The paired-like homeodomain (HD) protein Chx10 is distinguished by the presence of the CVC domain, a conserved 56 amino acid sequence C-terminal to the HD. In mammals, Chx10 is essential both for the proliferation of retinal progenitor cells and for the formation or survival of retinal bipolar interneurons. We describe the cloning and characterization of a mouse Chx10 homologue, Vsx1; phylogenetic analysis suggests that Vsx1 and its putative vertebrate orthologues have evolved rapidly. Vsx1 expression in the adult is predominantly retinal. Whereas Chx10 is expressed both in retinal progenitors in the developing eye and apparently in all bipolar cells of the mature retina, Vsx1 expression is first detected in the eye at postnatal day 5, where it is restricted to cone bipolar cells.  相似文献

Vsx1是第一个在金鱼中发现的编码含有同源异型框(homeodomain)和CVC结构域蛋白的基因。该基因在胚胎发育的不同阶段在胚胎的不同区域和不同组织中表达,并已经证明它在视网膜视锥双极细胞的分化和正常功能的维持中具有重要作用。为了进一步研究该基因在不同发育阶段组织特异性表达的调节机制,实验用PCR方法分析了金鱼Vsx1的基因组结构和内含子多态性。结果表明:金鱼Vsx1基因由5个外显子和4个内含子组成,与斑马鱼、人、小鼠的Vsx1基因结构相同。4个内含子中,第一内含子有两种序列差异明显的类型,但第二、三、四内含子无明显差异。第一内含子的一种类型比另外一种类型多39个碱基,这39个碱基中包括了真核生物增强子的核心序列。这一观测结果提示金鱼Vsx1第一内含子可能与该基因的发育阶段特异性和组织特异性表达调节有关。同时,第一内含子序列的明显差异也为分析Vsx1不同等位基因或基因拷贝的表达活性,以及组织特异性表达调节方式提供了合适的探针序列。  相似文献

Using PCR analysis and immunofluorescence staining, we have investigated the expression of homeobox genes Vsx1/Chx10-1 and Vsx2/Chx10 from the Vsx family (visual system homeobox) during retinal morphogenesis in the chicken Gallus domesticus. It was found that the expression of the studied genes starts at the early stages of embryogenesis. It was shown that the proteins of Vsx1 and Vsx2 are localized in the bipolar cells of the inner nuclear layer of the forming retina. The participation of Vsx1/Chx10-1 and Vsx2/Chx1 in the regulation of retinal cell differentiation in various species of vertebrates and in humans was discussed.  相似文献

Cone bipolar cells of the vertebrate retina connect photoreceptors with ganglion cells to mediate photopic vision. Despite this important role, the mechanisms that regulate cone bipolar cell differentiation are poorly understood. VSX1 is a CVC domain homeoprotein specifically expressed in cone bipolar cells. To determine the function of VSX1, we generated Vsx1 mutant mice and found that Vsx1 mutant retinal cells form but do not differentiate a mature cone bipolar cell phenotype. Electrophysiological studies demonstrated that Vsx1 mutant mice have defects in their cone visual pathway, whereas the rod visual pathway was unaffected. Thus, Vsx1 is required for cone bipolar cell differentiation and regulates photopic vision perception.  相似文献

Vertebrate retinal progenitor cells (RPCs) undergo a robust proliferative expansion to produce enough cells for the retina to form appropriately. Vsx2 (formerly Chx10), a homeodomain protein expressed in RPCs, is required for sufficient proliferation to occur. Sonic Hedgehog protein (SHH), secreted by retinal ganglion cells (RGCs), activates Hedgehog (Hh) signaling in RPCs and is also required for sufficient proliferation to occur. Therefore, we sought to determine if reduced Hh signaling is a contributing factor to the proliferation changes that occur in the absence of Vsx2. To do this, we examined Shh expression and Hh signaling activity in the homozygous ocular retardation J (orJ) mouse, which harbors a recessive null allele in the Vsx2 gene. We found that Shh expression and Hh signaling activity are delayed during early retinal development in orJ mice and this correlates with a delay in the onset of RGC differentiation. At birth, reduced expression of genes regulated by Hh signaling was observed despite the production of SHH ligand. orJ RPCs respond to pre-processed recombinant SHH ligand (SHH-N) in explant culture as evidenced by increased proliferation and expression of Hh target genes. Interestingly, proliferation in the orJ retina is further inhibited by cyclopamine, an antagonist of Hh signaling. Our results suggest that reduced Hh signaling contributes to the reduced level of RPC proliferation in the orJ retina, thereby revealing a role for Vsx2 in mediating mitogen signaling.  相似文献

