The effects of host age and parasitoid female age on the occurrence of 'Pseudoparasitism', using the Spodoptera littoralis-Microplitis rufiventris host-parasitoid system were investigated. The first four larval instars of the host are not equally suitable for parasitoid development. The proportion of pseudoparasitized hosts significantly increases when: (1) the age of the female parasitoid increases; (2) oviposition occurs mostly in fourth instar larvae; (3) a later age of the host instar is used; (4) the mandibles of the newly hatched parasitoid larvae mistakenly attack host interior organs (e.g. Malpighian tubules); and (5) an imperfect growth pattern of teratocytes occurs. The reluctance of female wasps to parasitize fourth instar host larvae is not due to the thickness of host cuticle but possibly due to the unfavourable physiological state of the host larvae. The age of host larvae at the time of parasitization may influence the adverse effects of parasitoid factors (e.g. polydnavirus, venom and teratocytes) on the growth of host larvae. It is suggested that females of M. rufiventris are able to determine the suitability of a potential host instar for the development of their offspring. The cell diameter of M. rufiventris teratocytes increases with increasing age of host larvae at the time of oviposition. The association within the host of living parasitoid larvae and functional teratocytes may be important for the survival of each other and consequently for successful parasitism.

The influence of sublethal dietary levels (0.4 p.p.m.) of a chitin synthesis inhibitor lefenuron { N -[2,5-dichloro-4-(1,1,2,3,3,3-hexafluoropropoxy)-phenylaminocarbonyl]-2,6-difluorobenzamide} on the development of Microplitis rufiventris teratocytes was investigated. Observations and data were taken on maturing teratocytes, i.e. at the end of parasitoid development. When embryogenesis of the parasitoid eggs was initiated in treated hosts, the dissociated cells of the embryonic membrane of some of the parasitoid larvae did not liberate in the haemolymph of the host, whereas in the other hosts, the released ones as individual cells scored the largest mean number per host larva. When young teratocytes were exposed to the treatment they were found in some cases clumping in masses or clustered around the parasitoid larvae. In treated hosts, different size classes of teratocytes co-existed. Larger and morphological abnormal cells were more common in hosts which contained older teratocytes at the time of treatments. Abnormal teratocytes were associated with developmental abnormalities of parasitoid larvae, possibly due to deterioration of the host environment. This observation was common in treated hosts and in few cases of untreated ones. A significant decrease in teratocyte size occurred 24 h after parasitoid emergence. Observations and results obtained in the present work suggested that: (1) teratocyte may function in controlling the growth and successful development of parasitoid larvae; (2) they might have a protective function through sequestering abnormal materials issuing in the haemolymph of the host either during the course of parasitoid development or due to external stress; and (3) number of teratocyte cells liberated from the embryonic membrane of parasitoid egg is not constant.

Reproductive biology including mating, adult longevity, fecundity and development of the tachinid fly Zenillia dolosa was investigated for optimizing rearing procedures using Mythimna separata as a host in the laboratory. Females lay microtype eggs containing a first instar larva on food plants of the host and then the eggs must be ingested by the host for parasitization. Mating success was 58.5% with mating duration of 80.7 min. Mating was most successful when day 0–1 females were kept with day 2–4 male flies. Female body size was positively correlated with its fecundity but not with longevity. However, females that survived longer produced more eggs during their lifetime. Parasitoids successfully developed in 4th to 6th instar host larvae. Host instars at the time of parasitoid egg ingestion significantly influenced development time of the immature parasitoid, but did not affect body size of the emerging parasitoid. We suggest that pairing newly emerged females with day 2–4 males should result in higher mating success and using the last instar hosts for parasitization should minimize development time of the parasitoid for rearing.

