The success of bioaugmentation processes for groundwater bioremediation requires efficient transport of bacteria in the subsurface environment. In this paper, the factors that influence transport of bacterial cells in porous media are reviewed and the effects of surfactants on the transport are discussed. Movement of bacterial cells in porous media is a process driven by advection and hydrodynamic dispersion forces of fluids. Immobilization of bacterial cells takes place due to processes such as adsorption and straining. Blocking and ripening along with bacterial migration process decrease and increase the retention of cells in porous media, respectively. Physicochemical properties of the porous media, groundwater chemistry, and properties of the bacterial cells affect the transport behavior. Surfactants have the potential to modify bacterial surface properties for both bacterial cells and medium solids, and thus enhance bacterial transport.  相似文献

As a result of the recently proposed mandatory groundwater disinfection requirements to inactivate viruses in potable water supplies, there has been increasing interest in virus fate and transport in the subsurface. Several models have been developed to predict the fate of viruses in groundwater, but few include transport in the unsaturated zone and all require a constant virus inactivation rate. These are serious limitations in the models, as it has been well documented that considerable virus removal occurs in the unsaturated zone and that the inactivation rate of viruses is dependent on environmental conditions. The purpose of this research was to develop a predictive model of virus fate and transport in unsaturated soils that allows the virus inactivation rate to vary on the basis of changes in soil temperature. The model was developed on the basis of the law of mass conservation of a contaminant in porous media and couples the flows of water, viruses, and heat through the soil. Model predictions were compared with measured data of virus transport in laboratory column studies and, with the exception of one point, were within the 95% confidence limits of the measured concentrations. The model should be a useful tool for anyone wishing to estimate the number of viruses entering groundwater after traveling through the soil from a contamination source. In addition, model simulations were performed to identify parameters that have a large effect on the results. This information can be used to help design experiments so that important variables are measured accurately.  相似文献

A systematic examination of cases on file with the Massachusetts Department of Environmental Protection was undertaken to identify a universe of sites with volatile organic compound (VOC) contamination in groundwater in close proximity to buildings. Such locations were grouped according to site variables, such as contaminants of concern and concentrations in various media; soil type; depth to groundwater; distance to building; and building construction. Indoor air, soil gas, and/or groundwater field data collected from these sites was then assembled and used to: (1) evaluate available transport models which describe the intrusion of vapors into buildings and predict indoor air contaminant concentrations resulting from the volatilization of VOC s in the subsurface; (2) examine the validity of established regulatory criteria; (3) identify specific trends and field conditions which appear to most influence vapor phase contaminant migration and intrusion processes; and (4) evaluate the possibility of vapor migration being inhibited by a "freshwater lens".  相似文献

In this article, a new mechanism influencing the transport of microorganisms through unsaturated porous media is examined, and a new method for directly visualizing bacterial behavior within a porous medium under controlled chemical and flow conditions is introduced. Resting cells of hydrophilic and relatively hydrophobic bacterial strains isolated from groundwater were used as model microorganisms. The degree of hydrophobicity was determined by contact-angle measurements. Glass micromodels allowed the direct observation of bacterial behavior on a pore scale, and three types of sand columns with different gas saturations provided quantitative measurements of the observed phenomena on a porous medium scale. The reproducibility of each break-through curve was established in three to five repeated experiments. The data collected from the column experiments can be explained by phenomena directly observed in the micromodel experiments. The retention rate of bacteria is proportional to the gas saturation in porous media because of the preferential sorption of bacteria onto the gas-water interface over the solid-water interface. The degree of sorption is controlled mainly by cell surface hydrophobicity under the simulated groundwater conditions because of hydrophobic forces between the organisms and the interfaces. The sorption onto the gas-water interface is essentially irreversible because of capillary forces. This preferential and irreversible sorption at the gas-water interface strongly influences the movement and spatial distribution of microorganisms.  相似文献

