Stratiolaelaps ( = Hypoaspis) miles (Berlese) (Acari: Mesostigmata: Laelapidae) is a polyphagous soil-dwelling predatory mite that is widely marketed for use in greenhouse production systems to manage populations of dark-winged fungus gnats, Bradysia spp. (Diptera: Sciaridae) and for supplemental control of thrips. The suggestion by Walter and Campbell (2003, Biol. Control 26: 253–269) that North American commercial cultures of S. miles may actually be S. scimitus was confirmed. The development and reproduction at 21–23 °C of S. scimitus provided ad libidum with one of three different prey – the fungus gnat Bradysia aff. coprophila (Lintner), potworms (Enchytraeidae), or Sancassania aff. sphaerogaster (Zachvatkin, 1937) (Acari: Astigmata: Acaridae) – were compared. Developmental duration of the egg and non-feeding larval stages were 2.47 and 1.11 days, respectively; mortalities were 8.3 and 5.5%. Stratiolaelaps scimitus failed to develop beyond the protonymphal stage when provided with S. aff. sphaerogaster alone, although some feeding was observed. Development and reproduction of S. scimitus was successful on both fungus gnat larvae and enchytraeids, with no influence of prey on protonymphal duration (4.70 days) and mortality (8.3%), or on deutonymphal duration (4.61 days) and mortality (6.1%). Adult female S. scimitus feeding on potworms, compared to those feeding on fungus gnat larvae, had a significantly shorter pre-oviposition period (2.69 vs. 4.59 days). However, diet did not influence other adult female developmental or reproductive characteristics (oviposition period, 18.6 days; post-oviposition period, 6.2 days; total adult longevity, 27.3 days; total number of eggs, 26.5). S. scimitus reared on potworms tended (p = 0.06) to have a higher intrinsic rate of increase, a higher finite rate of increase and a shorter doubling time (r m = 0.142 day−1, λ = 1.153, Dt = 4.85 days) than those reared on fungus gnat larvae (r m = 0.105 day−1, λ = 1.110, Dt = 6.58 days), but differences in net reproductive rate (R 0) and generation time (G) were not significant.  相似文献

This study investigates the molecular phylogeny of seven lionfishes of the genera Dendrochirus and Pterois. MP, ML, and NJ phylogenetic analysis based on 964 bp of partial mitochondrial DNA sequences (cytochrome b and 16S rDNA) revealed two main clades: (1) “Pterois” clade (Pterois miles and Pterois volitans), and (2) “Pteropterus–Dendrochirus” clade (remainder of the sampled species). The position of Dendrochirus brachypterus either basal to the main clades or in the “Pteropterus–Dendrochirus” clade cannot be resolved. However, the molecular phylogeny did not support the current separation of the genera Pterois and Dendrochirus. The siblings P. miles and P. volitans are clearly separated and our results support the proposed allopatric or parapatric distribution in the Indian and Pacific Ocean. However, the present analysis cannot reveal if P. miles and P. volitans are separate species or two populations of a single species, because the observed separation in different clades can be either explained by speciation or lineage sorting. Molecular clock estimates for the siblings P. miles and P. volitans suggest a divergence time of 2.4–8.3 mya, which coincide with geological events that created vicariance between populations of the Indian and Pacific Ocean.  相似文献

Subterranean predatory mites are important biological control agents of pests in soil. In order to understand the population characteristics of two predatory mites, Macrocheles glaber Miiller and Stratiolaelaps scimitus Womersley, we studied their development, survival and fecundity data under laboratory conditions using Coboldia fuscipes Meigen as a food source and analyzed them with the age-stage, two-sex life table. Macrocheles glaber had a significantly shorter developmental time, oviposition period, longevity and lower fecundity than those of S. scimitus. The intrinsic rate of increase (λ), finite rate of increase (r), net reproductive rate (C0),net predation rate (C0), and finite predation rate (ω) of M. glaber were significantly lower than those of S. scimitus. Both population parameters and computer simulation implied that S. scimitus is a potential powerful biocontrol agent compared to M. glaber.  相似文献

