从沈阳市南部污水处理厂活性污泥中分离获得同时具备异养硝化和好氧反硝化能力的新型菌株,研究其脱氮特性,为改善污水厂的脱氮处理工艺奠定基础。对菌株进行形态学观察和16S rRNA基因鉴定;分别以NH4Cl、NaNO2、KNO3为唯一氮源探究菌株的脱氮能力;以碳源、C/N比、pH值、温度、转速、接种量(V∶V)等因素对菌株脱氮效果的影响进行研究。获得一株新型异养硝化-好氧反硝化菌株,经16S rRNA基因序列比对为副球菌属(Paracoccus),命名为Paracoccus sp. QD-19。菌株对初始氨氮浓度在300 mg/L以下的低浓度氨氮去除率能够达到100%,去除速率为8.707 mg/(L·h)且在脱氮过程中几乎没有亚硝态氮和硝态氮的积累。以亚硝态氮和硝态氮作为唯一氮源时,对此两种氮源的去除率36 h内均能达到99%,去除速率分别为4.944和5.666mg/(L·h)。确定了去除氨氮的最佳脱氮条件:琥珀酸钠为碳源,C/N比为10,pH值为7,接种量(V:V)为1%,温度为30℃,转速为140 r/min。菌株Pa...  相似文献

【背景】基于硝化菌群的富集培养技术可高效稳定地去除养殖水体中的有害氮素,而当前在水产养殖领域有关硝化菌群定向培育及硝化功能菌株的研究较少。【目的】研究不同盐度、pH、温度、通气量条件下硝化菌群分离菌株XH1的生长及其对氨氮和亚硝氮的去除效果。【方法】设置不同梯度的盐度、pH、温度、通气量条件,通过计数菌量、测定氨氮及亚硝氮的浓度变化,比较不同条件下菌株XH1的生长及其对氨氮和亚硝氮的影响。【结果】菌株XH1可在盐度5‰-35‰、pH 6.0-9.0、温度15-45°C和通气量0.5-1 V/(V·min)的条件下生长良好,菌量最高可达2.34×109cells/mL;在盐度5‰-35‰、pH 6.0-9.0、温度15-30°C、通气量0.5 V/(V·min)的条件下,对氨氮的去除效果显著(P0.05),在第1-3天对培养液中氨氮的最高去除率可达86%-97%,但培养液中的氨氮浓度先降后升;对亚硝氮的最高去除率达68%。【结论】菌株XH1对盐度、pH、温度等主要环境因子具有良好的适应性,其对水体氨氮的去除效果良好,可作为中低盐度养殖池塘水体氨氮防控菌剂产品研发的备选菌株。  相似文献

【背景】深海海域具有高压、低温、无光等环境条件,蕴含着丰富而独特的微生物资源。【目的】从深海沉积物中定向分离、筛选脱氮效率高的好氧脱氮菌株资源,并揭示其脱氮特性,为开发水体脱氮微生物技术提供物质基础。【方法】以东太平洋、南大西洋、西南印度洋共10个站位的深海沉积物为研究材料,在28°C下使用无机氮源连续进行两轮富集培养,然后定性筛选可以脱除氨氮、亚硝态氮和硝态氮的菌株,并通过形态学和16S rRNA基因序列分析进行初步分类鉴定;对优选得到的功能菌株,分别采用以氨氮、亚硝态氮、硝态氮为唯一氮源的培养基定量研究其生长和脱氮性能。【结果】从10份大洋深海沉积物样品中共分离得到49株好氧反硝化菌,其中3株在有氧条件下反硝化效率较高,分别命名为Pseudomonassp.G111、Pseudomonassp.G112和Dietziamaris W023a,其中菌株G111和G112与模式菌株博岑假单胞菌Pseudomonas bauzanensis BZ93T的16S rRNA基因序列相似度为99.2%,菌株W023a与模式菌株海洋迪茨氏菌DietziamarisATCC35013T的16SrRNA基因序列相似度为99.9%。菌株G111、G112和W023a培养48h后,对氨氮的脱除率分别为98.0%、85.2%和97.6%;对亚硝态氮的脱除率分别为71.9%、67.5%和34.7%;对硝态氮的脱除率分别为66.0%、52.6%和56.3%。菌株G111、G112和W023a均为异养硝化-好氧反硝化菌,可通过好氧反硝化作用将亚硝态氮和硝态氮还原为含氮气体,也可通过异养硝化-好氧反硝化作用将氨氮转化为含氮气体。【结论】从深海沉积物中分离筛选得到3株高效好氧反硝化菌,所获得的菌株在水体净化、污水处理、生态系统修复等领域具有应用潜力。  相似文献

