In pH 5.0–5.4 HAc–NaAc buffer solution, lincomycin (Linco) reacted with Pd(II) to form 1:1 cationic chelate, which could further react with erythrosine (Ery) to form 1:1 ion‐association complexes (Pd–Linco)Ery. As a result, not only were the absorption and fluorescence spectra changed, but also the resonance Rayleigh scattering (RRS) intensity was greatly enhanced. These phenomena offered useful means for the determination of Linco by spectrophotometry, fluorescence and RRS methods. The linear range and detection limit of Linco were 0.20–3.00 µg/mL and 0.057 µg/mL, 0.20–4.80 µg/mL and 0.061 µg/mL, 0.05–2.70 µg/mL and 0.015 µg/mL for the spectrophotometric, fluorescence quenching and RRS methods, respectively. Among these, the RRS method obtained the highest sensitivity. Therefore, the optimum reaction conditions and the influences of coexisting substances were investigated using the RRS method. A simple, sensitive and rapid method has been developed for the determination of Linco in either the pharmaceutical form or human body fluids, and the reasons for RRS enhancement are discussed. The interaction of benproperine phosphate (BPP) with eriochrome blue black R (EBBR) in the presence of sodium dodecylbenzene sulphonate (SDBS) was studied using resonance light scattering (RLS) technology and ultraviolet‐visual (UV‐vis) spectrophotometry. Under optimum conditions, BPP reacts with EBBP and SDBS to form a three‐component complex, which results in strong RLS signal and a new RLS peak. The enhanced RLS intensities are proportional to the concentration of BPP over the range 0.6–28.0 µg/mL, with a detection limit of 0.053 µg/mL. The affecting factors as well as the influence of coexisting substances were investigated. The results indicate that this assay method could be applied to the determination of BPP in pharmaceuticals, serum and urine samples with satisfactory results. A simple, sensitive and rapid spectrofluorimetric method was developed for the determination of esomeprazole (EMZ) and pantoprazole (PRZ) in their pharmaceutical formulations and human plasma. The proposed method is based on the fluorescence spectral behavior of EMZ in methanol in the presence of 0.1 m NaOH containing 0.5% methyl cellulose (MC) at 306/345 nm. The fluorescence intensity of EMZ was enhanced about 1.3‐fold and good linearity in the range 0.4–4.0 µg/mL with a lower detection limit of 0.04 µg/mL and lower quantification limit of 0.14 µg/mL. For PRZ, its methanolic solution exhibited marked native fluorescence at 290/325 nm after enhancement (about 2.1‐ or 1.4‐fold) using either 0.025% sodium dodecyl sulfate (SDS) or 0.05% MC in the presence of 0.2 m borate buffer of pH 9.5. The fluorescence–concentration plots of PRZ were rectilinear over the ranges 0.2–2.0 and 0.3–3.0 µg/mL with lower detection limits of 0.02 and 0.03 µg/mL and lower quantification limits of 0.07 and 0.09 µg/mL using sodium dodecyl sulfate and MC, respectively. The method was successfully applied to the analysis of EMZ and PRZ in their commercial dosage forms and the results were in good agreement with those obtained with the comparison method. Furthermore, in a preliminary investigation, the proposed method was extended to the in vitro determination of the two drugs in spiked human plasma and the results were satisfactory. The fluorescence intensity of the morin-Al(3+) complex was greatly enhanced by proteins in the presence of sodium dodecyl benzene sulphonate (SDBS). Based on this, a new fluorimetric method for the determination of protein was developed. Under optimum conditions, the enhanced intensity of fluorescence was in proportion to the concentration of proteins in the range 1.0 x 10(-8)-1.3 x 10(-5) g/mL for bovine serum albumin (BSA), 4.0 x 10(-8)-1.2 x 10(-5) g/mL for egg albumin (EA) and 5.0 x 10(-8)-1.2 x 10(-5) g/mL for human serum albumin (HSA). Their detection limits (S:N = 3) were 5.0 x 10(-9), 1.8 x 10(-8) and 1.6 x 10(-8) g/mL, respectively. The interaction mechanism was also studied.  相似文献

