The tolerance of low oxygen stress in insecticide-resistant and susceptible populations of mosquitofish (Gambusiaaffinis) was investigated. Dissolved oxygen was reduced chemically with formaldehyde and sodium sulfite treatment. Available oxygen was reduced physically through barriers, both by preventing oxygen exchange at the air-water interface and by preventing the fish from contacting the surface of the water. In all cases, the tolerance to low oxygen stress was reduced in the insecticide-resistant population compared to the susceptible population. The results indicate that although tolerance of pesticides in the resistant population has been dramatically increased by environmental selective pressures, tolerance to other stresses has not been concomitantly increased.  相似文献

Polysome and ribosome preparations from normal rat liver and from a series of transplantable rat hepatomas of different growth rates were compared. All the hepatomas had a significantly higher percentage of RNA in a polysome preparation than did the normal liver, and the polysome preparations from the tumors, with the exception of the Dunning hepatoma which has a high lipid content, gave a greater yield of RNA and protein per gram of wet tissue than the liver did. Heavier polysomes were considerably less prevalent in the tumors than in the liver, and the tumors contained a larger proportion of monomer and dimer ribosomes than the liver did. Evidence is presented that the increased monomer and dimer ribosome population of the hepatomas studied is not an artifact of preparation, but represents the true intracellular distribution. Ribosomes from normal liver and Morris 5123-D hepatoma were readily dissociated by 20 min'' treatment with 1.0 mM EDTA, but ribosomes from the Dunning, Novikoff ascites, and McCoy MDAB hepatomas were little affected by such treatment. With higher concentrations of EDTA, the ribosomes from the Novikoff ascites and McCoy MDAB hepatomas broke down and did not form specific subunits as did ribosomes from liver and the Morris 5123-D hepatoma but rather gave rise to a variety of small degradation products. This behavior is ascribed to a higher RNase content of the Novikoff and McCoy MDAB hepatomas. Dunning hepatoma ribosomes were resistant to 4 mM EDTA.  相似文献

为研究高温对抗性和敏感小菜蛾Plutella xylostella生物学适合度及抗药性进化的影响,本研究测定了温度对杀虫剂抗性和敏感小菜蛾实验种群生命表参数及后代对阿维菌素敏感性的影响。 结果表明:与25℃相比,33.5℃条件下抗性和敏感种群的平均世代历期(D)、净生殖率(R0)、内禀增长率(rm)、周限增长率(λ)值、总产卵量、雌蛾平均寿命、日产卵量和孵化率均显著下降。 在25℃下,抗性和敏感种群的rm,λ,D和日产卵量无显著差异,但抗性种群的R0略低于敏感种群(为敏感种群的87%)。 在33.5℃下,抗性种群的rm和R0显著低于敏感种群(分别为敏感种群的75%和64%),抗性种群雌成虫寿命、总产卵量和孵化率显著也低于敏感种群。 抗性种群在33.5℃下饲养一代后,后代对阿维菌素的抗性水平大幅下降,而相同处理下敏感种群对阿维菌素的抗性水平没有显著下降。 上述结果表明,无论在常温还是高温条件下,抗性种群生物学适合度均低于敏感种群,尤其是高温下抗性种群适合度远低于敏感种群。 抗性种群经高温饲养一代后对阿维菌素的抗性水平大幅下降可能与高温下抗性种群适合度显著较低有关。  相似文献

We investigated the effect of a herbicide resistance-conferring mutation on fitness in Amaranthus powellii. Morphological and histological observations were made. Growth and leaf appearance were recorded for six resistant and six susceptible populations. The competitiveness of a susceptible population was compared with that of a resistant population using a replacement series experiment. Leaves of the resistant plants were distorted and much smaller than those of susceptible plants. Additionally, they exhibited an abnormal morphological and structural pattern consisting of a mosaic of heterogeneous areas in the same leaf blade. The roots and stems had similar structures in susceptible and resistant plants, but the former were up to four times more developed. The resistant plants were slower to develop and produced 67% less biomass and 58% lower leaf area than susceptible plants. Under competitive conditions, one susceptible population outperformed one resistant population by 7-15 times. The Trp(574)Leu acetohydroxyacid synthase (AHAS) mutation appears to have considerable pleiotropic effects on the early growth and development of the plants which, in competitive conditions, greatly reduce fitness.  相似文献