The minimum life cycle of Dermacentor silvarum Olenev had a mean duration of 87.5 days (range 74–102 days) under laboratory conditions [(27±1 °C), 70% RH, 6 L: 18 D]. The mean time in (days) for the different stages of its cycle was as follows: incubation period of eggs was 15.3 days; prefeeding, feeding and premoulting periods of larvae and nymphs averaged 5.5, 4.0 and 7.3 days, and 5.2, 5.0 and 14.6 days, respectively; prefeeding, feeding, preoviposition and oviposition periods of female adults lasted for 7.8, 4.5, 4.3 and 14.0 days, respectively. A study, involving laboratory choice tests, was conducted to determine the feeding behavior, based on food preference, of the adult and larval stage of the rove beetle, Atheta coriaria Kraatz when presented with both fresh moistened oatmeal and second instar fungus gnat, Bradysia sp. nr. coprophila (Lintner) larvae in Petri dishes. Rove beetles used in this study came from a laboratory-reared colony. A rating scale from 1 to 5, based on percent missing (1 = 0 to 10%, 2 = 11 to 30%, 3 = 31 to 50%, 4 = 51 to 75%, and 5 = 76 to 100%), was used to objectively assess the amount of oatmeal and number of fungus gnat larvae consumed by each rove beetle adult and larva. In all the choice tests, A. coriaria adults and larvae preferred to feed on fungus gnat larvae (78% and 69%, respectively) significantly more so than oatmeal (9% and 5%, respectively) based on the amount of oatmeal and number of fungus gnat larvae consumed after 4 and 6 hours. There were relatively minimal differences in the amount of food consumed for both adults and larvae after 4 and 6 hours. The results of this study indicate that oatmeal may be an inexpensive supplemental food source, during the rearing process, which will not inhibit the effectiveness of rove beetles to control fungus gnat larvae when released into greenhouses.  相似文献

Allozyme variation in viviparid snails from the genus Mekongia in Thailand were examined across the different species, subspecies and geographical locations (river drainage systems). Using horizontal starch gel electrophoresis, 11 presumed allozyme loci (eight polymorphic) from eight enzyme systems were screened. Heterozygosity was moderately low (Hexp = 0.000–0.109, mean = 0.037). One population of Mekongia pongensis (Nong Khai) was monomorphic at all 11 examined loci in contrast to the other two populations, suggesting bottleneck within this population. Populations were more differentiated in the Mekongia sphaericula complex (FST = 0.587) than in either the Mekongia swainsoni complex (FST = 0.161) or M. pongensis species (FST = 0.073). Mekongia sphaericula sphaericula and Mekongia sphaericula extensa exhibited fixed allele differences at two loci, a high genetic distance (D = 0.265–0.300) and a potential polyphyletic relationship, suggesting two distinct lineages (species).  相似文献

This study was conducted to determine the attractiveness of two growing media, commonly utilized in greenhouses, to fungus gnat, Bradysia sp. nr. coprophila adults. The constituents of the most attractive growing medium tested were determined by gas chromatography analysis using a steam-distillation procedure. We found that fungus gnat adults were more attracted to the growing medium, SB300 Universal Professional Growing Mix, which contains composted bark, than to another growing medium (Sunshine LC1 Mix) and their components when tested in a series of laboratory experiments using multiple-choice experimental arenas. A higher percentage of fungus gnat adults were attracted to moist SB300 (92%) than SB300 growing medium that had been oven dried (8%). In addition, fungus gnat adults preferred SB300 although they had been reared on Sunshine LC1 Mix. When comparing the SB300 fresh from the bag to growing medium that had been pasteurized and moistened with water, gas chromatographic-mass spectroscopic data showed there were declines in several terpenoid constituents as well as an increase in fatty acids and cyclosulfur. The results of this study indicate that B. sp. nr. coprophila adults prefer certain growing media, which may assist greenhouse producers in managing fungus gnats in crop production systems.  相似文献

Synchronization of estrous and fixed-time artificial insemination (FTAI) was conducted during the reproductive season of 2008 (March–April) in a local red deer breeding farm in Argentina. Multiparous suckling hinds (n = 38) were artificially inseminated following hormonal treatment (intravaginal sponge containing 100 mg of medroxiprogesterone acetate). At the time of sponge removal (day 12) 250 IU of eCG and 500 μg of PGF2α were given to each hind. The FTAI was performed at 48–55 h after device removal with cryopreserved semen imported from New Zealand. Rectal-transcervical AI method (similar to that in cattle) was performed and semen was deposited within the uterine body (n = 28) or the cervix (n = 10). Pregnancy was diagnosed by means of ultrasonography 44 days after FTAI. The overall pregnancy rate was 36.8% (14/38). Percentage of does that became pregnant with intrauterine seminal deposition was 42.9% (12/28) whereas pregnancy rate in the hinds with intracervical AI was 20% (2/10; P = 0.27).  相似文献

