Analysis has been made of testicular biopsies from 13 volunteers, 7 of whom received over 100 periods of 20–30 W of 915 or 2450 MHz microwave radiation. The other 6 subjects received fewer than 100 treatments. The radiation was localized to the scrotum for contraception, and the surface temperature was raised to 40–42° C. The results of the biopsies shows that human testes possess a greater tolerance to microwave than those of other species, such as rabbits or rats. Examined 0.5 years after cessation of treatment, it is evident that spermatogenesis resumes, even though they have been irradiated more than 100 times. No individuals were found to be sterile, though spermatogenesis had been severely inhibited during the period of exposure. Vestiges of damage remained, however. The number of degenerate sperm-atogenic cells was increased. and there was a thickening of the limiting membrane of the seminiferous tubules. Some hyaline degeneration was also present. There were local increases or decreases in the numbers of interstitial cells, but ultrastructural comparisons revealed no significant morphological abnormalities.  相似文献

Fixation and staining of planaria can affect the interpretation of histopathological changes following their exposure to various agents. We assessed several fixation protocols with various stains in planaria to determine an optimal combination. Planaria were fixed in each of the following: 10% neutral buffered formalin, 2.5%, glutaraldehyde, Bouin's, Zenker's, 70% ethanol, and relaxant. In addition, planaria were fixed in relaxant and postfixed in each of the fixatives above. Paraffin embedded sections from each fixation protocol were stained with hematoxylin and eosin (H & E), toluidine blue, periodic acid-Schiff (PAS), or phosphotungstic acld-hematoxylin (PTAH). Relaxant fixed planaria were also stained with Steiner's, Holmes, trichrome, Giemsa, Grocott's methenamine silver (GMS) and antibodies for intermediate filaments (cytokeratin, vimentin and desmin). Relaxant and Zenker's gave the best fixation with minimal artifacts. Formalin, glutaraldehyde, and ethanol were unacceptable because they caused contortions of the body, crenation, and a darkly pigmented epidermis. Gastroderm could be differentiated from stroma best when stained with H & E, toluidine blue and PTAH. Other organ systems differentially stained included the epidermis, marginal adhesion gland, nervous tissue, and muscle. PAS, Steiner's, Holmes, trichrome and the intermediate filament stains were not useful for planaria staining. The most morphological information was obtained with relaxant fixative and a combination of sections stained with H & E and PTAH.  相似文献

A technique for microwave fixation of inflated rat lung is described. Conventional intratracheal fixation with instillation of fixative into the airways at a constant pressure results in pressure artifacts as well as flushing and disruption of cells and exudates. Microwave fixation fixes these elements in situ without disruption and thus is valuable when evaluating the distribution of inflammatory infiltrates. Exudative pneumonitis was produced in the rat using intratracheal instillations of either endotoxin or silica and comparisons were made between histologic sections fixed using either standard formalin fixation or microwave fixation.  相似文献

Ethanol allows the skeleton of fetuses to be processed rapidly but fixes tissues incompletely. Formalin, on the other hand, preserves tissues well but retards maceration, thereby extending the time between sacrifice and skeletal examination.  相似文献

Because a technique was needed for good fixation of blood smears without the use of solvents such as methanol, the effect of heat fixation on human and rat blood smears was studied. Slides were placed in a fused silica dish, covered by a Pyrex petri dish lid, and heated for 5 min in an oven at 150° C. After such fixation, smears stained by Giemsa' s solution showed that the blood elements retained their characteristic morphology. Thus, even though volatile substances may be lost and heat-labile constituents may be modified, this simple method avoids contact of the cells with solvents.  相似文献

All fetuses were obtained from the same pregnant rat; half of them were placed in neutral formalin for 15 min at room temperature, then irradiated for 2.5 min in a domestic microwave oven. The remaining fetuses were placed in neutral formalin at room temperature for 48 hr as a control. Both experimental and control groups were exposed to routine tissue processing for light microscopy and embedded in paraffin wax. Sections 5 μm thick were stained with hematoxylin and eosin. Our results showed that the microwave technique reduced the fixation time while providing thin sections that were equal to or better in quality than those in the control group.  相似文献