In this paper, a sensitive resonance light scattering (RLS) method for the determination of protein is reported. In the Tris–HCl (pH 7.50) buffer, protein enhanced the RLS intensity of the Y3+–2‐thenoyltrifluoroacetone (TTA)–sodium dodecyl sulphate (SLS) system. The enhanced RLS intensities were in proportion to the concentrations of proteins in the range 8.0 × 10?9–1.0 × 10?5 g/mL for BSA, 1.0 × 10–8–1.0 × 10?5 g/mL for HSA and 1.0 × 10–8–1.0 × 10?6 g/mL for EA, and their detection limits were 5.0, 5.4 and 6.7 ng/mL, respectively. A simple and sensitive spectrofluorimetric method was developed for the determination of Ribavirin in pharmaceutical formulations. The proposed method was based on the fluorescence spectral behaviour of Ribavirin in a sodium dodecyl sulfate (SDS) micellar system. In an aqueous acetate buffer solution of pH 4.0, the fluorescence intensity of Ribavirin was significantly enhanced by about 217% in the presence of SDS. Fluorescence intensity was measured at 396 nm after excitation at 270 nm for Ribavirin. The fluorescence‐concentration plot was rectilinear over the range of 0.01‐3.0 µg/mL for Ribavirin with a lower detection limit of 5.02 x 10‐3 µg/mL. The method was successfully applied to the analysis of the drug in its commercial capsules. Results were in good agreement with those obtained with the official method. The application of the proposed method was extended to stability studies of Ribavirin after exposure to different forced degradation conditions such as acidic, alkaline, photo and oxidative conditions according to ICH guidelines. The fluorescence of acridine orange (AO) is greatly quenched by the anionic surfactant sodium dodecyl benzene sulphonate (SDBS), but when protein is added into the AO-SDBS system, the fluorescence intensity of the latter is enhanced again. Based on this, a new fluorimetric method of determination of protein was developed. Under optimum conditions, the enhanced intensity of fluorescence is in proportion to the concentration of protein, such as bovine serum albumin (BSA), human serum albumin (HSA) and egg albumin (EA), over a wide range with detection limits at the 10(-9) g/mL level. This method has been satisfactorily used for the determination of protein in samples. We compared results using 280 nm and 490 nm excitation wavelengths and the mechanism of the assay.  相似文献

A europium‐sensitized fluorescence spectrophotometry method using an anionic surfactant, sodium dodecyl benzene sulphonate (SDBS), was developed for the determination of gatifloxacin (GFLX). The GFLX–Eu3+–SDBS system was studied and it was found that SDBS significantly enhanced the fluorescence intensity of the GFLX–Eu3+ complex (about 25‐fold). The optimal experimental conditions were determined as follows: excitation and emission wavelengths of 338 and 617 nm, pH 7.5, 3.0 × 10–6 mol/L europium(III), and 5.0 × 10–5 mol/L SDBS. The enhanced fluorescence intensity of the system (ΔIf) showed a good linear relationship with the concentration of GFLX over the range 1.0 × 10–8–8.0 × 10–7 mol/L with a correlation coefficient of 0.9990. The detection limit (S:N = 3) was determined as 1.0 × 10–9 mol/L. This method has been successfully applied for the determination of GFLX in pharmaceuticals and human urine/serum samples. Compared with most other methods reported, the rapid and simple procedure proposed here offered higher sensitivity, wider linear range and good stability. This is the first report of terbium(III) as a probe of second-order scattering (SOS) for the determination of proteins in human serum at nanogram levels. A sensitive method has been developed using light scattering, based on the fact that the weak SOS of proteins can be enhanced in the presence of terbium(III) and sodium dodecyl sulphonate (SDS). With this method, 7.0 x 10(-9)-1.0 x 10(-5) g/mL human serum albumin (HSA) and 5.0 x 10(-9)-5.0 x 10(-6) g/mL gamma-globulin can be determined; the detection limits were 4.4 ng/mL for HSA and 3.1 ng/mL for gamma-globulin. The method has been applied to the detection of total proteins in human serum samples, and the results are consistent with those obtained by the Coomassie brilliant blue (CBB) G-250 assay.  相似文献

