Re-suspension of heavy metal residuals generated by former mining activities conducted at the Tar Creek Superfund site may result in exposures of nearby residents. Currently, remediation and mitigation activities include removal of yard soil containing high concentrations of heavy metals, removal of selected chat piles, and providing assistance for families with young children. Although these are important activities to reduce potential risks to Tar Creek residents, a qualitative assessment of the site that uses mental models identifies other potential contaminants of concern, transport, and exposure pathways that may require further investigation. Mental models structure judgments about hazards and risk by visually representing a contaminated site through the use of influence diagrams. A mental model was applied to the Mayer Ranch area of the Tar Creek Superfund site in Ottawa County, Oklahoma, to support a qualitative assessment of the health risks from heavy metals exposure. Documents were reviewed to develop influence diagrams illustrating the processes, interactions, and potential exposures that could subject a hypothetical individual recreating at Mayer Ranch to various health risks. The diagrams were modified to reflect expert judgments. The mental model includes four influence diagrams: source term, fate, and transport processes that may influence human health risks due to exposure to heavy metals; natural wetland processes that could remove heavy metals; net alkaline constructed wetland processes that could remove heavy metals; and potential future impacts from the removal of sediment and land development that could increase risk. Policies, regulations, and technologies that could most reduce the probability of exposures can be determined and evaluated by focusing on the environmental transport pathways and contaminants that pose the greatest potential risks and determining the difference between the risks. Also evident are the knowledge gaps with respect to the processes within wetlands.  相似文献

Mining activities lead to the destruction of soil properties and productivity. Accomplishment of soil properties which existed before mining can be used for successful reclamation of the mined out area. With this aim proposed bauxite mines in the Eastern Ghats, India, were studied with respect to the exchangeable fraction of Na, K, Li, Ca and Mg. Na in the soils ranged from 4 to 82 mg/kg, K 15.2–746 mg/kg, Ca 119.6–2875.2 mg/kg, Li 1.2–14 mg/kg and Mg 349.8–2391.9 mg/kg. The elements studied varied significantly among the locations (ANOVA, P < 0.05). Cation Exchange Capacity (CEC) was negatively correlated with most of the variables. Principal Component Analysis (PCA) accounted for 95.6% of the total variance. PC1 (first principal component) formed of pCa, rMg, Mg:Ca, rCa and pMg, indicated Ca type enrichment in the system. PC2 and PC3 reflected the influence of SAR, pNa, rNa and Na, and Mg and CEC, respectively. The study, aimed at documenting the background concentrations of base cations in the Araku soil system, India, will be useful in later years during mine restoration programme. It would also form a base document for the mine managers during mine restoration.  相似文献

A study on the seasonal changes in the ranging area of Brown-eared pheasant and its affecting factors was conducted in the Huanglongshan Nature Reserve, Huanglong County, Shaanxi Province, China, from March 2006 to January 2007. Forty used sites and forty random plots were selected along 8–9 transects crossing the study area. Fifteen factors related to the changes of altitude were measured in each site. The results showed that the altitude of home range of Brown-eared pheasant varied with seasons: the highest home range was found in summer, and the lowest one was found in winter, while the home range in autumn was higher than that in spring. According to the frequency of occurrences of Brown-eared pheasant in different seasons, we found that its home range occurred mainly at an altitude below 1400 m in spring, above 1500 m in summer, between 1200 m and 1500 m in autumn and below 1300 m in winter. The possible reason that Brown-eared pheasants preferred to live at lower altitude in spring was to have access to water sources and the abundance of food, correlated to the slope locations, number of trees and number of shrubs; in summer, they tended to appear at highest altitude, and the average height of shrubs, average height of grasses, sheltering class and distance to edge of woods, and human disturbance were affecting factors; in autumn, they appeared at higher altitude which was correlated with the covers of trees, shrubs and grasses, and ultimately related to the abundance of food; in winter, they lived at lowest altitude, which was correlated to the distance from the edge of woods, cover of trees and sheltering class, and ultimately related to the abundance of food and shelter.  相似文献

