A very low, for the most part unmeasurable glutamic-aspartio transminase activity and a very high glutamic-alanine transaminase activity was found in the overground parts and roots of young wheat plants. The roots had a higher glutamic-alanine transaminase activity than the overground parts in the first and second leaf stage. Plants cultivated in Knop’s nutrient solution (variant with humate and without) showed a higher glutamic-alanine transaminase activity than poorly growing plants, cultivated in distilled water (with humate and without). In plants cultivated in nutrient solutions, transaminase activity increased with the age of the wheat plants. As in the previous experiments, the effect of humate was only significant, in the roots of plants cultivated in distilled water with humate, where transamination activity was greater than in the control without humate. The roots of this variant with a stimulatory growth effect showed a large accumulation of free sugars in the previous experiments. The connection between these effects of humate on the roots of young winter wheat plants is discussed.  相似文献

The enzymatic transamination reactions between aspartic and α-ketoglutaric acid and between aspartic and pyruvic acid were studied in fresh dialysed extracts of young wheat plants cultivated under various trophical conditions, in mineral solution (Knop), in the solution of an soil organic substance (potassium humate) and without nutrients (H2O). Simultaneously, the level of endogenic aspartic acid, glutamic acid and the growth values were determined. The enzymatic reactions were characterized by determining the optimum pH, the time course, and the effect of coenzyme and of inhibitors. The activity of the aspartate-glutamate transaminase from the root system of plants was considerably higher than the activity of the overground organs. The enzymatic activity from both parts of the plant was inversely proportional to the growth rate: intensive growth of the plants from the Knop variant was connected with their low enzymatic activity; the level of endogenic glutamic acid was high. The slow growth of the plants without nutrients was connected with a higher enzymatic activity; the level of endogenic glutamic acid was low. The plants from the potassium humate variant had an intermediate position between these two variants from the point of view of growth as well as from that of enzymatic activity. The plants with insufficient nutrition (slow growth, low level of endogenic glutamic acid) apparently have a low capacity for supplementing the glutamic acid deficit, which is essential for the metabolic processes, by increasing the activity of the reactions leading to glutamic acid synthesis (Asp-Glu) and, on the other hand, by decreasing the reactions utilizing it (Glu-Ala). For wheat plants the active aspartate-glutamate reaction is obviously physiologically more important than the direct reaction glutamate-aspartate and the reaction aspartate-alanine which in all cases had a very low activity.  相似文献

In dialyzed extracts from winter wheat plants transamination reactions occurred between asparagine and α-ketoglutaric acid (L-asparagine+2-oxoacid=2-oxosuccinamate+ +amino acid; 2. 6. 1. 14). Reactions with pyruvate exhibited a very low activity. Besides transamination products,i. e. glutamate and alanine, aspartic acid was formed in both reactions. Deamidation was more intensive in the weak reaction asparagine-alanine and less intensive in the asparagine-glutamate reaction. When calculated per dry weight unit the activity was the same in plants of all variants (three experimental variants—Knop, potassium humate, water). A higher, activity was found in root dialysates; however, a highly significant difference could be observed only between shoots and roots of Knop variant. When evaluating results in terms of protein content we found a significant difference between mineral variant (Knop—the lowest activity) and both deficient variants (potassium humate, water—the highest activity). Thus the highest growth activity was in connection with the lowest transamination activity and vice versa, which was the same as in transaminations of aspartic acid. In the case of asparagine, too, one can consider the possibility of its utilization via transamination for biosynthesis of glutamic acid in plants which have, for reasons of nutrition, a low level of this metabolically important amino acid.  相似文献

