 Embryogenic cell masses of three Scots pine (Pinus sylvestris) cell lines K779, K884 and K1009 were cultivated with the ectomycorrhizal (ECM) fungi Laccaria bicolor, L. proxima, Pisolithus tinctorius, Paxillus involutus and two strains of Suillus variegatus. The average growth ratio of the slowly proliferating cell line K1009 was improved by L. proxima and S. variegatus strain H, while of the rapidly proliferating lines K779 and K884 the non-mycorrhizal controls grew best. The fungi caused two distinct reactions in embryogenic cultures. In the positive reaction, the shape and light yellow colour of the cultures resembled the controls, while in the negative reaction the embryogenic cells became brown and necrotic and the fungi grew aggressively over them. These reactions to the fungi did not correlate completely with effects on the growth ratio. All the cell lines enhanced the radial growth of S. variegatus H and of P. tinctorius, while the Laccaria species and S. variegatus strain 1 thrived better alone. This study shows that early-stage embryogenic cells of Scots pine and ECM fungi are able to interact. As some fungi produced a positive reaction or even increased proliferation, they could be used to enhance somatic embryogenesis of Scots pine. Specific fungi might be used to induce the growth of slowly proliferating cell lines, and knowledge of positive cell line-fungus interactions could be useful in work with later stages of somatic embryogenesis, such as rooting. The frost hardiness (FH) of mycorrhizal [ectomycorrhizal (ECM)] and non-mycorrhizal (NM) Scots pine (Pinus sylvestris) seedlings was studied to assess whether mycorrhizal symbiosis affected the roots’ tolerance of below-zero temperatures. ECM (Hebeloma sp.) and NM seedlings were cultivated in a growth chamber for 18 weeks. After 13 weeks’ growth in long-day and high-temperature (LDHT) conditions, a half of the ECM and NM seedlings were moved into a chamber with short-day and low-temperature (SDLT) conditions to cold acclimate. After exposures to a range of below-zero temperatures, the FH of the roots was assessed by means of the relative electrolyte leakage test. The FH was determined as the inflection point of the temperature-response curve. No significant difference was found between the FH of mycorrhizal and non-mycorrhizal roots in LDHT (?8.9 and ?9.8 °C) or SDLT (?7.5 and ?6.8 °C). The mycorrhizal treatment had no significant effect on the total dry mass, the allocation of dry mass among the roots and needles or nutrient accumulation. The mycorrhizal treatment with Hebeloma sp. did not affect the FH of Scots pine in this experimental setup. More information is needed on the extent to which mycorrhizas tolerate low temperatures, especially with different nutrient contents and different mycorrhiza fungi.  相似文献

The aim of this study was to investigate the potential of different isolates of ectomycorrhizal (EM) fungi to enhance the growth of Pinus sylvestris seedlings in five natural peat substrates with different nitrogen concentrations, and the effect of the Scots pine seedlings and fungal inoculum on the formation of dissolved inorganic and organic nitrogen in peat. Utilization of different organic nitrogen compounds by microbial community in the peat was also investigated using Biolog MT MicroPlates. Inoculation of the seedlings with EM fungi enhanced seedling growth. Piloderma croceum increased root growth especially, whereas Lactarius rufus increased needle growth and Suillus variegatus I, II and III improved both root and needle growth. All the EM fungi also significantly affected stem growth. Nitrogen concentration of the peat did not affect seedling growth as much as the EM fungi. At the lowest peat N concentration (1.17%) NH 4 + mineralisation was lower and DON (dissolved organic nitrogen) accumulation higher than at higher peat N concentrations. The EM fungal isolates had different effects on NH 4 + and DON accumulation/degradation in peat. The EM fungal isolates significantly increased NH 4 + formation in the peat, whereas L. rufus and P. croceum had an opposite effect on DON accumulation. S. variegatus I significantly decreased the DON concentrations during peat incubation. The N concentration of the peat slightly affected the utilization of amino acids and polyamines by the microbial community, whereas inoculation with S. variegatus I, II or III had no effect.  相似文献