Aprostocetus vaquitarum (Wolcott) causes 78–91 percent mortality to eggs of Diaprepes abbreviatus (L.), under field conditions in southern Florida. In the laboratory, A. vaquitarum was reared on D. abbreviatus eggs at 25 °C, a photoperiod of 12:12 (L:D) and with abundant hosts, A. vaquitarum adult females lived around 15 days. Oviposition was significantly affected by the age of the host egg mass. Egg masses aged 0- to 3-day-old were accepted significantly better than those aged 4–6 days. The mean number of eggs deposited per female was around 53, with extreme values of 124 and 19 eggs per female. Using these data in combination with the sex ratio observed in the field (0.16) and the duration of the preimaginal stages, rm (0.168–0142 day−1), T (22.39–22.89 days), and R0 (43.03–25.81 females per female) were calculated.  相似文献

Visceral leishmaniasis is a zoonosis whose primary vector in Brazil is the sandfly Lutzomyia longipalpis Lutz & Neiva. Presently, efforts to control the vector have not been effective in reducing the prevalence of disease. A possible alternative to current strategies is the biological control of the vector using entomopathogenic fungi. This study evaluates the effects of the fungus, Beauveria bassiana (Bals.) Vuilleman, in different developmental stages of L. longipalpis. Five concentrations of the fungus were utilized ranging from 104 to 108 conidia/ml, with appropriate controls. The unhatched eggs, larvae and dead adults exposed to B. bassiana were sown to reisolate the fungus. The fungus was subsequently identified by polymerase chain reaction (PCR) and DNA sequencing. Exposure to B. bassiana reduced the number of eggs that hatched by 59% (< 0.01). The longevity of infected adults was 5 days, significantly lower than that of the negative control which was 7 days (< 0.001). The longevity of the adult sandfly exposed to the positive chemical (pyrethroid, cypermetherin) control was less than 1 day. The effects of fungal infection on the hatching of eggs laid by infected females were also significant and dose-dependent (< 0.05). With respect to fungal post-infection growth parameters, only germination and sporulation were significantly higher than the fungi before infection (< 0.001). The identity of the reisolated fungus was confirmed by automated DNA sequencing post-passage in all insect stages. These data show that B. bassiana has good pathogenic potential, primarily on L. longipalpis larvae and adults. Consequently, the use of this fungus in sandfly control programs has potential in reducing the use of chemical insecticides, resulting in benefits to humans and the environment.  相似文献

The white grub, Hoplia philanthus Füessly (Coleoptera: Scarabaeidae), is a major pest of turf and ornamental plants in Belgium. Previously, the combination of lethal concentration of the entomopathogenic nematodes Heterorhabditis megidis or Steinernema glaseri with the entomopathogenic fungus Metarhizium anisopliae (strain CLO 53) caused additive or synergistic mortality to third-instar H. philanthus in the laboratory and greenhouse. In this present study, we examined this interaction under field conditions and compared a combination of a commercial formulation of Heterorhabditis bacteriophora (Nema-green®) and M. anisopliae. Controls were M. anisopliae, chlorpyrifos (Dursban 5 Granules) and H. bacteriophora. Field applications (surface or subsurface) were made against a mixed population of second/third-instar H. philanthus at a sport field and lawn infested in the province of West-Flanders. In both trials, the combination of M. anisopliae with H. bacteriophora at 5 × 1012 conidia/ha +2.5 × 109 infective juveniles/ha resulted in additive or synergistic effects, causing more than 95% grub mortality when the nematodes was applied 4 weeks after the application of fungus. However, application of nematode, chlorpyrifos or fungus alone provided 39–66%, 42–60% (surface) and 33–76%, 82–100% or 37–65%, (subsurface) control of H. philanthus. We concluded that the pathogen combinations we tested are compatible elements of integrated pest management and are likely to improve control of H. philanthus larvae and perhaps other insect pests beyond what is expected from single application of the pathogen.  相似文献