Changes in the nucleic acid (NA) content were studied in roots of young wheat plants cultivated under various nutritive conditions, namely in a nutrient solution, in distilled water and in a solution of sodium humate in distilled water. Changes in the ribosomal ribonucleic acid (RNA) in particular and their connection with growth rate were investigated. The amount of the NA fraction investigated (more than 90 per cent of which is represented by the ribosomal RNA) changed substantially under the cultivation conditions studied. In roots of one plant cultivated in water the content of the NA fraction investigated was at the most about 25 μg and it began to decrease as early as from the second day of cultivation. After 12 days of cultivation it decreased to 15 μg. When cultivated in Na-humate the roots contained at the most 33 μg NA, between the 5th–7th day, followed by an intensive decrease after 12 days, to 17 μg. The content of the fraction investigated in wheat plants cultivated in a nutrient solution was about double the value in comparison to these two cases with the maximal value about 60 μg between the 7th–9th day. After 12 days this amount decreased to 45 μg. The ratio between the growth rate and the NA content presented positive values only when plants were cultivated in a nutrient solution and in Na-humate until the day when the NA content ceased to increase. In the case of Na-humate this took place on the 6th day and in the case of a nutrient solution on the 9th day. Under conditions favourable for growth (in a nutrient solution and in field conditions) the precultivation of wheat plants in Na-humate resulted in a more intensive growth of roots in comparison with the root growth of plants precultivated in distilled water. In plants precultivated in distilled water for 4 days the growth rate continuously increased under favourable nutrition conditions in contrast to plants transferred from distilled water as late as on the 6th day. Their growth stopped after the transplantation and was restored only after a 2 day lag phase. On the other hand, in plants precultivated in Na-humate the transfer to favourable nutritive conditions resulted in both cases in a short term cessation of growth.  相似文献

Tomato plants were grown to the five-leaf stage under uniformconditions in a growth room with a daily light period of 15h. Plants were sampled at intervals through 24 h periods andthe free ninhydrin-positive compounds determined in roots, bleedingsap, stems and shoots (mainly leaves), using ion-exchange columnchromatography and a lithium-buffer separation system. The compoundspresent and their range of concentrations are given for twooccasions: after illumination for 8 hand after 5 h of darkness. Data for -aminobutyric acid (GAB), glutamic acid, glutamine,alanine, aspartic acid and ammonia are summarized graphicallyfor all occasions and for all parts of the plant; asparaginefor sap only. The data were examined for correlations betweenthese substances for both light and dark conditions. Relative amounts of free acids were: root glutamine> glutamicand GAB > aspartic > alanine; bleeding sap glutamine >asparagine > GAB > aspartic> alanine; stem glutamine> glutamic > GAB and aspartic > alanine; shoot (leaf)GAB and glutamine > aspartic > alanine and glutamic. Patternsof change were as follows: in the root GAB and glutamic weresimilar and unlike glutamine; alanine did not change;sap ammonia,GAB and alanine were parallel, glutamine was similar to theseonly in light; in the stem glutamine and glutamic tended toaccumulate in parallel in light, but GAB did not; in the shoot(leaf) GAB and glutamine were similar except that the formeraccumulated more rapidly in the initial light period; glutamicacid and alanine were similar to each other but distinct fromGAB and glutamine. The relatively large amounts of GAB in tomato plants and themagnitude of the changes occurring in light and darkness seemindicative of its importance as a temporary storage productfor protein amino acids, but the factors controlling accumulationand utilization in different parts of the plant are unknown.  相似文献

Summary The effect of fertilization with nitrogen and copper on the amino acid composition of oat straw has been studied.The plants (Avena sativa cv Yielder) were grown in peat with a very low copper content and supplied with two levels of nitrogen (NH4 or NO3) and three levels of copper sulphate.The higher level of nitrogen stimulated growth only when copper was added, whereas, without copper, it had an adverse effect on growth and prevented grain formation altogether. The higher level of nitrogen increased the nitrogen content of the straw at all levels of copper, but particularly in plants receiving no copper.Total amino acids in the straw hydrolysate of copper sufficient oats accounted for about 50% of the total N and was about 20% higher in copper-deficient tissues. The addition of copper caused a decrease in the amounts of all amino acids. The relative proportions of most of the amino acids to glycine remained fairly constant. Threonine, serine, alanine, iso-leucine, histidine and arginine showed small significant differences with copper treatment, whereas valine, tyrosine, phenylalanine, proline, lysine and cysteic acid (derived from cysteine and cystine) showed no differences. The proportion of aspartic acid relative to glycine in the straw hydrolysate was greatly increased in copper deficient plants supplied with the higher level of nitrogen, particularly as ammonium. The proportion of glutamic acid was also increased by the higher level of nitrogen, but showed no effect of added copper. Most of the difference in aspartic acid could be accounted for as free asparagine. The possible reasons for higher proportions of asparagine are discussed in relation to the metabolism of the oat plant.  相似文献

