Commercial chitosan and laminarin, as well as -glucans, isolated from either Phytophthora megasperma f.sp. glycinea or Saccharomyces cerevisiae, were applied to decapitated tomato (Lycopersicon esculentum Mill.) plants and evaluated for their potential to induce defense mechanisms in root tissues infected by Fusarium oxysporum f.sp. radicis-lycopersici. A significant decrease in disease incidence was monitored in elicitor-treated plants as compared to water-treated plants. No difference was detected in the capacity of the elicitors under study to confer enhanced protection against pathogen attack. Ultrastructural investigations of the infected root tissues from watertreated (control) plants showed a rapid colonization of all tissues including the vascular stele. Fungal ingress was lways associated with marked host cell disorganization and cell wall alteration. In root tissues from elicitortreated plants, restriction of fungal growth to the epidermis and the outer cortex, decrease in pathogen viability, and formation of numerous wall appositions at sites of attempted penetration were the main features of the hostpathogen interaction. The wall appositions were found to vary greatly in their appearance from multi-textured to multi-layered structures, from elongated deposits to hemispherical protuberances. Application of various goldcomplexed probes to root tissue sections revealed that callose, pectin and phenolic-like compounds (likely lignin) were the main components of the newly-formed barriers. By contrast, cellulose appeared confined to outer or intermediate layers resembling the host cell wall in terms of structure and architecture. In the absence of fungal challenge, the cytologically visible consequences of elicitation were restricted to a discrete deposition of electron-opaque substances in the vacuoles of some cells, and wall appositions were not detected. The key importance of fungal challenge in the elaboration of defense mechanisms is discussed in relation to the possibility that an alarm ignal provided by the pathogen itself is required for the expression of resistance in plants previously sensitized by an exogenous elicitor.Abbreviations AGL Aplysia gonad lectin - FORL Fusariumoxysporum f.sp. radicis-lycopersici The authors wish to thank Sylvain Noël for excellent technical assistance and Drs. Isolates of non-pathogenic Fusarium moniliforme (Fu3, Fu7 and Fu24), F. oxysporum (Fu2, Fu4), F. solani (Fu25) and F. merismoides (Fu1) that were found to be effective in reducing wilt incidence in tomato were tested for their potential to elicit induced systemic resistance (ISR) in tomato. Talc formulations of these isolates derived from liquid fermentation as well as cell elicitors of these cultures were tested. Changes in the phenol and total protein contents and activities of peroxidase and polyphenol oxidase were studied. Isolate Fu3 induced more phenol and total protein contents as well as activities of peroxidase and polyphenol oxidase. Elicitors of Fu2 induced more of these compounds and enzymes. Although Fu1, Fu4 and Fu24 were found to give good control against Fusarium wilt incidence in an earlier study, they were less effective in inducing these defense related compounds. Peroxidase activity was increased when plants were treated with Fu3, Fu4, Fu7, Fu24 and Fu25, whereas polyphenol oxidase activity was increased only with the isolate Fu3 and elicitor of Fu2. It is suggested that ISR was the mode of action for the isolates Fu2 and Fu3, whereas for the other isolates, the mode of action may be root colonisation, competition for nutrition and so on. The role of ISR with non-pathogenic isolates of Fusarium spp. is discussed.  相似文献

课题组前期报道了一株对马铃薯具有促生防病作用的内生砖红镰刀菌Fusarium lateritium (FL617)。为拓展该菌株的应用范围,本研究以同为茄科作物的烟草为研究对象,探究了砖红镰刀菌对其生长和抗病的影响。结果表明,与对照组相比,处理组叶表面积、主根数、叶片数、叶绿素a和叶绿素b含量分别提高了5.0、3.9、1.4、1.3和1.3倍;该结果表明砖红镰刀菌对烟草具有促生作用。生测结果表明,砖红镰刀菌增强了烟草对青枯病的抗病性,其青枯病病情指数下降约30%植物激素合成相关基因表达模式分析,发现处理组植物激素合成相关基因表达显著上调(1.6-39.9倍);用青枯病菌Ralstonia solanacearum感染寄主植物后分析其水杨酸(SA)、茉莉酸(JA)和R基因信号相关基因的转录模式,发现与对照组相比,处理组SA、JA相关基因均显著上调(1.2-8.3倍),仅有一个R基因显著下调(50%)。进一步用GFP标记的菌株进行荧光定殖观察,发现植物根系周围簇生着带有绿色荧光信号的真菌菌丝,表明砖红镰刀菌可以定殖于烟草根系。综上所述,推测砖红镰刀菌F. lateritium能够通过定殖于烟草根系介导植物激素、免疫防御相关基因的表达从而影响植株的生长发育和抗病性。  相似文献

