A hyphomycete with black synnema and appendaged phragmoconidia was collected on Rhododendron brachycarpum showing twig blight symptom in Aomori Prefecture, Japan. The twig blight is macroscopically similar to the disease symptoms caused by Pycnostysanus azaleae. The fungus, however, has determinate synnemata, (1–)3-level verticillately branched conidiophores, and Pestalotia-like conidia. It cannot be accommodated by any existing genus of anamorphic fungi. A new genus Synnemapestaloides is established to accommodate the fungus, which is named S. rhododendri. The pathogenicity of the fungus on Rhododendron leaves and shoots is confirmed by inoculation. Synnemapestaloides twig blight is proposed for the disease of R. brachycarpum caused by the fungus.  相似文献

The anamorphic fungus Synchaetomella lunatospora gen. et sp. nov. is described from a culture isolated from leaf litter in Singapore. The genus is characterized by the production of synnematous conidiomata with brown stipes, phialidic conidiogenesis with terminal and acropleurogenous conidiogenous cells, and falcate, 1-septate conidia, aggregating in white, mucilaginous heads. Its phylogenetic relationships, inferred from the partial nuclear small ribosomal subunit (18S) sequence, are with the coelomycetous genera Sphaerographium, Chaetomella, and Hainesia. Neosartorya indohii and N. tsurutae, two new Neosartorya species isolated from tropical rainforest soil in the Amazonian area, Brazil, are described and illustrated. Neosartorya indohii is characterized by its spreading growth on Czapeks and malt extract agars, light yellow cleistothecia, broadly lenticular ascospores with two conspicuously serrate-incised equatorial crests and tuberculate convex surfaces, and globose to subglobose conidia with a smooth wall. Neosartorya tsurutae is characterized by its spreading growth on Czapeks and malt extract agars, white cleistothecia, broadly lenticular ascospores with four equatorial crests and rugulose-ruminate convex surfaces, and ovoid to broadly ellipsoidal conidia with a smooth wall.  相似文献

During a survey of bryophilous fungi from boreal and montane habitats in central Alberta, a hitherto undescribed species of Cladophialophora was recovered from Polytrichum juniperinum, Aulacomnium palustre, and Sphagnum fuscum. On potato dextrose agar (PDA) colonies grew slowly, attaining a diameter of 25 mm after 30 d, were dark grey, velvety, radially sulcate, and convolute and cracked at the centre. Micronematous conidiophores gave rise to branched chains of small (1–2 × 8–22 μm), cylindrical to fusiform conidia with truncate, swollen scars at each end. Phylogenies built on the ITS and ribosomal SSU regions indicate the isolates form a monophyletic clade within the family Herpotrichiellaceae (Chaetothyriales) that is composed of two geographically based groups, each with 99 % within-group sequence similarity and 97–98 % between-group sequence similarity. A teleomorph has not been found but would likely be similar to species of Capronia. In vitro inoculation of the isolates onto axenically grown P. juniperinum produced no discernible host symptoms, and host penetration could not be detected using light microscopy. The production of polyphenol oxidases by the fungus and the role of other Cladophialophora species as latent endophytes and saprobes suggest that a potential role for the fungus is the degradation of the polyphenol-rich cell walls of mosses. A dichotomous key to species of the genus Cladophialophora is provided.  相似文献

Conidia of two morphologically different types, one with a basal appendage only and the other with appendage at both ends, were isolated from the stems of Paeonia suffruticosa. Single conidial isolates of both types of conidia yield identical colonies, which then produced both types of conidia on agar media depending on temperature, thus showing that both types of conidia belong to the same fungus. Seimatosporium botan is described based on its morphological characteristics. The teleomorph of the fungus was first found on sterilized P. suffruticosa stems placed on water agar, when grown at 5°C for 2 months in 12-h photoperiod. Discostroma botan is described for this fungus. The teleomorph is also found on the same host in the field.  相似文献