The effects of supplementary far-red sidelight on the formation of mycorrhizas and on the accumulation and allocation of dry weight and mineral nutrients were studied in Scots pine (Pinus sylvestris L.) seedlings. Starting one week after germination the seedlings were subjected to two different light quality regimes: control and simulated sparse-canopy conditions (FR+). In the FR+ regime, light reflected by neighbouring plants was simulated by means of supplementary far-red light sources, which reduced the horizontal red/far-red photon ratio (R:FR) without affecting PAR. Seedlings were harvested after three months of treatment. FR+ increased stem height and decreased the total dry weight of seedlings. Dry weight allocation to needles was not affected, whereas dry weight allocation to roots was reduced and that to stems was increased in FR+ treated seedlings. The total number of short root tips and developing mycorrhizas per seedling were lower in FR+ than in control plants. Most short roots were developing mycorrhizas, while non-mycorrhizal short roots and mycorrhizas with mantle or external mycelium were very scarce. Changes in the allocation of nutrients in general followed the changes in dry weight allocation, and changes in nutrient content followed those in total dry weight. However, mismatches among these changes resulted in significant changes in nutrient concentrations in some organs: the concentrations of nitrogen and potassium in needles and the concentration of nitrogen in stems were higher in FR+ than in control seedlings. Changes in biomass and nutrient allocation under low R:FR may promote rapid height growth during early development in stands of Scots pine seedlings, but concomitant reductions in growth of the root system and mycorrhizas may negatively affect tree performance over the long term.  相似文献

The proportion of planted forests in the Mediterranean Basin is one of the largest in the world. These plantations are dominated by pine species and present a series of characteristics such as low elevation, high competition or small tree size that make them more vulnerable to droughts. However, quantitative assessments of their post-drought growth resilience in accordance with species, site factors and tree characteristics are lacking. In this study we sampled 164 trees at four forest sites located in the drought-prone Sierra Nevada, southeastern Spain. We compared growth responsiveness to drought in rear-edge planted vs. relic natural Scots pine (Pinus sylvestris) and coexisting Pyrenean oak (Quercus pyrenaica) stands. Our objective was to characterize and compare the different growth responses to drought between species and sites and the effect of the main physiographic factors (altitude, aspect, and slope) on these responses since the influence of these factors on post-drought resistance and resilience has received little attention to date. Our results reveal that the planted pine sites with the lowest mean growth rates displayed greater resistance during drought, and that higher altitude was associated with improved resistance and/or resilience for all species and sites. Natural pine and Pyrenean oak stands were better adapted to the dry climatic conditions of the Mediterranean region where the study was undertaken, displaying greater resistance and/or resilience and lower influence of drought on growth in comparison to stands of planted pines. These results suggest that promoting the conservation of high-elevation pine plantations and enhancing the regeneration of natural pine and oak may improve the resistance and resilience of these drought-prone forest ecosystems.  相似文献

Global warming is driving plant range shifts towards higher latitudes, where plants encounter different light environment (photoperiod and light spectral quality) than that to which they are adapted. Light environment may indirectly influence the belowground part of the plant, where trees associate with ectomycorrhizal (ECM) and other biotrophic fungi. We studied joint impacts of warming and light climate on Scots pine (Pinus sylvestris) and its root-associated fungi in a reciprocal transplantation study, where pine seedlings from southern and northern tree populations were grown under similar experimental temperature in southern (60°N) and northern (69°N) Finland. Based on fungal ITS rDNA, the abundance of Basidiomycota, and ECM fungi in particular, was highest in the roots of southern pines in the south and in northern pines in the north, and seedling biomass was determined by population origin. Our results imply that root-associated fungi may respond differentially in native vs. non-native light environment of the host plant.  相似文献

