Underwater observations on fish and asteroid consumers (i.e. predators and scavengers) of sea urchins, Paracentrotus lividus and Arbacia lixula, were carried out at several locations in shallow Mediterranean rocky reefs. Observations conducted in the marine reserve of Torre Guaceto (Adriatic Sea) revealed that sparid fishes, Diplodus sargus and D. vulgaris, are the main fish predators of small (<1 cm in test diameter) and medium (1–4 cm) sea urchins, whereas the labrids Coris julis and Thalassoma pavo preyed only upon small sea urchins. Large D. sargus were able to prey upon small and medium, and occasionally large (>4 cm) sea urchins, whereas medium and small Diplodus preyed mainly upon small sea urchins. The number of sea urchins preyed upon by fishes was negatively related to sea urchin size for both species. P. lividus appeared to be subject to higher predation levels than A. lixula. The scavenger guild comprised 11 fish species, with D. sargus, D. vulgaris, Coris julis and Chromis chromis accounting for about 80% of scavenger fishes. Observations performed at several locations in the Mediterranean on the predatory asteroid Marthasterias glacialis revealed that only 3% of the detected individuals were preying upon sea urchins. Due to the importance of sea urchins for assemblage structure and functioning of Mediterranean rocky reef ecosystems, these results may have also important implications for management of fishing activities.Communicated by H.-D. Twelve species of intertidal algae and molluscs from Ras Beirut, Lebanon have been investigated for their heavy metal content. Atomic absorption spectrophotometric analysis showed that the algae and molluscs concentrated similar levels of most metals. However, of the twelve organisms, Brachydontes variabilis had the highest copper, Patella coerulea and Colpomenia sinuosa had the highest iron, and Pinctada radiata had the highest zinc values with rather elevated cadmium. Cystoseira spinosa concentrated the lowest levels of zinc, and nickel was variable in all organisms.Seasonal comparisons of metal levels were also considered in three algae and three molluscs studied previously. The only signs of seasonal variation in the algae were with nickel and iron in Halimeda tuna and lead in Pterocladia pinnata. Both cadmium and lead were generally lowest in the spring samples of the algae. Lead and nickel concentrations in the molluscs were generally highest in the summer and lowest in the spring. Slight seasonal trends were seen with lead and iron in Brachydontes variabilis and lead in Monodonta turbinata.The lack of comparison data from the Mediterranean, particularly the eastern basin, makes it essential that studies of this nature be continued.  相似文献

Summary Freeze-substitution was used to study the distribution of calcium in sea urchin eggs, and the validity of the technique was assessed. We followed the fate of both total and exchangeable calcium of sea urchin eggs in two species (Paracentrotus lividus and Arbacia lixula) after the various treatments needed for freeze-substitution and embedding. We compared the calcium content either by X-ray microanalysis of Epon-embedded sections of freeze-substituted eggs (6.2±0.71 mmoles/kg of Epon-embedded tissue) or by flame spectrometry analysis of living eggs (32.3±1.30 nmoles/mg protein). After standardization of units, both values lead to similar total calcium content. We also measured the movements of 45Ca from prelabelled eggs. Exchangeable 45Ca as well as total calcium appeared unaffected by the preparative treatment for X-ray microanalysis. In conclusion, our preparative technique for X-ray microanalysis can be considered appropriate for our material and allows us to undertake a subcellular quantification of calcium in various organelles.  相似文献

The aim of this study was to analyze male and female gonad fatty acids of two sea urchin species, Paracentrotus lividus and Arbacia lixula, from the south coast of Spain. Additionally, we investigated possible differences between two locations. The ovaries of both species showed higher percentages of 14:0, 16:0, 16:1n-7, 18:2n-6, 18:3n-3 and 18:4n-3 than testes and lower levels of 18:0, 22:1n-9, 20:4n-6 and 22:5n-3. In P. lividus but not in A. lixula, the level of 20:5n-3 was higher in testes than in ovaries. These differences between sexes probably indicate different requirements of males and females during gametogenesis although the presence of a large number of gametes in the mature gonad may also have influences on fatty acid composition. Significant differences in gonad fatty acid profiles where also found when individuals of P. lividus collected at a location of the Mediterranean region were compared with specimens collected at the Atlantic coast. The most remarkable changes were the lower levels of 14:0, 18:1n-7, 20:1n-9, 20:4n-6 and 22:4n-6 and the higher values of 20:1n-11, 20:5n-3 and 22:6n-3 found in males and females of the Mediterranean specimens compared to those of the Atlantic coast. Metallothionein presence and amount were determined in the unfertilized eggs of six sea urchin species by silver saturation assay and gel-chromatography of cell extracts. The results showed high levels of metallothionein in the egg cytoplasm of the two Mediterranean species Paracentrotus lividus and Sphaerechinus granularis. No metallothionein was found either in the eggs of Arbacia lixula, or in those of the three Eastern species Strongylocentrotus intermedius, Temnopleurus hardwickii and Clypeaster japonicus. However, the extracts of the latter three species revealed the presence of zinc bound in a macromolecular form, thus suggesting the existence of metal-binding proteins distinct from metallothioneins.  相似文献

