Quantitative data on modes of transmission are a crucial element in understanding the ecology of microorganisms associated with animals. We investigated the transmission patterns of a γ-proteobacterium informally known as pea aphid Bemisia-like symbiont (PABS), also known as T-type, which is widely but not universally distributed in natural populations of the pea aphid, Acyrthosiphon pisum. The vertical transmission of PABS to asexual and sexual morphs and sexually produced eggs was demonstrated by a diagnostic PCR-based assay, and the maximum estimated failure rate was 2%. Aphids naturally lacking PABS acquired PABS bacteria administered via the diet, and the infection persisted by vertical transmission for at least three aphid generations. PABS was also detected in two of five aphid honeydew samples tested and in all five siphuncular fluid samples tested but in none of 15 samples of salivary secretions from PABS-positive aphids. However, PABS-negative aphids did not acquire PABS when they were cocultured with PABS-positive aphids; the maximal estimated level of horizontal transmission was 18%. A deterministic model indicated that the force of infection by a horizontal transmission rate of 3% is sufficient to maintain a previously described estimate of the prevalence of PABS-positive aphids (37%), if the vertical transmission rate is 98%. We concluded that PABS infections in A. pisum can be maintained by high vertical transmission rates and occasional horizontal transmission, possibly via the oral route, in the absence of selection either for or against aphids bearing this bacterium.  相似文献

Five different isolates of beet yellows virus were maintained without any changes in their properties onTetragonia expansa Murr. syn.T. tetragonoides Pall. for a long period of time. According to their characteristics and different properties especially in a diploid inbred line of sugar beet the isolates are considered to be strains of BYV and are classified into three groups: group of mild strains (the mild masked and mild strains), normal strains (the common strain) and necrotic strains (the severe necrotic and necrotic strains). The necrotic strains of BYV were relatively easily transmissible manually to sugar beet plants and other indicator species. The common strain can be transmitted to sugar beet,Chenopodium quinoa Willd. but not toC. capitatum L. Asch. Mild strains are transmissible with difficulty andC. quinoa is the only species which develops a larger number of local lesions after inoculation. In contrast to the mild masked and common strains it is manually transmissible toC. capitatum. The mild masked strain can not be transmitted to sugar beet.Nicotiana quadrivalvis Pursh. is not susceptible to mechanical inoculation with BYV. Aphid transmission withMyzus persicae (Sulz.) was positive in experiments with necrotic strains only. Mechanical transmission of BYV was successful also toC. foliosum (Moench) Asch.,C. murale L. andClaytonia perfoliata Donn. The last two species were susceptible to inoculation by aphids as well. The free cyclitols pinitol, ononitol and myo-inositol occur in the honeydew (excreta) of pea aphids (Acyrthosiphon pisum) which feed on pea aphid-susceptible alfalfa (Medicago sativa cv Caliverde). These cyclitols also occur in the leaves and stems of alfalfa. Aphids were incapable of de novo synthesis of these cyclitols. Honeydew production by the pea aphid results from ingesting phloem-sap, so the occurrence of cyclitols in honeydew results from their translocation in the phloem. The relatively high content of myo-inositol in honeydew indicates that it is selectively translocated. The most abundant alfalfa cyclitol, pinitol, had no effect on aphid feeding behavior at concentrations up to 1% (w/v; artificial diet).  相似文献

Members of several bacterial lineages are known only as symbionts of insects and move among hosts through maternal transmission. Such vertical transfer promotes strong fidelity within these associations, favoring the evolution of microbially mediated effects that improve host fitness. However, phylogenetic evidence indicates occasional horizontal transfer among different insect species, suggesting that some microbial symbionts retain a generalized ability to infect multiple hosts. Here we examine the abilities of three vertically transmitted bacteria from the Gammaproteobacteria to infect and spread within a novel host species, the pea aphid, Acyrthosiphon pisum. Using microinjection, we transferred symbionts from three species of natural aphid hosts into a common host background, comparing transmission efficiencies between novel symbionts and those naturally infecting A. pisum. We also examined the fitness effects of two novel symbionts to determine whether they should persist under natural selection acting at the host level. Our results reveal that these heritable bacteria vary in their capacities to utilize A. pisum as a host. One of three novel symbionts failed to undergo efficient maternal transmission in A. pisum, and one of the two efficiently transmitted bacteria depressed aphid growth rates. Although these findings reveal that negative fitness effects and low transmission efficiency can prevent the establishment of a new infection following horizontal transmission, they also indicate that some symbionts can overcome these obstacles, accounting for their widespread distributions across aphids and related insects.  相似文献

