The meroblastic egg of the teleost, Fundulus heteroclitus, was studied electrophysiologically from cleavage to mid-gastrula stages. The yolk is an intracellular inclusion surrounded by a membrane of high resistivity (50 kΩcm2). This membrane generates a cytoplasm-negative resting potential in later stages. Cells of all stages studied are coupled electrically. In gastrulae, coupling is both by way of specialized junctions between cells and by way of intra-embryonic extracellular space, the segmentation cavity. The latter mode is present because the segmentation cavity is sealed off from the exterior by a high resistance barrier, and the outer membrane of surface cells is of high resistance (50–100 kΩcm2) compared to the inner membrane. It can be inferred that clefts between surface cells are occluded by circumferential junctions. Isolated cells from late cleavage stages develop coupling in vitro, confirming the existence of coupling by way of intercellular junctions. Both modes of coupling could mediate communication between cells that is important in embryonic development.  相似文献

In all animals examined, somatic cells of the gonad control multiple biological processes essential for germline development. Gap junction channels, composed of connexins in vertebrates and innexins in invertebrates, permit direct intercellular communication between cells and frequently form between somatic gonadal cells and germ cells. Gap junctions comprise hexameric hemichannels in apposing cells that dock to form channels for the exchange of small molecules. Here we report essential roles for two classes of gap junction channels, composed of five innexin proteins, in supporting the proliferation of germline stem cells and gametogenesis in the nematode Caenorhabditis elegans. Transmission electron microscopy of freeze-fracture replicas and fluorescence microscopy show that gap junctions between somatic cells and germ cells are more extensive than previously appreciated and are found throughout the gonad. One class of gap junctions, composed of INX-8 and INX-9 in the soma and INX-14 and INX-21 in the germ line, is required for the proliferation and differentiation of germline stem cells. Genetic epistasis experiments establish a role for these gap junction channels in germline proliferation independent of the glp-1/Notch pathway. A second class of gap junctions, composed of somatic INX-8 and INX-9 and germline INX-14 and INX-22, is required for the negative regulation of oocyte meiotic maturation. Rescue of gap junction channel formation in the stem cell niche rescues germline proliferation and uncovers a later channel requirement for embryonic viability. This analysis reveals gap junctions as a central organizing feature of many soma–germline interactions in C. elegans.  相似文献

Lanthanum tracer and freeze-fracture electron microscope techniques were used to study junctional complexes between granulosa cells during the differentiation of the rabbit ovarian follicle. For convenience we refer to cells encompassing the oocyte, before antrum and gap junction formation, as follicle cells. After the appearance of an antrum and gap junctions we call the cells granulosa cells. Maculae adherentes are found at the interfaces of oocyte-follicle-granulosa cells throughout folliculogenesis. Gap junctions are first detected in follicles when the antrum appears. In early antral follicles typical large gap junctions are randomly distributed between granulosa cells. In freeze-fracture replicas, they are characterized by polygonally packed 90-Å particles arranged in rows separated by nonparticulate A-face membrane. A particle-sparse zone surrounds gap junctions and is frequently occupied by small particle aggregates of closely packed intramembranous particles. The gap junctions of granulosa cells appear to increase in size with further differentiation of the follicle. The granulosa cells of large Graafian follicles are adjoined by small and large gap junctions; annular gap junctions are also present. The large gap junctions are rarely surrounded by a particle-free zone on their A-faces, but are further distinguished by particle rows displaying a higher degree of organization.  相似文献

