2-ethyl-1-hexanol exhibited the strongest activity, with an inhibitory rate of 100% using 10?µL/plate of this VOC. The activity of benzaldehyde was lower. 2-decanone showed the weakest activity among the compounds tested. The inhibitory activity of an artificial mixture of three VOCs against the C. acutatum increased with the amount of artificial mixture used. The inhibition rate reached 100% using 30?µL/plate of this artificial mixture in the plate test. Taken together, these results show that the antifungal VOCs produced by Lysinibacillus sp. FJAT-4748 are potentially useful as agents for controlling anthracnose caused by Colletotrichum acutatum.  相似文献

Filamentous fungi are being globally explored for the production of industrially important bioactive compounds including pigments. In the present study, a cold and pH tolerant fungus strain Penicillium sp (GEU_37), isolated from the soil of Indian Himalaya, is characterized for the production of natural pigments as influenced by varying temperature conditions. The fungal strain produces a higher sporulation, exudation, and red diffusible pigment in Potato Dextrose (PD) at 15 °C as compared to 25 °C. In PD broth, a yellow pigment was observed at 25 °C. While measuring the effect of temperature and pH on red pigment production by GEU_37, 15 °C and pH 5, respectively, were observed to be the optimum conditions. Similarly, the effect of exogenous carbon and nitrogen sources and mineral salts on pigment production by GEU_37 was assessed in PD broth. However, no significant enhancement in pigmentation was observed. Chloroform extracted pigment was separated using thin layer chromatography (TLC) and column chromatography. The two separated fractions i.e., fractions I and II with Rf values 0.82 and 0.73, exhibited maximum light absorption, λmax, at 360 nm and 510 nm, respectively. Characterization of pigments using GC–MS showed the presence of the compounds such as phenol, 2,4-bis (1,1-dimethylethyl) and eicosene from fraction I and derivatives of coumarine, friedooleanan, and stigmasterole in fraction II. However, LC-MS analysis detected the presence of derivatives of compound carotenoids from fraction II as well as derivative of chromenone and hydroxyquinoline as major compounds from both the fractions along with other numerous important bioactive compounds. The production of such bioactive pigments under low temperature conditions suggest their strategic role in ecological resilience by the fungal strain and may have biotechnological applications.  相似文献

虫草素是一种核苷类似物,具有多种药理作用。利用虫草素产生菌发酵的方式获得虫草素,可能是产生虫草素的一种新方法。为筛选能产虫草素的内生真菌,从中国传统的中药冬虫夏草子实体中共分离得到42株真菌,经PDB液体培养后,采用高效液相色谱(HPLC)鉴定其代谢产物。结果表明:菌株OS17的代谢产物中含有虫草素,且虫草素产量为40.16 mg/L。基于形态学特征和ITS序列分析,菌株OS17被鉴定为淡色生赤壳菌(Bionectria ochroleuca)。这是首次从冬虫夏草中分离得到能产虫草素的生赤壳属,表明了内生真菌具有产生多种生物活性物质的潜力。  相似文献

An Acinetobacter strain, given the code name LCH001 and having the potential to be an endophytic antagonist, has been isolated from healthy stems of the plant Cinnamomum camphora (L.) Presl, guided by an in vitro screening technique. The bacterium inhibited the growth of several phytopathogenic fungi such as Cryphonectria parasitica, Glomerella glycines, Phytophthora capsici, Fusarium graminearum, Botrytis cinerea, and Rhizoctonia solani. Biochemical, physiological, and 16S rDNA sequence analysis proved that it is Acinetobacter baumannii. When the filtrate from the fermentation broth of strain LCH001 was tested in vitro and in vivo, it showed strong growth inhibition against several phytopathogens including P. capsici, F. graminearum, and R. solani, indicating that suppression of the growth of the fungi was due to the presence of antifungal compounds in the culture broth. Moreover, the antifungal activity of the culture filtrate was significantly correlated with the cell growth of strain LCH001. The active metabolites in the filtrate were relatively thermally stable, but were sensitive to acidic conditions. Three antifungal compounds were isolated from the culture broth by absorption onto macropore resin, ethanol extraction, chromatography on silica gel or LH-20 columns, and crystallization. The structures of the bioactive compounds were identified by spectroscopic methods as isomers of iturin A, namely, iturin A2, iturin A3, and iturin A6. The characterization of an unusual endophytic bacterial strain LCH001 and its bioactive components may provide an alternative resource for the biocontrol of plant diseases.  相似文献

Bacillus subtilis KB-1111 and KB-1122 were studied to illustrate their phenotypic and biological properties. Comparison of KB-1111 with KB-1122 in morphology was carried out by microscopy and agar plate assays. Biological assay of the test strains showed that they may possess different physiological pathways from those of reference strain ATCC6501. The assessment of antagonism against the indicator fungi showed that both test strains had broad antifungal characteristics against eight phytopathogenic fungi. Of those fungal species, Magnaporthe grisea P131, Sclerotinia sclerotiorum, and F. oxysporium exhibited high sensitivity to the test strains.  相似文献

