I examined the activity of fungi associated with yellow poplar (Liriodendron tulipifera) and white oak (Quercus alba) leaves in two streams that differed in pH and alkalinity (a hard water stream [pH 8.0] and a soft water stream [pH 6.7]) and contained low concentrations of dissolved nitrogen (<35 microg liter(-1)) and phosphorus (<3 microg liter(-1)). The leaves of each species decomposed faster in the hard water stream (decomposition rates, 0.010 and 0.007 day(-1) for yellow poplar and oak, respectively) than in the soft water stream (decomposition rates, 0.005 and 0.004 day(-1) for yellow poplar and oak, respectively). However, within each stream, the rates of decomposition of the leaves of the two species were not significantly different. During the decomposition of leaves, the fungal biomasses determined from ergosterol concentrations, the production rates determined from rates of incorporation of [(14)C]acetate into ergosterol, and the sporulation rates associated with leaves were dynamic, typically increasing to maxima and then declining. The maximum rates of fungal production and sporulation associated with yellow poplar leaves were greater than the corresponding rates associated with white oak leaves in the hard water stream but not in the soft water stream. The maximum rates of fungal production associated with the leaves of the two species were higher in the hard water stream (5.8 mg g(-1) day(-1) on yellow poplar leaves and 3.1 mg g(-1) day(-1) on oak leaves) than in the soft water stream (1.6 mg g(-1) day(-1) on yellow poplar leaves and 0.9 mg g(-1) day(-1) on oak leaves), suggesting that effects of water chemistry other than the N and P concentrations, such as pH or alkalinity, may be important in regulating fungal activity in streams. In contrast, the amount of fungal biomass (as determined from ergosterol concentrations) on yellow poplar leaves was greater in the soft water stream (12.8% of detrital mass) than in the hard water stream (9.6% of detrital mass). This appeared to be due to the decreased amount of fungal biomass that was converted to conidia and released from the leaf detritus in the soft water stream.

Effects of Cadmium on Aquatic Hyphomycetes
Two kinds of experiments, sporulation and growth experiments, were carried out to demonstrate the effect of cadmium on aquatic hyphomycetes. Oak (Quercus petraea L.) leaves were exposed in a hard-water stream (Lüssel, Swiss Jura) and a soft-water stream (Ibach, Black Forest) for 2 months. In the laboratory, fungal sporulation on the leaves in stream water enriched with cadmium (as CdCl2) was studied. A measurable effect was found when the cadmium concentration exceeded 0.1 ppm (0.1 mg/liter). Concentrations higher than 100 ppm inhibited conidium production completely. This toxic effect of cadmium was species dependent and much higher in soft water (water with low concentrations of calcium and magnesium) than in hard water. Growth experiments with Alatospora acuminata Ingold, Clavariopsis aquatica De Wildeman, Flagellospora curvula Ingold, Heliscus lugdunensis Saccardo and Therry, and Tetracladium marchalianum De Wildeman showed the same pattern of cadmium sensitivity as that seen in the sporulation experiments. Mycelial growth was less sensitive to cadmium than was fungal sporulation. High concentrations of competing cations (e.g., calcium and zinc) or potential ligands could reduce cadmium toxicity. Calcium content seems to be the most important factor responsible for the different sensitivity of aquatic hyphomycetes in hard and soft water.

In several experimental techniques D2O rather then H2O is often used as a solvent for proteins. Concerning the influence of the solvent on the stability of the proteins, contradicting results have been reported in literature. In this paper the influence of H2O-D2O solvent substitution on the stability of globular protein structure is determined in a systematic way. The differential scanning calorimetry technique is applied to allow for a thermodynamic analysis of two types of globular proteins: hen's egg lysozyme (LSZ) with relatively strong internal cohesion ("hard" globular protein) and bovine serum albumin (BSA), which is known for its conformational adaptability ("soft" globular protein). Both proteins tend to be more stable in D2O compared to H2O. We explain the increase of protein stability in D2O by the observation that D2O is a poorer solvent for nonpolar amino acids than H2O, implying that the hydrophobic effect is larger in D2O. In case of BSA the transitions between different isomeric forms, at low pH values the Nm and F forms, and at higher pH values Nm and B, were observed by the presence of a supplementary peak in the DSC thermogram. It appears that the pH-range for which the Nm form is the preferred one is wider in D2O than in H2O.