Forty-five bacterial isolates were collected from surface-sterilized leaves of mulberry ( Morus alba L.). By screening their antagonistic activities against Ralstonia solanacearum in vitro , four isolates showed a remarkable inhibitory effect. The evaluation of the antagonistic strains against bacterial wilt of mulberry indicated that the strain Lu144 effectively reduced disease incidence. In the greenhouse, Lu144 displayed effective biological control against bacterial wilt of mulberry when it was applied to sterile or nonsterile soil before the infection by the pathogen. Based on bacteriological properties and 16S rRNA gene sequencing, Lu144 was identified as a strain of Bacillus subtilis . The endophytic population and infection process of Lu144 in mulberry seedlings was explored following recovery of the green fluorescent protein (GFP)-labeled Lu144 and examination of the labeled strain by confocal laser scanning microscopy. Interestingly, the infection of GFP-labeled Lu144 cells into the mulberry seedlings occurred through the cracks formed at the lateral root junctions and the zone of differentiation and elongation, and the cells were able to develop and transfer in mulberry and mainly in the intercellular spaces of different tissues. The population of the GFP-labeled Lu144 inoculant was larger and more stable in leaves than that in roots and stems.  相似文献

Plant growth promoting rhizobacteria (PGPR) strains Rb29 (B. amyloliquefaciens MF352007), Bs1 (B. subtilis MF352017) and Bt1 (B. tequilensis MF352019) were tested for growth promotion and for their ability to induce systemic resistance against Fusarium wilt, a vascular disease of chickpea, using two methods that include whole plant and a split-root system. Bacillus strains and Fusarium oxysporum f. sp. ciceris (FOC) were inoculated on separate halves of roots of chickpea seedlings at the same time and then planted in separate pots either in superposition or one side of the other. All Bacillus strains systemically induced resistance against FOC, and significantly (p < 0.05) reduced the wilt disease by 98–100%. Application of Bacillus strains effectively enhanced plant growth, leading to increased plant height, root length, a fresh and dry weight of shoots and roots. These results help to explain the role of strains of Bacillus in growth promotion and biological control of Fusarium wilt in chickpea. This is the first report of systemic-induced resistance against Fusarium wilt in chickpea obtained by application of Bacillus strains to a root system spatially separated from the FOC-inoculated root.  相似文献

【目的】优化比基尼链霉菌HD-087摇瓶发酵条件,提高菌株发酵液对黄瓜枯萎病菌HU-M的抑制率,并通过拮抗试验初步评价发酵液的抑菌作用效果。【方法】采用单因素筛选及正交试验对HD-087的发酵培养基和发酵条件进行优化。经发酵液处理后,光学显微镜观察HU-M菌丝形态和孢子萌发抑制率,测定HU-M菌丝电导率。【结果】改进的发酵培养基配方为:淀粉1.00%、黄豆粉0.80%、酵母粉0.12%、CaCO30.40%。对发酵条件的研究表明:pH为6.8,180 r/min、28°C条件下,250 mL三角瓶装液量为40 mL,接种1 mL种龄为2 d的种子,发酵5 d为最佳培养条件。抑菌结果表明,HD-087产抗菌物质能造成病原菌HU-M菌丝细胞质渗漏,菌丝畸形,分生孢子萌发受抑制,5倍发酵稀释液孢子抑制率达72.1%;除此之外还能引起菌丝电解质渗漏,造成菌丝细胞膜受损。【结论】优化后的摇瓶发酵条件能提高生防菌HD-087发酵液抑菌效果,并且发酵液可破坏细胞膜明显抑制病原菌HU-M生长,具有较大开发应用潜力。  相似文献