A new pest of Welsh onion and carrot, the dark-winged fungus gnat, Bradysia sp., was found in the northern region of Saitama Prefecture, Japan. DNA sequences of mitochondrial cytochrome c oxidase subunit I revealed that Bradysia sp. is identical to the Chinese chive gnat, Bradysia odoriphaga Yang and Zhang. This suggests that Bradysia sp. may be an invasive species that originates from continental East Asia. Direct sequencing analysis and PCR using a species-specific primer were found to be useful tools for discriminating Bradysia sp. from other native Japanese fungus gnats, such as Bradysia impatiens (Johannsen), Pnyxia scabiei (Hopkins), and Lycoriella ingenua (Dufour).  相似文献

The diversity and genetic structure of the cycad Dioon sonorense (De Luca, Sabato & Vázq. Torres) Chemnick, T.J. Greg. & S. Salas-Mor. were evaluated from the four known populations in tropical dry forests of Sonora, Mexico. The estimates of genetic diversity in the 19 loci analysed were: mean number of alleles per locus (A = 2.0), percentage of polymorphic loci (P = 81.6) and expected heterozygosity (HE = 0.314). The subpopulations were found to be in Hardy–Weinberg equilibrium at the local level (Fis), even though we found a global excess of homozygotes (Fit = 0.13). Genetic variation attributed to differences among populations was 15%. Our results indicate that the geographical isolation caused by the historical effects of the Pleistocene among populations has probably generated clinal variation of the allelic frequencies at two loci, in relation to their latitudinal distribution. We compare our results with the long-lived southern hemisphere trees Fitzroya and Nothofagus in relationship to the Pleistocene glaciations. We also infer that the populations have become fragmented recently due to increasing pressure of habitat conversion, disturbance and illegal extraction of plants and seeds for the international horticultural trade. We therefore recommend three aspects: (i) propagation of this species for reintroduction, (ii) declare the localities of its northern distribution as a protected area, and (iii) an amendment of the current International Union for Conservation of Nature Red List categorisation for this species to ‘Critically Endangered’.  相似文献

A chitinase was purified from the stomach of a fish, the silver croaker Pennahia argentatus, by ammonium sulfate fractionation and column chromatography using Chitopearl Basic BL-03, CM-Toyopearl 650S, and Butyl-Toyopearl 650S. The molecular mass and isoelectric point were estimated at 42 kDa and 6.7, respectively. The N-terminal amino acid sequence showed a high level of homology with family 18 chitinases. The optimum pH of silver croaker chitinase toward p-nitrophenyl N-acetylchitobioside (pNp-(GlcNAc)2) and colloidal chitin were observed to be pH 2.5 and 4.0, respectively, while chitinase activity increased about 1.5- to 3-fold with the presence of NaCl. N-Acetylchitooligosaccharide ((GlcNAc)n, n = 2–6) hydrolysis products and their anomer formation ratios were analyzed by HPLC using a TSK-GEL Amide-80 column. Since the silver croaker chitinase hydrolyzed (GlcNAc)4–6 and produced (GlcNAc)2–4, it was judged to be an endo-type chitinase. Meanwhile, an increase in β-anomers was recognized in the hydrolysis products, the same as with family 18 chitinases. This enzyme hydrolyzed (GlcNAc)5 to produce (GlcNAc)2 (79.2%) and (GlcNAc)3 (20.8%). Chitinase activity towards various substrates in the order pNp-(GlcNAc)n (n = 2–4) was pNp-(GlcNAc)2 >> pNp-(GlcNAc)4 > pNp-(GlcNAc)3. From these results, silver croaker chitinase was judged to be an enzyme that preferentially hydrolyzes the 2nd glycosidic link from the non-reducing end of (GlcNAc)n. The chitinase also showed wide substrate specificity for degrading α-chitin of shrimp and crab shell and β-chitin of squid pen. This coincides well with the feeding habit of the silver croaker, which feeds mainly on these animals.  相似文献

