Cadmium uptake by maize from polluted river sediments covered with a clean top layer of variable thickness is discussed in relation to root distribution. Two pathways for uptake are distinguished: roots penetrating the contaminated layer or contaminants moving into the root zone. Relative Cd uptake proved to be roughly proportional to the fraction of total root length found in the contaminated layer. A deeper water table induced a deeper root development and more Cd uptake for a given thickness of clean topsoil. A model based on exponential decrease of root length density with depth is acceptable as first approximation only. Little or no evidence was found for contaminants moving into the root zone during the ten years of the experiment.  相似文献

城市河道受污染底泥对水体产生大的二次污染,为有效的控制底泥污染物的释放和开发有效的水体治理技术方法,本研究利用人工模拟河道在连续流条件下,对河道污染底泥的生物修复及其对水体生物修复的影响进行了试验研究。研究结果表明:采用曝气增氧和投加底泥生物修复制剂联合处理的方式能达到好的底泥生物修复效果,能有效的提高底泥的生物降解活性(G值),削减底泥氮磷污染物的释放量,底泥G值与底泥氮、磷污染物释放量呈显著负相关性:单独采用曝气增氧和投加土著微生物制剂联合实施的生物修复方法对河道污染水体均具有良好的修复效果,但底泥的生物修复将对河道水体生物修复的效果产生大的影响。底泥生物修复后,对水体进行生物修复的处理效果更加显著,在相同实验条件下,对水体COD、氨氮的最大去除率由65.0%和16.30%提高到72.0%和41.0%,水体生物修复的周期由13 d缩短为6 d。  相似文献

Plant roots penetrate into the subsoil from which they may take up heavy metals, resulting in accumulation in the plant. The subsoil may contain heavy metals, possibly as a result of ploughing down contaminated topsoil. This is why the effect of Cd in the subsoil on Cd plant uptake was studied in outdoor miniplots in eight consecutive years. Cd (as Cd(NO3)2.4H2O) was added to four different layers of two soils (a sand and a sandy loam) at the start of the experiment. Nine arable and vegetable crops were grown and analysed for Cd. For most investigated crops Cd uptake increased linearly with increasing depth of Cd addition to the soils and for some crops (spinach and potato) the increase was exponential (decreasing increase). Cd uptake by crops on sand was higher than on sandy loam. As most investigated crops accumulated Cd from the subsoil, ploughing down a Cd-contaminated topsoil into the subsoil is a risk for the safe production of food and feed crops. It is concluded that acceptable Cd concentration levels in soils should not only be based an the Cd concentration in the topsoil but also on that in the rooted subsoil.  相似文献

DNA extraction from soil samples is a critical step for molecular biology analyses. The present study compared the efficiency of two DNA isolation methods from non-polluted and polluted soils with or without the presence of a plant. Both applied methods used chemical and physical lyses, but method 1 had an additional physical disruption. The main difference between these two methods was the humic acid purification technique as it was carried out during cell lysis for method 1 and after cell lysis for method 2. Samples were assessed on the basis of their yield and DNA purity as well as their bacterial quantity and diversity. Based on our results, method 1 proved to be more effective at removing protein and RNA, whereas method 2 proved to be more effective at removing humic acids. Although no differences were obtained in terms of the DNA yield, both the bacterial quantity and community structure were affected by the method used. Method 1 allowed for the recovery of more information than method 2, and polluted soil was more sensitive to the DNA extraction procedure. We recommend carefully selecting the DNA extraction method, especially when soil is disturbed.  相似文献

Summary The chance for exposure of man to Pt has increased because of the loss of Pt by the catalytic converter used to reduce levels of harmful auto emissions. Whether Pt can be taken up by plants and introduced into man's primary food chain was studied. Pt uptake was analyzed by plasma emission spectroscopy from a week's exposure to 0.057, 0.57, or 5.7 ppm Pt inHoaglands solution. Roots and shoots of nine horticulturally important crops were tested. Chlorosis of young tissue was usually evidenced and dry weights of five species were significantly reduced by 5.7 ppm Pt. Several species showed increases in dry weight at the lowest concentration of Pt. All species accumulated Pt in significantly high amounts in the roots at all concentrations of Pt (for cauliflower and tomato > 1000 ppm at 5.7 ppm Pt). Only pepper, cauliflower, and radish accumulated Pt in their tops. Water loss was increased at low Pt treatment levels and decreased at high levels. The results indicate that Pt can enter into man's food chain directly and probably in large amounts from root crops. Floating beds of canna can remove nitrogen from polluted river water, but the removal efficiency is not very high in a short time, so some enhanced methods must be used to improve nitrogen removal efficiency by the system. Immobilized denitrifying bacteria and aeration were added into the canna floating bed. The experimental results showed that these enhancements substantially improved the nitrogen removal efficiency of the floating beds. With the enhancements, total nitrogen removal was 72.1%, ammonia nitrogen oxidation was 100%, nitrate nitrogen removal was 75.8%, nitrite nitrogen removal was 95.9%, and COD removal was 94.6% in 5 days. Without the enhancements, the canna floating bed system removed only 50.4% of the total nitrogen, 22.4% of the nitrate nitrogen, 5.3% of the nitrite nitrogen and 39.9% of the COD, respectively, in 5 days, but did accomplish 100% oxidation of the ammonia nitrogen. The experimental results provide an improved method for the treatment of polluted river water.  相似文献