一株高效异养硝化菌的选育、鉴定及其硝化条件   总被引:3,自引:0,他引:3  
【目的】针对现阶段异养硝化菌硝化速率较低的问题,选育更高效的异养硝化菌,进而鉴定该菌株的种属,了解其硝化特性和硝化条件。【方法】分别从污水处理厂活性污泥、化肥厂土壤以及农田土壤中取样,以柠檬酸钠为碳源,NH4Cl为氮源,采用污泥驯化、驯化过程中驯化液连续梯度稀释、平板划线分离及颜色指示剂快速硝化效果检测等步骤,筛得一株高效的异养硝化菌。经生理生化和16SrDNA序列的系统发育分析鉴定其种属;将该菌接入人工氨氮废水,定时检测水中含氮化合物的变化,了解其硝化特性;通过改变培养基碳源、溶氧量、C/N比、温度和pH考察其硝化条件。【结果】获得的高效异养硝化菌为革兰氏阴性杆菌,不利用葡萄糖发酵,氧化酶、接触酶阳性,不产吲哚,能由有机酸盐产碱;其与产碱菌属菌株Alcaligenessp.ES-SDK-3的16SrDNA同源性高达99.7%。用该菌株处理初始氨氮浓度为182.30mg/L的废水,30h后氨氮去除率为99.8%,指数期平均氨氮去除速率为9.61mg-N/L/h,其在硝化过程中几乎没有亚硝酸盐氮和硝酸盐氮产生;最佳碳源为柠檬酸钠;高的溶氧量和高的C/N比有利于其降解氨氮,当C/N比为12时即可达到较好的效果;该菌株在温度为30℃-35℃,pH为5.0-9.0范围内均能较彻底地降解氨氮。【结论】该菌株为产碱菌属,命名为Alcaligenessp.HN-S;其在硝化速率与处理的氨氮浓度方面均高于目前国内外筛出的大多数异养硝化菌;通过考察其硝化条件,为其走向实际污水脱氮工艺提供了依据。  相似文献

【背景】城市垃圾渗滤液是一种成分复杂的有机废水,含氮量高,如果未经处理直接排放到环境中会造成严重的环境污染。【目的】筛选可以耐受垃圾渗滤液中高浓度氨氮并高效去除污水中氮素的异养硝化好氧反硝化菌株,为解决垃圾渗滤液的氮素污染提供功能菌株。【方法】从垃圾渗滤液中筛选分离能耐受高氨氮浓度的菌株,通过测定各菌株的脱氮能力,筛选到一株脱氮能力最强的菌株,命名为U1,通过测定16S rRNA基因序列和生理生化特性确定该菌株为铜绿假单胞菌。进一步研究了菌株U1在不同初始氨氮浓度、碳源、转速、初始pH、碳氮比等单因素变量下的脱氮能力,并结合L9(34)正交试验研究了菌株U1的最佳脱氮条件。【结果】分离出一株铜绿假单胞菌并命名为U1。该菌株的最优脱氮条件为:初始氨氮浓度为1 000 mg/L,红糖和柠檬酸三钠的混合碳源,pH 6.0,C/N为10,转速为130 r/min,菌株U1的最大总氮去除率为64.37%,最大氨氮去除率为76.73%。对于总氮和氨氮含量分别是2 345 mg/L和1 473.8 mg/L的垃圾渗滤液,菌株U1最大总氮去除率为27.86%...  相似文献