The variability of the incubation period of the beet mosaic virus [Beta virus 2(Lind) Smith] was investigated in a three year experiment. The virus was transmitted by the black bean aphid (Aphis fabae Scop.) or by a mechanical inoculation onAmaranthus caudatus L. Variability characterized by the standard deviation indicates a closer distribution of measurements when the virus was transmitted by mechanical inoculation. The differences in variability of the incubation period between the two methods of virus tramission are statistically significant; dispersion and standard deviation are lower after mechanical inoculation. The average length of the incubation period after virus transmission by aphids and by mechanical inoculation was 10.8 and 7.7 days, respectively. The difference was found statistically significant and may be explained by the higher number of viral particles transmitted by mechanical inoculation which may accelerate the process of infection. A significant correlation between the temperature of the environment and the length of the incubation period was also found in some experiments.  相似文献

This study examined the induction of the defence-related hormones jasmonic acid (JA), salicylic acid (SA) and abscisic acid (ABA) and the phytoalexin medicarpin in Medicago truncatula when challenged by the pea aphid Acyrthosiphon pisum. There was some induction of hormones in the compatible interaction between A. pisum clone N116 and M. truncatula cultivar DZA315, whereas JA, SA and medicarpin exhibited more significant increases in foliage concentration during the incompatible interaction between A. pisum clone PS01 and M. truncatula cultivar Jemalong A17. Foliar concentration of JA, SA and medicarpin exhibited a positive relationship with aphid density after 3-day feeding, whereas ABA was not affected by the presence of aphids. When aphids were restricted to a single leaf using plastic tubes, JA, SA and medicarpin displayed strong local induction, whereas there were no significant systemic increases in uninfested leaves. Medicarpin and SA appeared to increase with duration of aphid feeding, whereas JA showed a more transient increase in concentration 24 h after challenge commenced. Results suggest that increases in JA, SA and medicarpin are associated with M. truncatula resistance to particular clones of A. pisum. The variation in concentration of the defence-related compounds recorded with regard to aphid density, duration of challenge, genotypes of plant and aphids, and between locally challenged and distant leaves reinforces the need for consideration of these experimental factors when generalizing about the plant defence processes that occur during aphid–plant interactions.  相似文献

The occurrence of a secondary bacterial symbiont (PASS) of pea aphid, Acyrthosiphon pisum (Harris), was detected by polymerase chain reaction (PCR) with specific nucleotide primers based on PASS 16S rDNA nucleotide sequences from over 80% (50/57) of clones of pea aphid collected from widely separated locations in California. PASS was also detected by PCR in both red and green phenotypes of rose aphid, Macrosiphum rosae (L.), but not in six other species of aphids examined, including blue alfalfa aphid (A. kondoi Shinji). The nucleotide sequences of the PCR-amplified, partial 16S rDNAs (1060 bp) from pea aphid and rose aphid were identical and 99.9% similar to the published 16S rDNA of PASS. PASS and a recently described new rickettsia of pea aphid (PAR) were transmitted by needle injection of hemolymph from positive pea aphid clones into negative clones and into blue alfalfa aphids. Bathyplectes curculionis (Thomson) is an introduced natural enemy of the alfalfa weevil in North America. The wasp requires carbohydrate foods as an adult. Adult wasps have increased longevity and fecundity when provided access to pea aphid, Acyrthosiphon pisum (Harris), honeydew in the laboratory, and adults respond positively to the presence of pea aphids in alfalfa fields. However, it is unknown how these wasps find aphid honeydew in the field. In a series of Y-tube olfactometer experiments, we evaluated the response of naïve and experienced adult female B. curculionis to odors from pea aphids, alfalfa, and pea aphids on alfalfa. Naïve adult females did not respond positively to pea aphid odor even when hungry. But adult females were able to learn aphid odor, and the mechanism of learning appears to be associative rather than by sensitization. Naïve females also showed no preference for alfalfa odor but learned alfalfa odor through sensitization. The wasps did not distinguish between alfalfa with aphids and alfalfa without aphids, even after exposure to aphids or alfalfa with aphids. However, they preferred pea aphid odor to alfalfa odor after a feeding experience in the presence of pea aphid odors. But after exposure to mixed odors of aphids and alfalfa while feeding, B. curculionis females preferred the odor of alfalfa to the odor of pea aphids. These results suggest that alfalfa odors mask or override aphid odors when aphids are associated with alfalfa (as happens naturally), thus interfering with the wasp's ability to respond to learned aphid odors. Therefore, although the wasps are capable of learning to find pea aphids and their honeydew in a simplified laboratory setting, it appears unlikely that they do so in the field.  相似文献