It is assumed that small herbivores produce negligible amounts of methane, but it is unclear whether this is a physiological peculiarity or simply a scaling effect. A respiratory chamber experiment was conducted with six rabbits (Oryctolagus cuniculus, 1.57 ± 0.31 kg body mass) and six guinea pigs (Cavia porcellus, 0.79 ± 0.07 kg) offered grass hay ad libitum. Daily dry matter (DM) intake and DM digestibility were 50 ± 6 g kg? 0.75 d? 1 and 55 ± 6% in rabbits and 59 ± 11 g kg? 0.75 d? 1 and 61 ± 3% in guinea pigs, respectively. Methane production was similar for both species (0.20 ± 0.10 L d? 1 and 0.22 ± 0.08 L d? 1) and represented 0.69 ± 0.32 and 1.03 ± 0.29% of gross energy intake in rabbits and guinea pigs, respectively. In relation to body mass (BM) guinea pigs produced significantly more methane. The data on methane per unit of BM obtained in this study and from the literature on the methane output of elephant, wallabies and hyraxes all lay close to a regression line derived from roughage-fed horses, showing an increase in methane output with BM. The regression, including all data, was nearly identical to that based on the horse data only (methane production in horses [L d? 1] = 0.18 BM [kg]0.97 (95%CI 0.92–1.02)) and indicates linear scaling. Because feed intake typically scales to BM0.75, linear scaling of methane output translates into increasing energetic losses at increasing BM. Accordingly, the data collection indicates that an increasing proportion of ingested gross energy is lost because relative methane production increases with BM. Different from ruminants, such losses (1%–2% of gross energy) appear too small in non-ruminant herbivores to represent a physiologic constraint on body size. Nevertheless, this relationship may represent a physiological disadvantage with increasing herbivore body size.  相似文献

A comparative study of hypotensive effects of binuclear forms of dinitrosyl iron complexes (DNICs) with glutathione, S-nitrosoglutathione (GS-NO) and sodium nitrite (NaNO2) on rats has been carried out. The latter appeared to be the least efficient, viz., mean arterial pressure (MAP) decreased by 10 and 30 mm Hg at 25 and 100 μmoles/kg of NaNO2. In contrast, DNIC and GS-NO produced an appreciable hypotensive effect when used at much lower concentrations. GS-NO reduced MAP to the same extent, viz., to 90 mm Hg, on a hundredfold dose scale (from 0.4 up to 50 μmoles/kg) with subsequent restoration of MAP within the next 6–15 min. A similar effect was observed for DNIC except that the amplitude of the MAP drop was lower and the duration of hypotension was essentially greater. DNIC with glutathione were selected as a basic material for pilot-scale production of a hypotensive drug (commercial name Oxacom®). Preliminary pharmacological testing of Oxacom did not establish any adverse or deleterious side effects.Clinical trials of Oxacom® were performed on 14 healthy male volunteers in whom single intravenous infusion of the drug (5 mg/kg or 0.2 μmoles/kg of DNIC, respectively) evoked a characteristic response manifested as a 3–4 min drop by 24–27 mm Hg of both diastolic and systolic AP with its subsequent slow restoration within the next 8–10 h. The heart rate was quickly normalized after an initial increase. Cardiac output was unchanged despite reduced cardiac filling. A comprehensive analysis of clinical and biochemical data failed to establish any significant pathological changes in these parameters. The data obtained suggest that Oxacom® can be recommended for the second phase of clinical trials.  相似文献

