The toxicity of three common antibiotics (streptomycin sulfate, tetracycline hydrochloride, and tylosin tartrate) to the freshwater rotifer Brachionus calyciflorus and brackish-water rotifer B. plicatilis was investigated using full-lifespan exposure durations. Effects of each antibiotic on lifespan, lifetime reproduction, and Malthusian parameter were assessed at seven nominal concentrations (ranging from 5.6 mg l−1 to 2,000 mg l−1) and a negative control. Lowest Observed Effect Concentrations (LOECs) were determined for reproduction and lifespan, while 1%, 10%, 25%, and 50% Inhibitory Concentrations (IC1, IC10, IC25, IC50) and 95% confidence intervals were estimated for all three endpoints. LOECs ranged from 5.6 mg l−1 to 90 mg l−1, with all LOECs less than 90 mg l−1 occurring in B. calyciflorus. The lowest IC1 concentrations were 3.91 mg l−1 for the effect of tetracycline on lifetime reproduction in B. calyciflorus and 4.06 mg l−1 for the effect of tylosin on lifetime reproduction in B. plicatilis. Overall, lifetime reproduction was the most sensitive endpoint and the Malthusian parameter was the least sensitive. We confirmed the existence of growth-promoting substances in the conditioned media (CM) from the rotifer Brachionus plicatilis at an early exponential growth phase and isolated a novel protein with a growth-promoting activity from the crude extract (CE) of rotifer cells. CM was prepared from the culture media where rotifers had been cultured at an early exponential growth phase and filtered through a 0.22-μm filter membrane. The growth-promoting activity was determined using rotifers in CM for 5 days. As a result, the increase of rotifers added with CM was significantly higher than that of the control in artificial seawater (P < 0.001). Moreover, the growth-promoting activity of CM was dose-dependent and inactivated by heat treatment at 80°C for 60 min. Meanwhile, CM filtered through a <10 kDa ultrafiltration membrane showed a low activity, whereas proteinase K treatment resulted in a complete inactivation. These results suggest that the rotifer secrets growth-promoting proteins into CM. CE also contained a protein with the activity and properties similar to those found in CM. Then, CE was subjected to purification of a growth-promoting protein for convenience using various types of chromatography after fractionation with 30–80% saturated ammonium sulfate. Subsequently, a protein with an approximate molecular weight of 25000 was isolated, and its N-terminal amino acid sequence was determined to be PAVVDFTAVWFGPLQMIKP. An orthologue was found in the EST database of B. plicatilis, the full sequence of which showed about 50% identity to the corresponding regions of thioredoxins from other organisms.  相似文献

Population decline of the rotifer Brachionus plicatilis has been evaluated as a sensitive and reliable bioassay for assessing toxicity of marine sediment interstitial water. Three environmental conditions were examined using both interstitial and seawater cultures: (a) adverse effects from dissolved contaminant concentrations; (b) effects from particulate organic matter; and (c) increase or decrease of bacterial biomass from addition of mixtures of antibiotics. Three different parameters were measured to characterize decline: (i) time required for 50% of inoculated population to die (TL50); (ii) curve of the decline rate of Brachionus plicatilis BP and (iii) graphical area contained below plot of the egg:female ratio curve against time (A BPM ).Results indicated that, for each of the different assays, the three parameters of the Brachionus plicatilis population decline test were sensitive to concentrations of contaminants dissolved in interstitial waters (principally: total ammonia, Cu, Cr and alkylbenzenesulphonates) but not to particulate organic matter. Nevertheless, the presence or absence of mixed antibiotics with the contaminants may influence the Brachionus plicatilis population decline test, principally by retarding the hatching of eggs. Based on these results, Brachionus plicatilis is confirmed as an appropriate organism for use as an indicator of interstitial water contamination, using either decline rate, TL50 or both parameters. The presence of particulate matter has no effect on these parameters, but the bacterial population may be an influence, although to a lesser extent than the toxicants.  相似文献

There has been a substantial leap forward in the quantity and quality of genomic resources available for research on rotifers in recent years. We used available genomic and bioinformatics resources to identify variable regions of the genome to design PCR and sequencing primers for the Brachionus plicatilis complex. We then tested their suitability for the study of systematics of this group. Eight amplified successfully for members of the B. plicatilis complex. We sequenced the amplified products, constructed a concatenated alignment (5,511 bp) and carried out phylogenetic analyses. The resulting tree, based on mitochondrial and nuclear genes for 11 clones of six species, was well supported even at relatively deep nodes, contrasting with results of previous studies using only mitochondrial genes, which provide little phylogenetic information for the deepest nodes. The same procedure could be used to design primers for more conserved regions to be used in a wider range of rotifer groups.  相似文献