We present the global phylogeography of the black sea urchin Arbacia lixula, an amphi-Atlantic echinoid with potential to strongly impact shallow rocky ecosystems. Sequences of the mitochondrial cytochrome c oxidase gene of 604 specimens from 24 localities were obtained, covering most of the distribution area of the species, including the Mediterranean and both shores of the Atlantic. Genetic diversity measures, phylogeographic patterns, demographic parameters and population differentiation were analysed. We found high haplotype diversity but relatively low nucleotide diversity, with 176 haplotypes grouped within three haplogroups: one is shared between Eastern Atlantic (including Mediterranean) and Brazilian populations, the second is found in Eastern Atlantic and the Mediterranean and the third is exclusively from Brazil. Significant genetic differentiation was found between Brazilian, Eastern Atlantic and Mediterranean regions, but no differentiation was found among Mediterranean sub-basins or among Eastern Atlantic sub-regions. The star-shaped topology of the haplotype network and the unimodal mismatch distributions of Mediterranean and Eastern Atlantic samples suggest that these populations have suffered very recent demographic expansions. These expansions could be dated 94–205 kya in the Mediterranean, and 31–67 kya in the Eastern Atlantic. In contrast, Brazilian populations did not show any signature of population expansion. Our results indicate that all populations of A. lixula constitute a single species. The Brazilian populations probably diverged from an Eastern Atlantic stock. The present-day genetic structure of the species in Eastern Atlantic and the Mediterranean is shaped by very recent demographic processes. Our results support the view (backed by the lack of fossil record) that A. lixula is a recent thermophilous colonizer which spread throughout the Mediterranean during a warm period of the Pleistocene, probably during the last interglacial. Implications for the possible future impact of A. lixula on shallow Mediterranean ecosystems in the context of global warming trends must be considered.  相似文献

The species and stage specificities of membrane components active in promoting reaggregation of cells dissociated from embryos of the two Mediterranean sea urchin species Paracentrotus lividus and Arbacia lixula have been examined. Membrane proteins extracted with butanol either from purified membranes or from dissociated cells without significant reduction of viability promoted reaggregation of both the homologous and heterologous species. Extracts from plutei and blastulae were equally effective in promoting reaggregation of blastula cells. By contrast, Fab's prepared from IgG raised against these extracts or purified membranes are strictly species specific because they prevent reaggregation of cells and actively dissociate live embryos of only the homologous species. No corresponding stage specificity of the Fab was observed: Fab against extracts from blastula embryos also caused dissociation of plutei. Antigenic analysis of the extracts by the Ouchterlony test revealed the presence of components specific for each species as well as others common to both.  相似文献

The ultrastructural localization of B-1,3-glucanase in three species of sea urchin eggs was determined using a monospecific antibody in an electronmicroscopic immunogold procedure. In all three species, Lytechinus variegatus, Strongylocentrotus purpuratus, and Arbacia punctulata, B-1,3-glucanase was localized specifically to the cortical granules. No other organelle within the egg contained significant label. During the fertilization reaction, B-1,3-glucanase was released from cortical granules into the perivitelline space and became associated with the hyaline layer. No significant label was found in association with the fertilization envelope.  相似文献

Three species of sea urchins, Echinometra lucunter, Lytechinus variegatus, and Arbacia lixula, common on the southeastern coast of Brazil, were selected for experiments on seaweed survival after passage through their digestive tract. L. variegatus and E. lucunter have more than 80% of their diet composed of algae. C. 24% of the species of algae present in the digestive tract survived. A. lixula has only 29% of its diet composed by algae, but the survival rate of the algae is c. 50%.Although Rhodophyta represent 51% of the ingested algae, their survival rate in the faeces is only 19%, while the Chlorophyta, with a survival rate of 63%, comprise only about 26% of the diet. Most species of algae that survived ingestion were r-strategists.  相似文献

