In order to regulate cell volume during hyperosmotic stress, the intertidal copepod Tigriopus californicus, like other aquatic crustaceans, rapidly accumulates high levels of intracellular alanine, proline, and glycine. Glutamate-pyruvate transaminase (GPT; EC 2.6.1.2), which catalyzes the final step of alanine synthesis, is genetically polymorphic in T. californicus populations at Santa Cruz, California. Spectrophotometric studies of homogenates derived from a homozygous isofemale line of each of the two common GPT alleles indicated that the GPTF allozyme has a significantly higher specific activity than the GPTS allozyme. Under conditions of hyperosmotic stress, individual adult copepods of GPTF and GPTF/S genotypes accumulated alanine, but not glycine or proline, more rapidly than GPTS homozygotes. When young larvae were subjected to the same hyperosmotic conditions, GPTS larvae suffered a significantly higher mortality than GPTF or GPTF/S larvae. These results suggest that the biochemical differences among GPT allozymes result in specific physiological variation among GPT genotypes and that this physiological variation is manifested in differential genotypic survivorships under some naturally occurring environmental conditions.This work was supported in part by a grant from the Lerner Fund for Marine Research of the American Museum of Natural History, an NIH Training Grant in Integrative Biology, and NIH Grants GM 28016 and GM 10452.  相似文献

Glutamate-oxaloacetate transaminase (GOT; EC 2.6.1.1) occurs as two electrophoretically distinguishable isozymes in the copepod Tigriopus californicus. The slower-migrating form, referred to as GOT2, is shown to be associated with the mitochondrial cell fraction. GOT2 phenotypes are inherited in typical Mendelian fashion, indicating that they are encoded by a nuclear gene. Allelic frequencies for electrophoretic variants of the two Got loci in 12 California populations of T. californicus show a sharp differentiation of local populations. We investigated population dynamics, genetic diversity and spatial structure in the aphid species Macrosiphoniella tanacetaria, a specialist herbivore feeding on tansy, Tanacetum vulgare. Tansy plants (genets) consist of many shoots (ramets), and genets are grouped in sites. Thus, aphids feeding on tansy can cluster at the level of ramets, genets and sites. We studied aphid population dynamics in 1997 and 2001 and found that within sites: (i). at any time, aphids used only a fraction of the available ramets and genets; (ii). at the level of ramets, most aphid colonies survived only one week; (iii). at the level of genets, mean survival time was less than 4 weeks; and (iv). colonization and extinction events occurred throughout the season. We sampled aphids in seven sites in the Alsace region, France (4-45 km apart) and two sites in Germany in 1999 to study genetic structure within and between populations. Genetic analyses using nine microsatellite loci showed that: (i). genotypic variability was high, (ii). none of the populations was in Hardy-Weinberg equilibrium, (iii). heterozygote deficits and linkage disequilibria were frequent, and (iv). all populations were genetically differentiated, even at a small geographical scale. Renewed sampling of the Alsace sites in 2001 showed that three populations had become extinct and significant genetic changes had occurred in the remaining four populations. The frequencies of extinction and colonization events at several spatial scales suggest a hierarchical metapopulation structure for M. tanacetaria. Frequent population turnover and drift are likely causes for the genetic differentiation of M. tanacetaria populations.  相似文献

The results of several experiments on reproductive behaviour of Tigriopus fulvus, indicate that females release a sex pheromone promoting sexual recognition and attraction in males. This compound has been verified to be not only species-specific, but also slightly modified in specimens belonging to different geographical populations. The compound possesses a mass of between 100 and 1000 daltons. A male can invest from one to 15 days in the precopulatory phase; delayed fertilization causes a strong reduction in the average number of offspring produced by parental specimens belonging to the oldest agegroups, but does not have any effect on the sex ratio.  相似文献

Extinctions can dramatically reshape biological communities. As a case in point, ancient mass extinction events apparently facilitated dramatic new evolutionary radiations of surviving lineages. However, scientists have yet to fully understand the consequences of more recent biological upheaval, such as the megafaunal extinctions that occurred globally over the past 50 kyr. New Zealand was the world''s last large landmass to be colonized by humans, and its exceptional archaeological record documents a vast number of vertebrate extinctions in the immediate aftermath of Polynesian arrival approximately AD 1280. This recently colonized archipelago thus presents an outstanding opportunity to test for rapid biological responses to extinction. Here, we use ancient DNA (aDNA) analysis to show that extinction of an endemic sea lion lineage (Phocarctos spp.) apparently facilitated a subsequent northward range expansion of a previously subantarctic-limited lineage. This finding parallels a similar extinction–replacement event in penguins (Megadyptes spp.). In both cases, an endemic mainland clade was completely eliminated soon after human arrival, and then replaced by a genetically divergent clade from the remote subantarctic region, all within the space of a few centuries. These data suggest that ecological and demographic processes can play a role in constraining lineage distributions, even for highly dispersive species, and highlight the potential for dynamic biological responses to extinction.  相似文献

