猴头菌(Hericium erinaceus)多糖是猴头菌发挥药理作用的主要活性成分,猴头菌生长过程中多糖的合成代谢及其结构的变化目前还鲜有研究报道。该试验研究了猴头菌生长发育过程中多糖的产生及其结构和活性的变化。对猴头菌0605菌株生长发育6个时期的子实体进行了多糖和β-葡聚糖含量的测定,并对提取的粗多糖进行了体外免疫活性的研究。结果表明:在猴头菌的整个生长发育过程中,子实体多糖的含量呈现上升的趋势,成熟期含量最高,达1.54%,分裂期含量最低仅0.89%;β-葡聚糖含量整体上呈上升的趋势,中菌刺期含量最高33.58%,成熟期最高,达到50.67%;体外免疫活性试验表明,现蕾期、分裂期、中菌刺期和成熟期的多糖均能很好地刺激巨噬细胞RAW264.7产生NO,活性均优于无菌刺期和小菌刺期的多糖。分别采用体积分数为30%、50%和70%的乙醇对子实体发育后3个时期的热水浸提物进行分级醇沉,对分级分离多糖进行理化性质研究。结果表明:小菌刺期30%醇沉的粗多糖得率高于中菌刺期和成熟期,其大分子量多糖的比例也较高;3个时期50%和70%醇沉多糖分子量相差不大;分级分离多糖的单糖种类基本相同,都含有岩藻糖、半乳糖、葡萄糖和甘露糖,但摩尔比有差异;3个时期分级分离多糖均能刺激巨噬细胞分泌NO,30%醇沉多糖比50%和70%醇沉多糖的活性好;成熟期多糖的活性优于其他生长阶段。对小菌刺期、中菌刺期和成熟期50%醇沉多糖(H5FP4B、H5FP5B、H5FP6B)进行Sephacryl S300凝胶层析纯化,共得到9个组分:H5FP4B-1、H5FP5B-1、H5FP6B-1、H5FP4B-2、H5FP5B-2、H5FP6B-2、H5FP4B-3、H5FP5B-3、H5FP6B-3。对这9个组分的分子量分布进行研究,发现每个时期的主峰(H5FP4B-3、H5FP5B-3和H5FP6B-3)分子量基本相同,分子量分布差异较大的是3个时期的第1组分,在体外免疫活性上也表现出较大差异。主峰H5FP4B-3的分子量为1.59万,由岩藻糖、半乳糖和葡萄糖组成,摩尔比为5.2∶23.9∶1。经红外光谱、甲基化和核磁共振综合分析,解析出H5FP4B-3均一多糖的结构单元是以α-D-1,6连接的半乳糖和α-D-1,2,6连接的半乳糖构成主链,侧链为α构型的端基岩藻糖。研究结果表明:猴头菌不同发育阶段产生的多糖结构和活性有一定的差异,生长初期,大分子量的多糖较多,后期分子量分布差异变小并趋稳定。多糖主成分变化不大,较稳定;到成熟期,多糖的活性更高。  相似文献

The purpose of this study was to evaluate the change of surface roughness and the development of the film during the film coating process using laser profilometer roughness measurements, SEM imaging, and energy dispersive X-ray (EDX) analysis. Surface roughness and texture changes developing during the process of film coating tablets were studied by noncontact laser profilometry and scanning electron microscopy (SEM). An EDX analysis was used to monitor the magnesium stearate and titanium dioxide of the tablets. The tablet cores were film coated with aqueous hydroxypropyl methylcellulose, and the film coating was performed using an instrumented pilot-scale side-vented drum coater. The SEM images of the film-coated tablets showed that within the first 30 minutes, the surface of the tablet cores was completely covered with a thin film. The magnesium signal that was monitored by SEM-EDX disappeared after ∼15 to 30 minutes, indicating that the tablet surface was homogeneously covered with film coating. The surface roughness started to increase from the beginning of the coating process, and the increase in the roughness broke off after 30 minutes of spraying. The results clearly showed that the surface roughness of the tablets increased until the film coating covered the whole surface area of the tablets, corresponding to a coating time period of 15 to 30 minutes (from the beginning of the spraying phase). Thereafter, the film only became thicker. The methods used in this study were applicable in the visualization of the changes caused by the film coating on the tablet surfaces.  相似文献

Pteris vittata, a fern able to hyperaccumulate arsenic (As) in its fronds, has been object of a number of studies aimed to understand the mechanisms involved in As absorption and tolerance. This study has focused on a new mechanism, As leaching, already observed in P. vittata, but not explained, based on the possible involvement of hydathodes, not yet described in this fern; moreover, the results contained in this article will provide information on a more detailed frond anatomy of P. vittata. A combination of light and electron microscopic techniques (transmission electron microscopy and environmental scanning electron microscopy with energy dispersive X-ray) and chemical analyses (inductively coupled plasma atomic emission spectroscopy) was used. The results suggest that in phytoremediation processes under field conditions there could be the need to know the atmospheric conditions before harvesting plants, to avoid the loss of As in the environment.  相似文献

