Transition proteins and protamines are highly basic sperm-specific nuclear proteins that serve to compact the DNA during late spermiogenesis. To understand their sequential role in this function, transition protein 1 (TP1), transition protein 2 (TP2), and protamine 1 (P1) were assayed by polyacrylamide gel electrophoresis in pools of microdissected, staged seminiferous tubule segments in the rat. The results were compared with immunocytochemical analyses of squash preparations from accurately identified stages of the epithelial cycle. TP2 was the first to appear as a faint band at stages IX–XI, followed by high levels at stages XII–XIV of the cycle. TP1 showed a low expression at stage XII of the cycle and peaked at stages XIII–I, whereas protamine 1 first appeared at stage I of the cycle and remained high throughout the rest of spermiogenesis. Immunocytochemical analyses and Western blots largely confirmed these results: TP2 in steps 9–14, TP1 in steps 12–15, and P1 from late step 11 to step 19 of spermiogenesis. We propose that TP2 is the first nucleoprotein that replaces histones from the spermatid nucleus, and its appearance is associated with the onset of nuclear elongation. TP1 shows up along with the compaction of the chromatin. The two transition proteins seem to have distinct roles during transformation of the nuclei and compaction of spermatid DNA.  相似文献

The present study describes the morphological features of the eight stages of the seminiferous epithelium in Landrace boars according to the tubular morphology method, as well as their relative frequency, length, and duration. In Landrace boars the pre-meiotic stages occupied the 31.9 +/- 19.9% of the spermatogenic cycle and had a total length of 1788.8 +/- 1153.0 microm and a duration of 2.78 days; they were mainly characterised by the presence of leptotene and pachytene spermatocytes and round spermatids. Meiotic stages, with a relative frequency of 16.4 +/- 6.8%, a length of 787.1 +/- 603.1 microm and a duration of 1.41 days, contained spermatocytes in advanced meiosis I and/or in meiosis II and elongating spermatids grouped in bundles. Post-meiotic stages occupied the 50.6 +/- 20.4% of the spermatogenic cycle and had a length of 2096.8 +/- 1175.0 microm and a duration of 4.37 days; the most important event of these stages was the spermiation, which included the complete remodelling of sperm head and tail and the releasing of spermatozoa into the lumen, as well as the formation of residual bodies. From data obtained we concluded that both germ cell associations of the stages maintain constant among Landrace boars, and that the relative frequency, length and duration of the stages were directly dependent of the cytological transformations on the seminiferous tubules.  相似文献

Absolute and relative testis mass of two species of conilurine rodents, the plains rat (Pseudomys australis) and the hopping mouse (Notomys alexis), were markedly different. In plains rats the testis comprised approximately 3% of body mass while in hopping mice they rarely accounted for more than 0.2%. In both species, 8 cellular associations, or cycle stages, were recognized in the seminiferous epithelium. The relative frequencies, and hence durations, of Stages I to VIII were 5.1, 7.0, 12.5, 10.5, 5.4, 25.9, 4.4 and 29.0% of one cycle for plains rats (N = 14), and 20.4, 10.9, 9.6, 8.1, 5.1, 20.4, 14.7 and 10.8% for laboratory-bred hopping mice (N = 35). Generally, the relative durations of the cycle stages were very similar between plains rat individuals, but were variable between hopping mice. Also, organization of the seminiferous epithelium was less rigid in hopping mouse testes, and several anomalies were observed. These included: the occurrence of 2-4 cellular associations in about 20% (range 11.3-33.9%) of tubular cross-sections, deviations in cellular composition in certain cycle stages from that usually observed, and an increased incidence of degenerating cells, together with the presence of multinucleated "giant' cells, within the seminiferous epithelium.  相似文献