该研究利用SSR分子标记,对肋果沙棘(Hippophae neurocarpa)分布区北缘青海祁连地区5个自然居群进行分析,旨在了解小地理尺度下肋果沙棘北缘居群的遗传多样性和片段化分布居群的遗传结构,为肋果沙棘居群的资源保护提供了依据。采用6对微卫星引物对107个样本DNA进行扩增,共检测到27个等位变异,变幅为2~9个,平均每个位点有4.67个,平均观测杂合度(Ho)和期望杂合度(He)分别为0.142和0.230,Shannon信息指数(I)的变幅介于0.280~0.567之间,平均值为0.374,说明肋果沙棘北缘居群的遗传多样性较为丰富。遗传分化系数Fst=0.483,分子方差分析(AMOVA)表明肋果沙棘有48.33%的变异存在于居群间,51.67%存在于居群内。对5个居群之间的遗传距离与地理距离做Mental检验,结果表明遗传距离与地理距离相关性不显著,对基因流检测发现居群间的Nm为0.328,推测遗传漂变是居群分化的关键因素之一。Structure分析把5个居群分为2组。UPGMA聚类分析表明5个居群聚为2个分支,其中居群ARX为单独的一支,与主坐标分析的结果一致。基于分布区北缘青海祁连肋果沙棘自然居群的遗传结构分析,建议应尽可能多地保护不同的地方居群。  相似文献

During the oceanographic campaign, ALMOFRONT I (April 24–May 26, 1991) a total of 50 vertical zooplankton hauls was carried out in the Alboran Sea using a triple net. Leg A (18 stations; 18 hauls) described a widely spaced grid in the region of the Atlantic current and some northern eddies, while Leg B (16 stations; 32 hauls, 6 hydrodynamically characterized sites sampled during a two day period) covered a more restricted area in the frontal zone. Ninety-one species of copepods were found in 100 samples collected by either 200 µm or 500 µm mesh size nets, including 32 genera and 26 families. Copepod abundance, structure indexes (species richness, evenness, Shannon species diversity index, standardized for unequal sample counts) and species abundance patterns (as rank-frequency diagrams) are presented and compared among the sites of leg-B. Copepod abundance was found to be the highest in the more productive sites of the Atlantic current. Structure indexes values and the changing shape of rank-frequency diagrams give a coherent view of the ecological succession stages of the copepod community. Juvenile stage (1) develops from the Atlantic divergence zone, left side of the jet. More mature stages (1 and 2) occur on the right side of the jet influenced by cross frontal flow. Both abundance and structure indexes decrease within an anticyclonic gyre south to the current. A situation more influenced by oligotrophic conditions was observed in a Mediterranean anticyclonic gyre north of the current. The importance of lateral displacement and meandering of the Atlantic current across the Alboran Sea to the community structure is discussed.  相似文献

应用ISSR分子标记技术,对来自广西、贵州3个种源的马尾松开展遗传多样性、遗传结构及遗传距离等研究。结果表明:从100条引物中筛选出12条引物,共扩增出92个条带,86条具有多态性。 POPGENE分析显示:马尾松群体水平上的Nei’ s基因多样性指数的变化范围为0.1824~0.2065,Shannon遗传多样性指数的变化范围为0.2818~0.3178,3个群体的多态性水平差异不大;物种水平上的多态性百分率为93.48%, Nei’ s基因多样性指数为0.2842,Shannon信息指数为0.4381;表明马尾松在物种水平上具有较高水平的遗传多样性。遗传结构分析显示:马尾松的基因分化系数( Gst)为0.3153,表明遗传变异主要来源于群体内;基因流Nm为1.0853,表明不同群体间存在一定的基因流动。 AMOVA分析显示:马尾松的遗传分化指数( Fst)为0.246( P=0.001),表明群体间已出现明显的遗传分化。 UPGMA聚类和Mantel检测结果显示:每个群体内的个体均能很好地首先聚集为一个分支,群体间的遗传距离与地理距离之间存在显著相关性( r=0.972, P=0.001)。这说明马尾松在裸子植物界中具有较高水平的遗传多样性,遗传变异主要分布于群体内,群体间已出现了明显的遗传分化,这种分化并非由遗传漂变引起,可能与地理生境的差异有关。  相似文献

