Gravity-driven membrane (GDM) filtration is a promising tool for low-cost decentralized drinking water production. The biofilms in GDM systems are able of removing harmful chemical components, particularly toxic cyanobacterial metabolites such as microcystins (MCs). This is relevant for the application of GDM filtration because anthropogenic nutrient input and climate change have led to an increase of toxic cyanobacterial blooms. However, removal of MCs in newly developing GDM biofilms is only established after a prolonged period of time. Since cyanobacterial blooms are transient phenomena, it is important to understand MC removal in mature biofilms with or without prior toxin exposure. In this study, the microbial community composition of GDM biofilms was investigated in systems fed with water from a lake with periodic blooms of MC-producing cyanobacteria. Two out of three experimental treatments were supplemented with dead biomass of a MC-containing cyanobacterial strain, or of a non-toxic mutant, respectively. Analysis of bacterial rRNA genes revealed that both biomass-amended treatments were significantly more similar to each other than to a non-supplemented control. Therefore, it was hypothesized that biofilms could potentially be ‘primed’ for rapid MC removal by prior addition of non-toxic biomass. A subsequent experiment showed that MC removal developed significantly faster in mature biofilms that were pre-fed with biomass from the mutant strain than in unamended controls, indicating that MC degradation was a facultative trait of bacterial populations in GDM biofilms. The significant enrichment of bacteria related to both aerobic and anaerobic MC degraders suggested that this process might have occurred in parallel in different microniches.  相似文献

【目的】将厌氧的膜生物反应器(MBR)与微生物燃料电池(MFC)耦合的厌氧电辅助膜生物反应器(E-MBR)应用于实际工业焦化废水处理。【方法】通过正交实验优化了反应器进水的培养条件为PO_4~(3–)14.3 mg/L、Fe~(2+)0.2 mg/L、Fe~(3+)0.1 mg/L、Co~(2+)0.1 mg/L和Mn~(2+)0.2 mg/L。在此条件下考察了该反应器对系统中有机污染物的去除效率及厌氧污泥的污泥特性、产电性能、胞外聚合物(EPS)、微生物群落结构及膜污染的影响。【结果】结果表明,与未优化的培养条件相比,工业焦化废水COD的去除率提高了23%;污泥浓度(MLSS)、比重、沉降速度增加,污泥体积指数(SVI)降低,表明污泥颗粒化及沉降性能提高;污泥中溶解性EPS (SMP)、松散态EPS (LB-EPS)及紧密结合态EPS (TB-EPS)这3种组分中的蛋白质与多糖的比例(P/C)分别降低0.12、0.25和0.16,表明污泥更易于被降解;厌氧污泥的产电性能增强;高通量分子测序结果表明,反应器中污泥的群落结构发生了明显的变化,优势菌群突出;经扫描电镜(SEM)对比结果表明,反应器阴极膜的污染情况也得到了一定的减缓。【结论】优化进水培养条件可以达到使反应器污水处理效率提高、清理周期缩短和运行更稳定等效果,对于工业废水处理技术的节能环保方面提供一定的理论依据。  相似文献

1. The fraction of living and dead phytoplankton cells in seven Florida lakes was assessed by using the cell digestion assay, a non‐staining membrane permeability test. The cell digestion assay is an effective method to analyse cell viability in complex natural phytoplankton communities. 2. The lakes examined ranged widely in phytoplankton abundance and community composition. The variability in the percentage of living cells (% LC) was high among the taxonomic groups forming the different phytoplankton communities, ranging from 19.7% to 98% LC. 3. All cells within single cyanobacteria filaments were determined to be either dead or alive, suggesting physiological integration of the cells within colonies. 4. Within each lake, the dominant taxa generally exhibited the highest proportion of living cells. A high proportion of living cells was found to be a characteristic of the different taxa forming the communities of eutrophic lakes. The average value for the % LC for all groups comprising the phytoplankton communities in each of the lakes ranged from 29.9 ± 7.2 to 80.4 ± 4.0 (mean ± SE) and varied strongly and positively with chlorophyll a concentration. 5. These results suggest phytoplankton cell death to be an important process structuring phytoplankton communities in lakes, particularly in oligotrophic ones.  相似文献

