Microsatellite variation was studied at 48 microsatellite loci in 10 Drosophila melanogaster populations to investigate the population structure on the Australian east coast. Low, but statistically significant population differentiation was observed among most populations. The populations on the Australian mainland did not show evidence for isolation by distance. We conclude that the population structure of D. melanogaster on the Australian mainland is probably the result of a shared history (recent colonization). The observed differences between local D. melanogaster populations probably reflect variation in effective population sizes rather than patterns of gene flow. Two populations from Tasmania were more differentiated from the Australian mainland than a population from Israel, raising the question whether they are derived from the Australian mainland or colonized from a different source population.  相似文献

Flowering Locus C (FLC) and Frigida are two interacting genes controlling flowering time variation in Arabidopsis thaliana. Variation at these genes was surveyed in 12 A. thaliana populations sampled in France. These populations were also screened for variation at molecular markers [12 microsatellites and 19 cleaved amplified polymorphic sequence (CAPS) markers] and at seven quantitative traits measured with and without vernalization. Seven populations were highly polymorphic at markers (H(S) = 0.57 at microsatellites, 0.24 at CAPS) and showed heritable variation for bolting time and some other traits. Five populations were genetically fixed or nearly fixed. Q(ST) for bolting time without vernalization was significantly higher than F(ST), suggesting local divergent selection. One of the two haplotype groups at FLC (FLC(A)) was very predominant (frequency of 99%). The first exon of Frigida showed elevated nonsynonymous variation, and nine loss-of-function mutations were found throughout the gene. The association between loss-of-function and earlier bolting was confirmed. Overall, 18 Frigida haplotypes were detected. The pattern of variation at Frigida was largely similar to that found at markers and traits, with the same populations being fixed or highly diverse. Metapopulation dynamics is thus probably the main factor shaping genetic variation in A. thaliana. However, F(ST) for functional (FRI) vs. nonfunctional (FRI(Delta)) haplotypes was significantly higher than F(ST) at markers. Plants can achieve an appropriate phenotype in particular conditions either constitutively or plastically, depending in part on the grain size of the environmental conditions being considered. Coarse-grained environmental variation should result in selection for local adaptation and no selection on plasticity to novel levels of the coarse-grained environmental factors. We tested the hypotheses that natural populations of the well-studied model system Arabidopsis thaliana are locally adapted to spatially coarse-grained environmental variation, and that the photoperiodic regime per se is at least partially responsible for that local adaptation, by exposing natural populations to photoperiodic regimes characteristic of their native and foreign (novel) environments. We also tested the hypothesis that plasticity to novel photoperiodic regimes should appear random. We found that populations showed evidence of local adaptation at a spatially coarse grain, although not to photoperiodic regime per se. We also found that the plasticities to novel photoperiodic regimes appeared random and did not generally show evidence of adaptive divergence. Our study highlights the need for caution in extrapolating from the finding of local adaptation to the causes of local adaptation.  相似文献

Q(ST) is a commonly used metric of the degree of genetic differentiation among populations displayed by quantitative traits. Typically, Q(ST) is compared to F(ST) measured on putatively neutral loci; if Q(ST)=F(ST), this is taken as evidence of spatially heterogeneous and diversifying selection. This paper reviews the uses, assumptions and statistics of Q(ST) and F(ST) comparisons. Unfortunately, Q(ST)/F(ST) comparisons are statistically challenging. For a single trait, Q(ST) must be compared not to the mean F(ST) but to the distribution of F(ST) values. The sources of biases and sampling error for Q(ST) are reviewed, and a new method for comparing Q(ST) and F(ST) is suggested. Simulation results suggest that the distribution of neutral F(ST) and Q(ST) values are little affected by various deviations from the island model. Consequently, the distributions of Q(ST) and F(ST) are well approximated by the Lewontin-Krakauer prediction, even with realistic deviations from the island-model assumptions.  相似文献