AimTumor necrosis factor α (TNF-α) influences the pathogenesis of lung-fibrosis and carcinogenesis in normal cells. Polymorphisms of this gene are suggested to be associated with susceptibility to lung-diseases. Additionally TNF-α is postulated to play a significant role in regulating. Transforming growth factor (TGF-β1) expression Therefore we investigated if the TNF-α or TGF-β1 gene expression level is different within the ?308 TNF-α genotypes.MethodsQuantitative Real-time PCR of TNF-α and TGF-β1 was performed in 178 Germans. Calculations of expression were made with the 2?ΔΔCT method. Detection of the ?308 promoter polymorphism of the TNF-α gene was performed by rapid capillary PCR with melting curve analysis.ResultsThe relative TNF-α mRNA expression revealed significant differences between the TNF-α ?308 homozygote wild-type G/G (0.00079 ± 0.00011; n = 113) and the heterozygote genotype G/A (0.0005 ± 0.00008; n = 52; p = 0.030) as well as between homozygote wild-type G/G and the homozygote mutant A/A (0.00029 ± 0.00009; n = 5; p = 0.004). The relative TGF-β mRNA expression showed, similar to TNF-α, the highest mRNA expression was seen within the TNF-α ?308 homozygote wild-types, while the lowest mRNA expression lay within the homozygote mutant-types.ConclusionOur findings suggest that the G-allele of TNF-α ?308 is associated with a significantly higher TNF-α mRNA expression compared to the A-allele and that this also reflects in TGF-β expression. Therefore we support the thesis that TGF-β is regulated by TNF-α.  相似文献

BackgroundSeveral studies have attempted to identify risk factors for the development of an electrical storm (ES), which is defined as ⩾3 separate ventricular tachyarrhythmic (VT/VF) events, but in the majority of studies no triggers have been found. However, little is known about the role of inflammation and NT-proBNP in patients with ES. The aim of this study was therefore to assess the relationship of Interleukin-6 (IL-6), high-sensitivity C-reactive protein (hs-CRP) and NT-proBNP serum concentrations in ICD-patients with or without single spontaneous ventricular tachyarrhythmic events (VT/VF) and in ES.MethodsMarkers were determined in 51 patients without ICD-intervention, in 15 ICD-patients with single VT/VF-episodes during 9-months follow-up and in 20 ICD-patients with ES (blood sampling performed within 60 min after fulfilling ES criteria). VT/VF-episodes were analysed by stored ICD-electrograms.ResultsAll patients had idiopathic dilated cardiomyopathy (n = 23) or coronary artery disease (n = 63). Patients with ES revealed significantly higher mean serum concentrations of all markers (IL-6 15.19 ± 10.34 pg/mL, hs-CRP 20.12 ± 14.4 mg/L, NT-proBNP 4799 ± 4596 pg/mL) compared to baseline values of patients with single VT/VF-events during follow-up (IL-6 8.37 ± 5.8 pg/mL (p = 0.03), hs-CRP 4.7 ± 5.3 mg/dL (p < 0.001), NT-proBNP 1913 ± 2665 pg/mL (p = 0.04)) and compared to baseline values of ICD-patients without device intervention (IL-6 4.62 ± 3.66 pg/mL (p < 0.001), hs-CRP 4.1 ± 3.4 mg/L (p < 0.001), NT-proBNP 1461 ± 2281 pg/mL (p < 0.001)). In 9/20 patients presenting with ES (45%) baseline values were available. All markers were significantly higher during ES compared to event-free determination (IL-6 14.54 ± 10.43 vs. 7.03 ± 2.83 pg/mL (p = 0.04), hs-CRP 19.07 ± 16.07 vs. 6.5 ± 3.9 mg/L (p = 0.02), NT-proBNP 4218 ± 2561 vs. 2099 ± 1279 pg/mL (p = 0.03)).ConclusionsElectrical storm is associated with significantly elevated IL-6, hs-CRP and NT-proBNP serum concentrations in ICD-patients with structural heart disease. Thus, ES may be triggered by proinflammatory activity. Combined intraindividual elevation of determined markers might help to identify patients at risk of impending electrical storm.  相似文献

