Bacteriol. 90:1420-1425. 1965.-When toluene is added at appropriate levels to exponentially growing cultures of Escherichia coli, a time-dependent loss of turbidity is observed which is concurrent with a loss of material to the medium and with unmasking of beta-galactosidase. In addition, the galactoside permease system is totally destroyed. Electron micrographs confirm the indications that the cells are not being lysed by toluene, although the cytoplasm collapses to the interior of the cell. Included in the material lost from the cell after toluene treatment is 85% of the total ribonucleic acid (RNA), the principal source of which appears to be the ribosomes. The loss of RNA is temperature-dependent. Protein is also lost to the medium as a function of both temperature and available toluene. Up to 25% of the total protein is found in the medium, the precise amount depending on the level of toluene employed. Zone centrifugation studies of extracts from treated cells indicate that toluene elicits a rapid disaggregation of ribosomes that is terminated, at any stage, by disruption of the cells. The disaggregation is temperature-dependent and does not occur at 4 C. It appears to be distinct from the actual degradation of ribosomal RNA and is accompanied by an accumulation of small particles during the initial phases of treatment at 21 C. Toluene added to crude extracts of normal E. coli cells is unable to cause detectable ribosome destruction.  相似文献

The idea that plasmids replicate within hosts at the expense of cell metabolic energy and preformed cellular blocks depicts plasmids as a kind of molecular parasites that, even when they may eventually provide plasmid-carrying strains with growth advantages over plasmid-free strains, doom hosts to bear an unavoidable metabolic burden. Due to the consistency with experimental data, this idea was rapidly adopted and used as a basis of different hypotheses to explain plasmid-host interactions. In this article we critically discuss current ideas about plasmid effects on host metabolism, and present evidence suggesting that the complex interaction between plasmids and hosts is related to the alteration of the cellular regulatory status.  相似文献

Valsa ceratosperma, which is the pathogenic fungus of apple canker, was grown in a synthetic medium. The neutral extract from the culture filtrate was chromatographed on a silica gel column to give five isocoumarins. Their structures were determined by MS, UV, IR, 1H and 13C NMR, and CD spectra. Three of them were known compounds; ( ? )-5-methylmellein (1), ( ? )-5-carboxylmellein (2) and ( ? )-5-hydroxylmethylmellein (3). Since the absolute configurations at C-3 in 2 and 3 were not known until now, both were determined to be R by chemical correlations. The two were new compounds; ( + )-(3R,4S)-trans-4-hydroxy-5-methylmellein (4) and ( ? )-(3R,4R)-cis-4-hydroxy-5-methylmellein (5). All the five compounds showed phytotoxicity in a bioassay using detached apple shoots and lettuce seedlings.  相似文献

An attempt has been made to use low-voltage alternating current to kill microorganisms such as Escherichia coli. The bactericidal effect depends on the energy passing through the suspension and on the time during which the cells are left standing in the medium after the treatment. Most of the toxicity is due to an indirect effect developed with unalterable electrodes in the presence of chlorides in the medium. Tea tree oil (TTO) stimulates autolysis in exponential and stationary phase cells of Escherichia coli . Electron micrographs of cells grown in the presence of TTO showed the loss of electron dense material, coagulation of cell cytoplasm and formation of extracellular blebs. Stationary phase cells demonstrated less TTO-stimulated autolysis and also had greater tolerance to TTO-induced cell death, compared to exponentially grown cells. It was also revealed that a subpopulation of stationary phase cells demonstrated increased tolerance to TTO-bactericidal effects.  相似文献

Increasing amounts of tetradecyldimethylbenzyl ammonium chloride (TAC) were lethal to an increasing proportion of an actively growing culture of Escherichia coli. The loss of nucleic acid material by actively growing E. coli did not appear to play a major role in the lethal effect. It was found that lag-phase cells were more sensitive than logarithmic-phase cells to the lethal effect of TAC. The effect of TAC on the lysozyme sensitivity of the test organism was compared with that obtained using disodium dihydrogen ethylenediaminetetraacetate (EDTA). Although TAC was found to render the test organism susceptible to lysozyme, the degree of lysis never reached that attained with EDTA.  相似文献

