Shoot and flower primordia of vegetative and flower buds ofextremely or very hardy conifers belonging to the subfamilyAbietoideae of the Pinaceae, survived between –40 and–70?C by extraorgan freezing, which differed greatly dependingupon species. The water in these organs gradually froze outwith decreasing temperatures when cooled very slowly, whichenabled these organs to survive %40?C or below. The same icesegregation in shoot and flower primordia by extraorgan freezingwas observed in most of the temperate conifers belonging toTaxaceae, Cephalotaxaceae, Taxodiaceae and Cuppressaceae, makingthem resistant to temperatures between –15 and –25?C.In these conifers, scales acted as an ice sink, unlike the conifersof Abietoideae. The rates of cooling and exosmosis of waterin the shoot or flower primordia, their size, and their abilityto tolerate freeze-dehydration or its related stress play animportant role in determining whether death is caused by freeze-dehydrationor intraorgan freezing. Even in very hardy conifers, low temperature exotherms fromfreezing within the shoot primordia appeared between –30and –35?C on the DTA profiles when cooled continuouslyunder laboratory conditions from 5?C to –50?C at 2 to5?C/h. Appearance of low temperature exotherms always resultedin death. However, in the coldest area of Hokkaido, where theair temperature cools down to –40?C or below nearly everyyear, such an intraorgan freezing seems seldom to occur, especiallyin natural stands. On the other hand, low temperatures below–25?C seldom occur in warm-temperate climates. Thus, itmay be considered that in both boreal and temperate coniferstheir shoot and flower primordia seem to tolerate freeze dehydrationby extraorgan freezing under natural conditions. 1 Contribution No. 2431 from the rnstitute of Low TemperatureScience. Babesia rodhaini parasites in murine blood containing 1.5 m DMSO were frozen at two rates, as judged by the duration of the “freezing plateau”, then cooled to ?196 °C and rewarmed at two rates to detect interactions between the duration of the plateau and rates of subsequent cooling and rewarming. Infectivity tests showed that fast and slow freezing (plateau times of about 1 sec and 30 sec, respectively) had similar effects on parasite survival when cooling was at 130 °C/min and warming was at 800 °C/min. However, when either the cooling rate was increased to 3500 °C/min or the warming rate was decreased to 2.3 °C/min, fast freezing decreased parasite survival more than did slow freezing. It is suggested that fast freezing accentuated the damaging effects of fast cooling and slow warming by increasing intracellular ice formation.  相似文献

Tomato seeds of 49 per cent moisture content have been thawedfrom liquid nitrogen (—196 °C) with no loss of germinationfollowing both slow (12 °C min–1) and rapid (850 °Cmin–1) cooling, using 35 per cent dimethyl sulphoxide(DMSO) as a cryoprotectant. Lower concentrations of DMSO resultedin reduced germination. High proportions of seedlings from frozen/thawedseed failed to survive transplanting into soil, especially ifthey had been cooled rapidly. Seedlings grown to maturity afterthe slow cooling treatment showed a greater proportion of morphologicalabnormalities than a control population or those that had beenrapidly cooled. It is suggested that if cryopreservative techniques are to beevaluated for the storage of recalcitrant seeds, false impressionsof efficacy might be gained by reliance solely on germinationtesting. The changes in morphology of the unicellular algae Cylindrocystisbrebissonii and two species of Micrasterias during freezingand thawing were observed on a light microscope fitted witha temperature controlled stage. At slow rates of cooling extensiveshrinkage of the protoplast was observed. The response of thecell wall varied with cell-type. In C. brebissonii plasmolysiswas not observed and the cell wall and protoplast shrank together.In Micrasterias the cell wall did not contract and a distinctplasmolysis was observed. Following freezing to and thawingfrom –25?C cells of C. brebissonii were non-viable butremained osmotically responsive. Cooling at faster rates inducedintracellular