Botryosphaeria spp. are common endophytes of woody plants, and they also include some serious pathogens of Eucalyptus and Acacia species. Numerous anamorphs have been associated with Botryosphaeria, of which the species Fusicoccum are amongst the most common. Here, we characterize two new Fusicoccum species, isolated from Eucalyptus and Acacia trees in Venezuela, based on morphological features in culture and comparisons of DNA sequence data. The two taxa named Fusicoccum andinum and F. stromaticum spp. nov, reside in two well-supported clades (BS values = 100 %) based on a combined data set of the ITS of the rDNA operon and translation elongation factor 1-α (EF1- α) gene sequences. The conidia of F. andinum are unusually large amongst Botryosphaeria anamorphs, and peripherally resemble those of B. mamane and B. melanops. F. stromaticum is characterized by large conidiomata in cultures, growth at 35 °C and slightly thickened conidial walls, which is different to most other Fusicoccum spp. No teleomorphs were observed for these fungi, but DNA sequence data show that they are anamorphs of Botryosphaeria.  相似文献

Pezicula cinnamomea (DC.: Fr.) Sacc. was frequently isolated from dead twigs of flowering cherry (Prunus × yedoensis Matsum.) in Aomori Prefecture, Japan. Inoculations of flowering cherry tree twigs with the fungus resulted in longitudinal browning of heartwood tissues 3–4cm long after 7 months, showing no apparent symptoms on the bark. The fungus were reisolated at high rates from the affected tissues. Apothecia were formed on cut affected twigs when they were placed in a moist Petri dish and kept under near-UV (Black Light Blue; BL-B) light irradiation. On potato sucrose agar (PSA) plates, the fungus failed to form apothecia in the dark, while many apothecia were formed under BL-B and, to a lesser extent, under white light. In nature, the light, especially near-UV, might be an important factor in apothecium formation. Under near-UV from BL-B conidiomata with conidia formed after 5–6 days and mature apothecia after about 14 days. The developmental processes of the apothecium were histologically studied under light and electron microscopes. Pezicula cinnamomea, which is homothallic and fruits easily in culture, may be very useful for life cycle studies and the elucidation of ecological role of this group of fungi.  相似文献

Fusarium verticillioides and other Fusarium species were examined for their spore germination phenotypes. In general, germinating spores of F. verticillioides formed germ tubes that immediately penetrated into agar. Such invasive germination was the predominant growth phenotype among 22 examined field isolates of F. verticillioides from a broad range hosts and locations. However, two of the field isolates were unique in that they formed conidial germ tubes and hyphae that grew along the surface of agar before penetration eventually occurred. Conidia of 22 other Fusarium species were assessed for their germination phenotypes, and only some strains of F. annulatum, F. fujikuroi, F. globosum, F. nygamai, and F. pseudoanthophilum had the surface germination phenotype (21 % of the strains assessed). Sexual crosses and segregation analyses involving one of the F. verticillioides surface germination strains, NRRL 25059, indicated a single locus, designated SIG1 (surface vs. invasive germination), controlled the germ tube growth phenotypes exhibited by both conidia and ascospores. Perfect correlation was observed between an ascospore germination phenotype and the germination phenotype of the conidia produced from the resulting ascospore-derived colony. Recombination data suggested SIG1 was linked (7 % recombination frequency) to FPH1, a recently described locus necessary for enteroblastic conidiogenesis. Corn seedling blight assays indicated surface germinating strains of F. verticillioides were less virulent than invasively germinating strains. Assays also indicated pathogenicity segregated independently of the FPH1 locus. Invasive germination is proposed as the dominant form of spore germination among Fusarium species. Furthermore, conidia were not necessary for corn seedling disease development, but invasive germination may have enhanced the virulence of conidiating strains.  相似文献