Cell surface changes during mitosis and cytokinesis of epithelial cells   总被引:4,自引:0,他引:4  
Summary PtK2 cells were studied with scanning electron microscopy to record changes on the cell surface during mitosis and cytokinesis. During prophase, prometaphase and metaphase, the cells remain very flat with few microvilli on their surfaces. In anaphase cells, there is a marked increase in the number of microvilli, most of which are clumped over the separating chromosomes and polar regions of the mitotic spindle leaving the surface of the interzonal spindle region relatively smooth. Microvilli appear over the interzonal spindle region in telophase and the cells also increase in height. At the beginning of cleavage, the distribution of microvilli is roughly uniform over the surface but it becomes asymmetric at the completion of cleav-age when the daughter cells begin to spread. At this time most microvilli are over the daughter nuclei and the surfaces that border the former cleavage furrow. The regions of the daughter cells distal to the furrow are the first to spread and their surfaces have very few microvilli. When chromosome movement is inhibited by either Nocodazole or Taxol, microvilli formation is inhibited on the arrested cells. Nevertheless cell rounding still takes place in the normal time period. It is concluded from these observations that the signal for the onset of chromosome movement in anaphase is accompanied by a signal for the formation of microvilli. It is suggested that there is also a separate signal for the cell-rounding event in mitosis and that microvilli do not play a role in this contractile process.  相似文献

In the lactating breast, the development of secretory alveoli consisting of differentiated cells arranged around a central lumen is dependent on signals from the extracellular environment of the cells. There are few cell lines that model this process. We previously showed that the human breast carcinoma line PMC42-LA can be induced to form organoids, reminiscent of secretory alveoli found in the lactating human breast. In this report, we used high-resolution scanning electron microscopy to show that the formation of organoids is accompanied by development of cell surface microvilli. Extracellular matrix-induced formation of microvilli occurred on the internal and external surfaces of cells in the organoids and not on surfaces in contact with the extracellular matrix. Organoid formation of PMC42-LA cells induced a rearrangement of the extracellular matrix, seen in the form of radiating fibers from the organoids. In summary, there is an interaction between PMC42-LA cells and the underlying extracellular matrix, which leads to the formation of polarized cells with well-developed microvilli. This is accompanied by organization of the extracellular matrix. PMC42-LA is a relevant model of the human breast for investigations into cell-cell and cell-matrix interactions.  相似文献

利用扫描电镜技术,对4种补血草属植物大叶补血草(Limonium gmelinii (Willd.) Kuntze)、耳叶补血草(Limoniu otolepis (Schrenk) Kuntze)、繁枝补血草(Limonium myrianthum (Schrenk) Kuntze)和簇枝补血草(Limonium auream)的叶表面进行了研究,以探讨植物微形态结构与生态环境的关系。结果表明,这4种植物的上下表皮都分布有盐腺,且上表皮的盐腺密度大于下表皮,盐腺的基本结构相同,多由20个细胞构成,其中4个分泌细胞顶端的角质层中央有一小孔,植物体靠盐腺的泌盐孔泌盐。但4种植物在盐腺的密度、盐腺的大小、盐腺与周围表皮细胞的位置等方面存在差异。植物在对盐碱生境的长期适应过程中,促使其强烈地分化出泌盐结构,因而,具有明显的生态适应性。  相似文献

Summary

Surface architecture and selected ultrastructural features of normal embryos of the marine gastropod Ilyanassa obsoleta have been investigated using scanning and transmission electron microscopy. A re-examination of the classic account of Anthony Clement on temporal and division patterns in normal and lobeless embryos confirms most of his observations; a few differences, involving the size and positioning of micromere 4d and its derivatives, are noted. Several hypotheses are presented to account for zonular distribution of longer microvilli on some blastomere surfaces and progressive diminution of microvilli on micromere surfaces, especially the leading edge micromeres (LEMs), during postulated epibolic spreading movements. Microfilaments, discovered in the cortical regions of the LEMs, are described, and a role for these structures in gastrulation is proposed.  相似文献