A simple, rapid and precise flow‐injection–chemiluminescence (FI–CL) method is presented for the determination of tenoxicam in pharmaceutical preparations and biological samples. The method is based on the weak chemiluminescence signal arising from the reaction of cerium(IV) in a nitric acid medium with sodium hyposulphite being significantly increased by tenoxicam in the presence of sodium dodecyl benzene sulphonate. Several experimental parameters affecting the CL reaction were examined and optimized systematically. Under the optimum conditions, the CL intensity was proportional to the concentration of tenoxicam in the range 7.0 × 10–11–5.0 × 10–8 g/mL. The detection limit was 2.3 × 10–11 g/mL tenoxicam and the relative standard deviation (RSD) was 2.1% for 1.0 × 10–9 g/mL tenoxicam solution (n = 11). The proposed method was applied to the determination of tenoxicam in pharmaceutical preparations, serum and human urine, with satisfactory results. A highly sensitive and simple spectrofluorimetric method was developed for the determination of loratadine (LRT) and desloratadine (DSL) in their pharmaceutical formulations. The proposed method is based on investigation of the fluorescence spectral behaviour of LRT and DSL in a sodium dodecyl sulphate (SDS) micellar system. In aqueous solution of acetate buffer of pH 4.5, the fluorescence intensities of both LRT and DSL were greatly enhanced (240%) in the presence of SDS. The fluorescence intensity was measured at 438 nm after excitation at 290 nm for both drugs. The fluorescence–concentration plots were rectilinear over the range 0.05–2.0 µg/mL for both LRT and DSL, with lower detection limits of 5.13 × 10?3 and 6.35 × 10?3 µg/mL for LRT and DSL, respectively. The method was successfully applied to the analysis of the two drugs in their commercial tablets, capsules and syrups, and the results were in good agreement with those obtained with the official or comparison methods. The proposed method is specific for the determination of LRT in the presence of other co‐formulated drugs, such as pseudoephedrine. The application of the proposed method was extended to stability studies of LRT and DSL after exposure to different forced degradation conditions, such as acidic, alkaline and oxidative conditions, according to ICH guidelines. A highly sensitive, simple and rapid spectrofluorimetric method was developed for the determination of Amlexanox (AMX) in its bioadhesive buccal tablets. The proposed method is based on measuring the native fluorescence of the methanolic solution of AMX at 400 nm after excitation at 242 nm in 0.2 M borate buffer (pH 10) and 0.5% w/v sodium dodecyl sulfate (SDS) solution. The interaction of AMX with SDS was studied, and the enhanced fluorescence intensity was exploited to develop an assay method for the determination of AMX. The relative fluorescence intensity–concentration plot was rectilinear over the range 5.0–80.0 ng/mL, with a lower detection limit of 0.57 ng/mL and a lower quantification limit of 1.74 ng/mL. The proposed method was successfully applied to the analysis of AMX in its commercial tablets. Moreover, content uniformity testing was conducted by applying official USP guidelines. Statistical evaluation and comparison of the data obtained using the proposed and comparison methods revealed good accuracy and precision for the proposed method. A sensitive and simple chemiluminescent (CL) method for the determination of diclofenac sodium has been developed by combining the flow injection technique and its sensitizing effect on the weak CL reaction between formaldehyde and acidic potassium permanganate. A calibration curve is constructed for diclofenac sodium under optimized experimental parameters over the range 0.040–5.0 µg/mL and the limit of detection is 0.020 µg/mL (3σ). The