In C4 grasses belonging to the NADP-malic enzyme-type subgroup, malate is considered to be the predominant C4 acid metabolized during C4 photosynthesis, and the bundle sheath cell chloroplasts contain very little photosystem-II (PSII) activity. The present studies showed that Flaveria bidentis (L.), an NADP-malic enzyme-type C4 dicotyledon, had substantial PSII activity in bundle sheath cells and that malate and aspartate apparently contributed about equally to the transfer of CO2 to bundle sheath cells. Preparations of bundle sheath cells and chloroplasts isolated from these cells evolved O2 at rates between 1.5 and 2 mol · min–1 · mg–1 chlorophyll (Chl) in the light in response to adding either 3-phosphoglycerate plus HCO 3 or aspartate plus 2-oxoglutarate. Rates of more than 2 mol O2 · min–1 · mg–1 Chl were recorded for cells provided with both sets of these substrates. With bundle sheath cell preparations the maximum rates of light-dependent CO2 fixation and malate decarboxylation to pyruvate recorded were about 1.7 mol · min–1 · mg–1 Chl. Compared with NADP-malic enzyme-type grass species, F. bidentis bundle sheath cells contained much higher activities of NADP-malate dehydrogenase and of aspartate and alanine aminotransferases. Time-course and pulse-chase studies following the kinetics of radiolabelling of the C-4 carboxyl of C4 acids from 14CO2 indicated that the photosynthetically active pool of malate was about twice the size of the aspartate pool. However, there was strong evidence for a rapid flux of carbon through both these pools. Factors influencing the rate of superoxide (O 2 - ) production by thylakoids were investigated to determine if increased production of the radical was related to injury induced by chilling at a moderate photon flux density (PFD). Plants used were Spinacia oleracea L., Cucumis sativus L. and Nerium oleander L. grown at either 200° C or 45° C. Superoxide production was determined by electron-spin-resonance spectroscopy of the (O 2 - )-dependent rate of oxidation of 2-ethyl-1-hydroxy-2,5,5-trimethyl-3-oxazolidine (OXANOH) to the corresponding oxazolidinoxyl radical, OXANO ·. For all plants, the steady-state rate of O 2 - production by thylakoids, incubated at 25° C and 350 mol photon · m–2 · s–1 (moderate PFD) with added ferredoxin and NADP, was between 7.5 and 12.5 mol · (mg chlorophyll)–1 · h–1. Incubation at 5° C and a moderate PFD, decreased the rate of O 2 - production 40% and 15% by thylakoids from S. oleracea and 20° C-grown N. oleander, chillinginsensitive plants, but increased the rate by 56% and 5% by thylakoids from C. sativus and 45° C-grown N. oleander, chilling-sensitive plants. For all plants, the addition of either ferredoxin or methyl viologen increased the rate of O 2 - -production at 25° C by 75–100%. With these electron acceptors, lowering the temperature to 5° C caused only a slight decrease in O 2 - production. In the absence of added electron acceptors, thylakoids produced O 2 - at a rate which was about 45% greater than that when ferredoxin and NADP were present. The addition of 3-(3,4-dichlorophenyl)-1,1-dimethylurea reduced O 2 - production under all conditions tested. The results show that the rate of O 2 - production increases in thylakoids when the rate of electron transfer to NADP is reduced. Thylakoids isolated from cells of the red alga Porphyridium cruentum exhibit an increased PS I activity on a chlorophyll basis with increasing growth irradiance, even though the stoichiometry of Photosystems I and II in such cells shows little change (Cunningham et al. (1989) Plant Physiol 91: 1179–1187). PS I activity was 26% greater in thylakoids of cells acclimated at 280 mol photons · m–2 · s–1 (VHL) than in cells acclimated at 10 mol photons · m–2 · s–1 (LL), indicating a change in the light absorbance capacity of PS I. Upon isolating PS I holocomplexes from VHL cells