Although extensive studies have been conducted on the genetic structure of Arabidopsis thaliana (A. thaliana) populations worldwide, the populations from China have never been studied. In this study, we collected 560 individuals from 19 natural populations of A. thaliana distributed in East China along the lower reaches of the Yangtze River, and two populations from northwest China (Xinjiang Province). We adopted two kinds of molecular marker, inter-simple sequence repeats (ISSRs) and random amplified polymorphic DNA (RAPDs) to investigate the genetic diversity within and among populations, and the correlation between the genetic and geographic distances. Thirteen ISSR primers produced 165 polymorphic bands (PPB) (96%) and 11 RAPD primers produced 162 polymorphic bands (98%) in about 560 individuals. The two marker systems generated similar patterns of genetic diversity in these natural populations. The AMOVA analysis indicated about 42-45% of the total genetic variation existed within populations, and found possible geographic structure. The Mantel test revealed a significant correlation between the geographic distance and the genetic distance of these populations in general. A close genetic relationship was found among four populations in the Jiangxi Province, and these always appeared clustered together as a monophyletic group in unweighted pair-group method with arithmetic averages dendrograms based on both ISSR and RAPD data sets. Based on the observation of recolonization and extinction of naturally distributed populations of A. thaliana, and the pattern of their genetic differentiation, the distribution of this species in China might be a result of natural dispersal under the strong influence of human activity.  相似文献

Population genetic structure of Arabidopsis lyrata in Europe   总被引:2,自引:0,他引:2  
Population genetic theory predicts that the self-incompatible and perennial herb, Arabidopsis lyrata, will have a genetic structure that differs from the self-fertilizing, annual Arabidopsis thaliana. We quantified the genetic structure for eight populations of A. lyrata ssp. petraea in historically nonglaciated regions of central Europe. Analysis of 20 microsatellite loci for 344 individuals demonstrated that, in accordance with predictions, diploid populations had high genome-wide heterozygosity (H(O) = 0.48; H(E) = 0.52), high within-population diversity (83% of total) compatible with mutation-drift equilibrium, and moderate differentiation among populations (F(ST) = 0.17). Within a single population, the vast majority of genetic variability (92%) was found at the smallest spatial scale (< 3 m). Although there was no evidence of biparental inbreeding or clonal propagation at this scale (F(IS) = 0.003), significant fine-scale spatial autocorrelation indicated localized gene flow presumably due to gravity dispersed seeds (Sp = 0.018). Limited gene flow between isolated population clusters (regions) separated by hundreds of kilometres has given rise to an isolation by distance pattern of diversification, with low, but significant, differentiation among regions (F(ST) = 0.05). The maintenance of geographically widespread polymorphisms and uniformly high diversity throughout central Europe is consistent with periglacial survival of A. lyrata ssp. petraea north of the Alps in steppe-tundra habitats during the last glacial maximum. As expected of northern and previously glaciated localities, A. lyrata in Iceland was genetically less diverse and highly differentiated from central Europe (H(E) = 0.37; F(ST) = 0.27).  相似文献

Melipona quadrifasciata is a stingless bee widely found throughout the Brazilian territory, with two recognized subspecies, M. quadrifasciata anthidioides, that exhibits interrupted metasomal stripes, and M. quadrifasciata quadrifasciata, with continuous metasomal stripes. This study aimed to estimate the genetic variability of these subspecies. For this purpose, 127 colonies from 15 Brazilian localities were analyzed, using nine species-specific microsatellite primers. At these loci, the number of alleles ranged from three to 15 (mean: 7.2), and the observed heterozygosity (Ho) ranged from 0.03–0.21, while the expected heterozygosity (He) ranged from 0.23–0.47. The genetic distances among populations ranged from 0.03–0.45. The FST multilocus value (0.23) indicated that the populations sampled were structured, and the clustering analysis showed the formation of two subgroups and two more distant populations. The first group contained the subspecies M. quadrifasciata quadrifasciata, and the other, the subspecies M. quadrifasciata anthidioides and the two M. quadrifasciata populations with continuous metasomal stripes from northern Minas Gerais. These results confirmed that the yellow metasomal stripes alone are not a good means for correctly identifying the different subspecies of M. quadrifasciata.  相似文献

