The effect of phlorizin has been tested on hexose transport and hexose-induced changes of electrical potential (m) and conductance (g m) across the plasmalemma of rhizoid and thallus cells of the aquatic liverwort Riccia fluitans. The decrease of m (depolarization) and g m induced by 1 mM 3-oxymethyl-D-glucose (3-OMG) is substantially inhibited by simultaneous addition of 2 mM phlorizin, whereas no significant response was observed when phlorizin was added alone or several minutes after the sugar. Current-voltage data show that the 3-OMG-generated electrical inward current of 0.016 A m-2 drops to 0.010 A m-2 when phlorizin is present. Uptake as well as efflux of [14C]-3-OMG is strongly and reversibly inhibited by phlorizin between 0.2 and 5 mM. The results are consistent with our hypothesis that the hexose carrier has one binding site with competitive inhibition of glucose uptake by phlorizin (k i=0.08 mM). In the aquatic liverwort Riccia fluitans, membrane depolarization (m), change in membrane conductance (gm), and current-voltage (I-V) characteristics in the presence of different amino acids as well as the uptake of 14C-labeled amino acids were measured. L-isomers of the tested amino acids generate larger electrical effects (m, gm) than D-isomers, and the I-V characteristics show that the positive electrical inward-current of 20 mA m-2 generated by 0.5 mM D-serine is only about 50% of the current generated by adding 0.5 mM L-serine. Whereas - and -amino acids rapidly depolarize the membrane to the same extend, with -aminobutyric acid (-AB) and dipeptides no significant electrical effects have been measured. The uptake kinetics of 14C-labeled amino acids display three components: (I) A saturable high-affinity component with Ks-values of 48 M D-alanine, 12 M -aminoisobutyric acid (AIB), 9 M L-alanine, 8 M L-proline, and 6 M L-serine, respectively; (2) an apparently linear low-affinity component, and (3) an also linear but unspecific component at concentrations >20 times the given Ks-value. Uptake of 14C-labeled AIB can be inhibited competitively by all tested neutral amino acids, the L-isomers being more effective than the D-isomers, as well as by ammonium or methylamine. Vice versa, AIB competitively inhibits uptake of L-serine and L-alanine. It is concluded that an uncharged stereospecific carrier for the investigated amino acids exists in the plasmalemma of Riccia fluitans. Accumulation ratios of about 50 suggest secondary active transport driven by a transmembrane electro-chemical gradient (mainly m) which is generated by the electrogenic proton pump. By means of pH-sensitive microelectrodes, cytoplasmic pH has been monitored continuously during amino-acid transport across the plasmalemma of Riccia fluitans rhizoid cells under various experimental conditions. (i) Contrary to the general assumption that import of amino acids (or hexoses) together with protons should lead to cytoplasmic acidification, an alkalinization of 0.1–0.3 pHc units was found for all amino acids tested. Similar alkalinizations were recorded in the presence of hexoses and methylamine. No alkalinization occurred when the substrates were added in the depolarized state or in the presence of cyanide, where the electrogenic H+-pump is inhibited. (ii) After acidification of the cytoplasm by means of various concentrations of acetic acid, amino-acid transport is massively altered, although the protonmotive force remained essentially constant. It is suggested that H+-cotransport is energetically interconnected with the proton-export pump which is stimulated by the amino-acid-induced depolarization, thus causing proton depletion of the cytoplasm. The transport of several amino acids with different side-chain characteristics has been investigated in the aquatic liverwort Riccia fluitans. i) The saturation of system I (neutral amino acids) by addition of excess -aminoisobutyric acid to the external medium completely eliminated the electrical effects which are usually set off by neutral amino acids. Under these conditions arginine and lysine significantly depolarized the plasmalemma. ii) L- and D-lysine/arginine were discriminated against in favour of the L-isomers. iii) Increasing the external proton concentration in the interval pH 9 to 4.5 stimulated plasmalemma depolarization, electrical net current, and uptake of [14C]-basic amino acids. iv) Uptake of [14C]-glutamic acid took place only at acidic pHs. v) [14C]-histidine uptake had an optimum between pH 6 and 5.5. vi) Overlapping of the transport of basic, neutral, and acidic amino acids was common. It is suggested that besides system I, a second system (II), specific for basic amino acids, exists in the plasmalemma of Riccia fluitans. It is concluded that the amino-acid molecule with an uncharged side chain is the substrate for system I, which also binds and transports the neutral species of acidic amino acids, whereas system II is specific for amino acids with a positively charged side chain. The membrane potential (pd) of duck weed (Lemna gibba G1) proved to be energy dependent. At high internal ATP levels of 74 to 105 nmol ATP g-1 FW, pd was between -175 and -265 mV. At low ATP levels of 23 to 46 nmol ATP g-1 FW, pd was low, about -90 to -120 mV at pH 5.7, but -180 mV at pH 8. Upon addition of glucose in the dark or by light energy the low pd recovered to the high values. The active component of the pd was depolarized by the addition of hexoses in the dark and in the light. Hexose-dependent depolarization of the pd (= pd) followed a saturation curve similar to active hexose influx kinetics. Depolarization of the pd recovered in the dark even in the presence of the hexoses and with a 10fold enhancement in the light. Depolarization and recovery could be repeated several times with the same cell. Glucose uptake caused a maximum depolarization of 133 mV, fructose uptake half that amount, sucrose had the same effect as glucose. During 3-O-methylglucose and 2-deoxyglucose uptake the depolarizing effect was only slightly lower. The pd remained unchanged in the presence of mannitol. The glucose dependent pd and especially the rate of pd recovery proved to be pH-dependent between pH 4 and pH 8. It was independent of the presence of 1 mM KCl. Isolated internodes of Nitella (N. opaca, N. flexilis) and Nitellopsis spec. were punctured with single microelectrodes and their membrane potentials were recorded continuously during various light treatments. In red light the initial response was always a depolarization. This depolarization began with a lag-time of 0.4-3.5s and reached a steady state within 1–2 min of continuous illumination. Repolarization began within several seconds after turning off the light. The magnitude of the red-light-induced depolarization increased with the Ca2+-concentration of the medium. The largest depolarizations were recorded in 5 m mol l-1 Ca2+. Ca2+ could not be replaced in this function by Na+, Mg2+, La3+ or mannitol. Far-red light alone had no effect on the resting membrane potential. Far-red light applied immediately after red light accelerated the repolarization of the membrane potential. Far-red light applied simultaneously with red light reduced the amount of depolarization and increased the rate of repolarization. The results indicate that phytochrome and Ca2+ are involved in the light-induced depolarization of the membrane. This review treats some examples of electrogenic transport across the outer plasmamembrane (plasmalemma) of plant cells. The selection includes primary active uniport by membrane ATPases (e.g., the proton pump), secondary active transport of hexoses by proton-dependent cotransport, and passive uniport of amines. Summary The uptake of glucose and fructose from the medium by Catharanthus roseus cell suspensions was strongly inhibited by high medium salt concentration, such as found in LS (Linsmaier and Skoog 1965) medium. After inoculation into standard LS nutrient medium with less than 5 mM hexose no uptake occurred, while in low salt medium hexose was completely depleted. At a hexose concentration of 50 mM the uptake rate was higher in low salt medium than in standard medium. The lower rate of uptake at high salt concentration was not the result of a pH or osmotic effect of the salts. Probably the affinity of the hexose carrier is affected by the ion concentration of the medium. Lemna gibba L., grown in the presence or absence of Fe, reduced extracellular ferricyanide with a V max of 3.09 mol · g-1 fresh weight · h-1 and a K m of 115 M. However, Fe3+-ethylenediaminetetraacetic acid (EDTA) was reduced only after Fe-starvation. External electron acceptors such as ferricyanide, Fe3+-EDTA, 2,6-dichlorophenol indophenol or methylene blue induced a membrane depolarization of up to 100 mV, but electron donors such as ferrocyanide or NADH had no effect. Light or glucose enhanced ferricyanide reduction while the concomitant membrane depolarization was much smaller. Under anaerobic conditions, ferricyanide had no effect on electrical membrane potential difference (Em). Ferricyanide reduction induced H+ and K+ release in a ratio of 1.16 H++1 K+/2 e- (in +Fe plants) and 1.28 H++0.8 K+/2 e- (in -Fe plants). Anion uptake was inhibited by ferricyanide reduction. The cellular pathway of postphloem sugar transport was elucidated in the outer pericarp of tomato (Lycopersicon esculentum Mill cv. Floradade) fruit at 13–14 and 23–25 days after anthesis (DAA). These developmental stages are characterized by phloem-imported sugars being accumulated as starch and hexose, respectively. The symplasmic tracer, 5(6)-carboxyfluorescein, loaded into the storage parenchyma cells of pericarp discs, moved readily in the younger fruit but was immobile in fruit at 23–25 DAA. Symplasmic mobility of [14C]glucose was found to be identical to 5(6)-carboxyfluorescein. For the older fruit, the pericarp apoplasm was shown to be freely permeable to the apoplasmic tracer, trisodium 3-hydroxy-5,8,10-pyrenetrisulfonate. Indeed, the transport capacity of the pericarp apoplasm was such