对从塔里木盆地苦豆子中分离得到的内生细菌进行皿内涂布拮抗实验、对峙培养法拮抗实验、胞外分泌物拮抗性测定和盆栽控病实验等研究,结果表明塔里木盆地苦豆子根瘤中存在大量的棉花枯萎病菌拮抗性内生细菌。皿内涂布法筛选结果表明60株苦豆子根瘤内生细菌中有48株相对抑菌率超过50%。对峙培养法对48株拮抗性内生细菌进一步筛选的结果表明,对棉花枯萎病菌抑菌距离超过20 mm的菌株有40株。40株拮抗性内生细菌胞外分泌物对棉花枯萎病菌的抑菌距离超过5 mm的有26株。KDRE12和KDRE41对棉花枯萎病的盆栽控病平均防效分别为67.11%和72.65%,具有应用潜力。  相似文献

【背景】香蕉枯萎病是一种真菌土传毁灭性病害,由于抗病品种产量普遍不佳和化学防治易污染环境等系列问题,生物防治是其理想的防治方法。【目的】筛选抗香蕉枯萎病的深色有隔内生真菌(Dark septate endophytes,DSE)菌株,丰富生防菌株资源库。【方法】采用平皿和盆栽实验方法评价5株DSE对香蕉的促生作用和对香蕉枯萎病的防治效果,采用形态学和分子生物学相结合的方法对优良菌株进行分类鉴定。【结果】接种DSE可有效促进香蕉植株的生长,尤以菌株LS1作用最显著,接种后鲜重与干重分别比对照增加47.36%与42.40%;接种DSE可有效提高植株对香蕉枯萎病的抗性,其中菌株LS1处理的香蕉植株表现的防治效果显著优于其它菌株,平皿中的防效为86.19%,盆栽实验防效为63.19%;结合形态学和分子鉴定技术,将菌株LS1鉴定为绿头枝孢菌Cladosporium chlorocephalum。【结论】LS1是一株具有开发利用价值的香蕉枯萎病生防菌株。  相似文献

In the present study, we investigated the role of Trichoderma virens (TriV_JSB100) spores or cell‐free culture filtrate in the regulation of growth and activation of the defence responses of tomato (Solanum lycopersicum) plants against Fusarium oxysporum f. sp. lycopersici by the development of a biocontrol–plant–pathogen interaction system. Two‐week‐old tomato seedlings primed with TriV_JSB100 spores cultured on barley grains (BGS) or with cell‐free culture filtrate (CF) were inoculated with Fusarium pathogen under glasshouse conditions; this resulted in significantly lower disease incidence in tomato Oogata‐Fukuju plants treated with BGS than in those treated with CF. To dissect the pathways associated with this response, jasmonic acid (JA) and salicylic acid (SA) signalling in BGS‐ and CF‐induced resistance was evaluated using JA‐ and SA‐impaired tomato lines. We observed that JA‐deficient mutant def1 plants were susceptible to Fusarium pathogen when they were treated with BGS. However, wild‐type (WT) BGS‐treated tomato plants showed a higher JA level and significantly lower disease incidence. SA‐deficient mutant NahG plants treated with CF were also found to be susceptible to Fusarium pathogen and displayed low SA levels, whereas WT CF‐treated tomato plants exhibited moderately lower disease levels and substantially higher SA levels. Expression of the JA‐responsive defensin gene PDF1 was induced in WT tomato plants treated with BGS, whereas the SA‐inducible pathogenesis‐related protein 1 acidic (PR1a) gene was up‐regulated in WT tomato plants treated with CF. These results suggest that TriV_JSB100 BGS and CF differentially induce JA and SA signalling cascades for the elicitation of Fusarium oxysporum resistance in tomato.  相似文献

从病区健康烟草植株茎杆内分离到一株对烟草青枯罗尔氏菌(Ralstonia solanacarum)有强拮抗作用的内生细菌,命名为B-001菌株.拮抗性研究表明,B-001菌株对多种革兰氏阳性细菌、革兰氏阴性细菌以及病原真菌均有较强的抑制作用.形态和生理生化特征初步表明菌株B-001为芽孢杆菌属(Bacillus)细菌.经扩增、测序得到B-001的16S rDNA序列,GenBank接收号为DQ444283.用ClustalX进行多重序列对比,并通过MEGA3方法构建16S rDNA系统发育树,表明:菌株B-001与Bacillus subtilis (DQ415893)的相似性为99.2%,并处于同一分支;结合形态和生理生化指标,将其鉴定为枯草芽孢杆菌(B. subtilis).2005和2006年在湖南省桂阳县、宁乡县进行了田间试验,防效在40.03%~78.14%,防治效果良好,且明显优于农用链霉素.  相似文献