, , , and 1988. Schistosoma japonicum (Chinese): changes of the tegument surface in cercariae, schistosomula and juvenile parasites during development. International Journal for Parasitology18: 1093–1104. The surface of cercariae of S. japonicum (Chinese) and changes following their transformation into schistosomula were observed at 0.5,1, 3,6,12,24 and 48 hand 5,7,10 and 15 days by scanning electron microscopy. The cercarial surface shows two distinct regions: on the head and the body it is mildly undulating and covered throughout with numerous spines that are partially embedded in a thick layer of glycocalyx; on the tail it is highly folded into tall and thin circumferential ridges, spines are confined to the dorsal and ventral aspects and are prominent due to a thinner glycocalyx. Between 0.5 and 12 h following skin penetration, the surface of schistosomula is gradually cleared of glycocalyx, while small blebs and slender microvilli project out from the glycocalyx-free area and from spines. Subsequently, these structures are detached from the surface and thus responsible for the shedding of the original cercarial membrane and glycocalyx. Between 24 and 48 h, schistosomula show little change in the body size in comparison to earlier stages, and most of the surface is still covered with spines but devoid of blebs and microvilli. From day 2 onwards schistosomula are lengthened but with little net growth. Spines decrease in the middle region of the body; and the surface is invaginated by small pits and folded into thin ridges, which result in rapid increase of the surface area. Between days 5 and 7 schistosomula elongate further, and the disappearance of the spines in the middle region becomes even more pronounced, while those remaining are confined only to the anterior and posterior extremities. Ridges and pits are more developed, and the former begin to show regular foldings throughout all parts of the body. Between days 7 and 10 schistosomula grow rapidly and the body is quadrupled in size. Between days 10 and 15 sexes can be differentiated. The male has a flat ovoid shape body, whose ridges and pitting on the surface are highly developed almost to the same extent as in adult parasites, while spines remain mostly on the posterior end. Sensory papillae greatly increase in number, especially on the ventral and lateral aspects and around the suckers. The surface of the female is relatively smooth; ridges are developed only on the posterior part, while the remaining surface is flat and extensively pitted. BACKGROUND AND AIMS: The generic delimitations of Ficinia and Isolepis, sister genera in the Cypereae, are blurred. Typical Ficinia flowers have a lobed gynophore, which envelops the base of the nutlet, whereas in Isolepis the character is considered to be absent. Some former species of Isolepis, lacking the gynophore, were recently included in Ficinia. The floral ontogeny of representative taxa in Ficinia and Isolepis were investigated with the aim of evaluating the origin and nature of the gynophore in the Cypereae. METHODS: The spikelet and floral ontogeny in inflorescences collected in the field was investigated using scanning electron microscopy (SEM) and light microscopy (LM). KEY RESULTS: SEM images of Isolepis setacea and I. antarctica, Ficinia brevifolia, F. minutiflora, F. zeyheri and F. gracilis, and LM sections of F. radiata, show that the gynoecium in Ficinia is elevated above the flower receptacle by the development of a hypogynous stalk. From its apex, a (often three-)lobed cup is formed, which envelopes the basal part of the later nutlet. In developing flowers of I. antarctica, a rudimentary hypogynous stalk appears. In I. setacea, rudiments of a hypogynous stalk can be observed at maturity. In F. radiata and F. zeyheri, intralocular hairs are present in the micropylar zone. At the surface of developing gynoecia in flowers of F. gracilis, star-shaped cuticular structures appear which disappear again at maturity. CONCLUSIONS: The overall floral ontogeny of all species studied occurs following a typical scirpoid pattern, though no perianth primordia are formed. The gynophore in Ficinia originates as a hypogynous stalk, from which the typical gynophore lobes develop. The gynophore is not homologous with the perianth.  相似文献

BackgroundThe threat of cadmium (Cd), which is the cause of itai-itai disease in Japan, is still complicated and confusing, especially for digestive system, such as liver disease. One of the most keys of this problem is demonstrating that the hepatotoxicity is indeed induced by Cd. Therefore, we attempt detecting Cd at microscale during ultrastructural imaging of liver tissue.Methods12 rats were divided randomly into two experimental groups: control and Cd-treated. Treated rats were intraperitoneal injected with 1 mg/kg body weight cadmium chloride (CdCl2) for 4 weeks (5 P.M each day for 6 days/week). At the end of the exposure period, liver tissue samples were processed into ultrathin sections for analysis of advanced analytical transmission electron microscopy and X-ray energy dispersive spectroscopy (TEM/X-EDS) investigations. Ultrastructural images and X-ray energy dispersive spectrum were acquired at microscale.ResultsCd can cause changes in the structure of the organelle, including the collapse of the membrane structure in the cell, the destruction of the internal structure of the organelle, the mitochondrial swelling, the expansion of the endoplasmic reticulum, and the appearance of inclusions. Cadmium bioaccumulation is detected in the mitochondria at microscale by TEM/X-EDS, which is the visual evidence of morphological changes of mitochondria related to Cd.ConclusionThe combination of detailed ultrastructure and microscale X-ray energy dispersive spectroscopy (X-EDS) characterization of cadmium hepatotoxicity demonstrate that cadmium indeed leads to mitochondrial damage, which is helpful for further investigation of the pathological mechanism of cadmium hepatotoxicity.  相似文献
