低温胁迫对水稻幼苗不同叶龄叶片叶绿素荧光特性的影响   总被引:6,自引:0,他引:6  
以‘蜀恢162’(‘Shuhui 162’)、‘糯89-1’(‘Nuo 89-1’)、‘蜀恢162/糯89-1’(‘Shuhui 162/Nuo 89-1’)、‘奇妙香’(‘Qimiaoxiang’)和早黄矮(‘Zaohuang’ai’)5个水稻(Oryza sativa L.)品种(系)为研究对象,采用叶绿素荧光成像系统研究了低温(4℃)胁迫对水稻3叶期幼苗不同叶龄叶片叶绿素荧光特性的影响。结果表明:经低温胁迫处理后,5个水稻品种(系)幼苗3个叶龄叶片的各叶绿素荧光参数变化有明显差异,其中第一叶的各项参数均降至0。经低温处理后5个水稻品种(系)幼苗3片叶片的PSⅡ最大光化学量子产量(Fv/Fm)均明显小于对照(25℃),其中第一叶的降低幅度最大、第三叶最小。经低温胁迫处理后,5个水稻品种(系)幼苗第三叶的非光化学淬灭系数(qN)均显著大于对照,耐冷性品种‘糯89-1’幼苗第二叶的qN较对照显著增大,而其他水稻品种(系)幼苗第二叶的qN均显著小于对照;‘糯89-1’幼苗第二叶的光化学淬灭系数(qP)较对照略有增大,第三叶的qP显著大于对照;‘早黄矮’幼苗第三叶的qP也大于对照但差异不显著,而其余水稻品种(系)幼苗第二叶和第三叶的qP均显著小于对照。经低温胁迫后5个水稻品种(系)幼苗3片叶片的PSⅡ最大相对电子传递速率(rETRmax)和半饱和光强(Ik)均显著小于对照;除‘糯89-1’幼苗第三叶外,5个水稻品种(系)幼苗3片叶片的快速光响应曲线初始斜率(α)也均显著小于对照,总体上第一叶的rETRmax、Ik和α下降幅度最大、第三叶最小。研究结果揭示:受低温胁迫后,叶片自身生理差异是导致水稻幼苗不同叶龄叶片受伤害程度不同的主要因素。  相似文献

Physiological and biochemical changes related to methyl jasmonate (MeJA)-induced chilling tolerance of rice (Oryza sativa L. cv. Taichung Native 1) seedlings were investigated. Treatment of whole plants with 10 mmol m?3 MeJA for 48 h before chilling (5 °C) was optimal for the induction of chilling tolerance. MeJA greatly improved the survival ratio of chilled seedlings and ameliorated chilling injury such as demolition of membrane structure (estimated by electrolyte leakage). MeJA also prevented water loss in chilled seedlings by reducing the opening of stomata and decreasing the root bleeding rate. Putrescine and spermine levels in shoots increased but spermidine levels decreased on exposure to MeJA. In roots, putrescine levels also increased and spermidine levels increased transiently on exposure to MeJA. Activities of arginine decarboxylase (ADC; EC 4.1.1.19) and S-adenosylmethionine decarboxylase (SAMDC; EC 4.1.1.50) in both shoots and roots increased on exposure to MeJA, while the activity of ornithine decarboxylase (ODC; EC 4.1.1.17) remained unchanged. The MeJA-induced putrescine increase was inhibited by 50 mmol m?3α-difluoromethylarginine (DFMA), an irreversible inhibitor of ADC, but not by 50 mmol m?3α-difluoromethylornithine (DFMO), an irreversible inhibitor of ODC. The effect of MeJA on the induction of chilling tolerance was also reduced by 50 mmol m?3 DFMA. The effects of DFMA were partly prevented by 1 mol m?3 putrescine. This indicates that putrescine accumulation is required for the induction of chilling tolerance of rice seedlings by MeJA.  相似文献