THE STRUCTURAL ORGANIZATION OF THE SEPTATE AND GAP JUNCTIONS OF HYDRA   总被引:10,自引:8,他引:2  
The septate junctions and gap junctions of Hydra were studied utilizing the extracellular tracers lanthanum hydroxide and ruthenium red. Analysis of the septate junction from four perspectives has shown that each septum consists of a single row of hexagons sharing common sides of 50–60 A. Each hexagon is folded into chair configuration. Two sets of projections emanate from the corners of the hexagons. One set (A projections) attaches the hexagons to the cell membranes at 80–100-A intervals, while the other set (V projections) joins some adjacent septa to each other. The septate junctions generally contain a few large interseptal spaces and a few septa which do not extend the full length of the junction. Basal to the septate junctions the cells in each layer are joined by numerous gap junctions. Gap junctions also join the muscular processes in each layer as well as those which connect the layers across the mesoglea. The gap junctions of Hydra are composed of rounded plaques 0.15–0.5 µ in diameter which contain 85-A hexagonally packed subunits. Each plaque is delimited from the surrounding intercellular space by a single 40-A band. Large numbers of these plaques are tightly packed, often lying about 20 A apart. This en plaque configuration of the gap junctions of Hydra contrasts with their sparser, more widely separated distribution in many vertebrate tissues. These studies conclude that the septate junction may possess some barrier properties and that both junctions are important in intercellular adhesion. On a morphological basis, the gap junction appears to be more suitable for intercellular coupling than the septate junction.  相似文献

Some transposable DNA elements in higher organisms are active in somatic cells, as well as in germinal cells. What effect does the movement of DNA elements in somatic cells have on life history traits? It has previously been reported that somatically active P and mariner elements in Drosophila induce genetic damage and significantly reduce lifespan. In this study, we report that the movement of P elements in somatic cells also significantly reduces fitness, mating activity, and locomotion of Drosophila melanogaster. If other elements cause similar changes in life history traits, it is doubtful if transposable DNA elements remain active for long in somatic cells in natural populations.  相似文献

The intercellular junctions of the epithelium lining the hepatic caecum of Daphnia were examined. Electron microscope investigations involved both conventionally fixed material and tissue exposed to a lanthanum tracer of the extracellular space. Both septate junctions and gap junctions occur between the cells studied. The septate junctions lie apically and resemble those commonly discerned between cells of other invertebrates. They are atypical in that the high electron opacity of the extracellular space obscures septa in routine preparations. The gap junctions are characterized by a uniform 30 A space between apposed cell membranes. Lanthanum treatment of gap junctions reveals an array of particles of 95 A diameter and 120 A separation lying in the plane of the junction. As this pattern closely resembles that described previously in vertebrates, it appears that the gap junction is phylogenetically widespread. In view of evidence that the gap junction mediates intercellular electrotonic coupling, the assignment of a coupling role to other junctions, notably the septate junction, must be questioned wherever these junctions coexist.  相似文献

An electrophysiological and anatomical study of the guinea pig taenia coli is reported. Changing the membrane potential of single cells cannot modulate the rate of firing action potentials but does reveal electrical coupling between the cells during propagation. The amplitude of the junction potentials which occur during transmission from inhibitory nerves is unaffected in many cells during alteration of the membrane potential, indicating electrical coupling during transmission. The taenia coli is shown to consist of smooth muscle bundles which anastomose. There are tight junctions between the cells in the bundles, and these probably provide the pathway for the electrical coupling. The smooth muscle cells towards the serosal surface of the taenia coli are shown electrophysiologically to have an extensive intramural inhibitory innervation, but a sparse sympathetic inhibitory and cholinergic excitatory innervation. These results are in accordance with the distribution of these nerves as determined histochemically. As single axons are only rarely observed in the taenia coli, it is suggested that the only muscle cells which undergo permeability changes during transmission are those adjacent to varicosities in the nerve bundles. The remaining muscle cells then undergo potential changes during transmission because of electrical coupling through the tight junctions.  相似文献

The occurrence of low resistance junctions between normal chick embryo fibroblasts and between fibroblasts transformed with Rous sarcoma virus in tissue culture was studied with intracellular microelectrodes. The results showed that these junctions were present between normal chick fibroblasts in proliferating cultures as well as between cells in ‘density dependent inhibited cultures’. Mechanical injury to a fibroblast within a small group of coupled cells caused the injured fibroblast to uncouple immediately from its neighboring cells without interrupting coupling between the healthy uninjured cells. In the case of fibroblasts transformed with a Rous sarcoma virus, the results further showed that low resistance junctions were present when the transformation appeared in the infected cells and remained present thereafter.  相似文献

