After negatively staining with 1% (w/v) sodium phosphotungstate (pH 6·5) or 1% (w/v) ammonium molybdate (pH 6·5), the cell wall, cytoplasmic membrane and mesosomes of the RSD-associated bacterium obtained from the fibrovascular fluid of infected sugarcane were usually clearly displayed. The cells measured 0·19–0·39 μm (av. 0·27 μm) in width and 0·6–3·4 μm in length. Few mesosomes were visible and the cells were approx. 40% wider (0·27–0·52 μm, av. 0·38 μm) when stained with 1% (w/v) uranyl acetate (pH 3·0–4·2). Freezing and thawing the suspension before negative staining with sodium phosphotungstate did not greatly affect the size of the cells or resolution of the mesosomes. Glycine (0·25 M) as the suspending medium, fixation in 2% (w/w) glutaraldehyde, or placing wet instead of dry specimen grids in the electron microscope resulted in wider cells usually lacking mesosomes.  相似文献

In the mammalian ovary, there is a striking difference in the distribution of blood vessels to individual follicles, suggesting that a microvascular network affects the selective growth of oocytes and follicles. In the present study the role of microvascular networks and angiogenic factors on the selective growth of oocytes and follicles was evaluated histologically in fetuses and newborns of ICR strain mice. Apparent selective growth of oocytes and follicles was observed in the ovaries of 1 day old newborns and, at this time, microvascular networks were recognized electronmicroscopically around the follicle that had completed the formation of its follicular structure and contained oocytes more than about 20 μm in diameter. In 3 day old newborns, oocytes more than 30 μm in diameter were detected where blood capillaries were well vascularized. Immunoreactivity to epidermal growth factor (EGF) and a strongly negative charged (colloidal iron-positive) substance (glycosaminoglycans; GAG), which have angiogenic activity, were detected in the ovaries of 3 day or older newborns and were identified more often around growing follicles containing oocytes more than 30 (GAG) and 40 (EGF) μm in diameter. Ovaries removed from 20 day old fetuses and cultured for 4 and 6 days in vitro showed a different distribution of growing follicles. A proportion of oocytes 20.0–24.9 μm in diameter increased during 4 and 6 days of incubation. However, the majority of oocytes did not grow further. These findings indicate that microvascular networks and angiogenic factors are deeply involved in selective oocyte growth beyond approximately 20–30 μm in diameter in mouse ovaries.  相似文献

Euspermatozoa and paraspermatozoa of a copulatory (internal insemination with external sperm transfer) cottoid fish Blepsias cirrhosus were observed ultrastructurally. Euspermatozoa of B. cirrhosus consisted of an acrosome‐less, thin, disk‐like sperm head (1·6-2·0 μm in length and 1·3-1·6 μm in width), a long middle piece, and a long flagellum ( c . 30 μm). Aberrant spermatids, which were rich in cytoplasm and possessed two nuclei, occurred in testicular lobules. They were also observed in semen and were round (5·0-5·3 μm in diameter) and biflagellate, suggesting that they are released along with euspermatozoa at ejaculation. The nuclei of aberrant spermatids developed into masses of highly electron‐dense globules. Judging from their form, nuclear condition, and connection with normal spermatids by intercellular bridges during spermiogenesis, aberrant spermatids of B. cirrhosus are considered hyperpyrenic paraspermatozoa formed by incomplete cytokinesis at the second meiotic division.  相似文献

Image analysis of sagittal sections of gill lamellae of diploid and triploid tench Tinca tinca revealed the blood and water diffusion distance in diploids (2·07 μm) to be significantly higher than that of their triploid siblings (1·46 μm; P < 0·01). Lamellae of diploids compared to triploids were found to be significantly shorter (105·84 v. 132·11 μm) and thicker (18·47 v. 14·21 μm; all at P < 0·05) than those of their triploid siblings but with similar mean sectional areas (1965·44 v. 1910·86 μm2).  相似文献