Changes of ABA levels in chilled rice (Oryza sativa L.) seedlings of two varieties were determined. On exposure to chilling, ABA concentration rapidly increased in the chilling-tolerant cultivar (cv. Tainung 67, TNG.67) but not in the chilling-sensitive cultivar (cv. Taichung Native 1, TN.1). Both detached shoots and roots of TNG.67 seedlings showed a significant ABA increase after exposure to chilling. TN.1 seedlings could not accumulate ABA under low temperature but well-watered status. Exogenous application of the ABA biosynthetic inhibitor, fluridone, reduced ABA accumulation, as well as survival ratio of chilled TNG.67 seedlings. Electrolyte leakage and leaf conductance were also increased by the inhibitor and the effects could be reversed by exogenously applied ABA. ABA concentrations in xylem sap of TNG.67 seedlings increased within 4 h after chilling, and this was temporally coincident with the reduction of leaf conductance. The roles of endogenous ABA in the tolerance of rice seedlings to chilling on a whole plant basis are discussed and suggested.  相似文献

杂草稻落粒粳(Oryza sativa)发生在我国辽宁丹东.落粒粳植株明显高于当地大多数栽培品种,颖果呈中长型,成熟后容易掉粒;果壳稻草色或黄间黑灰色,小穗无芒或有芒,芒长4~12 cm;颖果千粒重235 g,种皮桔红色.落粒粳种子在13~38 ℃条件下的发芽率均大于88%,水层2.5~10 cm处理,落粒粳植株干重减少50%~69%.在幼苗期,落粒粳对无芒稗的各项影响因子均明显大于化感潜力品种I-kung-pao,表明落粒粳无化感作用.落粒粳可以忍耐0.5%的盐碱.  相似文献

Changes in abscisic acid (ABA) contents in Cd-treated rice (Oryza sativa L.) seedlings of two cultivars were investigated. On treatment with CdCl2, the ABA content rapidly increased in the leaves and roots of Cd-tolerant cultivar (cv. Tainung 67, TNG67) but not in the Cd-sensitive cultivar (cv. Taichung Native 1, TN1). The reduction of transpiration rate of TN1 caused by Cd was less than that of TNG67. Exogenous application of ABA reduced transpiration rate, decreased Cd content, and enhanced Cd tolerance of TN1 seedlings. Exogenous application of the ABA biosynthesis inhibitor, fluridone, reduced ABA accumulation, increased transpiration rate and Cd content, and decreased Cd tolerance of TNG67 seedlings. Fluridone effect on Cd toxicity of TNG67 seedlings was reversed by the application of ABA. The roles of endogenous ABA in Cd tolerance of rice seedlings are discussed and suggested.  相似文献

The 20S proteasome is the proteolytic complex that is involved in removing abnormal proteins, and it also has other diverse biological functions. Its structure comprises 28 subunits arranged in four rings of seven subunits, and exists as a hollow cylinder. The two outer rings and two inner rings form an 7β7β77 structure, and each subunit, and β, exists as seven different types, thus giving 14 kinds of subunits. In this study, we report the primary structures of the 14 proteasomal subunit subfamilies in rice (Oryza sativa), representing the first set for all of the subunits from monocots. Amino acid sequence homology within the rice family (-type: 28.9–42.1%; β-type: 17.2–31.9%) were lower than those between rice subunits and corresponding orthologs from Arabidopsis and yeast (-type: 49.2–94.5%; β-type: 34.8–87.7%). Structural features observed in eukaryotic proteasome subunits, i.e., - or β-type signature at the N-termini, Thr active sites in β1, β2 and β5 subunits, and nuclear localization signal-like sequences in some -type subunits, were shown to be conserved in rice.  相似文献

Complete submergence of plants prevents direct O(2) and CO(2) exchange with air. Underwater photosynthesis can result in marked diurnal changes in O(2) supply to submerged plants. Dynamics in pO(2) had not been measured directly for submerged rice (Oryza sativa), but in an earlier study, radial O(2) loss from roots showed an initial peak following shoot illumination. O(2) dynamics in shoots and roots of submerged rice were monitored during light and dark periods, using O(2) microelectrodes. Tissue sugar concentrations were also measured. On illumination of shoots of submerged rice, pO(2) increased rapidly and then declined slightly to a new quasi-steady state. An initial peak was evident first in the shoots and then in the roots, and was still observed when 20 mol m(-3) glucose was added to the medium to ensure substrate supply in roots. At the new quasi-steady state following illumination, sheath pO(2) was one order of magnitude higher than in darkness, enhancing also pO(2) in roots. The initial peak in pO(2) following illumination of submerged rice was likely to result from high initial rates of net photosynthesis, fuelled by CO(2) accumulated during the dark period. Nevertheless, since sugars decline with time in submerged rice, substrate limitation of respiration could also contribute to morning peaks in pO(2) after longer periods of submergence.  相似文献