Summary Inclusions have been found within lipid droplets in cells of a number of air-dry seeds when investigated by freeze-etching. The inclusions invariably appear as depressions in the replicas. Toxicological and neurophysiological studies were performed to characterize the resistance mechanism in a cyclodiene-resistant strain of Drosophila melanogaster (Maryland strain). Dieldrin had an LC50 of 0.058 ppm against the larvae of susceptible D. melanogaster (Oregon-R wild type) when formulated in the rearing media. The LC50 of the resistant Maryland strain was 10.8 ppm, giving a resistance ratio (LC50-Maryland/LC50-susceptible) of 186-fold. Suction electrode recordings were made from peripheral nerves of the larval central nervous system to test whether reduced nerve sensitivity played any role in the observed resistance. In susceptible preparations (n = 5), inhibition of nerve firing by 1 mM gamma-aminobutyric acid (GABA) was effectively antagonized within 3-10 min by 10 microM dieldrin. In contrast, 30 min incubations with 10 microM dieldrin had no effect on preparations from cyclodiene-resistant individuals (n = 5). Similarly, 10 microM picrotoxinin blocked GABA-dependent inhibition in susceptible nerve preparations (n = 3). In recordings from resistant insects (n = 4), picrotoxinin displayed either weak antagonism of GABA or hyperexcitation indistinguishable from susceptible preparations. These results demonstrate that cyclodiene resistance in the Maryland strain of D. melanogaster 1) is expressed in immature stages, 2) is present at the level of the nerve, and 3) extends to picrotoxinin, albeit at a reduced level compared with dieldrin. The possible role of an altered GABA receptor in this resistance is discussed.  相似文献

Cell walls in 2 strains of Staphylococcus aureus 209P, i.e. actinomycin D susceptible and resistant ones were comparatively investigated. The resistant cells contained much more wall material per a unit of the biomass weight vs the susceptible strain cells, that conformed to thickening of the resistant cell walls detected by electron microscopy and a sharp increase of their electron density. Investigation of peptidoglycans and teichoic acids did not reveal any significant alterations in the structure of the wall components in the actinomycin D resistant cells. Only some increase of glucosamine in the peptidoglycan fraction of the resistant cells vs the susceptible ones was observed. It was shown that preparations of the resistant cell walls and peptidoglycan isolated from the resistant cells were able to bind somewhat lower quantities of actinomycin D vs the analogous preparations of the susceptible cells. The significant decrease of the antibiotic binding by live cells of the resistant strain probably slightly depended on the structure characteristics of the main wall components. The barrier properties of the walls in resistant staphylococci are most likely defined by the wall thickening and consolidation while adapting to actinomycin D.  相似文献

Chloroplasts were isolated from triazine-sensitive and triazine-resistant biotypes of common groundsel (Senecio vulgaris L.), common lambsquarter (Chenopodium album L.), and redroot pigweed (Amaranthus retroflexus L.). Chloroplast lipids were extracted and analyzed for differences among sensitive and resistant biotypes. The distribution of lipid between major lipid classes differed in chloroplasts from resistant and susceptible biotypes. Chloroplasts from resistant biotypes contained higher proportions of monogalactosyl diglyceride and phosphatidyl ethanolamine and lower proportions of digalactosyl diglyceride and phosphatidyl choline than did chloroplasts from susceptible biotypes. Monogalactosyl diglyceride and phosphatidyl ethanolamine were also quantitatively higher in membranes of resistant versus susceptible biotypes. The major lipid classes of resistant chloroplast membranes contained lipids comparatively richer in unsaturated fatty acids with the exceptions of digalactosyl diglyceride from all three biotypes and phosphatidyl ethanolamine from common groundsel. Results correlated changes in triazine sensitivity with qualitative and quantitative differences in the lipid composition of chloroplast membranes.  相似文献