that the steady-state rate of in-vitro glucose uptake by pericarp discs accounted fully for the estimated rate of in-vivo glucose accumulation. For fruit at 23–25 DAA, the inhibitory effects of the sulfhydryl group modifier, p-chloromer-curibenzenesulfonic acid (PCMBS), on [14C]glucose and [14C]fructose uptake by the pericarp discs depended on the osmolality of the external solution. The inhibition was most pronounced for pericarp discs enriched in storage parenchyma. Consistent with the PCMBS study, strong fluorescent signals were exhibited by the storage parenchyma cells of pericarp discs exposed to the membrane-impermeable thiol-binding fluorochrome, mono-bromotrimethylammoniobimane. The fluorescent weak acid, sulphorhodamine G, was accumulated preferentially by the storage parenchyma cells. Accumulation of sulphorhodamine G was halted by the ATPase inhibitor erythrosin B, suggesting the presence of a plasma-membrane-bound H+-ATPase. A linkage between the putative H+-ATPase activity and hexose transport was demonstrated by an erythrosin-B inhibition of [14C]glucose and [14C]fructose uptake. In contrast, comparable evidence for an energy-coupled hexose porter could not be found in the pericarp of younger fruit at 13–14 DAA. Overall, the data are interpreted to indicate that: (i) The postphloem cellular pathway in the outer fruit pericarp shifts from the symplasm during starch accumulation (13–14 DAA) to the apoplasm for rapid hexose accumulation (23–25 DAA). (ii) An energy-coupled plasma-membrane hexose carrier is expressed specifically in storage parenchyma cells at the latter stage of fruit development.  相似文献

When grown in fructose or glucose the cells of Zygosaccharomyces bailii were physiologically different. Only the glucose grown cells (glucose cells) possessed an additional transport system for glucose and malate. Experiments with transport mutants had lead to the assumption that malate and glucose were transported by one carrier, but further experiments proved the existence of two separate carrier systems. Glucose was taken up by carriers with high and low affinity. Malate was only transported by an uptake system and it was not liberated by starved malate-loaded cells, probably due to the low affinity of the intracellular anion to the carrier. The uptake of malate was inhibited by fructose, glucose, mannose, and 2-DOG but not by non metabolisable analogues of glucose. The interference of malate transport by glucose, mannose or 2-DOG was prevented by 2,4-dinitrophenol, probably by inhibiting the sugar phosphorylation by hexokinase. Preincubation of glucose-cells with metabolisable hexoses promoted the subsequent malate transport in a sugar free environment. Preincubation of glucose-cells with 2-DOG, but not with 2-DOG/2,4-DNP, decreased the subsequent malate transport. The existence of two separate transport systems for glucose and malate was demonstrated with specific inhibitors: malate transport was inhibited by sodium fluoride and glucose transport by uranylnitrate. The incubation of Chlorella cells with glucose causes the induction of an uptake system, which is specific for the basic amino acids arginine and lysine. Both amino acids are taken up in the positively charged form and with high affinity (K m values 2 M and 7 M, respectively). The transport of arginine depolarizes the membrane by 20–30 mV. The charge compensation is achieved within a few seconds after arginine addition by the proton pump, later on K+ efflux serves for charge compensation. No evidence for arginine-proton symport was found, neither by inhibitor studies nor by use of other Chlorella strains which have a slower-responding proton pump. The accumulation of arginine is appreciably higher than it should be according to the thermodynamic force of the membrane potential. Summary Small, spherical cell fragments derived from macroplasmodia of the acellular slime moldPhysarum polycephalum by incubation in a 15 mM caffeine solution were investigated with morphological and electrophysiological techniques. Analysis of cell surface composition with the fluorescence microscope and different RITC-conjugated lectins revealed strong binding of ConA and RCA-I, weak binding of PEA, DBA and WGA and no binding of UEA-I. In addition, binding sites for external calcium ions were detected by chlorotetracycline-fluorescence. Electron microscopical staining with ruthenium red, iron or lanthanum delivered evidence for localization of lectin and calcium binding sites in a thin mucous layer on the cell surface.Electrical recordings by means of intracellular microelectrodes yielded an average membrane potential (MP) of –113 mV. Spontaneous depolarizations of the MP, with amplitudes between 10 and 80 mV and a duration of 20–30 s, failed to show a correlation with contractile activity. The ionic nature of MP was studied by varying the composition of the perfusing medium. The MP was not much affected by changes in external [Ca2+], [K+], or [Na+] but was sensitive to changes in [Cl] or [H+], with a linear