Bacterial chemotaxis can enhance the bioremediation of contaminants in aqueous and subsurface environments if the contaminant is a chemoattractant that the bacteria degrade. The process can be promoted by traveling bands of chemotactic bacteria that form due to metabolism-generated gradients in chemoattractant concentration. We developed a multiple-relaxation-time (MRT) lattice-Boltzmann method (LBM) to model chemotaxis, because LBMs are well suited to model reactive transport in the complex geometries that are typical for subsurface porous media. This MRT-LBM can attain a better numerical stability than its corresponding single-relaxation-time LBM. We performed simulations to investigate the effects of substrate diffusion, initial bacterial concentration, and hydrodynamic dispersion on the formation, shape, and propagation of bacterial bands. Band formation requires a sufficiently high initial number of bacteria and a small substrate diffusion coefficient. Uniform flow does not affect the bands while shear flow does. Bacterial bands can move both upstream and downstream when the flow velocity is small. However, the bands disappear once the velocity becomes too large due to hydrodynamic dispersion. Generally bands can only be observed if the dimensionless ratio between the chemotactic sensitivity coefficient and the effective diffusion coefficient of the bacteria exceeds a critical value, that is, when the biased movement due to chemotaxis overcomes the diffusion-like movement due to the random motility and hydrodynamic dispersion.  相似文献

Vanadium, a hazardous pollutant, has been frequently detected in soil and groundwater, however, its transport behavior in porous media were not clearly understood. In this study, the effects of solution pH, ionic strength (IS) and the effect of clay mineral on the transport of vanadium in saturated porous media were investigated. Laboratory experiments using a series of columns packed with quartz sand were carried out to explore the retention and transport of vanadium with a range of ionic-strength (0.001–0.1 M) and pH (4–8) and two different types of clay minerals montmorillonite and kaolinite. Results of the breakthrough experiments showed that vanadium was highly mobile in the saturated porous media. The increase in pH rendered a higher transport of vanadium in saturated porous media. The study also indicated an easier transfer of vanadium with an increase in IS. Montmorillonite enhanced the mobility of vanadium in the column when compared to kaolinite. A mathematical model based on advection-dispersion equation coupled with equilibrium and kinetic reactions was used to describe the retention and transport of vanadium in the columns very well.  相似文献

Column experiments were conducted to evaluate the effect of pore velocity on the extent of biodegradation of cis-dichloroethene (cis-DCE) during transport in porous media. Columns were filled with homogeneous glass beads and inoculated with a culture capable of complete dechlorination of tetrachloroethene to ethene. A constant concentration of cis-DCE was maintained in the columns’ influent. Three different pore velocities were tested in duplicate, subjecting each column to a constant velocity. At high flow velocity, degradation of cis-DCE to ethene was nearly complete within the residence time of the columns. However, at medium and low flow velocities, incomplete dechlorination was observed. After 7 weeks, DNA was harvested from the columns to determine differences in the microbial populations. Results suggest that Dehalococcoides sp. were present in higher quantities in the high-velocity columns, consistent with the observed dechlorination. These results suggest that, at contaminated groundwater sites, heterogeneity of groundwater velocity may be one factor that contributes to heterogeneous distribution of biological activity.  相似文献

Many studies have been performed to accelerate osteoinduction and osteoconduction into porous ceramic scaffolds by seeding them with cells. In this study, we compared available cell-seeding methods on a porous β-tricalcium phosphate (β-TCP) scaffold and evaluated the effects of cell-seeding on the mechanical properties of the porous β-TCP scaffold. Three types of porous bioceramic scaffolds were used: dry scaffold, scaffold wetted with media, and scaffold cultivated with normal human osteoblasts (NHOs). Cell-seeding into the porous β-TCP scaffolds was performed by conventional, centrifuge, high-density, and vacuum methods. After confirming cell proliferation with MTT assay and cell staining, a compressive test was performed after 2 and 4 weeks of cell culture. The vacuum method based on the high-density cell culture inserted effectively NHOs into the β-TCP scaffolds. The compressive elastic modulus of wetted β-TCP scaffolds decreased significantly (p < 0.05) about 20∼30% after 2 and 4 weeks of incubation in comparison with that of the dry scaffold. However, the compressive strength of the scaffolds cultivated with NHOs for 3 weeks was significantly (p < 0.05) higher than that of scaffolds without NHOs. The vacuum with the high-density of cell-seeding seems to be a suitable method for seeding cells into complex porous ceramic scaffolds. Cell proliferation and uniform distribution in the scaffolds can change the initial mechanical properties of porous ceramic scaffolds.  相似文献

