对绿色木霉接种到啤酒糟固态发酵产纤维素酶的培养基和培养条件进行优化,考察发酵物料起始含水量、发酵时间、起始pH值等发酵条件,以及啤酒糟培养基中添加麸皮、氮源种类对产酶的影响。结果表明,以啤酒糟为发酵基质接种绿色木霉生产纤维素酶是可行的。经单因素和正交试验获得最适固态发酵的培养条件为:起始pH 5~6,培养温度28~30℃,发酵4 d;最佳发酵培养基组合为:麸皮比例30%,培养基起始含水量50%,(NH4)2SO4添加量为2.0%~2.5%。  相似文献

探讨了嗜热真菌Chaetomium thermophile产生纤维素酶的液体发酵条件及滤纸酶(FPA)的特性。采用液体发酵培养法,通过对碳源、氮源、培养时间、培养基的起始pH值及产酶过程中pH值和蛋白质含量变化的研究发现:在2%纤维素、1%可溶性淀粉为碳源;2.0%KNO3 0.2%酵母粉为氮源;起始pH值为6.5,50℃下培养9d后,各种酶活最高。发酵过程中,pH值和蛋白质的含量均在前3d下降,后升高。FPA的反应最适温度和pH值分别为60℃和5.5~6.0;且具有较高的热稳定性和DH稳定性。  相似文献

本对嗜线虫致病杆菌(Xenorhadbus nematophilus)产生抗生素的发酵培养基和发酵条件进行了研究,同时对该菌代谢过程pH值、还原糖、总糖、氨基氮与抗生素产量的关系进行了分析,通过筛选该菌对碳源和氮源的要求,用正交试验初步确定了该菌产素的最佳发酵培养基和条件为:玉米粉1%,大豆粉3%,蔗糖1%,蛋白胨1.5%,KH2PO40.02%,MgSO40.2%,活化剂T0.1%;发酵培养基的起始pH值在6.0-8.0,种龄16h,接种量4%,500mL摇瓶装量15-150mL的条件下培养72h可获得较高的抗生素产量;产素量与菌代谢过程中pH、还原糖、总糖和氨基氮的变化有一定关系,通过培养基和培养条件的研究使该菌的产抗生素能力提高了56.3%。  相似文献

以亚硒酸钠为无机硒源,研究硒的加入对荷叶离褶伞菌丝体胞外粗多糖的影响并优化其发酵条件,选择培养基灭菌条件、添加硒浓度、发酵时间和初始pH值作为优化因素,研究硒在各因素不同水平下对荷叶离褶伞胞外粗多糖含量的影响。通过单因素试验和响应曲面法得到最佳发酵条件:在121℃,20 min对培养基进行灭菌,发酵液中添加2μg/m L Na_2SeO_3,发酵244 h,培养基初始pH值为7.0的条件下,发酵产生的胞外粗多糖含量为0.760 g/100 m L,较未加入硒(0.218 g/100 m L)和加硒未优化前的含量(0.755 g/100 m L)有所提高。研究结果表明,响应曲面法优化富硒荷叶离褶伞发酵条件可使胞外粗多糖含量大幅度提高。  相似文献

对戊糖乳杆菌31-1产细菌素的条件进行了优化,分别研究了培养温度,培养基起始pH值,培养基碳源、氮源,刺激因子等因素对细菌素产量的影响。组合因素优化结果得到最佳培养基与培养条件为:乳糖30g、胰胨15g、豆胨20g、牛肉膏30g、蛋白胨20g、吐温80 1mL、磷酸氢二钾2g、乙酸钠5g、柠檬酸铵2g、硫酸镁0.58g、硫酸锰0.25g,蒸馏水定容至1000mL,30℃培养24h,培养起始pH为6.5。在此条件下培养细菌素效价可达到640AU/mL,与起始培养基相比细菌素产量提高了8倍。  相似文献

[目的]通过优化内生真菌枝状枝孢霉MD2的发酵条件,提高10-去乙酰巴卡亭Ⅲ (10-DAB)和紫杉醇(Taxol)的产量.[方法]采用单因素试验分析不同的培养基初始pH值、培养温度、摇床转速和培养时间对10-DAB和紫杉醇产量的影响,优化枝状枝孢霉MD2的培养条件;以YES为基本培养基,采用单因素试验和正交试验分析添加苯甲酸钠、苯丙氨酸、丝氨酸和甘氨酸4种前体物对10-DAB和紫杉醇产量的影响,优化枝状枝孢霉MD2的培养基组分.[结果]优化后发酵条件为:在初始pH为5.0的300 mL YES培养基中,添加15 mg/L苯甲酸钠、25 mg/L苯丙氨酸、5 mg/L丝氨酸、15 mg/L甘氨酸,接种1 mL枝状枝孢霉MD2的孢子悬液(107-10s个孢子/mL),28.0℃、220 r/min发酵培养12d.在此条件下,枝状枝孢霉MD2的生物量、10-DAB和紫杉醇的产量分别为15.5 g/L、471.5 μg/L和569.5 μg/L,与初始发酵条件相比,分别提高了1.3、3.6和3.4倍.[结论]首次获得了枝状枝孢霉MD2生产10-DAB和紫杉醇的较适摇瓶发酵条件,可为进一步放大发酵培养提供参考.  相似文献