【目的】从污水厂的活性污泥中获得一株高效反硝化细菌。【方法】采用低温驯化,进行初筛、复筛选取一株反硝化活性最高的菌株,命名为L2,通过形态学、生理生化特征及16S r RNA基因序列分析研究其分类地位,系统研究理化因素对该菌株反硝化性能的影响。【结果】菌株在低温条件下能够稳定高效地进行反硝化,鉴定该菌株为荧光假单胞杆菌(Pseudomonas fluorescens),其反硝化最适接种量为10%,温度为20°C,p H为7.0,盐浓度为0.5%,碳源为葡萄糖,C/N为5.0,能够耐受较高初始硝态氮浓度。【结论】菌株L2是一株耐低温、耐高浓度初始硝态氮、耐低C/N、兼性厌氧、高效反硝化的荧光假单胞杆菌。  相似文献

【目的】分离获得耐碱反硝化菌株,确定其反硝化活性和耐碱能力。【方法】分离、纯化,获得耐碱反硝化菌株;通过形态观察、生理生化试验和16S rRNA基因测序分析,确定菌株分类地位;试验起始硝酸盐浓度和起始pH对分离菌株反硝化活性的影响。【结果】从实验室稳定运行的高效反硝化反应器中分离获得耐碱反硝化菌株R9,经鉴定归于Diaphorobater nitroreducens;菌株R9能够以甲醇为电子供体、硝酸盐为电子受体进行异养生长,当起始硝氮浓度为50 mg/L、起始pH为9.0时,288 h内硝氮去除率达93.25%;高浓度硝氮可抑制其反硝化活性,半抑制常数Ki为202.73 mg N/L;菌株R9的耐碱性良好,起始pH为11.0时的硝氮去除率是pH为9.0时的86%。【结论】菌株R9归于Diaphorobater nitroreducens,最适生长pH为9.0左右,是一株耐碱反硝化菌。  相似文献

【目的】筛选高效脱氮且N_2O释放量少的好氧反硝化细菌,并对菌株的反硝化特性进行研究,可为河口湿地富营养化水体的生物修复提供技术支撑。【方法】经BTB培养基初筛和反硝化能力测定,从辽河河口区芦苇湿地土壤中分离得到1株具有较高反硝化能力的好氧反硝化菌C3。经形态观察、生理生化鉴定和16S rRNA序列分析,对菌株进行鉴定。研究温度、碳源、pH及C/N对其生长量、反硝化能力及N2O释放的影响。【结果】筛选得到的高效好氧反硝化细菌C3,经鉴定属于假单胞菌属(Pseudomonas sp.)。反硝化特性研究结果表明,该菌最适碳源为柠檬酸三钠,在温度为30°C、pH为7.0、C/N为10时生长速率和脱氮效率最高且N_2O释放量较少。在此条件下,该菌在36 h内使NO_3~–由179.55 mg/L降至5.08 mg/L,脱氮率高达97.17%。该菌株在整个反硝化过程中中间产物N_2O的最大累积量较低,为0.22 mg/L。【结论】从湿地土壤中分离所得好氧反硝化菌C3为假单胞菌属的1个种(Pseudomonas sp.),该菌株在高效除氮和低N_2O累积方面均具有明显优势,对后续河口湿地富营养化水体治理具有重要意义。  相似文献

【背景】石化工业废水具有高盐含氮的特点,高盐度会对微生物代谢造成抑制,导致普通反硝化微生物难以在高盐环境下有效脱氮。【目的】筛选在高盐条件下仍能保持反硝化能力的菌株并研究其特性。【方法】富集筛选出一株耐盐反硝化细菌,对其进行生理生化特性和16S rRNA基因序列鉴定,对其生长条件进行优化并测定该菌株反硝化能力,对菌株在高盐环境下的产物进行定性定量分析。【结果】经鉴定菌株YA16-1为表皮短杆菌(Brevibacterium epidermidis),可对硝态氮进行反硝化作用,在盐度为3%、初始氮浓度为55 mg/L的条件下,18 h的硝态氮转化率达到97%;初始硝态氮浓度为250 mg/L时,24 h内硝态氮转化率达到100%。该菌株的最适生长条件为:2% NaCl,碳源为玉米芯粉,氮源为酵母粉,pH值为6.0,培养温度为30 ℃。菌株在盐度为2%-15%的培养基内生长良好。在15%盐度下,菌株通过产四氢嘧啶维持渗透压,产量为0.89 mg/mL。【结论】菌株YA16-1具有良好的耐盐能力和反硝化能力,在高盐废水处理、保护生态环境和四氢嘧啶的制备具有潜在的应用价值。  相似文献