it was found that they contained 132±9 Chl/P700 while those obtained from LL cells had 165±4 Chl/P700. Examination of the polypeptide composition of PS I holocomplexes on SDS-PAGE showed a notable decrease of three polypeptides (19.5, 21.0 and 22 kDa) in VHL-complexes relative to LL-complexes. These polypeptides belong to a novel LHC I complex, recently discovered in red algae (Wolfe et al. (1994a) Nature 367: 566–568), that lacks Chl b and includes at least six different polypeptides. We suggest that the decrease in PS I Chl antenna size observed with increasing irradiance is attributable to changes occurring in the LHC I-antenna complex. Evidence for a Chl-binding antenna complex associated with PS II core complexes is lacking at this point. LHC II-type polypeptides were not observed in functionally active PS II preparations (Wolfe et al. The growth of the anaerobic acetogenic bacterium Acetobacterium woodii DSM 1030 was investigated in fructose-limited chemostat cultures. A defined medium was developed which contained fructose, mineral salts, cysteine · HCl and Ca pantothenate (1 mg · 1–1) supplied in a vitamin supplement. Growth at high dilution rates was dependent on the presence of CO2 in the gas phase. The max was found to be 0.16 h–1 and the fructose maintenance requirement was 0.1 to 0.13 mmol fructose · (g dry wt)–1 · h–1. A growth yield of 61 g dry wt · (mol fructose)–1, corrected for the cell maintenance requirement and for incorporation of fructose carbon into cell biomass, was determined from the fructose consumption. A corresponding growth yield of 69 g dry wt · (mol fructose)–1 was calculated from the acetate production assuming that fructose fermentation was homoacetogenic. A YATP of 12.2 to 13.8 g dry wt · (mol ATP)–1 was calculated from these growth yields using a value of 5 mol ATP · (mol fructose)–1 as an estimate of the amount of ATP synthesised from fructose fermentation. The addition of yeast extract (0.5 g · 1–1) to the medium did not influence the max or cell yield. The uptake of ammonium, nitrate and phosphate by laboratory-grown young sporophytes of Laminaria abyssalis was measured in a perturbed system (batch mode) at 18 °C and 35 ± 5 µE m–2 s–1 photon flux density. Uptake of all appeared to follow saturation-type nutrient uptake kinetics. The NO inf3 sup– (K s = 14.0 µM, V max = 5.0 µmol h–1 g–1 dry wt) and NH inf4 sup+ (K s = 4.6 µM, V max= 2.0 µmol h–1 g–1 dry wt) were taken up simultaneously, although NH inf4 sup+ was taken up more rapidly. Values of K 3 and V max for phosphate were, respectively, 2.21 µM and 0.83 µmol h–1 g–1 dry wt. Nitrate and phosphate were both consumed in similar rates (V max /Ks 0.37) at low concentrations. NH inf4 sup+ , thus, might be a more efficient form of N fertilizer if artificial enrichment of seawater is used.  相似文献

Adjustment in leaf area : mass ratio called leaf area ratio (LAR) is one of the strategies to optimize photon harvesting. LAR was recorded for 10 genotypes of Hevea brasiliensis under high irradiance and low temperature and the genotypes were categorized into two groups, i.e. high LAR and low LAR types. Simultaneously, the growth during summer as well as winter periods, photosynthetic characteristics, and in-vitro oxidative damage were studied. Low LAR (19.86±0.52 m2 kg–1) types, recorded an average of 18.0 % chlorophyll (Chl) degradation under high irradiance and 7.1 % Chl degradation under low temperature. These genotypes maintained significantly higher net photosynthetic rate (P N) of 10.4 mol(CO2) m–2 s–1 during winter season. On the contrary, the high LAR (24.33±0.27 m2 kg–1) types recorded significantly lower P N of 4 mol(CO2) m–2 s–1 and greater Chl degradation of 37.7 and 13.9 % under high irradiance and low temperature stress, respectively. Thus LAR may be one of the physiological traits, which are possibly involved in plant acclimation process under both stresses studied.  相似文献