The dogwhelk Nucella lapillus is a predatory marine gastropod populating North Atlantic rocky shores. As with many other gastropod species, N. lapillus was affected by tributyltin (TBT) pollution during the 1970s and 1980s, when local populations became extinct. After a partial ban on TBT in the United Kingdom in 1987, vacant sites have been recolonized. N. lapillus lacks a planktonic larval stage and is therefore expected to have limited dispersal ability. Relatively fast recolonization of some sites, however, contradicts this assumption. We compared levels of genetic diversity and genetic structuring between recolonized sites and sites that showed continuous population at three localities across the British Isles. No significant genetic effects of extinction/recolonization events were observed in SW Scotland and NE England. In SW England we observed a decrease in genetic diversity and an increase in genetic structure in recolonized populations. This last result could be an artefact, however, due to the superposition of other local factors influencing the genetic structuring of dogwhelk populations. We conclude that recolonization of vacant sites was accomplished by a relatively high number of individuals originating from several source populations (the 'migrant-pool' model of recolonization), implying that movements are more widespread than expected on the basis of development mode alone. Comparison with published data on genetic structure of marine organisms with contrasted larval dispersal supports this hypothesis. Our results also stress the importance of local factors (geographical or ecological) in determining genetic structure of dogwhelk populations.  相似文献

We investigated the genetic population structure of two rare myrmecophilous lycaenid butterflies, Maculinea nausithous and M. teleius, which often live sympatrically and have similar biology. In Europe, both species occur in highly fragmented populations and are vulnerable to local extinction. The proportion of variable allozyme loci, average heterozygosity and genetic differentiation among populations was higher in M. nausithous than in sympatrically living M. teleius populations. We hypothesise that the differences in heterozygosity are mainly due to the known higher efficiency of typical host ant nests in rearing M. nausithous pupae compared to M. teleius pupae. This implies a larger probability of larval survival in M. nausithous, which buffers populations against environmental and demographic stochasticity. In contrast, the lower carrying capacity of ant nests in rearing M. teleius pupae requires higher nest-densities and makes M. teleius populations more prone to losing genetic variation through drift if this condition is not fulfilled. The single investigated Russian population of M. teleius showed much higher levels of heterozygosity than any of the Polish populations, suggesting a more viable and still intact metapopulation structure.  相似文献

For a study of diversity and genetic structuring in Melipona quadrifasciata, 61 colonies were collected in eight locations in the state of Minas Gerais, Brazil. By means of PCR analysis, 119 ISSR bands were obtained, 80 (68%) being polymorphic. He and H B were 0.20 and 0.16, respectively. Two large groups were obtained by the UPGMA method, one formed by individuals from Januária, Urucuia, Rio Vermelho and Caeté and the other by individuals from São João Del Rei, Barbacena, Ressaquinha and Cristiano Otoni. The Φst and θB values were 0.65 and 0.58, respectively, thereby indicating high population structuring. UPGMA grouping did not reveal genetic structuring of M. quadrifasciata in function of the tergite stripe pattern. The significant correlation between dissimilarity values and geographic distances (r = 0.3998; p < 0.05) implies possible geographic isolation. The genetic differentiation in population grouping was probably the result of an interruption in gene flow, brought about by geographic barriers between mutually close geographical locations. Our results also demonstrate the potential of ISSR markers in the study of Melipona quadrifasciata population structuring, possibly applicable to the studies of other bee species.  相似文献

Recent taxonomic revisions in the genus Diaphanosoma have made the uncertainty in species discrimination with morphological characters obvious. Therefore species characterization on a genetic basis seems to be required.Here we examined the genetic structure of two Diaphanosoma mongolianum populations and three Diaphanosoma brachyurum populations in Central Europe by allozyme electrophoresis. A genetic differentiation between both species was evident. 5 out of 9 tested loci carried diagnostic alleles. Both species differed in their habitat choice: D. mongolianum was adapted to higher trophic levels than D. brachyurum. Co-occurrence was observed in a eutrophic lake. Populations sampled from their preferred habitats showed high genotype diversities.Most genotype frequency distributions were in agreement with Hardy-Weinberg expectations and temporal (between years) changes in genotypic compositions were not common. This indicated annually occurring bouts of sexual reproduction, similar to intermittent Daphnia populations. Genetic differentiation among lakes suggested restricted gene flow.  相似文献