拟南芥(Arabidopsis thaliana)是植物生物学的模式植物, 在分子遗传学方面已经积累了丰富的研究成果, 但目前对拟南芥自然分布的生境特点、表型变化的环境依赖特征等研究很少, 极大地限制了对拟南芥进化动力和机制的理解。为了了解在微环境下拟南芥种群分布和表型性状的变化特点, 对天山北部分布于塔尔巴哈台山、阿尔泰山和天山的10个拟南芥种群的分布特征、表型的变化特点, 以及与综合环境因子的相互关系进行了分析。结果表明: 除分枝数外, 株高、株重、根重、单个果实重量、单株果数、单株果重、果长、果实开裂力度、单株果重/总重9个特征在种群间变化显著, 可塑性能力较强; 但方差分析和变异系数结果显示, 角果长度、果实开裂力度在种群内和种群间的变化相对较小。表型特征在山系间、经纬度和海拔间的变化规律不明显。拟南芥主要分布于pH值和HCO3 -含量低, 有机质丰富, 且有一定坡度的沙土地块上。种群内拟南芥分布频度很低, 在1.56%-10.69%之间, 空间自相关距离在15.4-46.7 cm之间变化较大, 10个种群均呈现极显著集群分布, 分布的集群性受果实开裂力度的影响显著, 而果实开裂力度随环境胁迫而极显著增加。总结认为: 天山北部拟南芥生长和分布主要受微环境的影响, 在干旱环境下, 拟南芥主要通过增加繁殖分配比例, 产生难开裂的果实, 促使种子短距离扩散于母株周围, 确保子代利用原适宜生境来生存繁衍。  相似文献

Currently, there exists a limited knowledge on the extent of temporal variation in population genetic parameters of natural populations. Here, we study the extent of temporal variation in population genetics by genotyping 151 genome-wide SNP markers polymorphic in 466 individuals collected from nine populations of the annual plant Arabidopsis thaliana during 4 years. Populations are located along an altitudinal climatic gradient from Mediterranean to subalpine environments in NE Spain, which has been shown to influence key demographic attributes and life cycle adaptations. Genetically, A. thaliana populations were more variable across space than over time. Common multilocus genotypes were detected several years in the same population, whereas low-frequency multilocus genotypes appeared only 1 year. High-elevation populations were genetically poorer and more variable over time than low-elevation populations, which might be caused by a higher overall demographic instability at higher altitudes. Estimated effective population sizes were low but also showed a significant decreasing trend with increasing altitude, suggesting a deeper impact of genetic drift at high-elevation populations. In comparison with single-year samplings, repeated genotyping over time captured substantially higher amount of genetic variation contained in A. thaliana populations. Furthermore, repeated genotyping of populations provided novel information on the genetic properties of A. thaliana populations and allowed hypothesizing on their underlying mechanisms. Therefore, including temporal genotyping programmes into traditional population genetic studies can significantly increase our understanding of the dynamics of natural populations.  相似文献

Understanding existing levels of genetic variability of camel populations is capital for conservation activities. This study aims to provide information on the genetic diversity of four dromedary populations, including Guerzni, Harcha, Khouari and Marmouri. Blood samples from 227 individuals belonging to the aforementioned populations were obtained and genotyped by 16 microsatellite markers. A total of 215 alleles were observed, with the mean number of alleles per locus being 13.4 ± 6.26. All loci were polymorphic in the studied populations. The average expected heterozygosity varied from a maximum of 0.748 ± 0.122 in Guerzni population to a minimum of 0.702 ± 0.128 in Harcha population; Guerzni population showed the highest value of observed heterozygosity (0.699 ± 0.088), whereas Harcha population the lowest (0.646 ± 0.130). Mean estimates of F-statistics obtained over loci were FIS = 0.0726, FIT = 0.0876 and FST = 0.0162. The lowest genetic distance was obtained between Guerzni and Khouari (0.023), and the highest genetic distance between Harcha and Marmouri (0.251). The neighbour-joining phylogenetic tree showed two groups of populations indicating a cluster of Guerzni, Khouari and Marmouri, and a clear isolation of Harcha. The genetic distances, the factorial correspondence analysis, the analysis of genetic structure and the phylogenetic tree between populations revealed significant differences between Harcha and other populations, and a high similarity between Guerzni, Khouari and Marmouri. It is concluded from this study that the camel genetic resources studied are well diversified. However, the herd management, especially the random selection of breeding animals, can increase the level of genetic mixing between different populations, mainly among Guerzni, Khouari and Marmouri, that live in the same habitat and grazing area.  相似文献