Fathead minnows (Pimephales promelas) comprise a species-of-choice for the hazard assessments of various environmental contaminants, including compounds capable of disrupting endocrine function. Towards this end, the use of liquid chromatography coupled with mass spectrometry (LC–MS) and/or tandem mass spectrometry (MS/MS) is gaining common use for the quantification of steroid hormones as biomarkers of endocrine stress in small-fish toxicological studies. In this work, 2-hydrazinopyridine (2-HP) was used to derivatize and quantify the physiologically relevant steroid hormones of: 17α-hydroxypregnenolone, progesterone, 11-ketotestosterone, 11-deoxycortisol and 17α,20β-dihydroxypregnenone, in the blood plasma of male and female fathead minnows. Liquid chromatographic separation was achieved using a Waters? Sunfire C18 column (2.1 mm × 50 mm with a 3.5 μm particle size) and Milli-Q water:methanol (both with 0.1% formic acid) mobile phase over a gradient of 15 min. All mass analyses were conducted using electrospray ionization in the positive mode with tandem mass spectrometry (ESI+/MS/MS). This is the first such application of 2-HP derivatization for the quantifications of the structurally and functionally diverse C19 androgen of 11-ketotestosterone; C21 progestogens of 17α-hydroxypregnenolone, progesterone and17α,20β-dihydroxypregnenone; and C21 corticosteroid of 11-deoxycortisol, in fathead minnow blood plasma. The limits of detection (LOD) were set to the lowest calibration standard that gave a signal-to-background response of ≥3, and were: 0.16 ng/ml for progesterone, 0.63 ng/ml for 17α-hydroxypregnenolone, 11-deoxycortisol and 17α,20β-dihydroxypregnenone, and 1.25 ng/ml for 11-ketotestosterone. This study demonstrates the application of 2-HP derivatization for the analysis of a variety of steroid hormones representative of endocrine function in a species of fish commonly used in toxicological studies.  相似文献

Effects of grassland conversion to cropland and forest on soil organic carbon (SOC), dissolved organic carbon (DOC) in the farming-pastoral ecotone of Inner Mongolia were investigated by direct field sampling. SOC content and DOC content in soil decreased after grassland were shifted to forest or cropland, in the sequence of grassland soil > forest soil > cropland soil. SOC stock declined by 18% after grassland shifted from to forest. Reclamation of cropland for 10 years, 15 years and 20 years lost SOC in 0–30 cm soil layer, by 34%, 14% and 18%, respectively, compared with that of grassland. DOC in 3 soil layers was within 21.1–26.5 mg/L in grassland, 12.1–14.6 mg/L in forest soil, and 8.0–14.0 mg/L in cropland soil. Correlation analysis indicated that SOC content and DOC content were positively dependent on total nitrogen content (p < 0.05), but negatively on bulk density or land use type (p < 0.05). DOC was positively correlated SOC (p < 0.01). Moreover, SOC content could be quantitatively described by a linear combination of land use types (p = 0.000, r2 = 0.712), and DOC content by a linear combination of two soil-related variables, land use types and SOC (p = 0.000, r2 = 0.861).  相似文献