Frequency response functions were measured between the cells of Xenopus laevis embryos during the first two cleavage stages. Linear systems theory was then used to produce electronic models which account for the electrical behavior of the systems. Coupling between the cells may be explained by models which have simple resistive elements joining each cell to its neighbors. The vitelline, or fertilization, membrane which surrounds the embryos has no detectable resistance to the passage of electric current. The electrical properties of the four-cell embryo can only be explained by the existence of individual junctions linking each pair of cells. This arrangement suggests that electrotonic coupling is important in the development of the embryos, at least until the four-cell stage.  相似文献

TLR7 enhances germinal center maturation and migration of B cells to the dark zone where proliferation and somatic hypermutation occur. Our goal was to determine how Tlr7 dose influences selection of the autoreactive B cell repertoire in NZW/BXSB. Yaa mice bearing the site-directed heavy chain transgene 3H9 that encodes for the TLR7 regulated anti-CL response. To create a physiologic setting in which autoreactive B cells compete for survival with non-autoreactive B cells, we generated bone marrow chimeras in which disease onset occurred with similar kinetics and the transferred 3H9+ female non-Yaa, male Yaa or male TLR7-/Yaa cells could be easily identified by positivity for GFP. Deletion of 3H9 B cells occurred in the bone marrow and the remaining 3H9 follicular B cells manifested a decrease in surface IgM. Although there were differences in the naïve repertoire between the chimeras it was not possible to distinguish a clear pattern of selection against lupus related autoreactivity in TLR7-/Yaa or female chimeras. By contrast, preferential expansion of 3H9+ B cells occurred in the germinal centers of male Yaa chimeras. In addition, although all chimeras preferentially selected 3H9/Vκ5 encoded B cells into the germinal center and plasma cell compartments, 3H9 male Yaa chimeras had a more diverse repertoire and positively selected the 3H9/Vκ5-48/Jκ4 pair that confers high affinity anti-cardiolipin activity. We were unable to demonstrate a consistent effect of Tlr7 dose or Yaa on somatic mutations. Our data show that TLR7 excess influences the selection, expansion and diversification of B cells in the germinal center, independent of other genes in the Yaa locus.  相似文献

We identified adhesive junctions and gap junctions between Sertoli cells, between Sertoli and germ cells and between germ cells in the testis of P. fasciatum, a catfish of commercial relevance. To investigate the role of these junctions in spermatogenesis, as well as the molecular composition of the junctions, we performed an immunohistochemistry light microscopy as well as an immunogold labelling electron microscopy study with antibodies to adhesive and gap junctions proteins. Testes that were at different stages of spermatogenesis were used. Based on our morphological studies we speculate that Sertoli–germ and germ–germ cell adhesive junctions are important for maintaining the three-dimensional structure of the germinal cysts and an organized arrangement of the germ cells inside the cysts. Connexin 32 was identified in the germ cells and in the cysts walls. Our observations also suggest that Sertoli–germ and germ–germ cells gap junctions may be involved in the mechanism of synchronous development of germ cells.  相似文献