【目的】揭示碳源对活性污泥微生物细胞膜特性(磷脂脂肪酸组成和流动性)以及群落结构的影响规律。【方法】采用原子力显微镜、磷脂脂肪酸(PLFA)、荧光漂白恢复(FRAP)和Mi Seq分析技术,考察以葡萄糖、乙酸钠、蛋白胨、葡萄糖?蛋白胨(1?1)和乙酸钠?蛋白胨(1?1)为基质的序批式活性污泥(SBR)反应器中,活性污泥微生物表面粘附力、细胞膜PLFA组成和流动性及群落结构的差异。【结果】含有蛋白胨为碳源时相比于葡萄糖和乙酸钠为单一碳源,细胞膜磷脂流动性增加,且18?1ω9c、15?0iso和17?0iso的含量提高了53.1%-354.7%、135.6%-407.9%和88.1%-264.3%;同时其微生物表面粘附力和群落多样性均增大。优势菌群对碳源的响应呈现出不一致的规律:葡萄糖和乙酸钠为单一碳源时其优势菌门分别为放线菌门和变形菌门,Nakamurella和Flavobacterium分别为其优势菌属,群落多样性指数分别为3.65和4.25;蛋白胨为碳源时促进了Candidatus Saccharibacteria门的累积,群落多样性指数在4.96-5.09。【结论】主成分分析(PCA)表明,含有蛋白胨为碳源时微生物细胞膜PLFA组成具有相似性;冗余分析(RDA)表明,不同碳源驯化出不同的微生物群落,进而也对细胞膜磷脂组成产生了影响。  相似文献

Effects of temperature and temperature shock on the performance and microbial community structure of a submerged anaerobic membrane bioreactor (SAnMBR) treating thermomechanical pulping pressate were studied for 416 days. The results showed that the SAnMBR system were highly resilient to temperature variations in terms of chemical oxygen demand (COD) removal. The residual COD in treated effluent was slightly higher at 55 °C than that at 37 and 45 °C. There were no significant changes in biogas production rate and biogas composition. However, temperature shocks resulted in an increase in biogas production temporarily. The SAnMBR could tolerate the 5 and 10 °C temperature shocks at 37 °C and the temperature variations from 37 to 45 °C. The temperature shock of 5 and 10 °C at 45 °C led to slight and significant disturbance of the performance, respectively. Temperature affected the richness and diversity of microbial populations.  相似文献

A key issue in evolutionary biology is whether selection acting at levels higher than the individual can cause evolutionary change. If it can, then conceptual and empirical studies must consider how selection operates at multiple levels of biological organization. Here, we test the hypothesis that estimates of broad‐sense community heritability, HC2, can be used to predict the evolutionary response by community‐level phenotypes when community‐level selection is imposed. Using an approach informed by classic quantitative genetics, we made three predictions. First, when we imposed community‐level selection, we expected a significant change in the average phenotype of arthropod communities associated with individual tree genotypes [we imposed selection by favoring high and low NMDS (nonmetric multidimensional scaling) scores that reflected differences in arthropod species richness, abundance and composition]. Second, we expected HC2 to predict the magnitude of the community‐level response. Third, we expected no significant change in average NMDS scores with community‐level selection imposed at random. We tested these hypotheses using three years of common garden data for 102 species comprising the arthropod communities, associated with nine clonally replicated Populus angustifolia genotypes. Each of our predictions were met. We conclude that estimates of HC2 account for the resemblance among communities sharing common ancestry, the persistence of community composition over time, and the outcome of selection when it occurs at the community level. Our results provide a means for exploring how this process leads to large‐scale community evolutionary change, and they identify the circumstances in which selection may routinely act at the community level.  相似文献