蔓割病是我国南方薯区甘薯主要病害之一,本研究采用直接观察、显微和超微结构观测等方法,对高抗、中感和高感蔓割病的3个甘薯品种(高抗品种:金山57,中感品种:热薯1号,高感品种:新种花)接种蔓割病菌28 d植株充分发病后其茎基部细胞的侵染结构进行了观察。结果表明,在用清水作对照处理时,3个品种茎部组织的细胞形态和结构正常且完整,细胞代谢强。高抗品种金山57无论是接种组还是对照组,均未发现病原菌丝的存在,其茎下部、中部和上部细胞结构相对完整。中感品种热薯1号,蔓割病菌从其茎基部侵入后导致茎基部细胞破损坏死,而中部寄主-病原互作较为活跃和典型,造成养分运输受阻,茎基部接种后病原菌丝会沿着寄主茎部的维管束和其他组织一直向寄主的茎部末端蔓延,遭受侵染后的部位其细胞反应与高感品种新种花类似。高感品种新种花遭受蔓割病原菌侵染后,病原菌菌丝从茎基部新鲜剪口侵入后进入表皮细胞、皮层、维管束并在甘薯茎基部蔓延至中部、上部,直至寄主整株枯死,甘薯茎的木质部导管出现侵填体,质壁分离;与此同时,细胞壁的沉积物及乳突在病原菌的入侵处形成,各种无定型物质或纤丝构成的织网迅速包围入侵菌丝。  相似文献

【目的】了解抗感品种香蕉植株中内生细菌与香蕉枯萎病间的关系,从而为香蕉枯萎病的发生与防控提供一定的理论基础。【方法】通过基于16S rRNA的末端标记限制性片段长度多态性技术(Terminal restriction fragment length polymorphism,T-RFLP)分析健康和感枯萎病香蕉植株不同品种各组织,以及香蕉植株不同发病时间根组织中内生细菌的多样性和群落结构。【结果】抗病品种"农科一号"植株根、假茎、叶片各组织中内生细菌的种类基本都比感病品种"巴西蕉"的相应组织中的要丰富。感枯萎病香蕉植株根、假茎、叶片各组织中内生细菌种类基本都比健康植株各组织的丰富。在植株发病前、发病初期、再到发病后一个月的不同时期,对于抗病品种而言,其内生细菌的多样性都基本保持稳定,而感病品种则变化幅度较大。同时发现不同品种不同组织的内生细菌的优势种群有所不同,且不同品种健康和发病植株都存在一些特有优势种群。【结论】香蕉抗病品种比感病品种植株中内生细菌的多样性更丰富且更稳定;感枯萎病植株中的内生细菌种类比健康的丰富;而且不同抗性品种中健康和感病植株内的优势种群存在明显差异。  相似文献

The objective of this study was to examine the effects of Bacillus amyloliquefaciens Y1 on the control of Fusarium wilt disease and subsequent improvement in the growth of tomato plants. The Y1 strain strongly inhibited Fusarium oxysporum f. sp. lycopersici in vitro and also produced indole-3-acetic acid (IAA) in both the presence and absence of tryptophan. Over 96% of tomato seeds germinated when treated with either water, tryptone soy broth, or Y1 cultures, whereas root (5.40?cm) and shoot (5.15?cm) lengths were greatest in tomato seedlings treated with Y1 cultures that lacked tryptophan. Three experimental treatments – Black White medium (BW), BW medium with a commercial fungicide (BW?+?F), and Y1 culture inoculated in BW medium (Y1) – were applied to control Fusarium wilt disease under in vivo conditions. Application of Y1 culture and BW?+?F led to significantly lower disease incidence than did BW; moreover, shoot length and fresh and dry weight of both roots and shoots were greater in plants treated with Y1 than in plants treated with either BW or BW?+?F. A similar trend was observed for chitinase and β-1,3-glucanase activities in roots and leaves of tomato plants in all treatment groups over most of the experimental period. Finally, the presence of Y1 in the rhizospheric soils of Y1-treated plants resulted in a significant reduction in the populations of other bacteria. The results of our study demonstrated the effectiveness of Y1 not only in the control of Fusarium wilt disease but also for the enhancement of plant growth in cultivated tomato.  相似文献

