Powdery mildew is one of the most important limiting factors for cucurbits production in Spain, its management being strongly dependent on chemicals. The aim of this work was to evaluate the possibility of exploiting antagonistic bacteria in the biological control of the cucurbit powdery mildew fungus Podosphaera fusca (syn. Sphaerotheca fusca). Among a collection of bacterial strains isolated from distinct cucurbit powdery mildew diseased plants and rhizospheric soils, four isolates were selected, by means of a screening method based on antibiotic production, and identified as Bacillus spp. These isolates proved to be efficacious in the control of cucurbit powdery mildew in in vitro detached leaves and seedling biocontrol assays, where reductions of disease severity of up to 80% were obtained. Furthermore, bacterial populations on melon leaves remained at similar levels (105 cfu cm–2) over the 16-day period studied and, as observed by scanning electron microscopy analysis, they were able to establish microcolonies associated with an extracellular matrix, which reveals that these isolates efficiently colonize melon phylloplane. These results indicate that the bacterial isolates selected are promising candidates for biological control agents of cucurbit powdery mildew in southern Spain.  相似文献

The stem borer, Busseola fusca (Fuller), is an important pest of maize Zea mays L. and sorghum Sorghum bicolor (L.) in eastern and southern Africa. To control this pest, biological control methods including the use of entomopathogenic fungi are being considered. The pathogenicity of one isolate of Metarhizium anisopliae (Metsch.) Sorok. and one isolate of Beauveria bassiana Bals. (Vuill.) were first tested on different developmental stages of B. fusca including eggs, neonate, 2nd and 3rd-instar larvae. Both fungal isolates were pathogenic to all the stages tested. However, differences in mortality were observed among larvae that hatched from treated egg masses. Experiments were conducted thereafter to test whether B. fusca males could serve as a vector for fungal conidia to contaminate B. fusca females and subsequently eggs and larvae. Results demonstrated that B. fusca males successfully transferred inoculum to females during copulation, which in turn transmitted it to the eggs they laid on maize plants, resulting in the decrease of leaf damages.  相似文献

Phytoviruses including tospoviruses are known to affect the behavior and fitness of their vectors both positively and negatively. In this study, we investigated the effects of Tomato spotted wilt virus (TSWV) (family Bunyaviridae, genus Tospovirus) infection on the fitness and feeding ability of tobacco thrips, Frankliniella fusca (Hinds) (Thysanoptera: Thripidae) using peanut, Arachis hypogaea L. (Fabaceae), as a host. Potentially viruliferous F. fusca laid more eggs than non‐viruliferous F. fusca. In contrast, fewer potentially viruliferous F. fusca developed into adults and required a longer developmental time than non‐viruliferous F. fusca, indicating a direct negative effect of the virus on thrips fitness. In addition, no‐choice feeding tests indicated that non‐viruliferous F. fusca fed more rapidly than potentially viruliferous F. fusca. Typically, phytovirus infections are known to enhance the availability of vital nutrients such as free amino acids in infected host plants and to affect other important physiological processes negatively. Free amino acids are known to play a vital role in egg production and development. Further investigations in this study revealed that leaflets of infected plants had ca. 15 times more free amino acids than non‐infected leaflets. TSWV‐infected leaflets were used to rear potentially viruliferous thrips. Higher amino acid levels in TSWV‐infected leaflets than in non‐infected leaflets could have contributed to increased oviposition by potentially viruliferous F. fusca compared to non‐viruliferous F. fusca. Taken together, these results suggest that increased concentrations of free amino acids in TSWV‐infected plants might serve as an incentive for thrips feeding on otherwise unsuitable hosts, thereby facilitating TSWV acquisition and transmission.  相似文献

The powdery mildew fungus Podosphaera fusca (synonym Podosphaera xanthii) is the main causal agent of cucurbit powdery mildew and one of the most important limiting factors for cucurbit production worldwide. Despite the fungus' economic importance, very little is known about the physiological and molecular processes involved in P. fusca biology and pathogenesis. In this study, we isolated and characterised the β-tubulin-encoding gene of P. fusca (PfTUB2) to develop molecular tools with different applications in powdery mildew research. PfTUB2 is predicted to encode a protein of 447 amino acid residues. The coding region is interrupted by six introns that occur at approximately the same positions as the introns present in other fungal TUB2-like genes. Once cloned, the PfTUB2 sequence information was used in different applications. Our results showed that the TUB2 gene is a good marker for molecular phylogenetics in powdery mildew fungi but it is unsuitable for the analysis of intraspecific diversity in P. fusca. The expression of PfTUB2 was proven to be stable in different temperature conditions, supporting its use as a reference gene in quantitative gene expression studies. Furthermore, an allele-specific PCR assay for the detection of resistance to methyl-2-benzimidazole carbamate (MBC) fungicides in P. fusca was developed based on the correlation between the single amino acid change E198A in β-tubulin and the MBC resistance phenotype. Lastly, PfTUB2 was used as a target gene in the development of a high-throughput method to quantify fungal growth in plant tissues.  相似文献