Protein kinases in plants have not been examined in detail, but protein phosphorylation has been shown to be essential for regulating plant growth via the signal transduction system. A Ca2+- and phospholipid-dependent protein kinase, possibly involved in the intracellular signal transduction system from rice leaves, was partially purified by sequential chromatography on DE52, Phenyl Superose and Superose 12. This protein kinase phosphorylated the substrate, histone III-S, in the presence of Ca2+ and phosphatidylserine. The apparent molecular mass of the Ca2+- and phosphatidylserine-dependent protein kinase (Ca2+/PS PK), determined by phosphorylation in SDS-polyacrylamide gel containing histone III-S, was 50 kDa. The protein kinase differed from Ca2+-dependent protein kinase (CDPK) in rice leaves in that Ca2+/PS PK showed phospholipid dependency and the molecular mass of Ca2+/PS PK exceeded that of CDPK. Investigations were carried out on changes in Ca2+/PS PK and CDPK activity in the cytosolic and membrane fractions during germination. The maximum activity of Ca2+/PS PK in the cytosolic fraction was observed before imbibition and that of CDPK in the membrane fraction was noted at 6 days following imbibition. Protein kinases are likely to regulate plant growth through protein phosphorylation.  相似文献

Comparative analysis revealed that a xantha rice mutant (cv. Huangyu B) had higher ratios of chlorophyll (Chl) a/b and carotenoids/Chl, and higher photosynthetic efficiency than its wild type parent (cv. II32 B). Unexpectedly, the mutant had higher net photosynthetic rate (P N) than II32 B. This might have resulted from its lower non-photochemical quenching (qN) but higher maximal photochemical efficiency (FV/FM), higher excitation energy capture efficiency of photosystem 2 (PS2) reaction centres (FV′/FM′), higher photochemical quenching (qP), higher effective PS2 quantum yield (ΦPS2), and higher non-cyclic electron transport rate (ETR). This is the first report of a chlorophyll mutant that has higher photosynthetic efficiency and main Chl fluorescence parameters than its wild type. This mutant could become a unique material both for the basic research on photosynthesis and for the development of high yielding rice cultivars.  相似文献

用RACE方法获得了全长的水稻谷胱甘肽合成酶(GS)基因的cDNA,并命名为OsGS(GeneBankaccession No.:AY453405)。该cDNA全长1 892 bp,编码一个由540个氨基酸组成的多肽,预测其氨基端(N端)含有一段定位叶绿体的信号肽。比较水稻基因组定位结果表明OsGS基因位于水稻12号染色体短臂上,转录区全长6 321 bp,由12个外显子和11个内含子组成。通过RT-PCR对OsGS在水稻正常生长条件和逆境条件下的表达进行了研究。结果表明,在正常生长条件下,OsGS在水稻幼苗的根和叶以及抽穗期水稻的根中表达;但不在抽穗期水稻的叶、茎和幼穗中表达,这显示OsGS在水稻中的表达具有发育和组织特异性。利用抽穗期水稻的叶片为材料,经高温、干旱和重金属逆境处理后,OsGS在抽穗期叶片中的转录被诱导表达;而在盐、低温、伤害逆境下则不被诱导。在轻度和中度干旱胁迫4 h后OsGS基因可被诱导表达。外源ABA处理也能够提高OsGS的转录水平,这显示OsGS可能是依赖ABA信号途径的环境胁迫诱导基因。  相似文献

为鉴定水稻(Oryza sativa)响应缺铁的根冠长距离信号转导物质, 采用TMT标记技术分析了不同浓度铁处理下水稻韧皮部汁液的蛋白质组学变化, 共鉴定出206个差异蛋白, 其中54个蛋白表达丰度上调, 152个蛋白表达丰度下调。差异蛋白的KEGG通路分类主要包括激素信号代谢、谷胱甘肽代谢、碳代谢以及mRNA转运等代谢途径。此外, 对差异蛋白对应的生理指标进行测定, 发现激素、蔗糖、谷胱甘肽和转运蛋白等在缺铁条件下变化显著, 后续对这些差异蛋白的功能研究有助于揭示水稻响应铁素营养的长距离信号途径。  相似文献

