云南红豆杉愈伤组织的诱导频率为White〉MS〉B5,其中最高达80%,叶片和种子之间差异不大,但不同时期的叶片差异较大,以嫩叶诱导为佳,激素配比以2,4-D2.0-3.0mg/L,BAP0.5-1.0mg/L为宜,在继代培养阶段,B5比White和MS更适合细胞的快速生长、繁殖,并且有利于克服组织的褐化,适合于细胞生长的激素配比也以2,4-D2.0-3.0mg/L,BAP0.5-1.0mg/Lo  相似文献

针对药用植物红芽大戟结实率和种子萌发率较低的情况,探讨了利用组织培养方法解决生产中繁殖的问题。结果表明,在茎、叶和芽3种外植体中,芽的出愈率最高,愈伤组织褐化率低,生长最好,且易被诱导出不定芽,是较理想的快速繁殖材料;而茎和叶的愈伤组织褐化率高,难以诱导出不定芽。较适宜的诱导愈伤组织的激素组合是6-BA2.0mg/L+NAA0.4mg/L,诱导不定芽的适宜激素组合是6-BA5.0mg/L+NAA0.05mg/L,根的诱导则在1/2MS+NAA0.2+庶糖1.5%+琼脂0.8%培养基上进行  相似文献

大蒜分生组织培养脱病毒和快速繁殖技术   总被引:10,自引:1,他引:9  
采用茎尖分生组织培养技术,获得了大蒜(Allium sativum L.)的无病毒试管苗。通过基本培养基和激素配比试验,筛选出最佳的培养基组成,进行脱病毒苗的快速繁殖。结果表明:诱导愈伤组织的最适培养基为:MS+BA0.2mg/L+NAA0.5mg/L,月生长率达12.70倍;诱导丛生芽的最适培养基为:B5+BA0.5mg/L+IAA0.2mg/L,丛生芽繁殖系数高达25.5倍,技术上达到了快速繁  相似文献

研究了离体条件下胡杨(Populus euphratica Oliver)茎段、叶片及愈伤组织的器官发生和植株再生技术。离体培养以MS为基本培养基并附加40mg/L腺嘌呤和500mg/L水解乳蛋白。离体叶片和茎段在BA为0.5mg/L和NAA为0.5mg/L的培养基上诱导产生愈伤组织,并在含0.25mg/LBA和0.5mg/LNAA的培养基上继代增殖。BA为0.5mg/L和NAA为0.1mg/L可诱导叶片和愈伤组织发生不定芽,诱导频率分别为100%和82.9%,对于茎段,BA和NAA分别为0.1mg/L和0.01mg/L时诱导不定芽频率可达83%。试管苗在大量元素减半并附加0.015mg/LNAA的MS培养基上诱导生根,生根率达86.2%。  相似文献

从抱茎獐牙菜的胚轴、幼叶及未成熟种子诱导出愈伤组织并再生植株,试验选用MS、B5和N6三种培养基,其中以附加2.4-D3.0mg/L+6-BA0.5mg/L的MS培养基诱导率最高;以附加6-BA0.5mg/L+NAA0.2mg/L的MS培养基分化苗频率最高;以附加2.4-D2.0mg/L+6-BA0.5mg/L的MS培养基愈伤组织的生长最好。结果表明,外植体,培养基,激素等对愈伤组织诱导、继代和分化均有明显影响。采用高压液相色谱法(HPLC)测定抱茎獐牙菜愈伤组织中齐墩果酸含量的结果表明,愈伤组织中齐果墩酸含量因培养基、继代培养时间的不同而有所差异  相似文献

报道了不同激素浓度对无融合生殖没菜花托器官分化效果的研究,结果显示:(1)以MS为基本培养基,以带有子房和花柄的花托为外植体离体培养,花托、花柄切口部位直接芽诱导的最佳激素配比为4.0mg/L6-BA+0.01mg/L NAA,频率为58.82%,花托、花柄部位先形成愈伤组织,继而分化出丛生芽的最佳激素配比为5.0mg/L 6-BA+0.5mg/L NAA,频率为84.00%;(2)腋芽增殖的最佳  相似文献

采用单因子比较、正交设计和均匀设计法,分别考察影响杜仲愈伤组织诱导、生长及其超低温保存的因素。试验表明:B5+NAA0.5mg/L+6—BA1mg/L有利于意伤组织诱导。MS+2.4—D3mg/L+6—BA0.5mg/L则有利于愈伤组织快速生长。超低温保存的最佳材料是2周龄的愈伤组织。愈伤组织超低温保存的较好冷冻保护剂是12.5%Gly+7.5%PEG+2.5g/LLH。  相似文献