Wind tunnel and Y‐tube olfactometer studies are useful for demonstrating the responsiveness of male moths to sex pheromones. However, in the cereal stem borer, Busseola fusca (Fuller) (Lepidoptera: Noctuidae), wind tunnel bioassays yielded poor results when the behavioural responses of females to plant odours were tested. We demonstrated that for B. fusca females, the Y‐tube olfactometer was better suited for measuring the attractiveness of plant odours, compared with the wind tunnel. In particular, we showed B. fusca preference for odours of a host over a non‐host plant species using this apparatus. Behavioural responses for B. fusca after host finding are also discussed.  相似文献

I observed 22 species of birds visiting flowering Erythrina fusca trees at Matamatá, Amazonas, Colombia. The large orange flowers of E. fusca are adapted for pollination by birds and are protected from illegitimate visits by a petal that covers the nectaries and anthers until displaced by a foraging bird. Experiments with flowers bagged to exclude potential pollinators demonstrated that the flowers do not open without assistance. At Matamatá, parrots are the most frequent visitors to flowering E. fusca, and two species, Dusky‐headed Parakeet (Aratinga weddellii) and Cobalt‐winged Parakeet (Brotogeris cyanoptera), appear to be the main pollinators. This is only the fourth report of Neotropical parrots acting as pollinating agents. At least five other parrot species fed on the nectar or flowers of E. fusca but destroyed the flowers in the process. Orange‐backed Troupials (Icterus jamacaii) were the only other species observed opening E. fusca flowers nondestructively and are likely to be pollinators. Hummingbirds were common visitors to E. fusca flowers and some species were found to carry E. fusca pollen; however, hummingbirds were unable to open the flowers themselves and relied on other visitors to open the flowers for them. The number of hummingbird visits to a flowering E. fusca tree was positively correlated with the number of visits by parrots and icterids, but not with the number of mature flowers, indicating that legitimate visitors facilitate access by hummingbirds.  相似文献

Myxogastria is a group of protozoa characterized by cellular uninucleate amoeboflagellates (myxamoebae and flagellated swarm cell), acellular multinucleate plasmodia, and stationary spore‐bearing sporocarps. The Stemonitales is a large order in the Myxogastria and contains approximately 230 species, but only 13 species have their completed life cycles observed so far. Here, we described the life cycles of two species in Stemonitales, Stemonitopsis typhina and Stemonitis fusca by culturing in water agar medium and observing the morphogenesis of their spore germination, plasmodium, and sporocarp development. The spore‐to‐spore life cycles of Ste. typhina and S. fusca were completed in approximately 67 and 12 d, respectively. Both species possessed an aphanoplasmodium. However, the spores of Ste. typhina and S. fusca germinated by the V‐shape split and pore methods, respectively. Unlike S. fusca with an evanescent peridium, Ste. typhina produced a shiny persistent peridium which was continuous with the membrane surrounding its stalk. The information will contribute to a better understanding of their taxonomy and phylogeny.  相似文献