The effect of coumarin (1,2 benzopyrone) on glucose utilisation by the anaerobic rumen fungi Neocallimastix frontalis and N. patriciarum has been compared with the effect of p-coumaric acid. Both compounds largely inhibited glucose utilisation by N. patriciarum strain Cx when present in the medium at a concentration of 2.5 mM, and had a similar effect on N. frontalis strain RE1 at 5 mM. Although in earlier studies co-culturing rumen fungi with Methanobrevibacter smithii enhanced resistance to ionophores, no comparable protective effect of M. smithii was found in the present study.  相似文献

In vitro assays were undertaken to evaluate the control of two sapstain fungi, Leptographium procerum and Sphaeropsis sapinea by a combination of chitosan or chitosan oligomer and an albino strain of Trichoderma harzianum. Spore germination and hyphal growth of the test fungi were assessed on media amended with chitosan or chitosan oligomer with and without T. harzianum using either simultaneous inoculation with test fungus or inoculation 1, 2, or 3 days after pre-infection with test fungus.There was no mycelial growth of the test fungi regardless of chitosan concentrations used when either L. procerum or S. sapinea was simultaneously inoculated with T. harzianum. However, the dose–response of chitosan or chitosan oligomer on the test fungi was apparent when T. harzianum was not simultaneously inoculated with test fungus but introduced later. There was a greater growth reduction at higher concentrations (0.075–0.1% v/v) of chitosan, and overall chitosan oligomer was more effective than chitosan aqueous solution.Chitosan alone was able to restrict or delay the germination of spores but the combination of chitosan and T. harzianum inhibited spore germination and hence colony formation of test fungi regardless of time delay.  相似文献

Bacillus subtilis RP24, a promising plant growth-promoting rhizobacterium and a potent biocontrol agent isolated from pigeonpea rhizosphere was mutagenized with ethyl methanesulphonate to study the possible mechanism/s involved in the potential antagonistic properties of the strain. Over 10,000 mutants were screened against the phytopathogenic fungus Macrophomina phaseolina on potato dextrose agar plates to select ten mutants showing partial antagonism as compared to the parent strain and one negative mutant showing no antagonism. The parent strain RP24 was compared with its mutants for the presence of different possible mechanisms behind antagonism. Production of hydrogen cyanide, ammonia, siderophores, and hydrolytic enzymes like lipase, amylase, and protease were detected in all the mutants as well as the parent strain, whereas fungal cell-wall-degrading enzymes, β-1, 3-glucanase and chitosanase were not detected in any of the mutants and the parent strain, indicating that none of these mechanisms was involved in the antagonistic trait of the strain. Two possible mechanisms detected behind the antifungal trait of the strain RP24 were production of antifungal volatiles and extra-cellular diffusible antibiotics. An attempt was made for extraction, partial characterization of the extra-cellular diffusible antifungal metabolite/s by thin layer chromatography and sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS–PAGE). The extracellular, methanol soluble, hydrophobic, ninhydrin-negative, thermostable and pH-stable antifungal metabolites were characterized as cyclic lipopeptides belonging to the iturin group of peptide antibiotics.  相似文献

该研究从广西钦州市采集健康的柳树桑寄生的根、茎和叶中分离并纯化内生真菌,对真菌进行形态学鉴定,提取内生真菌的DNA,采用真菌ITS序列对内生真菌进行分子鉴定。利用A549和H460细胞作为抗肿瘤活性指示细胞,采用MTT法测定真菌乙酸乙酯提取物的抗肿瘤活性。经过初步分离分析,从柳树桑寄生中纯化出27株内生真菌,经鉴定它们分别属于7个目9个属15个种。拟盘多毛孢属和间座壳属为优势属,其中拟盘多毛孢属全部定植于寄生根;其次为新壳梭孢属、拟茎点霉属和球座菌属,各分离到3株;其他包括青霉属、镰刀菌属、炭疽菌属和派伦霉属,各分离到1株。抗肿瘤活性研究表明,有一株与Pestalotiopsis protearum的ITS序列相似性达100%的拟盘多毛孢属菌株Gen24表现有抑制肿瘤细胞A549和H460生长的特性,在真菌乙酸乙酯浸提物浓度为800μg·mL~(-1)时,对A549细胞的抑制率达到了56.92%,对H460细胞的抑制率达到了70.11%。该研究结果表明广西柳树桑寄生内生真菌较丰富,在寄主中的分布表现了一定的组织特异性,而且还存在一些具有抗肿瘤活性的菌株及其活性物质可供进一步深入研究。  相似文献

Summary Mycoparasitic behaviour ofF. udum on two soil-inhabiting microfungi,Aspergillus luchuensis andSyncephalastrum racemosum, was studied in dual cultures.Fusarium udum coiled and penetrated the host fungi and formed chlamydospores inside their hyphae and reproductive structures. The vegetative hyphae ofA. luchuensis showed swellings due to diffusible toxic substances ofF. udum in the medium and formed vesicle-like structures. This is the first record of these fungi being hosts of the mycoparasiteF. udum.  相似文献