Laboratory-scale column experiments were performed to investigate the effects of membrane-supplied H2 on tetrachloroethene (PCE) dechlorination and microbial community composition. Columns were filled with aquifer material from one of two TCE-contaminated sites and fed a PCE-spiked anaerobic minimal medium for approximately 1 year. For each experiment, one or more experimental columns were supplied with H2 via gas-permeable hollow-fiber membranes with one control column not receiving any H2. After approximately 1 year of operation, aquifer material samples were collected along the length of the columns. Bacterial communities in the samples were analyzed by amplifying the highly variable V3 region of the 16S rRNA gene and separating amplicons using denaturing gradient gel electrophoresis. Microbial community profiles in H2-fed (continuous or pulsed delivery) columns were compared with those in untreated control columns and microbial community profiles were also compared with dechlorination profiles. Selected bands were sequenced for identification. Supply of the simple electron donor H2, changed the microbial community composition, but did not decrease overall diversity. Continuous H2 addition via hollow-fiber membranes enriched for Dehalococcoides-like species, whose relative abundance correlated with enhanced dechlorination activity. PCE was completely dechlorinated to ethene in columns packed with aquifer material from Cape Canaveral, Florida; PCE was dechlorinated to only cis-dichloroethene, however, in columns packed with aquifer material from a TCE-contaminated wetland near Minneapolis, Minnesota. Unexpectedly, Dehalococcoides-like populations were detected in samples from both sets of column experiments. These results suggest that the mere detection of Dehalococcoides-like species in a sample of aquifer material is not a sufficient indicator of the potential to dechlorinate PCE to ethene via biostimulation by H2.  相似文献

Members of the YidC/Oxa1/Alb3 protein family mediate membrane protein insertion, and this process is initiated by the assembly of YidC·ribosome nascent chain complexes at the inner leaflet of the lipid bilayer. The positively charged C terminus of Escherichia coli YidC plays a significant role in ribosome binding but is not the sole determinant because deletion does not completely abrogate ribosome binding. The positively charged cytosolic loops C1 and C2 of YidC may provide additional docking sites. We performed systematic sequential deletions within these cytosolic domains and studied their effect on the YidC insertase activity and interaction with translation-stalled (programmed) ribosome. Deletions within loop C1 strongly affected the activity of YidC in vivo but did not influence ribosome binding or substrate insertion, whereas loop C2 appeared to be involved in ribosome binding. Combining the latter deletion with the removal of the C terminus of YidC abolished YidC-mediated insertion. We propose that these two regions play an crucial role in the formation and stabilization of an active YidC·ribosome nascent chain complex, allowing for co-translational membrane insertion, whereas loop C1 may be involved in the downstream chaperone activity of YidC or in other protein-protein interactions.  相似文献

Abstract

Biological membranes encompass and compartmentalize cells and organelles and are a prerequisite to life as we know it. One defining feature of membranes is an astonishing diversity of building blocks. The mechanisms and principles organizing the thousands of proteins and lipids that make up membrane bilayers in cells are still under debate. Many terms and mechanisms have been introduced over the years to account for certain phenomena and aspects of membrane organization and function. Recently, the different viewpoints – focusing on lipids vs. proteins or physical vs. molecular driving forces for membrane organization – are increasingly converging. Here we review the basic properties of biological membranes and the most common theories for lateral segregation of membrane components before discussing an emerging model of a self-organized, multi-domain membrane or ‘patchwork membrane'.  相似文献

Field trials were carried out from June to August in 2004 at Wuyishan (Wuyi Mountains), Fujian province, China, to determine the effects of habitat diversification on arthropod communities. Two Chinese cabbage, Brassica chinensis, field 1 (F1) and field 2 (F2) surrounded by diverse vegetable cultivars were selected, while a monoculture of Chinese cabbage served as the control field (CK). The results showed that: (i) when comparing insect abundance of each order between different habitats, significantly higher numbers of lepidopterous insects (39.76% from the each order) and lower densities of Hymenoptera (19.82%) were found in CK than in F1 and F2; (ii) compared with CK, F1 and F2 had a lower percentage of species richness and an abundance of herbivorous insects, but increased richness, abundance and biodiversity of predatory insects; (iii) no differences were observed in neutral insects' guild between different fields; and (iv) the dominant species for each guild depends on the habitat types and sampling dates. This study suggests that intercropping could conceivably be used in these habitats to increase the population of natural predators, thus achieving desirable and ecologically friendly results in vegetable fields.  相似文献