Malic acid is mainly used as an acidulant and taste enhancer in the beverage and food industry. Previously, a mutant strain Thermobifida fusca muC, obtained by adaptive evolution was found to accumulate malic acid on cellulose with low yield. In this study, the malic acid synthesis pathway in T. fusca muC was confirmed to be from phosphoenolpyruvate to oxaloacetate, followed by reduction of oxaloacetate to malate. To increase the yield of malic acid by the muC strain significantly, the carbon flux from pyruvate was redirected to oxaloacetate by expressing an exogenous pyruvate carboxylase (PCx) gene from Corynebacterium glutamicum ATCC 13032 in the chromosome of T. fusca muC‐16. The yield of malic acid in the engineered strain muC‐16 was increased by 47.9% compared to the parent strain muC. The muC‐16 strain was then grown on ~100 g/L cellulose and the highest titer of malic acid was 62.76 g/L by batch fermentation. T. fusca muC‐16 strain converted milled corn stover to malic acid with the highest titer of 21.47 g/L with minimal treatment. © 2016 American Institute of Chemical Engineers Biotechnol. Lepidopteran stemborers are the most destructive insect pests of cereal crops in sub‐Saharan Africa. In nature, these insects are often exposed to multiple environmental stressors, resulting in potent impact on their thermal tolerance. Such environmental stressors may influence their activity, survival, abundance and biogeography. In the present study, we investigate the effects of acclimation to temperature, starvation and desiccation on thermal tolerance, measured as critical thermal limits [critical thermal minima (CTmin) and maxima (CTmax)] on laboratory‐reared economic pest species Chilo partellus Swinhoe (Lepidoptera: Crambidae), Busseola fusca (Fuller) and Sesamia calamistis Hampson (Lepidoptera: Noctuidae) using established protocols. Low temperature acclimation results in improved CTmin for B. fusca and C. partellus, whereas high temperature acclimation enhances the same trait for B. fusca and S. calamistis. Similarly, high temperature and starvation pretreatment improve CTmax for C. partellus relative to S. calamistis and B. fusca. In addition, starvation and desiccation pretreatments improve CTmin for all stemborer species. Furthermore, rapid cold‐hardening (RCH) enhancs CTmin for B. fusca and C. partellus, whereas rapid heat‐hardening (RHH) improves the same trait for C. partellus. However, RCH and RHH impair CTmax for all stemborer species. These findings show differential thermal tolerances after exposure to heterogeneous environmental stress habitats. Chilo partellus, of exotic origin, shows a higher magnitude of basal thermal tolerance plasticity relative to the indigenous African species S. calamistis and B. fusca. This indicates that C. partellus may have a fitness and survival advantage under climate‐induced heterogeneous environments, and also have a greater chance for geographical range expansion and invasion success compared with the indigenous B. fusca and S. calamistis.  相似文献

Aphis gossypii Glover shows obvious host specialization, with cucurbit‐ and cotton‐specialized biotypes or host races in many regions. Because its annual natal host crops senesce earlier the cucurbit‐specialized biotype may suffer food deficiency. The method this biotype uses to overcome this challenge is still poorly understood. In order to understand the potential of the cucurbit‐specialized biotype aphids in host shift and usage, the performance of this biotype on cotton (Gossypium hirsutum), a common but poor quality host plant, was explored in this study. The cucurbit‐specialized aphids could establish populations on cotton only when these plants had at least nine leaves, and subsequent populations developed rather slowly. The presence of whitefly populations on cotton improved the success rate of cucurbit‐specialized aphids. The cucurbit‐specialized aphids were mainly distributed on the older leaves of cotton, with only a few settling on the upper leaves. The cucurbit‐specialized aphids reared on cotton for 40, 54 and 61 days still maintained strong preference for their natal host plant, cucumber (Cucumis sativus), rather than cotton, and their net reproductive rates and intrinsic rates of natural increase were dramatically lower when they were transferred onto new six‐leaf cotton plants or detached leaves. Therefore, we concluded that the cucurbit‐specialized aphids have the potential to utilize mature or whitefly‐stressed cotton plants, but that this feeding experience on cotton did not alter their specialization for cucurbits. Some cotton plants could act as a temporary host for the cucurbit‐specialized aphids to overcome food deficiency arising from senescing cucurbits.  相似文献

Tree pollen, especially Pinus spp. (Pinaceae), is shed in large quantities every spring in North America. Pine pollen deposition onto leaves was found to significantly influence the ovipositional behaviors of certain thrips species (Thysanoptera: Thripidae) in peanut and tomato leaf choice and no‐choice tests. Pine pollen (Pinus elliottii Engelm.) increased the oviposition rate 2.9‐fold for Frankliniella occidentalis (Pergande) (western flower thrips) and 1.6‐fold for Frankliniella fusca (Hinds) (tobacco thrips) in choice tests averaged over both plant species. These results support the idea that pollen has a greater impact on F. occidentalis behavior than on F. fusca behavior. The most dramatic increase was in peanut, where F. occidentalis only oviposited on leaves dusted with pollen, suggesting that the addition of pollen stimulated this flower thrips to lay eggs on a poor host‐plant part. The impact of pollen on the rate of oviposition by thrips is important because it is the early‐instar nymphs that acquire tomato spotted wilt virus (TSWV), which these two thrips species vector. In a laboratory bioassay, the addition of pine pollen to TSWV‐infected peanut foliage increased the percentage of infected F. fusca after one generation.  相似文献