Colonization of overlaying water by bacteria from dry river sediments   总被引:1,自引:0,他引:1  
We studied the diversity, community composition and activity of the primary microbial colonizers of the water above freshly re-wetted sediments from a temporary river. Dried sediments, collected from Mulargia River (Sardinia, Italy), were covered with sterile freshwater in triplicate microcosms, and changes of the planktonic microbial assemblage were monitored over a 48 h period. During the first 9 h bacterial abundance was low (1.5 x 10(4) cells ml(-1)); it increased to 3.4 x 10(6) cells ml(-1) after 28 h and did not change thereafter. Approximately 20% of bacteria exhibited DNA de novo synthesis already after 9 h of incubation. Changes of the ratios of (3)H-leucine to (3)H-thymidine incorporation rates indicated a shift of growth patterns during the experiment. Extracellular enzyme activity showed a maximum at 48 h with aminopeptidase activity (430.8 +/- 22.6 nmol MCA l(-1) h(-1)) significantly higher than alkaline phosphatase (98.6 +/- 4.3 nmol MUF l(-1) h(-1)). The primary microbial colonizers of the overlaying water - as determined by 16S rRNA gene sequence analysis - were related to at least six different phylogenetic lineages of Bacilli and to Alphaproteobacteria (Brevundimonas spp. and Caulobacter spp.). Large bacterial cells affiliated to one clade of Bacillus sp. were rare in the dried sediments, but constituted the majority of the planktonic microbial assemblage and of cells with detectable DNA-synthesis until 28 h after re-wetting. Their community contribution decreased in parallel with a rise of flagellated and ciliated protists. Estimates based on cell production rates suggested that the rapidly enriched Bacillus sp. suffered disproportionally high loss rates from selective predation, thus favouring the establishment of a more heterogenic assemblage of microbes (consisting of Proteobacteria, Actinobacteria and Cytophaga-Flavobacteria). Our results suggest that the primary microbial colonizers of the water above dried sediments are passively released into the plankton and that their high growth potential is counteracted by the activity of bacterivorous protists.  相似文献

Aim:  The goal of this study was to identify functional targets to detect polycyclic aromatic hydrocarbon (PAH)-degrading bacterial populations in cold marine ecosystems.
Methods and Results:  We designed a degenerate primer set targeting genes encoding the α subunit of PAH-dioxygenases from Gram-positive bacteria. This primer set was used to amplify gene fragments from metagenomic DNA isolated from Subantarctic marine sediments (Ushuaia Bay, Argentina). These gene fragments were cloned and sequenced. We identified 14 distinct groups of genes, most of them showing significant relatedness with dioxygenases from Gram-positive bacteria of the genera Rhodococcus , Mycobacterium , Nocardioides , Terrabacter and Bacillus . The level of identity with these genes, however, was low to moderate (33–62% at the amino acid level).
Conclusion:  These results indicate the presence of a high diversity of hitherto unidentified dioxygenase genes in this cold polluted environment.
Significance and Impact of the Study:  Subantarctic marine ecosystems are particularly vulnerable to hydrocarbon pollution, and the development of environmental restoration strategies for these environments is pressing. The information obtained in this work will be the starting point for the design of quantitative molecular tools to analyse the abundance and dynamics of these aromatic hydrocarbon-degrading bacterial populations in the marine environment.  相似文献

