Behavioral and adrenocortical reactivity to stressful stimulation was examined in 12- and 13-day-old chicks of two lines of Japanese quail selected over several generations for exaggerated (HS: high stress) or reduced (LS: low stress) plasma corticosterone (B) response to brief immobilization stress. Plasma B concentrations and tonic immobility (TI) fear reactions were measured in unstressed (control) and stressed (overnight cooping) chicks of both lines. The stress treatment was applied over a period of 12-20 hr and it involved capture by the experimenter, inescapable exposure to an unfamiliar environment and to strange conspecifics, reductions in ambient temperature and floor space, and the deprivation of food and water. Chicks of the HS line were more susceptible to the induction of TI and they remained immobile longer than did LS chicks. Therefore HS chicks were considered to be more fearful than their LS counterparts. Stress treatment elicited a marked adrenocortical response that was more pronounced in HS than in LS chicks. Stress treatment also increased susceptibility to TI but did not significantly affect the duration of immobility. These findings suggest that selecting the quail for differential corticosterone response to a particular stressor had exerted an unconscious and concomitant effect on underlying fearfulness as well as on their adrenocortical reactivity to several other stressful situations. The results are further discussed in terms of a putative relationship between adrenocortical activation and fearfulness.  相似文献

The right and left femora of three groups of rodents, cold stressed, noise stressed, and heat stressed, were sectioned transversely at the most inferior point of the third trochanter. Cortical bone thickness of the proximal section for all three groups was determined by measuring an enlargement obtained with a Wild stereo microscope with a camera lucida drawing attachment. Cold stressed animals were found to have significantly thinner cortical bone than did controls, noise stressed animals did not differ from controls, and heat stressed animals had thinner cortical bone than did controls. It is concluded that stress may be responsible for the thinner cortex of cold and heat stressed animals, but that other factors may be at work and that it is not possible to tell whether different types of stress act through the same or through different mechanisms.  相似文献

Animals perceiving repeated aversive events can become chronically stressed. Chronic activation of the hypothalamic–pituitary–adrenal (HPA) axis can have deleterious consequences on physiological parameters (e.g. BW, blood chemistry) and behaviour (e.g. emotional reactivity, stereotypies, cognition). Environmental enrichment (EE) can be a mean to reduce animal stress and to improve welfare. The aim of this study was first, to assess the effects of EE in battery cages on the behaviour of young Japanese quail and second, to evaluate the impact of EE on quail exposed to chronic stress. The experiment involved quail housed in EE cages and submitted or not to a chronic stress procedure (CSP) (EE cages, control quail: n=16, CSP quail: n=14) and quail housed in standard cages and exposed or not to the CSP (standard non-EE cages, control quail: n=12, CSP quail: n=16). Our procedure consisted of repeated aversive events (e.g. ventilators, delaying access to food, physical restraint, noise) presented two to five times per 24 h, randomly, for 15 days. During CSP, EE improved quail’s welfare as their stereotypic pacing decreased and they rested more. CSP decreased exploration in all quail. After the end of CSP, quail presented increased emotional reactivity in emergence test. However, the effect of EE varied with test. Finally, chronic stress effects on comfort behaviours in the emergence test were alleviated by EE. These results indicate that EE can alleviate some aspects of behavioural alterations induced by CSP.  相似文献

Experimental liver injury with different stages was induced to adult female test rats with daily injection of thioacetamide (ThAA). The doses administered intraperitoneally were 50 mg/kg body weight. In the liver sections progressive changes of damage, regeneration and fat substitution were noticed. Kidney sections revealed enhanced glomerular atrophy, particularly in the cortical tubules, provoked in the 3-week traumatization period. The influence of ThAA on female rat blood was assayed using standard biochemical methods. The analyses done were: the percentage of blood obtainable and the serum/blood ratio; the serum alanine transferase; serum alkaline phosphatase; serum creatinine; serum hydroxyproline and serum beta-glucuronidase activity in the acute, subacute, chronic and highly chronic stage of liver injury. The biochemical findings show continuously progressing damages when traumatization proceeds. In the 3-week test period the histological findings processed showed an increase in osteoclastic resorption in the alveolar bone around the occlusally stressed tooth simultaneously with a horizontal bone loss. Some indications of recovering incidents were seen, too. Only in the histological findings was no difference seen in the deterioration between both sexes, contrarily to the biochemical results also discussed in this study.  相似文献