The paper describes in detail the cytomorphology of different types of germ cells, the 10 typical cellular associations or stages of the cycle of seminiferous epithelium (CSE), frequency of appearance of these stages, pattern of spermatogonial stem cell renewal and per cent degeneration of various germ cells in R. leschenaulti. Of the 14 steps of spermiogenesis (stained with PAS-haematoxylin) the first 10 were associated with the stages I-X, whereas, the remaining were found in association with one of the first six stages. The frequency of appearance of the various stages ranged from 3.84% (stage V) to 19.84% (stage I). These observations indicate that stage V is of shortest duration and stage I is of the longest duration in the bat. Five types of spermatogonia (A1, A2, A3, In and B) were identified based on their shape, size and nuclear morphology. Type A spermatogonia are oval with a large nucleus containing 1 or 2 nucleoli. The chromatin showed progressive condensation from A1 to A3 so that the latter appeared darkest among all the A type spermatogonia. The In type derived from A3 are smaller but appear darker than A3 due to heterochromatin crusts along the inner border of the nucleus. The B type spermatogonia derived from In are round and possess single nucleolus. The B type spermatogonia divided mitotically before entering meiosis or the actual production of the primary spermatocytes. A regular, well defined spermatogenic cycle was found in the Japanese quail by examining thin sections of isolated lengths of seminiferous tubules embedded in epoxy resin to resolve the structure of developing spermatids. The stages of the cycle initially were identified in studies using a preparatory method for fixation which separated adjacent cellular associations. The cycle was divided into 10 stages with relative frequencies (%) of Stages I to X respectively of: 11.9, 14.8, 24.1, 10.3, 8.2, 6.4, 9.4, 5.5, 3.8 and 5.4. The duration of one cycle was 2.69 +/- 0.08 days (mean +/- s.e.m.) as determined by intraventricular injection of [3H]thymidine and autoradiographic examination of the testes 1-4 days later. It was estimated that lifespans were 2.01 days for type B spermatogonia, 3.86 days for primary spermatocytes, 0.15 days for secondary spermatocytes, and 4.54 days for spermatids. The results suggest that the kinetics of spermatogenesis in the quail are fundamentally similar to the pattern in mammals.  相似文献

The purpose of this study was to determine whether species differences in neonatal vocalizations of rodent pups could be observed. Ultrasonic vocalizations of pups of 5 rodent species, mouse (ICR), vole (Microtus arvalis), Syrian hamster, rat (Wistar-Imamichi), and Mongolian gerbil were recorded from 3 to 15 or 21 days of age. Recordings were made under conditions of separation from mothers and litter mates in a cooled chamber (approximately 10 degrees C). The major species differences observed were age specific and species specific frequencies. The Mongolian gerbil displayed a different frequency change with age. Namely, the day on which ultrasonic vocalizations ceased was delayed in Mongolian gerbil compared with the other rodents. The modal peak frequencies of ultrasound emitted from pups at 3 days of age were low (around 35 kHz) in the vole and the Syrian hamster, medium (around 45 kHz) in the rat and the Mongolian gerbil, and high (around 55 kHz) in the mouse.  相似文献

The paper describes in detail the morphology and kinetics of germ cell associations, pattern of mitotic divisions, frequency distribution of different cellular associations (stages) and percent degeneration of various germ cells in the squirrel in which spermatogenesis in adults occurs all year round. Eighteen steps of spermiogenesis were identified based on the development of the acrosomal system using PAS-haematoxylin. These were appropriately divided into Golgi, acrosome, cap and maturation phases. Thirteen types of cellular associations or stages (I-XIII) were characterized along the length of the seminiferous tubule which repeated itself in space and time constituting the seminiferous epithelial cycle (CSE). Of the 18 steps of spermiogenesis, the first 13 were associated with stages I-XIII, respectively, and the rest with the first 9 stages. Spermiation occurred in stage IX. Seven types of spermatogonia [A0, A1, A2, A3, A4, intermediate (In) and B type] were identified based on their shape, size and nuclear morphology. A0 spermatogonia are pale in appearance with homogeneously distributed chromatin surrounded by a thin nuclear membrane. These are present in all stages. A1 are oval in shape and possess a thicker nuclear membrane. They are found in stages VI-X. The chromatin material undergoes progressive condensation from A1 to A4 making the last generation of spermatogonia appear darker. The In spermatogonia which are derived from A4 are morphologically similar to them but smaller in size. The B-type spermatogonia derived from the In types possess a typically round nucleus with uniformly condensed chromatin material underneath the nuclear membrane. The spermatogonia divide mitotically at fixed stages of the CSE giving rise to their next generations. Thus, A-type spermatogonia divide at stages X, XIII/I, IV and V, while In divide at stage VI. During each CSE of the squirrel, 5 peaks of mitosis occur. There is a single generation of B-type spermatogonia. These differentiate into primary spermatocytes and undergo meiosis or maturation divisions which enter to form spermatids. The A4 which divide differentially in stage VI give rise to In- and A1-type spermatogonia. Therefore, A4 spermatogonia form renewing stem cells. Based on the above pattern of spermatogonial mitosis a model for stem cell renewal in the squirrel is proposed. The percentage degeneration of germ cells varied with the cell type. During a single CSE of the squirrel, a total of 42.09% germ cells were found to degenerate. An attempt is made to compare and contrast the kinetics of spermatogenesis in the wild squirrel with that of the other rodents studied so far.  相似文献