ice formation in all cell-types. The criticalrate of cooling varied with cell-type and was determined bycell volume and suface area. Intracellular gas bubbles wereobserved during thawing following both rapid and slow cooling. Following cooling in dimethylsulphoxide cells of C. brebissoniiwere protected against freezing injury. The recovery on thawingfrom –196?C being determined by the rate of cooling, anoptimum rate of 1?C min–1 was observed. During slow ratesof cooling (<2?C min–1) cells remained unshrunken,at faster rates (10?C min–1) the loss of cell viabilitywas related to osmotic shrinkage during cooling rather thanto nucleation of intracellular ice. Spring wheat (Triticum aestivum cv. Warimba) plants were grownin a controlled environment (20°C) in two photoperiods (8or 16 h). In the first instance, plants were maintained in eachof the photoperiods from germination onwards at the same irradiance(375 µE m–2 s–1). In the second case, allplants were grown in a long photoperiod until 4 days after double-ridgeinitiation when half the plants were transferred to a shortphotoperiod with double the irradiance (16 h photoperiod at225 or 8 h at 475 µE –2 s–1). The rates of growth and development of the apices were promotedby the longer photoperiod in both experiments. Shoot dry weightgain was proportional to the total light energy received perday whereas the dry weight of the shoot apex increased withincreasing photoperiod even when the total daily irradiancewas constant. The principal soluble carbohydrate present in the shoot apexwas sucrose, although low concentrations of glucose and fructosewere found in the apices of long photoperiod plants late indevelopment. Sucrose concentration was invariably greater inthe slow-growing apices of short photoperiod plants, but roseto approach this level in the long photoperiod plants when theterminal spikelet had been initiated. Single-cell suspensions of splenic lymphocytes from 5- to 6-month-old C57BL/6 mice were cryopreserved using cooling rates ranging from ?0.25 to ?10.0 °C/min with the microprocessor-controlled cooling system developed in our laboratory. The cells (30 × 106 cells/ml) were suspended in RPMI 1640 containing 10% FCS and 10% DMSO, and a total volume of 1.75 ml was frozen. Fluorescein-diacetate staining identified viable cells in unfrozen controls and frozen-thawed suspensions. Functional capacity was assessed in vitro by the incorporation of [3H]thymidine by dividing cells activated with graded concentrations of the T-lymphocyte mitogens, PHA-P and Con A, and the B-lymphocyte mitogen, LPS. High percentages of viable cells were recovered after cooling at rates ranging from ?0.5 to ?5.0 °C/min, as compared with those of unfrozen control suspensions. Incorporation of [3H]thymidine by T and B cells reached similar levels after cooling at rates ranging from ?0.25 to ?5.0 °C/min, and the percentage incorporation of [3H]thymidine as compared with that of unfrozen cells was 80–100%, except for T lymphocytes activated with PHA-P after cooling at ?5.0 °C/min. The relative response of cell suspensions to T- and B-cell mitogens, the relative mitogenic index, was unchanged from that of unfrozen controls in suspensions cooled at all rates including two (?0.25 and ?10.0 °C/min), which permitted recovery of only 55% of unfrozen cells. The importance of the constant cooling rates and rapid compensation for heat released at the phase change using the microprocessor-controlled system and of precise determinations of cellular viability and function are discussed and related to the apparent protection conferred on subpopulations of murine lymphocytes using cooling rates ranging from ? 0.25 to ?10.0 °C/min.  相似文献