Chloroplasts with high rates of photosynthetic O2 evolution (up to 120 mol O2· (mg Chl)-1·h-1 compared with 130 mol O2· (mg Chl)-1·h-1 of whole cells) were isolated from Chlamydomonas reinhardtii cells grown in high and low CO2 concentrations using autolysine-digitonin treatment. At 25° C and pH=7.8, no O2 uptake could be observed in the dark by high- and low-CO2 adapted chloroplasts. Light saturation of photosynthetic net oxygen evolution was reached at 800 mol photons·m-2·s-1 for high- and low-CO2 adapted chloroplasts, a value which was almost identical to that observed for whole cells. Dissolved inorganic carbon (DIC) saturation of photosynthesis was reached between 200–300 M for low-CO2 adapted chloroplasts, whereas high-CO2 adapted chloroplasts were not saturated even at 700 M DIC. The concentrations of DIC required to reach half-saturated rates of net O2 evolution (Km(DIC)) was 31.1 and 156 M DIC for low- and high-CO2 adapted chloroplasts, respectively. The light-dependent modulation of ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) activity was studied in two species: Phaseolus vulgaris L., which has high levels of the inhibitor of Rubisco activity, carboxyarabinitol 1-phosphate (CA1P), in the dark, and Chenopodium album L., which has little CA1P. In both species, the ratio of initial to fully-activated Rubisco activity declined by 40–50% within 60 min of a reduction in light from high a photosynthetic photon flux density (PPFD; >700 mol · m–2 · s–1) to a low PPFD (65 ± 15 mol · m–2 · s–1) or to darkness, indicating that decarbamylation of Rubisco is substantially involved in the initial regulatory response of Rubisco to a reduction in PPFD, even in species with potentially extensive CA1P inhibition. Total Rubisco activity was unaffected by PPFD in C. album, and prolonged exposure (2–6 h) to low light or darkness was accompanied by a slow decline in the activity ratio of this species. This indicates that the carbamylation state of Rubisco from C. album gradually declines for hours after the large initial drop in the first 60 min following light reduction. In P. vulgaris, the total activity of Rubisco declined by 10–30% within 1 h after a reduction in PPFD to below 100 mol · m–2 · s–1, indicating CA1P-binding contributes significantly to the reduction of Rubisco capacity during this period, but to a lesser extent than decarbamylation. With continued exposure of P. vulgaris leaves to very low PPFDs (< 30 mol · m–2 · s–1), the total activity of Rubisco declined steadily so that after 6–6.5 h of exposure to very low light or darkness, it was only 10–20% of the high-light value. Summary From acetylene reduction assays over a 10-month period starting in April 1979, nodule activities averaged 18.78 (se 4.67) moles C2H4 g nodule dw–1 h–1 forAlnus rubra and 59.95 (se 12.14) moles C2H4 g nodule dw–1 h–1 forCytisus scorparius. Plant rates were 1.91 (se. 47) moles C2H4 plant–1 h–1 forA. rubra and 0.55 (se. 17) moles C2H4 plant–1 h–1 forC. Scoparius. Plant activity and total leaf N were strongly correlated with the dw of other plant parts, but nodule activity and percent leaf N were not. Plant and nodule activities were not associated with temperature, moisture stress, precipitation events or percent light for either species over the growing season nor for 54A. rubra sampled in mid-season 1979 on one replication. After 5 to 6 growing seasons, 14A. rubra on the same site ranged from 30 to 332 cm in height and showed strong correlation between nodule dw, leaf dw, plant size and total leaf N. Results from this study and others indicate logistic equations may be modified to predict the effect of adding a N2 fixing plant to a population of non N2 fixing trees.  相似文献