Studying the amino acid absorption by higher plants we observed a stimulation of 14C-alanine absorption by wheat roots preloaded with inactive alanine. Within the limits of concentration examined the increase of stimulation is more likely to depend on the free alanine content of the roots than on the alanine concentration of the solutions used for pretreatment. The stimulation is retained even after incubating the pretreated roots in diluted CaSO4 solution before the uptake period. Besides the alanine only the glycine gave stimulation, while pretreatment with valine, arginine or aspartic acid gave an inhibition. (The aspartic acid is likely to cause a general rise in the permeability of cells.) We suppose that the phenomenon of flow driven by counterflow is likely to serve as an explanation to the stimulatory preloading effect observed in the alanine absorption by wheat roots.  相似文献

Cultivar TMV-2 of groundnut plant {Arachis hypogaea L.) was grown in a nutrient solution containing fluchloralin at the rate of either 2 mg litre-1 or 4 mg litre“1. Protein synthesis and hydroxyproline content in the cell walls of roots, stem and leaves were determined. Free amino acids content and total ammonia in leaves and roots were also analysed. Presence of fluchloralin did not adversely affect protein synthesis. No significant effect of herbicide was observed on hydroxyproline content of a purified cell wall fraction of groundnut roots, stem and leaves. The total amount of ammonia increased in roots and leaves of plants which received the higher concentration of fluchloralin. With the exception of aspartic acid, asparagine, glutamic acid and glutamine, free amino acids content decreased considerably with herbicide treatment. Alanine and glycine were strongly reduced. It is suggested that transamination reactions could be affected and the process of senescence may be enhanced.  相似文献

Nicotiana rustica plants were grown under two grades of salinity conditions, 50 mM and 100 mM NaCl. High levels of NaCl added to the nutritive solution produced a progressive absorption of Na in detriment of K, thereby causing an ionic disequilibrium, particularly in leaves. The weight and longitudinal growth of plants grown under salinity conditions were lower than control plants, while their protein content was higher. Deficit of K induced by salinity increased the levels of free amino acids, especially of aspartic acid, glutamic acid and proline. On the other hand, the levels of nicotine in leaves of treated plants were lower than controls. In contrast, treatment of the plants with 100 mM NaCl induced in general an increase of nicotine in the roots. These results indicate that there was an unclear effect of salinity either on synthesis or on translocation of nicotine from roots to leves.  相似文献

In vitro root formation was studied in petiole explants from Pelargonium×hortorom Bailey cv. Radio exposed to different irradiance (2.5. 11.6 or 23.0 W/m2). Optimal root formation was found when stock plants were subjected to 2.5 W/m2 and explants were grown on a medium with 20 g/I of sucrose and 5.0 mM of nitrogen. The number of roots per explant decreased with increasing nitrogen or sucrose concentrations in the growth medium and with increasing irradiance applied to the stock plants. Variation in temperature (17, 21 or 25°C) did not affect root formation. The number of roots per explant was higher in explants from young petioles than in explants from older petioles. The initial nitrogen content of the explants was highest at the lowest irradiance. The content of reducing sugars and sucrose increased with increasing irradiance. The endogenous levels of lysine, arginine and ornithine were highest at 2.5 W/m2 and the levels of proline. alanine, γ-aminobutyric acid, glutamic acid, glutaminc, aspartic acid and asparagine were highest at 23.0 W/m2. With rising nitrogen concentration in the medium, the final endogenous content of nitrogen in the explants increased, whereas the content of reducing sugars decreased. When the sucrose concentration in the medium increased, the endogenous content of reducing sugars rose, and the nitrogen level was lowered. Variations in the nitrogen concentration in the growth medium influenced the final level of free amino acids in the explants.  相似文献

The content of the carbohydrates glucose, fructose and sucrose was determined in spring barley anthers at different stages of maturity. During maturation the sucrose content of the anthers increased markedly. The following 17 free amino acids were detected in anthers of different stages of maturity: aspartic acid, glutamic acid, serine, alanine, arginine, leucine, isoleucine, lysine, α-aminobutyric acid, glutamine, proline, tyrosine, phenylalanine, valine, threonine, cystine and glycine. Quantitative analysis was only carried out in amino acids present in higher concentrations in the analysed samples. These were: aspartic acid, glutamic acid, α-aminobutyric acid, proline, serine, valine and glutamine, and a mixture of amino acids (leucine, isoleucine, valine and phenylalanine). The total content of free amino acids increased with increasing maturity of the anthers. However, not all amino acids followed contributed to this increase, but only proline, glutamic acid, aspartic acid and glutamine. A small difference was found in the variety Gopal in which the aspartic acid content did not increase significantly, but the content of the mixture of amino acids and serine did. With the exception of green anthers of the variety Firlbecks Union, proline was present in the highest concentration in all samples analysed.  相似文献