dependence on pH0 in the range between 7 and 5. Metabolic inhibition by potassium cyanide or low temperature (11°C) as well as application of the protonophore CCCP caused a depolarization of the MP. When membrane vesicles from maize (Zea mays L.) coleoptiles are extracted at high buffer strength, a pH-driven, saturable association of [14C] indole-3-acetic acid is found, similar to the in-vitro auxin-transport system previously described for Cucurbita hypocotyls. Previous work on heterotrophic suspension-cultured cells has failed to detect the electrogenic processes normally associated with the plasma membranes of non-animal cells. This study reports measurements on heterotrophic cells from soybean (Glycine max L.) suspension cultures, which are shown to be amenable to impalement with microelectrodes. The plasma membrane clearly exhibits fundamental characteristics which are common to many other plant cell types: (i) a resting membrane potential significantly more negative than-100mV (measured value:121±4mV); (ii) obvious electrogenic activity, as evidenced by the marked depolarization of the membrane (87±6mV) by cyanide, and by the fact the membrane potential was frequently more negative than the equilibrium potential for K+; (iii) a finite permeability to K+ ions; (iv) electrophoretic transport of glucose. The development of a recording medium consisting primarily of 1:5 diluted growth medium was critical for successful impalement of these cells. It is proposed that the novel identification of electrogenic processes in heterotrophic suspension-cultured cells results from the deployment of electrodes with relatively dilute filling solutions, thus avoiding substantial changes in intracellular ion concentrations.The overwhelming majority of cells in soybean suspension cultures exist in small clusters, and the possibility of intercellular coupling potentially precludes assessment of membrane specific resistance and current density. Furthermore, as with most higher-plant cells, the vacuole occupies a large fraction of the intracellular volume. However, a model in which the measuring electrode is cytosolically located and the cells are electrically well-coupled is the only one which satisfactorily generates values for membrane specific resistance in a manner which is not strongly dependent on the number of cells in the cluster: other models in which the electrode tip is located in the vacuole and-or the impaled cell is electrically isolated from the others do not seem to apply. The measured values of membrane specific resistance are in the range 5.4 to 8.4 ·m2, which is in excellent agreement with comparable measurements on other plant and fungal cells. The conversion of the submerged form ofRiccia fluitans to the landform either by transfer to a moist solid surface or by treatment with abscisic acid (ABA), is accompanied by the formation of a set of new polypeptides and concomitant down-regulation of other polypeptides. Changes in gene expression were analyzed by twodimensional separations of proteins and differential screening of a cDNA library. One of the landform-specific proteins might depend on the expression of the newly discoveredRic 1 gene. The deduced amino acid sequence of the isolatedRic 1 cDNA clone codes for a protein with a molecular mass of 30.1 kDa. This polypeptide possesses two amino acid sequences which are repeated five times each and it is largely hydrophilic with the exception of a hydrophobic carboxyl-terminal region. Under ABA treatment the expression of theRic 1 mRNA had already reached its maximum after 1 h of incubation. Transferring submerged thalli onto an agar surface resulted in a slower induction. TheRic 1 gene product shows homology to an embryo-specific polypeptide of carrot seeds and to the group 3 of late-embryogenesis-abundant (LEA) proteins. Interestingly, ABA treatment improved the desiccation tolerance of the submerged thalli. Additionally, ABA stimulated the synthesis of a protein which is immunologically related to a tonoplast protein. Since was found to be extremely sensitive to air, a special reaction vessel was developed to maintain strict anaerobiosis. The dependence of the membrane potential (Em) and the membrane resistance (Rm) of Chara australis R. Brown on the pH of the external medium (pH0) was studied by controlling the activity of the plasmamembrane H+ pump under both light and dark conditions. The activity of the pump was controlled by regulating the internal ATP or Mg2+ concentration in tonoplast-free cells prepared by vacuolar perfusion. In these cells, which contained Mg · ATP (mgATP cells), Em and Rm were very sensitive to pH0, as in normal cells. Em was more negative in light than in the dark at all pH0 values tested. Tonoplast-free cells with very low [ATP]i (-ATP cells) or [Mg2+]i (-Mg cells) showed very weak dependence of Em and Rm on pH0. Thus, the active and not the passive component of Em was sensitive to pH0. At the same time, the high permeability of the plasma membrane to H+ was questioned.