Xanthoria parietinathalli were collected from a ‘ clean-air ’ location and from a polluted area.Ramalina duriaeithalli were collected from the same ‘ clean-air ’ location and some thalli were transplanted to air-polluted locations, whereR. duriaeino longer occurs. The effects of air contaminants on these two lichens were compared under controlled laboratory conditions and in field experiments. Air contaminants and exposure to bisulphite ions had little or no damaging effect onX. parietina, whereas severe damage was caused toR. duriaei, as judged by chlorophyll degradation, autofluorescence of photobionts, photosynthetic activity, membrane integrity and ATP content. AimsLate phase ischemic preconditioning (LPC) protects the heart against ischemia–reperfusion (I/R) injury. However, its effect on myocardial tissue oxygenation and related mechanism(s) is unknown. The aim of the current study is to determine whether LPC attenuates post-ischemic myocardial tissue hyperoxygenation through preserving mitochondrial oxygen metabolism.Main methodsC57BL/6 mice were subjected to 30 min coronary ligation followed by 60 min or 24 h reperfusion with or without LPC (3 cycles of 5 min I/5 min R): Sham, LPC, I/R, and LPC + I/R group. Myocardial tissue Po2 and redox status were measured with electron paramagnetic resonance (EPR) spectroscopy.Key findingsUpon reperfusion, tissue Po2 rose significantly above the pre-ischemic level in the I/R mice (23.1 ± 2.2 vs. 12.6 ± 1.3 mm Hg, p < 0.01). This hyperoxygenation was attenuated by LPC in the LPC + I/R mice (11.9 ± 2.0 mm Hg, p < 0.01). Activities of NADH dehydrogenase (NADH-DH), succinate-cytochrome c reductase (SCR) and cytochrome c oxidase (CcO) were preserved or increased in the LPC group, significantly reduced in the I/R group, and conserved in the LPC + I/R group. Manganese superoxide dismutase (Mn-SOD) protein expression was increased by LPC in the LPC and LPC + I/R mice compared to that in the Sham control (1.24 ± 0.01 and 1.23 ± 0.01, p < 0.05). Tissue redox status was shifted to the oxidizing state with I/R (0.0268 ± 0.0016/min) and was corrected by LPC in the LPC + I/R mice (0.0379 ± 0.0023/min). Finally, LPC reduced the infarct size in the LPC + I/R mice (10.5 ± 0.4% vs. 33.3 ± 0.6%, p < 0.05).SignificanceThus, LPC preserved mitochondrial oxygen metabolism, attenuated post-ischemic myocardial tissue hyperoxygenation, and reduced I/R injury.  相似文献

Acute kidney injury (AKI) frequently afflicts patients undergoing cardiopulmonary bypass and independently predicts death. Both hemoglobinemia and myoglobinemia are independent predictors of postoperative AKI. Release of free hemeproteins into the circulation is known to cause oxidative injury to the kidneys. This study tested the hypothesis that postoperative AKI is associated with both enhanced intraoperative hemeprotein release and increased lipid peroxidation assessed by measuring F2-isoprostanes and isofurans. In a case–control study nested within an ongoing randomized trial of perioperative statin treatment and AKI, we compared levels of F2-isoprostanes and isofurans with plasma levels of free hemoglobin and myoglobin in 10 cardiac surgery AKI patients to those of 10 risk-matched controls. Peak plasma free hemoglobin concentrations were significantly higher in AKI subjects (289.0 ± 37.8 versus 104.4 ± 36.5 mg/dl, P = 0.01), whereas plasma myoglobin concentrations were similar between groups. The change in plasma F2-isoprostane and isofuran levels (repeated-measures ANOVA, P = 0.02 and P = 0.001, respectively) as well as the change in urine isofuran levels (P = 0.04) was significantly greater in AKI subjects. In addition, change in peak plasma isofuran levels correlated not only with peak free plasma hemoglobin concentrations (r2 = 0.39, P = 0.001) but also with peak change in serum creatinine (r2 = 0.20, P = 0.01). Postoperative AKI is associated with both enhanced intraoperative hemeprotein release and enhanced lipid peroxidation. The correlations among hemoglobinemia, lipid peroxidation, and AKI indicate a potential role for hemeprotein-induced oxidative damage in the pathogenesis of postoperative AKI.  相似文献

Buffer zones along rivers and streams can provide water quality services by filtering nutrients, sediment and other contaminants from the surface. Redundancy analysis was used to determine the influence of the landscape pattern at the entire catchment scale and at multiple buffer zone scales (100 m, 300 m, 500 m, 1000 m and 1500 m) on the water quality in a highly urbanised watershed. Change-point analysis was further applied to estimate the specific locations along a gradient of landscape metric that result in a sudden change in the water quality variable. The landscape characteristics for 100 m buffer zones appeared to have a slightly greater influence on the water quality than the entire catchment. The patch density of urban land and the large patch index of water were recognised as the dominant variables influencing the water quality for a 100 m buffer zone. The result of change-point analysis indicated key interval values of the two landscape metrics within the 100 m buffer zone. When the patch density of urban land was >30–40 n/100 ha and the largest patch index of water was >2.5–3.5%, the watershed water quality appeared to be better protected.  相似文献