1. Three floods (July 2000, August 2002, September 2003) and a hurricane (October 2001) that occurred in a lowland forest in the southern Maya Mountains of Belize presented an opportunity to evaluate the influence of these disturbances on the structure of a small mammal assemblage. 2. Four terrestrial and four primarily scansorial/arboreal species were trapped July 2000-March 2005 in six grids over 14 irregularly spaced trapping periods. 3. Community dynamics were characterized more by changes in species composition than changes in diversity. The dynamics were driven by species-specific variation in abundance, with changes in composition generally, but not exclusively, due to the occurrence or disappearance of species at low abundance. Despite the disturbances, species richness remained relatively constant. Evenness within the assemblage was consistently low, primarily as a result of dominance by one species, Heteromys desmarestianus. 4. Effects of flooding on community structure were direct but relatively brief (< 1 year), and varied with the duration and intensity of flooding. Effects from the hurricane were indirect but long-lasting and strongly related to severely reduced food resources. 5. This study suggests that long-term dynamics in the structure of many animal communities in the tropics often results from interactions between direct and indirect effects of disturbance. It also suggests that community resistance will depend on variation in disturbance type and regime, but resilience will be determined by the life-history characteristics of each species.  相似文献

如何解释群落中物种的丰富与稀少,并对物种多样性和群落稳定性进行合理的量化评价是群落生态学研究中的一个热点问题。20世纪中期,MacArthur将影响自然群落稳定性的因素归结为物种数量多少以及物种间相互作用的大小,20世纪末Doak等学者提出群落的容纳能力和物种间的维持机制是决定群落稳定性的关键因素。同时对群落结构及物种间维持机制的研究也有了新的突破,Hubbell提出"生物多样性与生物地理学统一的中性理论(Unified Neutral Theory of Biodiversity and Biogeography)"为群落生态学研究提供了新的思路和方法。从群落中性理论的基本假设出发,对Hubbell中性理论中局域群落的物种多度动态模型进行分析,归纳得出群落中性理论中物种多样性与群落稳定性之间的量化关系。封闭的局域群落中,出现物种灭绝或单物种独占的时间与群落大小及物种相对多度成正比,物种多样性程度的增加可延长物种灭绝或独占的时间;开放的局域群落中,物种多度期望值与局域群落大小、物种在集合群落中的物种相对多度成正比,周围群落中物种的灭绝会引起局域群落中相应物种的灭绝,最终导致整个生态群落物种多样性的降低;群落中物种多度的方差与局域群落大小、迁移率、物种在集合群落中的物种相对多度相关,局域群落物种多度的波动幅度随着群落间生态隔离的减弱或物种多样性程度的增加而减小。由此,集合群落物种多样性是影响局域群落物种多样性的重要因素,生态隔离程度的减弱及物种多样性的增加都将增强群落的稳定性。  相似文献

There are contradictory results in the literature relating to the effect oftamoxifen on membrane fluidity. The present work investigates the effect oftamoxifen on membrane dynamics to find out whether the concentration oftamoxifen can be one of the factors in this discrepancy. Turbidity(absorbance at 440 nm) and Fourier transform infrared spectroscopicstudies reveal that tamoxifen causes opposite effects on membranefluidity at low (1 mol.%) and high (30 mol.%) tamoxifen concentrations. Lowtamoxifen concentrations increase the absorbance in the gel and liquidcrystalline phase, whereas high tamoxifen concentrations decrease theabsorbance in gel and liquid crystalline phase, whereas tamoxifenconcentrations decrease the absorbance. Observations on both phasesshow that the bandwidth of the CH2 stretching bands decreases with1 mol.% tamoxifen and increases with 30 mol.% tamoxifen present, indicatinga decrease in membrane fluidity at low tamoxifen concentrations and anincrease in fluidity at high tamoxifen concentrations. It is seen that theapparent discrepancy in the literature on the effect of tamoxifen onmembrane fluidity mainly arises from the tamoxifen concentration used andthe confusion on the concept of lipid fluidity and lipid order.  相似文献