Lignin content, composition, distribution as well as cell wall thickness, structures, and type of tissue have a measurable effect on enzymatic hydrolysis of cellulose in lignocellulosic feedstocks. The first part of our work combined compositional analysis, pretreatment and enzyme hydrolysis for fractionated pith, rind, and leaf tissues from a hybrid stay-green corn, in order to identify the role of structural characteristics on enzyme hydrolysis of cell walls. The extent of enzyme hydrolysis follows the sequence rind < leaves < pith with 90% conversion of cellulose to glucose in 24 h in the best cases. Physical fractionation of corn stalks or other C(4) grasses into soft and hard tissue types could reduce cost of cellulose conversion by enabling reduced enzyme loadings to hydrolyze soft tissue, and directing the hard tissue to other uses such as thermal processing, combustion, or recycle to the land from which the corn was harvested.

This study examined the effects of water hardness on the physiological responses associated with high pH exposure in multiple strains of diploid and triploid rainbow trout Oncorhynchus mykiss. To accomplish this, three wild strains and one domesticated strain of diploid and triploid O. mykiss were abruptly transferred from control soft water (City of Vancouver dechlorinated tap water; pH 6·7; [CaCO3] < 17·9 mg l?1) to control soft water (handling control), high pH soft water (pH 9·5; [CaCO3] < 17·9 mg l?1), or high pH hard water (pH 9·5; [CaCO3] = 320 mg l?1) followed by sampling at 24 h for physiological measurements. There was a significant effect of ploidy on loss of equilibrium (LOE) over the 24 h exposure, with only triploid O. mykiss losing equilibrium at high pH in both soft and hard water. Furthermore, exposure to pH 9·5 resulted in significant decreases in plasma sodium and chloride, and increases in plasma and brain ammonia with no differences between soft and hard water. There was no significant effect of strain on LOE, but there were significant differences between strains in brain ammonia and plasma cortisol. Overall, there were no clear protective effects of hardness on high pH exposure in these strains of O. mykiss.

Cardiovascular surveys were conducted in areas with different water supplies. In two areas with hard water 243 men were seen and in three areas with soft water 357 men were seen. There was no evidence of an important difference between the two areas in blood pressure, serum cholesterol, blood sugar, or packed cell volume. A variety of measurements were made on an E.C.G. The P-R interval was significantly longer in the men in the hard water area, and a 40-complex trace showed ventricular extrasystoles in a significantly higher proportion of these men than in those in the soft water area.