The distribution patterns and genetic structure of the Pellioditis marina species complex in Belgium and The Netherlands were compared between four consecutive seasons. Different types of habitats (coast, estuary, semi-estuary and lake) with different degrees of connectivity were sampled. In addition, each habitat type was characterised by either temporal or permanent algal deposits. We screened 426 bp of the mitochondrial cytochrome oxidase c (COI) gene with the single-strand conformation polymorphism (SSCP) method in 1615 individuals of Pellioditis marina. The 51 haplotypes were divided into four (sympatric) lineages, with divergences ranging from 0.25 to 10.6%. Our results show that the lineages have different temporal dynamics, which may be linked to abiotic factors. Analysis of Molecular Variance (AMOVA) indicated a significant structuring in the PmI lineage, which correlated with habitat characteristics and which changed over time (Mantel, r = 0.51; p = 0.126). Intrapopulational diversity was similar in all locations, and temporal changes in haplotype frequencies were not higher in temporary than in permanent algal deposits. Instead, the results of the temporal survey indicated that (some) P. marina populations are characterised by a metapopulation structure. It is emphasized that a complete and correct interpretation of processes causing genetic structuring within species and of the genetic structure itself can only be done when analyses are performed at several time points.  相似文献

海草是生长在潮间带和潮下带的单子叶植物,由海草植物组成的海草床是生态系统服务价值最高的生态系统之一.然而,近几十年人类活动干扰、全球气候变化等因素导致海草床衰退严重.海菖蒲是分布于热带、体型最大的雌雄异株海草,我国位于该物种的分布北缘,本文对其克隆多样性和遗传结构进行研究,以期为该海草的保护提供参考.采用4对多态微卫星标记对采自海南岛4个地点的现存海菖蒲种群的样品进行基因型分型.结果表明:海菖蒲种群克隆多样性和遗传多样性较低,这与所研究种群处于分布区北缘有关;种群间遗传分化值范围较大(0.073~0.309),这可能是由于分布于不同港湾的种群间距离范围较大以及局域绝灭/再拓殖的遗传漂变效应所致;各种群未发现近期经历种群瓶颈的信号,很可能是由于种群内遗传多样性已经很低,种群减小未能导致遗传多样性明显降低.根据种群遗传特征,提出了重点保护种群的建议,鉴于目前我国海菖蒲等海草快速衰退的局面,应强化海草保护并实施海草床生态恢复.  相似文献

A new species of cyclopoid copepod, Diacyclops biceri sp. nov., is described from Lake Baikal. It was found in the interstitial water of a sandy beach at Buchta Peschanaya on the western shore of the central basin of Baikal. The new species is unique in possessing 2-segmented endopods in swimming legs 3 and 4. Swimming legs 2 to 4 have 3-segmented exopods. The slender body form, the lack of the antennal exopodal seta, and the presence of a secondary pseudosomite anterior to the genital double somite of the adult female are interpreted as adaptations to the interstitial habitat. The harpacticoid Epactophanes richardi Mrazek was found in the same interstitial habitat as D. biceri.  相似文献

The effects of inbreeding on fitness and themaintenance of genetic load in metapopulationsof the endangered Glanville fritillarybutterfly (Melitaea cinxia) were examinedin four laboratory experiments. In FinlandM. cinxia occurs as a large metapopulationconsisting of small local populations with fastturnover, whereas in southern France thespecies has a more continuous populationstructure. In the experiments, we compared theperformance of crosses between full sibs,crosses between members of different familieswithin populations, and crosses betweenindividuals from different populations. Theseexperiments were replicated using insects fromtwo different regions, Finland and southernFrance, between which the frequency of naturalinbreeding should differ substantially becauseof differing population structure. In Finnishbutterflies, the rate of successful mating waslower among insects derived from small thanfrom large natural populations, probablyreflecting the effect of past inbreedinghistory. Mating between full sibs lowered egghatching rate in all experiments. Thisreduction of egg hatching rate was more severeamong French butterflies with a more continuouspopulation structure than among Finnishbutterflies with small naturally fragmentedpopulations and with a history of repeatedrounds of inbreeding in the past. This resultsuggests that recurrent inbreeding has led topartial purging of deleterious recessives fromthe Finnish metapopulation. Nonetheless,substantial genetic load still remains in thismetapopulation, and we discuss possible reasonswhy this should be the case.  相似文献