在城市化的背景下,探明不同大型底栖动物群落对植物群落变化的响应,对红树林生态系统的健康评价和退化生态系统的修复具有重要意义。以福建漳江口国家级红树林自然保护区为研究对象,揭示红树植物群落周转对不同大型底栖动物群落结构(蟹类和软体动物)的影响,并分析其与环境参数的关联性。结果表明:在1.6 km的地理尺度上,漳江口红树林湿地的环境变化较大,表现出7.19个环境梯度。在此地理尺度的环境梯度上,红树植物群落由5种红树植物组成,其平均周转率为76%;软体动物群落由12种软体动物以模块化和非随机的方式聚集,其平均周转率为83%,并随红树植物群落周转出现显著非线性增加,盐度和老鼠簕(Acanthus ilicifolius)周转是其主要影响因素;蟹类群落则由20种蟹类以更高的模块化和非随机的方式分散,平均周转率为89%,并随红树植物群落周转而呈线性增加,主要受pH和白骨壤(Avicennia marina)周转的影响。研究揭示了大型底栖动物与植物群落和环境之间的关系,为城市化背景下红树林生态系统的保护和恢复提供基础资料。  相似文献

Interactions between spectrin and the inner surface of the human erythrocyte membrane have been implicated in the control of lateral mobility of the integral membrane proteins. We report here that incubation of “leaky” erythrocytes with a water-soluble proteolytic fragment containing the membrane attachment site for spectrin achieves a selective and controlled dissociation of spectrin from the membrane, and increases the rate of lateral mobility of fluorescein isothiocyanate-labeled integral membrane proteins (> 70% of label in band 3 and PAS-1). Mobility of membrane proteins is measured as an increase in the percentage of uniformly fluorescent cells with time after fusion of fluorescent with nonfluorescent erythrocytes by Sendai virus. The cells are permeable to macromolecules since virus-fused erythrocytes lose most of their hemoglobin. The membrane attachment site for spectrin has been solubilized by limited proteolysis of inside-out erythrocyte vesicles and has been purified (V). Bennett, J Biol Chem 253:2292 (1978). This 72,000-dalton fragment binds to spectrin in solution, competitively inhibits association of 32P-spectrin with inside-out vesicles with a Ki of 10?7M, and causes rapid dissociation of 32P-spectrin from vesicles. Both acid-treated 72,000-dalton fragment and the 45,000 dalton-cytoplasmic portion of band 3, which also was isolated from the proteolytic digest, have no effect on spectrin binding, release, or membrane protein mobility. The enhancement of membrane protein lateral mobility by the same polypeptide that inhibits binding of spectrin to inverted vesicles and displaces spectrin from these vesicles provides direct evidence that the interaction of spectrin with protein components in the membrane restricts the lateral mobility of integral membrane proteins in the erythrocyte.  相似文献

Summary Single-channel analysis of electrical fluctuations induced in planar bilayer membranes by the purified human complement proteins C5b6, C7, C8, and C9 have been analyzed. Reconstitution experiments with lipid bilayer membranes showed that the C5b-9 proteins formed pores only if all proteins were present at one side of the membrane. The complement pores had an average single-channel conductance of 3.1 nS at 0.15m KCl. The histogram of the complement pores suggested a substantial variation of the size of the single channel. The linear relationship between single-channel conductance at fixed ionic strength and the aqueous mobility of the ions in the bulk aqueous phase indicated that the ions move inside the complement pore in a manner similar to the way they move in the aqueous phase. The minimum diameter of the pores as judged from the conductance data is approximately 3 nm. The complement channels showed no apparent voltage control or regulation up to transmembrane potentials of 100 mV. At neutral pH the pore is three to four times more permeable for alkali ions than for chloride, which may be explained by the existence of fixed negatively charged groups in or near the pore. The significance of these observations to current molecular models of the membrane lesion formed by these cytolytic serum proteins is considered.  相似文献