The rare earth cerium was found to bind rapidly to Escherichia coli. Cerium inhibited oxygen uptake in the presence of glucose as well as the endogenous respiration of glucose-grown cells. For a cell concentration of 4 mg per ml, maximal inhibition was obtained at 120 mug per ml. Greater concentrations did not increase the inhibitory effect. Cerium inhibited (14)CO(2) evolution and (14)C uptake from uniformly labeled glucose. Marked changes in the distribution of (14)C incorporated into different chemical fractions of the cell were noted. The most striking changes occurred in the alcohol- and alcohol ether-soluble fractions, in which the (14)C activity was increased 5- to 20-fold in the presence of cerium.  相似文献

Exposure of Escherichia coli to critical acridine orange (AO) concentrations did not result in loss of viability. However, the deoxyribonucleic acid (DNA) of cells exposed to such agents was rapidly degraded and repolymerized. On the other hand, a bacterium deficient in DNA repair (pol A1, lacking DNA polymerase) was sensitive to the action of AO. The DNA of such cells was also degraded but it was not repaired.  相似文献

Exposure of growing cells to low levels of N-hydroxyurethan (approximately 0.05 m) resulted in loss of colony-forming ability. This was accompanied by degradation of cellular deoxyribonucleic acid. When the concentration of N-hydroxyurethan was increased to 0.2 m, bacteriostasis was the primary manifestation. The lethal action of the drug was prevented by contreatment with chloramphenicol or azauracil or by uracil-deprivation of a bacterial strain requiring this pyrimidine for growth.  相似文献

Silver resistance of sensitive Escherichia coli J53 and resistance plasmid-containing J53(pMG101) was affected by halides in the growth medium. The effects of halides on Ag+ resistance were measured with AgNO3 and silver sulfadiazine, both on agar and in liquid. Low concentrations of chloride made the differences in MICs between sensitive and resistant strains larger. High concentrations of halides increased the sensitivities of both strains to Ag+.  相似文献

When cells of Escherichia coli B growing in a glucose-synthetic medium were treated with mitomycin C, the effects produced by the antibiotic varied, depending on the concentration. When the concentration was reduced to less than 0.1 mug/ml, the action of the antibiotic was bacteriostatic; cell elongation resulted, but no effect on the synthesis of cellular macromolecules was apparent. At higher levels (more than 5 mug/ml), mitomycin C was highly bactericidal and inhibited deoxyribonucleic acid synthesis almost completely. The exposure of growing cells to a bactericidal level of mitomycin C resulted also in a delayed inhibition of the synthesis of ribonucleic acid (RNA) and protein. The capacity of the treated cells to synthesize beta-galactosidase inducibly in a medium free from a carbon source remained constant for the first 30 min and then was destroyed progressively with time. Prolonged incubation with the bactericidal level of mitomycin C caused a degradation of cellular nucleic acids, particularly RNA. The degraded nucleic acid components were eventually released into the medium.  相似文献

The effects of high temperatures (46 to 50 degrees C) on the production of F pili by Escherichia coli were studied by electron microscopy. Attached F pili rapidly disappeared at 48 and 50 degrees C but not at 46 degrees C. Free pili were not denatured at these temperatures. The pili that disappeared from the cells at 50 degrees C did not appear as free pili in the culture supernatant fluid, indicating that the pili had retracted to the cell surface or into the cell. The adsorption of either R17 phage or F pili antibody to the sides of pili prevented retraction. The disappearance of pili was accompanied by a loss in the ability to adsorb R17 phage but not M13 phage, suggesting that the tip of a pilus remains exposed after retraction.  相似文献