Heat and stable isotope tracers were used to study axial and radial water transport in relation to sapwood anatomical characteristics and internal water storage in four canopy tree species of a seasonally dry tropical forest in Panama. Anatomical characteristics of the wood and radial profiles of sap flow were measured at the base, upper trunk, and crown of a single individual of Anacardium excelsum, Ficus insipida, Schefflera morototoni, and Cordia alliodora during two consecutive dry seasons. Vessel lumen diameter and vessel density did not exhibit a consistent trend axially from the base of the stem to the base of the crown. However, lumen diameter decreased sharply from the base of the crown to the terminal branches. The ratio of vessel lumen area to sapwood cross-sectional area was consistently higher at the base of the crown than at the base of the trunk in A. excelsum, F. insipida and C. alliodora, but no axial trend was apparent in S. morototoni. Radial profiles of the preceding wood anatomical characteristics varied according to species and the height at which the wood samples were obtained. Radial profiles of sap flux density measured with thermal dissipation sensors of variable length near the base of the crown were highly correlated with radial profiles of specific hydraulic conductivity (k(s)) calculated from xylem anatomical characteristics. The relationship between sap flux density and k(s) was species-independent. Deuterium oxide (D(2)O) injected into the base of the trunk of the four study trees was detected in the water transpired from the upper crown after only 1 day in the 26-m-tall C. alliodora tree, 2 days in the 28-m-tall F. insipida tree, 3 days in the 38-m-tall A. excelsum tree, and 5 days in the 22-m-tall S. morototoni tree. Radial transport of injected D(2)O was detected in A. excelsum, F. insipida and S. morototoni, but not C. alliodora. The rate of axial D(2)O transport, a surrogate for maximum sap velocity, was positively correlated with the predicted sapwood k(s) and with tree height normalized by the relative diurnal water storage capacity. Residence times for the disappearance of the D(2)O tracer in transpired water ranged from 2 days in C. alliodora to 22 days in A. excelsum and were positively correlated with a normalized index of diurnal water storage capacity. Capacitive exchange of water between stem storage compartments and the transpiration stream thus had a profound influence on apparent rates of axial water transport, the magnitude of radial water movement, and the retention time in the tree of water taken up by the roots. The inverse relationship between internal water exchange capacity and k(s) was consistent with a trade-off contributing to stability of leaf water status through highly efficient water transport at one extreme and release of stored water at the other extreme.

Leaf decomposition in an experimentally acidified stream channel
Decomposition of Alnus rugosa and Myrica Gale leaves immersed in artificial stream channels fed by a small headwater creek was followed over a three month period. At the end of experiment, remaining weights of both leaf types confined in litter bags were significantly higher after immersion in experimentally acidified water (pH 4.0) than when immersed in control water (pH 6.2–7.0). For both types of leaves and for all sampling times, there was generally no difference in the C:N ratios between leaves in acidified and those in control water. In control water, oxygen uptake by microorganism on A. rugosa leaves was significantly higher after 46 days of immersion, whereas differences between treatments appeared only after 69 days for M. Gale leaves. Transfer of A. rugosa leaves from acid to control water led to a rapid increase in microbial activity; this increased activity was reflected in a fast weight loss of the leaves. For both leaf types, total numbers of macroinvertebrates were usually higher in litter bags immersed in control water. Macroinvertebrates colonizing the litter bags were mainly collector-gatherers: Chironomidae were numerically dominant in control leaf packs whereas Oligochaeta dominated in acid leaf packs. Macroinvertebrate biomass in M. Gale litter was higher in control than in acidified water, which contrasted with macroinvertebrate biomass in A. rugosa leaf packs which was not significantly different between treatments. Macroinvertebrate contribution to the breakdown of leaf litter was thus considered less important than the microbial contribution. This study demonstrated that decomposition of leaf litter in acidic headwater streams can be seriously reduced, mainly as a result of a lower microbial activity.

1. After it enters streams, terrestrially derived organic matter (OM) rapidly absorbs water. Using field and laboratory experiments, we examined how this process affected the buoyancy, settling velocity, transport distance and retention locations of four types of organic matter typically found in Pacific coastal streams (‘flexible’ red alder leaves and three ‘stiff’ particle types – Douglas‐fir needles, red cedar fronds and Douglas‐fir branch pieces). 2. Immersion in water rapidly changed the physical characteristics of alder leaves, Douglas‐fir needles and red cedar fronds, which all reached constant still‐water settling velocities after only a few days of soaking. In contrast, the settling velocity of branch pieces continued to increase for 13 days, eventually reaching much higher values than any other OM type. Dried alder leaves became negatively buoyant after only two days of immersion, while other types took substantially longer (up to 24 days) before the specific gravity of all particles was >1. 3. We released saturated OM particles in an experimental channel and found that all particle types travelled further in a fast, shallow ‘riffle’ than a slow, deep ‘pool’. Comparisons with a passive settlement null model indicated that leaves were retained more rapidly than expected in the riffle (by large protruding stones), while the three stiff particle types travelled further than expected (probably due to turbulent suspension) and were retained when they settled in deeper water between larger stones. In pools, passive settlement appeared to dominate the retention of all OM types, with leaves travelling furthest. 4. These retention patterns corresponded well with those observed when saturated OM particles collected in the field were released in two pools and two riffles in a second‐order coastal stream. 5. When the experimental channel and in‐stream data were combined, the retention rates of the three stiff OM types were closely related to calculated Rouse numbers (Rouse number = particle settling velocity/shear velocity), whereas the retention rate of alder leaves was not. This suggests that different physical mechanisms are responsible for the retention of leaves and stiff OM types in shallow streams.