Benzo (1,2,3) thiadiazole-7-carbothioic acid S-methyl ester (BTH, CGA 245704), a non-toxic, synthetic chemical, was applied as a foliar spray to cucumber plants and evaluated for its potential to induce defense mechanisms in root tissues infected by the soilborne pathogen, Pythium ultimum Trow. In non-treated cucumber plants, fungal colonization was intense and paralleled marked host tissue damage, whereas in BTH-treated plants, pathogen ingress towards the vascular stele was apparently halted by the massive deposition of a phenolic-enriched material which occluded a large number of cortical and vascular parenchyma cells. This considerable increase in the accumulation of phenolics was accompanied by cytological disorders of the invading pathogen at a time when the wall-bound cellulose component was preserved. In addition to phenolic compounds, the occluding material contained large amounts of beta-glucoside residues. These residues gradually decreased in the areas neighboring fungal cells whereas phenolic deposition appeared to be more uniformly distributed throughout the occluded host cells. Pathogen penetration in non-occluded cucumber root cells coincided with other changes, mainly characterized by both the deposition onto the inner surface of the cell walls of some heterogeneous wall appositions and the coating of some intercellular spaces with an electron-opaque material. Evidence is provided in this study that BTH has the ability to induce SAR in cucumber. Exogenous, foliar applications of the chemical sensitize susceptible cucumber plants to react more rapidly and more efficiently to P. ultimum attack, mainly through the massive accumulation of phenolic compounds at sites of attempted pathogen penetration.  相似文献

A putative endophytic Bacillus cereus str. S42 (KP993206), recovered from surface-sterilised stems of Nicotiana glauca was assessed in vitro and in vivo for its antifungal potential towards Fusarium oxysporum f. sp. lycopersici (FOL). Pathogen sporulation was significantly inhibited by B. cereus str. S42. FOL mycelial growth was reduced using its whole-cell suspensions, cell-free culture supernatant and chloroform extract. Its extracellular metabolites remained effective after heating at 50–100 °C with a decline in their activity was observed beyond 100 °C, when added with proteinase K and/or after pH adjustment to 2 and 12. Chitinase gene was detected using PCR amplification. Gas chromatography–mass spectrometry analysis of its chloroform extract matched phthalic acid, dibutyl ester with high level of similarity. B. cereus str. S42 cell-free culture supernatant and whole-cell suspensions had significantly suppressed Fusarium wilt severity by 87–96% and enhanced tomato growth by 39–79% compared to FOL-inoculated and untreated control.  相似文献

The tomato I‐3 and I‐7 genes confer resistance to Fusarium oxysporum f. sp. lycopersici (Fol) race 3 and were introgressed into the cultivated tomato, Solanum lycopersicum, from the wild relative Solanum pennellii. I‐3 has been identified previously on chromosome 7 and encodes an S‐receptor‐like kinase, but little is known about I‐7. Molecular markers have been developed for the marker‐assisted breeding of I‐3, but none are available for I‐7. We used an RNA‐seq and single nucleotide polymorphism (SNP) analysis approach to map I‐7 to a small introgression of S. pennellii DNA (c. 210 kb) on chromosome 8, and identified I‐7 as a gene encoding a leucine‐rich repeat receptor‐like protein (LRR‐RLP), thereby expanding the repertoire of resistance protein classes conferring resistance to Fol. Using an eds1 mutant of tomato, we showed that I‐7, like many other LRR‐RLPs conferring pathogen resistance in tomato, is EDS1 (Enhanced Disease Susceptibility 1) dependent. Using transgenic tomato plants carrying only the I‐7 gene for Fol resistance, we found that I‐7 also confers resistance to Fol races 1 and 2. Given that Fol race 1 carries Avr1, resistance to Fol race 1 indicates that I‐7‐mediated resistance, unlike I‐2‐ or I‐3‐mediated resistance, is not suppressed by Avr1. This suggests that Avr1 is not a general suppressor of Fol resistance in tomato, leading us to hypothesize that Avr1 may be acting against an EDS1‐independent pathway for resistance activation. The identification of I‐7 has allowed us to develop molecular markers for marker‐assisted breeding of both genes currently known to confer Fol race 3 resistance (I‐3 and I‐7). Given that I‐7‐mediated resistance is not suppressed by Avr1, I‐7 may be a useful addition to I‐3 in the tomato breeder's toolbox.  相似文献