从5龄家蚕肠道分离筛选得到一株产纤维素酶菌株BMC-2,以羧甲基纤维素酶(CMCase)比活力为响应值,通过Plackett-Burman试验设计、最陡爬坡试验和Box-Behnken试验设计对菌株BMC-2产纤维素酶发酵条件进行优化,结果表明,发酵时间、发酵温度、培养基初始pH和转速对CMCase比活力具有显著影响,其影响程度由大到小依次为发酵时间、培养基初始pH、发酵温度、转速.确定菌株BMC-2产纤维素酶最优发酵条件为:发酵时间94.35 h,发酵温度30.3℃,培养基初始pH 7.01,转速179 r/min.在此条件下, CMCase比活力理论值为25.801 U/mg,验证值为25.526 U/mg,较产酶条件优化前提高了1倍,预测模型可靠性高,可应用于菌株BMC-2产纤维素酶条件的优化.  相似文献

对前期筛选得到的在田间试验中防治根结线虫效果较好的蜡质芽孢杆菌(Bacillus cereus)AR156,通过单因素筛选及正交试验的方法进行了发酵培养基优化,得到的最佳配比为:麦芽糖0.25%,玉米粉0.5%,黄豆粉0.5%,胰蛋白胨0.5%,CaCl2·2H2O0.05%,MnSO4·H2O0.05%,K2HPO40.1%。同时对实验室摇瓶条件下液体发酵的主要影响因子温度、转速、初始pH值等进行实验探讨,确定了最佳培养条件:初始pH值7.0,装液量200mL/L,接种量5%,发酵温度28℃,转速200r/min,发酵时间48h。优化后芽孢产量为1.03×109CFU/mL,芽孢生成率在97%以上,明显高于初始发酵培养基发酵结果。  相似文献

α-乙酸乳酸脱羧酶(α-acetolactatedecarboxylase,简称ALDC,EC,4.1.1.5)用于啤酒生产中,可将啤酒酵母在主发酵期间形成的α-乙酰乳酸直接转化为3-羟基丁醇,由此可大大缩短啤酒熟化期,从而提高设备利用率,增加产值.我们通过反复诱变已筛选到一株ALDC高产菌株(编号为ZIT-6-1-2)。为了进一步提高酶产量,对该菌株进行了发酵条件的优化组合研究.通过对发酵培养基组分、碳氮比、培养基起始pH值、通气量、培养温度、培养时间和微量元素等进行了一系列研究.结果表明上述诸因素对该ALDC高产菌株的酶产量均有不同程度的…  相似文献

Statistical experimental design was used to optimize medium constituents and the conditions of fermentation, viz., temperature, pH?and the time of fermentation. Higher yields of L-glutamic acid (37.1?kg/m3) was obtained after optimizing medium components and the conditions of fermentation. The optimal levels of medium components were: 61.5575?kg/m3 glucose, 7.3272?kg/m3 urea and 1.783?μg/dm3 biotin. The optimum productivity was achieved using optimized medium at the fermentation temperature of 33.7?°C, initial pH?7.74, and at the time of fermentation of 58.4?h.  相似文献

【目的】从红景天根部筛选并鉴定一株产酪醇的细菌,初步研究其产酪醇特性,为寻找红景天替代资源提供新途径。【方法】用NA培养基从大花红景天根部中分离内生细菌,通过薄层层析(TLC)、高效液相色谱(HPLC)、气相色谱-质谱联用(GC-MS)筛选出产量最大的菌株,经菌落形态分析、革兰氏染色分析及16S rRNA基因序列分析其分类学地位。单因素实验确定初始pH、培养温度、发酵时间及接种量对菌株产酪醇活力的影响。【结果】从大花红景天根部分离出14株内生细菌,其中8株能产酪醇,筛选出酪醇产量最大的菌株B3,经菌落形态分析、革兰氏染色分析及16S rRNA基因序列分析初步鉴定为水生拉恩氏菌(Rahnella aquatilis)。研究其发酵条件,其最适pH为6.0,最适温度为32 °C,最佳发酵时间为42 h,最佳接种量为15%。在最适发酵条件下,用改良NA培养基发酵,B3菌株酪醇的产量为15.68 mg/L。【结论】B3菌株是一株具有产酪醇能力的细菌,在最适发酵条件下酪醇产量达到15.68 mg/L,具有潜在的开发价值。  相似文献