The rate of carbon import by tomato fruits has been relatedto their carbon metabolism by examining the effects of fruittemperature on the metabolism of imported assimilates. 14C–sucrose,–glucose, –fructose, –malic acid and –citricacid were injected individually into young growing tomato fruitswhich were subsequently maintained at 25 or 5 °C for 48h. Fruit temperature greatly affected the proportions of 14Clost from the fruits by export and respiration. Only 40 percent of the injected 14C from 14C–sugars and 20 per centfrom 14C–acids was recovered from fruits at 25 °C.Less than 10 per cent of the injected 14C was exported, thebalance being respired. In contrast, more than 50 per cent ofthe injected 14C was recovered from cooled fruits, in whichthe import rate of carbon was presumably reduced, and 20–36per cent of injected 14C was exported. Cooling enhanced thesynthesis of 14C–sucrose from injected 14C–hexosesand inhibited the incorporation of 14C into starch and insolubleresidue. When 14C–sugars were injected, radioactivityexported from the cooled fruits was detected as sucrose in thephloem of the peduncles; radioactivity was also detected instems and roots when fruits were cooled. In almost fully–grownfruits injected 14C–compounds were metabolized less readilythan in smaller fruits. Conversion of 14C–hexoses to 14C–sucrosewas again enhanced by cooling (5 °C, but was less in fruitsmaintained at 35 °C than in controls. The factors that affect the survival of mouse lymphocytes throughout a procedure for storage at ?196 °C have been studied both for the improvement of recovery and the possible extension to the mouse system of cell selection by freezing. After thawing, the survival of cells cooled at different rates in dimethyl sulphoxide (DMSO, 5 or 10%, vv) was assessed from the [3H]thymidine incorporation in response to phytohaemagglutinin and concanavalin A. Before freezing the protection against freezing damage increased with time (up to 20 min) in DMSO (5%, vv) at 0 °C. Superimposed upon this effect was toxicity due to the DMSO. During freezing and thawing the cooling rate giving optimal survival was 8 to 15 °C/min for cells in DMSO (5%) and 1 to 3 °C/min for DMSO (10%). Omission of foetal calf serum was detrimental. Rapid thawing (>2.5 °C/min) was superior to slow thawing. After thawing dilution at 25 or 37 °C greatly improved cell survival compared with 0 °C; at 25 °C survival was optimal (75%) at a moderate dilution rate of 2.5 min for a 10-fold dilution in FCS (10%, vv) followed by gentle centrifugation (50g).Dilution damage during both thawing and post-thaw dilution may be due to osmotic swelling as DMSO and normally excluded solutes leave the cell. The susceptibility of the cell membrane to dilution damage may also be increased during freezing. The need to thaw rapidly and dilute at 25 °C after thawing is probably due to a decrease in dilution stress at higher temperatures. Optimisation of dilution procedures both maximised recovery and also widened the range of cooling rates over which the cells were recovered. These conditions increase the possibility of obtaining good recovery of a mixed cell population using a single cooling procedure. Alternatively, if cell types have different optimal cooling rates, stressful dilution may allow their selection from mixed cell populations.  相似文献

Rates of absorption were relativelystable at 3?C and 5?C compared with those at 17?C and 25?C whichincreased sharply after 10 d. Tissue concentration of N in theshoot, expressed on a fresh weight basis, was independent ofroot temperature throughout, but doubled between 3–25?Cwhen expressed on a dry weight basis. The present study employed cryomicroscopy to derive an optimal sperm freezing protocol for guppy (Poecilia reticulata) sperm. Evaluation criteria during the freezing-thawing process were assessed for nucleation temperature (Tn), temperature when more than 50% of sperm display bending mid-piece (Tb), temperature when more than 80% of sperm stop moving (Tm), thawing temperature (Tt), and post-thaw motility. We compared four different cryoprotectants: 5% N-dimethyl formamide (DMF), 6% methanol (MEOH), 10% dimethyl sulfoxide (DMSO), and 14% glycerol, as well as glycerol at different concentrations of 7-50%; cooling and rewarming rates ranged from 5 to 100 °C/min. The protocol that yielded the highest post-thaw motility was samples suspended in 14% glycerol, cooled at 25 °C/min, and thawed at 100 °C/min, which was in complete agreement with our previous findings derived from a controlled-rate freezer. In addition, Tb and Tm were found to be negatively correlated with post-thaw motility, suggesting their possible role in predicting freezing success. The present study for the first time demonstrated the usefulness of cryomicroscopy in deriving an optimal sperm freezing protocol for aquatic species.  相似文献