A double isotope DNA labelling method has been used to determine the duration of DNA synthesis (S) in bone marrow lymphoid cells classified by their nuclear diameters in smears. Incorporation of 3H-thymidine was confined almost entirely to marrow lymphoid cells of 8·0-15·0 μm nuclear diameter (large lymphoid cells). After exposure to 3H-thymidine in vivo and 14C-thymidine 40-104 min later in vitro , the proportion of cells labelled with 3H alone to those labelled with 14C(±3H) in radioautographic smears, plotted against time indicated the efflux from S per hour. Collectively, 28·3 ± 1·1% of all large lymphoid cells were in S and the efflux from S was 15·1% per hour. With decreasing cell size (nuclear diameter) the efflux fell progressively from 28·3% per hour (11·0 μm) to 9·2% per hour (8·0-8·9 μm) and the proportion of cells in S declined from 54·9 ± 2·3% to 14·8 ± 1·6%. Influx into S, measured in vitro by reversing the sequence of isotopes, closely resembled the corresponding efflux values in vivo relative to cell size. Most DNA synthesizing marrow large lymphoid cells belonged to a subgroup with deeply basophilic cytoplasm. The results demonstrate that basophilic large lymphoid cells in the marrow are actively proliferating and have a mean S phase duration of 6·6 hr. The largest marrow lymphoid cells (11·0 μm) proliferate most rapidly (S phase, 3·5 hr; maximum cell cycle time, 6·4 hr) while S duration is prolonged progressively to 10·9 hr for the smaller cells (8·0-8·9 μm).  相似文献

Abstract— A method is described for quantifying the GABA distribution in cat spinal cord at 200–500 μn resolution. Isolated spinal cord (L5–S1) was frozen and sectioned at about 150 μm thickness. The frozen tissue section was cut into 200 or 500 μm square blocks. The GABA content of each square tissue block was determined by enzymic micromethods and GABA distribution was mapped quantitatively. Average GABA concentrations were: 0·4 mmol/l. in white matter, 1·2 mmol/l. in ventral horn and 1·7 mmol/l. in dorsal horn. The highest concentrations of GABA (2–3 mmol/l.) were found in the dorsolateral part of dorsal horn. In order to destroy the interneurons of dorsal horn, the blood vessels supplying the dorsal horn of the lumbar enlargement were unilaterally cauterized. Seven to 30 days after operation, both the size of dorsal root potential and the GABA level in the dorsal horn were markedly reduced on the cauterized side. These results suggest that GABA is highly concentrated in the interneurons of dorsal horn and functions as a transmitter of presynaptic inhibition.  相似文献

The ventricle of the burbot Lota lota heart comprised 0·148 ± 0·006% of the body mass which is nearly two-fold heavier than the relative ventricular mass ( M V) of other similarly sized teleosts. The shape of the ventricle is pyramidal and the wall is exclusively composed of spongious muscle without a distinct compact layer. The atrium forms 0·017 ± 0·002% of the body mass. Length, width, sarcolemmal surface area and volume of enzymatically isolated myocytes from burbot ventricle were 147·2 ± 10·2 μm, 6·3 ± 0·4 μm, 2440·8 · 251·5 μm2 and 2356·8 ± 316·6 μm3, respectively. The myofibrils were peripherally located and their volume density was remarkably high: 65 ± 2 and 68 ± 3% in ventricle and atrium, respectively ( P >0·05). Although not particularly conspicuous, some nonjunctional and junctional sarcoplasmic reticulum (SR) was present in both atrial and ventricular myocytes. The SR formed peripheral couplings with the sarcolemma and the junctional clefts were frequently occupied by foot processes. These findings suggest that cold-adaptation is achieved by cardiac enlargement, high volume density of myofibrils and well-developed peripheral couplings in the SR in the heart of stenothermal burbot.  相似文献