The objective of the present work was to develop a method for the preservation of T. harzianum conidia at room temperature and the immobilised conidia propagation in submerged culture. This was accomplished by immobilising the strain in sodium alginate capsules (white capsules) and subsequently propagating them in a column bubble reactor (green capsules). Three capsule diameters were tested (micro, medium and large capsules), which were produced by emulsion internal gelation and dripping methods. Tested variables were the immobilised conidia propagation in submerged culture for free conidia production, the immobilised conidia viability throughout the time (two years), the resistance of the encapsulated conidia to the UV irradiation of short and long wavelength, and the antagonistic effect of the encapsulated T. harzianum against four phytopathogenic fungi. It was found that the medium capsules (1.5?±?0.3?mm) favoured the massive production of released conidia in submerged culture and that the higher the density of conidia per capsule, the greater the protection against the ultraviolet irradiation. Regarding the conidia preservation in calcium alginate, a viability loss of around 30% was observed two years after storage at environmental temperature in both white and green capsules; along the two years that the viability of conidia was analyzed, the purity of the formulation was corroborated. The results presented here show the efficacy of the green and white capsules for T. harzianum preservation at room temperature for a long period of time.  相似文献

The present study was aimed at characterizing differences in mate acceptance and host plant recognition between Busseola fusca (Lep.: Noctuidae) reared for several generations under laboratory conditions and wild conspecifics, directly collected from maize stems in the field. The mating success was significantly higher in laboratory reared when compared with the wild B. fusca population. Oviposition on artificial stems was significantly higher for laboratory‐reared insects than for the wild ones. Moreover, unlike adults of the wild strain, laboratory‐reared B. fusca showed no preference to oviposit on surrogate stems impregnated with maize extracts. Long‐range attraction to the host plant was significantly lower for laboratory‐reared insects. Furthermore, the polymerase chain reaction‐restriction fragment length polymorphism (PCR‐RFLP) analysis of mitochondrial DNA showed that the wild type and laboratory populations belonged to the same genetic strain, indicating that these differences between the populations were mainly phenotypic.  相似文献

Brain investment is evolutionarily constrained by high costs of neural tissue. Several ecological factors favour the evolution of increased brain investment; we predict reduced brain region investment will accompany the evolution of organismal or social parasitism when parasites rely on host behaviour and cognition to solve ecological problems. To test this idea we investigated whether brain region investments differed between obligate slave‐making Polyergus mexicanus ant workers and their Formica fusca slave workers. Polyergus workers perform little labour for their colonies; enslaved workers of Formica host species forage, excavate nests and tend the brood. We focused on the calyces of the mushroom bodies, central processing brain regions that are larger in social insect workers that perform complex tasks. As predicted we found lower relative investment in mushroom body calyx in P. mexicanus workers than in F. fusca workers; by contrast, enslaved and free F. fusca workers did not differ in mushroom body calyx volume. We then tested whether slave‐makers and hosts differed in brain investment among sensory modalities. Polyergus slave‐makers employ several unique classes of pheromones during raids, and eye size relative to head size was smaller in P. mexicanus workers than in F. fusca workers. The size of antennal brain tissues relative to visual tissues was greater in Polyergus, both in the peripheral sensory lobes and in the mushroom body calyx, suggesting greater relative investment in antennal processing by slave‐makers. © 2014 The Linnean Society of London, Biological Journal of the Linnean Society, 2014, 113 , 415–422.  相似文献

1. Full sunlight conditions in open clear‐cuts may limit the activity of ants as soil surface temperatures reach lethal levels. Therefore, differences may be expected between the diurnal and nocturnal activity of ants, and in the interactions between ant species. These predictions, however, have been poorly investigated so far. 2. The circadian activity of ants in clear‐cuts in managed forests in Poland was investigated. Repeated counts of ants were performed during the day and the following night at the clear‐cut edge and in the clear‐cut interior. Interspecific interactions and the effect of plant coverage were also considered. 3. Abundances of Formica fusca Linnaeus and red wood ants were higher during the day, whereas Myrmica were more common at night. Formica fusca, Lasius and red wood ants were more common at the clear‐cut edge than in the interior. Myrmica showed the opposite pattern, but at night, its numbers increased at the edge. Plant coverage positively affected F. fusca and red wood ants. 4. Red wood ants tended to be negatively associated with Lasius, whereas they were neutral for F. fusca. The negative association of red wood ants and Myrmica was stronger during the day compared to night. 5. The time of day was a strong driver of ant activity in the clear‐cuts, whereas the distribution of red wood ants was of lesser importance. It is concluded that circadian activity may substantially contribute to niche separation between coexisting species, therefore, studies performed exclusively during the day cannot reflect the real structure of the community.  相似文献