Reduced fitness among resistant versus susceptible individuals slows resistance evolution and makes it easier to manage. A loss of resistance costs could indicate novel adaptations or mutations contributing to resistance. We measured costs of resistance to imidacloprid in a Massachusetts resistant population compared with a Massachusetts susceptible population in 1999 in terms of fecundity, hatching success, egg development time, and sprint speed. Resistance was additive and seemed to be polygenic with high heritability. The fecundity cost appeared overdominant in 1999, and the hatch rate cost was partly recessive in 1999, but neither was significantly different from dominant or recessive. In 2004, we repeated our measures of resistance costs in Massachusetts in terms of fecundity and hatching success, and we added a new resistant population from Maine. In 2005, we compared development time of Maine resistant and the laboratory susceptible colony eggs. Significant fecundity costs of resistance were found in both population in both 1999 and 2004, and significant egg developmental time costs were found in 1999 and 2005. However, the hatching success costs of resistance were significant in 1999 and not apparent in 2004, suggesting some modification or replacement of the resistance genes in the intervening time.  相似文献

The molecular mechanism of genetic resistance of inbred mouse strains to mouse hepatitis virus, a murine coronavirus, was studied by comparing virus binding to plasma membranes of intestinal epithelium or liver from susceptible BALB/c and resistant SJL/J mice with a new solid-phase assay for virus-binding activity. Virus bound to isolated membranes from susceptible mice, but not to membranes from resistant mice. F1 progeny of SJL/J X BALB/c mice had an intermediate level of virus-binding activity on their enterocyte and hepatocyte membranes. This correlated well with previous studies showing that susceptibility to mouse hepatitis virus strain A59 is controlled by a single autosomal dominant gene (M. S. Smith, R. E. Click, and P. G. W. Plagemann, J. Immunol. 133:428-432). Because virus binding was not prevented by treating membranes with sodium dodecyl sulfate, the virus-binding molecule could be identified by a virus overlay protein blot assay. Virus bound to a single broad band of Mr 100,000 to 110,000 in membranes from hepatocytes or enterocytes of susceptible BALB/c and semisusceptible C3H mice, but no virus-binding band was detected in comparable preparations of resistant SJL/J mouse membranes. Therefore, SJL/J mice may be resistant to mouse hepatitis virus A59 infection because they lack a specific virus receptor which is present on the plasma membranes of target cells from genetically susceptible BALB/c and semisusceptible C3H mice.  相似文献

应用食蚊鱼(Gambusia affinis)的卵黄蛋白原(VTGa)基因和雌/雄激素受体(ERa/ARa)基因转录水平为指标, 评价广东东莞寒溪河受雌/雄激素物质污染的现状。结果显示, 与对照点从化流溪河(LX)相比, 寒溪河松山湖(SM)、杨屋村(YW)、横沥镇(HL)、樟村污水处理厂上游(ZU)和樟村污水处理厂下游(ZC)各采样点雄性成年食蚊鱼肝脏中的VTG转录水平都具有显著或极显著水平的升高(p0.05和p0.001), 分别为7.67、169.43、148.69、152.01和98.12倍; 而较之对照点, 雄鱼臀鳍的雌激素受体ER转录水平仅HL位点有显著性升高(p0.05), 其余位点则不明显(p0.05)。此外, 寒溪河SM、YW、HL、ZU和ZC各采样点雌鱼臀鳍雄激素受体AR转录水平分别是对照点从化流溪河(LX)的1.21、0.82、0.34、0.41和0.89倍, 但是只有HL和ZU点有显著性减小(p0.05), 其余各点的变化差异不显著(p0.05)。研究结果显示, 生活在东莞寒溪河中的食蚊鱼受雌/雄激素物质干扰明显, 普遍出现显著的雌激素效应, 表明寒溪河水体受雌激素物质污染更严重。此外证明, 鱼类肝脏VTGa基因相对于臀鳍ERa/ARa基因转录水平更适合作为监测水体环境雌/雄激素物质的生物标志物。    相似文献