The absolute amount of microbial biomass and relative contribution of fungi and bacteria are expected to vary among types of organic matter (OM) within a stream and will vary among streams because of differences in organic matter quality and quantity. Common types of benthic detritus [leaves, small wood, and fine benthic organic matter (FBOM)] were sampled in 9 small (1st-3rd order) streams selected to represent a range of important controlling factors such as surrounding vegetation, detritus standing stocks, and water chemistry. Direct counts of bacteria and measurements of ergosterol (a fungal sterol) were used to describe variation in bacterial and fungal biomass. There were significant differences in bacterial abundance among types of organic matter with higher densities per unit mass of organic matter on fine particles relative to either leaves or wood surfaces. In contrast, ergosterol concentrations were significantly greater on leaves and wood, confirming the predominance of fungal biomass in these larger size classes. In general, bacterial abundance per unit organic matter was less variable than fungal biomass, suggesting bacteria will be a more predictable component of stream microbial communities. For 7 of the 9 streams, the standing stock of fine benthic organic matter was large enough that habitat-weighted reach-scale bacterial biomass was equal to or greater than fungal biomass. The quantities of leaves and small wood varied among streams such that the relative contribution of reach-scale fungal biomass ranged from 10% to as much as 90% of microbial biomass. Ergosterol concentrations were positively associated with substrate C:N ratio while bacterial abundance was negatively correlated with C:N. Both these relationships are confounded by particle size, i.e., leaves and wood had higher C:N than fine benthic organic matter. There was a weak positive relationship between bacterial abundance and streamwater soluble reactive phosphorus concentration, but no apparent pattern between either bacteria or fungi and streamwater dissolved inorganic nitrogen. The variation in microbial biomass per unit organic matter and the relative abundance of different types of organic matter contributed equally to driving differences in total microbial biomass at the reach scale.

A yellow-pigmented Gram-negative bacterium, Chryseobacterium indologenes, was found in the gut contents of about 65% of soft ticks Ornithodoros moubata from a perishing laboratory colony. The isolated putative pathogen, C. indologenes, was susceptible to cotrimoxazol and addition of this antibiotic (Biseptol 480) to the blood meal significantly decreased the tick mortality rate. The artificial infection of healthy O. moubata by membrane feeding on blood contaminated with C. indologenes was lethal to all ticks at concentrations 10(6) bacteria/ml. On the contrary, a similar infection dose applied to the hard tick Ixodes ricinus by capillary feeding did not cause significant mortality. Examination of guts dissected from infected O. moubata and I. ricinus revealed that C. indologenes was exponentially multiplied in the soft tick but were completely cleared from the gut of the hard ticks within 1 day. In both tick species, C. indologenes were found to penetrate from the gut into the hemocoel. The phagocytic activity of hemocytes from both tick species was tested by intrahaemocoelic microinjection of C. indologenes and evaluated by indirect fluorescent microscopy using antibodies raised against whole bacteria. Hemocytes from both tick species displayed significant phagocytic activity against C. indologenes. All O. moubata injected with C. indologenes died within 3 days, whereas the increase of the mortality rate of I. ricinus was insignificant. Our results indicate that hard ticks possess much more efficient defense system against infection with C. indologenes than the soft ticks. Thus, C. indologenes infection has the potential to be a relevant comparative model for the study of tick immune reactions to transmitted pathogens.

