The activity of visual cortical neurons (area 17) was recorded in anaesthetized cats in response to sinusoidal drifting gratings. The statistical structure of the discharge of simple and complex cells has been studied as a function of the various parameters of a drifting grating: spatial frequency, orientation, drifting velocity and contrast. For simple cells it has been found that the interspike interval distributions in response to drifting gratings of various spatial frequencies differ only by a time scale factor. They can be reduced to a unique distribution by a linear time transformation. Variations in the spatial frequency of the grating induce variations in the mean firing rate of the cell but leave unchanged the statistical structure of the discharge. On the contrary, the statistical structure of the simple cell activity changes when the contrast or the velocity of the stimulus is varied. For complex cells it has been found that the invariance property described above for simple cells is not valid. Complex cells present in their activity in response to visual stimuli two different firing patterns: spikes organized in clusters and spikes that do not show this organization ('isolated spikes'). The clustered component is the only component of the complex cell discharge that is tuned for spatial frequency and orientation, while the isolated spike component is correlated with the contrast of the stimulus. 相似文献

Circulating Tumour Cells (CTCs) can be released from the primary tumour into the bloodstream and may colonize distant organs giving rise to metastasis. The presence of CTCs in the blood has been documented more than a century ago, and in the meanwhile various methods have been described for their detection. Most of them require an initial enrichment step, since CTCs are a very rare event. The different technologies and also the differences among the screened populations make the clinical significance of CTCs detection difficult to interprete. Here we will review the different assays up to now available for CTC detection and analysis. Moreover, we will focus on the relevance of the clinical data, generated so far and based on the CTCs analysis. Since the vast majority of data have been produced in breast cancer patients, the review will focus especially on this malignancy. 相似文献

This paper reports the results of computer modeling of microtubules that end up in the cortical region of a one-cell amphibian embryo, prior to the first cell division. Microtubules are modeled as initially randomly oriented semi-flexible rods, represented by several lines of point-masses interacting with one another like masses on springs with longitudinal and transverse stiffness. They are also considered to be space-filling rods floating in a viscous fluid (cytoplasm) experiencing drag forces and buoyancy from the fluid under a variable gravity field to test gravitational effects. Their randomly distributed interactions with the surrounding spherical container (the cell membrane) have a statistical nonzero average that creates a torque causing a rotational displacement between the cytoplasm and the rigid cortex. The simulation has been done for zero and normal gravity and it validates the observation that cortical rotation occurs in microgravity as well as on Earth. The speed of rotation depends on gravity, but is still substantial in microgravity. 相似文献

This paper explores the determinants of extinction of endemic plant taxa in mediterranean-climate regions in South Africa and southwestern Australia. Major threats to biodiversity in these areas include agriculture, deforestation, fragmentation, invasive alien organisms and urbanization. Case studies from the two regions show that synergisms between factors can lead to discontinuous, or non-linear, responses that have increased extinction rates (or threaten to) beyond predictions based on simple deterministic processes. 相似文献

Receptive fields of neurons of the rabbit visual cortex selective for stimulus orientation were investigated. These receptive fields were less well differentiated than those of the analogous neurons of the cat visual cortex (large in size and circular in shape). Two mechanisms of selectivity for stimulus orientation were observed: inhibition between on and off zones of the receptive field (sample type) and oriented lateral inhibition within the same zone of the receptive field (complex type). Lateral inhibition within the same zone of the receptive field also took place in unselective neurons; "complex" selective neurons differed from them in the orientation of this inhibition. A combination of both mechanisms was possible in the receptive field of the same neuron. It is suggested that both simple and complex receptive fields are derivatives of unselective receptive fields and that "complex" neurons are not the basis for a higher level of analysis of visual information than in "simple" neurons.A. Achondroplasia is the most common form of dwarfism and has an incidence of approximately 1/7500. In more than 97% of cases, it is caused by a recurrent point mutation, a G to A substitution at nucleotide position 1138 (G1138A) of the fibroblast growth factor receptor 3 gene. Although this is an autosomal dominant condition, more than 90% of all mutations occur sporadically making this one of the most mutagenic sites in the human genome. The reasons for the high spontaneous G1138A mutation rate are not known. This investigation was performed by developing a simple and rapid semi-quantitative allele specific PCR based assay capable of reliably detecting more than 25 mutant G1138A copies in a pool of 300 000 wild type molecules. Using this assay, the G1138A mutation frequency was measured in cell lines deficient in mismatch repair (LoVo, SW48) and comparing it with controls. No differences were found in the frequency of this point mutation between the mismatch repair deficient and wild type cell lines. 相似文献