Allozyme variation was examined in 223 samples of the operculate land snail Cyclophorus fulguratus from 13 localities across three regions of Thailand. Using horizontal starch gel electrophoresis, 13 allozyme presumed loci (12 polymorphic) were screened. Heterozygosity was moderate in C. fulguratus (Hexp = 0.008–0.127) with a high genetic heterogeneity among samples (Fst = 0.734). Populations showed a greater genetic differentiation in central Thailand (Fst = 0.380) than in northeastern Thailand (Fst = 0.108), suggesting frequent gene flow among populations in northeastern Thailand. C. fulguratus exhibits a strong pattern of isolation by distance over the entire tested species range in Thailand and may potentially have been involved in an extensive local fragmentation. Results of the distance analysis revealed that large genetic divergence has occurred among the central, northeastern and eastern Thailand groups [D = 0.361–0.701], strongly suggesting populations from these three geographical regions may actually represent or else be evolving into separate species.  相似文献

The effect of ozone on the respiration of three species of adult stored-product Coleoptera was tested in an air-tight flask. Sitophilus oryzae (L.), Rhyzopertha dominica (F.) and Tribolium castaneum (Herbst) adults were exposed to atmosphere containing 0.1, 0.2 or 0.4 μg/ml initial ozone at 23–25 °C and 50% r.h. Carbon dioxide (CO2) production reflected the respiration rates of insects and was determined with a gas chromatograph (GC). The experiments showed that the effects of ozone on respiration had two distinct phases. Phase 1 involved a lower respiration rate of the adult stored-product Coleoptera under ozone atmosphere and reflected the need for insects to reduce ozone toxicity. After 1 h, CO2 production of S. oryzae was 3.19, 2.63, 2.27 and 1.99 μl/mg for the ozone concentration of 0, 0.1, 0.2 and 0.4 μg/ml, respectively. The results also showed that there were decreases in the rate of respiration in R. dominica and T. castaneum with an increase in ozone concentration. During phase 2, respiration of S. oryzae, R. dominica, and T. castaneum adults treated with ozone increased as the ozone degraded to oxygen. After 7 h, the effect of ozone on CO2 production, relative to the control, changed from a decrease to an increase. The findings in relation to control strategies were discussed.  相似文献

Feltiella acarisuga (Vallot) is a common predatory gall midge, which feeds on many species of spider mites. All major life history and life table parameters of F. acarisuga were determined using the carmine spider mite, Tetranychus cinnabarinus (Boisduval) eggs as prey under laboratory conditions [26.7 ± 2 °C, 75 ± 5% RH, and a photoperiod of l4:10 (L:D) h]. Developmental times of F. acarisuga were 2.6, 7.1, and 6.7 d for eggs, larvae, and pupae, respectively, with an average of 16.4 d from egg to adult emergence. Female immatures development took ≈1 d longer than male immatures did. Adult F. acarisuga lived an average of 12.8 d, and the female adults (13.3 d) lived significantly longer than male adults did (11.9 d). After an average of 1.5 d preoviposition period, each female laid an average of 27.3 eggs in its life span with an average of 2.1 eggs per day and an average of 2.8 eggs on each of its oviposition day. The raw data were analyzed using an age-stage, two-sex life table method that takes into consideration of the variable developmental rates among individuals and between sexes. The intrinsic rate of natural population increase (r), net reproductive rates (Ro), gross reproductive rate (∑mx), generation time (T), and doubling time (DT), and the finite rate of increase (λ) of F. acarisuga were estimated using the age-stage, two-sex (male and female) life table analysis as 0.122 d−1, 16.19 eggs per female, 20.81 eggs per female, 22.81 d, 1.1298 d, and 5.7 d−1, respectively. Each of the first, second, and third instar larvae of F. acarisuga consumed an average of 35.5, 54.0 and 86.9 T. cinnabarinus eggs per day, respectively. Larvae of F. acarisuga could consume an average of 175.4, T. cinnabarinus eggs, and female larvae consumed 14% more spider mite eggs (187.6 eggs) than male larvae (165.1 eggs). The significance of other life table parameters related to the population and the potential of using F. acarisuga as a biological control agent are discussed.  相似文献