Summary Rape, cucumber, wheat, oats and tomato were grown for one to two weeks in nutrient solutions with heavy metals added. Of the metal ions tested (Cr3+, Cu2+, Co2+, CrO4 2-, Ni2+, Cd2+, Pb2+, Mn2+, Zn2+ and Ag+), manganese, nickel and lead exhibited the greatest mobility in cucumber plants, which resulted in the highest shoot/root concentration ratio. Silver was not translocated to the shoots of cucumber plants in measurable amounts.When the plants were grown with 1.0, 10 and 100 M cadmium or nickel in the solution, the shoot and root concentration increased 5–10 times if the metal ion concentration of the solution was increased 10 times.The plants showed great differences in cadmium and nickel uptake. In the shoot, the cadmium concentration increased in the order: oats = wheat < cucumber = rape < tomato, and in the root in the order: oats = wheat < cucumber = rape < tomato. The great uptake of cadmium and nickel by tomato is notable and agrees with other reports.The nickel, and especially the cadmium, concentration in roots and shoots increases with the age of the plant.The results are discussed and related to other investigations. Denitrification by the sessile microbial community of the River Tamagawa was studied in laboratory experiments. Inorganic nitrogen loss was observed when river water was incubated with sessile microbial community of the river in a continuously circulating system. It was confirmed by the 15N tracer technique that both sessile microbial communities of unpolluted and polluted areas had denitrifying activity, even though they were incubated in oxygenated river water. The denitrification rate of the sessile microbial community taken from a polluted area, measured by the 15N tracer technique, was 8–16 mg N/m2/day in October and December, 1977, and it was enhanced 10-fold by raising the water temperature from 14 to 30° C. Denitrification in the river was also suggested by determining the N2: Ar ratio of gases evolved from the river bed.  相似文献

Hexavalent chromium [Cr(VI)] is a known carcinogen and mutagen; however, the actual mechanisms of Cr toxicity are unknown. Two approaches were used to isolate Cr(VI)-resistant bacteria from metal-contaminated river sediments. Diluted sediments were plated directly onto a peptone-yeast extract (PYE) medium containing 0 to 100 micrograms of Cr(VI) ml-1. Approximately 8.4 x 10(5) CFU g-1 were recovered on 0 microgram of Cr(VI) ml-1, whereas 4.0 x 10(2) CFU g-1 were recovered on PYE plus 100 micrograms of Cr(VI) ml-1. Alternatively, continuous culture enrichment techniques were employed using PYE and 100 micrograms Cr(VI) ml-1 input at dilution rates of 0.02 and 0.10 h-1. After six residence periods, 10(9) CFU were recovered on PYE agar containing 0 microgram of Cr(VI) ml-1 and 10(7) CFU on PYE agar plus 100 micrograms of Cr(VI) ml-1. Of 89 isolates obtained by direct plating onto PYE, 47% were resistant to 100 micrograms of Cr(VI) ml-1, and 29% were resistant to 250 micrograms of Cr(VI) ml-1. When the same isolates were plated onto PYE containing Cr(III), 88% were resistant to 100 micrograms ml-1 but only 2% were resistant to 250 micrograms ml-1. Cr, Co, Sb, and Zn were found in significantly higher concentrations at an industry-related contaminated site than at a site 11 km downstream. Total Cr in the sediments at the contaminated site averaged 586 micrograms (dry weight) g-1, and the downstream site averaged 71 micrograms (dry weight) g-1. The Cr recovered from acid-digested Ottawa River sediment samples was predominantly hexavalent. Five acid digestion procedures followed by atomic absorption spectroscopy were compared and found to be 30 to 70% efficient for recovery of Cr relative to neutron activation analysis. Strains 19SMN4T and ST27MN3 were isolated from marine sediments after enrichment with 2-methylnaphthalene and were classified as Pseudomonas stutzeri genomovar 4. Four other strains, BG 2, HT20, HT24, and A7, were isolated from sulphide-oxidizing bioreactors or activated sludge affiliated with the same clade in the 16S rRNA phylogenetic tree. P. stutzeri has been recently reclassified as a new genus, Stutzerimonas, and a preliminary analysis indicated that the strains in this study were distinct from any classified Stutzerimonas and are considered representatives of phylogenomic species 4 (pgs4). Strains 19SMN4T and ST27MN3 were extensively characterized with phenotypic, chemotaxonomic, genomic and phylogenomic data. Strain 19SMN4T had a well-characterized naphthalene degradative plasmid that has been compared with other plasmids, while in strain ST27MN3, the naphthalene degradative genes were detected in the chromosome sequence. Phylogenomic analysis of the core gene sequences showed that strains 19SMN4T and ST27MN3 shared 3,995 genes and were closely related to members of the species “Stutzerimonas songnenensis” and Stutzerimonas perfectomarina, as well as to the Stutzerimonas phylogenomic species, pgs9, pgs16 and pgs24. The aggregate average nucleotide identity (ANI) indicated that strains 19SMN4T and ST27MN3 belonged to the same genomic species, whereas the genomic indices with their closest-related type strains were below the accepted species threshold (95 %). We therefore conclude that strains 19SMN4T and ST27MN3 represent a novel species of Stutzerimonas, for which the name Stutzerimonas decontaminans is proposed; the type strain is 19SMN4T (=CCUG44593T = DSM6084T = LMG18521T).  相似文献