在家蚕疾病中,病毒性疾病是危害茧产品的主要且最普遍的疾病。病毒性蚕软腐病是由BmIFV, BmDNV1 和 BmDNV2引起的。对印度种质库(Indian Germ plasm stock)中的BmDN V1有抗性和敏感的家蚕品种进行了鉴定。利用标准方法对在BmDNV1感染过程中抗性和敏感品系的主要有机成分的变化(包括总蛋白、碳水化合物和脂类)进行了检测。结果表明: 随着幼虫年龄的增长,对照组和处理组中有机成分(即蛋白质和碳水化合物)的含量也随之增长,但是处理组的增长水平明显地低于各自对照组的增长水平。接种BmDNV1后,与对照组比较,敏感品种体内的血淋巴和中肠组织的总蛋白的量显著地下降。在抗性品种,接种4天后的总蛋白的量有了显著地下降,之后,下降水平小于各自的对照组。在抗性和敏感品种中,血淋巴和中肠组织中的总碳水化合物的量略有下降。在变化不显著的抗性和敏感品种中,血淋巴和中肠组织中的脂质的量有显著地提高。在敏感家蚕品种,生化变化清晰地显示: BmDNV1感染消耗作为主要能量来源的总蛋白和碳水化合物。这些成分的损耗导致了被感染家蚕的生长受阻。  相似文献

Summary Analysis of phyllosphere microflora showed that in the resistant cultivar (PHB-14) there was a significantly higher population of fungi, gram positive and gram negative bacteria, compared to susceptible cultivar (NHB-3) under healthy and diseased situations. The cultivars during earhead stage supported maximum phyllosphere fungal and gram negative bacterial populations.The rhizosphere of the susceptible cultivar under downy mildew influence supported maximum fungal and gram negative bacterial populations and the rhizosphere effect was the highest in downy mildew colonized susceptible NHB-3 at the earhead stage (95 days after planting).Among the fungi isolated Cladosporium dominated the phyllosphere of resistant, susceptible and susceptible cultivar under the influence of downy mildew.Among the rhizosphere microflora the population ofAspergillus sp. was found to be greatest in resistant, susceptible and the susceptible cultivar under the influence of downy mildew.  相似文献

Chronic ethanol intoxication leads to the development of a resistance to lipid disordering by ethanol, a phenomenon known as "membrane tolerance". In the absence of the added ethanol, the lipid order, as measured by ESR and fluorescence techniques, does not necessarily change as a result of chronic ethanol ingestion (as in liver microsomes, for example). This suggests that the spectroscopic techniques detect tolerance somewhat indirectly, in that the modification responsible may reside in a region distinct from that being probed and also raises the question of whether membrane tolerance is necessarily associated with an alteration in the membrane lipid structure. Here we show that liver microsomes from rats treated chronically with ethanol are rendered relatively resistant to the hydrolytic action of exogenous phospholipase A2, compared to preparations from control animals. This resistance persists in reconstituted lipid vesicles prepared from extracted phospholipids. Since the same substrate (1-palmitoyl-2-N-(4-nitrobenzo-2-oxa-1,3-diazole)amino caproylphosphatidylcholine) was used in both membranes from ethanol-treated animals and controls, the modification appears to reside in the structure and/or organization of the membrane. Further evidence that the lipid structure is modified by chronic ethanol treatment is provided by the observation that perturbance of the membrane structural integrity by increasing levels of oleic acid led to a progressive loss of the ethanol-induced relative resistance to hydrolysis by phospholipase A2. The results of this study support the idea that membrane tolerance involves a modification to lipid structure probably at the bilayer surface. The use of exogenous phospholipase A2 provides a new method for probing the structural modifications induced by chronic ethanol ingestion.  相似文献