对丙酮丁醇梭菌发酵木薯产溶剂进行研究,分别考察了N源、木薯含量、酶处理条件和培养基pH对发酵产丁醇的影响。结果表明:最佳的产丁醇发酵培养基为木薯粉120g/L,乙酸铵6g/L;木薯粉先用高温淀粉酶按酶量20U/g、90℃水解60min,再糊化30min;发酵初始pH为6.0,发酵96h。在此条件下,5L发酵罐中丁醇产量达到13.5g/L,总溶剂达到22.8g/L。  相似文献

This article addresses the issue of effect of fermentation parameters for conversion of glycerol (in both pure and crude form) into three value-added products, namely, ethanol, butanol, and 1,3-propanediol (1,3-PDO), by immobilized Clostridium pasteurianum and thereby addresses the statistical optimization of this process. The analysis of effect of different process parameters such as agitation rate, fermentation temperature, medium pH, and initial glycerol concentration indicated that medium pH was the most critical factor for total alcohols production in case of pure glycerol as fermentation substrate. On the other hand, initial glycerol concentration was the most significant factor for fermentation with crude glycerol. An interesting observation was that the optimized set of fermentation parameters was found to be independent of the type of glycerol (either pure or crude) used. At optimum conditions of agitation rate (200 rpm), initial glycerol concentration (25 g/L), fermentation temperature (30°C), and medium pH (7.0), the total alcohols production was almost equal in anaerobic shake flasks and 2-L bioreactor. This essentially means that at optimum process parameters, the scale of operation does not affect the output of the process. The immobilized cells could be reused for multiple cycles for both pure and crude glycerol fermentation.  相似文献

Effect of pH on the batch fermentation of pullulan from sucrose medium   总被引:3,自引:0,他引:3  
Two strains of the yeast-like fungus Aureobasidium pullulans 2552 and 140B have been used for the fermentative production of the polysaccharide pullulan from a sucrose synthetic medium. In the batch fermentation, either in Erlenmeyers or in the fermentor, the pH of the culture medium was decreased rapidly from its initial pH value of 5.5 to the self-stabilized final value of 2.5 within 24 h. Experiments on the effect of initial pH on the fermentation revealed that at very low initial pH values, such as at pH 2, the polysaccharide production was in-significant. However, the biomass concentration obtained was very high at this very low initial pH value. This interesting phenomenon was served as the basic principle for the development of the bistaged pH fermentation process for the production of pullulan. In this process the first stage of fermentation was conducted at the very acidic pH for the best production of biomass. When the biomass concentration reached its maximum value, the second stage of fermentation was initiated by adjusting the medium pH to a higher value for promoting the synthesis of the polysaccharide. Experiments conducted in Erlenmeyers and in the fermentor confirmed this concept. The bistaged pH process enhanced the polysaccharide concentration in the medium, influenced the rheological properties of the fermentation broth, and has a potential of operation under nonsterile and nonaseptic conditions.  相似文献

优化了尖孢镰刀菌液体发酵生产蒽醌类红色素的发酵条件。通过单因素实验和正交优化实验,确定最佳产色素发酵培养基为:可溶性淀粉30%,(NH4)2SO4 3%,MgSO4 0.3%,KH2PO4 4%,pH 6.0。产色素最适培养条件为:初始pH6.0,装液量20%,接种量10%,吐温-80添加量1%,温度28℃,摇床转速200r/min,发酵周期120h。此条件下,色素效价即可达到8.184U/ml,比优化前提高了1.8倍。国内首次对尖孢镰刀菌所产蒽醌色素进行研究,为其进一步应用奠定基础。  相似文献

采用单因素试验和正交试验研究发酵培养基组成,优化Streptomyces sp. FIM-16-06产他克莫司的发酵条件,探讨摇瓶发酵的主要影响因子初始p H、装液量、转速等发酵参数的影响。确定了适宜的发酵培养基和发酵参数:6. 0%玉米淀粉、2. 0%黄豆粉、2. 0%葡萄糖和0. 5%玉米浆,初始pH 7. 5,装液量50 m L/500 m L三角瓶,种子菌龄48 h,接种量10%,摇床转速250 r/min,发酵温度27℃,发酵周期7 d。优化后的发酵水平较原生产工艺提高60%以上。他克莫司的优化发酵工艺为其工业化生产奠定了基础。  相似文献