Relative effects of parasitism by Microplitis rufiventris on the development of the third instar Spodoptera littoralis (preferable, optimal host) with the development of penultimate (5th) and last (6th) instars (suboptimal hosts) were investigated. Newly molted 6th instar hosts were more acceptable for parasitization by the wasp female than older hosts. In singly parasitized 3rd instar hosts, 82.0 +/- 3.9% of the parasitoid eggs developed to full-grown instar wasp larvae. However, parasitoid eggs deposited singly in 73.9 +/- 3.3% of 5th and 100% of 6th instar hosts failed to develop. Superparasitization in the 3rd instar hosts reduced the production of pseudoparasitized larvae and, conversely, all parasitized hosts yielded viable parasitoid offspring. In suboptimal hosts, the development interaction between the parasitoid and its host larvae was highly influenced by the age of hosts at parasitism, load of deposited eggs, and other parasitoid factors. The latter factors, e.g., mainly calyx fluid particles, might be involved in establishing parasitoid eggs in the suboptimal hosts. In the last two host instars, superparasitization significantly increased the number of parasitoid larvae successfully reaching their final instar. Variation in host quality, e.g., physiological status, might be attributed, in part, to the partial breakdown of the solitary habit observed in the earlier instars. More parasitoid eggs developed to mature parasitoid larvae in hosts superparasitized as 6th instar than parasitoid eggs laid in 5th instar hosts. Superparasitization significantly lengthened the developmental period of 5th and 6th host instars and inhibited their development to the pupal stage. Studying parasitoid development in suboptimal instars of its habitual host provided physiological insight, as shown here. The results may have implication for biological control and in vitro mass rearing programs with solitary parasitoids.

We compared the production of pseudoparasitization by Microplitis rufiventris females in most (third) and less (fourth) preferred instars of Spodoptera littoralis larvae at 20+/-1 and 27+/-1 degrees C. The parasitized hosts were classified into hosts producing parasitoids (type A hosts) and hosts producing no parasitoids, i.e., pseudoparasitized hosts (type B hosts). The latter were further classified into: (a) pseudoparasitized hosts with "well" arrested development (type B1 hosts); (b) pseudoparasitized hosts with partially arrested development (type B2 hosts); and (c) pseudoparasitized hosts that successfully pupated to apparently normal host pupae (type B3 hosts). The present series of experiments showed that parasitization by M. rufiventris was clearly affected by host instar, age within an instar and rearing temperature. Production of type B hosts was less when third instar S. littoralis larvae were exposed to the wasp females than when the host larvae were in fourth instar. The production of type A hosts was much greater when early or mid ages of an instar was stung by the wasp females comparing with stung late age of the same instar. Production of type B hosts may be due to one or overall of the following: (a) dosage dilution of M. rufiventris female's factors in the different age classes of the instar; (b) endocrine system (physiological state) at parasitization time, i.e., early vs late age of the instar; (c) growth rate of host larvae. The lowest production of type B hosts was at highest growth rate; and (d) temperature, larger proportions of type B hosts were produced at 27+/-1 than at 20+/-1degrees C. The three types host development (B1, B2 and B3) are possibly representing three levels of host resistance (host control) resulting in partial or complete failure of parasitoid control. Type A hosts represent complete success of parasitoid control. The results suggest that the impact of parasitoid factor(s) on developmental arrest is affected by host age at the time of parasitism and/or by temperature.

Abstract. Laboratory studies investigated the development of teratocytes derived from the eggs of the parasitoid Meteous gyrator (Thun.) in its host, the tomato moth Lacanobia oleracea (L.). At hatching, each parasitoid egg produced an average of approximately 1000 teratocytes, but this number declined to approximately 400 during the course of parasitism. The teratocytes increased in size markedly, such that 7 days after egg hatch their mean diameter was approximately four times that of the cells immediately after dissociation. The haemolymph of parasitized hosts had reduced phenoloxidase activity, and teratocytes inhibited phenoloxidase activity when coincubated with plasma from nonparasitized hosts. The injection of teratocytes into nonparasitized fifth‐instar L. oleracea larvae suppressed growth and induced a supernumerary moult in some larvae. A number of parasitism‐specific proteins were detected in the haemolymph of parasitized hosts, and incubation of teratocytes in culture media indicated that these cells were a source of at least two of these proteins.