The fetal skeleton double staining method is used to reveal developmental abnormalities in the skeletal system. We used alizarin red S and al-cian blue successfully with microwave irradiation for skeletal double staining. The fixation time was reduced from 4-7 days to 2-2.5 min and the staining time was reduced from 4 days to 23 min.  相似文献

On the Use of Microwave Radiation Energy for Brain Tissue Fixation   总被引:3,自引:3,他引:0  
Abstract: Focused microwave irradiation (MWR) is an increasingly accepted method of sacrifice of laboratory animals such as the mouse or rat. By fixing the brain within a fraction of a second with heat inactivation, the investigation of fast neurochemical events may be obtained. Even though the technique is widely utilized, its application is inconsistent. This report illustrates some of the requirements necessary for the proper application of MWR for the sacrifice of animals, particularly those related to the length of time MWR is applied and the efficiency with which generated MWR power is coupled to the brain tissue. Studies were performed on the mouse, using either a 2.5 KW or 6.3 KW generator with a focused, closed system waveguide at time intervals of 350 or 500 ms or 1.4 s. During each of these intervals MWR was varied so that core brain temperatures for all groups were held between 83 and 95°C. In contrast with reported studies that used full animal restraint, all animals were minimally restrained for less than 1 s before sacrifice. Tissue content of cyclic AMP, an index of neuronal activity grossly affected by subtle changes in the activity of adenylate cyclase and/or phosphodiesterases, was monitored. No differences in tissue cyclic AMP content in any of 12 brain regions were detected after MWR, either at 350 or 500 ms. A substantial increase in cyclic AMP content occurred in 8 of 12 brain regions examined following microwave irradiation for 1.4 s. On the basis of these experiments, accurate determination of cyclic AMP in rodent brain requires that the maximum time interval of MWR exposure should not exceed 500 ms.  相似文献

Recent research on the chemical nature of the red dyes isolated from Pterocarpus santalinus and certain West African plants, viz., Baphia nitida, Pterocarpus osun and Pterocarpus soyauxii, have been reviewed. P. santalinus contains santalins A, B and C., but no santarubin. Santalins and santarubins have been found in P. osun, P. soyauxii and B. nitida. The structural formulae of the santalins are presented and their differences from santarubins indicated. Santalins A and B have some similarities in structure with hematein. This is probably responsible for their staining properties; the possible mechanism of staining is discussed.  相似文献

采用微波炉加热,在敞口容器中,进行甲壳素脱乙酰反应,制备壳聚糖。考察了碱溶液浓度和微波加热时间对壳聚糖脱乙酰度的影响。固定微波加热时间30min,随NaOH溶液浓度增加,脱乙酰度先增加,后减小;NaOH溶液浓度为45%时,壳聚糖的脱乙酰度最高。固定NaOH溶液浓度为45%,随着微波加热时间延长,壳聚糖的脱乙酰度增加。微波加热的最佳时间为30min。加热时间继续延长,壳聚糖变黑。碱溶液浓度和微波加热时间对壳聚糖的粘均分子量影响都不大。本文试图从微波场的能量分布和微波加热机理方面解释实验结果。  相似文献

在微波辐射条件下,以2-萘酚和氯化苄为原料,用氢氧化钠作碱剂,碘化钾作催化剂,以水和N,N-二甲基甲酰胺(DMF)为溶剂合成了苄基-2-萘基醚;采用单因素实验法,考察了反应物的摩尔比、催化剂用量、微波功率、辐射时间等因素对苄基-2-萘基醚产率的影响。实验结果表明:在n(2-萘酚):n(氢氧化钠):n(氯化苄):n(碘化钾)=1:1.1:1:0.018,水7 ml,DMF 25 ml,微波功率320 W和辐射时间75 s的优化条件下,苄基-2-萘基醚的产率可达88.03%。  相似文献