Two sets of irrigated and water-stressed swards of perennial ryegrass (Lolium perenne) growing in the field were compared. One set of swards was grown normally (field swards) and the other was established in boxes of low water-holding capacity (simulated swards). Rain covers were used to exclude rain from half of both sets of swards: the others were kept within 20 mm of field capacity. Daily minimum values of leaf water potential fell to —12 bars (—1.2 MPa) in the irrigated swards, —16 bars in the stressed field swards and —20 bars in the stressed simulated swards. Dry-matter production was reduced in the stressed field swards and leaf extension declined markedly in both the field and simulated swards. Canopy photosynthesis at saturating light intensity was reduced by about half in the stressed field swards and by more than 80% in the stressed simulated swards. In the former case a proportion of this reduction was due to the lower leaf area but the mean rate of leaf photosynthesis at saturating light intensity (Pmax/LAI) was reduced by about 40% and this was attributable to increased stomatal resistance calculated from canopy transpiration rates or measured with a porometer. The more marked decline in photosynthesis in the stressed simulated swards was not only the consequence of more complete stomatal closure but also a decrease in the quantum efficiency of photosynthesis. When water stress occurs naturally in the field it appears that changes in the leaves take place as stress is developing and these changes result in a less severe effect of water stress on leaf photosynthesis. When stress is imposed with unnatural rapidity, as in the simulated swards, there is no opportunity for these changes in the leaf to occur and stress leads to a rapid and severe decline in leaf photosynthesis. The importance of these findings for the grass sward under water stress is discussed.  相似文献

In 8 patients with idiopathic myelofibrosis (IM) T and B cells were studied in view of the possibility that immunological dysfunctions are involved in initiating or contributing to the bone marrow fibrosis. In peripheral blood the absolute numbers of E-SRBC and OKT3+ lymphocytes were significantly reduced; in addition a significant decline was observed in the proportion and absolute numbers of OKT8+ cells, resulting in a reversed Leu-3a/OKT8 ratio. An impaired B cell function was observed in 4 of the 8 patients, characterized by a disturbed in vitro pokeweed mitogen stimulated immunoglobulin synthesis and low serum immunoglobulin levels. Immuno-histological studies of the bone marrow demonstrated a scarcity of T cells but normal numbers of B cells. However, no correlation was noted between the observed deviations of B and T cells and the degree of bone marrow fibrosis determined by means of bone marrow histology and serum procollagen-III levels. These data are not sufficient to support the hypothesis that immunological changes in IM are primarily involved in the process of bone marrow fibrosis.  相似文献

The aims of this study were to examine the effect of implant neck design and cortical bone thickness using 3D finite element analysis and to analyse the stability of clinical evidence based on micromotion and principal stress. Four commercial dental implants for a type IV bone and maxillary segments were created. Various parameters were considered, including the osseointegration condition, loading direction and cortical bone thickness. Micromotion and principal stresses were used to evaluate the failure of osseointegration and bone overloading, respectively. It was found that the maximum stress of the peri-implant bone decreased as cortical bone thickness increased. The micromotion level in full osseointegration is less than that in non-osseointegration and it also decreases as cortical bone thickness increases. The cortical bone thickness should be measured before surgery to help select a proper implant. In the early stage of implantation, the horizontal loading component induces stress concentration in bone around the implant neck more easily than does the vertical loading component, and this may result in crestal bone loss.  相似文献

Free radicals have been shown to be involved in bone resorption in vitro and in rodents. We studied the effect of oxidative stress on bone mineral density (BMD) in 48 women and 53 men from a population-based study. The levels of 8-iso-PGF(2alpha) (a major F(2)-isoprostane and a biomarker of oxidative stress) and a control, 15-keto-dihydro-PGF(2alpha) (a biomarker of inflammatory response), were measured in urinary samples and their association with BMD and quantitative ultrasound (QUS) measurements were examined. In multivariate linear regression analyses, 8-iso-PGF(2alpha) levels were negatively associated with both BMD and QUS. In contrast, no association was found for 15-keto-dihydro-PGF(2alpha). Our findings establish a biochemical link between increased oxidative stress and reduced bone density and provide a rational for further studies investigating the role of pro- and antioxidants in osteoporosis.  相似文献