Abstract:  Earlier research has shown that the koinobiont parasitoid, Microplitis rufiventris , attacks and can develop on early instars of Spodoptera littoralis larvae with preference to third instars. However, the present study was carried out using the newly moulted sixth instar larvae at two different temperatures (20 ± 1 and 27 ± 1°C) to study the developmental interaction between the parasitoid and the last instar host larvae. Parasitoid eggs laid in singly parasitized host larvae invariably died. As the number of parasitoid eggs/host larvae increased, the proportion of eggs that hatched and number of viable parasitoid larvae successfully reached to their final instar increased. The effect of superparasitization seems to be dose (no. of eggs + parasitoid factors)-temperature-dependent. The results demonstrate a kind of 'Allee effect' suggesting that superparasitized last instar S. littoralis larvae provide a better host environment than singly parasitized hosts for the parasitoid, M. rufiventris . This may be due to host's hormone and/or low dose of factors injected with parasitoid eggs. The supernumerary individuals of wasp larvae developed normally to the point of emergence but most did not successfully emerge from the host. The improvement of both hatchability and post-embryonic development of parasitoid wasp was significantly (P < 0.01) greater at 20°C than at 27°C. The results of the present study are useful in understanding the evolution of life-history strategies and host range in parasitic hymenoptera.

The solitary endoparasitoid Meteorus pulchricornis can parasitize many lepidopteran host species successfully. In the case of parasitization of Pseudaletia separata, developmental duration of M. pulchricornis was 8-9 days from egg to larval emergence and 6 days from prepupa to adult emergence. Successful parasitism by M. pulchricornis decreased with host age. Following parasitization of day-0 4th host instar, the parasitoid embryo, whilst still enclosed in serosal cell membrane, hatched out of the egg chorion 2 days after oviposition. Subsequently, the 1st instar parasitoid emerged from the surrounding serosal cell membrane. Serosal cells dissociated and developed as teratocytes 3.5 days after oviposition. One embryo of M. pulchricornis gave rise to approximately 1200 teratocytes, a number that remained constant until 6 days after parasitization, but decreased drastically to 200 at 7 days post-oviposition. The teratocytes of M. pulchricornis were round- or oval-shaped and grew from 65 microm at 4 days to 200 microm in the long axis at 6 days post-parasitization. At 4 days post-parasitization, many cells or cell clusters with lipid particles were observed in the hemocoels of parasitized hosts. In addition, paraffin sections of parasitized hosts revealed that many teratocytes were attached to the host's fat body and contributed to disrupting the fat body tissue. Further, examination of the total hemocyte count (THC) during parasitization revealed that THC was maintained at low levels. Surprisingly, a temporal decrease followed by restoration of THC was observed in hosts injected with virus-like particles of M. pulchricornis (MpVLPs) plus venom, which contrasts with the constant THC suppression seen in parasitized hosts. This indicates that MpVLP function is temporal and is involved in regulation of the host during early parasitism. Therefore, teratocytes, a host regulation factor in late parasitism, could be involved in keeping THC at a low level.

Microplitis croceipes teratocytes placed into nonparasitized Heliothis virescens larvae survived in the absence of a parasitoid larva and caused developmental changes in the host. Expressions of these changes included delayed larval mortality, incomplete larval-pupal ecdysis, or delayed pupation. Two day old 4th stadium H. virescens larvae were more sensitive to injected teratocytes than were 5th stadium larvae. Three day old teratocytes were more effective than were 6 day old teratocytes. The degree of response was related to the number of injected teratocytes. For example, 750 three day old teratocytes (the approximate number from a single parasitoid egg) caused delayed larval mortality in 96% of the treated larvae whereas 175 three day old teratocytes caused delayed larval mortality in only 33% of the treated larvae. Even a dose of 80 teratocytes resulted in 15% incomplete larval-pupal ecdysis compared to 0% for controls. Treatment with hemocyte-and teratocyte-free hemolymph from parasitized larvae, hemocytes from nonparasitized H. virescens, unfertilized M. croceipes eggs, Cotesia congregata teratocytes, or Micrococcus lysodeikticus cells all had very little effect either on larval growth or development time.