The effect of microwave irradiation on the survival of bacteriophage PL-1, which is specific for Lactobacillus casei, was studied using a commercial 2,450 MHz microwave oven. The phages were inactivated by microwave irradiation according to almost first-order reaction kinetics. The rate of phage inactivation was not affected by the difference in the continuous or intermittent irradiation, nor by the concentrations of phages used, but was affected by the volume of phage suspensions, which prevented the loss of generated heat. Microwave irradiation of phage suspensions produced a number of ghost phages with empty heads, but fragmentation of the tail was hardly noticed. The breakage of phage genome DNA was primarily caused by the heat generated by microwave irradiation, whereas the phage DNA was not affected by the same temperature achieved by heat from outside. Thus we concluded that the phage-inactivating effect of microwave irradiation was mainly attributed to a thermal microwave effect, which was much stronger than a simple thermal exposure.  相似文献

It has long been known that the level of radiosensitivity between individuals covers a considerable range. This range is reflected in analysis of patient cell lines with some cell lines showing significantly reduced sensitivity to in vitro radiation exposure. Our increased exposure to radiation from diagnostic medical procedures and other life style changes has raised concerns that there may be individuals who are at an elevated risk from the harmful impact of acute or chronic low dose radiation exposure. Additionally, a subset of patients show an enhanced normal tissue response following radiotherapy, which can cause significant discomfort and, at the extreme, be life threatening. It has long been realised that the ability to identify sensitive individuals and to understand the mechanistic basis underlying the range of sensitivity within the population is important. A reduced ability to efficiently repair DNA double strand breaks (DSB) and/or activate the DSB damage response underlies some, although not necessarily all, of this sensitivity. In this article, we consider the utility of the recently developed γH2AX foci analysis to provide insight into radiation sensitivity within the population. We consider the nature of sensitivity including the impact of radiation on cell survival, tissue responses and carcinogenesis and the range of responses within the population. We overview the current utility of the γH2AX assay for assessing the efficacy of the DNA damage response to low and high dose radiation and its potential future exploitation.  相似文献

We describe a method in which microwave irradiation is used to reduce substantially the incubation time for immunoperoxidase staining of antigens in cryostat sections of pso-riatic skin. An incubation time of 5-9 min irradiation at 80 W generated similar or better staining intensity for all antibodies used compared to the standard methods using 30-60 min incubation at room temperature. Although we found that microwave irradiation could be used with all antibodies tested, independent of whether they recognized extracellular, membrane or cytoplasmic antigens in skin, the conditions needed to be optimized for each antibody.  相似文献

The Central Tissue Bank in Warsaw was established in 1963 and since then ionising radiation has been routinely applied to sterilise tissue grafts. Connective tissue grafts such as bone, cartilage, tendons, sclera, pericardium, skin, acellular dermis and amnion irradiated with a dose of 35 kGy in a 60Co source and/or with an electron beam 10 MeV accelerator are prepared in our Tissue Bank and two other multi-tissue banks operating in Poland. Over 250,000 radiation-sterilised tissue grafts have been prepared and used in hospitals throughout Poland and no infectious disease transmission or other adverse post-transplantation reactions have been reported up to today. It should be kept in mind however, that high doses of ionising radiation can evoke numerous chemical and physical changes that may affect the biological quality of tissue allografts. Therefore, interdisciplinary research has been undertaken at the Central Tissue Bank in Warsaw to establish the origin and stability of free radicals and other paramagnetic entities induced by irradiation in bone. The effects of various preservation procedures (e.g. lyophilisation, deep-freezing) and irradiation conditions (doses, temperature of irradiation) on the osteoinductive potential and mechanical properties of bone and on the degradation of collagen, a major constituent of all connective tissue grafts, have been also studied. The results of these studies indicate that radiation-induced changes can be diminished by modification of tissue preservation methods and that, to some extent, it is possible to reduce undesired radiation-induced damage to the tissue grafts.  相似文献