The bacterium Serratia marcescens is pathogenic for the pea aphid, Acyrthosiphon pisum. When S. marcescens cells are inoculated into the hemocoel of the aphid, there is a variable lag period of 2–3 hr duration. This phase is followed by a rapid increase in pathogen numbers causing a general septicemia and the death of the aphid. The LD50 for fourth-instar pea aphid nymphs was estimated as 190 viable S. marcescens cells (95% confidence limits, 123–288). It is suggested that being sucking insects aphids are protected against infection by bacterial pathogens in the field.  相似文献

Viral diseases non-persistently transmitted by aphids are of great economic importance in several annual crops. Transmission efficiency of these non-persistent phytoviruses is dependant on vector efficiency (i.e. vector intrinsic ability to transmit the virus) but also on the vector activity that implies the early steps of aphid host plant selection process (i.e. brief intracellular stylet punctures after landing) and to their interplant movement ability. In Europe, Macrosiphum euphorbiae (Thomas 1878) is considered as one of the most serious virus vectors on potato (Solanum tuberosum L. 1753). Nevertheless, several alate aphid species that do not colonise potato plants are trapped in potato crops. Therefore, we investigated, through laboratory experiments, vector activity of one potato colonising aphid, M. euphorbiae, and two non-colonising potato aphids, the bird cherry-oat aphid Rhopalosiphum padi (L. 1758) and the pea aphid Acyrthosiphon pisum (Harris 1776). A settling experiment was used to evaluate dispersal activity, and the electrical penetration graph (EPG) technique was used to investigate probing activity on potato plants. Results showed that M. euphorbiae exhibited a better vector activity than other two aphid species in terms of landing and probing. By contrast, interplant movements were only recorded on non-colonising aphids, suggesting a better vector activity than M. euphorbiae in terms of locomotive behaviour. These data confirm the involvement of A. pisum and R. padi in the spread of non-persistent viruses.  相似文献

The impact of host nutrition on symbiont regulation in the pea aphid Acyrthosiphon pisum was investigated. The population density of the obligate symbiont Buchnera aphidicola positively correlated with dietary nitrogen levels. The innate immune system of insects provides effective defence against a range of parasites and pathogens. The pea aphid, Acyrthosiphon pisum, is a novel study system for investigating host-parasite interactions due to its complex associations with both well-characterised bacterial symbionts and a diversity of pathogens and parasites, including several important biological control agents. However, little is known about the cellular and humoral immune responses of aphids. Here we identify three morphologically distinct types of haemocytes in circulation that we name prohemocytes, granulocytes and oenocytoids. Granulocytes avidly phagocytose Gram negative Escherechia coli and Gram positive Micrococcus luteus while oenocytoids exhibit melanotic activity. Prohaemocytes increase in abundance immediately following an immune challenge, irrespective of the source of stimulus. Pea aphids form melanotic capsules around Sephadex beads but do not form cellular capsules. We also did not detect any antimicrobial peptide activity in the haemolymph using zone of inhibition assays. We discuss these results in relation to recent findings from the pea aphid genome annotation project that suggest that aphids have a reduced immune gene repertoire compared to other insects.  相似文献