Experiments were carried out to investigate interspecific interactions between the rotifer Brachionus plicatilis and two harmful algal bloom (HAB) species using single and mixed culture methods. B. plicatilis populations and the growth of two algae were compared at different algal cell densities. The results demonstrate that B. plicatilis obtained sufficient nutrition from Alexandrium tamarense to support net population increase. When exposed to a density of 8 × 104 cells ml−1 A. tamarense, the number of B. plicatilis increased faster than it did when exposed to other four algal densities (16 × 104, 24 × 104, 32 × 104, and 40 × 104 cells ml−1). Cell densities of A. tamarense decreased due to the grazing of B. plicatilis. In contrast, Heterosigma akashiwo had an adverse effect on the B. plicatilis population and its growth was largely unaffected by rotifer grazing. In this case, the B. plicatilis population decreased and H. akashiwo grew at a rate similar to that of a control without addition of rotifers. Mixed culture experiments showed that A. tamarense could partly counteract the effect of H. akashiwo in limiting the rate of population increase of rotifer. In addition, the effect of different initial cell densities on interspecific competition between A. tamarense and H. akashiwo in mixed culture(s) was also investigated. The interaction between sexual reproduction and population growth in the rotifer Brachionus plicatilis was examined using exponential and logistic growth models. A computer simulation was used to explore the effects of the frequency of sex and the proportion of a female's daughters reproducing sexually on population growth rate and resting egg production. Within the parameters of the simulation, the proportion of sexual daughters required for maximum resting egg production increased as the frequency of sex decreased. Adding density-dependence to the model also shifted this optimum towards a higher proportion of sexual daughters. When population growth was unconstrained by density, the optimum proportion of sexual daughters varied between 10–50%, depending on the frequency of sex. This compares to 25–64% when the greatest density restraints were applied at K = 500 females. As the population growth rate (r) increases, the optimum proportion of sexual daughters increases nonlinearly and these effects are exaggerated as sex becomes less frequent. A compilation of published data from laboratory populations of four Brachionus plicatilis strains reveals that the average proportion of sexual daughters in 21% and in close agreement with that predicted by simulation. The limitations of the simulation and its generalizability to other rotifers are discussed.  相似文献

A combination of founder effects and local adaptation – the Monopolization hypothesis – has been proposed to reconcile the strong population differentiation of zooplankton dwelling in ponds and lakes and their high dispersal abilities. The role genetic drift plays in genetic differentiation of zooplankton is well documented, but the impact of natural selection has received less attention. Here, we compare differentiation in neutral genetic markers (FST) and in quantitative traits (QST) in six natural populations of the rotifer Brachionus plicatilis to assess the importance of natural selection in explaining genetic differentiation of life‐history traits. Five life‐history traits were measured in four temperature × salinity combinations in common‐garden experiments. Population differentiation for neutral genetic markers – 11 microsatellite loci – was very high (FST = 0.482). Differentiation in life‐history traits was higher in traits related to sexual reproduction than in those related to asexual reproduction. QST values for diapausing egg production (a trait related to sexual reproduction) were higher than their corresponding FST in some pairs of populations. Our results indicate the importance of divergent natural selection in these populations and suggest local adaptation to the unpredictability of B. plicatilis habitats.  相似文献

The monogonont rotifer Brachionus plicatilis produces resting eggs through sexual reproduction (mixis), which is affected by external and internal factors. We collected resting eggs from rotifers cultured at 15 and 25°C and hatched them with and without 14-day dormancy in the dark. Stem females hatched from both conditions were further cultured at 15, 20 and 25°C. We obtained two phenotypes, one with high mixis (more than 50%), which was hatched from resting eggs formed at 15°C without dormancy, and another in which sexual reproduction occurred at 25°C and resting eggs were formed at 15°C with a 14-day dormancy. In the latter phenotype, mictic females appeared at 15°C, but not at 25°C. Using subtractive hybridization, we isolated one gene from the latter phenotype of females that had no significant similarity to known genes in BLAST searches. We propose that this gene is unique to rotifer mictic reproduction. Ongoing characterization of this gene attempts understand its role in mixis.  相似文献