The dynamics of increase of the mosaic muscle in hatchery reared rainbow trout (2·3 to 61·3 cm fork length) are investigated. In trout <5 cm, all fibres are <40 μm in diameter; from 5 to 20 cm the diameter mode remains in the 0–39·9 μm class and there is some extension in range of diameter, thereby suggesting that mosaic muscle increase up to 20 cm is mainly by recruitment of new (small) fibres. When trout exceed 20 cm, mode of fibre diameter shifts to the 40–79·9 um class and fibres of larger diameter (> 100 μrn) appear but the subsequent overall fibre diameter frequency distribution changes little until 50 cm. Increase in muscle during the phase 20–50 cm seems partly attributable to increase in fibre diameter, but remains largely the result of recruitment of small fibres, although the mechanism of the latter process appears less clear than in fish of <20 cm. The recruitment of new fibres stops at 55 cm and further increase in mosaic muscle evidently depends upon the ability of existing fibres to increase in diameter. Possible means by which increasing muscle fibre diameter may limit the ultimate size which trout may achieve are discussed. A phenomenon of apparent reduction in fibre diameter in winter among fish 20–39·9 cm long is noted and its significance is considered.  相似文献

Adult female specimens of Liza ramada were pinealectomized and sham-pinealectomized, and the development of their ovaries was followed over a period of 14 weeks and compared with those of untreated controls. Pinealectomized specimens exposed to long photoperiod (16L/8D) for 14 weeks, showed undeveloped ovaries, with a maximum oocyte diameter of less than 100 μm, and a gonadosomatic index of 0·6 similar to that of sham-pinealectomized and control specimens. In pinealectomized females exposed to short photoperiod (8L/16D) for 6 weeks, the mean diameter of oocytes was 270 μm v . 155 μm in control and sham-pinealectomized specimens. After 9 weeks, the oocytes in pinealectomized specimens reached 430 μm as against 265 μm in the controls. Within 14 weeks of pinealectomy, mean oocyte diameter was 480 um while it was 400 μm in controls and sham-pinealectomized specimens. It is tentatively concluded that the pineal complex has an inhibitory effect on ovarian function in Liza ramada exposed to short photoperiod.  相似文献

The prevalence of Myxobolus dahomeyensis in ovaries of Tilapia zillii was 31·6% and 18·3% in Sarotherodon melanotheron melanotheron . The parasite induced destruction of the oocytes and host castration. Its ovoid spore was 6·5–12·0 × 6·0–8·0 μm and the polar capsules averaged 3·6 × 2·2 μm ( n = 30).  相似文献

The effect of lysine amino acid supplementation on the growth characteristics and morphological pattern of skeletal muscle tissue in Nile tilapia Oreochromis niloticus larvae was evaluated. There were four treatments (T) with increasing levels of lysine supplement (T1 = 0·0%; T2 = 1·1%; T3 = 1·7%; T4 = 4·0%) and one treatment with a commercial diet (T5). In all treatments, morphological and histochemical muscle tissue analyses were similar. Two distinct layers were identified: a superficial red layer, more developed in the lateral line region, formed by fibres with intense to moderate NADH‐TR reaction and strong acid‐stable mATPase activity, and a deep white one, most of the muscle mass, formed by fibres with weak NADH‐TR reaction and strong alkali‐stable mATPase activity. There was an intermediate layer between these two layers with fibres exhibiting either weak acid‐stable or acid‐labile mATPase activity. Body mass increase was significantly higher in T5 than in the lysine treatments (T1–T4). There was no difference in number and diameters of muscle fibres between lysine treatments. In T5, muscle fibre diameter and number were higher. The frequency of red fibres with diameters ≤8 μm was higher in the lysine treatments, and with diameters between 16 and 24 μm, was higher in T5. Most white fibre diameters in T5 were significantly larger than 24 μm and in T1–T4 were between 8 and 16 μm. Cell proliferation was higher in the lysine treatments and muscle growth in T5 was mainly by fibre hypertrophy.  相似文献