To demonstrate the efficacy of direct DNA extraction from hyphal ingrowth bags for community profiling of ectomycorrhizal (ECM) mycelia in soil, we applied the method to investigate the influence of long-term repeated prescribed burning on an ECM fungal community. DNA was extracted from hyphal ingrowth bags buried in forest plots that received different prescribed burning treatments for 30 yr, and denaturing gradient gel electrophoresis (DGGE) profiles of partial fungal rDNA internal transcribed spacer (ITS) regions were compared. Restriction fragment length polymorphism (RFLP) and sequence analyses were also used to compare clone assemblages between the treatments. The majority of sequences derived from the ingrowth bags were apparently those of ECM fungi. DGGE profiles for biennially burned plots were significantly different from those of quadrennially burned and unburned control plots. Analysis of clone assemblages indicated that this reflected altered ECM fungal community composition. The results indicate that hyphal ingrowth bags represent a useful method for investigation of ECM mycelial communities, and that frequent long-term prescribed burning can influence below-ground ECM fungal communities.  相似文献

Exogenous spermidine (Spd) and the ectomycorrhizal (ECM) fungus Pisolithus tinctorius (Pers.) Coker and Couch had a synergistic effect on the maturation of Scots pine (Pinus sylvestris L.) somatic embryos. Induced maturation was expressed as a higher number of cell masses able to form embryos and a greater number of embryos formed per cell mass. In contrast, treatment with P. tinctorius alone on the hormone-free medium resulted in the lowest embryo-forming capacity. Retarded proliferation growth appeared to be required for maturation, but did not explain the synergistic effect of the fungus and exogenous Spd. Simultaneous treatment did not result in lower concentrations of putrescine (Put), Spd or spermine (Spm) in the embryogenic cell masses relative to the separate treatments. Our study is the first report on the use of a specific ECM fungus to induce maturation of somatic embryos, and it indicates that P. tinctorius was able to modify the maturation media in a way that, together with exogenous Spd, positively affected embryogenic cultures of Scots pine. Our study also shows that it is possible to enhance plant development other than root formation by using specific ECM fungi.  相似文献

Summary Ultraviolet-B light (UV-B) and ultraviolet-A light (UV-A) at higher doses exert a strong inhibitory (toxic) effect on axis growth in Scots pine (Pinus sylvestris L.) seedlings. This effect is unrelated to control of growth rate by phytochrome. Rather, after a toxic UV dose growth of the pine seedling no longer responded to phytochrome. Both, the effect of UV-B as well as the inhibiting effect of UV-A could be photoreactivated by blue light (B). The action of UV-A was 2 fold: (i) it exerted a toxic effect which could be photoreactivated by B, and (ii) applied after UV-B it photoreactivated to some extent the toxic UV-B effect. Obviously, the UV-A range causes a toxic effect, and at the same time is capable of photoreactivating the toxic UV effect. At higher doses the toxic effect prevails.  相似文献

The formation of phloem was studied for two years in stems of 50 to 60 year old trees of Scots pine (Pinus sylvestris L.) growing in nature. The development of phloem of the current year begins 10 to 20 days before the xylem formation and is completed with the termination of shoot growth in the end of June. Observations over the seasonal activity of cambium producing sieve-like cells of phloem and duration of their differentiation as compared to the xylem derivatives of cambium have shown that the maxima of formation of phloem and xylem cells could coincide or not coincide by season, while the activities of their differentiation were always at antiphase. The sieve-like cells of early phloem were separated from those of late phloem by a layer of tannin-containing cells, which are formed simultaneously with the formation of late xylem cells by the cambium. Seasonal dynamics of accumulation of starch grain in structural elements of the phloem is related to the xylem development. The content of metabolites in differentiating and mature phloem elements, in the cambium zone, and in the xylem cells growing in the radial direction depended on cell specificity, stage of their development, and type of forming wood, early or late, which differ in the cell wall parameters and, hence, requirement of assimilates. Significant differences were described between the content of low molecular weigh carbohydrates, amino acids, organic acids, and phenol compounds using two methods of calculation: per dry weight and per cell.  相似文献