Abstract.  In some parasitoid species, the serosa membrane breaks apart at hatching and produces teratocyte cells that assume various functions (immunossupression, secretion and nutrition) mediating host–parasitoid relationships. Teratocyte growth pattern may thus reflect the host suitability for a parasitoid. The teratocyte growth pattern (increase in size and number of teratocytes as a function of time) is studied and used as an indirect measure of fitness to compare the development of the endoparasitoid Dinocampus coccinellae in a marginal host, the coccinellid Harmonia axyridis , and in a suitable host, Coleomegilla maculata . Indirect measures of fitness recorded in both host species confirm that C. maculata is a suitable host for D. coccinellae contrary to the marginal host H. axyridis. According to regression analysis, teratocyte numbers decrease linearly whereas teratocyte size increases linearly with time in the suitable host C. maculata (larvae or adults). In the marginal host, parasitism occurs only in the larval stage where a delay in the parasitoid larval development is observed. Increase in teratocyte size is also highly variable. The teratocyte growth pattern of the parasitoid in the marginal host does not follow the linear model found in the suitable host. Teratocyte growth pattern may be a useful criterion to evaluate host-suitability and host range of parasitoids.

Second instar larvae of Lymantria dispar were parasitized or injected with parasitoid-derived factors such as venom, calyx fluid or parasitoid eggs from Glyptapanteles porthetriae . Growth and development of the host larvae were affected in all different groups compared to control larvae of the same age, injected with Ringer solution. The greatest impact on host growth and on the duration of the 3rd instar was caused by injecting parasitoid eggs. Treated larvae showed melanized capsules or nodules in the hemocoel. While the wasp age had no effect on parasitization efficiency or on the percentage of melanized particles in the hemocoel, significantly more encapsulations were found in larvae parasitized by old wasps as opposed to young wasps. Superparasitization (double or quadruple oviposition) increased the parasitization efficiency markedly. While none of the control larvae showed melanized particles, in the groups of single and superparasitized (2× and 4×) hosts a high percentage of melanized particles (capsules and nodules) occurred.

Two states of parasitization in the Pseudaletia separata-Cotesia kariyai system were examined: one that was lightly parasitized and one that was heavily parasitized. We predicted that the consumption of fat body and hemolymph nutrients depends on the number of parasitoid larvae in the host. Lightly parasitized hosts (average clutch size+/-S.E.: 42.5+/-16.2, N=15) and heavily parasitized hosts (average clutch size+/-S.E.: 230.2+/-8.8, N=15) were prepared artificially. Eight days after parasitization, perivisceral fat body was depleted in the heavily parasitized host, although peripheral fat body was not yet consumed, but by day 10 most of the peripheral fat body was consumed. In lightly parasitized hosts, perivisceral fat body was not consumed by day 10. The parasitoid larvae deplete the perivisceral fat body first and then consume the peripheral fat body in the heavily parasitized host. The amount of trehalose, the major carbohydrate in the hemolymph, was related to the number of parasitoid larvae developing in the host. In a heavily parasitized host, trehalose concentrations remained low. However, in lightly parasitized hosts, the amount of trehalose increased 8 days after parasitization and then decreased by day 10. Protein and total lipid concentrations in the hemolymph of the heavily parasitized host were significantly lower than in lightly parasitized host on day 10, suggesting that the large number of parasitoid larvae depleted the fat body and hemolymph nutrients by day 10. High concentrations of total lipid on day 8 and 10 in lightly parasitized hosts and on day 8 in heavily parasitized host are likely to be attributed to the teratocytes.

The developmental interactions between the gregarious endoparasitoidApanteles ruficrus Hal. and the army worm,Leucania separata Walker were investigated. The parasitoid preferred young host larvae and developed in 9.5 days irrespective of host age at the time of parasitization. The growth of parasitized host larvae were depressed. The net maximum weight of the host larva was positively correlated with the number of parasitoid eggs laid when the 2nd instar was parasitized. And when parasitizing in 2nd instar, the weight of parasitoid was negatively correlated with the number of eggs laid. The parasitoid has an ability to regulate the size of the host and the parasitoid itself according to the number of eggs laid when the host larva is very small.