We studied population genetic variation and structure in the fire ant Solenopsis invicta using nuclear genotypic and mitochondrial DNA (mtDNA) sequence data obtained from samples collected throughout its native range. Geographic populations are strongly differentiated at both genomes, with such structure more pronounced in Brazil than in Argentina. Higher-level regional structure is evident from the occurrence of isolation-by-distance patterns among populations, the recognition of clusters of genetically similar, geographically adjacent populations by ordination analysis, and the detection of an mtDNA discontinuity between Argentina and Brazil coinciding with a previously identified landform of biogeographical relevance. Multiple lines of evidence from both genomes suggest that the ancestors of the ants we studied resembled extant northern Argentine S. invicta , and that existing Brazilian populations were established more recently by serial long-distance colonizations and/or range expansions. The most compelling evidence for this is the corresponding increase in F K (a measure of divergence from a hypothetical ancestor) and decrease in genetic diversity with distance from the Corrientes population in northern Argentina. Relatively deep sequence divergence among several mtDNA clades, coupled with geographical partitioning of many of them, suggests prolonged occupation of South America by S. invicta in more-or-less isolated regional populations. Such populations appear, in some cases, to have come into secondary contact without regaining the capacity to freely interbreed. In 2004, the term 'ghost population' was introduced to summarize the effect of unsampled subpopulations that exchange migrants with other subpopulations that have been sampled. Estimated long-term migration rates among populations sampled will be affected by ghost populations. Although it would be convenient to be able to define an apparent migration matrix among sampled populations that incorporate the exchange of migrants with ghost populations, no such matrix can be defined in a way that predicts all features of the coalescent process for the true migration matrix. This paper shows that if the underlying migration matrix is symmetric, it is possible to define an apparent migration matrix among sampled subpopulations that predicts the same within-population and between-population homozygosities among sampled populations as is predicted by the true migration matrix. Application of this method shows that there is no simple relationship between true and apparent migration rates, nor is there a way to place an upper bound on the effect of ghost populations. In general, ghost populations can create the appearance of migration between subpopulations that do not actually exchange migrants. Comparison with published results from the application of the program, MIGRATE, shows that the apparent migration rates inferred with that program in a three-subpopulation model differ from those based on pairwise homozygosities. The apparent migration matrix determined by the method described in this paper probably represents the upper bound on the effect of ghost populations.  相似文献

利用RAPD技术对浙江省境内的七子花9个天然种群遗传多样性和遗传分化进行研究.结果表明,12种随机引物对180棵植物进行检测,共得到164个可重复的位点.多态位点百分率在14.60%~27.44%(平均为20.73%),以括苍山种群最高,其次是四明山种群,最低是观音坪种群.Shannon指数和Nei指数均反映出七子花各种群具有较低的遗传多样性,但遗传分化明显.Shannon指数显示种群内遗传多样性只占总遗传多样性的27.28%,而种群间遗传多样性却占72.72%;Nei指数表明种群内的遗传变异较小,种群间的遗传变异较大,种群间的遗传分化系数为0.7157.七子花种群间的基因流为0.1987,遗传相似度平均为0.7306,遗传距离平均为0.3150,各种群间的遗传分化明显.根据遗传距离聚类分析,大致可以将9个七子花种群分为东部和西部两大类群.  相似文献