Neurons in somatosensory cortex of primates process sensory information from the hand by integrating information from large populations of receptors to extract specific features. Tactile neurons in areas 1 and 2 are shown to select features such as contact area, edge orientation, motion across the skin, or direction of movement. Features coded by kinesthetic neurons in areas 3a and 2 relate to joint movement, the joint angle around which the movement occurs, or coordinated postures of the hand and arm. An even higher order cortical cell integrates tactile and kinesthetic information; these haptic neurons respond optimally to contact of objects actively grasped in the hand. These global features are coded at the expense of loss of information concerning fine-grained spatial detail. 相似文献

A model for the folding of the cerebral cortex is described which effects a direct link between the geometry of the brain and its folding pattern. Using some simple approximations to the brain's geometry the model is able to predict folding patterns. Circulating tumor cells (CTCs) might not only serve as prognostic marker but could also be useful for monitoring treatment efficacy. A multicolor flow cytometry protocol for their detection and molecular characterization in peripheral blood was developed which consisted of erythrocyte lysis followed by staining of cells with fluorochrome-labeled antibodies against CD45 and the epithelial markers EpCam and cytokeratin 7/8. For reducing the number of events acquired by flow cytometry, an electronic threshold for the fluorescent signals from the epithelial markers was applied. After establishment of the protocol by using spiking experiments, its suitability to determine the absolute number of CTCs as well as their expression of epidermal growth factor receptor (EGFR) and its phosphorylated form (phospho-EGFR) in blood samples from patients with squamous cell carcinoma of the head and neck (SCCHN) was validated. Spiking experiments demonstrated an excellent recovery (mean 85%) and a linear performance (R(2) = 0.98) of the protocol. Sensitivity and specificity were comparable to our former protocol using immunomagnetic CTC pre-enrichment. The analysis of 33 SCCHN patient samples revealed the presence of CTCs in 33.3% of cases with a mean ± SD of 1.5 ± 0.5 CTCs per 3.75 ml blood. EGFR was expressed in 100% and phospho-EGFR in 36.4% of the CTC+ cases. We have established a simple and sensitive multicolor flow cytometry protocol for detection of CTCs in patients with epithelial cancers including SCCHN which will allow their detailed molecular characterization. 相似文献

Non-linearities in apparent mass and transmissibility during exposure to whole-body vertical vibration 总被引:1,自引:0,他引:1
The causes of low back pain associated with prolonged exposure to whole-body vibration are not understood. An understanding of non-linearities in the biomechanical responses is required to identify the mechanisms responsible for the dynamic characteristics of the body, to allow for the non-linearities when predicting the influence of seating dynamics, and to predict the adverse effects caused by various magnitudes of vibration. Twelve subjects were exposed to six magnitudes, 0.25-2.5ms(-2) rms, of vertical random vibration in the frequency range 0.2-20Hz. The apparent masses of the subjects were determined together with transmissibilities measured from the seat to various locations on the body surface: the upper and lower abdominal wall, at L3, over the posterior superior iliac spine and the iliac crest. There were significant reductions in resonance frequencies for both the apparent mass and the transmissibilities to the lower abdomen with increases in vibration magnitude. The apparent mass resonance frequency reduced from 5.4-4. 2Hz as the magnitude of the vibration increased from 0.25-2.5ms(-2) rms. Vertical motion of the lumbar spine and pelvis showed resonances at about 4Hz and between 8 and 10Hz. When exposed to vertical vibration, the human body shows appreciable non-linearities in its biodynamic responses. Biodynamic models should be developed to reflect the non-linearity. 相似文献

A method has been developed for growing cultured cells in 200-μl capillary pipets in order to measure the uptake and release of materials by the cells. Feeding is accomplished by means of continuous superfusion of culture medium over the cells. The technique virtually eliminates problems caused by cultured cells detaching from the substratum during experiments and has been used to study the depolarization-dependent processes of [3H]-acetylcholine release and 22Na+ uptake by NG108-15 neuroblastoma × glioma hybrid cells. 相似文献

The extended classical receptive field (ECRF) of retinal ganglion cells has been modelled as a combination of three zero-mean Gaussians at three different scales that has been shown to be equivalent to a Biharmonic or Bi-Laplacian of Gaussian filter. It has also been shown that the ECRF can be approximated by a combination of Laplacian of Gaussian (LoG) and the Dirac-delta function. Zero-crossings detected with this operator are more informative than those detected by the traditional filters like LoG or Difference of Gaussians (DoG) that had been devised using the classical receptive field of the ganglion cells. We have also explained that such an additional information processing is not in contradiction with the recent experimental findings on the physiology of retinal ganglion cells. 相似文献