An indigenous strain of Pseudomonas putida capable of degrading 3-chlorobenzoic acid as the sole carbon source was isolated from the Riachuelo, a polluted river in Buenos Aires. Aerobic biodegradation assays were performed using a 2-l microfermentor. Biodegradation was evaluated by spectrophotometry, chloride release, gas chromatography and microbial growth. Detoxification was evaluated by using Vibrio fischeri, Pseudokirchneriella subcapitata and Lactuca sativa as test organisms. The indigenous bacterial strain degrades 100 mg l−1 3-chlorobenzoic acid in 14 h with a removal efficiency of 92.0 and 86.1% expressed as compound and chemical oxygen demand removal, respectively. The strain was capable of degrading up to 1,000 mg of the compound l−1. Toxicity was not detected at the end of the biodegradation process. Besides initial concentration, the effect of different factors, such as initial pH, initial inoculum, adaptation to the compound and presence of other substrates and toxic related compounds, was studied.  相似文献

Spring wheat was grown in the field under deficient and sufficient levels of soil K and with high and low supplies of fertiliser nitrogen. Measurements were made of K uptake, soil nutrient supply parameters, root growth and, in solution culture, root influx parameters. Mechanistic models predicted uptake reasonably well under K-deficient conditions, but over-predicted uptake, by as much as 4 times, under K-sufficient conditions. The over-prediction was apparently due to poor characterisation of plant demand.  相似文献

The contribution of erosion of bed sediment to the load of metals leaving Lake Ketelmeer, a shallow lake in the Netherlands fed by the IJssel branch of the River Rhine, is reported. Transport of suspended matter and associated trace metals was measured using both centrifuges and sediment traps at several locations in the lake. Mass balances of suspended matter and heavy metals were calculated using data from these field measurements.Metal/scandium-ratios were used to identify the source of the suspended matter in the lake. Since the bed sediment is more polluted than the suspended sediment entering the lake, higher metal/scandium-ratios were found for bottom sediment in the lake compared with those for suspended matter entering the lake from the River IJssel. Using the metal/scadium-ratio in suspended matter from the lake, it was calculated that bottom sediments made up 43% of the suspended matter leaving the lake. This implies an erosion flux of bottom sediment of 16 g m–2 d–1. For cadmium, mercury, chromium and zinc, this erosion process accounts for more than 50% of the pollutant load leaving Lake Ketelmeer and entering Lake IJsselmeer.  相似文献

红树林真菌由于其独特的生态特征、多样性特点和丰富的新型生物活性代谢产物而引起了广泛的关注。本文以漳江口红树林保护区4个采样点的沉积物为研究对象,采用8种筛选培养基(RBM、PDA、CDA、Martin、YM、SDA、ISP2和R2A)分离可培养真菌,根据ITS序列分析对其进行物种鉴定,并利用多样性指数分析评价不同采样点真菌群落的差异。结果共分离到274株真菌,隶属于2门12纲23目39科52属,其中优势属为枝顶孢属(Acremonium)(20.8%)和青霉属(Penicillium)(11.3%),其次是枝孢属(Cladosporium)(7.3%)和帚枝霉属(Sarocladium)(7.3%)。8种筛选培养基中,分离出真菌种属类型最丰富的培养基是RBM,其次是PDA。根据Shannon-Wiener多样性指数(H′)、Simpson优势度指数(D)、Magalef丰富度指数(R)和Pielou均匀度指数(J)的分析结果,在属的水平上,距海洋的位置越近,真菌的群落多样性越高,真菌分布越均匀。  相似文献

1. The Ogeechee River, in south-eastern Georgia, U.S.A, is a blackwater river with an extensive floodplain that is inundated regularly during winter months. Previous studies have shown that low to moderate bacterial production rates cannot support the relatively high suspended bacteria concentrations observed (107?-108 cells ml?1), suggesting an allochthonous source of bacteria. We report the results of a combination of field and flume experiments which demonstrate that river sediments and floodplain soils are significant sources of suspended bacteria during seasonal flooding. Benthic bacteria are also entrained by normal discharges. There are sizeable fluxes of POC and DOC from river sediments and floodplain soils. 2. Bacterial, POC and DOC fluxes (14, 250, and 790 mg Cm?2 h?1, respectively) were substantial when water was percolated upward through floodplain soils. 3. Simulation of overland flow using a flume demonstrated further fluxes of bacteria and POC from floodplain soils (up to 61 and 10700 mg Cm?2h?1, respectively) and river sediments, but did not yield additional DOC from floodplain soils. 4. These laboratory results are supported by experiments in which we measured significant increases in concentrations of bacteria and DOC in a downstream direction in (i) the main river channel during a winter flood in 1986, and (ii) a floodplain slough (channel side-arm) which re-entered the main channel 800m from its initial divergence. Inputs of bacteria and DOC from the surrounding floodplain were estimated to be up to 3500 kg DOC h?1, and 4000 kg bacteria Ch?1 over a 50-km reach. 5. These previously unmeasured fluxes of organic carbon help to explain the high concentrations of suspended bacteria in the Ogeechee River.  相似文献