In vitro enumeration of diploid human cell variants that are resistant to purine analogues is a possible method of detecting mutagenesis. Their incidences can be increased by the known mutagens, X-rays and N-methyl-N′-nitro-N-nitrosoguanidine (MNNG). Usefulness of this method depends on the kinds of hereditary changes that confer analogue-resistance on somatic cells. If resistance usually results from changes in genetic material, in vitro studies could be useful indicators of mutagenic effects on somatic cells and germ cells in vivo. If epigenetic changes are primarily responsible for analogue-resistant variants, their enumeration might not provide information relevant to germinal mutations but would still be a useful way to detect induction of general kinds of stable phenotypic changes that could cause cancer. This article outlines hypothetical epigenetic and genetic causes of somatic cell variation and a prospective genetic analysis of human cell variants that are resistant to 8-azaguanine (AG) or 2,6-diaminopurine ( (DAP).Recent evidences and arguments favoring epigenetic origins of resistance to base-analogues are inconclusive. The often cited high rate of changes causing impermeability to BUdR in hamster cells is based on one improperly executed determination. Comparisons of rates of variation conferring BUdR-resistance on cultured haploid and diploid frog cells included diploid variants that did not behave as mutants and ignored major sources of error in estimating mutation rates. AG-resistance could result from recessive mutations in X-chromosomal genes but comparisons of rates of mutation in hamster cells of different ploidies did not provide information about the numbers of X-chromosomes in the variants. Reports that normal rodent HGPRT reappeared in hybrids of enzyme-deficient rodent cells and HGPRT-containing cells of other species or in the rodent cells alone in response to the conditions of cell hybridization did not include adequate controls for reversions in mutant genes of the rodent cells. Questions about the epigenetic and genetic origins of analogue-resistance are mostly unanswered. It remains possible that some kinds of abnormal epigenetic changes cause somatic disease. Specific methods for detecting their occurrence and responsiveness to environmental factors should be devised by focusing efforts on traits that are normally subject to epigenetic regulation. Derepression of genes on the inactive X-chromosome and of liver phenylalanine hydroxylase production are presented as possible examples of abnormal epigenetic changes that could be quantitatively studied by direct selection in vitro.  相似文献

Sixty-seven specimens of the common North Atlantic asteroid, Asterias vulgaris, were collected at seasonal intervals over a 2-year period and their testes observed with both light and electron microscopy. In the germinal epithelium, a predictable series of interactions between versatile somatic cells and germinal cells is repeated annually in relation to sequential events in spermatogenesis. For example, massive proliferation and differentiation of spermatogenic cells depend on the elaboration of thousands of spermatogenic columns, which are distinct cellular subdivisions of the germinal epithelium. Each fully developed column is composed of at least one somatic cell surrounded by ≈? 400 germinal cells. Such columns form only after intensive spermatogonial mitosis begins in the germinal epithelium. Single annual periods of spermatogenic proliferation and differentiation are initiated from 1 to 3 months out of phase in different individuals and overlap incompletely. Therefore, it is possible to observe testes that are entirely in the proliferative phase, entirely in the differentiative phase, or in both phases simultaneously. Detailed ultrastructural observations and preliminary autoradiographic data demonstrate that columns maintain their height for a variable period of time as germinal cells are generated near their bases, pass along their lengths, and differentiate near their tips; therefore, simultaneous proliferation and differentiation of more than one generation of germinal cells occur in the same column. Finally, formation of primary spermatocytes ceases basally, (terminating proliferation), and remaining columns degrade completely as germinal cells composing them differentiate or are phagocytized (terminating differentiation and spermatogenesis); resulting spermatozoa ultimately accumulate in the expandable lumen. It is proposed that spermatogenic columns provide the structural basis for organization of the microenvironment of small groups of spermatogenic cells (≈? 400 at a time) during proliferation and differentiation. Preliminary evidence from A. vulgaris and other species also suggests that somatic cells are temporally pluripotent and are variously involved in the formation, structure, and activities of columns, in extensive phagocytosis, and probably in contributing intrinsic (e.g., 1-methyl adenine and steroids) and mediating extrinsic (e.g., gamete shedding substance and nutrients) microenvironmental factors influential during spermatogenesis in asteroids. The prodigious spermatogenic capabilities of asteroids apparently depend on the generation of spermatogenic columns, on the progressive interaction of germinal and somatic cells before, during, and after columns form, and on the predictable effects of microenvironmental factors received and interpreted at the structural level of the spermatogenic column.  相似文献