AimsThis study was to determine the effects of zinc plus arachidonic acid (ZA) treatment on the insulin action in the specific ZA target organs using hyperinsulinemic euglycemic clamp method.Main methods18 Sprague–Dawley rats weighing ~ 130 g were divided into 3 groups of 6 rats and treated them with 1) normal rat chow, 2) high fructose (60.0%) diet only, or 3) the same fructose diet plus drinking water containing 10 mg zinc plus 50 mg arachidonic acid (AA)/L. In a separate study, male Wistar rats weighing ~ 250 g were fed normal rat chow (n = 4) or high fat (66.5%) diet with drinking water containing zero (n = 9) or 10 mg AA plus 20 mg zinc /L (n = 9). After 4 week treatment, insulin action was assessed using the hyperinsulinemic eguglycemic clamp technique.Key findingsHigh fructose feeding impaired suppression of hepatic glucose output by insulin compared to controls during the clamp procedure (4.39 vs. 2.35 mg/kg/min; p < 0.05). However, ZA treatment in high fructose-fed rats showed a significant improvement of hepatic insulin sensitivity compared to non-treatment controls (4.39 vs. 2.18 mg/kg/min; p < 0.05). Glucose infusion rates in Wistar rats maintained on a high fat diet (HFD) were significantly lower compared to control rats (22.8 ± 1.3 vs. 31.9 ± 1.4 mg/kg/min; p < 0.05). ZA treatment significantly improved (~ 43%) peripheral tissue insulin sensitivity in HFD fed animals (26.7 ± 1.3 [n = 9] vs. 22.8 ± 1.3 mg/kg/min; p < 0.05).SignificanceThese data demonstrate that ZA treatment is effective in improving glucose utilization in hyperglycemic rats receiving either a high-fructose or a high-fat diet.  相似文献

In Sichuan Ruoergai Wetland National Nature Reserve, the changes in landscape patterns over a period of 17 years – from 1990 to 2007 – have been studied by means of RS, GIS technologies and field validation. In the reserve, the seven landscape patterns – river, lake, swamp, seasonal swamp, meadow, shrub and desert – were subject to the current study. The results show that the swamp, seasonal swamp, and meadow are the dominant landscape types occupying over 90% of the reserve’s total area. Over the past 17 years, the distribution area of swamp has been decreased by 3.81%, while that of seasonal swamp and meadow has been increased by 2.58% and 2.09%, respectively. In addition, the desert area has expanded by 1.19% since 2000. The nature reserve can be characterized by patch diversity, causing landscape fragmentation over the years; the total number of patches increased to 585 from 544 and the average area of the patches decreased to 283.49 h m2 from 304.29 h m2 – to the detriment of the wetland functions such as carbon sink and habitats for wild animals. In the study, the causes of the alternations are discussed as mainly resulting from climate change and human disturbances such as overgrazing, drainage, and unmanaged tourism. The study results provide a scientific basis at a strategic scale for the efficient conservation and management implementation of nature reserve zoning and assessment of landscape ecological functions and conservation values.  相似文献

We have optimized a simple and rapid preparation procedure for mushroom DNA extraction from colonies on media or from fruiting bodies for PCR amplification. The protocol combines microwaving twice for 1 min, cooling for 10 min, and centrifuging for 5 min. By using this procedure, more than 100 samples of mushroom DNA can be prepared within 1 h. The DNA obtained can be used for (1) identifying mushroom species by PCR and subsequent sequencing, (2) amplifying low copy number genes (at least 2,000 bp), and (3) screening genetic transformants. This technique will contribute to the mycology of mushroom species.  相似文献