An integrated simulation-assessment modeling approach for analyzing environmental risks of groundwater contamination is proposed in this paper. It incorporates an analytical groundwater solute transport model, an exposure dose model, and a fuzzy risk assessment model within a general framework. The transport model is used for predicting contaminant concentrations in subsurface, and the exposure dose model is used for calculating contaminant ingestion during the exposure period under given exposure pathways. Both models are solved through the Monte Carlo simulation technique to reflect the associated uncertainties. Based on consideration of fuzzy relationships between exposure doses and cancer risks, risk levels of different exposure doses for each contaminant can be calculated to form a fuzzy relation matrix. The overall risks can then be quantified through further fuzzy synthesizing operations. Thus, probabilistic quantification of different risk levels (possibilities) can be realized. Results of the case study indicate that environmental risks at the waste landfill site can be effectively analyzed through the developed methodology. They are useful for supporting the related risk-management and remediation decisions.  相似文献

In this study, we investigated the treatability of co-mingled groundwater contaminated with polycyclic aromatic hydrocarbons (PAHs), gasoline hydrocarbons, and methyl tert-butyl ether (MtBE) using an ex-situ aerobic biotreatment system. The PAHs of interest were naphthalene, methyl-naphthalene, acenaphthene, acenaphthylene, and carbazole. The gasoline hydrocarbons included benzene, toluene, ethyl benzene, and p-xylene (BTEX). Two porous pot reactors were operated for a period of 10 months under the same influent contaminant concentrations. The contaminated groundwater was introduced into the reactors at a flow rate of 4 and 9 l/day, resulting in a hydraulic retention time (HRT) of 32 and 15 h, respectively. In both reactors, high removal efficiencies were achieved for the PAHs (>99%), BTEX and MtBE (>99.7%). All the PAHs of interest and the four BTEX compounds were detected at concentrations less than 1 μg/l throughout the study duration. Effluent MtBE from both reactors was observed at higher levels; nevertheless, its concentration was lower than the 5 μg/l Drinking Water Advisory for MtBE implemented in California.  相似文献

The Savannah River Site (SRS) is a 780 km2 U.S. Department of Energy facility near Aiken, South Carolina, established in 1950 to produce nuclear materials. SRS streams are “integrators” that potentially receive water transportable contaminants from all sources within their drainage basins, necessitating a watershed approach to organize contaminant distribution data and characterize the effects of multiple contaminants on aquatic organisms. This study used several lines-of-evidence to assess the ecological effects of metals in SRS streams, including contaminant exposure models for apex predators and bioassessments of fish and macroinvertebrate assemblages. Concentrations of metals in sediments, fish, and water were elevated in streams affected by SRS operations, but contaminant exposure models for the river otter Lontra Canadensis and belted kingfisher Ceryle alcyon indicated that toxicological reference values were exceeded only by Hg and Al. Macroinvertebrate assemblage structure was unrelated to sediment metal concentrations. Fish assemblage data were inconclusive. This study indicated that (1) modeling studies and field bioassessments provide a complementary basis for addressing the individual and cumulative effects of contaminants, (2) habitat effects must be controlled when assessing contaminant impacts, (3) sensitivity analyses of contaminant exposure models can help to apportion sampling effort, and (4) most individual metals in SRS streams are unlikely to have significant ecological effects.  相似文献

微生物在地下水和土壤环境中的迁移与地下水资源保护、地下水污染修复及土壤污染防治等息息相关。自然界中多孔介质具有结构复杂性和空间异质性。这导致微生物在其中的迁移易受多重环境因素的影响。本文总结了几种典型多孔介质中微生物迁移模型、理论与研究方法,并对多孔介质中影响微生物迁移行为的3种因素——物理、化学和生物因素进行了梳理。其中物理因素的影响主要包括多孔介质的粒径、表面粗糙度、饱和度、环境温度、水体流速等相关;化学因素主要包含流体pH、离子种类与强度、可溶性有机物含量、多孔介质自身化学性质等;生物因素不但涉及微生物种类、细胞大小和细胞表面特性,还与胞外聚合物的分泌、鞭毛运动及趋化性等相关。本综述旨在总结近年来有关微生物在多孔介质中迁移的相关研究,深入理解微生物在多孔介质中的迁移行为,为其在地下水和土壤污染修复中的实际应用提供理论依据。  相似文献