Fifty-one strains of Fusarium verticillioides and F. proliferatum isolated from corn collected from four different geographic areas in Iran, namely Fars, Khuzestan, Kermanshah and Mazandaran (an endemic oesophageal cancer (OC) area) were evaluated for their ability to produce fumonisins B1 (FB1), B2 (FB2) and B3 (FB3) in corn culture. Fumonisin levels were determined by high-performance liquid chromatography. All tested strains of F. verticillioides and F. proliferatumproduced fumonisins within a wide range of concentrations, 197–9661 g/g, 18–1974 g/g, and 21–1725 g/g for FB1, FB2, and FB3, respectively. The highest mean concentrations of FB1, FB2, and FB3 were 3897, 806 and 827 g/g, respectively. Overall, 61% of the F. verticillioides and F. proliferatum strains produced higher levels of FB3 than FB2. The mean ratios of FB1:FB2, FB1:FB3 and FB1:total fumonisins were 8, 7 and 0.7 for F. verticillioides and 5.7, 10.7 and 0.7 for F. proliferatum, respectively. Significant differences in some of the meteorological data (rainfall, relative humidity and minimum temperature) from the four provinces were observed. Fumonisin levels produced by F. verticillioides strains isolated from Khuzestan province (tropical zone) were significantly (P < 0.01) higher than the other three provinces. This is the first report of the fumonisin-producing ability of F.verticillioides and F. proliferatum strains isolated from corn harvested from different geographic areas in Iran.  相似文献

Hairy roots were induced from androgenic embryos of horse chestnut (Aesculus hippocastanum L.) by infection with Agrobacterium rhizogenes strain A4GUS. Single roots were selected according to their morphology in the absence of antibiotic or herbicide resistance markers. Seventy-one putative transformed hairy root lines from independent transformation events were established. Regeneration was induced in MS liquid medium supplemented with 30 6-benzylaminopurine (BA), and the regenerants were multiplied on MS solid medium containing 10 M BA. Following elongation on MS medium supplemented with 1 M BA and 500 mg/l polyvinylpyrrolidone, the shoots were subjected to a root-inducing treatment. Stable integration of TL-DNA within the horse chestnut genome was confirmed by Southern hybridization. The copy number of transgenes was estimated to be from two to four.Communicated by E.D. Paecilomyces fumosoroseus, monospore culture EH-506/3, isolated in Mexico from Bemisia tabaci whitefly was tested for acute oral intragastric pathogenicity and toxicity in CD-1 mice. Animals were inoculated by gavage with only one dose (108 conidia/animal) of viable (72 mice), heat-killed (24 mice) fungus and compared to 18 control mice. Clinical observations were done daily; mycological and histological tests were performed during necropsies at days 3, 10, 17, and 21 after the inoculation. No mice were clinically ill or died. At the end of the study, their mean weight corresponded to healthy adults. Positive fungal cultures of feces were obtained only 24 h after inoculation. Positive cultures were found in 15 out of 360 organs (liver, spleen, kidney, brain, lung) in 12 of 72 mice inoculated with viable conidia. Gross pathology exhibited splenomegaly and liver paleness in mice inoculated with viable and heat-killed fungus. Non-germinated conidia were observed in studied organs, without any pathological tissue reaction, suggesting no mycological or histopathological evidence of fungal multiplication. The fungus was able to persist, but did not cause permanent damage to the host. This study supports the non-pathogenic/toxic status of P. fumosoroseus EH-506/3 when administered intragastrically in mice.  相似文献

Two xylariaceous fungi were isolated from a nest of a termite, Odontotermes formosanus, that was incubated in a laboratory after collecting from Iriomote Is., Okinawa Pref., in Japan. One of the two fungi was identified as Xylaria angulosa on the basis of the morphology of branched stroma produced on medium, tiny asci, and ascospores having a germ slit. Another fungus is an anamorphic fungus that produces synnemata up to 50 mm long from which dendritic conidiophores branch out. Unicellular conidia are holoblastically produced on a sympodially proliferating conidiogenous cell. Such morphological characters resemble those of the genus Geniculosporium. However, its distinctive synnema formation and dendritic conidiophores do not assign the fungus to Geniculosporium or other known genera and warrant establishment of a new genus. The phylogenetic tree based on the ITS regions of rDNA shows that the fungus is nested in the cluster of the genus Nemania (Xylariaceae), whose species have mainly Geniculosporium-like anamorphs. We describe here the present anamorphic fungus as Geniculisynnema termiticola gen. et sp. nov., and discuss its phylogenetic and ecological relationships to xylariaceous fungi, especially termiticolous species.  相似文献