inter‐assay relative standard deviation for 0.040 µg/mL diclofenac sodium (n = 11) is 2.0%. This method is rapid, sensitive, simple, and shows good selectivity and reproducibility. The proposed method has been successfully applied to the determination of the studied diclofenac sodium in pharmaceutical preparations with satisfactory results. This work proposes a simple and sensitive solid substrate–room temperature phosphorimetry (SS–RTP) for the selective determination of carvedilol (CV). The method is based on the sensitizing effect of sodium dodecyl benzene sulphonate (SDBS) on CV to activate the oxidation between NaClO and amaranth, resulting in the intense quenching of room temperature phosphorescence (RTP) of the system. Compared with non‐SDBS system, the reduction of phosphorescence intensity (ΔIp) with SDBS is 16.5 times higher and is directly proportional to the content of CV, covering a wide range 0.080–16.00 fg/spot. The regression equation of the working curve can be expressed as ΔIp = 0.7780 + 7.057 mCV (fg/spot) (correlation coefficient (r) = 0.9976, n = 8), with a detection limit (LD) of 0.020 fg/spot (corresponding concentration is 5.1 × 10−14 g/mL, sample volume is 0.40 μL/spot). This sensitive method has also been applied to determine trace CV in human plasma and the results agreed with synchronous fluorimetry (SF). The activation energy (E) and rate constant (k) of this activating reaction were 69.04 kJ/mol and 3.580 × 10−4 s−1, respectively. The reaction mechanism is also discussed. A terbium‐sensitized spectrofluorimetric method has been developed for determination of catecholamines such as norepinephrine (NE), epinephrine (EP) and dopamine (DA), using sodium dodecyl benzene sulphonate (SDBS). Fluorescence sensitization of terbium ions (Tb3+) by complexation with catecholamines in the presence of SDBS was observed. The fluorescence intensities of the Tb3+–catecholamine complexes were highly enhanced by introducing SDBS with an emission maximum at 545 nm after excitation at 290 nm. The conditions for the complex formation of Tb3+–catecholamine were investigated systematically and optimized to determine catecholamines in a serum sample. Under the optimum conditions, the fluorescence intensities of the Tb3+–catecholamine complexes were increased linearly with the concentration of NE, EP and DA over the ranges 2.5 × 10–10–1.0 × 10–8, 2.5 × 10–10–1.0 × 10–8 and 2.5 × 10–9–1.0 × 10–7 g/mL with correlation coefficients of 0.999, 0.999 and 0.9996, respectively. The limits of detection (3δ) of NE, EP and DA were found to be 4.6 × 10–11, 7.8 × 10–11 and 8.38 × 10–10 g/mL, respectively. Precision of the method was tested at the concentration level of 1.2 × 10?7 g/mL for five replicate measurements of NE, EP and DA, giving relative standard deviations (RSDs) of 1.41%, 1.23% and 1.89%, respectively. The interaction mechanism of the Tb3+–catecholamine complexes system was investigated and presented with ultraviolet absorption spectra. The proposed method has been applied for the quantitative determination of NE, EP and DA in a spiked serum sample and a pharmaceutical preparation sample. A simple and selective aptamer (ssDNA)‐modified nanogold probe (AussDNA) was prepared for the determination of trace As(III) in HEPES buffer solution (pH 8.2) containing 0.05 mol/L NaCl. The method coupled the aptamer reaction of AussDNA–As(III) and the resonance Rayleigh scattering (RRS) of nanogold aggregations at 278 nm. When the As(III) concentration increased, the RRS intensity at 278 nm increased to form more nanogold aggregation and a stable As(III)–ssDNA complex. Under selected conditions, the increased RRS intensity (ΔI) was linear to the concentration of As(III) in the range 3.8–230.4 ng/mL, with a detection limit of 1.9 ng/mL. An enhanced thiosemicarbazide(TSC)–H2O2 chemiluminescence (CL) system was established and proposed as a new analytical method for determination of β‐lactam antibiotics, ampicillin sodium and amoxicillin at microgram levels. The