In this article we propose models and a numerical method for pattern formation on evolving curved surfaces. We formulate reaction-diffusion equations on evolving surfaces using the material transport formula, surface gradients and diffusive conservation laws. The evolution of the surface is defined by a material surface velocity. The numerical method is based on the evolving surface finite element method. The key idea is based on the approximation of Γ by a triangulated surface Γ h consisting of a union of triangles with vertices on Γ. A finite element space of functions is then defined by taking the continuous functions on Γ h which are linear affine on each simplex of the polygonal surface. To demonstrate the capability, flexibility, versatility and generality of our methodology we present results for uniform isotropic growth as well as anisotropic growth of the evolution surfaces and growth coupled to the solution of the reaction-diffusion system. The surface finite element method provides a robust numerical method for solving partial differential systems on continuously evolving domains and surfaces with numerous applications in developmental biology, tumour growth and cell movement and deformation.  相似文献

 The urine concentrating mechanism of mammals and birds depends on a counterflow configuration of thousands of nearly parallel tubules in the medulla of the kidney. Along the course of a renal tubule, cell type may change abruptly, resulting in abrupt changes in the physical characteristics and transmural transport properties of the tubule. A mathematical model that faithfully represents these abrupt changes will have jump discontinuities in model parameters. Without proper treatment, such discontinuities may cause unrealistic transmural fluxes and introduce suboptimal spatial convergence in the numerical solution to the model equations. In this study, we show how to treat discontinuous parameters in the context of a previously developed numerical method that is based on the semi-Lagrangian semi-implicit method and Newton's method. The numerical solutions have physically plausible fluxes at the discontinuities and the solutions converge at second order, as is appropriate for the method. This study presents a method for identifying cost effective sampling designs for long-term monitoring of remediation of groundwater over multiple monitoring periods under uncertain flow conditions. A contaminant transport model is used to simulate plume migration under many equally likely stochastic hydraulic conductivity fields and provides representative samples of contaminant concentrations. Monitoring costs are minimized under a constraint to meet an acceptable level of error in the estimation of total mass for multiple contaminants simultaneously over many equiprobable realizations of hydraulic conductivity field. A new myopic heuristic algorithm (MS-ER) that combines a new error-reducing search neighborhood is developed to solve the optimization problem. A simulated annealing algorithm using the error-reducing neighborhood (SA-ER) and a genetic algorithm (GA) are also considered for solving the optimization problem. The method is applied to a hypothetical aquifer where enhanced anaerobic bioremediation of four toxic chlorinated ethene species is modeled using a complex contaminant transport model. The MS-ER algorithm consistently performed better in multiple trials of each algorithm when compared to SA-ER and GA. The best design of MS-ER algorithm produced a savings of nearly 25% in project cost over a conservative sampling plan that uses all possible locations and samples.  相似文献

The impact of bacterial chemotaxis on in situ ground-water bioremediation remains an unanswered question. Although bacteria respond to chemical gradients in aqueous environments and under no-flow conditions, it is unclear whether they can also respond in porous media with advective flow to improve overall contaminant degradation. The effect of chemotaxis is most profound in regions with sharp chemical gradients, most notably around residual nonaqueous phase liquid (NAPL) ganglia and surrounding clay lenses or aquitards with trapped contamination. The purpose of this study is to simulate bacterial transport through a two-dimensional subsurface environment, containing one region of low permeability with trapped contaminant surrounded above and below by two regions of higher permeability. Using mathematical predictions of the effect of pore size on measured bacterial transport parameters, the authors observe a 50% decrease in both motility and chemotaxis in the finer-grained, low-permeability porous medium. The authors simulate how chemotaxis affects bacterial migration to the contaminated region under various flow and initial conditions. Results indicate that bacteria traveling through a high-permeability region with advective flow can successfully migrate toward and accumulate around a contaminant diffusing from a lower permeability region.  相似文献