Ophiocordyceps unituberculata, a new insect pathogenic fungus from southwestern China, is described using molecular phylogenetic and morphological data. This fungus differs from other Ophiocordyceps species by its enormously long monophialidic conidiogenous cells and a large periclinal protuberance often growing near the apex of conidiogenous cells, single conidia (lanceolate to fusiform) embedded in mucous sheath, and much larger conidial dimensions. Four-locus (nrSSU, nrLSU, tef-1α and rpb1) and ITS data phylogenetic analyses show that O. unituberculata belongs to the Hirsutella nodulosa clade within the genus Ophiocordyceps of Ophiocordycipitaceae and is a separate clade from other allied species. Molecular phylogeny and morphology both strongly support the distinctiveness of this taxon. The interspecific relationships in the H. nodulosa clade are discussed.  相似文献

A new rust fungus,Spumula serispora, is described. The morphology of the teliospores and the telia was studied. In the teliospores ofS. serispora andS. quadrifida, the type species of the genus, sterile cells could be found, which are comparable to apical cells of otherRaveneliaceae. Similar sterile cells were also present in the telial heads ofRavenelia texensis. The ontogeny of the teliospores ofS. serispora and the teliospore heads ofRavenelia texensis was studied and compared. In both species the ontogeny of the spores heads was similar. During an extended search in Western Canada for fungal root endophytes useful as biocontrol agents against soil-borne pathogens, we isolated Heteroconium chaetospira, as well as Phialocephala fortinii or similar taxa, from seven samples of forest soil using herbaceous seedlings of four different species (i.e., barley, Chinese cabbage, eggplant, and melon) as bait plants. Our results support a previous observation that eggplant is a particularly effective species for baiting H. chaetospira from soil and confirm the ability of this fungus to grow as an endophyte in the roots of axenically reared host plants. Cultural characters show that this species is similar to P. fortinii and other melanized fungi in the dark septate endophyte (DSE) group (e.g., Leptodontidium orchidicola, P. sphaeroides, and Cadophora finlandica) in that it produces darkly pigmented colonies on agar media. Heteroconium chaetospira differs from P. fortinii and other melanized members of the Leotiomycetes in the DSE group in that its conidia are fusiform and develop in blastic acropetal chains. Heteroconium chaetospira is phylogenetically distant from most DSE taxa because DNA sequences for the nuclear small subunit (SSU) ribosomal RNA gene (rDNA) indicate that the taxon is affiliated with the Herpotrichiellaceae of the Chaetothyriales rather than with the Leotiomycetes.  相似文献

Summary Microscopic examination of the vascular cambium of Eucalyptus globulus, from four trees, at nine height levels, revealed a non-storied cambial structure with both ray and fusiform initials. Average fusiform initial length increased from 443 m at 70% of tree height from the base to a maximum of 484 m at 10% of tree height. At 2.5% of tree height average fusiform initial length sharply declined to 438 m. A strong positive correlation (r = 0.757, df = 7, P <0.05) existed between average fusiform initial length and average fibre length at each height, giving quantitative support to the notion that variation in fusiform initial length is an important mechanism influencing fibre length in hardwoods. However, 62% of the longitudinal variation in average fibre length was explained by the extent of elongation during differentiation, which increased linearly down the stem. At 70% of tree height, the average length of fibres was 1.84 times that of the fusiform initials, compared to 2.11 times at 2.5% of tree height. Concomitant with this longitudinal gradient of elongation was a gradient of increasing tangential area of the cambium occupied by cambial rays. This was accounted for by dimensional increases in the ray initial cells with decreasing height. Possible mechanisms that regulate the elongation of developing fibres and the proportion of cambial rays are discussed.  相似文献