The Turkish hamster ( Mesocricetus brandti ) has become a desirable species for experimentation in testicular function, photoperiod, reproductive hormones and hibernation. Basic data on the kinetics of the seminiferous epithelium have not yet been published. In the present study, the cycle of the seminiferous epithelium was divided into eight stages based on the overall cellular associations of 1540 cross sections of tubules. The mean relative frequencies for stages 1 through 8 were 5.9, 3.3, 11.7, 6.7, 7.2, 28.5, 21.6 and 15.1%, respectively. The absolute duration of the cycle of the seminiferous epithelium was determined by administration of 3H-thymidine, removal of testes at intevals after injection and autoradiography. The mean duration of one cycle was estimated at 8.0 days and the duration of stages 1 through 8 was 0.5, 0.3, 0.9, 0.5, 0.6, 2.3, 1.7 and 1.2 days, respectively. The duration of meiotic prophase was 11.5 days and of spermiogenesis was 13.8 days. The life span of preleptotene cell was estimated at 1.21 days, leptotene, 0.73 days, zygotene, 0.94 days and pachytene, 7.37 days. The total cycle length of spermatogenesis as usually calculated was 32.0 days.  相似文献

The stages of the cycle in the rat seminiferous epithelium are illustrated for testes fixed by vascular perfusion and embedded in plastic resins. Improved cellular resolution in plastic sections permitted a clearer demarcation of the stages than in paraffin. Quantitative data are presented to support the recognition of stages, particularly those in transition. Stages IV, V, VII, XI, and XII had the highest frequencies of transitional characteristics. Stage IV was redefined to be more consistent with the occurrence of a high percentage of mitotic figures and to clarify transitions in this stage. Although the resolution of cellular detail was greatly improved with the use of plastics, the thinner sections contained fewer identifying features together within a single tubule cross section and sometimes major characteristics were absent. Therefore, additional characteristics were used for stage classification, such as nuclear diameter and the presence or absence of mitotic figures. A binary decision key is provided to improve consistency among laboratories in the identification of the stages in plastic-embedded testes.  相似文献

Vertebrates are able to perceive the pitch of a series of harmonics, even when the fundamental frequency has been removed from the acoustic stimulus. Neural periodicity responses corresponding to the “missing fundamental” frequency of sonic stimuli have been observed in the auditory system of several animal species, including our own. This paper examines periodic cochlear neural responses of the gerbil. Periodicity responses to both sonic and ultrasonic stimuli originate within the cochlea of this animal. Acoustic stimuli, consisting of 2–12 successive harmonic frequencies, were used to generate an ensemble cochlear nerve periodicity response that was recorded from the round window of the cochlea. This response had a frequency equal to that of the missing fundamental, and not to those of the harmonic stimuli. Forward masking of the stimuli used to produce the periodicity response was used to generate sharp tuning curves, with tip frequencies corresponding to the harmonics and not to the periodicities. The sharpness of these functions increased as the frequencies of the harmonics increased, up to at least 38 kHz. Summary Spermiogenesis and cellular associations in the seminiferous epithelium of the guinea fowl were studied and described in sexually active adult birds. PAS stain was found to be useful in the recognition of steps of spermatid differentiation only in the first early stages. Nuclear morphological changes were subsequently found to be more reliable in tracing steps of spermiogenesis. It was observed that haematoxylin-eosin stained tissue can be used in the study of spermiogenesis in the bird. Various stages of the seminiferous epithelium were observed in any cross-section of the seminiferous tubules. Distinct cellular associations were observed, but intermix of adjacent germ cells or heterogenous cellular associations were frequently encountered.  相似文献