Growth dynamics of white fibres from axial muscle has been investigated in Argentine hake, Merluccius hubbsi , discriminating between sexes for the first time. The frequency distributions of fibre diameters are remarkably different in both sexes at sizes between 42 and 63·9 cm total length (T.l.). Males have a much lower proportion of newly recruited fibres (0–10 μm) than females; at 52–53·9 cm T.l., females have 4% of fibres in that category and males 0·5%. It appears that, from this size interval onwards, the increase in muscle mass is due only to the increase in diameter of individual fibres, which may exceed 300 μm. The lower recruitment rate of new fibres in males, and its relationship to lower growth rates and smaller final sizes, are discussed, and possible effects of reproductive activity are considered.  相似文献

Sperm-storage glands in the zebra finch are described; the first passerine for which such information is available. Sperm-storage glands occurred only in paired females, and were located in folds throughout the vagina and on the vaginal side of the utcro-vaginal region. Mean dimensions of the glands were: internal diameter 17·5 μm and length 254 μm; these values are similar to those found for two other (much larger) bird species. The need for passerines to possess sperm-storage glands is discussed.  相似文献

A filtration system was designed to sterilize large volumes of Mycobacterium bovis BCG Tokyo culture safely, needed to purify protein antigens for immunodiagnosis of bovine tuberculosis. A closed system consists of culture bottles connected to three disposable filter capsules of decreasing pore size in series : a depth prefilter over a 1·2 μm filter ; a 0·8 μm prefilter over a 0·45 μm filter ; and a 0·2 μm sterile filter. Low air pressure (3 psi) forces liquid from below the bacillary pellicle. The system features a stainless steel clamp to hold rubber stoppers on the culture bottles, pleated filters to exclude bacillary clumps, a quick disconnector to minimize aerosols, and a closed system with plastic disposable filters that can be autoclaved as a unit without dismantling.  相似文献

Spermatozoa in triploids of the rosy bitterling Rhodeus ocellatus ocellatus   总被引:1,自引:0,他引:1  
Artificially induced triploid male Rhodeus ocellatus ocellatus showed typical nuptial colorations, irrespective of spermiation. In milt from triploids, abnormal spermatozoa (malformation of the head and mitochondrion, excessive formation of the head, mitochondrion and flagellum, and no flagellum) occurred at 78°4% frequency. Spermatozoa with multiflagella were most common, often with a saccate-like organ. Many triploid spermatozoa moved actively as long as those of diploids (10·92±0·91 min=mean±S.D., P >0·05), but did not advance like diploids, spinning around until movement ceased. The sperm density in triploids was < 2% of that from diploids. In triploid testes, deformed and variously sized spermatids were often observed, and normal spermatids and spermatozoa were seldom recognized. The DNA content of triploid spermatozoa varied greatly, compared with that of diploids. Peak of sperm DNA content differed slightly between two triploid samples with two peaks at 1·5 n and 1·9 n ( P <0·0001 in both), respectively. Triploids had the greatest average sperm head diameter of 2·25±0·67 μm (mean±S.D.), while that of diploids was 1·83±0·15 μm ( P =0·002). In the fertilization test using the eggs of diploids ( n =1500, 30 trials), only one egg developed. The embryo chromosome number was 60 (2·5 n) and the ploidy of spermatozoa contributing to fertilization appears to be 1·5 n. The extremely low fertility of triploid R. o. ocellatus spermatozoa seems to be caused by the reduced motility and large head size of spermatozoa, and the low sperm density of the milt. The ploidy of spermatozoa that are successful in fertilization is likely to be related to the distribution pattern in the DNA content of cells.  相似文献