Previous data from our laboratory showed that the reticulum of the sea cucumber smooth muscle body wall retains both a sarco/endoplasmic reticulum Ca2+-ATPase (SERCA) and a sulfated polysaccharide. In this invertebrate, the transport of Ca2+ by the SERCA is naturally inhibited by these endogenous sulfated polysaccharides. The inhibition is reverted by K+ leading to an enhancement of the Ca2+ transport rate. We now show that vesicles derived from the endoplasmic reticulum of unfertilized eggs from the sea urchin Arbacia lixula retain a SERCA that is able to transport Ca2+ at the expense of ATP hydrolysis. As described for the sea cucumber SERCA isoform, the enzyme from the sea urchin is activated by K+ but not by Li+ and is inhibited by thapsigargin, a specific inhibitor of SERCA. A new sulfated polysaccharide was identified in the sea urchin eggs reticulum composed mainly by galactose, glucose, hexosamine and manose. After extraction and purification, this sulfated polysaccharide was able to inhibit the mammal SERCA isoform found in rabbit skeletal muscle and the inhibition is reversed by K+. These data suggest that the regulation of the SERCA pump by K+ and sulfated polysaccharides is not restricted to few marine invertebrates but is widespread.  相似文献

Fibronectin-like proteins were purified from ovaries of the sea urchin species, Paracentrotus lividus (PI), Sphaerechinus granularis (Sg), Arbacia lixula (Al), Pseudocentrotus depressus (Pd), and Anthocidaris crassispina (Ac), by gelatin-Sepharose affinity chromatography. The major component had a molecular mass of 180 kDa and was eluted by 1 M NaCl or 8 M urea, depending on the species used. By substrate adhesion assay, we tested the biological activity of the 180 kDa protein purified from Paracentrotus lividus (P1-180K) and showed that it promotes the adhesion of homologous embryonic cells to the substrate. An antiserum, developed against Temnopleurus hardwickii fibronectin-like protein (Th-180K), was used in Western blots of the proteins purified from the five species. The antibody cross-reacted with Pl-180K, Pd-180K and Ac-180K. A peptide map of P1-180K, obtained by V8 protease partial digestion, was compared with those obtained from the other four proteins and showed an homology between 40 and 56%. This report confirms that fibronectin-like proteins can be purified from sea urchins on the basis of their binding to gelatin-Sepharose; the proteins differ for their binding affinity to gelatin and share different epitopes, suggesting that they are members of a sea urchin fibronectin super family.  相似文献

Control and γ-irradiated sea urchin (Arbacia punctulata) eggs were incubated with H3-leucine. No incorporation of the amino acid into the proteins of most unirradiated eggs was detectable by autoradiography. The distribution of pigment granules in eggs of three species of sea urchins is described with reference to developmental stage and an egg's animal-vegetal axis of organization. Polarity in unfertilized sea urchin eggs has been a debated subject; present evidence demonstrates that the animal-vegetal axis is established before fertilization. The pigment pattern in some batches of Paracentrotus eggs exhibiting the celebrated “pigment band,” originally described by Theodor Boveri, is revised and is interpreted as a comparatively precocious expression of the underlying egg polarity. “Unbanded” Paracentrotus eggs and eggs of Arbacia lixula and Arbacia punctulata can be induced to exhibit the same pigment pattern by artificial activation. The induced pigment pattern aligns with an axis defined by polar bodies and the jelly canal, which are two external markers of the animal pole which are only rarely seen. It is therefore concluded that all of these eggs possess an animal-vegetal axis before fertilization even though it usually remains unexpressed until later developmental stages. Polarized changes in pigmentation are consistent with the following general mechanism: A change is triggered in the cortex of the vegetal pole; the change is programmed for a time which corresponds to the fourth mitotic division, even though mitosis itself is not involved; activation at fertilization initiates the “clock” in most cases, although in “banded” Paracentrotus eggs the “clock” is apparently started before ovulation; only the vegetal hemisphere's pigment is affected by the change. The nature of the underlying axis which defines animal and vegetal poles is discussed. Aspects of the axis have been tentatively traced back to the primary oocyte stage, but its fundamental nature remains unknown.  相似文献