Barley plants (Hordewn vulgare L. cv. Atem) were grown fromseed for 28 d in flowing solution culture, during which timeroot temperature was lowered decrementally to 5?C. Plants werethen subjected to root temperatures of 3, 5, 7, 9, 11, 13, 17or 25 ?C, with common air temperature of 25/15 ?C (day/night).Changes in growth, plant total N, and NO3 levels, andnet uptake of NH4+ and NO3 from a maintained concentrationof 10 mmol m–3 NH4NO3 were measured over 14 d. Dry matterproduction increased 6-fold with increasing root temperaturebetween 3–25 ?C. The growth response was biphasic followingan increase in root temperature. Phase I, lasting about 5 d,was characterized by high root specific growth rates relativeto those of the shoot, particularly on a fresh weight basis.During Phase I the shoot dry weight specific growth rates wereinversely related to root temperature between 3–13 ?C.Phase 2, from 5–14 d, was characterized by the approachtowards, and/or attainment of, balanced exponential growth betweenshoots and roots. Concentrations of total N in plant dry matterincreased with root temperature between 3–25 ?C, moreso in the shoots than roots and most acutely in the youngestfully expanded leaf (2?l–6?9% N). When N contents wereexpressed on a tissue fresh weight basis the variation withtemperature lessened and the highest concentration in the shootwas at 11 ?C. Uptake of N increased with root temperature, andat all temperatures uptake of NH4+, exceeded that of NO3,irrespective of time. The proportions of total N uptake over14 d absorbed in the form of NH4+ were (%): 86, 91, 75, 77,76, 73, 77, and 80, respectively, at 3, 5, 7, 9, Il, 13, 17,and 25 ?C. At all temperatures the preference for NH4+ overNO3 uptake increased with time. An inverse relationshipbetween root temperature (3–11 ?C) and the uptake of NH4+as a proportion of total N uptake was apparent during PhaseI. The possible mechanisms by which root temperature limitsgrowth and influences N uptake are discussed. The percentages of Babesia rodhaini parasites that survived different rates of cooling to −79 °C were determined by titrating infectivity in CBA mice before freezing and after thawing. The cryoprotective effect of DMSO and the effect of warming rate were also assessed.When parasitized blood containing 1.5 DMSO was cooled at nominal rates of 2.5 °, 265 °, and 2785 °C/min and warmed at 4320 °C/min, the respective survival rates were 0.075, 4.9, and 0.1%, indicating the existence of an optimal cooling rate. Blood without DMSO cooled and warmed under the same conditions was over 1000 times less infective. When parasitized blood containing DMSO was cooled at 2785 °C/min and warmed at 4320 °, 24.5 °, and 1.84 °C/ min, infectivity decreased progressively with the warming rate. The degrees of haemolysis in frozen and thawed blood indicated that cooling rate was more important than an intact host cell to survival of the parasite.The growth rate of B. rodhaini in CBA mice, estimated to be one binary fission in 8.5 hr, was not affected by the addition of DMSO followed by freezing and thawing.  相似文献

The air temperature was 20/15?C day/nightand nitrogen was supplied automatically to maintain 10 mmolm–3 NH4NO3 in solution. Total dry matter production wasexponential with time and similar at all root temperatures givinga specific growth rate of 0?0784 g g–1 d–1. Partitioningof dry matter was influenced by root temperature; shoot: rootratios increased during treatment at 17?C and 25?C but decreasedafter 5 d at 3?C and 5?C. The ratio of shoot specific growthrate: root specific growth rate increased with the ratio ofwater soluble carbohydrates (shoot: root). Concentrations ofwater soluble carbohydrates in shoot and root were inverselyrelated to root temperature; at 3, 5 and 7?C they increasedin stem + petioles throughout treatment, coinciding with a decreasein the weight of tissue water per unit dry matter. These resultssuggest that the accumulation of soluble carbohydrates at lowtemperature