Increases of 23- (5.6 mmol acetylene reduced mg dry wt–1) and 16- (4 mmol acetylene reduced mg dry wt–1) fold in nitrogenase activity and 12- (671 l H2 mg dry wt–1 h–1) and 6- (349 l mg dry wt–1 h–1) fold in H2 photoproduction in Rhodopseudomonas palustris JA1 over 24 h were achieved with pyrazine 2-carboxylate (3 mM) and 3-picoline (3 mM), respectively, and were higher than earlier reports of enhancement (1.5 to 5- fold) in biological H2 production using various alternative methods.  相似文献

The photosynthetic properties of a range of lichens (eight species) containing green algal primary photobionts of either the genus Coccomyxa, Dictyochloropsis or Trebouxia were examined with the aim of obtaining a better understanding for the different CO2 acquisition strategies of lichenized green algae. Fast transients of light/dark-dependent CO2 uptake and release were measured in order to screen for the presence or absence of a photosynthetic CO2-concentrating mechanism (CCM) within the photobiont. It was found that lichens with Trebouxia photobionts (four species) were able to accumulate a small pool of inorganic carbon (DIC; 70–140 nmol per mg chlorophyll (Chl)), in the light, which theoretically may result in, at least, a two to threefold increase in the stromal CO2 concentration, as compared to that in equilibrium with ambient air. The other lichens (four species), which were tripartite associations between a fungus, a cyanobacterium (Nostoc) and a green alga (Coccomyxa or Dictyochloropsis) accumulated a much smaller pool of DIC (10–30 nmol·(mg Chl)–1). This pool is most probably associated with the previously documented CCM of Nostoc, inferred from the finding that free-living cells of Coccomyxa did not show any signs of DIC accumulation. In addition, the kinetics of fast CO2 exchange for free-living Nostoc were similar to those of intact tripartite lichens, especially in their responses to the CCM and the carbonic anhydrase (CA) inhibitor ethoxyzolamide. Trebouxia lichens had a higher photosynthetic capacity at low and limiting external CO2 concentrations, with an initial slope of the CO2-response curve of 2.6–3.9 mol·(mg Chl)–1·h–1·Pa–1, compared to the tripartite lichens which had an initial slope of 0.5–1.1 mol-(mg Chl)–1·h–1·-Pa–1, suggesting that the presence of a CCM in the photobiont affects the photosynthetic performance of the whole lichen. Regardless of these indications for the presence or absence of a CCM, ethoxyzolamide inhibited the steady-state rate of photosynthesis at low CO2 in all lichens, indicating a role of CA in the photosynthetic process within all of the photobionts. Measurements of CA activity in photobiont-enriched homogenates of the lichens showed that Coccomyxa had by far the highest activity, while the other photobionts displayed only traces or no activity at all. We studied the export of inorganic carbon and nitrous oxide (N2O) from a Danish freshwater wetland. The wetland is situated in an agricultural catchment area and is recharged by groundwater enriched with nitrate (NO3 ) (1000 M). NO3 in recharging groundwater was reduced (57.5 mol NO3 m–2 yr) within a narrow zone of the wetland. Congruently, the annual efflux of carbon dioxide (CO2) from the sediment was 19.1 mol C m–2 when estimated from monthly in situ measurements. In comparison the CO2 efflux was 4.8 mol C m–2 yr–1 further out in the wetland, where no NO3 reduction occurred. Annual exports of inorganic carbon in groundwater and surface water was 78.4 mol C m–2 and 6.1 mol C m–2 at the two sites, respectively. N2O efflux from the sedimenst was detectable on five out of twelve sampling dates and was significantly (P < 0.0001) higher in the NO3 reduction zone (0.35–9.40 mol m–2 h–1, range of monthly means) than in the zone without NO3 reduction (0.21–0.41 mol m–2 h–1). No loss of dissolved N2O could be measured. Total annual export of N2O was not estimated. The reduction of oxygen (O2) in groundwater was minor throughout the wetland and did not exceed 0.2 mol 02 m–2yr–1. Sulfate (SO4 ––) was reduced in groundwater (2.1 mol SO4 –– m–2 yr–1) in the zone without NO3 reduction. Although the NO3 in our wetland can be reduced along several pathways our results strongly suggest that NO3 loading of freshwater wetlands disturb the carbon balance of such areas, resulting in an accelerated loss of inorganic carbon in gaseous and dissolved forms.  相似文献