In this paper, we present an integrated account of the diurnal variation in the stable isotopes of water (δD and δ18O) and dry matter (δ15N, δ13C, and δ18O) in the long‐distance transport fluids (xylem sap and phloem sap), leaves, pod walls, and seeds of Lupinus angustifolius under field conditions in Western Australia. The δD and δ18O of leaf water showed a pronounced diurnal variation, ranging from early morning minima near 0‰ for both δD and δ18O to early afternoon maxima of 62 and 23‰, respectively. Xylem sap water showed no diurnal variation in isotopic composition and had mean values of ?13·2 and ?2·3‰ for δD and δ18O. Phloem sap water collected from pod tips was intermediate in isotopic composition between xylem sap and leaf water and exhibited only a moderate diurnal fluctuation. Isotopic compositions of pod wall and seed water were intermediate between those of phloem and xylem sap water. A model of average leaf water enrichment in the steady state (Craig & Gordon, pp. 9–130 in Proceedings of a Conference on Stable Isotopes in Oceanographic Studies and Palaeotemperatures, Lischi and Figli, Pisa, Italy, 1965; Dongmann et al., Radiation and Environmental Biophysics 11, 41–52, 1974; Farquhar & Lloyd, pp. 47–70 in Stable Isotopes and Plant Carbon–Water Relations, Academic Press, San Diego, CA, USA, 1993) agreed closely with observed leaf water enrichment in the morning and early afternoon, but poorly during the night. A modified model taking into account non‐steady‐state effects (Farquhar and Cernusak, unpublished) gave better predictions of observed leaf water enrichments over a full diurnal cycle. The δ15N, δ13C, and δ18O of dry matter varied appreciably among components. Dry matter δ15N was highest in xylem sap and lowest in leaves, whereas dry matter δ13C was lowest in leaves and highest in phloem sap and seeds, and dry matter δ18O was lowest in leaves and highest in pod walls. Phloem sap, leaf, and fruit dry matter δ18O varied diurnally, as did phloem sap dry matter δ13C. These results demonstrate the importance of considering the non‐steady‐state when modelling biological fractionation of stable isotopes in the natural environment.

Summary Immigration and colonization of isolates of naturally occurring stream bacteria and hyphomycetes on beech leaves were studied in a laboratory stream microcosm. Fungal spores were more successful immigrants, especially on new leaves, than bacteria, which were more repelled than attracted by the substrate. Fewer bacteria immigrated to older leaves than to new, and bacteria multiplied faster in water than on leaves. Fungi and bacteria showed synergistic relationships so that each group grew significantly faster in presence of the other group. If one considers, differences in immigration, colonization and synergism patterns, fungal mycelia doubled about 10 times faster than bacterial cells which might explain the dominance of fungi usually found on leaves in early decay. The individual fungal species could be assigned to one of three colonization groups; one of fugitive species, preceding a second group of species that grew from rarity to dominance, and a third group of very slow colonizers. The leachate was fractionated in different molecular size classes by gel chromatography, and the fraction around 2500D in the new leaf leachate was associated with a high concentration of polyphenols. High-pressure liquid chromatography (HPLC) and gas chromatography-mass spectrometry (GC-MS) revealed the presence of 16 phenolic acids in the new leaf leachate at concentrations ranging from < 1 to 640 g l–1. All fungi except the species with the slowest growth rate grew faster on leachate with the fraction around 2500D removed, and the density of bacteria was significantly reduced when pure stream water was supplemented with compounds from the same fraction.