枸杞髓组织在4种MS培养基上都能诱导出愈伤组织,诱导率53.7%~100%。在培养基MS+6-BA0.1mg/L+NAA0.5mg/L获得的愈伤组织,呈颗粒状,分散性能好,胚性细胞多.将其转移到MS+6-BA0.5mm/L+NAA0.01mg/L的分化培养基上获得大量绿色小芽,小芽在MS+6-BA0.2mg/L的培养基上得到快速繁殖,繁殖系数50~150株/芽·月。丛生芽在MS+NAA0.2tmg/L的培养基上形成完整植株  相似文献

The in vitro studies with Cardiospermum halicacabum indicated that the different explants, i.e cotyledon, hypocotyl, cotyledonary node, leaf, internode and node had the potential to produce calli on Murashige and Skoog (MS) medium supplemented with benzylaminopurine (BAP) and napthalene acetic acid (NAA). Calli of different explant origin showed variable growth responses on different BAP concentrations. The shoots were favourably formed from the calli of leaf and cotyledon explants. The maximum number of shoots were produced from calli subcultured on MS + BAP (17.8 µM). The roots were initiated on growth regulator free MS medium.  相似文献

The regeneration of natural tetraploid T. pratense, originated from Erzurum-Turkey, is reported in this study. This plant has low seed setting and hard seed problems due to polyploidy. Hypocotyl, cotyledon, apical meristems, epicotyl and young primary leaves were inoculated on MS and PC-L2 media containing different concentrations of BAP and NAA as growth regulators. The best shoot formation has been observed on explants initiated from apical meristem placed on PC-L2 medium that includes 2 mg dm-3 BAP and 1 mg dm-3 NAA. 94.4% of the shoots originated from calli were rooted on PC-L2 medium with 1 mg dm-3 NAA. In vitro organogénesis has been accomplished in the natural tetraploid T. pratense regenerated plants successively transferred to the field.  相似文献

Saffron calli were induced from ovary explants on Murashige-Skoog (MS) medium supplemented with beyzyladenine (BA) and naphthalene acetic acid (NAA) as growth factors. MS medium with 5 mg l?1 BA and 10 mg l?1 NAA was selected for calli induction and undifferentiated calli growth, while MS medium with 1 mg l?1 BA and 1 mg l?1 NAA was the most appropriate for stigma differentiation. On this medium, stigma-like structures measuring 0.5–1.5 cm were obtained. Initially they were colourless, but yellow pigmentation, due to the presence of crocin, progressively increased with calli growth. Extracts of stigma-like structures were analysed by HPLC and the presence of saffron secondary metabolites was demonstrated. In addition, calli also showed yellow pigmentation.  相似文献

The influence of culture medium and explant on callus and shoot formation of lentil (Lens culinaris Medik.) has been studied. Three different explants (shoot-tip, first node and first pair of leaves) from three Spanish lentil cultivars were cultivated on two basal media: Murashige and Skoog medium (MS) and medium with mineral salts of MS medium plus vitamins of Gamborg's B5 medium (MSB), supplemented with growth regulators. Media with 2,4-D induced the formation of calli in all explants, but no organ regeneration was obtained from these calli. Multiple shoot formation was obtained from 33% to 92% of the explants in media supplemented with 2.25 mg l–1 of BA and 0.186 mg l–1 NAA+2.25 mg l–1 BA; in the other media one to two shoots per explant were formed in 10 to 98% of the explants. Root formation from explants was achieved only in media with NAA or IAA. Of the explants tested, the best morphogenetic responses were obtained from nodes and the poorest from leaves.  相似文献

Callus cultures of 5 genotypes of S. scabra Vog. were optimally established from leaf tissue on Murashige and Skoog (MS) basal medium supplemented with 0.5–2.0 mg l-1 2, 4-Dichlorophenoxy acetic acid (2, 4-D) and 1.0–2.0 mg l-1 6-benzylaminopurine (BAP). On media containing 2, 4-D only, calli were soft, and rhizogenesis occurred on calli of 4 genotypes. Calli formed on media containing BAP only, but not with kinetin only. In the presence of 2, 4-D, BAP inhibited rhizogenesis and promoted better callus growth than kinetin. High frequency shoot induction was achieved for 3 genotypes on MS +2.0 mg l-1 BAP. Roots formed on shoots when sub-cultured on half-strenght MS without growth regulators. The form of cytokinin used in the callus induction media appeared to affect subsequent shoot organogenesis. Genotypic differences were observed for shoot organogenesis. There was some morphological variation evident among regenerants.  相似文献