In this study, we surveyed the long term effects of liming and fertilizing in old Scots pine stands on the ectomycorrhiza (ECM) colonization, tree growth and needle nutrient concentration 35 years later. Four mature stands of Scots pine on low productive mineral soil were limed in 1959 and 1964 with total doses of limestone ranging from 3 to 15 Mg ha?1 and fertilized with nitrogen (N) in 1970. Thirty-five years after the first liming treatment, all stands were analysed for tree growth and needle nutrient concentrations and two of the stands were also analysed for ECM colonization. ECM colonization increased significantly with liming from 61.5% in the control plots to 88% in the plot with the highest limestone dose. ECM colonization increased with increasing pH in the humus layer from 62% colonization at pH?=?3.5 to 90% at pH?=?6.5 and decreased with increasing amount of extractable phosphorus (P) in the humus. Liming did not affect the frequencies of different ECM morphotypes or dead short root tips, the fine root biomass or necromass. ECM colonization was uncorrelated with needle nutrient concentrations or tree increment. Liming did not significantly affect tree growth. However, nutrient concentrations of current-year needles were affected by prior liming. Ca concentrations in current-year needles increased from approximately 15 mg g?1 in control treatments to more than 30 mg g?1 in limed plots, whereas concentrations of Mn, Al, Fe, and in two stands, B, decreased due to liming. In conclusion, liming with doses up to 15 Mg ha?1 was detectable in stands 35 years after treatment. The liming significantly increased the ECM colonization of Scots pine fine roots, increased the needle nutrient concentration of Ca and decreased the needle concentrations of Mn, Al, and Fe.  相似文献

This study experimentally analyses the response to simulated herbivory of juvenile Scots pine of two different ages in contrasting abiotic scenarios, focusing on the potential dual role of browsing ungulates: negative, by removing aerial biomass, and positive, by stimulating compensation capacity and providing nutrients by depositing their excrement. Compensation against herbivory was investigated by experimentally clipping a set of Scots pine (Pinus sylvestris L. nevadensis) juveniles, grown under different levels of light and nutrient availability. The responses analysed were survival, trunk-diameter growth, leader-shoot growth, increment in number of meristems, RGR, biomass of needles, shoots, root and whole plant, and root-to-shoot ratio. Clipping consistently resulted in a worse survival and performance of pines with respect to unclipped ones. From the factors analysed, light availability was responsible mainly for the variations in plant performance, while the addition of nutrients was much less important. Age was also important, with older pines showing in general better performance after clipping. Overall, clipping invariably had a negative effect on Scots pine, since none of the combinations of abiotic factors used resulted in overcompensation. However, the intensity on this negative effect proved quite variable, from almost an exact compensation in clipped older pines under full sunlight availability to very poor performance and high death probability in younger pines in shade. Scots pine cannot overcompensate after clipping, but, depending of the environmental conditions, the negative result of clipping varies from severe undercompensation to almost exact compensation. Also, small differences in sapling age can promote significant differences in sapling response to clipping and light environment.  相似文献

Lignosulfonate (LS) is a lignin-based polymer obtained as a by-product from paper industry, which may have potential as an amendment with macronutrients. We studied effects of LS on the interaction between Scots pine (Pinus sylvestris L.) seedlings and hypocotyl cuttings and the ectomycorrhizal (ECM) fungusPisolithus tinctorius (Pers.) Coker and Couch. The experiments were performed in vitroon the MMN agar medium containing Fe–LS chelate at the concentrations of 0, 5, 10 and 25 mg/L. Inoculation with P. tinctoriusincreased root growth of the seedlings. Fe–LS enhanced P. tinctorius induced formation of lateral roots and had a dose-dependent positive effect on the establishment of mycorrhizas on the seedlings. The growth of the fungal mycelium was improved by Fe–LS, which might cause faster and more intensive contact with the roots and, thus, better root growth and mycorrhiza formation. P.tinctorius enhanced also adventitious root formation and subsequent root growth of the hypocotyl cuttings but without any synergistic effect with Fe–LS. Our study with P. tinctorius and Scots pine in vitro indicates that a low-cost by-product Fe–LS, obtained from paper industry, may be a potential tool to improve the efficiency of fungal inoculations, thus, facilitating the early interaction between an ECM fungus and host seedling.  相似文献