缺磷条件下,供试粳稻品种植株的单株鲜重、干重、全磷含量和单株磷累积量等具有较大差异,从中筛选出磷高效品种TP309和优质8号,其中TP309较磷低效品种早88-1的全磷含量和单株磷累积量分别增加37.50%~40.00%和82.76%~102.00%。单株根数、平均根长和根系体积与单株磷素吸收量的相关程度较小,光合速率(Pn)、叶片可溶蛋白含量和叶片可溶性糖含量均表现为随着吸磷量的增加而不断增大。  相似文献

Mapping QTLs for phosphorus deficiency tolerance in rice (Oryza sativa L.)   总被引:23,自引:0,他引:23  
 The amplified fragment length polymorphism (AFLP) technique combined with selective genotyping was used to map quantitative trait loci (QTLs) associated with tolerance for phosphorus (P) deficiency in rice. P deficiency tolerant cultivar IR20 was crossed to IR55178-3B-9-3 (sensitive to P-deficiency) and 285 recombinant inbred lines (RILs) were produced by single-seed descent. The RILs were phenotyped for the trait by growing them in P-sufficient (10.0 mg/l) and P-deficient (0.5 mg/l) nutrient solution and determining their relative tillering ability at 28 days after seeding, and relative shoot dry weight and relative root dry weight at 42 days after seeding. Forty two of each of the extreme RILs (sensitive and tolerant) and the parents were subjected to AFLP analysis. A map consisting of 217 AFLP markers was constructed. Its length was 1371.8 cM with an average interval size of 7.62 cM. To assign linkage groups to chromosomes, 30 AFLP and 26 RFLP markers distributed over the 12 chromosomes were employed as anchor markers. Based on the constructed map, a major QTL for P-deficiency tolerance, designated PHO, was located on chromosome 12 and confirmed by RFLP markers RG9 and RG241 on the same chromosome. Rice ( Oryza sativa L.) is considered a drought-sensitive crop species; however, within this species, there are considerable varietal differences in sensitivity to this environmental stress. In the present work, the effect of water stress on germination, plant growth and root proteins in three rice cultivars (Sinaloa, IR10120 and Chiapas) was analyzed. Seed germination and plant growth were found to be significantly inhibited by polyethylene glycol (PEG)-imposed water deficit in cv. Sinaloa; cvs IR10120 and Chiapas were more tolerant to water stress. Fluorographs of two-dimensional electropherograms of in vivo-labeled polypeptides were analyzed to identify changes in the root protein patterns that resulted when plants were grown in the presence of 10% PEG for 10 days. The treatment induced or increased the synthesis of eight polypeptides or groups of polypeptides in cv. Sinaloa, seven in cv, IR10120 and four in cv. Chiapas. The synthesis of several polypeptides was decreased by the PEG treatment in cv. Sinaloa and cv. IR10120. Most of these PEG-induced changes in the root protein patterns were cultivar-specific and only one 26-kDa protein with a pI of 6.0 was induced by water deficit in the two cultivars Sinaloa and IR10120.  相似文献

应用常规石蜡切片和荧光显微镜观察水稻(Oryza sativa)受精过程中雌雄性细胞融合时的形态特征及时间进程,确定合子期,为花粉管通道转基因技术的实施提供理论依据。结果表明:授粉后,花粉随即萌发,花粉管进入羽毛状柱头分支结构的细胞间隙,继续生长于花柱至子房顶部的引导组织的细胞间隙中,而后进入子房,在子房壁与外珠被之间的缝隙中向珠孔方向生长,花粉与花粉管均具有明显的绿色荧光。花粉管经珠孔及珠心表皮细胞间隙进入一个助细胞,释放精子。精子释放前,两极核移向卵细胞的合点端:两精子释放于卵细胞与中央细胞的间隙后,先后脱去细胞质,然后分别移向卵核和极核,移向卵核的精核快于移向极核的精核:精核与两极核在向反足细胞团方向移动的过程中完成雌雄核融合。大量图片显示了雌雄性核融合的详细过程以及多精受精现象。水稻受精过程经历的时间表如下:授粉后,花粉在柱头萌发:花粉萌发至花粉管进入珠孔大约需要0.5小时:授粉后0.54,时左右,花粉管进入一个助细胞,释放精子:授粉后0.5—2.5小时,精卵融合形成合子:授粉后约10.0小时,合子第1次分裂,合子期为授粉后2.5-10.04,时:授粉后1.0-3.04,时,精核与两极核融合:授粉后约5.0小时,初生胚乳核分裂。’  相似文献