Different explants of Rauwolfia serpentina were tested for their capacity and root differentiation. MS sedium containing 2.0 mg I-1 BAP or 1.5 mg I-1 BAP with 0.5 mg I-* NAA gave the best shoot proliferation from shoot apices (84.12%). Multiple shoots subcultured on the same media gave higher number of shoots (6–9). Callus formed at the cut ends of explants produced shoots when subcultured on the above media. Rooting was achieved after transfer of shoots either to hormone free or half strength or full strength MS medium with different concentrations of IAA (0.5,1.0 mg I-1 and IBA (0.5,1.0 mg I-1).The complete regenerated plantlets have been successfully transferred to earthen pots in green house. Maximum root differentiation from leaves, stem and nodal cuttings derived calli was obtained on MS medium supplemented with NAA (2.0 mg I-1) and BAP (1.5 mg I-1). Identification of reserpine and other alkaloids from differentiated roots holds an interesting alternative for controlled production of alkaloids from this endangered, overexploited medicinal plant species.  相似文献

单一激素种类对银杏叶片,叶柄和幼茎愈伤组织的诱导中以NAA的效果最佳,2,4-D次之,6-BA最差,除胚乳外,胚,幼苗的胚根,子叶,幼茎,叶片和叶柄,以及成年树的嫩茎,叶片和叶柄各外植体在本试验条件下都能诱发愈伤组织,其中胚,子叶和叶柄的愈伤组织形成频率均可达到100.0%,叶片和幼茎在光照下的愈伤组织诱导频率比黑暗中的略高,而叶柄和胚根则相反,MS和DCR两种培养基都适合银杏幼苗叶片及叶柄愈伤组织的诱导,两者之间不存显著性差异,测得光照培养的3个组织系(ST1,ST2,ST3)中均含银杏黄酮甙元槲皮素,山柰素和异鼠李素,总含量分别为干重的0.35%,0.29%和0.14%,而黑暗中培养的这3个愈伤组织系则没有银杏黄酮的产生。  相似文献

本文报道了爪哇三七通过组织培养再生植株的方法。爪哇三七的叶片,叶柄或茎段外植体分别接种于6种分化培养基上,均难以直接分化。在附加1mgL~(12),4-D和0.1mgL~(-1)KT的B_5培养基上的叶片形成无色疏松的愈伤组织转接于MS附加1mgL~(-1)BAP和0.1mg L~(-1)NAA培养基上,5天后开始形成绿色球胚状结构,继而形成不定芽或丛生芽。这些不定芽或丛生芽在锈根培养基中迅速长成根系发达的完整植株。此外,本试验比较了不同浓度及不同组合的激素对爪哇三七外植体的脱分化、再分化的影响。并讨论了此项工作对柑桔裂皮病类病毒(简称CEV)的复制及致病机理研究的意义。  相似文献

In vitro regeneration of plantlets and multiplication of Sesbania bispinosa (Jacq.) W.F. Wight plants from cultured callus tissue were demonstrated. Callus was established from both cotyledons and mature leaflets on Murashige and Skoog (MS) basal medium supplemented with BAP (0.5 mg/l) and 2,4-D (2 mg/l). Callus mediated shoot bud differentiation was studied under defined nutritional, hormonal and cultural conditions. Various concentrations of BAP or kinetin (Kn) with coconut milk (CM) in MS media induced different levels of shoot bud differentiation as well as multiplication. Multiple shoot bud differentiation occurred in most of the primary calli. The best medium for shoot bud differentiation from cotyledon derived callus, contained BAP (2 mg/l) and 15% CM (V/V). More efficient shoot bud organogenesis was recorded with BAP than Kn. Supplementation with CM in MS media accelerated shoot bud organogenesis in differentiating callus tissue. Rooting of differentiated shoots was achieved by a three step culture procedure involving (a) MS solid medium containing IBA (2 mg/l), (b) growth regulator free half strength MS medium with 1% charcoal, and (c) half strength MS liquid medium free of vitamins, growth regulators and charcoal.Abbreviations IAA indoleacetic acid - IBA indole-3-butyric acid - NAA naphthaleneacetic acid - 2,4-D 2,4-dichlorophenoxyacetic acid - BAP 6-benzylaminopurine - Kn kinetin - CM coconut milk - MS Murashige and Skoog's medium - SBI shoot bud inducing medium  相似文献