is the result of metabolic imbalance and of osmoticadjustment to water stress. Low temperature tolerance was investigated in the imbibed seedof 15 seed lots compnsmg seven cultivars of Lactuca sativa L.During rapid cooling (20 °C h–1) some seeds of allseed lots survived to –16 °C but none to –20°C. The majority of seed lots retained over 50 per centviability above –14 °C due to isolation of the embryofrom external ice by the endosperm, and subsequent embryo super-cooling.Certain seed lots, including all three seed lots of cv. TomThumb, showed high mortality at temperatures above –10°C. Correlation of mortality with the formation of externalice suggested that the endosperm is not an effective nucleationbarrier in these seed lots. Survival to –20 °C was increased at slower coolingrates (6 to 1 °C h–1) due to freeze desiccation ofthe embryo, but seed lots varied considerably in their toleranceof specific cooling rates. A model to explain this variationwas developed incorporatmg (1) seed lot super-cooling limittemperature, (2) the rate at which freeze dehydration of thesupercooled embryo took place, (3) the moisture content at whichnucleation (at –20 °C) was no longer certain and (4)the.initial equilibrium moisture content of the fully imbibedseed. Factors (1), (2) and (3) were found to be relatively constant,but low (or artificially reduced) seed moisture content wasclosely correlated with high survival at natural cooling rates.Seed size fractions of similar moisture content from a singlecultivar showed that more small seeds survive cooling at 3 °Ch–1 to –20 °C than larger seed. Seed with pierced endosperms or ineffective nucleation barrierswere capable of surviving to at least –10 °C if cooledslowly (1 °C h–1) but were killed by rapid (20 °Ch–1) cooling. The effect of shoot demand for nutrients on nutrient uptakeand translocation in the xylem exudate was studied in maizegrowing in nutrient solution at uniform shoot zone (24/2C,day/night), but different root zone temperatures (RZT: 12C,18C, 24C). The shoot base (apical shoot meristem and zoneof leaf extension) was either kept within or lifted above thecooling zone. In plants with their shoot base above the coolingzone (RZT: 12C and 18C) shoot growth was significantly increasedbut not root growth. Therefore, at suboptimal RZT shoot freshweight increment d–1 g–1 root fresh weight, whichwas taken as a parameter for the shoot demand for nutrientsper unit root, varied strongly depending on the temperatureof the shoot base (shoot base temperature, SBT). In short-term studies (2 h and 1 d after onset of temperaturetreatment) rates of nutrient (nitrogen, N; potassium, K; phosphorus,P; calcium, Ca) uptake or translocation in the xylem exudatewere markedly decreased at suboptimal RZT (12C, 18C), irrespectiveof the SBT. In long-term studies (3, 5, and 10 d after onsetof temperature treatment) uptake and translocation of K, N,and Ca, but not P, increased in plants at suboptimal RZT whenthe shoot demand was high (shoot base above the cooling zone)but decreased when the shoot demand was low (shoot base withinthe cooling zone). These results suggest, that the increase of translocation ratesof N, K, and Ca after long-term exposure to suboptimal RZT wasa consequence of a higher shoot demand per unit root fresh weightand not due to a direct temperature effect on the nutrient uptakesystem. Concentrations of inorganic cations are often lower in plantssupplied with NH4+ as compared with NO3. To examine whetherthis is attributable to impaired root uptake of cations or lowerinternal demand, the rates of uptake and translocation of K,Mg, and Ca were compared in maize plants (Zea mays L.) withdifferent growth-related nutrient demands. Plants were grownin nutrient solution with either 1·0 mol m–3 NO3or NH4+ and the shoot growth rate per unit weight of roots wasmodified by varying the temperature of the shoot base (SBT)including the apical shoot meristem. The shoot growth rate per unit weight of roots, which was takenas the parameter for the nutrient demand imposed on the rootsystem, was markedly lower at 12°C than at 24°C SBT.As a consequence of the lower nutrient demand at 12°C SBT,uptake rates