Disentangling how variation in reproduction and growth is linked in plants across different ecological scales, and how allocation rules change in response to stress are fundamental aspects of life history theory. Although it is known that reproductive allocation is an allometric process and that environmental conditions can influence demographic traits, patterns of variation in vegetative and reproductive functions across and within individuals of tree species suffering drought-induced decline have rarely been documented. In this study we use Scots pine (Pinus sylvestris L.) as a model species to explore patterns of variation in cone production and growth in two declining populations at the southern edge of its distribution. A Bayesian approach was used to assess how these demographic traits vary as a function of drought effects and competition and covary across different ecological scales. The allometric trajectories relating tree size with cone production and growth differed along gradients of drought impacts and biotic interactions. Although reproduction and growth increased with tree size, cone production reached a maximum at intermediate sized trees and stabilized or decreased at larger sizes. Drought stress effects (defoliation at the tree level and overall decline at the plot level) and competition for resources reduced cone production and growth. Our results also showed differential effects of defoliation on cone production depending on tree size, with stronger effects on larger individuals. After accounting for these effects, much of the variation of demographic traits and correlations among them occurred at small ecological scales across individuals (i.e. within plots) and within individuals across years. This resulted in covariations between demographic traits among nearby individuals and within individuals through time, suggesting a consistent advantage in resource acquisition of some individuals within plots, and trade-offs between growth and cone production within trees across years. In conclusion, this study reports that drought-induced forest decline is associated with lower growth and cone production in Scots pine, which could contribute to explain the long-term impacts of drought in southern populations of this species and, in particular, its low regeneration capacity after severe drought.  相似文献

Effects of manipulation of litter and humus layers (removal, doubling and control treatments) on the colonization potential of ectomycorrhizal fungi were studied in two secondary stands of Pinus sylvestris (5 and 18 years old) in The Netherlands. Five-mont-hold, sterile-grown Scots pine seedlings, inoculated with Laccaria bicolor, Paxillus involutus or Rhizopogon luteolus and noninoculated seedlings were used as baits. The seedlings were harvested after one growing season. For comparison, sporocarps of ectomycorrhizal fungi were also investigated. Genus composition on the seedlings was independent of initial inoculum, but determined by both treatment and age of the stands. In both stands, removal of litter and humus layers increased, and addition of organic material decreased the number of ectomycorrhizal types on the seedlings. Not all indigenous genera were observed by either outplanting seedlings or sporocarp surveys.  相似文献

Abstract. 1. The survival, growth and development of larvae of the pine beauty moth, Panolis flammea (D & S) (Lepidoptera: Noctuidae) were examined as part of a study to determine (i) why damaging outbreaks of this insect in Scotland are frequent on lodgepole pine but do not occur on Scots pine, and (ii) why outbreaks are associated with areas of deep un-flushed peat.
2. Larvae were studied inside predator exclusion cages on mature pine trees in Elchies forest where a major infestation occurred in 1978–79.
3. Surprisingly, larval survival was generally greatest on Scots pine and lowest on lodgepole growing in deep peat. Also, larval growth and development were greater on Scots than lodgepole pine and were unaffected by the type of soil in which the lodgepole pine was growing.
4. Larval performance was generally better on lodgepole pine that had been thinned and received fertilizer.
5. Larval survival was affected by pine shoot stage during egg hatch in one of the two years in which this was examined.
6. Observations on P.flammea pupae were confused by disease, parasitism and probably by differences in weight loss caused by differences in larval development. Parasitism by ichneumonid parasitoids was recorded on Scots pine but not on lodgepole pine.
7. It was concluded that (i) the absence of pine beauty moth outbreaks on Scots pine, and (ii) the occurrence of outbreaks on lodgepole pine growing in deep unflushed peat were not the result of lodgepole pine growing in deep peat being a more suitable host plant than lodgepole pine and Scots pine growing elsewhere. It seems more likely that the observed outbreak behaviour of P.flammea in Scotland is associated with differences in predation and parasitism in plantations of difference host plants and in plantations of lodgepole pine growing in different soils.  相似文献