水溶性环氧化合物水解酶(Soluble Epoxide Hydrolase,SEH)是一组催化环氧化合物水解为相应邻位二醇的酶类,在哺乳动物、植物、昆虫和微生物体内广泛存在。通过BLAST对水稻基因组的蛋白质数据库进行搜索,获得10个水溶性环氧化物水解酶(Soluble Epoxide Hydrolase SEH)sEH蛋白的同源序列。经分析发现这些基因在水稻不同胁迫处理下各个部位都有所表达,而且不同成员之间的表达模式存在较大的差异。水稻sEH蛋白主要参与角质层形成,应激反应,以及病原防御等生理过程,特别在脱毒过程中扮演着重要的角色。对蛋白质多序列联配和三级结构预测结果表明:水溶性环氧化合物水解酶的核心结构域由3个催化残基Asp、His和Asp形成三位一体的催化活性构象。这类基因的表达受抗逆环境诱导,其功能与抗逆性有关,为基因工程抗逆育种提供了参考。  相似文献

Since apomixis has a close correlation with polyploidy and sterility, a number of autotriploids with no sexual reproductivity were induced and apomictic germplasm were screened in Oryza sativa L. As a result, an autotriploid line, named TAR, was cytoembryologically identified which possessed apomictic property, with an average seed-set rate of 10% per panicle. Karyotype analysis proved that all the progeny seeds of TAR carried 36 chromosomes in the generations tested. Priliminary cytological observations revealed that all the ovaries of TAR had embryo sac differentiation, 33% of which developed into normal megagametophyte, 9% with previous embryogenesis prior to anthesis, and about 58% differentiated abnormally, i.e. disordered polarization, absent female generative unit and more than 2 polar nuclei. In TAR, the frequencies of chromosome configuration of 12 Ⅲ, 11 Ⅱ + 1 Ⅱ +1 Ⅰ. L0Ⅲ +2Ⅱ +2 Ⅰ, 9Ⅲ+3Ⅱ +3 Ⅰ, 8Ⅲ+4Ⅱ +4 Ⅰ and 7Ⅲ+5 Ⅱ +5 Ⅰ were ll%, 17%, 15%, 26%, 20% and 11% respectively at metaphase Ⅰ . While in the check line T-15 of autotriploid only 7 % of the ovaries observed had embryo sac development, and the progenies of this triploid line were aneuploids with chromosome number of 25~27. In T-15, the frequencies of chromosome configuration of 12 Ⅲ, 11Ⅲ +1 Ⅱ +1 Ⅰ, 10 Ⅲ +2 Ⅱ +2 Ⅰ , 9Ⅲ+3 Ⅱ +3 Ⅰ and 8 Ⅲ+4 Ⅱ +4 Ⅰ were 24%, 16%, 36%, 17% and 7% respectively at metaphse Ⅰ . The above observations indicated that some megaspore mother cell in TAR might undergo apomeiosis and where it gave rise to unreduced embryo sac, the unreduced eggs or synergids developed into embryos without fertilization and polar nuclei produced endosperm by pseudogamy.  相似文献

与对低温不敏感的粳稻台北309和武育粳相比,对低温敏感的籼稻IR64、CA212和Pusa经光照条件下8℃处理后最大光合速率(Pmax)和原初光化学效率(Fv/Fm)下降较多,出现了O2-·、过氧化氢、氧化型谷胱甘肽(GSSG)和氧化型抗坏血酸(DHA)的大量累积,其GSSG和DHA的含量分别与叶绿素含量的下降呈极显著负相关,表明光照条件下低温胁迫下,还原态的谷胱甘肽(GSH)和抗坏血酸的再生受阻,不能有效地清除活性氧,导致其叶绿素含量降低和光合能力受抑,而汕优63的变化位于上述两种类型之间。其中AsA/DHA和GSH/GSSG的变化与叶绿素含量的变化呈极显著正相关。  相似文献