基于响应面法对一株好氧反硝化菌脱氮效能优化   总被引:2,自引:1,他引:1  
【目的】水体富营养化是当今我国水环境面临的重大水域环境问题,氮素超标排放是主要的引发因素之一。好氧反硝化菌构建同步硝化反硝化工艺比传统脱氮工艺优势更大。获得高效的好氧反硝化菌株并通过生长因子优化使脱氮效率达到最高。【方法】经过序批式生物反应器(Sequencing batch reactor,SBR)的定向驯化,筛选获得高效好氧反硝化菌株,采用响应面法优化好氧反硝化过程影响总氮去除效率的关键因子(碳氮、溶解氧、pH、温度)。【结果】从运行稳定的SBR反应器中定向筛选高效好氧反硝化菌株Pseudomonas T13,采用响应面法对碳氮比、pH和溶解氧关键因子综合优化获得在18 h内最高硝酸盐去除率95%,总氮去除率90%。该菌株的高效反硝化效果的适宜温度范围为25?30 °C;最适pH为中性偏碱;适宜的COD/NO3?-N为4:1以上;最佳溶解氧浓度在2.5 mg/L。【结论】从长期稳定运行的SBR反应器中筛选获得一株高效好氧反硝化菌Pseudomonas T13,硝酸盐还原酶比例占脱氮酶基因的30%以上,通过运行条件优化获得硝氮去除率达到90%以上,对强化废水脱氮工艺具有良好应用价值。  相似文献

The mature embryo of seven species belonging to five genera of Indian bamboos is described. In all these the basic pattern of embryo organisation is same: the scutellar and coleoptilar bundles are not separated by an internode, the epiblast is absent, the lower portion of the scutellum and the coleorhiza are separated by a cleft and the margins of embryonic leaves overlap. The features unique to fleshy fruited bamboos are: presence of a massive scutellum, the juxtaposition of plumule and radicle and the occurrence of a bud in the axil of the coleoptile. The fleshy fruit bearing bamboos should be classified into one group, the tribeMelocanneae. Evidence is provided to recognise additional groups in the subfamilyBambusoideae.  相似文献

Komárek has recently reviewed the various species assigned to the green algal genusNeochloris Starr (Chlorococcales, Chlorococcaceae) and removed those with uninucleate vegetative cells to a new genus,Ettlia. Watanabe & Floyd, unaware ofKomárek's work, also reviewed the species ofNeochloris and distributed them among three genera—Neochloris, Chlorococcopsis gen. nov., andParietochloris gen. nov.—on the basis of details of the covering of the zoospore and the arrangement of the basal bodies of the flagellar apparatus. This paper reconciles these two treatments and makes additional recommendations at the ranks of genus, family, order, and class.  相似文献

