Summary The distribution of renin and angiotensin II (ANG II) in juxtaglomerular epitheloid cells of control and adrenalectomized rats was studied, using specific antisera and the protein A-gold technique in Lowicryl- and glycol methacrylate-embedded tissue.The matrix of virtually all mature secretory granules of epitheloid cells contains not only renin, but also ANG II. On adrenalectomy, the concentration of both renin and ANG II in the granule internum increases markedly, as indicated by the density of the immunolabel.Given the coexistence of renin and ANG II in the granule matrix, it is quite probable that, with each secretory event, a certain amount of ANG II is released together with renin. Further experiments will have to show if this amount of ANG II cosecreted with renin is sufficient to elicit immediate local intrarenal actions.ANG I, as well as angiotensinogen and converting enzyme, were not found in epitheloid cells. It is therefore inferred that ANG II is not generated intracellularly, but within the extracellular space and subsequently taken up by pinocytosis and incorporated into the secretory granules of epitheloid cells.These studies were supported by the Deutsche Forschungsgemeinschaft within the SFB 90 Cardiovasculäres System  相似文献

We produced three monoclonal antibodies, SG1, SG2 and SG3, specific for human seminal -glutamyltransferase when characterized by enzyme-linked immunosorbent assay and immunoblotting. Seminal -glutamyltransferase was localized, by immunostaining, to the epithelial cells of the ductus epididymidis, seminal vesicle and prostate gland with SG1, those of the prostate gland with SG2, and those of the seminal vesicle with SG3. Rabbit polyclonal anti-seminal -glutamyltransferase serum reacted with the proximal convolution of the kidney and the bile capillaries of the liver, and with the epithelial cells of the reproductive organs. However, immunoreactivity was not observed in the kidney or liver with the monoclonal antibodies. Thus, these monoclonal antibodies are probably all specific to seminal -glutamyltransferase but recognize different epitopes.  相似文献

Summary We have localized the enzyme renin (EC 3.4.99.19) in the normal adult human kidney by immunohistology. Serial paraffin sections of kidneys were incubated with renin antisera and then processed by the peroxidase-antiperoxidase method. Renin immunoreactivity was observed in the juxta-glomerular epithelioid granular cells (JEG-cells) in the wall of the afferent and rarely of the efferent vessel of the glomerulus. JEG-cells have long cytoplasmic processes penetrating between adjacent cells. This suggests a possible paracrine release of renin. Staining of various segments of the tubular system was shown to be artifactual. The kidney proteins recognized by our anti-human renin antisera had similar characteristics to renin when determined by a combination of gel-electrophoretic and immunologic techniques. Renin immunostaining in the juxtaglomerular apparatus of the human kidney is discrete and reflects the low amount of extractable renin.Dedicated to Prof. G. Summary The PAP-method was used for immunocytochemical investigations with antisera against angiotensin (ang) I, ang II and renin in kidneys of rats and mice. In 14 rats, ang II was found in the media of the afferent arteriole — both in the region of the JGA and upstream until the interlobular artery. Serial sections alternately reacted for ang II and renin revealed that the octapeptide is contained in the well known renin positive epitheloid cells of the afferent arteriole and, beyond that, together with renin probably in the same specific granules. Fixation conditions were critical for the visualization of immunoreactivity. With ang I antisera, comparable in terms of titer and affinity to the ang II antisera, specific immunoreactivity could not be found in the kidneys of rats. With horse radish peroxidase and ferritin as tracers it could be shown that the epitheloid cells of the JGA have the ability to pinocytize and incorporate macromolecules into their granules. It is suggested that ang II is taken up by these cells through the same route. Intracellular generation of ang II appears unlikely as an explanation. Functionally the selective uptake of ang II by epitheloid cells might be a specific process, possibly connected with the negative feedback of the octapeptide on renin secretion. Negative results in mice may be explained by a small uptake or more rapid degradation of ang II by the epitheloid cells.These studies were supported by the Deutsche Forschungsgemeinschaft within the SFB 90 Cardiovasculäres System. Summary Angiotensin II (ANG II) was localized immunocytochemically in kidney and various other organs of the chinese hamster. In the kidney ANG II-like activity was found in the epitheloid cells of the juxtaglomerular apparatus as well as in the media i.e. the smooth muscle cells of arcuate and interlobular arteries and afferent arterioles. ANG II-like activity was also observed in the medial muscle cells of resistance vessels in other organs and tissues such as submandibular gland and brown adipose tissue. The site of synthesis of ANG II needs to be investigated but the data point to the possibility of an intracellular function of ANG II in smooth muscle cells of blood vessels.These studies were supported by the Deutsche Forschungsgemeinschaft within the SFB 90 Cardiovasculäres System  相似文献