Complete annual cycles of N2O and CH4 flux in forest soils at a beech and at a spruce site at the Höglwald Forest were followed in 1997 by use of fully automatic measuring systems. In order to test if on a microsite scale differences in the magnitude of trace gas exchange between e.g. areas in direct vicinity of stems and areas in the interstem region at both sites exist, tree chambers and gradient chambers were installed in addition to the already existing interstem chambers at our sites. N2O fluxes were in a range of –4.6–473.3 g N2O-N m–2 h–1 at the beech site and in a range of –3.7–167.2 g N2O-N m–2 h–1 at the spruce site, respectively. Highest N2O emissions were observed during and at the end of a prolonged frost period, thereby further supporting previous findings that frost periods are of crucial importance for controlling annual N2O losses from temperate forests. Fluxes of CH4 were in a range of +10.4––194.0 g CH4 m–2 h–1 at the beech site and in a range of –4.4––83.5 g CH4 m–2 h–1 at the spruce site. In general, both N2O-fluxes as well as CH4-fluxes were higher at the beech site. On a microsite scale, N2O and CH4 fluxes at the beech site were highest within the stem area (annual mean: 49.6±3.3 g N2O-N m–2 h–1; –77.2±3.1 g CH4 m–2 h–1), and significantly lower within interstem areas (18.5±1.4 g N2O-N m–2 h–1; –60.2±1.8 g CH4 m–2 h–1). Significantly higher values of total N, C and pH in the organic layer, as well as increased soil moisture, especially in spring, in the stem areas, are likely to contribute to the higher N2O fluxes within the stem area of the beech. Also for the spruce site, such differences in trace gas fluxes could be demonstrated to exist (mean annual N2O emission within (a) stem areas: 9.7±0.9 g N2O-N m–2 h–1 and (b) interstem areas: 6.2±0.6 g N2O-N m–2 h–1; mean annual CH4 uptake within (a) stem areas: –26.1±0.6 g CH4 m–2 h–1 and (b) interstem areas: –38.4±0.8 g CH4 m–2 h–1), though they were not as pronounced as at the beech site.  相似文献

The FB iron-sulfur cluster is destroyed preferentially by treating Photosystem I complexes with HgCl2(Kojima Y, Niinomi Y, Tsuboi S, Hiyama T and Sakurai H (1987) Bot Mag 100: 243–53). When FB is 95% depleted but FAis quantitatively retained in cyanobacterial PS I complexes, the reduction potential of FA remains highly electronegative (Em=–530 mV, n=1), the EPR spectral and spin relaxation properties of FA and FXremain unchanged, but NADP+ photoreduction rates decline from 552 to 72 mol mg Chl–1 h–1.When FB is reconstituted with FeCl3, Na2S and -mercaptoethanol, NADP+photoreduction rates recover to 528 mol mg Chl–1 h–1. Hydrogen production rates by Anabaena sp. strain TU37-1 obtained after an initial 1-day incubation period were approximately 70 to 80 and 3 to 9 µmol (mg chl)–1 h–1 under argon and nitrogen atmospheres, respectively. Hydrogen production under argon was not enhanced by addition of carbon dioxide, but was enhanced to some extent under nitrogen by increasing the initial carbon dioxide concentration. Rates of hydrogen and oxygen production during the initial 7-hour period were 15 and 220 µmol (mg chl)–1 h–1, respectively, in vessels with 18.5% initial carbon dioxide. Hydrogen production under nitrogen was enhanced by addition of carbon monoxide (1%). The rate obtained from the initial 1-day incubation period was about 40 µmol (mg chl)–1 h–1, which corresponded to about 60% of that under argon. On the basis of these observations, a possible strategy for hydrogen production by nitrogen-fixing cyanobacteria under nitrogen in the presence of carbon monoxide is indicated.  相似文献