Residual stress and strain in living tissues have been investigated from the viewpoint of mechanical optimality maintained by adaptive remodeling. In this study, the residual stresses in the cortical-cancellous bone complex of bovine coccygeal vertebrae were examined. Biaxial strain gages were bonded onto the cortical surface, so that the gage axes were aligned in the cephalocaudal and circumferential directions. Strains induced by removal of the end-plate and the cancellous bone were recorded sequentially. The results revealed the existence of compressive residual stress in the cortical bone and tensile residual stress in the cancellous bone in both the cephalocaudal and the circumferential direction. The observed strains were examined on the basis of the uniform stress hypothesis using a three-bar model for the cephalocaudal direction and a three-layered cylinder model for the circumferential direction. In this model study, the magnitude of effective stresses, which is defined as the macroscopic stress divided by the area fraction of bone material, was found not to differ significantly between cephalocaudal and circumferential directions, although they were evaluated using independent models. These results demonstrate that the uniform stress state of the cortical-cancellous bone structure is consistent with results obtained in the cutting experiment when the existence of residual stress is taken into account.  相似文献

The purpose of this study was to assess the inflammatory nature of obesity and its effect on blood and bone marrow endothelial cell populations. Obese patients (BMI ≥30) had significantly higher concentrations of the inflammatory marker C-reactive protein (CRP) (P = 0.03) and lower concentrations of the anti-inflammatory cytokine interleukin-10 (IL-10) (P = 0.05). This cytokine profile is consistent with obesity being an inflammatory condition and is further supported by the significant correlation between total white blood cell count and BMI (r = 0.15; P = 0.035). High BMI was associated with significantly lower numbers of early endothelial cells (CD45(-)/CD34(+)) in the bone marrow (r = -0.20; P = 0.0068). There was also a significant inverse correlation between BMI and a more mature endothelial cell phenotype (CD45(-)/31(+)) in the blood (r = -0.17; P = 0.02). In addition, there was a significant correlation between BMI- and endothelial-related cells of hematopoietic origin (CD133(+)/VEGFR-2(+)) in the bone marrow (r = -0.26; P = 0.0007). Patients with higher plasma IL-10 and insulin-like growth factor (IGF) concentrations had higher numbers of endothelial phenotypes in the bone marrow suggesting a protective effect of these anti-inflammatory cytokines. In conclusion, this work confirms the inflammatory nature of obesity and is the first to report that obesity is associated with reduced endothelial cell numbers in the bone marrow of humans. These effects of obesity may be a potential mechanism for impaired tissue repair in obese patients.  相似文献

Because insulin resistance is inevitably associated with cardiovascular complications, there is a need to further investigate the potential involvement of oxidative stress and the cyclo-oxygenase (COX) pathway in the vascular modifications associated to this pathological context. Endothelial function was evaluated in control and fructose-fed rats (FFR) by i) in vitro study of endothelium-dependent and -independent relaxations of aortic rings, and ii) in vivo telemetric evaluation of pressor response to norepinephrine. After 9 weeks of diet, FFR displayed hypertriglyceridemia, hyperinsulinemia and exaggerated response to glucose overload. Aortic rings from control rats and FFR exhibited comparable endothelium-dependent relaxations to Ach. In the presence of indomethacin, relaxations were significantly reduced. FFR showed exaggerated pressor responses to norepinephrine that were abolished with indomethacin. Urinary nitrites/nitrates, 8-isoprostanes and thromboxane B2 excretion levels were markedly enhanced in FFR, whereas the plasma levels of 6-keto prostaglandin F1alpha were unchanged. In conclusion, fructose overload in rats induced hypertriglyceridemia and insulin resistance associated with an enhanced oxidative stress. This was associated with COX pathway dysregulation which could be one of the contributors to subsequent vascular dysfunction. Consequently, reduction of oxidative stress and regulation of the COX pathway could represent new potential therapeutic strategies to limit vascular dysfunction and subsequent cardiovascular complications associated with insulin resistance.  相似文献