We screened a mouse germinal cell expression library with a probe derived from Sob1, a human testis-specific cDNA, and identified 2P1, a new mouse cDNA. A database search revealed that 2P1 was 91% identical to ORF1 of E3-3, a rat gene probably involved in the regulation of alternative splicing. Sequencing showed that 2P1 has a destabilization motif in its 3'-untranslated region. Northern blotting showed strong gene expression in the testis and weak expression in the epididymis, with no signal detected in other tissues. RT-PCR analysis confirmed testis and epididymis expression. In situ hybridization revealed that 2P1 mRNA was absent in spermatogonia but expressed in spermatocytes. This last result was confirmed by RT-PCR of FACS isolated primary spermatocytes (pachytene stage). Using RT-PCR, purified spermatids were also shown to express 2P1.  相似文献

Approaches for manipulating cell type-specific gene expression during development depend on the identification of novel genetic tools. Here, we report the generation of a transgenic mouse line that utilizes Vsx2 upstream sequences to direct Cre recombinase to developing retinal bipolar cells. In contrast to the endogenous Vsx2 expression pattern, transgene expression was not detected in proliferating retinal progenitor cells and was restricted to post-mitotic bipolar cells. Cre immunolabeling was detected in rod bipolar cells and a subset of ON and OFF cone bipolar cells. Expression was first observed at postnatal day 3 and was detectable between 24 hours and 36 hours after the last S-phase of the cell cycle. The appearance of Cre-immunolabeled cells preceded the expression of bipolar cell type-specific markers such as PKCα and Cabp5 suggesting that transgene expression is initiated prior to terminal differentiation. In the presence of a constitutive conditional reporter transgene, reporter fluorescence was detected in Cre-expressing bipolar cells in the mature retina as expected, but was also observed in Cre-negative Type 2 bipolar cells and occasionally in Cre-negative photoreceptor cells. Together these findings reveal a new transgenic tool for directing gene expression to post-mitotic retinal precursors that are mostly committed to a bipolar cell fate.  相似文献

小鼠子宫颈部位输卵管蛋白表达的初步研究   总被引:2,自引:0,他引:2  
用RT-PCR及免疫印迹法检测证实小鼠子宫颈粘膜上皮细胞具表达输卵管蛋白的能力,宫颈部所获得的输卵管蛋白转录产物, 310到 714的405bp的cDNA片段经序列测定及blast比较表明与输卵管部位表达的输卵管蛋白完全一致(100%)。小鼠子宫颈输卵管蛋白在动情周期的表达波动情况与输卵管粘膜上皮的类似,其表达于小鼠动情期明显增加。Westernblot显示小鼠子宫颈提取物中存在两种不同分子量(60KD,30KD)的输卵管蛋白。原位杂交结果则提示子宫颈粘膜上皮细胞含丰富的输卵管蛋白mRNA信号。通过"原体中风"实验未发现输卵管蛋白在体外对精子活动有影响,其功能尚须进一步分析。  相似文献

采用表达序列标签(EST)介导的基因克隆和表达谱分析,从小鼠心脏克隆了一个心脏特异新基因Lrrc10(GenBank Acc No. AF527781).该基因cDNA全长为1 410 bp,定位于小鼠染色体10D2,在基因组中无内含子.Lrrc10的最大开放阅读框编码的假想蛋白由274个氨基酸组成,含有7个亮氨酸重复基序.同源性检索未发现有整体同源性的已知基因.EST数据库中支持该基因cDNA序列的全部18条EST均来自小鼠心脏组织.对小鼠的不同组织cDNA的RT-PCR检测证实该基因主要在心脏中强表达,在肺低表达,而在其他组织中不表达或表达很弱.因此该基因是心脏特异的富亮氨酸重复超家族新成员.  相似文献

目的:研究印记基因Dlk1在小鼠胚胎发育过程中的动态表达模式,以揭示Dlk1与胚胎发育的关系。方法:通过半定量PCR和定量PCR分析Dlk1在小鼠胚胎发育E8.5~E19.5的基因表达模式,并选取Dlk1表达量最高的时期进行胚胎切片原位杂交和组织定量PCR分析。结果:在小鼠胚胎发育E8.5~E15.5时,Dlk1的表达逐渐升高,在E15.5时表达量达到最高;E15.5~E19.5时,Dlk1表达有所下降,但仍然维持较高水平。E15.5切片原位杂交显示,垂体、肺脏、软骨、舌和背侧肌肉组织中Dlk1表达较高,组织定量PCR实验进一步证实了原文杂交的结果。结论:Dlk1在小鼠胚胎发育中后期持续表达,并呈现一定的组织特异性,对胚胎发育可能起重要的调节作用。  相似文献