New streams formed following ice recession in Glacier Bay National Park, southeastern Alaska allow insights into the role of abiotic and biotic interactions in the assemblage of benthic communities. Reductions in abundance of a pioneer chironomid colonizer, Diamesa alpina/lupus, in one new stream, Wolf Point Creek, is considered to be a result of competitive interactions with subsequent colonizers, as opposed to intolerance of warmer water temperature as previously suggested. Reduced densities of potential competitors (25–50 larvae per 500 cm2) in a cobble transplant experiment between streams, allowed persistence of D. alpina/lupus at low densities. In addition, significantly more D. alpina/lupus larvae emigrated from artificial stream channels containing other chironomid taxa than channels without potential competitors while there was no significant correlation of emigration with water temperature. A small number of D. alpina/lupus transplanted from a cold stream (4–6 °C) survived at water temperatures of 10–15 °C for 1 week. These results infer that interference competition is the causal mechanism in the decline of D. alpina/lupus. Complete exclusion of D. alpina/lupus from the stream has not occurred and water temperature may play a role in partitioning D. alpina/lupus from other taxa on a temporal or a spatial basis. This revised version was published online in August 2006 with corrections to the Cover Date.

1. To assess whether the reported slow breakdown of litter in tropical Cerrado streams is due to local environmental conditions or to the intrinsic leaf characteristics of local plant species, we compared the breakdown of leaves from Protium brasiliense, a riparian species of Cerrado (Brazilian savannah), in a local and a temperate stream. The experiment was carried out at the time of the highest litter fall in the two locations. An additional summer experiment was conducted in the temperate stream to provide for similar temperature conditions. 2. The breakdown rates (k) of P. brasiliense leaves in the tropical Cerrado stream ranged from 0.0001 to 0.0008 day−1 and are among the slowest reported. They were significantly (F = 20.12, P < 0.05) lower than in the temperate stream (0.0046–0.0055). The maximum ergosterol content in decomposing leaves in the tropical Cerrado stream was 106 μg g−1, (1.9% of leaf mass) measured by day 75, which was lower than in the temperate stream where maximum ergosterol content of 522 μg g−1 (9.5% of leaf mass) was achieved by day 30. The ATP content, as an indicator of total microbial biomass, was up to four times higher in the tropical Cerrado than in the temperate stream (194.0 versus 49.4 nmoles g−1). 3. Unlike in the temperate stream, leaves in the tropical Cerrado were not colonised by shredder invertebrates. However, in none of the experiments did leaves exposed (coarse mesh bags) and unexposed (fine mesh bags) to invertebrates differ in breakdown rates (F = 1.15, P > 0.05), indicating that invertebrates were unable to feed on decomposing P. brasiliense leaves. 4. We conclude that the slow breakdown of P. brasiliense leaves in the tropical Cerrado stream was because of the low nutrient content in the water, particularly nitrate (0.05 mgN L−1), which slows down fungal activity and to the low density of invertebrates capable of using these hard leaves as an energy source.

Outdoor artificial streams were treated continuously with pentachlorophenol (PCP) for 88 days during the summer of 1983. The contributions of different stream compartments (microbial habitats) to microbial degradation of PCP were determined in a stream treated with 144 micrograms of PCP per liter. The 488-m long stream was composed of mud-bottomed pools alternating with gravel riffles. PCP loss in the stream attributable to microbial degradation after an adaptation period was in the range of 55 to 74%. Contributions to PCP loss were determined for rock surface (epilithic), macrophyte surface (epiphytic), sedimentary, and water column communities by measuring rates of PCP disappearance in stream water, containing ambient concentrations of PCP, in contact with representative compartmental samples. The specific capability, in units of micrograms of PCP per hour per square meter of stream cross-sectional area (macrophytes at maximum plant density, water column at mean depth, upper 10-cm layer of gravel), followed the order rock surface much greater than macrophytes greater than sediment approximately equal to water column. The compartmental contribution to total stream losses in units of grams per hour followed the same order, although the differences were smaller. The rate of PCP disappearance in the water column above sediment cores followed the order oxygen-rich greater than oxygen-poor approximately equal to anaerobic greater than sorption-only conditions. The large difference in specific capability between the rock surface and sediment compartments could be attributed to oxygen deficiency (because of chemical and biological oxygen demand) in the sediments. Free-floating and particle-attached organisms in the water column were important to PCP biodegradation.