Summary Monospecific antisera and the immunocytochemical PAP-method have been used to localize renin in the kidneys of mice. With this procedure, reaction product was not only observed in the epitheloid cells of kidney vessels but also in kidney tubules: in the apical portion of proximal tubule cells and in some cells of the connecting and the cortical collecting tubule. To answer the question, whether the occurrence of renin in kidney tubule cells is the consequence of tubular synthesis or that of glomerular filtration of plasma renin followed by its uptake from the tubular lumen, tracer experiments with radioiodinated renin and with horseradish peroxidase were undertaken. The results of these studies as well as other arguments suggest reabsorptive pinocytosis of the filtered hormone as the source of tubular renin.These studies were supported by the Deutsche Forschungsgemeinschaft within the SFB 90 Cardiovasculäres System  相似文献

Summary The present study was undertaken to investigate the effects of sarmesin, an analogue of [Sar1] angiotensin II (ANG II) where the tyrosine hydroxyl group in position 4 is methylated, on dopamine (DA)-related paradigms: locomotor and exploratory behaviour as well as apomorphine (3 mg/kg, ip)-induced stereotypy in rats. Sarmesin (0.5 and 1 g, icv) significantly decreased ambulation and rearing movements, and blocked the inhibitory effect of ANG II (0.1 g) on both types of activity. Sarmesin induced biphasic effects on apomorphine-induced stereotypy depending on the dose increase (0.5 and 5 g, icv) and decrease (10 g). Moreover, sarmesin (5 g) blocked the inhibitory effect of ANG II (2 g, icv) on apomorphine stereotypy. Taken together, these results suggest that sarmesin might interact with AT1 and AT2 receptor subtypes. The results further confirm the statement for ANG II-DA interaction in brain structures involved in these types of behaviour.  相似文献

The in vitro effects of -L-glutamyltaurine on different stages of excitatory aminoacidergic neurotransmission were tested with -D-glutamyltaurine as reference. -L-Glutamyltaurine enhanced the K+-stimulated release of [3H]glutamate from cerebral cortical slices (25% at 0.1 mM) and slightly inhibited the uptake by crude brain synaptosomal preparations (about 10% at 1 mM). -L-Glutamyltaurine was also a weak displacer of glutamate and its agonists from their binding sites in brain synaptic membrane preparations, being, however, less selective to quisqualate (QA) sites than -D-glutamyltaurine. The basal influx of Ca2+ into cultured cerebellar granular cells was not affected by 1 mM -L-glutamyltaurine, but the glutamate- and its agonist-activated influx was significantly inhibited in low-Mg2+ (0.1 mM) and Mg2+-free media. The glutamate-evoked increase in free intracellular Ca2+ and the kainate-activated formation of cGMP in cerebellar slices were both markedly inhibited by 0.1 mM -L-giutamyltaurine. We propose that -L-glutamyltaurine may act as endogenous modulator in excitatory aminoacidergic neurotransmission.  相似文献