对安徽省淮北市小麦植株根、茎、叶中内生细菌的数量进行了调查和筛选,并测定了其抑菌活性.小麦根、茎和叶中的内生细菌的数量分别为7.01×105、4.26×105和0.94×105CFU·g-1鲜重.从健康小麦植株体内分离到131株内生细菌,通过对峙实验,筛选到6株对禾谷镰刀菌有拮抗作用的菌株,占所分离内生细菌总数的4.58%.来自根系组织中的菌株HB022的抑菌效果最佳,抑菌半径为17.0mm.根据形态特征、生理生化特性和16SrDNA序列分析,将菌株HB022初步鉴定为多粘类芽孢杆菌(Paenibacillus polymyxa).抑菌试验结果表明,100倍稀释的菌株HB022无菌发酵滤液对禾谷镰刀菌菌丝生长具有明显抑制作用,5倍稀释的无菌发酵滤液可完全抑制禾谷镰刀菌分生孢子萌发.可见,内生细菌HB022具有潜在生防应用前景.  相似文献

【背景】香蕉枯萎病是由尖孢镰刀菌古巴专化型(Fusarium oxysporum f. sp. cubense,Foc)引起的一种真菌毁灭性土传病害,近年来施用生防菌被认为是一种有效的防治手段。【目的】从香蕉根际土壤中分离筛选具有良好防效的生防菌,并通过优化培养基及发酵条件,提高生防菌数量及抑菌效率。【方法】以福建省漳州蕉园中根际土壤为样品,以香蕉枯萎病致病菌(4号生理小种)为指示菌,通过稀释涂布、平板对峙法筛选得到一株具有较强抑菌活性的拮抗菌株HQB-1。通过形态观察、生理生化检测及16SrRNA基因序列分析,初步鉴定其种属,并采用单因素试验及正交设计优化菌株的发酵培养基及发酵条件。【结果】初步鉴定HQB-1菌株为Burkholderiastagnalis;最适培养基为:牛肉膏5.0 g/L,酵母浸粉10.0 g/L,NaCl 5.0 g/L;最佳发酵条件为:温度27°C,pH 7.0,转速200 r/min,接种量1%,培养时间36 h。【结论】使用该条件培养获得的有效活菌数及抑菌率较优化前明显提高,其中OD600由优化前的1.251提高至1.881,抑菌率由优化前的9.18%提高至34.60%。  相似文献

An endophytic fungal isolate (Fs-K), identified as a Fusarium solani strain, was obtained from root tissues of tomato plants grown on a compost which suppressed soil and foliar pathogens. Strain Fs-K was able to colonize root tissues and subsequently protect plants against the root pathogen Fusarium oxysporum f.sp. radicis-lycopersici (FORL), and elicit induced systemic resistance against the tomato foliar pathogen Septoria lycopersici. Interestingly, attenuated expression of certain pathogenesis-related genes, i.e. PR5 and PR7, was detected in tomato roots inoculated with strain Fs-K compared with non-inoculated plants. The expression pattern of PR genes was either not affected or aberrant in leaves. A genetic approach, using mutant tomato plant lines, was used to determine the role of ethylene and jasmonic acid in the plant's response to infection by the soil-borne pathogen F. oxysporum f.sp. radicis-lycopersici (FORL), in the presence or absence of isolate Fs-K. Mutant tomato lines Never ripe (Nr) and epinastic (epi1), both impaired in ethylene-mediated plant responses, inoculated with FORL are not protected by isolate Fs-K, indicating that the ethylene signalling pathway is required for the mode of action used by the endophyte to confer resistance. On the contrary, def1 mutants, affected in jasmonate biosynthesis, show reduced susceptibility to FORL, in the presence Fs-K, which suggests that jasmonic acid is not essential for the mediation of biocontrol activity of isolate Fs-K.  相似文献