The theanine content of the leaves of 27 species or varieties of Theaceae plants was investigated. Theanine was present in 21 species or varieties, but in much lower amounts (<0.2 μmol/g fresh weight) than the quantity detected in Camellia sinensis var. sinensis. The major free amino acids in leaves of four species belonging to the genera Schima and Eurya, were glutamic acid, aspartic acid, glutamine, asparagine, alanine and proline and content of these amino acids is similar to or higher than theanine. Accumulation of free amino acids in these plants was generally lower than in C. sinensis var. sinensis. The biosynthetic activity of theanine, assessed by the incorporation of radioactivity from [14C]ethylamine, was detected in seedlings of two species of Schima. The theanine biosynthetic activity in roots was higher than that of leaves.  相似文献

Influence on individual types (fractions) of nucleic acids (NA) was studied in roots of wheat plants grown in various cultivation media, namely in distilled water, in sodium humate and in a nutrient solution. NA’s were prepared by means of the phenol technique. Using separation on a methylalbumine column (MAK) five fractions were obtained, namely: fraction I.— low molecular weight substances, fraction II.—soluble RNA, fraction III.—DNA-RNA, and the ribosomal RNA in two fractions, IV.—(l r-RNA) and V.—(h r-RNA). Of the NA fractions investigated, the r-RNA fraction was noticeably influenced by the kind of nutrition, its amount varying in a certain proportion to the growth intensity affected by the cultivation medium. The other NA fractions were not apparently affected. The metabolical turnover of the individual NA types (as observed from the specific32P activity) was considerably influenced by the kind of nutrition as well. The specific32P activity in all NA fractions of wheat roots cultivated in a nutrient solution was approximately double that in roots of wheat plants cultivated in distilled water and Na-humate. Changes in the specific32P activity of r-RNA were again considerably evident. With regard to root growth their relative values appeared in an inverse proportion to the changes in the r-RNA content. The specific32P activity decreased with increasing growth intensity. Besides changes in the r-RNA fraction, changes in fraction I. were apparent. An especially striking decrease in the specific32P activity was found in roots of plants grown in H2O, namely by about one order in comparison with its specific activity in fraction I. from roots of plants grown in Na-humate and in a nutrient solution.  相似文献

The pathway of ammonium nitrogen assimilation, its incorporationinto amino acids and synthesis of protein was studied with theaid of nitrogen-15. The analysis of 15N involves the use ofoptical emission spectrometry. Kinetic analysis of nitrogen assimilation by the roots indicatesthat glutamine and glutamic acid were the primary products ofammonium assimilation. Possibly some of the amino acids, suchas aspartic acid and alanine received their amino nitrogen directlyfrom free ammonia in the roots. Seventeen protein-constitutive and free amino acids were isolated and determined from the fruit-bodies of Tricholoma giganteum . Aspartic acid and alanine were the most abundant protein constitutive amino acids. Among the free amino acids, alanine, glutamic acid, serine, aspartic acid and glycine were present in the largest amounts, with aspartic acid and glutamic acids especially dominant.  相似文献

Peptostreptococcus elsdenii, a strict anaerobe from the rumen, was grown on a medium containing yeast extract and [1-(14)C]- or [2-(14)C]-lactate. Radioisotope from lactate was found in all cell fractions, but mainly in the protein. The label in the protein fraction was largely confined to a few amino acids: alanine, serine, aspartic acid, glutamic acid and diaminopimelic acid. The alanine, serine, aspartic acid and glutamic acid were separated, purified and degraded to establish the distribution of (14)C from lactate within the amino acid molecules. The labelling patterns in alanine and serine suggested their formation from lactate without cleavage of the carbon chain. The pattern in aspartic acid suggested formation by condensation of a C(3) unit derived directly from lactate with a C(1) unit, probably carbon dioxide. The distribution in glutamic acid was consistent with two possible pathways of formation: (a) by the reactions of the tricarboxylic acid cycle leading from oxaloacetate to 2-oxoglutarate, followed by transamination; (b) by a pathway involving the reaction sequence 2 acetyl-CoA-->crotonyl-CoA-->glutaconate-->glutamate.  相似文献