Endothelin-1 (ET-1) has been found to increase cardiac -myosin heavy chain (-MyHC) gene expression and induce hypertrophy in cardiomyocytes. ET-1 has been demonstrated to increase intracellular reactive oxygen species (ROS) in cardiomyocytes. The exact molecular mechanism by which ROS regulate ET-1-induced -MyHC gene expression and hypertrophy in cardiomyocytes, however, has not yet been fully described. We aim to elucidate the molecular regulatory mechanism of ROS on ET-1-induced -MyHC gene expression and hypertrophic signaling in neonatal rat cardiomyocytes. Following stimulation with ET-1, cultured neonatal rat cardiomyocytes were examined for 3H-leucine incorporation and -MyHC promoter activities. The effects of antioxidant pretreatment on ET-1-induced cardiac hypertrophy and mitogen-activated protein kinase (MAPKs) phosphorylation were studied to elucidate the redox-sensitive pathway in cardiomyocyte hypertrophy and -MyHC gene expression. ET-1 increased 3H-leucine incorporation and -MyHC promoter activities, which were blocked by the specific ETA receptor antagonist BQ-485. Antioxidants significantly reduced ET-1-induced 3H-leucine incorporation, -MyHC gene promoter activities and MAPK (extracellular signal-regulated kinase, p38, and c-Jun NH2 -terminal kinase) phosphorylation. Both PD98059 and SB203580 inhibited ET-1-increased 3H-leucine incorporation and -MyHC promoter activities. Co-transfection of the dominant negative mutant of Ras, Raf, and MEK1 decreased the ET-1-induced -MyHC promoter activities, suggesting that the Ras-Raf-MAPK pathway is required for ET-1 action. Truncation analysis of the -MyHC gene promoter showed that the activator protein-2 (AP-2)/specificity protein-1 (SP-1) binding site(s) were(was) important cis-element(s) in ET-1-induced -MyHC gene expression. Moreover, ET-1-induced AP-2 and SP-1 binding activities were also inhibited by antioxidant. These data demonstrate the involvement of ROS in ET-1-induced hypertrophic responses and -MyHC expression. ROS mediate ET-1-induced activation of MAPK pathways, which culminates in hypertrophic responses and -MyHC expression. We examined physical condition, niche dimensions, and survival of New England cottontails (Sylvilagus transitionalis) that occupied 21 habitat patches of different sizes during winter. Rabbits on small patches (2.5 ha) were predominantly males, and both sexes had lower body mass than individuals on large patches (5.0 ha). Niche indices (, where ranges from 0 to 1. and values approaching 1 indicate generalized resource use) of habitat use revealed that rabbits on small patches used a greater variety of microhabitats (based on understory stem density: s, and proximity to cover: c) than rabbits occupying large patches (s=0.65, c=0.66). Rabbits on small patches also consumed low quality forage more often and fed at sites farther from escape cover than rabbits on large patches. There were no significant correlations between rabbit densities and niche dimensions. Niche expansion was not a result of compertitive release or relaxation of predator pressure. Rabbits on small patches apparently modified their niche dimensions in response to resource limitations. This response included occupying sites with limited understory cover that apparently resulted in rabbits on small patches having a lower survival rate (0.35) than rabbits on large patches (0.69) during a 10-week monitoring period. Skewed sex ratios and low survival rates among rabbits on small patches suggest that these habitats act as sinks to dispersing, juveniles from large (source) patches. As a result, local populations of New England cottontails may become vulnerable to extinction if larte patches of habitat are not maintained.  相似文献

Companion animal overpopulation is a problem of human creation with significant human costs that can only be addressed through human action. A model was constructed to understand the dynamics of canine overpopulation and the effectiveness of various policy options for reducing euthanasia. The model includes economic and ecological factors in human and dog populations. According to the model, a no-kill society is an achievable goal at an acceptable human cost. Spay/neuter programs were generally found to be the most effective, with increasing adoptions also being an effective option. However, spay/neuter policies need to be evaluated over a very long time horizon since full impact may not be achieved for 30 years or more. Spay/neuter efforts can have a large impact even if they only effect a small portion of the human population. Adoption and spay/neuter programs were found to work well in combination, and to continue being effective as society approaches no-kill dynamics.  相似文献