The cardinalfishes (Apogonidae) are a diverse clade of small, mostly reef-dwelling fishes, for which a variety of morphological data have not yielded a consistent phylogeny. We use DNA sequence to hypothesize phylogenetic relationships within Apogonidae and among apogonids and other acanthomorph families, to examine patterns of evolution including the distribution of a visceral bioluminescence system. In conformance with previous studies, Apogonidae is placed in a clade with Pempheridae, Kurtidae, Leiognathidae, and Gobioidei. The apogonid genus Pseudamia is recovered outside the remainder of the family, not as sister to the superficially similar genus Gymnapogon. Species sampled from the Caribbean and Western Atlantic (Phaeoptyx, Astrapogon, and some Apogon species) form a clade, as do the larger-bodied Glossamia and Cheilodipterus. Incidence of visceral bioluminescence is found scattered throughout the phylogeny, independently for each group in which it is present. Examination of the fine structure of the visceral bioluminescence system through histology shows that light organs exhibit a range of morphologies, with some composed of complex masses of tubules (Siphamia, Pempheris, Parapriacanthus) and others lacking tubules but containing chambers formed by folds of the visceral epithelium (Acropoma, Archamia, Jaydia, and Rhabdamia). Light organs in Siphamia, Acropoma, Pempheris and Parapriacanthus are distinct from but connected to the gut; those in Archamia, Jaydia, and Rhabdamia are simply portions of the intestinal tract, and are little differentiated from the surrounding tissues. The presence or absence of symbiotic luminescent bacteria does not correlate with light organ structure; the tubular light organs of Siphamia and chambered tubes of Acropoma house bacteria, those in Pempheridae and the other Apogonidae do not.  相似文献

Intracellular cysteine aspartate-specific proteases (caspases) play both signaling and effector roles in realizing the program of cell death. Caspases function as proteolytic cascades unique for each cell type and signal triggering apoptosis. All parts of the proteolytic cascades are duplicated and controlled by feedback signals. Amplification cycles between pairs of caspases (the third and the sixth, the ninth and the third, the twelfth and the sixth, and others) help multiply the initial apoptotic signal. The presence of physiological inhibitors of apoptosis that directly interact with caspases creates a multilevel regulatory network of apoptosis in cell. The caspase proteolytic cascades are also regulated by sphingolipid secondary messengers, among them ceramide, sphingosine, and their phosphates. Moreover, an association of the caspase signaling with ubiquitin-dependent proteolysis is shown in cells. In particular, the use of extracellular activators and inhibitors of caspases allows irreversible activation of apoptosis in tumor cells or the prevention of apoptosis in cortical neurons under neurodegenerative diseases.  相似文献

The Drosophila Pax6 homolog twin of eyeless (toy) is so far the first zygotically expressed gene involved in eye morphogenesis in Drosophila. The study of its expression during embryogenesis is therefore informative of the initial events of eye development in Drosophila. We have analyzed how the initial expression domain of toy at cellular blastoderm is regulated. We show that the three maternal patterning systems active in the cephalic region (the anterior, terminal and dorsal-ventral systems) cooperate with zygotically activated gap genes to shape the initial expression domain of toy. Whereas Bicoid, Dorsal and Torso signaling synergistically act as activators, Hunchback, Knirps and Decapentaplegic act as repressors.  相似文献

Twenty three species in 11 genera were examined in the field to determine hosts. OnlyStriga asiatica andSeymeria cassioides have a narrow host range being restricted to grasses and pines, respectively. A molecular phylogeny of Hebeloma species from Europe   总被引:2,自引:1,他引:1  
In order to widen the scope of existing phylogenies of the ectomycorrhizal agaric genus Hebeloma a total of 53 new rDNA ITS sequences from that genus was generated, augmented by sequences retrieved from GenBank, and analysed using Bayesian, strict consensus and neighbour joining methods. The lignicolous Hebelomina neerlandica, Gymnopilus penetrans, and two species of Galerina served as outgroup taxa. Anamika indica, as well as representatives of the genera Hymenogaster and Naucoria, were included to test the monophyly of Hebeloma, which is confirmed by the results. Hebeloma, Naucoria, Hymenogaster and Anamika indica cluster in a strongly supported monophyletic hebelomatoid clade. All trees largely reflect the current infrageneric classification within Hebeloma, and divide the genus into mostly well-supported monophyletic groups surrounding H. crustuliniforme, H. velutipes, H. sacchariolens, H. sinapizans, and H. radicosum, with H. sarcophyllum being shown at an independent position; however this is not well supported. The section Indusiata divides with strong support into three groups, the position of the pleurocystidiate Hebeloma cistophilum suggests the possible existence of a third subsection within sect. Indusiata. Subsection Sacchariolentia is raised to the rank of section.  相似文献