method is based on the inhibition of CL emission accompanying oxidation of TSC by H2O2 in alkaline medium. The effect of anionic, cationic, and non‐ionic surfactants on the CL emission of the system was studied. Both N‐cetyl‐N,N,N‐trimethylammonium bromide (CTMAB) and Triton X‐100, unlike sodium dodecyl sulfate (SDS), reinforced the CL intensity and were efficient to approximately the same level. The effect of the presence of eight non‐aqueous solvents on the CL system was also investigated. Upon addition of both of the non‐ionic surfactant, Triton X‐100, and the non‐aqueous solvent, N,N‐dimethyl formamide (DMF), the intensity of the CL reaction was increased 100‐fold. This method allows the measurement of 25–545 µg amoxicillin, and 35–350 µg ampicillin sodium. The detection limits are 8 µg for amoxicillin and 9 µg for ampicillin sodium. The relative standard deviations of six replicate measurements of 200 µg amoxicillin and 200 µg ampicillin sodium were 1.9 and 2.1%, respectively. The effect of foreign species on the determination of amoxicillin and ampicillin sodium was also examined. In pH 4.99‐6.06 Britton‐Robinson (BR) buffer medium, 6‐benzylaminopurine (6‐BA) reacted with Na2WO4 to form 1:1 anionic chelate (6‐BA·WO4)2‐, which further reacted with rhodamine 6G to form ternary ion complexes at room temperature. This resulted in a significant enhancement of resonance Rayleigh scattering (RRS) with a maximum RRS wavelength of 316 nm. Meanwhile, the fluorescence of the solution was quenched and excitation (λex) and emission (λem) wavelengths of the fluorescence were 290 and 559 nm, respectively. Intensities of RRS enhancing (ΔIRRS) and fluorescence quenching (ΔIF) were directly proportional to concentrations of 6‐BA. As a result, RRS and fluorescence quenching for determination of trace amounts of 6‐BA were developed. Under optimal conditions, linear ranges and detection limits of the two methods were 0.05‐15.00 µg/mL and 8.2 ng/mL (RRS), 0.50‐15.00 µg/mL and 17.0 ng/mL, respectively. It was found that the RRS method was superior to fluorescence quenching. The influence of these methods were investigated and results showed that RRS had good selectivity. RRS was applied to determine 6‐BA in vegetable samples with satisfactory results. Furthermore, the reaction mechanisms of the ternary ion‐association system are discussed. In addition, the polarization experiment revealed that the resonance light scattering (RLS) peak of Na2WO4‐6‐BA‐R6G consisted mainly of depolarized resonance fluorescence and resonance scattering. It was speculated that light emission fluorescence energy (EL) transformed into resonance light scattering energy (ERLS), which was a key reason for enhancement of RRS. A new flow injection chemiluminescent method has been developed for the determination of procaine hydrochloride, based on the inhibition of the chemiluminescence reaction of luminol–hydrogen peroxide by procaine hydrochloride. The influence of several surfactants and β‐cyclodextrin on the chemiluminescence intensity were studied. It was found that β‐cyclodextrin enhanced the decrease in chemiluminescence intensity. The method is simple, convenient and sensitive, with a detection limit (3 σ) of 0.08 µg/mL. The decreased chemiluminescence intensity is linear, with the concentration of procaine hydrochloride in the range 0.2–100.0 µg/mL and 100.0–400.0 µg/mL. The relative standard deviation for 10 repeated measurements were 4.5% and 3.4% for 1.0 and 20.0 µg/mL procaine hydrochloride, respectively. Graphite oxide (GO) was prepared by the Hummer procedure, and can be dispersed to stable colloid solution by ultrasonic wave. The GO exhibited an absorption peak at 313 nm, and a resonance Rayleigh scattering (RRS) peak at 490 nm. In pH 4.6 HAc‐NaAc buffer solution, human serum albumin (HSA) combined with GO probe to form large HSA‐GO particles that caused the RRS peak increasing at 490 nm. The increased RRS intensity was linear to HSA concentration in the range 0.50–200 µg/mL.