The I.29 cell line is a nonsecreting B-cell leukemia which bears two different immunoglobulin isotypes on its surface, IgM and IgX. The I.29 cells were hybridized with nonsecreting myeloma cells giving rise to dozens of immunoglobulin secreting hybridomas. These fall into three groups differing in the class of immunoglobulin they secrete. Cells of the first group secrete pentameric IgM (, ), those of the second group secrete an unknown immunoglobulin, IgX, which may constitute an allotype of IgA, and those of the third group produce light chains only. Summary In immunocytochemical experiments on human kidney tissue with an antiserum directed against the prosegment of renin, only juvenile granules were clearly labeled. Summary Imidazolides of dinucleotides such as ImpApA can be formed from the corresponding dinucleotides in a two-stage process, which gives up to 15% yields under potentially prebiotic conditions. First a solution of the dinucleotide and sodium trimetaphosphate is dried out at constant temperature and humidity. This produces polyphosphates such as pnApA in excellent yield (80%). The products are dissolved in water, imidazole is added, and the solution is dried out again. Photon yields of oxygen evolution at saturating CO2 were determined for 44 species of vascular plants, representing widely diverse taxa, habitats, life forms and growth conditions. The photonyield values on the basis of absorbed light ( a) were remarkably constant among plants possessing the same pathway of photosynthetic CO2 fixation, provided the plants had not been subjected to environmental stress. The mean a value ±SE for 37 C3 species was 0.106±0.001 O2·photon-1. The five C4 species exhibited lower photon yields and greater variation than the C3 species ( a=0.0692±0.004). The a values for the two Crassulaceanacid-metabolism species were similar to those of C3 species. Leaf chlorophyll content had little influence on a over the range found in normal, healthy leaves. Chlorophyll fluorescence characteristics at 77 K were determined for the same leaves as used for the photon-yield measurements. Considerable variation in fluorescence emission both at 692 nm and at 734 nm, was found 1) among the different species; 2) between the upper and lower surfaces of the same leaves; and 3) between sun and shade leaves of the same species. By contrast, the ratio of variable to maximum fluorescence emission at 692 nm (Fv/FM, 692) remained remarkably constant (The mean value for the C3 species was 0.832±0.004). High-light treatments of shade leaves resulted in a reduction in both a and the Fv/FM, 692 ratio. The extent of the reductions increased with time of exposure to bright light. A linear relationship was obtained when a was plotted against Fv/FM, 692. The results show that determinations of the photon yield of O2 evolution and the Fv/FM, 692 ratio can serve as excellent quantitative measures of photoinhibition of overall photosynthetic energy-conversion system and of photochemistry of photosystem II, respectively. Infant lesser bushbabies are typically able, from the earliest age, to utter distress vocalizations which appear to have major adaptive significance. Distress vocalizations of infant G. moholi were analyzed by means of a digital storage oscilloscope. Three kinds of sound analyses were carried out: analysis of the sound wave signal, sequential spectrum analysis, and fast Fourier transformation. We emphasize analysis of the frequency spectrum. Three distress calls were identified-the click call, the crackle, and the chirp-which we report for the first time. Click calls were typically composed of short, high-pitched pulses (clicks) uttered in fast, repetitive sequences with a broad band frequency spectrum containing frequencies up to 60 kHz. These acoustic characteristics appear to be particularly well suited for accurate spatial localization of the sound source. The main function of the click call is apparently to alert and to guide the mother to a lost or endangered infant. These basic acoustic characteristics-related to sound localization-are shared to some extent, with the other two distress calls. However, the three calls appear to represent different degrees of distress, with the click call being the basic, call, the crackle corresponding to a greater degree of distress, and the chirp possibly signaling extreme distress such as physical pain.We prefer the specific name,Galago moholi (A. Smith, 1836), for the southern African lesser bushbaby [instead ofGalago senegalensis moholi (A. Smith, 1839)] according to the new classification proposed by Olson (1979, 1986) and Nashet al. Following the widespread application and success of Bongaarts' proximate fertility framework in the 1980s, anthropologists and demographers have shown increased interest in the delineation of distal fertility variables, alternatively called higher-order by cultural ecologists or ultimate variables by evolutionary ecologists. This shift in focus raises at least four immediate issues: (1) confusion over the role and effect of culture on individual members' behavior, (2) whether the individual or the group forms the basic unit of analysis, (3) discordance between external and internal perspectives of demographic regimes, and (4) difficulty comparing and evaluating quantitative survey-based data with qualitative information derived from focus groups or key informants. This paper presents one approach to dealing with these problems, featuring the assessment of anthropological and demographic data collected for Rendille pastoralists of northern Kenya, a group long cited in both anthropological and demographic literature as regulating their fertility in relation to ecological factors.  相似文献