A sample of 27 males of Zoniopoda tarsata from Argentina was studied cytologically. The three largest autosomal pairs and the X were characterized by the presence of interstitial C-bands. Chiasma position relative to the bands was analyzed at diplotene and diakinesis. The frequency of interstitial, terminal and total chiasmata per cell was studied for the whole autosomal bivalent set, analysing the variations between stages and among individuals. The comparison of interstitial chiasma frequencies between stages and among individuals and the study of chiasma position relative to the bands in pairs 1, 2 and 3 indicated that chiasma distribution varied from diplotene to diakinesis. Therefore, terminalization does exist in this species and the movement may occur towards the centromere. The frequency of terminal associations at diplotene showed a high negative correlation (r=-0.89; p<10-5) with the number of interstitial chiasmata. This correlation would not be expected if the two kinds of association were produced by different (independent) mechanisms. Consequently, terminal associations were considered genuine chiasmata. The correlation between interstitial and total chiasmata was very much lower then the former (r=0.39; p=0.04). This fact, besides the relatively low variation for chiasma number, observed among individuals suggests that in this species the number of interestitial chiasmata, which are the most important in controlling the genetical recombination, is mainly regulated by changes in chiasma distribution, while variations in total chiasma frequency are of much lower magnitude.Member of Carrera del Investigador del Consejo Nacional de Investigaciones Cientificas y Técnicas (CONICET)  相似文献

The stages of the cycle of the seminiferous epithelium in the Japanese macaque are investigated using testes fixed by a mixture of formaldehyde and glutaraldehyde containing picric acid and embedded in a methacrylate resin, Quetol 523M. Sections, 1.0–2.0 μm in thickness, were cut with glass knives and stained with periodic acid-Schiff (PAS) and hematoxylin. Sections from such resin blocks illustrated cellular detail without structural distortion during the polymerization process. Furthermore, staining affinity with PAS and hematoxylin was excellent. In stained sections, typical germ cell associations were described, based on the nuclear morphology of type A (dark and pale) spermatogonium, type B spermatogonium, various developmental stages of primary spermatocytes during meiosis, and the development of the acrosomic system. In the Japanese macaque, two different steps of spermatids (steps 3 and 4) were constantly seen in the same area of the tubular epithelium during stage III. Therefore, a classification into ten stages is proposed for the cycle in this species. We know little about the process of spermatogenesis in bats, a great and diverse clade of mammals that presents different reproductive strategies. In the present study, spermatogenesis in six species of Neotropical bats was investigated by light microscopy. On the basis of chromatin condensation, nuclear morphology, relative position to the basal membrane and formation of the flagellum, three types of spermatogonia were recognized: dark type A (Ad), pale type A (Ap), and type B; the development of spermatids was divided into seven steps. With the exception of Myotis nigricans, the seminiferous epithelium cycle of the other five species studied was similar to those of other mammals, showing gradual stages by the tubular morphology method. Asynchrony was observed in the seminiferous epithelium cycle of M. nigricans, shown by overlapping stages and undefined cycles. The frequencies found in the three phases of the cycle were variable with the greatest frequency occurring in the postmeiotic phase (>50%) and the least in the meiotic phase (<10%). The similarities observed in the five species of Phyllostomidae appeared to be related to their phylogenetic relationship and shorter divergence times, whereas the differences in M. nigricans appeared to be related to its greater phylogenetic distance because the Vespertilionidae family diverged earlier. Shrub encroachment caused by overgrazing has led to dramatic changes of savannah landscapes and is considered one of the most threatening forms of rangeland degradation leading to habitat fragmentation. Although changes to plant assemblages are becoming better known, however, our understanding of how shrub encroachment affects rodent communities is low. In this study, we investigated relative abundance of five rodent species in sixteen southern Kalahari rangelands where shrub cover ranged from low (<5%) to high (>25%). Rodent abundance was determined on three trapping grids (40 × 100 m) for each site. Our results show that increasing shrub cover affected rodent species differently. The relative abundance of hairy‐footed gerbil, short‐tailed gerbil and bushveld gerbil declined with increasing shrub cover, whereas highveld gerbil and striped mouse exhibited hump‐shaped relationships with shrub cover. Overall, species richness decreased with increasing shrub cover and a negative impact of high shrub cover above 15% on rodent abundance was congruent for all species. We conclude that our results support the hypothesis that long‐term heavy grazing that results in area wide shrub encroachment, threatens the diversity of arid environments.  相似文献