The effect of TPA (12-O-tetradecanoylphorbol-13-acetate) upon ionic exchanges was investigated in eggs of the sea urchin Arbacia lixula. Ouabain-sensitive 86Rb uptake and amiloride-sensitive 24Na influx were dramatically stimulated after TPA addition, indicating an enhancement of total ionic permeabilities. Stimulation by TPA of both Na+/H+ and Na+/K+ exchanges was canceled by amiloride, suggesting that activation of protein kinase C elicits, via Na+/H+ activity, stimulation of the sodium pump. However, TPA did not stimulate sodium pump activity and Na+/H+ exchange at the same rate as fertilization, probably because of an absence of calcium-dependent events. Further fertilization of TPA-pretreated eggs triggered an enhancement of sodium pump activity when the TPA treatment duration did not exceed 10 min. It is suggested that TPA activates preexisting transporting mechanisms in plasma membranes of unfertilized eggs (Na+ pump, Na+/H+ exchange) without eliciting corresponding regulatory mechanisms (Na+ stat, pH stat).  相似文献

Recruitment is a principal controlling factor in population dynamics of marine species. In marine invertebrates with a planktonic larval stage, such as echinoids, recruitment is assured by larval supply, settlement and juvenile survival. Larval supply and juvenile survival are affected by a wide range of factors, including temperature, presence of predators, quality and quantity of food. Echinoid larval settlement is mainly conditioned by the finding of a suitable substrate to metamorphose. The sea urchins Arbacia lixula and Paracentrotus lividus are considered key species of the Mediterranean infralittoral rocky shores. At high densities, the grazing activity of both species can produce and maintain barren grounds, a particular habitat condition characterized by extremely low cover values of erect algae with high presence of naked substrates and encrusting corallinales, poor in biodiversity and ecosystem functions. We tested the role of different settlement substrates on the metamorphosis competent larvae of the two species. Furthermore, from our larval rearing trails we were able to identify strong temperature effects on larval development of the two species. P. lividus and A. lixula larvae have been reared at 18 °C but for the second species it was necessary to use higher temperatures (22 °C) to perform settlement experiments, as in the 18 °C set all larvae died in the first week. Both species larvae have been fed Cricosphaera elongata. Metamorphosis of competent larvae has been induced using different substrates: naked stones, Lithophyllum incrustans, Stypocaulon scoparium, Corallina elongata, turf forming algae and Posidonia oceanica. For each species, two larval batches were used for settlement experiments; for each larval batch two replicates/substrates were set up. No differences in the rate of metamorphosis on any of the tested substrates were observed for P. lividus, while A. lixula showed to prefer naked stones and encrusting coralline algae Considering that A. lixula population growth may trigger barren extension on rocky shores, this may lead to a positive feedback between barren extension and A. lixula population density. Furthermore, our results suggest that the predicted rise in seawater temperature may favor A. lixula larval survival and inhibit P. lividus. Combining information on temperature tolerance with other sources of information for these species in the Mediterranean, it is possible to develop a conceptual model of the interaction between the two species and the alternative state of their habitats.  相似文献

This work reports the evaluation of some toxic effects induced by mercury on sea urchin sperm. Spermatozoa of the sea urchin Arbacia lixula L., were treated with 0.003, 0.03 and 0.06 mg/l of Hg. We estimated the amount of ATP of sperm cells with the luciferin-luciferase reaction, and we also observed motility, fertilizing activity and number of sperm bound to the eggs. The results clearly show that ATP levels are strongly affected by mercury concentrations, already three hours after the beginning of treatment. We propose the use of ATP determination in sea urchin sperm as a bioassay, because they, even more than eggs and developmental stages, readily suffer the environmental stresses.  相似文献

Early embryos of A. lixula are 10–800 times more sensitive to several neuropharmaca than are early embryos of 6 other studied species of sea urchins. Of 53 neuropharmaca studied, 25 were found hyperactive; of 19 other inhibitors of development (mitotic and metabolic poisons) only antimycin A was hyperactive for A. lixula. Both hyperactive neuropharmaca and neuropharmaca with normal activity suppress cleavage divisions and inhibit protein biosynthesis acting as antagonists of intracellular acetylcholine and monoamines. The mechanism responsible for the normal sensitivity and hypersensitivity of early sea urchin embryos is discussed.  相似文献

The concentrations of Ca2+, Na+ and H+ in echinoderm oocytes and eggs were measured during maturation and activation using ion-selective microelectrodes. In both oocytes and eggs, from three species of starfish and two species of sea urchin, the resting level of cytosolic Ca2+ was about 10-7 M. We did not detect any change in Ca2+ concentration either during hormone-induced oocyte maturation (starfish) or during egg activation (starfish and sea urchin) induced by spermatozoa or chemical agents. During 1-methyl-adenine induced maturation of starfish oocytes the intracellular level of Na+ increased from 12–35 mM to 40–90 mM, while the pH changed from 6.6–6.8 to 7.0–7.2 Aged oocytes, with intact germinal vesicles, also had elevated levels of Na+ and pH.  相似文献