Amino acid requirements for the growth of Aedes albopictus, clone C6/36, cells and for the production of dengue (DEN) and Chikungunya (CHIK) viruses were examined by growing the cells or the viruses in media which were deprived of one of the 20 amino acids. Cell growth was markedly inhibited when cystine was omitted from the medium, and to a lesser extent by arginine deprivation. On the other hand, omission of alanine, asparagine, aspartic acid, and glutamic acid at the same time did not affect cell growth. Marked accumulation of alanine was observed in the medium when the cells were grown for 8 days in complete medium, with concomitant depletion of aspartic acid and glutamic acid. The production of CHIK virus was inhibited markedly by omission of cystine from the medium after virus infection, while the production of DEN viruses was more affected by glycine deprivation, although cystine deprivation also inhibited virus production to a lesser extent. On the other hand, production of CHIK and DEN viruses was not affected when alanine, asparagine, aspartic acid, and glutamic acid were omitted from the medium at the same time.  相似文献

Amino acids emitted and extracted from surface-sterilized larvae and adults of Heterodera glycines were identified by paper chromatography and quantitatively analyzed by column chromatography. Five amino acids (alanine, aspartic acid, glutamic acid, glycine and serine) were emitted by H. glycines larvae and eight others (asparagine, glutamine, leucine/isoleucine, lysine, methionine sulfoxide, threonine, tyrosine, valine/methionine) were found in extracts from crushed larvae.In addition to the amino acids emitted or extracted from larvae, four others were emitted by adults (γ-aminobutyric acid, histidine, phenylalanine, and proline). Four different amino acids (arginine, cystathionine, hydroxyproline, and ornithine) were found only in the extract from crushed adults. Greater quantities of alanine, aspartic acid and glycine were emitted than could be detected in nematode extracts suggesting selective emission.Subsamples of nematode populations were taken from growing plants 19, 26, 33, and 40 days after inoculation and extracted to determine whether changes in specific amino acid content correlated with aging. Proline content shifted most, increasing from 4.1% to 21.5% of the total amino acid complement from the 19th to the 40th days.  相似文献

The present investigation revealed the effect of the organochlorine insecticide dieldrin at the dose level 0.25 LD50 at different time intervals on the concentration of 11 rat brain amino acids, on the activities of glutamic oxyacetic transaminase (GOT), glutamic pyruvic transaminase (GpT) and cholinesterase. The study was also extended to include the total protein content during the tested periods. The daily injection of dieldrin caused a marked decrease in the levels of glutamic acid, glutamine and taurine and an increase in the levels of aspartic acid, asparagine, GABA, glycine, lysine, serine, alanine and histidine. However, the maximal increase and decrease were recorded for most of the tested amino acids at the end of the tested period. The activity of the transaminases increased significantly. The recorded values of GOT were usually higher than GPT. Cholinesterase activity was inhibited thoroughly during all the experimental periods. Total protein content was decreased in the experiment; the minimal value was given 3 days after the injection.  相似文献

The amino acids lysine and glycine are reported to react with glucose at physiological pH and temperature and undergo non-enzymic glycation. Three other amino acids present in relatively larger amounts in the lens i.e. alanine, aspartic acid and glutamic acid were also found to undergo non-enzymic glycation as found by incorporation of uniformly labelled (U-[14C]) glucose into the amino acids. The glucose incorporation was 1.6 to 2.5% for alanine, 35 to 50% for aspartic acid and 2.3 to 3.3% for glutamic acid. Each amino acid of varying concentrations lowered the extent ofin vitro glycation of lens proteins significantly in glucose-treated homogenates of normal lens from humans. The decrease in glycation for alanine was between 32 and 69%, that for aspartate was between 18 and 74%, and for glutamate was between 52 to 74%. Decreased glycation was greater for higher concentrations of glucose. Scavenging of intracellular glucose and decreasing the extent of glycation of lens proteins could be the mechanism of action by which the amino acids alanine, aspartic acid and glutamic acid could exercise a beneficial effect on cataract and diabetic retinopathy.  相似文献