Pre and Post-virus-acquisition starvation of Aphis craccivora Koch, and Acyrthosiphon pisum Harris resulted in appreciable increase in percentage of transmission of urdbean leaf crinkle virus. Highest transmission occured when aphids were starved for 90 min prior to virus-acquisition. A. pisum and A. craccivora on 20 and 80 min of post-acquisition starvation and 10 and 20 min of virus-acquisition, respectively inoculated more number of plants. Both the aphids transmitted the virus in probes lasting for one min. However, beyond 5 min of successive transfer on test plants loss in transmissibility was apparent. Viruliferous aphid off the plant retained the virus for a much longer period than on maize plant. The virus appears to be non-persistently borne in the aphids.  相似文献

We compared the settling preferences and reproductive potential of an oligophagous herbivore, the pea aphid, Acyrthosiphon pisum Harris (Hemiptera: Aphididae), in response to pea plants, Pisum sativum L. cv. ‘Aragorn’ (Fabaceae), infected with two persistently transmitted viruses, Pea enation mosaic virus (PEMV) and Bean leaf roll virus (BLRV), that differ in their distribution within an infected plant. Aphids preferentially oriented toward and settled on plants infected with PEMV or BLRV in comparison with sham‐inoculated plants (plants exposed to herbivory by uninfected aphids), but aphids did not discriminate between plants infected with the two viruses. Analysis of plant volatiles indicated that plants inoculated with either virus had significantly higher green leaf volatile‐to‐monoterpene ratios. Time until reproductive maturity was marginally influenced by plant infection status, with a trend toward earlier nymph production on infected plants. There were consistent age‐specific effects of plant infection status on aphid fecundity: reproduction was significantly enhanced for aphids on BLRV‐infected plants across most time intervals, though mean aphid fecundity did not differ between sham and PEMV‐infected plants. There was no clear pattern of age‐specific survivorship; however, mean aphid lifespan was reduced on plants infected with PEMV. Our results are consistent with predictions of the host manipulation hypothesis, extended to include plant viruses: non‐viruliferous A. pisum preferentially orient to virus‐infected host plants, potentially facilitating pathogen transmission. These studies extend the scope of the host manipulation hypothesis by demonstrating that divergent fitness effects on vectors arise relative to the mode of virus transmission.  相似文献

The distribution and mobility of infected aphid hosts can have a great effect on the ability of a pathogen to spread throughout a population. The distribution of dead and living pea aphids (Acyrthosiphon pisum) infected with Pandora neoaphidis was compared with that of their healthy conspecifics. Infected aphids were significantly more likely to be found on the undersides of alfalfa leaves and off of the plants than were healthy aphids. These two shifts in microhabitat location have potential costs and benefits for both the host and the pathogen.  相似文献

Pea enation mosaic virus (PEMV) was isolated from disea sed field pea (Pisum sativum L.ssp. arvense A.Gr.) and broad bean (Faba vulgaris Moench) plants grown as filed crops at Bohumilice in Bohemia. The virus proved to be pathogenic for the following plant species:Pisum sativum L. cv. Raman,Faba vulgaris Moench,Lens culinaris Med.,Vicia sativa L.,Lathyrus odoratus L.,Glycine soja L.,Phaseolus vulgaris L.,Chenopodium amaranticolor Coste andReyn,Nicotiana clevelandi Gray,Trifolium incarnatum L. The dilution end point of the isolate was higher in pea plants (10?4) than in broad bean plants (10?2). The thermal inactivation point was 65–68° and the longevityin vitro between 10 and 14 days. According to the host range, symptoms on pea plants and physical properties the virus isolate studied resembles some isolates described in the U.S.A. and represents a PEMV strain different from those reported so far in Czechoslovakia.  相似文献

The function of the pea aphid's primary symbiont, Buchnera aphidicola, has been well studied. However, the factors affecting the dynamics of Buchnera density are seldom studied simultaneously. A better understanding of these factors could provide insights into its symbiosis with aphids. This study evaluated the effects of host life stage and rearing temperature on Buchnera density. We measured Buchnera density in seven life stages of pea aphids (Acyrthosiphon pisum) reared at six constant temperatures. Both host life stage and temperature significantly affected Buchnera density, which tended to decrease as aphid age and rearing temperature increased.  相似文献