Pesticides have been major contributors to environmental pollution and they are now widely distributed in aquatic environments. Zooplankters are frequently used as test animals to detect aquatic contaminants because of their sensitivity and ecological importance. We investigated the effect of a 7-day exposure to four commonly used pesticides (diazinon, fenitrothion, methoprene and isoprothiolane) on reproduction of the rotifer Brachionus plicatilis. Pesticide concentrations of 3–7 times lower than the 24-h 50% lethal concentration (24-h LC50) were tested to determine the ‚no observed effect’ concentration (NOEC), ‚lowest observed effect’ concentration (LOEC), and the ‚50% effective’ concentration (EC50) on specific growth rate (r), sexual reproduction, fertilization, resting egg production, and hatchability of resting eggs. Results showed that the lowest EC50 value of r, mixis, fertilization, and resting egg production of 1.4 μM for diazinon was 63 times lower than its 24-h LC50 of 88.4 μM, while for fenitrothion it was 66 times (3.5 and 229.8 μM, respectively). For isoprothiolane, the lowest EC50 value of r, mixis, fertilization, and resting egg production of 8.9 μM was 25 times lower than its 24-h LC50 of 220.7 μM, while for methoprene was 37 times (2.7 and 100.8 μM, respectively). In all pesticides, hatching rate of resting eggs consistently gave the lowest EC50 values which is about 2–40 times lower than the lowest EC50 of r, mixis, fertilization, and resting egg production. Hatchability of resting eggs therefore is the most sensitive parameter in detecting effects of pesticides exposure in rotifer B. plicatilis.  相似文献

The mate recognition protein (MRP) gene is a member of a family of extracellular matrix protein genes, called MRP Motif Repeat (MMR) genes, with no known homologs. Two sets of MMR genes, designated MMR-A and MMR-B, were found in Brachionus manjavacas. MMR-B has previously been shown to encode the MRP in the Brachionus plicatilis species complex. MMR family genes share the same basic structure: a signal peptide sequence, followed by nearly identical 276 bp (MMR-A) or 261 bp (MMR-B) repeats, with a truncated final repeat. Each repeat of the predicted MMR-A and -B proteins is expected to have a secondary structure of 5 α-helices, ranging in length from 11 to 20 amino acids, separated by coils of 1–3 amino acids. Hydrophobic and hydrophilic amino acids are predicted to be partitioned to opposite sides of each α-helix, suggesting that MMR proteins are globular with a hydrophobic core. MMR-A and MMR-B proteins vary in their post-translational modifications, resulting in differences in size and charge, and likely causing differences in the physical properties of the proteins on the surface of the female, and their ability to be recognized by a receptor on a male rotifer. The identity of MMR gene repeats is theorized to be maintained by concerted evolution, through a process of unequal crossing over and/or gene conversion, with new mutations likely to be lost. Rarely, however, the same process of concerted evolution can rapidly spread a mutation across all of the repeats. When a mutation results in conformational changes in the protein detectable by males, it could lead to reproductive isolation and thereby to speciation. Thus, changes in MRP could be a driving force in the high degree of species diversity seen within the B. plicatilis cryptic species complex.  相似文献

Deductions from both evolutionary models and inductive argumentation from empirical data support the notion of intraspecific variability for the initiation of sexual reproduction (mixis) within rotifer populations. In this study, we focus on the time and density at which mixis is initiated in a growing population. Cyclical parthenogenetic clones of Brachionus plicatilis established by hatching of resting eggs, isolated from a natural habitat, have been tested at the start of their sexual phase. Clones exhibited great variation for this trait, their time of switching to sexual reproduction being correlated with population density. Most of the variation for mixis initiation has either low or no heritability and is caused by individual environmental factors.  相似文献