AxenicTrentepohlia odorata was cultured at three different NH4Cl levels (3.5 × 10–2, 3.5 × 10–3, 3.5 × 10–4 M) and three different light intensities (48, 76, 122 µmol m–2 s–1). Chloride had no effect on growth over this range of concentration. High light intensity and high NH4Cl concentration enhanced the specific growth rate. The carotenoid content increased under a combination of high light intensity and low N concentration. WhenD. bardawil was exposed to the same combination of growth conditions, there was an increase in its carotenoid content. The light saturation and the light inhibition constants (K s andK i, respectively) for growth, and the saturation constant (K m) for NH4Cl were determined. TheK s andK i values were higher inT. odorata (66.7 and> 122 mol m–2 s–1, respectively) than inD. bardawil (5.1 and 14.7 µmol m–2 s–1, respectively). Degradation of polyaromatic hydrocarbons by Burkholderia cepacia 2A-12   总被引:1,自引:1,他引:0  
A new strain of bacterium degrading polyaromatic hydrocarbons (PAHs), Burkholderia cepacia 2A-12, was isolated from oil-contaminated soil. Of three PAHs, the isolated strain could utilize naphthalene (Nap) and phenanthrene (Phe) as a sole carbon source but not pyrene (Pyr). However, the strain could degrade Pyr when a cosubstrate such as yeast extract (YE) was supplemented. The PAH degradation rate of the strain was enhanced by the addition of other organic materials such as YE, peptone, glucose, and sucrose. YE was a particularly effective additive in stimulating cell growth as well as PAH degradation. When 1 g YE l–1, an optimum concentration, was supplemented into the basal salt medium (BSM) with 215 mg Phe l–1, the specific growth rate (0.30 h–1) and Phe-degrading rate (29.6 mol l–1 h–1) were enhanced approximately ten and three times more than those obtained in the BSM with 215 mg Phe l–1, respectively. Both cell growth and PAH degradation rates were increased with increasing Phe and Pyr concentrations, and B. cepacia 2A-12 had a tolerance against Phe and Pyr toxicity at the high concentration of 730–760 mg l–1. Through kinetic analysis, the maximum specific growth rate ( max) and PAH degrading rate ( max) for Phe were obtained as 0.39 h–1 and 300 mol l–1 h–1, respectively. Also, max and max for Pyr were 0.27 h–1 and 52 mol l–1 h–1, respectively. B. cepacia 2A-12 could simultaneously degrade crude oil as well as PAHs, indicating that this bacterium is very useful for the removal of oils and PAHs contaminants.  相似文献

The role of oxygen in the photoinactivation of the photosynthetic apparatus of Spinacia oleracea L. was investigated. Moderate irradiation (1200 mol photons m-2s-1) of spinach leaves in an atmosphere of pure nitrogen caused strong inhibition of subsequently measured net CO2 assimilation, whereas considerably less photoinhibition was observed in the presence of low partial pressures (10–20 mbar) of O2. The decrease in activity caused by anaerobiosis in the light was not based on stomatal closure; the decline of assimilation represents a photoinhibition, as activity was not impaired by low irradiation (80 mol photos m-2s-1). In contrast, gassing with pure N2 in the dark caused strong inhibition. Electron-transport rates and chlorophyll-fluorescence data of thylakoids isolated from photoinhibited leaves indicated damage to the electron-transport system, in particular to photosystem II reaction centers. In vitro, photoinhibition in isolated thylakoid membranes was also strongly promoted by anaerobiosis. Photoinhibition of electron-transport rates under anaerobic conditions was characterized by a pronounced increase in the initial fluorescence level, F0, of chlorophyll-fluorescence induction, in contrast to photoinhibition under aerobic conditions. Hill activity (photoreduction