Fiber cells of the lens are electrically and diffusionally interconnected through extensive gap junctions. These junctions allow fluxes of small solutes to move between inner cells and peripheral cells, where the majority of transmembrane transport takes place. We describe here a method utilizing two intracellular microelectrodes to measure the cell to cell resistance between fiber cells at any given distance into the intact lens. We also use ion-sensitive microelectrodes to record intracellular pH at various depths in the intact lens. We find that gap junctions connecting inner fiber cells differ in pH sensitivity as well as normal coupling resistance from those connecting peripheral cells. The transition occurs in a zone between 500 and 650 microns into the lens. Fiber cells peripheral to this zone have a specific coupling resistance of 1.1 omega cm2, whereas those inside have a specific coupling resistance of 2.7 omega cm2. However, when the cytoplasm of fiber cells is acidified by bubbling with CO2, peripheral cells uncouple and the cell to cell resistance goes up more than 40-fold, whereas junctions inside this zone are essentially unaffected by changes in intracellular pH. In a normal frog lens, the intracellular pH in fiber cells near the lens surface is 7.02, a value significantly alkaline to electrochemical equilibrium. Our data suggest that Na/H exchange and perhaps other Na gradient-dependent mechanisms in the peripheral cells maintain this transmembrane gradient. Deep in the lens, the fiber cell cytoplasm is significantly more acidic (pHi 6.81) due to influx of hydrogen across the inner fiber cell membranes and production of H+ by the inner fiber cells. Because of the normally acid cytoplasm of interior fiber cells, their loss of gap junctional sensitivity to pH may be essential to lens survival.  相似文献

A two-element transposon system based on the maize elements Ac and Ds is currently being used for insertional mutagenesis in Arabidopsis. With the aim of making this system as efficient as possible we have continued to analyse several parameters which affect Ds activity in Arabidopsis. The influence of genomic position on Ds excision has been analysed in five lines carrying Ds integrated in different genomic locations. Differences in both somatic and germinal excision were observed between the different lines. The relationship between somatic and germinal excision, the timing of excision events and environmental influences on transposition frequency have been investigated. The effect of varying dosage of the different elements was also analysed. A strong positive dosage effect was observed for the transposase source, but not for the Ds element. Analysis of germinal excision events showed that the majority of them occurred very late in the development of the plant, resulting in the majority of Ds transpositions being independent events.  相似文献

Contralateral cerebellectomy can induce hypertrophy of olivary neurons in cat. In the present study we examined the ultrastructure of the cat hypertrophic inferior olive following GABA-, dopamine- and serotonin-immunocytochemistry, anterograde tracing from the mesodiencephalic junction, and intracellular labeling with HRP. Compared to normal olivary neurons the hypertrophic cells showed larger cell bodies, more and longer somatic spines which were linked by gap junctions, and longer distal dendrites with relatively few spines. The hypertrophic olivary neurons received less GABAergic boutons on their dendrites but an equal percentage was apposed to their somata as compared to normal cells. Relatively many mesodiencephalic terminals, a similar serotoninergic, and a slightly increased dopaminergic input were found. The axon of one intracellularly labeled hypertrophic cell gave off recurrent collaterals bearing varicosities filled with vesicles. These results indicated that 1) hypertrophic olivary cells are affected by trophic factors not only at the cell body but also at the level of the somatic spines, dendrites, and axon, 2) the ratio of excitatory to inhibitory terminals is increased in the hypertrophic neuropil, whereas the monoaminergic input remains stationary, and 3) the electronic coupling between hypertrophic olivary neurons has shifted from a dendritic to a more somatic location due to a relatively high number of gap junctions between the somatic spines.  相似文献

Cardiac cells with distinctive electrophysiological and morphological features were found at the junctional region between Purkinje and ventricular cells of the dog heart. The electrophysiological exploration of these "transitional" cells revealed action potentials markedly different in configuration from those generated by Purkinje or by ventricular cells. The impaled cardiac cells which generated transitional action potentials were identified in serial sections and studied with the light and the electron microscopes. The transitional cells were found to be characterized cytologically by: (a) their subendocardial location, (b) their small diameter, (c) the absence of T system and sarcoplasmic reticulum, and (d) the lack of intercalated discs under the light microscope and the sparsity of specialized intercellular junctions under the electron microscope. Purkinje, transitional, and ventricular cells were found to be joined by gap junctions permeable to lanthanum. A quantitative difference in the extent and distribution of specialized intercellular junctions may be one of the factors responsible for the slow velocity of conduction characteristic of the Purkinje-ventricular junctional region.  相似文献