The subterranean insect Bradysia cellarum Frey (Diptera: Sciaridae) is a notorious and major pest of Chinese chives, Allium tuberosum Rottler ex Sprengle (Amaryllidaceae) in China. Current chemical control of B. cellarum results in low insecticide efficacy, high cost and pesticide resistance, therefore there is an urgent need for sustainable management. Here, greenhouse experiments were conducted to evaluate the potential biocontrol agent Stratiolaelaps scimitus Womersley (Acari: Laelapidae) against B. cellarum. The number of B. cellarum larvae in soil declined from 17.6 to 0 in 4 months after releasing predatory mites in high density (5,000 adults per row); treatment was less effective under low densities of 2500 adults per row. To determine whether S. scimitus can be used in combination with soil solarization by film mulching over 40 °C for 4 h, we also evaluated heat tolerance of S. scimitus in laboratory and its control efficacy against B. cellarum after high-temperature treatment mimicking the film mulching in greenhouse. As our results showed that egg hatchability of S. scimitus was 2.6% at 38 °C and adult survival rate was 2% at 40 °C for 4 h, respectively, we concluded S. scimitus was largely inviable and could not reproduce at 40 °C. This temperature was the baseline of soil solarization, suggesting predatory mites should be released after soil solarization. When using S. scimitus after soil solarization or when using soil solarization as single treatment, fly larvae declined similarly from initial density of 18 to 0 or 17.2 to 0, respectively, within a month. Thus, our study suggests the potential of S. scimitus as a biocontrol agent of B. cellarum in greenhouse, and the most effective strategy is to combine film mulching and predatory mites (after soil heating) to control B. cellarum in chive productions.

The ladybird beetle, Stethorus gilvifrons, is a major predator of the red spider mite, Oligonychus coffeae, infesting tea. Biology, life table and predatory efficiency of S. gilvifrons were studied under laboratory conditions. Its average developmental period from egg to adult emergence was 19.2 days. After a mean pre-oviposition period of 5.3 days, each female laid an average of 149.3 eggs. Adult females lived for 117.3 days and males for 41.5 days. The life table of the beetle was characterized by an intrinsic rate of increase (r) of 0.066 day−1, net reproductive rate (R 0) of 72.2 eggs/female, gross reproduction rate (Σm x ) of 82.3 eggs/female, generation time (T) of 64.9 days, doubling time of 10.5 days and finite rate of increase (λ) of 1.07 day−1. Population dynamics of S. gilvifrons and its prey, O. coffeae, was monitored by sampling 25 tea leaves from each experimental block grown under the prevailing field conditions. Populations of S. gilvifrons reached a peak during January to March and had low incidence during June to November. Peaks in the populations of S. gilvifrons coincided with the abundance of O. coffeae in tea fields. Weather factors such as low temperature, high humidity and heavy rainfall adversely affected the populations of S. gilvifrons. The predatory efficiency of S. gilvifrons increased during the growth of larval instars. An adult female consumed 205.0 eggs, 92.2 larvae, 81.8 nymphs and 52.4 adult mites per day.  相似文献

Biological control of invasive saltcedars (Tamarix spp.) in the western U.S. by exotic tamarisk leaf beetles, Diorhabda spp., first released in 2001 after 15 years of development, has been successful. In Texas, beetles from Crete, Greece were first released in 2004 and are providing control. However, adults alight, feed and oviposit on athel (Tamarix aphylla), an evergreen tree used for shade and as a windbreak in the southwestern U.S. and México, and occasionally feed on native Frankenia spp. plants. The ability of tamarisk beetles to establish on these potential field hosts was investigated in the field. In no-choice tests in bagged branches, beetle species from Crete and Sfax, Tunisia produced 30–45% as many egg masses and 40–60% as many larvae on athel as on saltcedar. In uncaged choice tests in south Texas, adult, egg mass and larval densities were 10-fold higher on saltcedar than on adjacent athel trees after 2 weeks, and damage by the beetles was 2- to 10-fold greater on saltcedar. At a site near Big Spring, in west-central Texas, adults, egg masses and 1st and 2nd instar larvae were 2- to 8-fold more abundant on saltcedar than on athel planted within a mature saltcedar stand being defoliated by Crete beetles, and beetles were 200-fold or less abundant or not found at all on Frankenia. At a site near Lovelock, Nevada, damage by beetles of a species collected from Fukang, China was 12–78% higher on saltcedar than on athel planted among mature saltcedar trees undergoing defoliation. The results demonstrate that 50–90% reduced oviposition on athel and beetle dispersal patterns within resident saltcedar limit the ability of Diorhabda spp. to establish populations and have impact on athel in the field.  相似文献