of 2,6,dichlorophenol indophenol) of heat inactivated (40°C, 3 min) and Tris-washed (0.8M, pH 8.3) thylakoids of Beta vulgaris (beet-spinach) was partially restored if they were incubated with 150 mM MgCl2 prior to the assay. Mg(NO3)2 or MgSO4 were unable to restore this activity. The extent of this reactivation was dependent upon the degree of inactivation by heating and upon the composition of the isolation and the resuspension buffer used during the heat treatment. Washing of heat-treated thylakoids with phosphate-EDTA buffer prior to incubation with MgCl2 did not affect the extent of this reactivation. Chloride ions seem to be required for the reactivation of Hill activity damaged either by heat or by Tris.Most commonly used chloroplast isolation and resuspension media, except for Tris-HCl as resuspension medium, were suitable for restoration of Hill activity in heat-damaged thylakoids by preincubation with 150 mM MgCl2 prior to the assay. Pretreatment with MgCl2 stimulated Hill activity in Tris-treated and heat-damage thylakoids if phosphate buffer was used for their resuspension. However, the same pretreatment inhibited Hill activity in unheated thylakoids isolated in Tris medium and resuspended in the same medium. On the other hand, MgCl2 pretreatment induced restoration of the Hill activity of the heated thylakoids when Tricine or Hepes was used as the resuspension medium. It appears that the presence of Tris somehow hampers the Cl induced reactivation. In order to gain information about seasonal and interannual variations of CH4-fluxes at a spruce control site, a limed spruce site and a beech site of the Höglwald Forest, Bavaria, Germany, complete annual cycles of CH4-exchange between the soil and the atmosphere with 2-hourly resolution were followed for 4 consecutive years. The ranges of CH4 fluxes observed for the different sites were: +12.4 to –69.4 g CH4 m–2 h–1 (spruce control site), +11.7 to –51.4 g CH4 m–2 h–1 (limed spruce site), and –4.4 to –167.3 g CH4 m–2 h–1 (beech site). Lowest rates of atmospheric CH4-uptake or even a weak net-emission of CH4 by the soils were observed during winter/spring times, whereas highest rates of CH4-uptake were always found in summer/spring. Over the entire observation period of 4 years, mean CH4-uptake rates were –1.82 kg CH4-C ha–1 yr–1 at the spruce control site, –1.31 kg CH4-C ha–1 yr–1 at the limed spruce site, and –4.84 kg CH4-C ha–1 yr–1 at the beech site. The results obtained in this study demonstrate that in view of the huge interannual variations in CH4-fluxes of approx. 1 kg CH4-C ha–1 yr–1, multiple year measurements of CH4-fluxes are necessary to accurately characterize the sink strength of temperate forest for atmospheric CH4. By comparison of CH4-fluxes measured at the spruce control site and the limed spruce site, a significant negative effect of forest floor liming on CH4-uptake could be demonstrated. Compared to the spruce stand, the beech stand showed on average approx. 3 times higher rates of atmospheric CH4-uptake, most likely due to pronounced differences between both sites with regard to the organic layer structure and bulk density of the mineral soil. Regression analysis between CH4-fluxes and environmental parameters revealed that at all sites the dominating factors regulating temporal variations of CH4 fluxes were soil moisture and soil temperature. Field measurements of CH4 concentrations in the soil profile and laboratory measurements of CH4-oxidation and CH4-production activity on soil samples taken from different soil depths showed that the CH4-flux at the Höglwald Forest sites is the net-result of simultaneous occurring production and consumption of CH4 within the soil. Highest CH4-oxidation activity was found in the uppermost centimeters of the mineral soil, whereas highest potential CH4-production activity was found in the organic layer.  相似文献