of NO3 and NH4+ declined by more than 50%Compared with NO3 supply, NH4+ nutrition depressed theconcentrations of K and particularly of Ca in the shoot, bothin plants with high and with low nutrient demand. This indicatesa control of cation concentration by internal demand ratherthan by uptake capacity of the roots. Translocation rates of K, Mg and Ca in the xylem exudate werelower in NH4+- than in NO3-fed plants. Net accumulationrates of Ca in the shoot were also decreased, whereas net accumulationrates of K in the shoot were even higher in NH4+-fed plants.It is concluded that reduced cation concentrations in the xylemsap of plants supplied with NH4+ are due to the lower demandof cations for charge balance. The lower K translocation tothe shoot is compensated by reduced retranslocation to the roots.For Ca, in contrast, decreased translocation rates in NH4+-fedplants result in lower shoot concentration. Wheat plants were grown in a controlled environment with daytemperatures of 18 ?C and with 500 µ Einsteins m–28–1 of photosynthetically active radiation for 16 h. Beforeanthesis and 2 to 3 weeks after, rates of net photosynthesiswere measured for leaves in 2 or 21% O2 containing 350 vpm CO2at 13, 18, 23, and 28 ?C and with 500 µEinsteins m–2s–1 of photosynthetically active radiation. Also, underthe same conditions of light intensity and temperature, therates of efflux of CO2 into CO2-free air were measured and,for mature flag leaves 3 to 4 weeks after anthesis, gross andnet photosynthesis from air containing 320 vpm 14CO2 of specificactivity 39?7 nCi µmol–1. When the O2 concentration was decreased from 21 to 2% (v/v)the rate of net photosynthesis increased by 32 per cent at thelowest temperature and 54 per cent at the highest temperature.Efflux of CO2 into CO2-free air ranged from 38 per cent of netphotosynthesis at 13 ?C to 86 per cent at 28 ?C. Gross photosynthesis,measured by the 14C assimilated during 40 s, was greater thannet photosynthesis by some 10 per cent at 13 ?C and 17 per centat 28 ?C. Survival of the parasitic protozoan, Babesia bigemina, in blood cooled at widely different rates to ? 196°C. International Journal for Parasitology4: 169–172. The infectivity of Babesia bigemina in blood containing 2 m DMSO was tested in 99 cattle after the blood had been cooled to ? 196°C at eight rates ranging from 0·73–3070°C/min. Blood cooled at each rate was infective; 95 of the recipients became infected, the exceptions being four of the seven cattle inoculated with blood cooled at 3070° C/min. The infectivity of blood cooled at 39, 82 and 212°C/min was higher than that of blood cooled at slower or faster rates. Least depression of infectivity occurred at 82°C/min.  相似文献

Single, clonal plants of white clover were grown without inorganicnitrogen in four contrasting day/night temperature regimes,with a 12 h photoperiod, in controlled environments. Root andnodule respiration and acetylene reduction activity were measuredin a flow-through system during both day and night for plantsacclimated to day/night regimes of 23/18, 15/10 and 10/5 ?C.Similar measurements were made on plants acclimated to 20/15?C and stepwise at temperatures from 4 to 33 ?C. Peak rate of ethylene production, nitrogenase-linked respirationand basal root + nodule respiration increased approximatelylinearly from 5 to 23 ?C both in temperature-acclimated plantsand in plants exposed to varying measurement temperatures. Themeasured attributes did not vary significantly between day andnight. Temperatures above 23–25 ?C did not further enhancethe rate of ethylene production, which remained essentiallythe same up to the maximum measured temperature of 33 ?C. The measurements of nitrogenase-linked respiration between 5and 23 ?C, during both day and night, demonstrated a constant‘energetic cost’ of acetylene reduction of 2.9 µmolCO2 µmol C2H4–1,. Over the same temperature range,the approximate activation energy of acetylene reduction was60 kJ mol–1. The integrated day plus night nitrogenase-linkedrespiration accounted for 13.4–16% of the plant‘snet shoot photosynthesis in a single diurnal period: there wasno significant effect of temperature between 5 and 23 ?C.