Materials, including bone, often fail due to loading in the presence of critical flaws. The relative amount, location, and interaction of these flaws within a stressed volume of material play a role in determining the failure properties of the structure. As materials are generally imperfect, larger volumes of material have higher probabilities of containing a flaw of critical size than do smaller volumes. Thus, larger volumes tend to fail at fewer cycles compared with smaller volumes when fatigue loaded to similar stress levels. A material is said to exhibit a volume effect if its failure properties are dependent on the specimen volume. Volume effects are well documented in brittle ceramics and composites and have been proposed for bone. We hypothesized that (1) smaller volumes of cortical bone have longer fatigue lives than similarly loaded larger volumes and (2) that compared with microstructural features, specimen volume was able to explain comparable amounts of variability in fatigue life. In this investigation, waisted rectangular specimens (n=18) with nominal cross-sections of 3×4 mm and gage lengths of 10.5, 21, or 42 mm, were isolated from the mid-diaphysis of the dorsal region of equine third metacarpal bones. These specimens were subjected to uniaxial load controlled fatigue tests, with an initial strain range of 4000 microstrain. The group having the smallest volume exhibited a trend of greater log fatigue life than the larger volume groups. Each volume group exhibited a significant positive correlation between the logarithm of fatigue life and the cumulative failure probability, indicating that the data follow the two-parameter Weibull distribution. Additionally, log fatigue life was negatively correlated with log volume, supporting the hypothesis that smaller stressed volumes of cortical bone possess longer fatigue lives than similarly tested larger stressed volumes.  相似文献

In 1993 we proposed an empirical formula for describing the relaxation modulus of cortical bone based on the results of stress relaxation experiments performed for 1 x 10(5) sec: [E(t) = E0{A exp[ -(t/tau1)beta] + (1 - A) exp(-t/tau2)}, (0 < A, beta <1 and tau1 < tau2) where E0 is the initial value of the relaxation modulus, A is the portion of the first term, tau1 and tau2 are characteristic relaxation times, and beta is a shape factor [Sasaki et al., J. Biomechanics 26 (1993), 1369]. Although the relaxation properties of bone under various external conditions were described well by the above equation, recent experimental results have indicated some limitations in its application. In order to construct an empirical formula for the relaxation modulus of cortical bone that has a high degree of completeness, stress relaxation experiments were performed for 6 x 10(5) seconds. The second term in the equation was determined as an apparently linear portion in a log E(t) vs t plot at t>1 x 10(4) sec. The same plot for experiments performed for 6 x 10(5) seconds revealed that the linear portion corresponding to the second term was in fact a curve with a large radius of curvature. Learning and memory deficits occur in depression and other stress related disorders. Although the pathogenesis of cognitive impairment after stress has not been fully elucidated, factors such as oxidative stress and neurotrophins are thought to play possible roles. Here we investigated the effect of treatment with vitamin E (40 mg/kg) and vitamin C (100 mg/kg) on the effects elicited by chronic variable stress on rat performance in Morris water maze. Brain-derived neurotrophic factor (BDNF) immunocontent was also evaluated in hippocampus of rats. Sixty-day old Wistar rats were submitted to different stressors for 40 days (stressed group). Half of stressed group received administration of vitamins once a day, during the period of stress. Chronically stressed rats presented a marked decrease in reference memory in the water maze task as well as a reduced efficiency to find the platform in the working memory task. Rats treated with vitamins E and C had part of the above effects prevented, suggesting the participation of oxidative stress in such effects. The BDNF levels were not altered in hippocampus of stressed group when compared to controls. Our findings lend support to a novel therapeutic strategy, associated with these vitamins, to the cognitive dysfunction observed in depression and other stress related diseases.  相似文献