The Mongolian gerbil (Meriones unguiculatus) is known to be one of the most radioresistant animals. We have examined the X-ray sensitivity of normal diploid fibroblasts from Mongolian gerbil embryos compared with those of cultured embryo cells obtained from various laboratory animals and a normal human. There was a wide difference in X-ray sensitivity for cell killing among different mammalian species. The D0 values for Mongolian gerbil cells ranged from 2.08 to 2.28 Gy, values which are twice as high as those for human cells. The mean D0 value for human cells was 1.06 Gy. Mouse, rat, Chinese hamster, and Syrian/golden hamster cells showed similar D0 values ranging from 1.30 to 1.56 Gy. When cells were irradiated with X rays, ten times more chromosome aberrations were detected in human cells than in Mongolian gerbil cells. The frequencies of chromosome aberrations in other rodent cells were between the values for cells from humans and those from gerbils. These data indicate that the Mongolian gerbil cells are resistant to X-ray-induced cell killing and chromosome aberrations, and that the radiation sensitivity of mammalian cells in primary culture may be reflected by their radioresistance in vivo.  相似文献

This study determined the optimum number of tubules to be counted per testis cross section, and the number of animals per treatment group, when changes in stage frequencies in the cycle of the seminiferous epithelium are criteria for assessing effects of treatment on spermatogenesis. A data base of 9,672 observed and staged tubules was collected from testicular cross sections of 15 Sprague-Dawley rats. A significant variation between animals was found for the frequencies of Stages I, II, IV, VI, VIII, and XIII. Computer simulation was used to randomly select different combinations of animal and tubule numbers from the observed data. Stage frequency means from each simulation experiment were compared statistically to observed mean frequencies. A model that used data from all 14 stages was analyzed. The following conclusions were made: a) a minimum of 200 tubule cross sections/testis is recommended for estimating stage frequencies; b) for a fixed number of tubules scored, the number of animals sampled is more important than the number of tubules per animal in reducing variance; c) to detect a difference of 2 standard deviations from the mean with a 2% error rate and examining 200 tubules/testis, at least 12 animals must be used per group when assessing all 14 stages; d) when individual stages are examined using 10 animals per group, only Stage VII has 80% or greater power of test (alpha = 0.05) to detect a frequency difference; e) pooling stages into 3-4 groups is recommended to improve the power of detecting a treatment difference.  相似文献

Chloroplast DNA (cpDNA) markers and 12 nuclear (random amplified polymorphic DNA, or RAPD) markers were used to examine the distribution of genetic variation among individuals and the genetic and ecological associations in a hybrid iris population. Plants in the population occurred at various distances from the edge of a bayou in a relatively undisturbed mixed hardwood forest and in an adjacent pasture dominated by herbaceous perennials with interspersed oak and cypress trees. The majority of plants sampled possessed combinations of markers from the different Iris species. Genetic markers diagnostic for Iris fulva and I. brevicaulis occurred at high frequencies, whereas markers diagnostic for I. hexagona were infrequent. For the majority of the nuclear markers, significant levels of cytonuclear disequilibria existed because of intraspecific associations among the markers in both the pasture and the forest. The distribution of nuclear markers among individuals was bimodal; intermediate genotypes were absent and the majority of RAPD markers were associated with their intraspecific cpDNA haplotypes. Strong intraspecific associations existed among RAPD markers in the forest, but associations tended to be weaker in the pasture area. Ecological correlations were detected for all but one of the I. fulva and I. brevicaulis RAPD markers. The ecological associations of hybrids similar to I. brevicaulis resembled associations of I. brevicaulis parental genotypes, suggesting that these hybrid genotypes may be relatively fit in the same habitats. The hybrids similar to I. fulva, however, were distributed in habitats that were unique relative to the parental species. The patterns of genetic and environmental associations along with other available data suggest that (1) only advanced generation hybrids were present in the population; (2) formation of F1 hybrids among Louisiana irises is rare, leading to sporadic formation of hybrid populations; and (3) selection and assortative mating have contributed to the formation of hybrid genotypes that tend to be similar to parental genotypes. The patterns of ecological and genetic associations detected in this population suggest that assortative mating and environmental and viability selection are important in the structuring and maintenance of this hybrid zone.  相似文献