Divalent cation permeability of rat parotid gland basolateral plasma membranes was examined in dispersed parotid acini (by Ca2+ or Mn2+ entry) and in isolated basolateral plasma membrane vesicles (BLMV, by45Ca2+ influx). Mn2+ entry (fura2 quenching) was about 1.6 fold higher in internal Ca2+ pool-depleted acini (Ca2+-depl acini) than in unstimulated cells. Mn2+ entry into Ca2+-depl acini was increased at external pH>7.4 and decreased at pH<7.4. Pretreatment of Ca2+-depl acini with the relatively hydrophobic carboxylic group reagent, N,N-dicyclohexylcarbodiimide (DCCD, 50 M for 30 min) resulted in the inhibition of Mn2+ entry into Ca2+-depl acini to unstimulated levels. Another hydrophobic carboxyl group reagent, N-ethoxycarbonyl-2-ethoxy-1,2-dihydroquinoline (EEDQ) and the relatively hydrophilic carboxyl group reagents, 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide (EDC) and 1-cyclohexyl-3-(2-morpholinoethyl)carbodiimide (CMCD) did not affect Mn2+ entry.Similar to the effects in intact acini, Ca2+ influx into BLMV was decreased when the external pH was lowered below 7.4. Also DCCD (5 mM, 30 min), but not EEDQ, decreased (40%) Ca2+ influx in BLMV. However, unlike in acini, the hydrophilic reagents, EDC, EAC, and CMCD decreased Ca2+ permeability in BLMV and the effects were nonadditive with the decrease induced by DCCD. The aggregate effects of carboxyl group reagents on the Ca2+ and Mn2+ permeability in BLMV and intact acini, respectively, suggest that a critical carboxyl group (most likely accessible from the cytoplasmic side of the plasma membrane) is involved in divalent cation flux in rat parotid acinar cells.  相似文献

【目的】调查和分析银川不同湿地陆生昆虫群落结构组成、功能多样性及其稳定性变化。【方法】2016年5-10月选取银川黄河湿地(HH)、鸣翠湖湿地(MCH)、芦花台湿地(LHT)、水洞沟湿地(SDG)4种湿地生境,采用棋盘式扫网和陷阱捕捉的方法进行昆虫采集。根据调查数据进行昆虫群落结构多样性分析、主成分分析和聚类分析。【结果】4种湿地生境共采集昆虫38 668头,隶属13目97科370种。双翅目、同翅目、膜翅目在4种生境中均为优势类群。植食性(Ph)昆虫功能团的科数和种数在4种生境昆虫群落中占比均最大。植食性-中性昆虫(Ph-Ne)间个体数在4种生境下均存在显著负相关(P<0.01);植食性-捕食性昆虫(Ph-Pr)间个体数在MCH生境下存在显著正相关(P<0.05);中性-捕食性昆虫(Ne-Pr)间个体数在MCH生境下存在显著负相关(P<0.01),在LHT生境下存在显著负相关(P<0.05);4种功能团结构的其他组合的个体数在4种生境下均不存在显著相关性。主成分分析结果表明,捕食性类群是4个类群中影响构成昆虫群落组成的主要成分。同种功能团在4种生境下的多样性分析结果表明,捕食性功能团(Pr)与寄生性功能团(Pa)在4种生境群落中的群落丰富度、均匀程度[辛普森指数(Simpson index)]差异性显著(P<0.05),而植食性功能团(Ph)与中性功能团(Ne)的群落丰富度、均匀程度的差异性不显著(P>0.05);中性功能团(Ne)在4种生境群落中个体出现的不确定程度[香农-维纳指数(Shannon-Wiener index)]差异性不显著(P>0.05),在4种生境的均匀度指数(Pielou evenness index)差异性显著(P<0.05),其余3个功能团在4种生境下的香农-维纳指数均差异性显著(P<0.05),在4种生境下的均匀度指数均差异性不显著(P>0.05)。4种生境昆虫群落特征指数的聚类分析分为2类,永久性湿地HH和MCH为第1类,季节性湿地LHT和SDG为第2类。Ss/SiSa/Sp两个稳定性指标均显示MCH昆虫群落稳定性最高,4种生境昆虫群落最稳定的时期是5和9月。【结论】双翅目和膜翅目昆虫在银川不同湿地生境中均为优势类群,可作为环境指示参考;昆虫群落稳定性最好的湿地类型为永久性湖泊湿地;不同生境昆虫群落多样性随季节时序变化,在银川最稳定的时期是5和9月。气候因子、环境因素如何驱动各湿地昆虫群落多样性及稳定性的变化因素还有待开展研究。  相似文献