The mechanical properties of bone depend largely on its degree and distribution of mineralization. The present study analyzes the effect of an inhomogeneous distribution of mineralization on the stress and strain distributions in the human mandibular condyle during static clenching. A condyle was scanned with a micro-CT scanner to create a finite element model. For every voxel the degree of mineralization (DMB) was determined from the micro-CT scan. The Young's moduli of the elements were calculated from the DMB using constant, linear, and cubic relations, respectively. Stresses, strains, and displacements in cortical and trabecular bone, as well as the condylar deformation (extension along the antero-posterion axis) and compliance were compared. Over 90% of the bone mineral was located in the cortical bone. The DMB showed large variations in both cortical bone (mean: 884, SD: 111 mg/cm(3)) and trabecular bone (mean: 738, SD: 101 mg/cm(3)). Variations of the stresses and the strains were small in cortical bone, but large in trabecular bone. In the cortical bone an inhomogeneous mineral distribution increased the stresses and the strains. In the trabecular bone, however, it decreased the stresses and increased the strains. Furthermore, the condylar compliance remained relatively constant, but the condylar deformation doubled. It was concluded that neglect of the inhomogeneity of the mineral distribution results in a large underestimation of the stresses and strains of possibly more than 50%. The stiffness of trabecular bone strongly influences the condylar deformation. Vice versa, the condylar deformation largely determines the magnitude of the strains in the trabecular bone.  相似文献

A study was conducted to determine the effect of transportation stress on ovarian function in superovulated heifers. Thirty cyclic Hereford heifers of similar age and weight and in good body condition were randomly assigned to control and stress-treated groups. All animals received two daily injections of 5 mg follicle stimulating hormone (FSH) for 4 d beginning on Day 10 to 12 of the estrous cycle. A blood sample was collected at each FSH injection. On the fourth day of injections, heifers were given 25 mg prostaglandin F(2)alpha (PGF(2)alpha) in the morning and a second injection of 15 mg PGF(2)alpha in the afternoon. During superovulation, the stressed heifers were transported to a different location every 12 h whereas control animals remained at the pretrial site. Following 4 d of intermittent transporting and FSH treatment, stress-treated heifers were recombined with control animals. Ovaries were examined 8 d following the onset of standing estrus to determine length, width, thickness, and number of corpora lutea (CL). Peripheral plasma levels of cortisol were higher in the stressed group (P< 0.1). Least squares means for numbers of CL were 20.4 +/- 2.1 and 15.4 +/- 1.7 for control and treated heifers, respectively (P< 0.1). There were no treatment differences (P> 0.1) between length, width, or thickness of ovaries when the number of CL was held constant. These data suggest that stress of the type, intensity, and duration imposed in this study increased plasma levels of cortisol and reduced ovulation rate as determined by CL formation in superovulated heifers.  相似文献

Cortical bone growth is analyzed for 174 children from a Medieval Christian population at Kulubnarti in the Batn el Hajar of Sudanese Nubia (550–1450 AD ). Using the tibia as a representative long bone, total subperiosteal area, cortical area, medullary area, and percent cortical area at midshaft were calculated. While growth in total and cortical areas, as well as in length, appear to be fairly well maintained, percent cortical area reveals unusual growth patterns which reflect excessive endosteal resorption. Compared to the relative reduction in bone mass which has been observed in malnourished living children, as well as with previously reported evidence for stress in the Kulubnarti population, the present data support an interpretation of nutritionally related stress and of no major diachronic dietary change.  相似文献

The salt-induced accumulation of some nitrogen compounds (free amino acids, ammonium and urea) in shoots of eight rice cultivars differing in salt tolerance was investigated. Salt treatment (100 mM, 6 days) significantly increased the proline content of shoots but this appeared to be a reaction to stress damage and not associated with salt tolerance, because proline contents were higher in the more sensitive cultivars. Besides proline, some other free amino acids also accumulated leading to a significant increase in the total amino acid content of the stressed seedlings. High levels of free ammonium also accumulated under conditions of stress; this was highly correlated with the accumulation of Na+ in the shoots and negatively correlated with salt tolerance. The accumulation of ammonium was positively correlated with the accumulation of many free amino acids, and also associated with the production of urea in the stressed seedlings. Results from the present investigations suggest that an increase in the concentration of some free amino acids including proline, may be a result of the reassimilation of the stress-induced ammonium. A high capacity to assimilate ammonium may be an important factor in alleviating the consequence of stress because ammonium can be toxic at high concentrations.  相似文献