Observations on morphology, sterility and cytology of some interchangetrisomics obtained from the progeny of radiation induced interchangeheterozygotes in a 6-rowed barley (Hordeum vulgare L.) var. K12 are described. The different interchange trisomics show distinct phenotypic expression and have been classified as bushy, slender, robust, semi-erect and pseudonormal on the basis of their gross morphology, leaf characteristics, awn and spike lengths. All interchange trisomics show pollen and ovule sterility which varies between plants. Meiotic behaviour is described. The five different interchange trisomics appear to have different extra chromosomes.  相似文献

Summary Availability of seed and microsites, respectively, are two factors that potentially may limit recruitment in plant populations. Microsites are small-scale sites suitable for germination and survival of seedlings. We discuss this dichotomy of recruitment limitation both from a theoretical and empirical point of view. Investigations of recruitment in 14 woodland species showed that 3 species were seed limited, 6 species were limited by a combination of seed and microsite availability, and 5 species were found not to be seed limited, but the limiting factor was not identified. A combination of seed and microsite limitation implies that recruitment is promoted by increasing both seed and microsite availability. We suggest that the importance of seed limitation in plant populations has been underestimated, and that the operating limiting factors may be dependent on spatial and temporal scale. We expect that many species, if adequately studied, will turn out to be both seed and microsite limited. Experimental field studies that incorporate a range of seed and microsite densities in various spatial and temporal scales are needed to examine the extent to which plant populations are seed and microsite limited.  相似文献

We have been studying the potential of human fibroblastic cells (HFC) from periapical granulation tissue to form a calcified matrix. Recently, we reported that inflamed periapical granulation tissue contains osteogenic cells. In the present study, we tested the hypothesis that HFC, cultured with decalcified bone (DB) of rat, might form much greater calcified matrices than with rat decalcified boiled bone (DBB), which was originally prepared as a negative control. HFC were cultured with DB or DBB in the presence or absence of 2 mM -glycerophosphate (-GP) and 50 g/ml ascorbic acid. After six weeks of culture, a number of von Kossa-positive globular structures were unexpectedly observed inside DBB, but not DB. Without HFC, such structures were never seen in DBB incubated with 2 mM -GP and 50 g/ml ascorbic acid. DB cultured with HFC under the same conditions did not show these structures. Electron-microscopic observation revealed that matrix vesicles aggregated on collagen fibrils around globular structures in DBB. Energy dispersive X-ray microanalysis confirmed that these structures were calcified matrices composed of calcium and phosphate. These results suggest that human periapical granulation tissue contains cells responsible for the formation of calcified matrices in DBB, and that DBB could serve as an excellent scaffold for the calcification of HFC, rather than DB.This work was supported by grant-in-aid (project 15689024) for scientific research from the Ministry of Education, Culture, Sports, Science and Technology, Japan.  相似文献