The genetic diversity and population structure of Arabidopsis thaliana populations from Norway were studied and compared to a worldwide sample of A. thaliana to investigate the demographic history and elucidate possible colonization routes of populations at the northernmost species limit. We genotyped 282 individuals from 31 local populations using 149 single nucleotide polymorphism markers. A high level of population subdivision (F(ST) = 0.85 ± 0.007) was found indicating that A. thaliana is highly structured at the regional level. Significant relationships between genetic and geographical distances were found, suggesting an isolation by distance mode of evolution. Genetic diversity was much lower, and the level of linkage disequilibrium was higher in populations from the north (65-68°N) compared to populations from the south (59-62°N); this is consistent with a northward expansion pattern. A neighbour-joining tree showed that populations from northern Norway form a separate cluster, while the remaining populations are distributed over a few minor clusters. Minimal gene flow seems to have occurred between populations in different regions, especially between the geographically distant northern and southern populations. Our data suggest that northern populations represent a homogenous group that may have been established from a few founders during northward expansions, while populations in the central part of Norway constitute an admixed group established by founders of different origins, most probably as a result of human-mediated gene flow. Moreover, Norwegian populations appeared to be homogenous and isolated compared to a worldwide sample of A. thaliana, but they are still grouped with Swedish populations, which may indicate common colonization histories.  相似文献

The genetic structure of ten natural populations of Arabidopsis thaliana (L.) Heynh. at eight isozyme loci was studied. The populations were located in the northern part of the species range, 200 km from the north to the south along the Onega Lake coast in Karelia. Considerable genetic diversity (P99% = 43.7, Hobs = 0.003) was revealed that is not typical of populations of self-pollinating plant species. A direct correlation between the proportion of polymorphic loci and geographical latitude was shown (r = 0.68; P < 0.05). It is suggested that a high polymorphism level in Karelian Arabidopsis thaliana (L.) populations increasing from the south to the north is due to extreme environmental conditions in the northern part of the species range. The distribution of genetic diversity within and between populations is typical of self-pollinating species: the larger part of the total diversity resides among populations (GST = 0.583).  相似文献

The parasitic wasp Nasonia vitripennis has been used extensively in sex allocation research. Although laboratory experiments have largely confirmed predictions of local mate competition (LMC) theory, the underlying assumptions of LMC models have hardly been explored in nature. We genotyped over 3500 individuals from two distant locations (in the Netherlands and Germany) at four polymorphic microsatellite loci to validate key assumptions of LMC theory, in terms of both the original models and more recent extensions to them. We estimated the number of females contributing eggs to patches of hosts and the clutch sizes as well as sex ratios produced by individual foundresses. In addition, we evaluated the level of inbreeding and population differentiation. Foundress numbers ranged from 1 to 7 (average 3.0 ± 0.46 SE). Foundresses were randomly distributed across the patches and across hosts within patches, with few parasitizing more than one patch. Of the hosts, 40% were parasitized by more than one foundress. Clutch sizes of individual foundresses (average 9.99 ± 0.51 SE) varied considerably between hosts. The time period during which offspring continued to emerge from a patch or host correlated strongly with foundress number, indicating that sequential rather than simultaneous parasitism is the more common. Genetic differentiation at the regional level between Germany and the Netherlands, as estimated by Slatkin's private allele method (0.11) and Hedrick's corrected G' LT (0.23), indicates significant substructuring between regions. The level of population inbreeding for the two localities ( F IL = 0.168) fitted the expectation based on the average foundress number per patch.  相似文献

SUMMARY 1. Dispersal ability is an important ecological factor that can influence population structure. In an attempt to determine the extent that the pattern of genetic differentiation is correlated with dispersal ability in stream-dwelling aquatic insects, we used the amplified fragment length polymorphism (AFLP) technique to characterise genetic variation in four aquatic insect species: Gumaga griseola (Trichoptera: Sericostomatidae), Helicopsyche mexicana (Trichoptera: Helicopsychidae), Psephenus montanus (Coleoptera: Psephenidae) and Ambrysus thermarum (Hemiptera: Naucoridae). Individuals were sampled from several sites within two adjacent catchments in the Arizona White Mountains. In addition to the genetic analyses, a 20-week-long trapping study was used to determine the relative dispersal ability of adults of the four species examined.
2. We obtained hierarchical indicators of genetic differentiation for catchments, sites within catchments and sites across the region examined. Overall, average estimators of genetic differentiation ( F -statistics) were consistent with direct observations of organismal movement, although it was our direct observations on adult insect flight that permitted us to interpret our results correctly. This was because of the fact that a lack of genetic differentiation across watersheds can be interpreted in two ways.
3. In contrast to F -statistics, patterns of genetic isolation by distance for each species more clearly reflected dispersal ability, suggesting that such analytical approaches provide less ambiguous information about the importance of gene flow in the hierarchical partitioning of genetic variation in stream organisms.  相似文献