Aims:  To examine the diversity of cultivable 0·2 micron filtrate biofilm forming bacteria from drinking water systems.
Methods and Results:  Potable chlorinated drinking water hosts phylogenetically diverse ultramicrocells (UMC) (0·2 and 0·1  μ m filterable). UMC (starved or dwarf bacteria) were isolated by cultivation on minimal medium from a flow system wall model with polyvinyl chloride (PVC) pipes. All cultivated cells (25 different isolates) did not maintain their ultra-size after passages on rich media. Cultured UMC were identified by their 16S ribosomal DNA sequences. The results showed that they were closely related to uncultured and cultured members of the Proteobacteria, Actinobacteria and Firmicutes. The isolates of phylum Actinobacteria included representatives of a diverse set of Actinobacterial families: Micrococcaceae, Microbacteriaceae, Dermabacteraceae, Nocardiaceae and Nocardioidaceae.
Conclusions:  This study is the first to show an abundance of cultivable UMC of various phyla in drinking water system, including a high frequency of bacteria known to be involved in opportunistic infections, such as Stenotrophomonas maltophilia, Microbacterium sp., Pandoraea sp. and Afipia strains.
Significance and Impact of the Study:  Chlorinated tap water filtrate (0·2 and 0·1  μ m) still harbours opportunistic micro-organisms that can pose some health threat.  相似文献

S ummary . Bdellovibrio bacteriovorus strains which parasitize Rhizobium meliloti, R. trifolii, Agrobacterium tumefaciens and A. radiobacter have been found in Western Australian soils. Rhizobium lupini was not lysed by any strain. Some Bdellovibrio cells pass through 0·22 μm membrane filters, whilst the 0·45 μm filters allow several saprophytic soil bacteria to pass. The strength of the medium is shown to be of great importance for the detection or counting of Bdellovibrio cells. The Bdellovibrio strains are capable of destroying large numbers of rhizobia in the laboratory.  相似文献

The vascular bed in 25 rabbits and 25 white rats has been revealed by means of vital injection of Indian ink into the intraorganic vessels, staining of erythrocytes (benzidine reaction) and silver impregnation. In preparations of the aorta, inferior and superior vena cava diameter of the vessels, the wall and middle membrane thickness, amount of elastic membranes have been measured. Ramification of small vessels and capillaries are revealed only in the paravasal tissues, surrounding the aorta and veins. In serial sections it has been stated that at their greatest extent vasa vasorum has no walls and only in places, where the intercostal (or others) vessels get out of the aorta in its middle or external tunics, a limited, as to its spread, network of intramural vessels exists. The morphometrical analysis does not show any regular connection between the vascular diameter, wall thickness, or middle membrane thickness, amount of elastic membranes in it with presence or peculiarities in distribution of the intramural vessels.  相似文献

Summary Electron micrographs of the zone of separation in flower pedicels of Lycopersicon esculentum and Nicotiana tabacum are presented with particular reference to the indentation of epidermal tissue in the abscission zone, subcellular organelles, and the cell wall. The indentation or groove which delineates the abscission zone extends some distance into the pedicel with branchings off the main groove. These branches are approximately 20 m in width while the main groove averages approximately 200 m in width. Invaginations of the plasmalemma are observed with considerable frequency. within these invaginations one observes a material of about the same density as the cell wall except that it is more fibrillar. Plasmodesmata are also observed, with considerable branching into middle lamellae of cells comprising the abscission zones. Microbodies with crystalloid cores appear with considerable frequency in cells of the abscission zone. The crystalloids appear to be cubical in shape and are composed of parallel sheets of osmiophilic material. The sheets average about 6 m in thickness and are spaced at 4 m intervals. The microbodies with crystalloid cores are observed to be characteristically of two size groupings. In tobacco the microbodies average 900 m and 1,500 m in profile. In tomato they average 300 m and 500 m. Chloroplasts contain a granular component which is membrane-enclosed. The component is large in comparison with the plastid in which it occurs, averaging 1.2–1.4 in diameter in chloroplasts ranging from 1.6 to 2.2 in diameter. The inner membrane of the chloroplast is highly invaginated, and DNA- and phytoferritin-like materials are observed within the plastids. Microtubules with an average diameter of 20 m are observed adjacent and parallel to the plasmalemma, primarily in the corners of the cells. Micrographs of other normally occurring cytoplasmic inclusions are also presented.  相似文献