Summary As neighboring plants flower sequentially, do flower feeders preferentially remain in the area, rather than move to another area with flowering plants? I examined the movements of the meloid beetle Epicauta pennsylvanica, a flower predator specializing on Solidago, in four types of replicated experimental plots — monocultures of Solidago altissima, or S. altissima interplanted with members of the same genus, same family, or different taxonomic orders. I released marked beetles only in the genus plots, which contained four species of Solidago, two that bloom before S. altissima. The number of beetles in the genus plots declined steadily as S. altissima came into flower in all the plots; the total number of beetles in all the plots remained fairly constant. I found no evidence that plant neighborhoods affected beetle distribution. Beetles foraging on the early blooming Solidago species did not remain in the genus plots as S. altissima came into flower. In addition, beetles that left the genus plots did not differentially accumulate in any of the other plot types, even though one type of plot was a monoculture with four times the density of S. altissima than the other plots.  相似文献

Ascochyta caulina (P. Karst) v.d. Aa and v. Kest is aplant pathogenic fungus which is specific to Chenopodium albumL. It has been suggested as a potential mycoherbicide to this weed,which is important and wide spread in arable crops throughout Europe. Toinvestigate its potential as a biocontrol agent, the fungus has beentested in glasshouse and field experiments. Formulations containingdifferent combinations of A. caulina conidia, the phytotoxinsfrom the fungus and low doses of herbicides have been tested.Significant improvement in the efficacy of the fungus was achieved inglasshouse trials with an aqueous formulation containing PVA(0.1% v/v), Psyllium (0.4% w/v), Sylgard 309(0.1% v/v), nutrients and conidia (5 ×106/ml). The extracellular, hydrophilic phytotoxinsproduced by A. caulina were purified and their structuresdetermined. The main toxin, named ascaulitoxin, was characterised as theN2--D-glucopyranoside of the unusual bis-aminoacid2,4,7-triamino-5-hydroxyoctandioic acid. Two other toxins proved to betrans-4-amino-D-proline and the aglycone of ascaulitoxin. Thesetoxins have shown promising herbicidal properties. Field trials haveinvestigated the performance of A. caulina conidia applied atdifferent developmental stages of C. album either as a singletreatment or combined with sub-lethal doses of herbicides or with thefungal phytotoxins. With the available formulation, favourable weatherconditions are needed to obtain infection in the field. The efficacy ofthe strain of A. caulina used so far has proved to beinadequate to justify its development as a bioherbicide. This isprobably due to its low virulence.  相似文献

The left (5) inverted terminal repeat (ITR) of the Mos1 mariner transposable element was altered by site-directed mutagenesis so that it exactly matched the nucleotide sequence of the right (3) ITR. The effects on the transposition frequency resulting from the use of two 3 ITRs, as well as those caused by the deletion of internal portions of the Mos1 element, were evaluated using plasmid-based transposition assays in Escherichia coli and Aedes aegypti. Donor constructs that utilized two 3 ITRs transposed with greater frequency in E. coli than did donor constructs with the wild-type ITR configuration. The lack of all but 10 bp of the internal sequence of Mos1 did not significantly affect the transposition frequency of a wild-type ITR donor. However, the lack of these internal sequences in a donor construct that utilized two 3 ITRs resulted in a further increase in transposition frequency. Conversely, the use of a donor construct with two 3 ITRs did not result in a significant increase in transposition in Ae. aegypti. Furthermore, deletion of a large portion of the internal Mos1 sequence resulted in the loss of transposition activity in the mosquito. The results of this study indicate the possible presence of a negative regulator of transposition located within the internal sequence, and suggest that the putative negative regulatory element may act to inhibit binding of the transposase to the left ITR. The results also indicate that host factors which are absent in E. coli, influence Mos1 transposition in Ae. aegypti.Communicated by G. P. Georgiev  相似文献