In the Blackcap (Aves: Sylvia atricapilla), a widespread passerine noctural migrant, a “migratory divide” between SE- and SW-migrating populations exists in Central Europe at about 14° E and south of 52° N. The autumn migratory directions are known to have a genetic basis and are expressed in orientation cages in captivity. Migratory directions of birds in the contact zone between the two populations were studied by analysing ringing data and by testing three groups of hand-raised individuals in orientation cages. Available ringing data are insufficient to establish migratory directions in the contact zone north of the Alps. Hand-raised birds from south-west Germany and the most eastern part of Austria oriented SW and SE, respectively, confirming directions known from ringing recoveries. A sample of birds from the contact zone near Linz (Austria) oriented SW to NW (mean = 268°) and was significantly different from both adjoining populations. This contrasts with results of a cross-breeding experiment with mixed pairs of SW- and SE-migrants bred in captivity: The F1-offspring chose southerly directions, intermediate between both parental populations (Helbig, 1991). It is suggested, therefore, that a distinct subpopulation with a large fraction of birds wintering in the British Isles has established itself in the contact zone. Differences in directional choices between groups of siblings from this area indicate that intrapopulation genetic variability is present. This may have led to a rapid spread of the novel W-NW migratory direction, because north of the Alps strong selection seems to be acting against mixing of SE- with SW-migrating populations.  相似文献

为了解拟南芥在天山北部的分布状况及环境依赖特点, 分析拟南芥的自然选择特征, 本文对天山北部分布的13个拟南芥(Arabidopsis thaliana)生存群落结构、组成及其与环境关系进行了研究, 并分析了拟南芥与群落主要物种的种间联结性。结果表明: 拟南芥生存的群落结构简单, 其中天山北坡中段的石河子、一四三团、沙湾、独山子地区的8个群落均为草本类型, 优势种相似, 而与伊犁果子沟、额敏和阿勒泰的5个群落差别较大。属的区系成分分析表明世界分布、北温带分布以及地中海、西亚至中亚分布型成分占大多数, 具有典型的地中海旱生植物区系分布特征, 体现了本地拟南芥分布及演化的干旱、半干旱的地理环境特点。采用双向指示种分析(TWINSPAN)将13个群落分为新疆绢蒿–猪毛菜–角果毛茛(Seriphidium kaschgaricum–Salsola collina–Ceratocephalus testiculatus)、新疆绢蒿–猪毛菜(S. kaschgaricum–S. collina)、新疆绢蒿–狭果鹤虱(S. kaschgaricum–Lappula semiglabra)、新疆绢蒿–旱麦草(S. kaschgaricum–Eremopyrum triticeum)、勿忘草–草原苔草(Myosotis sylvatica–Carex liparocarpos)5个群落类型。去势典范对应分析(DCCA)表明纬度、坡向、土壤有机质及pH值是决定天山北部拟南芥种群分布的主导因子。拟南芥分布与群落内其他物种有极强的依赖关系, 与13个群落62个主要物种的种间联结性分析表明, 共有119个正关联性种对, 明显高于72个负关联性种对, 与各群落优势种呈显著正关联。拟南芥种群分布数量在群落间差异较大, 分布于降雨较少的天山中部浅山地带拟南芥种群数量均高于降雨较丰富的天山西部伊犁果子沟地区, 是否发生适应性分化需要深入研究。  相似文献