We investigated the nutritional effects of both food quantity and quality on Brachionus plicatilis. Decomposition of particulate and dissolved organic matter by rotifer digestive enzymes play a crucial role in rotifer nutrition. Among other enzymes, rotifers produce phosphatases, non‐specific enzymes that allow for the release of orthophosphate from a variety of organic phosphorus compounds. Phosphatase saturation was measured in B. plicatilis homogenates using the spectrofluorimetric method. We examined population growth rate, reproduction and phosphatase activity in the homogenate of rotifers (PARH) fed by nutrient‐replete algal food supplied at different quantities. Population growth rate, number of eggs per individual and PARH were affected by food quantity. Growth rate and number of eggs per individual significantly increased in rotifers fed by food supplied at the highest quantity. The highest population growth rate was reached by rotifers fed by nutrient‐replete food, while it did not significantly differ between rotifers fed on nitrogen (N)‐depleted and phosphorus (P)‐depleted food. The number of eggs per individual was more affected by N than P supply. PARH and rotifer RNA content were not influenced by different food quality. The results indicate that B. plicatilis is not able to regulate its digestive apparatus in terms of efficiently getting access to essential nutrients when scarce, but do this when nutrient‐replete food is available in different quantity. We quantified the predation of Acanthocyclops americanus from the shallow Mediterranean lake Albufera, using gut contents from field collections and laboratory feeding tests. For functional response studies, we used Brachionus plicatilis (at 6 concentrations, 400–4000 ind. 40 ml−1) and Diaphanosoma mongolianum (at 2–20 ind. 40 ml−1). Copepod feeding rates were also estimated using different proportions of rotifer prey and lake seston (0–67.5% of seston + 40 individuals of B. plicatilis). Prey selection studies were conducted using five zooplankton species: Brachionus angularis, Brachionus plicatilis, Keratella tropica, Daphnia magna and Diaphanosoma mongolianum. Gut contents of field-collected adult Acanthocyclops contained filamentous algae and cyanobacteria and 16 zooplankton species (Keratella cochlearis, unspined and spined forms, K. tropica, Brachionus plicatilis, Brachionus calyciflorus, Brachionus angularis, Brachionus variabilis, Asplanchna girodi, Polyarthra vulgaris, Synchaeta pectinata, Lepadella rhomboides, unidentified bdelloids, Alona rectangula, Chydorus sphaericus, Bosmina longirostris, D. magna, Ceriodaphnia dubia and copepod nauplii). When fed B. plicatilis or D. mongolianum, female A. americanus had higher prey consumption rates than males. Increased proportion of lake seston caused reduced consumption of brachionid prey. Our data suggest that A. americanus is omnivorous in nature.

A model of nitrogen flow is presented through the Brachionus plicatilis/Chlorella saccharophila mass (batch) culture system, from the initial input of inorganic nitrogen to the algal culture medium to the final production of rotifers, dissolved nitrogen and particulate nitrogen.A nitrogen budget was first formulated for B. plicatilis relating ingestion, excretion, egestion, somatic growth and reproductive growth. Measurements were made on rotifers from 20° and 10° cultures.The calculated model of nitrogen flow through the rotifer/algal batch culture system estimates the percentages of the original input nitrogen which will be incorporated into algal nitrogen, rotifer nitrogen and the particulate and dissolved nitrogen pools. It is suggested that the dissolved nitrogen pool could be recycled directly for use in subsequent algal culture.  相似文献

Population growth parameters in batch culture of Brachionus plicatilis under a continuous supply of freeze-dried microalgae powder have been determined. Two B. plicatilis strains (L- and S-types) and four microalgae species (Nannochloropsis oculata, Nannochloropsis gaditana, Nannochloris oculata and Tetraselmis suecica) have been tested, establishing the dynamics of growth at different daily food rations. Cultures showed a short lag phase, an exponential growth phase, a long post-exponential growth phase and long decline with episodic increases. In both rotifer strains, the best growth was obtained with Nannochloropsis oculata and the poorest with Nannochloris oculata.  相似文献

Superoxide dismutases (SODs) promote a conversion of harmful reactive oxygen species (ROS) to relatively moderate forms, resulting in the extension of lifespan in the nematode Caenorhabditis elegans under caloric restriction. The lifespan of the rotifer Brachionus plicatilis is also markedly extended by caloric restriction. We, therefore, cloned cDNA encoding SOD activated with Mn (Mn SOD) from B. plicatilis and examined its expression pattern in rotifers raised with energy restricted diet. The full length deduced amino acid sequence of the rotifer Mn SOD showed 61% identity with the C. elegans ortholog. Four amino acid residues that are essential to the binding of this enzyme to Mn were conserved in the rotifer Mn SOD. Subsequently we examined the mRNA expression patterns of Mn SOD using highly sensitive quantitative real-time PCR for various rotifer populations that are likely to differ in their lifespans in experiments on calorie restricted diets. The accumulated mRNA levels of Mn SOD were found to increase in supposedly long-lived rotifers. These results suggest that Mn SOD is possibly related to the aging of B. plicatilis.  相似文献