A bioassay was used to detect active site insensitivity (knock-down resistance [kdr]) in pyrethroid resistant larvae of the horn fly, Haematobia irritans (L.). The larvae of the resistant population had KD50's 42.0-, 28.1- and 29.2-fold greater to permethrin, fenvalerate and lambda-cyhalothrin, respectively, compared with the susceptible population. In filter paper bioassays, resistant adult horn flies were 17 to 39.1 times less susceptible to the pyrethroids than susceptible adults at LC50. These results further document active site insensitivity as the major mechanism of pyrethroid resistance in the horn fly.  相似文献

The molecular mechanisms that mediate fish reproduction and detoxification in response to steroid hormones were studied by using adult male western mosquitofish (Gambusia affinis) as sentinel species. The expression patterns of three vitellogenins (VtgA, VtgB and VtgC), two estrogen receptors (ERα and ERβ), two androgen receptors (ARα and ARβ), metallothionein (MT) and cytochrome P450 1A (CYP1A) in the liver and testis of adult male mosquitofish were assessed through exposure treatments with progesterone (P), testosterone (T) and 17β-estradiol (E2), alone and in combination for eight days. The results showed that expression patterns of Vtg subtype, ER subtype, AR subtype, MT and CYP1A genes in male mosquitofish varied according to tissue and specific hormone stress. Vtg subtype mRNA expression was induced in the liver in E2-added treatments, and an up-regulation of ERα mRNA expression was also observed. In addition, hormone treatments increased three Vtg subtype mRNA expression levels in the testis, at least to some extent. All hormone treatments significantly inhibited ERα, ERβ and ARβ mRNA expression in the testis. Some of hormone treatments could affect MT and CYP1A gene expression in mosquitofish. In general, multiple hormone treatments showed different effects on target gene expression compared with corresponding hormone alone. The results from the present study provided valuable information on the toxicological effects of steroid hormones in mosquitofish.  相似文献

本文对白纹伊蚊Aedes albopictus溴氰菊酯抗性品系和敏感品系羧酸酯酶的生物化学性质进行了比较。白纹伊蚊抗性品系和敏感品系羧酸酯酶随底物浓度(α-乙酸萘酯或β-乙酸萘酯)的变化比活力变化趋势一致,但抗性品系对这2种底物的比活力均高于敏感品系,抗性品系羧酸酯酶的米氏常数和最大反应速度与敏感品系有显著差异。胆碱酯酶抑制剂测定结果表明,抗性品系羧酸酯酶对敌敌畏和磷酸三苯酯的敏感性高于敏感品系,对残杀威的敏感性低于敏感品系。2个品系羧酸酯酶对脱叶磷的敏感性差异不大。说明羧酸酯酶可能与白纹伊蚊对溴氰菊酯抗性有关。  相似文献

Toxicity of toxic compounds in the culture filtrates of Exserohilum turcicum towards seed germination, inhibition of shoot and root growth, callus growth and on reduction in chlorophyll content and cell viability of maize was studied. There was no germination in unautoclaved extracts at 100% (undiluted) and 50% concentrations, whereas germination was normal at all the concentrations of autoclaved toxic compounds. Inhibition of root growth was higher compared with that of shoot growth in all the test preparations of toxic compounds. Reduction in total chlorophyll and also chlorophyll a and b contents of disease resistant (KWTC-17) and susceptible (DK-4) inbreds occurred at 50, 25 and 10% concentrations of toxic compounds, with maximum reduction in the susceptible compared with the resistant inbred. Chlorophyll a was found to be more sensitive, showing 7.2% reduction in content in susceptible inbred even at 5% toxin concentration. Callus cultures/cell suspensions derived from coleotible nodes of DK-4 were more sensitive compared with that of KWTC-17 in all the test concentrations of toxic compounds. Callus growth of susceptible inbred DK-4 was inhibited by 32.7% compared with 15.31% of resistant inbred KWTC-17 at 5% concentration of toxic compounds. Cell viability of resistant and susceptible inbreds was 62.0% and 20.2% at 10% toxic concentration, respectively. Spore germination assay with four fungal species or leaf puncture assay were not found useful.  相似文献