High-risk strains of human papillomaviruses (HPVs) cause nearly all cases of cervical cancer as well as a growing number of head and neck cancers. The oncogenicity of these viruses can be attributed to the activities of their two primary oncoproteins, E6 and E7. The E6 protein has among its functions the ability to prevent apoptosis of infected cells through its binding to FADD and caspase 8. A small molecule library was screened for candidates that could inhibit E6 binding to FADD and caspase 8. Flavonols were found to possess this activity with the rank order of myricetin > morin > quercetin > kaempferol = galangin ? (apigenin, 7-hydroxyflavonol, rhamnetin, isorhamnetin, geraldol, datiscetin, fisetin, 6-hydroxyflavonol). Counter screening, where the ability of these chosen flavonols to inhibit caspase 8 binding to itself was assessed, demonstrated that myricetin, morin and quercetin inhibited GST-E6 and His-caspase 8 binding in a specific manner. The structure–activity relationships suggested by these data are unique and do not match prior reports on flavonols in the literature for a variety of anticancer assays.  相似文献

Main objective of this study was to determine the interspecific relationships between two dominant species in terms of root distribution in a typical arid tree-herbage (Elaeagnus angustifolia–Achnatherum splendens) community at Xidatan, Pingluo County, Ningxia Autonomous Region, Northwest China. Eight concentric zones (namely, Z1–Z8) were set from the bases of E. angustifolia individuals to the open lands and five soil profiles were excavated in each zone. Each soil profile was divided into five layers at the depths of 0–10 cm, 10–30 cm, 30–60 cm, 60–100 cm and 100–150 cm. Roots were collected for each species, and soil water content (SWC) and soil bulk density (SBD) were measured for each layer. We found noteworthy roots layer separation in the sub-canopy zones (Z1–Z4). The soil layers with highest fine root biomass density (FRBD) of A. splendens was primarily in the 0–10 cm, which were significantly shallower than those of E. angustifolia; whereas in the inter-canopy zones (Z5–Z8), inconsistent separation, or even overlapping of highest-FRBD-layers emerged between the two dominant species. Correlation analyses showed that negative correlations of FRBD between the two species mainly occurred in those soil layers with relatively higher FRBD and lower SWC. In contrast, positive correlations corresponded with relatively lower FRBD and higher SWC.  相似文献

Background and AimsWith the advent of regenerative therapy, there is renewed interest in the use of bone marrow as a source of adult stem and progenitor cells, including cell subsets prepared by immunomagnetic selection. Cell selection must be rapid, efficient and performed according to current good manufacturing practices. In this report we present a methodology for intra-operative preparation of CD34+ selected autologous bone marrow for autologous use in patients receiving coronary artery bypass grafts or left ventricular assist devices.Methods and ResultsWe developed a rapid erythrocyte depletion method using hydroxyethyl starch and low-speed centrifugation to prepare large-scale (mean 359 mL) bone marrow aspirates for separation on a Baxter Isolex 300i immunomagnetic cell separation device. CD34 recovery after erythrocyte depletion was 68.3 ± 20.2%, with an average depletion of 91.2 ± 2.8% and an average CD34 content of 0.58 ± 0.27%. After separation, CD34 purity was 64.1 ± 17.2%, with 44.3 ± 26.1% recovery and an average dose of 5.0 ± 2.7 × 106 CD34+ cells/product. In uncomplicated cases CD34-enriched cellular products could be accessioned, prepared, tested for release and administered within 6 h. Further analysis of CD34+ bone marrow cells revealed a significant proportion of CD45? CD34+ cells.ConclusionsIntra-operative immunomagnetic separation of CD34-enriched bone marrow is feasible using rapid low-speed Hetastarch sedimentation for erythrocyte depletion. The resulting CD34-enriched product contains CD45? cells that may represent non-hematopoietic or very early hematopoietic stem cells that participate in tissue regeneration.  相似文献