Nests of social insects are an attractive resource in terms of nutrition and shelter and therefore targeted by a variety of pathogens and parasites that harness the resources of a host colony in their own reproductive interests. Colonies of the ants Formica fusca and F. lemani serve as hosts for mound‐building Formica species, the queens of which use host colonies during colony founding. Here, we investigate whether workers of the host species can mitigate the costs imposed on them by invading parasite queens by recognizing and selectively removing eggs laid by these queens. We used behavioural assays, allowing host workers to choose between con‐colonial eggs and eggs laid by the parasite species F. truncorum. We show that workers of both host species discriminate between the two types of eggs in favour of con‐colonial eggs. Moreover, workers of F. fusca rejected more con‐colonial eggs than F. lemani. This higher rate of error in F. fusca may reflect a greater selectivity or a greater difficulty in discriminating between the two egg types. Nevertheless, both host species removed parasite eggs at a similar rate, when these were artificially introduced into the colonies, although some eggs remained after 10 d. In addition, upon receiving parasite eggs, host workers started to lay unfertilized male‐destined eggs within 6 d, thus employing an alternative pathway to gain direct fitness when the resident queen is no longer present and the colony is parasitized.  相似文献

The African stem borer, Busseola fusca (Fuller) (Lepidoptera: Noctuidae), is an important pest of maize and sorghum in sub‐Saharan Africa. As in many other lepidopteran insects, the ability of B. fusca to recognize and colonize a variety of plants is based on the interaction between its sensory systems and the physical and chemical characteristics of its immediate environment. In this study, we tried to identify the behavioural steps of B. fusca leading to host selection and oviposition. Three Poaceae species commonly cultivated in Kenya for human consumption and animal forage were used in this study: the two most preferred hosts, maize (Zea mays L.) and sorghum [Sorghum bicolor (L.) Moench], and one non‐preferred host, Napier grass (Pennisetum purpureum Schumach). Wind tunnel observations revealed that volatiles produced by the different plant species did not appear to strongly influence the general orientation of B. fusca towards the plant, as similar behavioural steps were exhibited by the female moth regardless of the plant species involved. This indicated that the females were not able to recognize their preferred hosts from a distance. After landing, the female typically swept her ovipositor on the plant surface, simultaneously touching it with the tips of her antennae, and then oviposited. This behaviour was more frequently observed on maize and sorghum than on Napier grass, and indicated that both antennal and ovipositor receptors are used by the female moths to evaluate the plant surface before deciding to oviposit. As a result, the females laid more eggs on the two crops than on Napier grass. We conclude therefore that females recognized their preferred hosts only after landing. Tactile and contact‐chemoreception stimuli from the plants seemed to play a major role in oviposition decisions of B. fusca.  相似文献

The noctuid stem borer Busseola fusca occurs throughout sub‐Saharan Africa, where it is considered as a major pest of corn and sorghum. To understand B. fusca phylogeography, we isolated and characterized eight microsatellite loci in this species. We investigated these loci for polymorphism on a subset sample of individuals from different geographic populations. All loci were polymorphic, showing between four and 10 alleles. Cross‐species amplification of four of these loci was tested on 11 other noctuid species.  相似文献

Collaborative research was conducted at the USDA-ARS Subtropical Agricultural Research Center in southern Texas to assess the microbial control potential of Beauveria bassiana and Paecilomyces fumosoroseus against Bemisia whiteflies. Laboratory assays demonstrated the capacity of both pathogens to infect Bemisia argentifolii nymphs on excised hibiscus leaves incubated at relative humidities as low as 25% at 23 ± 2°C (ca. 35% infection by B. bassiana and P. fumosoroseus resulted from applications of 0.6–1.4 × 103 conidia/mm2 of leaf surface). In small-scale field trials using portable air-assist sprayers, applications at a high rate of 5 × 1013 conidia in 180 liters water/ha produced conidial densities of ca. 1–2.5 × 103 conidia/mm2 on the lower surfaces of cucurbit leaves. Multiple applications of one isolate of P. fumosoroseus and four isolates of B. bassiana made at this rate at 4- to 5-day intervals provided >90% control of large (third- and fourth-instar) nymphs on cucumbers and cantaloupe melons. The same rate applied at 7-day intervals also provided >90% control in zucchini squash, and a one-fourth rate (1.25 × 1013 conidia/ha) applied at 4- to 5-day intervals reduced numbers of large nymphs by >85% in cantaloupe melons. In contrast to the high efficacy of the fungal applications against nymphs, effects against adult whiteflies were minimal. The results indicated that both B. bassiana and P. fumosoroseus have strong potential for microbial control of nymphal whiteflies infesting cucurbit crops.  相似文献