Adult size, longevity, egg load dynamics and oviposition ofMicroplitis rufiventris Kok. which began their development in the first, second, third (preferred hosts) or fourth (non-preferred hosts) instar larvae of Spodoptera littoralis (Boisd.) were studied. The parasitoid size was largely determined by the initial host size at parasitism. Non-ovipositing females derived from older hosts lived for longer periods than those derived from younger ones. However, the ovipositing females, irrespective of their size, lived for almost the same periods. At emergence, the oviducts of adult females contain a significant amount of mature eggs available for oviposition for a few hours on eclosion day. Egg load increases during the early phase of adult life. The amount of additional mature eggs and rate of egg maturation per hour was greater for wasps derived from preferred hosts compared with those in females derived from non-preferred hosts. The pattern of egg production in M. rufiventris females depended on the availability of hosts for parasitization. Host-deprived females depleted the egg complement with aging; the longer the host deprivation, the lower the oviduct egg load. Marked reduction in both realized or potential fecundity of host-deprived females was observed following host availability. Host privation for more than 3 days induced a marked deficit fecundity pattern through the female' s life. The realized fecundity was determined by the interaction among host availability, the number of eggs that are matured over the female' s life span, oviposition rate and host size from which the female was derived. These results suggest that: (i) M. rufiventris wasp is a weak synovigenic species; (ii) the maturation of additional eggs is inhibited once the maximum oviduct egg load is reached; (iii) the egg load of the newly emerged female is significantly less than the realized fecundity; and (iv) because M. rufiventris females oviposit fewer eggs when they begin depleting their egg supply at 3 days, augmentative releases will require release immediately following emergence to ensure the highest parasitization rate in the field.

A laboratory study investigated development of teratocytes derived from the parasitoid Microctonus aethiopoides Loan in the natural host, Sitona discoideus Gyllenhal, and in three novel hosts, the introduced weed biological control agent Rhinocyllus conicus (Froehlich), and two New Zealand native species Nicaeana cervina Broun and Irenimus stolidus Broun. Weevils were exposed to parasitoids and then examined 6, 10 and 15 days post-parasitism for parasitoid stage and size, and teratocyte number and size. In all hosts, teratocyte numbers decreased and size increased as parasitoid development progressed, although 6 days after parasitism, fewer, larger teratocytes were found in I. stolidus than S. discoideus or N. cervina. In weevils containing second-third instar parasitoid larvae, the most permissive hosts, S. discoideus and N. cervina contained more teratocytes than the least permissive hosts I. stolidus and R. conicus. Host gender influenced some aspects of parasitoid and teratocyte development. Total teratocyte volume was greater in female than male S. discoideus at all sampling times, and at 10 days post-parasitism in N. cervina. A possible relationship between host suitability and teratocyte development is discussed.

Females of insect endoparasitoids inject substances along with the egg at oviposition, which can regulate or induce changes in a number of host physiological processes to benefit the developing parasitoid. These changes can be caused by substances such as venoms, calyx fluids, or symbiotic‐associated virus particles (polydnavirus), which are injected by females along with their eggs, and by substances secreted by parasitoid‐derived tissues (teratocytes) or the developing parasitoid larvae. Teratocytes (dissociated cells of the serosal membrane after parasitoid eclosion) release substances that have roles (i) in parasitoid nutrition, (ii) in the digestion of host tissues, and (iii) in the regulation of host development. Teratocytes of Toxoneuron nigriceps (Viereck) (Hymenoptera: Braconidae) have been implicated in the arrestment of the host development and in the regulation of circulating levels of host ecdysteroids. Here we describe the cDNA of a teratocyte‐secreted chitinase and its expression during parasitoid development, and discuss its putative role in this host–parasitoid association.

Adaptiveness of sex ratio control by the solitary parasitoid wasp Itoplectis naranyae (Hymenoptera: Ichneumonidae) in response to host size was studied, by examining whether differential effects of host size on the fitness of resulting wasps are to be found between males and females. The offspring sex ratio (male ratio) decreased with increasing host size. Larger hosts yielded larger wasps. Male larvae were less efficient in consuming larger hosts than female larvae. No significant interaction in development time was found between parasitoid sex and host size. Larger female wasps lived longer than smaller females, while longevity of male wasps did not increase with increasing wasp size. Smaller males were able to mate either with small or with large females, while larger males failed to mate with small females. Larger female wasps had a greater number of ovarioles and mature eggs at any one time than smaller females, although the number of eggs produced per host-feeding was not influenced by female wasps. Thus, the differential effect of host size on the fitness of males and females exists in I. naranyae. The basic assumption of the host-size model was therefore satisfied, demonstrating that sex ratio control by I. naranyae in response to host size is adaptive.
